Abstract
본 발명은 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질 및 이의 용도에 관한 것이다. 본 발명의 융합단백질은 우수한 혈뇌장벽 투과 효율을 가지므로, 신경계 질환의 효과적인 치료에 널리 활용될 수 있다.
Description
본 발명은 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질 및 이의 용도에 관한 것이다.
혈뇌장벽(blood-brain barrier; BBB)은 관련된 혈관주위세포 (pericytes) 및 성상세포 (astrocytes)와 접하는 혈관내피세포 간의 0.1 Ω·m 이상의 고도로 높은 전기적 저항성을 갖는 치밀한 연결 (tight junctions)로 구성된 세포 장벽이며, 중추신경계 (central nervous system; CNS)에서 뇌 세포 외액 (brain extracellular fluid)으로부터 순환하는 혈액을 분리하는 고도로 선택적인 투과성을 갖는 장벽으로서, 뇌로의 영양 및 다른 물질의 출입을 조절함으로써 중추신경계를 보호하는 관문 역할을 한다.
통상, 혈뇌장벽은 뇌 기능에 필수적인 글루코스 및 아미노산과 같은 분자들을 선택적으로 이동시킬 뿐만 아니라, 수동적 확산 (passive diffusion)에 의해 물, 몇 가지 기체 및 지용성 분자를 통과시킨다. 반면, 혈뇌장벽은 투과성 당단백질 (p-glycoprotein)에 의해 매개되는 능동적 전달 기전에 의해 친유성, 잠재적 신경독소의 출입을 차단한다. 따라서, 뇌 안쪽에서 작용해야 되는 분자량이 큰 약물 또는 낮은 뇌 투과율을 갖는 저분자 약물과 같은 신경 질환 약물들은 혈뇌장벽을 투과할 수 없다.
이와 같이, 혈뇌장벽은 혈액을 통해 운반될 수 있는 박테리아, 병원체 및 혈액 내 잠재적 위험물질이 뇌로 전달되는 것을 차단하는 역할을 하지만, 이러한 혈관 장벽으로 인해, 대부분의 중추신경계 약물도 경두개(transcranial) 전달에 대해 낮은 효율을 나타내며, 이를 보상하기 위하여 이들 약물을 고용량으로 투여하게 되므로 주변 장기에 심각한 부작용을 유발하기도 한다. 따라서, 연구가 지속되고 있으나 (KR 10-2211721), 부정적인 전신 효과를 방지하는 동시에, 화학 약물 (chemodrug)의 치료 효과를 보장하기 위하여 혈뇌장벽을 투과할 수 있는 효율적인 약물 전달 시스템의 발굴이 요구된다.
본 발명자들은 혈뇌장벽을 투과할 수 있으며, 약물 투여 시 지속적인 효능을 발휘하고, 신경계 질환 예방 및 진행 지연 효과를 발휘할 수 있는 융합단백질을 개발함으로써 본 발명을 완성하였다.
본 발명의 하나의 목적은 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질을 제공하는 것이다.
본 발명의 다른 하나의 목적은 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질을 유효성분으로 포함하는 신경계 질환의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 하나의 목적은 상기 조성물을 개체에 투여하는 단계를 포함하는, 신경계 질환 예방 또는 치료방법을 제공하는 것이다.
본 발명의 다른 하나의 목적은 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질의 신경계 질환의 예방, 개선 또는 치료 용도를 제공하는 것이다.
본 발명의 다른 하나의 목적은 상기 융합단백질을 포함하는 약학적 조성물의 신경계 질환의 예방 또는 치료 용도를 제공하는 것이다.
본 발명의 융합단백질은 새로운 형태의 혈뇌장벽 투과 효율이 증진된 신경계 질환 치료 후보물질을 제시하므로, 신경계 질환의 효과적인 치료에 널리 활용될 수 있다.

또한, 당해 기술분야의 통상의 지식을 가진 자는 통상의 실험만을 사용하여 본 발명에 기재된 본 발명의 특정 양태에 대한 다수의 등가물을 인지하거나 확인할 수 있다. 또한, 이러한 등가물은 본 발명에 포함되는 것으로 의도된다.
본 발명의 목적을 달성하기 위한 하나의 양태는 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질을 제공한다.
상기 목적을 달성하기 위하여 IGF-1(Insulin-like growth factor 1)를 포함하는 혈뇌장벽(Blood-Brain Barrier; BBB) 투과용 약물 전달체를 제공한다.
본 발명의 용어 "혈뇌장벽(blood-brain barrier; BBB)"이란, 관련된 혈관주위세포 (pericytes) 및 성상세포 (astrocytes)와 접하는 혈관내피세포 간의 0.1 Ω 이상의 고도로 높은 전기적 저항성을 갖는 치밀한 연결 (tight junctions)로 구성된 세포 장벽이며, 중추신경계 (central nervous system; CNS)에서 뇌 세포 외액 (brain extracellular fluid)으로부터 순환하는 혈액을 분리하는 고도로 선택적인 투과성을 갖는 장벽으로서, 뇌로의 영양 및 다른 물질의 출입을 조절함으로써 중추신경계를 보호하는 관문 역할을 한다.
