Abstract
La presente invención se relaciona con una composición con actividad antimicrobiana que comprende aceite esencial de Ciprés de las Guaitecas (Pilgerodendron uviferum) encapsulado en nanopartículas de quitosano con tripolifosfato de sodio, un gel que la comprende, métodos de producción del aceite esencial, la composición con actividad microbiana y el gel que la comprende y el uso del aceite esencial de Ciprés de las Guaitecas y la composición que la comprende para el tratamiento del acné y la enfermedad periodontal.

COMPOSICIÓN A BASE DE ACEITE DE CIPRÉS DE LAS GUAITECAS
PARA EL TRATAMIENTO DEL ACNÉ Y ENFERMEDAD PERIODONTAL
CAMPO DE LA INVENCIÓN La presente invención se relaciona con la industria del cuidado oral y facial. En particular, la presente invención se refiere a una composición que comprende aceite esencial de Ciprés de las Guaitecas encapsulado en nanopartículas de quitosano con tripolifosfato de sodio para el tratamiento, prevención y control del acné y la enfermedad periodontal.
ESTADO DEL ARTE
La formación de biopelículas bacterianas sobre superficies bióticas ha sido descrita como uno de los principales gatillantes y elicitores de las enfermedades periodontal y acné. La estructura de exopolisacarido laxo, unido débilmente a la pared bacteriana, le permite a los microorganismos patógenos -y potencialmente patógenos- colonizar y comunicarse entre sí generando un microambiente que favorece la expresión de factores de virulencia involucrados con la enfermedad propia, enfermedades sistémicas, así como expresar múltiples genes de resistencia a los antibióticos convencionales. El acné, como familia de trastornos de la piel, es una de las enfermedades dermatológicas más prevalentes en el mundo, afectando a la gran mayoría de la población durante la vida. Se ha determinado que los principales microorganismos involucrados en el desarrollo de la infección corresponden a Propionibacterium acnés y Staphylococcus aureus.
Por su parte, la enfermedad periodontal se relaciona con una enfermedad inducida por patógenos periodontales que se desarrolla en tejidos periodontales, tales como encía, membrana periodontal, cemento y huesos alveolares que soportan los dientes. Los principales microorganismos involucrados en el desarrollo de la infección corresponden a P. gigivalis y A. actinomycetemcomitans como agentes etiológicos y S. gordonii y S. sanguinis como agentes cariogénicos.
Existe una variedad de medicamentos para el acné que incluyen agentes tópicos, antibióticos orales, retinoides orales y terapias hormonales orales. Por su parte, para el tratamiento de la enfermedad periodontal, principalmente se utilizan productos colutorios con agentes antimicrobianos que deben ser utilizados previa o posteriormente al cepillado de dientes.
En ambos casos, la formación de biopelículas disminuye la expectativa de éxito de los tratamientos, generando resistencia a los agentes antimicrobianos existentes y prevaleciendo la infección. Durante la última década, en vista del aumento de la resistencia a antibióticos, los efectos secundarios y, en ocasiones, el alto costo de los distintos tratamientos, existe un interés creciente por el desarrollo de nuevos tratamientos que resulten eficaces en un amplio espectro de microorganismos, que sean bien tolerados por los pacientes y que sean amigables con el medio ambiente. Una de las estrategias más utilizadas y con más proyección en la actualidad es el uso de fitoquímicos bioactivos con acción antimicrobiana. La composición, estructura y grupos funcionales de los aceites
esenciales de las plantas, juegan un papel clave en la determinación de su actividad antimicrobiana.
El Ciprés de las Guaitecas ( PHgerodendron uviferum (D. Don) Florín) corresponde a una planta conifera endémica del sur de Chile y Argentina, su distribución geográfica comprende unos 1600 km entre 39 ° 36' y 54 ° 20'S lo que la convierte en la conifera nativa más austral del mundo. La calidad, durabilidad y resistencia a la pudrición de su madera aromática la ha convertido en una especie de valor comercial en la fabricación de muelles, construcciones navales, postes, pisos y mobiliario. Estudios preliminares han demostrado que la madera del Ciprés de las Guaitecas posee compuestos químicos con actividad antimicrobiana inhibitoria. Solis et al (2014) analizaron los extractos de corteza y madera cruda y determinaron que poseían actividad biológica inhibidora contra diferentes tipos de hongos y bacterias, reconociendo distintos metabolitos secundarios con actividad antimicrobiana.
