Изобретение относится к области биотехнологии. Штамм микроводоросли Chlorella sp. VADA 2020 выделен из образцов воды озера Тамаркуль Чановского района Новосибирской области и депонирован в Коллекции микроводорослей ИФР РАН под регистрационным номером IPPAS С-2048. Для культивирования штамма применяют доступные питательные среды. Изобретение позволяет получить биомассу с высоким содержанием белка и ненасыщенных жиров, пригодную для использования в пищевых целях. А именно продуцирует биомассу с содержанием белка 48,1% (мас.) и с содержанием ненасыщенных и мононенасыщенных жирных кислот в количестве 59,1% (от общего содержания) может быть использован в качестве сырья для производства пищевых добавок.

ШТАММ МИКРОВОДОРОСЛИ CHLORELLA SP. VADA 2020
Изобретение относится к микробиологии и биотехнологии и представляет собой новый штамм одноклеточной зеленой микроводоросли Chlorella sp. VADA 2020, предназначенный для получения биомассы, пригодной для использования в пищевых целях.
Микроводоросли являются признанным перспективным источником биомассы с высоким содержанием ценных химических веществ, пригодных к использованию в пищевой промышленности. Биомасса микроводорослей может содержать значительное количество липидов (с полиненасыщенными w-З и w-б кислотами), белков, хлорофилла, каротиноидов, витаминов, минералов, а также пробиотических соединений, улучшающих здоровье (Hayes М. et al. , Microalgal proteins for feed, food and health. // Microalgae-based biofuels and bioproducts: from feedstock cultivation to end-product // 2017. p. 347- 368). Однако не все штаммы обладают оптимальным химическим составом и способны давать высокий выход биомассы в условиях лабораторного и промышленного культивирования. Другой проблемой при культивировании является получение чистой биомассы и возможность ее длительного хранения в суспензии без загрязнения побочной микрофлорой.
Известен штамм одноклеточной зеленой микроводоросли Chlorella vulgaris ИФР N° С - 111 , предназначенный для получения биомассы и отвечающий требованиям промышленного культивирования. Клеточная оболочка состоит из внутреннего и внешнего слоев, причем внешний слой содержит обширный миофибриллярный материал (патент RU 1751981 , опубл. 10.02.1997).
Недостатками штамма Chlorella vulgaris ИФР N° С - 111 являются стойкий асинхронный цикл развития клеток, сезонность в развитии (с мая по декабрь), требовательность к составу питательной среды, прочная и сравнительно толстая клеточная оболочка, а также узкий спектр температуры культивирования (26-36°С).
Известен типовой штамм 132-1 одноклеточной зеленой микроводоросли Chlorella vulgaris f. Suboblonga, имеющий следующие
характеристики. Клетки эллипсоидные, 2,8-6, 1 мкм длиной и 1 ,7-5, 5 мкм шириной, при образовании автоспор 8,0 и 5,5 мкм соответственно. Оболочка тонкая. Хлоропласт широкий, поясковидный, незамкнутый или корытовидный. Положение хлоропласта может быть ориентировано по отношению к длинной оси клетки продольно или поперечно. Автоспоры по 2- 4, реже по 8, сразу после освобождения неправильно эллипсоидные. Типовой штамм 132-1 хранится в Ботаническом институте РАН и является музейным образцом (Андреева В.М. Род Chlorella. Морфология, систематика, принципы классификации. Л.: «Наука», Ленингр. отд., 1975. - 110 с.).
Известен штамм одноклеточной зеленой микроводоросли Chlorella vulgaris N А-29, предназначенный для получения биомассы, при культивировании которого на среде Тамийя при 42°С получают 6,38 г/л сухой биомассы. Штамм водоросли Chlorella vulgaris N получен методом мутационной селекции с использованием нитрозометилмочевины из дикого штамма 132. (Патент RU 2005777 С 1 , опубл. 14.06.1991).
Недостатком штамма Chlorella vulgaris N А-29 является узкий температурный оптимум), приспособленность к условиям только жаркого климата, например Средней Азии и культивирование в специальных аппаратах промышленного типа (стеклотрубных установках).
Наиболее близким к заявляемому штамму - прототипом, является штамм микроводоросли Chlorella vulgaris ВКПМ А1-24, который культивировался на питательной среде с включением в ее состав зерноматериала (голозерного овса) для получения раствора углекислого газа. В результате селекции был отобран штамм, наделенный тонкой оболочкой, способный интенсивно воспроизводить биомассу в синхронном режиме культивирования и поддерживать монокультуру в процессе производства (Патент RU 2644653 С 1 , опубл. 13.02.2018).
Недостатками штамма Chlorella vulgaris ВКПМ А1 -24 является высокий риск присоединения бактериальной, микрофлоры при культивировании с использованием раствора углекислого газа, приготовленного на основе голозерного овса, что ограничивает возможность использования штамма в пищевых целях.
Задачей изобретения является получение нового штамма микроводоросли Chlorella sp . , обладающего высокой скоростью роста в асептических условиях и оптимальным химическим составом биомассы,
пригодной для использования в пищевых целях в виде суспензии или сухого порошка.
Технический результат: получение биомассы с высоким содержанием белка и ненасыщенных жиров, пригодной для использования в пищевых целях.
Поставленная задача решается предлагаемым штаммом микроводоросли Chlorella sp. VADA 2020, обладающим высокой специфической скоростью роста биомассы с высоким содержанием белка и ненасыщенных и мононенасыщенных жирных кислот, пригодной для использования в пищевых целях.
Штамм микроводоросли Chlorella sp. VADA 2020 выделен из образцов воды озера Тамаркуль Чановского района Новосибирской области и депонирован в Коллекции микроводорослей ИФР РАН под регистрационным номером IPPAS С-2048.