본 발명의 용어, "약물 전달체"는, 목적하는 조직 또는 장기에 치료학적 활성을 나타내는 약물을 효율적으로 전달하기 위한 전달체로, 본 발명의 약물 전달체는 특히 혈뇌장벽을 투과하여 뇌에 약물을 높은 효율로 전달할 수 있는 것을 특징으로 한다.
구체적으로, 본 발명의 약물 전달체는 IGF-1을 포함하고, 보다 구체적인 양태로는 IGF-1과 면역글로불린 Fc가 연결된 단백질의 형태일 수 있으나, 이에 제한되지 않는다.
본 발명의 약물 전달체는 약물 전달체 자체로도 치료 효능을 나타낼 수 있을 뿐만 아니라, 약물 전달체에 연결되는 약물을 혈뇌장벽을 투과하여 효율적으로 전달하여 뇌에서 치료 활성을 나타내도록 할 수 있다. 또한, 약물의 반감기를 증가시켜 약효가 오래 지속되고 투여 횟수를 감소시킬 수 있는 효과를 갖는다.
본 발명의 일 구현예에서는 본 발명의 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질의 약동학 평가를 한 결과, 반감기를 증가시킴을 확인하였으며 (도 5), 형광 표지 된 물질을 마우스 복강 내로 투여한 후, 뇌를 관찰한 결과 형광 세기가 시간이 지남에 따라 증가함을 확인하여 BBB 투과율 또한 우수함을 확인하였다 (도 7)
본 발명의 용어 "IGF-1 (Insulin-like growth factor 1)"란, 인슐린과 구조가 유사한 세포증식인자로, 정상적인 성장과 건강유지에 중요한 역할을 한다. 사람 체내에 존재하는 펩타이드에서 유래하였다. 본 발명의 IGF-1은 천연형뿐만 아니라 이의 유도체 또는 변이체를 모두 포함한다. 상기 유도체, 변이체는 천연형 서열에서 하나 이상의 아미노산이 치환, 결실, 부가 등의 변이된 것으로, 고유의 활성을 유지하는 것을 의미한다. 상기 IGF-1의 아미노산 서열은 특별히 제한되지 않으나, 서열번호 9의 아미노산 서열을 포함하거나, 서열번호 9의 아미노산 서열을 기준으로 3번, 49번, 67번, 70번 및 이들의 조합 중 하나 이상의 위치에 상응하는 아미노산에서의 치환, 결실, 부가 등의 변이가 일어난 서열을 포함할 수 있다. 구체적으로, 3번째 위치에 상응하는 아미노산은 알라닌으로, 49번째 위치에 상응하는 아미노산은 알라닌으로, 67번째 위치에 상응하는 아미노산은 트레오닌으로, 70번째 위치에 상응하는 아미노산은 티민으로 치환된 것을 포함할 수 있다. 보다 구체적으로, 서열번호 10, 11 또는 12의 아미노산 서열을 포함할 수 있다. 상기 서열번호 9, 10, 11 또는 12의 아미노산 서열은 상기 서열번호 9, 10, 11 또는 12으로 기재된 아미노산 서열과 적어도 70%, 75%, 80%, 85%, 90%, 95%, 96%, 97%, 98%, 99%, 99.5%, 99.7% 또는 99.9% 이상의 상동성 또는 동일성을 가지는 아미노산 서열을 포함할 수 있다. 또한, 이러한 상동성 또는 동일성을 가지며 본 출원의 단백질에 상응하는 효능을 나타내는 아미노산 서열이라면, 일부 서열이 결실, 변형, 치환, 보존적 치환 또는 부가된 아미노산 서열을 갖는 단백질도 본 출원의 범위 내에 포함됨은 자명하다.
예를 들어, 상기 아미노산 서열 N-말단, C-말단 그리고/또는 내부에 본 출원의 단백질의 기능을 변경하지 않는 서열 추가 또는 결실, 자연적으로 발생할 수 있는 돌연변이, 잠재성 돌연변이 (silent mutation) 또는 보존적 치환을 가지는 경우이다.

본 출원에서 용어, '상동성 (homology)' 또는 '동일성 (identity)'은 두 개의 주어진 아미노산 서열 또는 핵산염기 서열 상호간 유사한 정도를 의미하며 백분율로 표시될 수 있다. 용어 상동성 및 동일성은 종종 상호교환적으로 이용될 수 있다.
보존된(conserved) 폴리뉴클레오티드 또는 단백질의 서열 상동성 또는 동일성은 표준 배열 알고리즘에 의해 결정되며, 사용되는 프로그램에 의해 확립된 디폴트 갭 페널티가 함께 이용될 수 있다. 실질적으로, 상동성을 갖거나(homologous) 또는 동일한(identical) 서열은 일반적으로 서열 전체 또는 일부분과 중간 또는 높은 엄격한 조건(stringent conditions)에서 하이브리드할 수 있다. 하이브리드화는 폴리뉴클레오티드에서 일반 코돈 또는 코돈 축퇴성을 고려한 코돈을 함유하는 폴리뉴클레오티드와의 하이브리드화 역시 포함됨이 자명하다.