La aplicación de fitoquímicos bioactivos suele ser limitada debido a su escasa solubilidad, estabilidad y biodisponibilidad. Con el objetivo de solucionar esta problemática, la encapsulación de los compuestos activos se ha convertido en una tecnología atractiva que permite mantener las propiedades fisicoquímicas de los aceites esenciales, evitar cambios indeseados y favorecer su biodisponibilidad.
La gelificación ionotrópica (IG) es una técnica que permite la producción de nanopartículas y micropartículas por interacciones electrostáticas entre dos especies iónicas en determinadas condiciones. Cuando se agrega un compuesto
bioactivo a la reacción, puede quedar atrapado entre las cadenas poliméricas, lo que resulta en ser capturado dentro de la estructura de nanopartículas / micropartículas.
Diversos compuestos han logrado ser encapsulados mediante este método, entre ellos figuran agentes antidiabéticos, drogas antiinflamatorias, antibióticos, proteínas y enzimas, probióticos, metales, antioxidantes, aceites esenciales, entre otros.
El quitosano (Qo) es el derivado N-desacetilado de la quitina siendo éste el polímero natural más abundante luego de la celulosa. Es un copolímero lineal compuesto por cadenas formadas por unidades de 2-acetamido-2-desoxi-D- glucopiranosa (N-acetilglucosamina) y 2-amino-2-desoxi-D-glucopiranosa ( N - glucosamina) unidas por enlaces glicosídicos b(1 4), con un grado de desacetilación no menor al 65%.
Debido a su versatilidad, biocompatibilidad, toxicidad, biodegradación y bioactividad ha encontrado muchas aplicaciones tecnológicas y biomédicas. Los biomateriales a base de quitosano se pueden utilizar en los campos farmacéutico, biomédico y biotecnológico como agente antimicrobiano natural.
Sao Pedro, et al (2009) describe la microencapsulación de compuestos bioactivos tales como aceites esenciales en matrices de quitosano para mejorar su estabilidad y permitir su liberación controlada mejorando así la eficacia de los tratamientos convencionales en enfermedades orales.
Por su parte, los documentos WO2012018519A1 y US2019175956A1 describen productos para el cuidado oral de origen natural tales como pasta de dientes, enjuague bucal, o varas blanqueadoras que incluyen agentes antimicrobianos
que corresponden a aceites esenciales o compuestos derivados de aceites esenciales.
Ninguno de estos documentos describe composiciones que puedan ser utilizadas en el tratamiento de distintas afecciones relacionadas con microorganismos productores de biopelículas, tales como la enfermedad periodontal y el acné, que sean efectivas para todos los agentes etiológicos involucrados y que solucionen los problemas de la técnica previamente descritos.
DESCRIPCIÓN RESUMIDA DE LA INVENCIÓN La presente invención se relaciona con una composición que comprende aceite esencial de Ciprés de las Guaitecas ( Pilgerodendron uviferum) encapsulado en nanopartículas de quitosano con tripolifosfato de sodio y su aplicación cosmética para el tratamiento del acné y la enfermedad periodontal.
En otra realización de la invención, se divulga un método para producir la composición que comprende aceite esencial de Ciprés de las Guaitecas encapsulado en nanopartículas de quitosano obtenido mediante gelificación iónica con tripolifosfato de sodio (TPP).
En otra realización de la invención, se divulga un método de producción de aceite esencial de Ciprés de las Guaitecas. En una realización preferente, la invención comprende una nanosuspensión que comprende aceite esencial de ciprés de las Guaitecas en una concentración de 0,1% a 0,5% p/v, encapsulado en nanopartículas de quitosano con tripolifosfato de sodio y su aplicación en un gel cosmético útil para el tratamiento del acné y
la enfermedad periodontal. De manera preferente, la concentración del aceite de Ciprés de las Guaitecas es de 0,4% p/v obtenido a partir de descartes madereros. La composición de la presente invención posee actividad antibacteriana relevante frente a cepas patógenas de la enfermedad del acné y periodontal.
DESCRIPCIÓN DE LAS FIGURAS

DESCRIPCIÓN DETALLADA DE LA INVENCIÓN
La formación de biopelículas bacterianas sobre superficies bióticas ha sido descrita como uno de los principales gatillantes y elicitores de las enfermedades periodontal y acné.