Штамм микроводоросли Chlorella sp. VADA 2020 имеет следующую морфо-физиологическую характеристику.
Клетки круглые, шаровидные, с тонкой оболочкой без слизи, размером 3-10 мкм, с одиночным пиреноидом, размером 1 мкм, размножение происходит 2-4 автоспорами. На агаризованной среде ВВМ образует колонии зеленого цвета с ровным краем. Штамм авто- и миксофототрофный. Температура культивирования от 15 до 35°С. Штамм растет на минеральной среде ВВМ, не требует для роста подачи в среду углекислого газа. Достаточный период освещения для роста - до 16-18 ч в сутки. Штамм не склонен к осаждению, как в культуре, так и на стенках культивационной установки, сохраняет аксеничность при хранении. Штамм развивается вне зависимости от сезона года.
Полученный штамм, по сравнению с ранее описанными штаммами, обладает повышенным содержанием белка (48,1 ) и липидов с мононенасыщенными жирными кислотами и полиненасыщенными жирными кислотами. Общее содержание данных жирных кислот, определенных методом газовой хромато-масс-спектрометрии, составляет 59,1%, как показано в таблице 2, что позволяет получать биомассу с высоким содержанием ненасыщенных жиров, что имеет преимущество для использования в пищевых целях.
Изобретение иллюстрируется следующими примерами:
Пример 1
Для культивирования штамма микроводоросли Chlorella sp. VADA 2020 использовали стерилизованный стеклянный панельный фотореактор, размером 800 мм c 312 мм c 500 мм (длинахширинахвысота) с рабочим объемом 100 л. Поступление смеси воздуха с 1 ,5% СОг осуществлялось со скоростью потока 200 л/ч через пористую трубку. Инокуляцию проводили культурой, находящейся в экспоненциальной фазе роста, с начальной концентрацией биомассы микроводорослей 3 млн. клеток/мл. Культивирование проводили на среде ВВМ в течение 5 суток при температуре 30±1°С, освещенности 9850 кЛк. Для оценки ростовых характеристик и состава биомассы в течение культивирования отбирали аликвоты суспензии клеток, затем осаждали клетки центрифугированием при 10000 g в течение 10 мин. Полученную биомассу высушивали до постоянной массы и использовали для анализа химического состава.
В таблице 1 представлены данные ростовых характеристик и состава биомассы штамма микроводоросли Chlorella sp. VADA 2020 при росте на среде ВВМ.
Из таблицы 1 видно, что штамм характеризуется быстрым накоплением биомассы (245,4±8,6 мг л-1 сут-1 ) с высоким содержанием белка (48,1 ±1 ,6%), а также высокой специфической скоростью роста 0,7±0,1 сут-1.
Предлагаемый штамм продуцирует биомассу с содержанием белка, сравнимым с содержанием белка в биомассе, продуцируемой штаммом С. vulgaris (47,82% белка), раскрытом в исследовании (Tokusoglu, О. et al. , Biomass nutrient profiles of three microalgae: Spirulina platensis, Chlorella vulgaris, and Isochrisis galbana // J. of Food Science. - 2003. - V. 68. - N. 4. - P. 1144-1148) и штаммом C. vulgaris с содержанием белка 46%, описанного в работе (Seyfabadi, J. et al. Protein, fatty acid, and pigment content of Chlorella
vulgaris under different light regimes // Journal of Applied Phycology. - 2011. - V. 23. - N. 4. - P. 721-726).
Пример 2
Для определения состава метиловых эфиров жирных кислот использовали метод ГХ-МС. Анализ проводили на приборе Agilent 7000 В с ионизацией электронным ударом (70 эВ), на колонке ZB-WAX (30 м c 0.25 мм х 0.25 мкм), с температурным градиентом от 100°С до 260°С со скоростью повышения 12°С/мин. Температура инжектора была установлена на 260°С. Поток газа-носителя (гелия) составлял 1 ,2 мл/мин. Идентификацию соединений проводили путем сравнения получаемых данных с библиотекой спектров N 1ST' 11. Относительное количество индивидуальных жирных кислот было посчитано интегрированием процентной площади от всех жирных кислот. Эксперименты были выполнены в трех повторностях и данные были представлены как среднее значение ± стандартное отклонение.
В таблице 2 представлен состав жирных кислот микроводоросли Chlorella sp. VADA 2020 при фотоавтотрофном культивировании по данным газовой хромато-масс-спектрометрии.
В таблице 2 использованы следующие сокращения: первое значение после символа «С» - число атомов углерода, второе значение после двоеточия - число двойных связей в молекуле жирной кислоты; НЖК -
насыщенные жирные кислоты, ПНЖК полиненасыщенные жирные кислоты, МНЖК - мононенасыщенные жирные кислоты.
Из таблицы 2 видно, что в составе липидов исследуемого штамма было обнаружено 11 жирных кислот с длиной углеродной цепи от С14 до С20. Основными ненасыщенными жирными кислотами биомассы Chlorella sp. VADA 2020 являются кислоты С 16:2, С 16: 3, С 18: 1 , С 18:2 и С 18:3. Суммарное содержание всех ПНЖК и МНЖК в исследуемом штамме после 5-ти суток культивирования составило от 59,1%. Это значение сравнимо с ранее опубликованными значениями жирных кислот для микроводорослей Chlorella, перспективными для использования в пищевых целях (Matos, et al. , Chemical Characterization of Six Microalgae with Potential Utility for Food Application. J. Am. Oil Chem. Soc, 93: 963-972).
Таким образом, использование предлагаемого штамма Chlorella sp. VADA 2020 позволит получать биомассу с высоким содержанием ненасыщенных жиров, пригодную для использования в пищевых целях.