본 발명에 있어서, 상기 IGF-1는 뉴런을 보호하며 흥분성 물질에 대한 신경세포 보호 및 혈뇌장벽(BBB, blood-brain barrier) 투과도를 증진시킬 수 있는 기능을 가짐으로써 신경계 질환 예방 또는 치료의 효과를 상승시킨다.
본 발명의 목적상, 본 발명의 IGF-1은 혈뇌장벽 투과를 위해 서열번호 9의 야생형 IGF-1 아미노산 서열에서 변형을 포함할 수 있다. 혈뇌장벽 투과를 위해 도입되는 변형의 종류는 제한되지 않으나, IGFBP와의 결합능 저해를 위해 도입된 변형이면 제한 없이 본 발명에 포함될 수 있다. 구체적으로, 본 발명의 IGF-1 은 IGFBP와 결합능이 저해되도록 아미노산 치환을 포함하는 변이체일 수 있으며, 보다 구체적으로 서열번호 10, 11 또는 12의 아미노산을 포함하는 변이체일 수 있다.
예를 들어, IGF-1의 서열에서 하나 이상의 아미노산 치환을 통해 IGFBP 단백질과 결합하는 것을 저해할 수 있는데, 본 발명의 IGF-1이 포함할 수 있는 아미노산의 제한되지 않는 예는 다음과 같다: E3A, F49A, A67T, A70T 등. 이 때, 상기의 표기는 순서대로 치환 전 아미노산, 치환 위치, 및 치환 후 아미노산을 의미한다. 예를 들어, E3A는 야생형 IGF-1의 서열을 기준으로 3번째에 상응하는 아미노산인 글루탐산(E)이 알라닌(A)으로 치환된 것을 의미한다. 상기 아미노산 치환에서 선택되는 하나 이상의 아미노산 치환을 포함함으로써 본 발명의 IGF-1을 포함하는 약물 전달체는 우수한 혈뇌장벽 투과능을 가질 수 있다.
본 발명의 용어 "IGFBP (Insulin-like growth factor-binding protein)"는 IGF-1(insulin-like growth factor 1)의 수송단백질 역할을 한다. 체내 IGF는 대부분이 IGFBP 단백질군과 결합한 상태로 존재함으로써 표적전달능이 저해된다. 따라서, 본 발명의 약물 전달체는 IGFBP와 결합하지 못하도록 설계되어 BBB 투과 효율을 높일 수 있다.
본 발명의 약물 전달체의 구체적인 양태로는, IGF-1이 면역글로불린 Fc와 연결된 Fc-IGF1의 구조를 갖는 약물 전달체를 제공할 수 있다.
본 발명에서 용어, "Fc-IGF1 약물 전달체"는 "Fc-IGF1 하이브리드(Hybrid)"와 혼용되어 사용될 수 있다. 상기 Fc-IGF1 약물 전달체는 IGFBP와의 결합이 저해되고 혈뇌장벽 투과능이 향상된 IGF-1을 포함함으로써 우수한 혈뇌장벽 투과 효율을 보일 뿐만 아니라, 면역글로불린 Fc와의 결합을 통해 약물의 반감기가 증대된 것일 수 있다.
본 발명의 일 구현예에서는 본 발명의 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질의 IGF-1 활성을 평가하기 위하여 Saos-2 세포주에서 proliferation assay 및 PathHunter HEK293 IGF-1R bioassay kit을 이용한 결과, 대조구(wild) 대비 IGF-1 활성이 우수함을 확인하였다 (도 2, 도 3).
본 발명의 용어 "면역글로불린 Fc"는 혈청성분 중 면역에 중요한 역할을 하고, 또 항체 작용을 하는 단백질의 총칭이다. 기본구조는 분자량 약 2만 3000의 L사슬(가벼운 사슬) 1쌍과 5만~7만의 사슬(무거운 사슬) 1쌍이 S-S 다리걸침에 의해 결합된 것으로, H 사슬의 종류에 의해 각각 IgG, IgA, IgM, IgD, IgE로 분류된다. 분자 모양은 Y자형이고, 2개의 상단이 등가의 항체 결합부위인데 대해, 하단(Fc 부분)은 항원과 결합하는 항체가 보체나 세포와 결합하는 등의 생물활성을 나타내는 부위이다. 본 발명에서 "면역글로불린 Fc"는 "면역글로불린 Fc 영역"과 혼용되어 사용될 수 있다.