Esta estructura de exopolisacarido laxo, unido débilmente a la pared bacteriana, le permite a los microorganismos patógenos -y potencialmente patógenos- colonizar y comunicarse entre sí generando un microambiente que favorece la expresión de factores de virulencia involucrado con la enfermedad propia,
enfermedades sistémicas, así como expresar múltiples genes de resistencia a los antibióticos convencionales.
Por tanto, existe una necesidad de generar nuevos y mejorados tratamientos para el control de los microorganismos patógenos y potencialmente patógenos capaces de formar biopelículas bacterianas causantes del acné y la enfermedad periodontal, que sean de fácil producción, amigables con el medio ambiente, antimicrobiano de alto espectro y que sean bien tolerados por los pacientes.
En la presente invención, se ha descubierto que el aceite esencial de Ciprés de las Guaitecas posee acción antimicrobiana bactericida y bacteriostática frente a los distintos microorganismos involucrados en las patologías del acné y la enfermedad periodontal. De manera específica, se ha demostrado que el aceite esencial de Ciprés de las Guaitecas es capaz de inhibir el desarrollo e interrumpir la formación de biopelículas de las cepas Propionibacterium acnés y Staphylococcus aureus (agentes causantes del acné) y P. gigivalis, A. actinomycetemcomitans, S. gordonii y S. sanguinis (agentes causantes de la enfermedad periodontal).
De manera aún más sorprendente, la nanoencapsulación del aceite esencial de Ciprés de las Guaitecas en un sistema que comprende quitosano nanoparticulado aumenta su eficacia antimicrobiana de manera sinérgica, superando los tratamientos actuales para el control, prevención y tratamiento del acné y la enfermedad periodontal.
Se caracterizó el aceite esencial de Ciprés de las Guaitecas mediante cromatografía de gases acoplada a espectrometría de masas utilizando un
equipo GC-MS/MS, GC modelo Trace 1300, MS/MS modelo TSQ 8000Evo y autosampler: Triplus RSH Thermo Scientific,
Se identificaron 32 compuestos que representan el 88% del área relativa y un total de 32 compuestos no se pudieron identificar, los que representan el 12% del área relativa. Se identificaron 11 compuestos con un área relativa superior al 1%, que representan el 77,9% del área relativa, estos compuestos se enumeran a continuación:
El aceite esencial de Ciprés de las Guaitecas ( Pilgerodendron uviferum) difiere significativamente del aceite esencial de Ciprés común ( Cupressus sempervirens), cuya composición química comprende esencialmente a-pineno (40-65%), b-pineno (0,5-3%), delta-3-carene (12-25%), b-mycerene (1 -3,5%),
limonene (1 ,8-5%), terpinene-4-ol (0,2-2%), acétate de terpenyle (1 -4%) y cedrol (0,8-7%).
La presente invención se relaciona con una composición que comprende aceite esencial de Ciprés de las Guaitecas ( Pilgerodendron uviferum) encapsulado en nanopartículas de quitosano con tripolifosfato de sodio y su aplicación cosmética para el tratamiento del acné y la enfermedad periodontal. La composición puede estar en forma de nanosuspensión, gel, nanoemulsión, emulsión, pasta, crema, loción, jalea u otros.
En una realización preferente, la invención comprende una nanosuspensión que comprende aceite esencial de ciprés de las Guaitecas en una concentración de 0,1% a 0,5% p/v, encapsulado en nanopartículas de quitosano con tripolifosfato de sodio y su aplicación en un gel cosmético útil para el tratamiento del acné y la enfermedad periodontal. De manera preferente, la concentración del aceite de Ciprés de las Guaitecas es de 0,4% p/v.
La composición de la presente invención posee actividad antibacteriana relevante frente a cepas patógenas de la enfermedad del acné y periodontal siendo competitivo frente a otros productos actualmente disponibles en el mercado para el tratamiento del acné y para el tratamiento de la enfermedad periodontal.
La composición de la presente invención posee actividad antibacteriana relevante frente a cepas patógenas de la enfermedad del acné y periodontal y el aceite de Ciprés de las Guaitecas presenta buenos parámetros de encapsulación. La eficiencia de encapsulación del aceite en nanosistemas está cercana o por
sobre el 50% y la capacidad de carga es razonable considerando la actividad antimicrobiana del aceite.