본 발명에서 면역글로불린 Fc 단편은 CH1, CH2, CH3 및 CH4 도메인으로 이루어진 군으로부터 선택된 1개 내지 4개 도메인으로 이루어진 것일 수 있으며, 상기 면역글로불린 Fc 단편은 힌지(hinge) 영역을 추가로 포함하는 것일 수 있다. 또한, 면역글로불린 Fc 단편은 IgG, IgA, IgD, IgE, IgM, 이들의 조합(combination) 및 이들의 하이브리드(hybrid)로 이루어진 군으로부터 선택되는 것일 수 있다. 본 발명의 구체적인 면역글로불린 Fc의 예로는 트라스투주맙(trastuzumab)이 사용될 수 있으나, 이에 제한되지 않는다. 또한, Fc를 포함하는 본 발명의 약물 전달체가 체내에서 감소된 이펙터 기능을 갖도록 변형을 포함하는 것일 수 있으나, 예를 들어, N297A의 변형을 포함하는 것일 수 있다.
본 발명의 용어 "IgG(immunoglobulin G, 면역글로불린 G)"는 면역글로불린의 1종으로서 태반통과성이 있다. 기본구조로 나타내는 H사슬의 constant부분의 상위에 따라서 4개의 서브클래스(subclass)IgG1, IgG2, IgG3, 및 IgG4로 나눠진다.
본 발명의 용어 "human IgG1 Fc"는 항체 개별형 중 하나로서 사람의 경우 혈청에 있는 면역글로불린 총량 중 75%를 차지한다. 본 발명에서는 사람 유래 항체(human IgG1)의 Fc 부위를 이용하였으며 해당 도메인은 물질의 수용성 증진 및 반감기 증진을 목적으로 다양한 의약품에 사용되는 단백질 기능단이다. 본 발명의 면역글로불린 Fc는 천연형뿐만 아니라 이의 유도체 또는 변이체를 모두 포함한다. 상기 유도체, 변이체는 하나 이상의 아미노산이 치환, 결실, 부가 등의 변이된 것으로, 고유의 활성을 유지하는 것을 의미한다. 상기 human IgG1 Fc의 아미노산 서열은 특별히 제한되지 않으나 서열번호 4의 아미노산을 포함하거나, 서열번호 4의 아미노산 서열을 기준으로 297번, 309번, 311번, 428번 및 이들의 조합 중 하나 이상의 위치에 상응하는 아미노산에서의 치환, 결실, 부가 등의 변이가 일어난 서열을 포함할 수 있다. 구체적으로, 297번째 위치에 상응하는 아미노산은 알라닌으로, 309번째 위치에 상응하는 아미노산은 타이로신으로, 311번째 위치에 상응하는 아미노산은 메티오닌으로, 428번째 위치에 상응하는 아미노산은 류신으로 치환된 것을 포함할 수 있다. 보다 구체적으로, 서열번호 5, 6, 7 또는 8의 아미노산 서열을 포함할 수 있다. 상기 서열번호 4, 5, 6, 7 또는 8의 아미노산 서열은 상기 서열번호 4, 5, 6, 7 또는 8으로 기재된 아미노산 서열과 적어도 70%, 75%, 80%, 85%, 90%, 95%, 96%, 97%, 98%, 99%, 99.5%, 99.7% 또는 99.9% 이상의 상동성 또는 동일성을 가지는 아미노산 서열을 포함할 수 있다. 상기 "상동성", "동일성"은 전술한 바와 같다.
본 발명에 있어서, 상기 human IgG1 Fc는 전달체의 수용성과 생체 내 반감기 증진 기능을 가짐으로써 신경계 질환 예방 또는 치료의 효과를 상승시킨다.
본 발명에서 FcRn은 IgG 항체 Fc 영역에 결합하는 단백질을 의미한다. FcRn은, 이로 제한하는 것은 아니나, 인간, 마우스, 래트, 토끼 및 원숭이를 포함하는 임의의 유기체 유래일 수 있다. 면역글로불린 (항체)의 생체 내 반감기가 FcRn에 대한 Fc의 결합에 의해 매개된다는 것이 보고되며, 항체의 혈중 반감기와 지속성은 항체의 Fc 부위와 IgG 결합 리간드 중의 하나인 FcRn (neonatal Fc receptor) 결합에 크게 의존한다. 또한, FcRn에 대한 Fc의 결합은 항체 운반에 있어서도 중요한 역할을 담당한다.
본 발명의 일 구현예에서는, GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질은 FcRn에 대한 결합력이 우수함을 확인하였다 (도 6).
본 발명의 약물 전달체는 혈뇌장벽 투과 효율이 높으므로, Fc-IGF1 구조를 갖는 약물 전달체의 치료학적 활성을 갖는 약물을 결합시킴으로써, 약물의 효능이 적절히 발휘할 수 있도록 할 뿐만 아니라, 증가된 반감기로 인해 효능이 오래 발휘되도록 하므로, 우수한 치료 효과를 가질 수 있다.
구체적으로, 혈뇌장벽을 투과하여 약물을 전달함으로써 우수한 치료 효과를 기대할 수 있는 신경계 질환의 예방 또는 치료를 위한 물질을 본 발명의 약물 전달체와 연결시킬 수 있으나, 혈뇌장벽을 투과하여 치료 효과를 나타내는 물질이라면 특정 질환의 치료제에 한정되지 않고 본 발명의 약물 전달체와 결합시킬 수 있다.
상기 물질은 신경계 질환 치료제가 될 수 있고, 상기 신경계 질환 치료제는 신경계 질환 치료용 저분자 의약품, 펩타이드, 효소, 항체, 단백질 등을 들 수 있다. 신경계 질환 치료제의 예로는 celecoxib, masitinib, (1-3) IGF1, Exenatide BDNF, GDNF, CNTF, Iduronate 2-sulfatase (IDS), Glucocerebrosidase (GBA), α-synuclein specific antibody 등이 있으며, 구체적인 한 예로는 GLP-1을 들 수 있고, 본 발명의 약물 전달체 Fc-IGF1는 GLP-1이 연결된 융합단백질의 형태일 수 있으나, 이에 제한되지 않는다.
본 발명의 용어 "GLP-1"는 글루카곤 유전자의 전사체로부터 유도된 위장유래 호르몬의 일종이다. 본 발명의 GLP-1은 천연형뿐만 아니라 이의 유도체 또는 변이체를 모두 포함한다. 상기 유도체, 변이체는 하나 이상의 아미노산이 치환, 결실, 부가 등의 변이된 것으로, 고유의 활성을 유지하는 것을 의미한다. 상기 GLP-1의 아미노산 서열은 특별히 제한되지 않으나 서열번호 1 내지 3 중 어느 하나의 아미노산 서열을 포함할 수 있다. 본 발명의 GLP-1은 천연형 서열 (서열번호 2)을 포함하거나, 이의 활성형인 7-36번 아미노산에 해당하는 서열번호 3을 포함하거나, 또는 이들 서열에서 하나 이상의 아미노산의 치환, 부가, 결실 등을 추가로 포함하는 GLP-1의 변이체일 수 있다.
또는, 천연형 서열에서 DPPIV에 대한 저항성을 가져 체내 분해가 저해되도록 변형된 것일 수 있다. 변형의 예로는 2번째 아미노산인 알라닌을 다른 아미노산(예를 들어, 글리신(G)) 또는 비천연형 아미노산으로 치환시키는 것이 될 수 있으나, 이에 제한되지 않는다.
본 발명의 일 구현예에서는 본 발명의 융합단백질의 GLP-1 활성을 평가하기 위하여 cAMP HunterTM Liraglutide bioassay kit을 이용한 결과, GLP-1 수용체 작용제인 엑센딘-4와 유사한 수준의 활성을 나타냄을 확인하였다 (도 4). 본 발명에 있어서, 상기 GLP-1는 전달체의 신경세포 내 항염증 작용의 기능을 가짐으로써 신경계 질환 예방 또는 치료의 효과를 상승시킨다.
본 발명에서 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질은 PF1802로 명명한다.
또한, 본 발명의 약물 전달체는 근위축성 측삭경화증(amyotrophic lateral sclerosis, ALS)의 치료제와 연결된 것일 수 있다. 상기 근위축성 측삭경화증의 치료제는 신경계 질환 치료용 저분자 의약품, 펩타이드, 효소, 항체, 단백질 등을 들 수 있다. 상기 근위축성 측삭경화증의 치료제의 예로는 celecoxib, masitinib, (1-3) IGF1, Exenatide BDNF, GDNF, CNTF, Iduronate 2-sulfatase (IDS), Glucocerebrosidase (GBA), α-synuclein specific antibody 등이 있으며, 구체적인 예로는 GLP-1을 들 수 있으나, 이에 제한되지 않는다.
본 발명의 일 구현예에서는 근위축성 축상경화증 유전자 변형 마우스 모델에 본 발명의 PF1802_M008 후보물질을 투여한 결과, 완전히 신경화된 신경근 접합부 (Fully innervated NMJ)는 증가하였으며, 탈신경 신경근 접합부 (denervated NMJ)는 감소함을 확인하여, 근위축성 측상경화증 질환에 대한 예방 및 진행 지연 효과가 우수함을 확인하였다 (도 8, 도 11).
또한, 미세아교세포 마커 Iba1가 무처리 대비 현저히 감소하고, ALS 치료제로 알려진 릴루졸(riluzole)과 유사한 활성을 나타내어 본 발명의 물질의 BBB 투과율이 우수함을 확인하였다 (도 12).
본 발명의 약물 전달체와 약물은 당업계의 공지된 다양한 방법으로 연결된 것일 수 있다. 예를 들어, 약물이 IGF-1 또는 Fc에 링커를 통해 연결되거나, 공유 또는 비공유 결합으로 직접적으로 연결된 형태를 모두 포함할 수 있으며, 혈뇌장벽 투과 및 치료 효과를 나타낼 수 있는 한, 특정한 연결 방법이나 최종 연결된 형태로 제한되지 않는다.
본 발명의 융합단백질은 N-말단부터 GLP-1, 제1링커, Fc영역, 제2링커 및 IGF-1이 연결된 형태일 수 있다.