La composición que comprende aceite esencial de Ciprés de las Guaitecas encapsulado en nanopatículas de quitosano de bajo peso molecular fabricadas por gelificación iónica con TPP evidencian un potente efecto bactericida y bacteriostático, incrementando la actividad del aceite esencial en estado puro, mostrando un importante efecto sinérgico tanto en la inhibición del crecimiento, proliferación y desarrollo de los principales agentes etiológicos de la enfermedad periodontal y el acné. La composición de la presente invención exhibe una excelente habilidad para inhibir sobre el 90% la formación de la biopelícula de las cepas de Staphylococcus aureus, Propionibacterium acnés, Agregregatibacter actinomycetemcomitans y Porphyromonas gingivalis al aplicar entre 12 y 24 mg/mL del nanoformulado. La composición de la presente invención exhibe una excelente capacidad bacteriostática y bactericida y de inhibición del crecimiento, independiente de la temperatura de almacenamiento y el tiempo de preparación, indicando una adecuada estabilidad de los activos que permiten el control de la proliferación de los patógenos relevantes en el acné y la enfermedad periodontal. Se realizaron estudios de estabilidad acelerado de tres meses a diferentes temperaturas de almacenamiento que demostraron que la composición mantiene sus propiedades antimicrobianas en el tiempo.
De manera preferente, la composición de la invención comprende aceite esencial de ciprés de las guaitecas encapsulado en nanopartículas de quitosano-TPP y al menos un surfactante. De manera preferente, el surfactante utilizado corresponde a Tween 80. En otra realización de la invención, se divulga un método para producir una composición de uso cosmético para el tratamiento del acné y la enfermedad periodontal que comprende los siguientes pasos: a) mezclar aceite esencial de Ciprés de las Guaitecas con surfactante en una relación entre 1 :0,5 a 1 :10 (p/v); b) agregar agua destilada y agitar por un periodo de 12 a 48 horas; c) agregar ácido acético y quitosano; d) en forma paralela, elaborar una solución de tripolifosfato de sodio; e) mezclar ambas soluciones en una relación Qo/Tpp 2:1 (v/v) mediante goteo por medio de una bomba de infusión en agitación constante; f) centrifugar a 21.000 x g por 30 a 60 min y conservar el sobrenadante.
De manera preferente, la composición comprende una relación de aceite/surfactante de 1/5. De manera aún más preferente, el surfactante utilizado corresponde a Tween 80.
De manera preferente, la carga del aceite esencial de Ciprés de las Guaitecas en la composición es de 0,1 a 0,5 mi aceite / 100 mi solución de quitosano. De manera aún más preferente, la carga del aceite es de 0,4 mi de aceite / 100 mi solución de quitosano.
De manera preferente el quitosano utilizado corresponde al de bajo peso molecular.
El método reivindicado permite obtener una composición que posee una potente actividad antimicrobiana frente a los agentes etiológicos de la enfermedad periodontal y del acné. En ambos casos se observa una actividad bactericida sinérgica entre la matriz nanoparticulada del quitosano y el aceite esencial.
El aceite esencial de Ciprés de las Guaitecas puede ser obtenido a partir de plantas vivas y ejemplares muertos, los que pueden ser remanentes de incendios pasados. Adicionalmente, puede obtenerse el aceite de ciprés a partir de descartes madereros y biomasa residual del procesamiento de la madera
El aceite esencial se obtiene mediante destilación por arrastre con vapor de agua. El proceso de obtención del aceite esencial comprende las siguientes etapas: a) agregar viruta de tronco de Ciprés de las Guaitecas y agua destilada en un destilador; b) calentar el caldero por un periodo de 1 -10 horas; c) condensar los vapores generados en un tubo de decantación; d) decantar la composición hasta que el aceite se separe del agua.
En una realización de la invención, la composición que comprende aceite esencial de Ciprés de las Guaitecas ( Pilgerodendron uviferum) encapsulado en nanopartículas de quitosano con tripolifosfato de sodio se utiliza en la preparación de un gel de uso cosmético con actividad antimicrobiana.