상기 링커는 펩타이드 링커로서, 본 발명의 약물 전달체는 IGF-1과 면역글로불린 Fc 영역이 펩타이드 링커를 통해 융합된 것일 수 있다. 상기 링커의 일 말단이 이량체의 면역글로불린 Fc 영역의 한 사슬에 연결될 수 있으나, 이에 제한되는 것은 아니다.
상기 펩타이드 링커는 1개 이상의 아미노산을 포함할 수 있으며, 예컨대 1개부터 1000개의 아미노산을 포함할 수 있으며, 당업계에 알려진 임의의 펩타이드 링커, 그 예로 [GS]x 링커, [GGGS]x 링커 및 [GGGGS]x 링커 등을 포함하며, 여기서 x는 1 이상의 자연수(예를 들어, 1, 2, 3, 4, 5, 또는 그 이상)일 수 있으나, 이에 제한되지 않는다.
구체적으로, 본 발명에서 링커는 GSAPAP(G4S) (서열번호 38), (G4S)4GAHS (서열번호 39), ASGAGSTTLEVLFQGP (서열번호 40), (G)8 (서열번호 41) 또는 (PA)5 (서열번호 42)일 수 있으나, 이에 제한되지 않는다.

본 발명의 용어 "신경계 근육질환"은 근육의 직접적인 질병으로 인한 또는 신경, 신경근육연접의 이상으로 인한 간접적인 근육의 기능 이상을 포괄하는 질병을 의미하는 것으로 신경근육장애라고도 한다. 중추신경계에 영향이 있는 경우 근육의 경련 및 마비 증상이 있고 침범한 뇌의 부분에 따라 증상이 다르게 나타나며, 뇌졸중, 다발성 경화중, 파킨슨병, 말초신경병증, 근육병, 중증근무력증, 근위축성 측삭경화증, 척수성 근위축증 등이 이에 해당한다.
본 발명의 상기 약학적 조성물은 신경근 접합부 기능 개선, 뉴런 보호 효과 및 혈뇌장벽(blood-brain barrier) 투과도 개선 및 체내 반감기 증진를 가져 신경계 질환의 예방 또는 치료 효과를 가지는 것일 수 있으나, 이에 제한되지 않는다. 본 발명에서 신경계 질환은 신경근 접합부의 기능 이상과 관련된 질환으로, 본 발명에 따른 융합단백질의 투여로 신경근 접합부의 기능 개선과 회복을 통해 신경계 질환 예방 또는 치료 효과를 얻을 수 있다.
본 발명의 용어 "예방"이란 본 발명의 조성물의 투여로 신경계 질환을 방지하거나 지연시키는 모든 행위를 의미하며, "치료"란 본 발명의 조성물의 투여로 신경계 질환의 증세가 호전되거나 이롭게 변경하는 모든 행위를 의미한다.
본 발명의 약학적 조성물은, 약학적으로 허용되는 담체, 부형제 또는 희석제를 추가로 포함할 수 있다. 이러한 약학적으로 허용되는 담체, 부형제, 또는 희석제는 비자연적으로 발생된 것일 수 있다. 구체적으로, 상기 조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명에서, 상기 약학적 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화 할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 추출물과 이의 분획물들에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘 카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는 데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 다른 하나의 양태는 상기 약물 전달체, 융합단백질 또는 이를 포함하는 약학적 조성물을 개체에 투여하는 단계를 포함하는 신경계 질환 예방 또는 치료 방법을 제공한다.

상기 용어, "약학적으로 유효한 양"은 의학적 용도에 적용 가능한 합리적인 수혜/위험 비율로 신경계 질환을 예방 또는 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 예를 들면, 상기 약물 전달체, 융합단백질 또는 이를 포함하는 약학적 조성물은 1일 0.01 내지 500 mg/kg으로, 구체적으로 10 내지 100 mg/kg의 용량으로 투여할 수 있으며, 상기 투여는 하루에 한 번 또는 수회 나누어 투여할 수도 있다.
본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 조성물은 신경계 질환 예방 또는 치료를 위하여 단독으로 사용할 수 있고, 수술, 호르몬 치료, 약물 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명에서 상기 조성물은 상기 조성물은 신경근 접합부 (Neuromuscular junction, NMJ)의 기능 회복 또는 개선 효과를 갖는 것일 수 있다.
본 발명에서 "신경근 접합부 (Neuromuscular junction, NMJ)"는 운동신경이 말단에서 근에 접속하고, 신경의 흥분을 근섬유의 막으로 전달하기 위한 특수한 구조를 말하고 시냅스의 일종을 의미한다. 신경근 접합부 질환은 중증근무력증(myasthenia gravis, MG)과 Lambert-Eaton 근무력증후군, 보툴리누스중독(botulism) 및 선천근무력증 (congenital myasthenic syndrome)을 포함한다.
본 발명의 다른 하나의 양태는 상기 약물 전달체 또는 융합단백질을 포함하는, 근위축성 측삭경화증(amyotrophic lateral sclerosis, ALS)의 예방 또는 치료용 약학적 조성물을 제공한다.
상기 "약물 전달체", “융합단백질” 및 "약학적 조성물"에 대해서는 전술한 바와 같다.
본 발명에서, 상기 융합단백질은 본 발명의 약물 전달체에 치료제인 GLP-1이 연결된 것일 수 있고, 구체적으로는 GLP-1, 면역글로불린 Fc, 및 IGF-1이 연결된 것일 수 있다. 본 발명에 따른 융합단백질은 뇌에 도달하는 효율이 높고 반감기가 증가된 것이어서, 근위축성 측삭경화증에서 우수한 치료 효과를 기대할 수 있다.
본 발명의 다른 하나의 양태는 GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질의 신경계 질환의 예방, 개선 또는 치료 용도를 제공한다.
본 발명의 다른 하나의 양태는 상기 융합단백질을 포함하는 약학적 조성물의 신경계 질환의 예방 또는 치료 용도를 제공한다.
상기 "GLP-1", "면역글로불린 Fc", "IGF-1", "신경계 질환", "약학적 조성물"은 전술한 바와 같다.
이하, 실시예를 통하여 본 발명을 보다 상세히 설명하고자 한다. 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 한정되는 것은 아니다.
실시예 1. 융합단백질의 제조
후보물질의 확보를 위해 GLP1-Fc-IGF1 또는 GLP1-Fc-mut. IGF1-TfR을 합성한 후, Nhe I/Hind III 사이트를 이용하여 pcDNA3.1 벡터에 클로닝 하였다. 클로닝 된 벡터를 CHO-S 세포에 형질 감염하여 5일차 상등액을 수거한 후, 원심분리와 0.22 μm 필터를 통해 세포와 부유물을 제거하였다. 그 후, protein A resin으로 정제하여 표 1과 같이 다양한 PF1802 후보물질_M003 내지 M032 (서열번호 13 내지 37)을 확보하였다 (도 1, 표 1).

실시예 2. PF1802 후보물질의 활성 평가
실시예 2-1. Saos-2 세포주를 이용한 PF1802 후보물질의 IGF-1 활성 평가
실시예 1.의 방법으로 제조된 PF1802 후보물질의 IGF-1 활성을 확인하기 위하여, Saos-2 세포주에서 IGF-1에 의한 proliferation assay를 수행하였다. Saos-2 세포주를 웰 당 1x104씩 96 well plate에 분주하여 37℃, 5% CO2에서 16시간 배양한 후 4시간 동안 starvation하였다. 이후, PF1802 후보물질을 최고농도 100 nM에서 0.00256 nM까지 5배씩 희석하여 각 웰에 처리한 후, 37℃, 5% CO2에서 48시간 동안 배양하였다. 배양한 세포를 SRB(sulforhodamine B) 염색을 진행 한 후, microplate reader기를 이용하여 540 nm에서 흡광도를 측정하였으며, PLA statics 기능을 적용하여 IGF-1을 reference로 설정한 뒤 PF1802 후보물질의 relative potency 값을 도출하였다 (표 2, 도 2).

그 결과, 상기 표 2에서 나타나듯이, wt.IGF1을 사용한 물질 대비 Mutant IGF-1 물질인 M007, M008의 IGF-1 활성을 확인하였다.
실시예 2-2. PF1802 후보물질의 IGF-1 활성 평가
실시예 1.의 방법으로 제조된 PF1802 후보물질의 IGF-1 활성을 확인하기 위하여, IGF1R 를 과발현시킨 세포주를 사용하는 키트인 PathHunter HEK293 IGF-1R bioassay kit(Discover X, 95-0505Y1-000070)을 이용하여 생산된 PF1802 물질의 IGF-1 활성 (인산화 활성)을 측정하였다. 구체적으로, 표 3에 나타낸 바와 같이 assay kit의 technical manual에 따라 진행하였으며, 4-parameter fit을 적용하여 결과를 도출하였다 (표 4, 도 3)

그 결과, 표 4에 나타나듯이 IGF1이 없는 물질인 M011(음성 대조구)와 비교하였을 때, 본 발명의 PF1802 후보물질인 M005, M008, 및 M010의 IGF-1 활성이 존재함을 확인하였다.
실시예 2-3. PF1802 후보물질의 GLP-1 활성 평가
실시예 1.의 방법으로 제조된 PF1802 후보물질의 GLP-1 활성을 확인하기 위하여, cAMP HunterTM Liraglutide bioassay kit (Discover X, 95-0062Y2-00100)을 이용하여 생산된 GLP-1 활성을 측정하였다. 구체적으로, 표 5에 나타낸 바와 같이 assay kit의 technical manual에 따라 진행하였으며, 4-parameter fit을 적용하여 결과를 도출하였다 (표 6, 도 4).

그 결과, 표 6에 나타나듯이 M005, M008, M010, M011의 GLP-1 활성은 엑센딘-4 (exendin-4)와 유사한 수준임을 확인하였으며, 이를 통하여 생산된 물질의 GLP-1 활성에 문제가 없음을 알 수 있었다.