En otra realización de la invención, se describe un proceso para producir el gel que comprende la composición. El proceso para producir un gel de uso cosmético con actividad antimicrobiana comprende las siguientes etapas: a) disolver 100 mi de la composición que comprende aceite esencial de Ciprés de las Guaitecas ( Pilgerodendron uviferum) encapsulado en nanopartículas de quitosano con tripolifosfato de sodio y 2 g de HPMC K15 a 2500 rpm; b) dejar reposar por 12-24 horas a 5°C.
En otra realización de la invención, se describe el uso del aceite esencial de Ciprés de las Guaitecas para preparar una composición útil para el tratamiento del acné y enfermedad periodontal.
EJEMPLOS DE APLICACIÓN
EJEMPLO 1. Encapsulación de aceite esencial de ciprés en Q-TPP Se mezcló aceite de ciprés de las guaitecas con Tween 80 en las siguientes relaciones 1 :0.5 - 1 :1 - 1 :5 - 1 :10 (v/), se agregó al agua destilada y se dejó agitando de un día para otro, luego se agregó ácido acético al 1% y quitosano al 0,3% p/v. En forma paralela se elaboró una solución de tripolifosfato de sodio al 0,1% p/v. Las soluciones se mezclaron en una relación Qo/Tpp 2:1 (v/v) mediante goteo (1 ,8 mL/min) por medio de una bomba de infusión en agitación constante. La dispersión obtenida se centrifugó a 21.000 x g a 14°C por 30 min y se conservó el sobrenadante.
Se estudió el efecto de la carga de aceite esencial de ciprés en el nanosistema de 0,1 a 0,5 mi aceite /100 mL solución de quitosano. El efecto en el tamaño, PDI, y potencial Z se muestra en la tabla siguiente: Tabla 1 : Tamaño, PDI y potencial Z a distintas concentraciones de aceite esencial en el nanosistema
Se observa que con el incremento de aceite se reduce el tamaño hidrodinámico y aumenta la polidispersidad, el potencial Z no se afecta significativamente.
EJEMPLO 2. Determinación de la concentración inhibitoria mínima (CIM) y la concentración bactericida mínima (CBM) de los aceites esenciales y sus formulaciones. Para el ensayo de la CIM, cada cepa bacteriana fue inoculada en medios de cultivos y disponibilidad de oxígeno (aerobiosis o anaerobiosis) selectivos para su crecimiento hasta alcanzar una Oϋboo de 1.745 ± 0.122, equivalente a 1x105 UFC/mL. Posteriormente, cada cultivo fue diluido hasta obtener una Oϋboo de 0.05 nm en donde fueron cultivados en un volumen final de 400 pL utilizando placas de poliestireno de 96 pocilios en donde las cepas ensayadas fueron enfrentadas a concentraciones crecientes de cada activo y en ausencia del activo (control). Luego de 24 h de incubación, la OD fue medida utilizando el lector de placas TECAN Infinite PRO m200. La CIM fue determinada por cuantificación de la OÜ6oo luego de la incubación con los activos y se estableció como la concentración más baja de los activos que evidencio Oϋboo similar al caldo de cultivo sin inoculo bacteriano (no turbiedad).
La CBM fue determinada mediante recuento de células viables (colonias) en placas de agar (UFC/mL) utilizando la misma metodología de incubación que la CIM. Brevemente, un volumen de 1000 pL expuesto a los activos fue inoculado en 9,0 mL de suero fisiológico (NaCI 0,9% p/v) estéril en donde diluciones seriadas fueron realizadas hasta 100000 veces (10-5). 1 mL de cultivos fueron sembrados en placas de agar TSA y fueron incubadas a 37°C durante 24 h. Luego de la incubación, se realizó el recuento de colonias de cada cepa expuesta a los activos. Los resultados fueron expresados como UFC/mL. La CBM fue
determinada como la concentración que es capaz de eliminar el 99.9% respecto al inoculo inicial (control de viabilidad). Cada ensayo se realizó en quintuplicado tres veces en forma independiente. Resultados:

Se observa que todos los extractos analizados poseen actividad inhibitoria frente a los distintos agentes etiológicos tanto del acné como de la enfermedad periodontal.

La tabla 3 muestra los resultados de la concentración bactericida mínima, que corresponde a la concentración del activo requerida para eliminar el 100% de la población bacteriana. Los resultados evidencian que todos los extractos analizados poseen actividad bactericida frente a todas las cepas estudiadas. Actividad antimicrobiana de composición que comprende aceite esencial de ciprés de las Guaitecas encapsulado en nanopartículas de quitosano con TPP: Fueron determinados los parámetros CIM y CBM para las nanopartículas fabricadas por gelificación iónica utilizando quitosano de bajo peso molecular y tripolifosfato de sodio cargadas con el aceite esencial de Ciprés de las Guaitecas (NQoAE) y se utilizó como control y parámetro de comparación el mismo tipo de nanopartículas sin aceite esencial encapsulado (NQo), los resultados de ambos ensayos son mostrados a continuación.
Se determinó la capacidad antimicrobiana de los geles formulados por la adición de diferentes proporciones de aceites esenciales nanoencapsulados en matrices de nanopartículas elaboradas a partir de quitosano de bajo peso molecular y 5 tripolifosfato de sodio (NQoTPP). Fueron ensayadas concentraciones crecientes (desde 0.5 hasta 125 mg/mL) de las diferentes formulaciones elaboradas las cuales contenían 0.2, 0.3, 0.4 y 0.5 % de NQoTPP en su composición. Estos resultados se muestran en la Tabla 5. Se evidencia que la concentración requerida para inhibir el desarrollo de las cepas bacterianas disminuye en función 10 a que incrementa la concentración de activos nanoencapsulados en la matriz del gel, por tanto, se evidencia una actividad antimicrobiana directamente proporcional a la presencia del nanoformulado en la matriz (% p/v).

EJEMPLO 3. Determinación de la capacidad de inhibición de la formación de biofilm de las cepas ensayadas enfrentadas a las bioactivos.
5 Cada cepa ensayada fue inoculada en condiciones óptimas de crecimiento (37°C y disponibilidad de oxígeno según requerimientos metabólicos específicos de los géneros ensayados) hasta alcanzar una fase exponencial temprana (Oϋboo de ~0,4). Una vez alcanzada la densidad óptica deseada, un volumen de 200 pL de cada cepa fue inoculada en placas de poliestireno de 96 pocilios en ausencia 10 (control) y presencia de diferentes concentraciones de los aceites esenciales y sus diferentes formulaciones. Para la exposición de las cepas a los activos, las placas fueron incubadas a 37°C por 24 h. Luego de la incubación, las células en estado planctónico fueron descartadas. Para la fijación del biofilm formado, se agregó metanol seguido de la tinción con cristal violeta (0,2 % p/v). Las bacterias
15 que fueron capaces de adherirse y formar las biopelículas en los pocilios de las
placas de poliestireno, fueron lavadas 3 veces con agua destilada estéril y fueron resuspendidas en ácido acético glacial (33 % v/v) y la densidad óptica (OD570) fue determinada en el lector de placas TECAN Infinite PRO m200, en donde la densidad óptica de las cepas control (no expuestas a los activos) se consideró como el 100 % en la capacidad de formación de biofilm y fue comparada con las OD570 de las cepas expuestas a las diferentes concentraciones de los activos, determinado así la capacidad de inhibición del biofilm. Cada ensayo se realizó en quintuplicado tres veces en forma independiente. Resultados:
Se evaluaron tanto las nanopartículas con AE nanoencapsulado (NQoAE) así como el control NQo, estos resultados en la inhibición del biofilm son mostrados en las tablas 6 y 7 correspondiente a los patógenos de la enfermedad periodontal y acné, respectivamente.

Se observa un potente efecto de inhibición en la formación de biofilm tanto para los patógenos de la enfermedad periodontal como de la enfermedad del acné. EJEMPLO 4: Determinación de la zona de inhibición del crecimiento.
Se realizó un ensayo de difusión en agar para determinar la zona de inhibición del crecimiento de todas las cepas ensayadas. Las placas de agar Müller-Hinton fueron inoculadas con una suspensión bacteriana de 100 pL con ~1 x106 UFC/mL.
Discos blancos fueron cargados con concentraciones determinadas de cada formulación ensayada y dispuesto equidistantes en la superficie del agar
sembrada con los inóculos bacterianos. Las placas fueron incubadas a 37°C por 24h y la zona de inhibición fue estimada por medición del diámetro de las áreas en donde no se evidenció el crecimiento bacteriano. Cada experimento fue realizado en triplicado. Las Figuras 1 y 2, muestran la zona de inhibición del crecimiento (ZIC) de las 2 formulaciones de nanopartículas ensayadas NQo y NQoAE. Para el caso de los patógenos del acné (Fig. 1) se evidencia el potente efecto en la inhibición tanto en S. aureus y P. acnés de la composición que comprende aceite esencial de ciprés de las guaitecas encapsulado en nanopartículas de quitosano con TPP (NQoAE), siendo significativamente superior a la inhibición generada por las nanopartículas control (NQo), las nanoformulaciones con aceite esencial evidenciaron un incremento del 47 a 56% la capacidad de inhibir el desarrollo de los agentes etiológicos estudiados. En el caso de los patógenos de la enfermedad periodontal, las formulaciones analizadas (Fig 2) evidencian un incremento del poder de inhibición de la formulación NQoDP en todas las cepas P. gingivalis, S. sanguinis y Agregregatibacter actinomycetemcomitans, resultando esta formulación entre 20 a 50% más potente que la NQo, ratificando el efecto sinérgico AE y quitosano.
Al comparar la eficiencia del nanoformulado (NQoAE) versús el aceite esencial en estado puro, se evidenció que para las cepas responsables de la enfermedad del acné, utilizando las mismas concentraciones (8 mg) cargadas en el sensidisco existe un significativo incremento de la capacidad inhibitoria del crecimiento . Similar efecto se evidencia en los microorganismos de la enfermedad periodontal.
EJEMPLO 5: Ensayos de estabilidad
Se determinó la CIM y la CBM y la ZIC de la composición almacenada a distintas temperaturas (25 °C, 30°C y 40°C). Los resultados se muestran en la Tabla 8 y Tabla 9, para el caso del ZIC. Los resultados evidenciaron que no existen diferencias significativas entre las CIM y las CBM de las formulaciones almacenadas durante 4 semanas a diferentes temperaturas al comprarlas con las formulaciones frescas (Tabla 8). Al igual que la preparación de los geles frescos, independiente al tiempo y temperatura de almacenamiento, esta mantiene la capacidad bacteriostática y bactericida para todas las cepas ensayadas.

La Tabla 9 muestra los resultados obtenidos desde el análisis de inhibición de la zona de crecimiento, tanto de la formulación almacenada a diferentes temperaturas (25, 30 y 40°C) durante un mes, así como la ZIC de la formulación fresca. Los resultados evidencian que, al igual que los resultados obtenidos en la CIM y CBM, la capacidad inhibitoria del gel, no se ve afectada por la temperatura y el almacenamiento.

EJEMPLO 6: Pruebas clínicas de la composición en el tratamiento del acné.
El objetivo del estudio es evaluar la tolerabilidad y eficacia de un producto cosmético para ser utilizado en sujetos propensos al acné con lesiones inflamatorias a un placebo después de 4 semanas de uso.
Se realizó una evaluación dermatológica del estado de la piel: eritema, sequedad, descamación, fisuras, pápulas, pústulas, edema, supuración, vesículas, otros; encuesta de parámetros subjetivos de la piel: picazón, ardor, sensación de sequedad, otros; re cuento de lesiones inflamatorias (pápulas y pústulas) y lesiones / comedones no inflamatorios (puntos negros y puntos blancos, dejando fuera la nariz); cuestionario de autoevaluación.
Se evaluó un panel de 24 sujetos con edades comprendidas entre 12 y 45 años con un mínimo de 3-4 lesiones inflamatorias por área sin tratamiento de retinoides durante 6 meses, sin tratamiento médico para el acné o antiseborreicos durante dos meses; sin tratamiento con antibióticos y sin tratamiento cosmético para pieles propensas al acné durante 2 semanas.
EJEMPLO 7: Pruebas clínicas de la composición en el tratamiento de la enfermedad periodontal.
El objetivo del estudio es evaluar la tolerabilidad y eficacia de la composición para ser utilizada en sujetos con enfermedad periodontal después de 2 y 6 semanas de uso.
Se consideró un total de 30 sujetos entre 12 y 45 años que cumplieran con los siguientes criterios de inclusión: Pacientes que tengan 20 o más dientes, paciente sistemáticamente sano sin comorbilidades, pacientes con gingivitis clínicamente confirmada (sangrado al sondaje del 10%) sin tratamiento con antibióticos en los últimos 30 días y sin ningún tipo de tratamiento médico que tenga efecto en condiciones gingivales. El grupo de estudio se dividió en dos grupos, ambos grupos fueron evaluados clínicamente antes de comenzar el tratamiento. En esta evaluación inicial se midió sitios de sangrado, profundidad de sondaje y puntuación del índice gingival para cada diente. Al grupo A se le indicó la aplicación de la composición en la encía marginal durante 14 días, dos veces al día después del cepillado de dientes por la mañana y por la noche. Al grupo B se le indicó la aplicación de un placebo en la encía marginal durante 14 días, dos veces al día después del cepillado de dientes por la mañana y por la noche. Luego de este periodo se realizó un examen clínico para los sitios de sangrado, profundidad de sondaje y puntuación del índice gingival para cada diente. Posteriormente se prosiguió con la aplicación de la composición y el placebo durante 4 semanas más, y se realizó el mismo examen clínico.
Claims
1. Una composición con actividad antimicrobiana CARACTERIZADA porque comprende aceite esencial de Ciprés de las Guaitecas ( Pilgerodendron uviferum) encapsulado en nanopartículas de quitosano con tripolifosfato de sodio.
2. La composición de la reivindicación 1 CARACTERIZADA porque comprende además al menos un surfactante.
3. La composición de la reivindicación 1 CARACTERIZADA porque se selecciona de una nanosuspensión, gel, nanoemulsión, emulsión, pasta, crema, loción, jalea.
4. La composición de la reivindicación 3 CARACTERIZADA porque corresponde a una nanosuspensión.
5. La composición de la reivindicación 1 CARACTERIZADA porque el aceite de Ciprés de las Guaitecas se obtiene a partir de descartes madereros.
6. La composición de la reivindicación 5 CARACTERIZADA porque el aceite se obtiene mediante destilación por arrastre con vapor de agua
7. La composición de la reivindicación 1 CARACTERIZADA porque el aceite de Ciprés de las Guaitecas está en una concentración de 0,1% a 0,5% p/v.
8. La composición de la reivindicación 7 CARACTERIZADA porque el aceite de Ciprés de las Guaitecas está en una concentración de 0,4% p/v.
9. La composición de la reivindicación 1 CARACTERIZADA porque el quitosano corresponde a quitosano de bajo peso molecular.
10. Uso de la composición de cualquiera de las reivindicaciones anteriores CARACTERIZADA porque sirve para uso en el tratamiento del acné y la enfermedad periodontal.
11. Un método de producción de una composición con actividad antimicrobiana CARACTERIZADO porque comprende las siguientes etapas: a. mezclar aceite esencial de Ciprés de las Guaitecas con surfactante en una relación entre 1 :0,5 a 1 :10 (p/v); b. agregar agua destilada y agitar por un periodo de 12 a 48 horas; c. agregar ácido acético y quitosano; d. en forma paralela, elaborar una solución de tripolifosfato de sodio; e. mezclar ambas soluciones en una relación Qo/Tpp 2:1 (v/v) mediante goteo por medio de una bomba de infusión en agitación constante; f. centrifugar a 21.000 x g por 30 a 60 min y conservar el sobrenadante g el sobrenadante se dializa por 2 horas frente a agua usando membranas de diálisis de celulosa de 12 kDa.
12. Un método de producción de aceite esencial de Ciprés de las Guaitecas CARACTERIZADO porque comprende las siguientes etapas: a. agregar viruta de tronco de Ciprés de las Guaitecas y agua destilada en un destilador; b. calentar el caldero por un periodo de 1 -10 horas; c. condensar los vapores generados en un tubo de decantación; d. decantar la composición hasta que el aceite se separe del agua.
13. Gel cosmético CARACTERIZADO porque comprende la composición de las reivindicaciones 1 a 9.
14. Un método de producción de un gel cosmético CARACTERIZADO porque comprende las siguientes etapas: a. disolver 100 mi de la composición que comprende aceite esencial de Ciprés de las Guaitecas (Pilgerodendron uviferum) encapsulado en nanopartículas de quitosano con tripol ¡fosfato de sodio y 2 g de HPMC
K15 a 2500 rpm; b. dejar reposar por 12-24 horas a 5°C.
15. Uso del aceite esencial de ciprés de las guaitecas CARACTERIZADO porque sirve para preparar una composición útil para el tratamiento del acné y enfermedad periodontal.