실시예 2-4. PF1802 후보물질의 약동학 (Pharmacokinetics, PK) 평가
혈액에서의 PF1802 후보물질의 약동학을 확인하기 위하여 마우스에서 5mpk 용량으로 피하투여 한 후 144시간까지 채혈하였다. 이후 ELISA 결과를 바탕으로 WinNolin 소프트웨어를 이용하여 PK parameter 값을 도출하였다 (표 7, 도 5).

그 결과, 표 7에 나타나듯이 후보물질인 M005 및 M010의 median Tmax는 각각 48시간, 16시간으로 천천히 흡수 되었음을 확인하였으며, 반감기는 각각 61시간, 29.4 시간임을 확인하였다.
이를 통해, 실제 약물 투여 시 본 발명의 후보물질은 지속적인 효능을 나타내며, 약물 투여 간격을 줄여 줄 수 있다는 점을 시사하는 바이다.
실시예 2-5. 혈중 반감기 증진 Fc 도입 물질
도입된 PF1802 후보물질의 Human FcRn에 대한 결합력 평가
PF1802_M008 후보물질에 3종의 혈중 반감기 증진 Fc를 도입한 PF1802_M020, M021, M022 물질의 human FcRn에 대한 결합력을 확인하기 위하여, ELISA를 수행하였으며, pH에 따른 결합력 차이 확인을 위해 pH6.0, pH7.4 두 가지로 준비하여 각각 진행하였다. 구체적으로, PF1802_M020, M021, M022 물질을 각각 4 ug/mL 로 96 well immunoplate에 코팅하고 4% skim milk가 포함된 PBS (pH6.0, 7.4)로 상온에서 1시간 동안 반응시켜 비특이적인 결합을 차단하였으며, 그 후 Human FcRn-GST 단백질을 1% skim milk가 포함된 PBS (pH6.0, 7.4)를 사용하여 최고농도 5 ug/mL 에서 0.0003 ug/mL까지 4배씩 희석하여 각 웰에 분주한 후 상온에서 1시간 동안 반응 하였다. 이 후, anti-GST Ab-HRP (cytiva, 27457701)를 처리한 다음, TMB 용액 및 H2SO4를 처리하여 450 nm에서 흡광도를 측정하여 결과를 도출하였다 (도 6).
그 결과, 혈중 반감기 증진 Fc를 도입한 PF1802_M020, M021, M022 후보물질이 PF1802_M008 보다 Human FcRn에 대한 결합력이 현저히 증가함을 확인하였다. 이를 통해, 혈중 반감기 증가에 중요한 pH6.0에서 Human FcRn에 대한 우수한 결합력을 확인 할 수 있었다.
더 나아가, 후술하는 실시예 4. 및 5.를 통하여 물질 최적화가 완료된 PF1802_M024 후보물질에도 상기와 동일한 3종 혈중 반감기 증진 Fc를 도입한 PF1802_M030, M031, M032 후보물질을 확보하였으며 (서열번호 35, 36, 37), PF1802_M030, M031, M032 후보물질 또한 결합력이 증가하고 Human FcRn에 대한 우수한 결합력을 가진다.
실시예 3. PF1802_M008 후보물질의 in vivo 효능 평가
실시예 3-1. PF1802_M008 후보물질의 Mouse Brain Blood Barrier (BBB) 투과율 평가
BBB 투과율 확인을 위하여, IVISense 680 NHS Fluorescent labeling kit (PerkinElmer)을 사용하였으며, IGF-1가 없는 M011 (GLP1-Fc) 후보물질을 음성 대조군으로 이용하였다. 구체적으로, 형광 표지 된 물질을 마우스 (Balb/C male) 복강 내로 1회 투여 한 후, 24, 72시간 마다 생리식염수로 perfusion하였고, 그 다음 마우스의 뇌를 적출하여 2차 광학영상장비(IVIS spectrum)로 이미징 분석을 수행하였다 (도 7).
그 결과, 뇌 조직 형광세기는 M008 > M005 > M011 순임을 확인하였고, M005의 경우 M011 대비 형광세기가 1.1배 (24시간), 1.2배 (72시간)로 높게 확인 되기는 하나 통계적으로 유의한 차이는 보이지 않은 반면, M008의 경우 M011 대비 형광세기가 1.7배 (24시간), 2.6배 (72시간)로 형광세기가 가장 높으며, 통계적으로 유의함을 확인하였다.
이를 통해, PF1802_M008 후보물질의 BBB 투과율이 우수함을 알 수 있었으며, wt.IGF1 대비 mut.IGF1의 BBB 투과율이 현저히 높음을 알 수 있었다.
실시예 3-2. 근위축성 측상경화증(ALS) 유전자 변형 마우스 모델에서 PF1802_M008 후보물질의 효능 검정
PF1802_M008 후보물질의 효능을 확인하기 위하여 표 8과 같이 생후 9주된 SOD-1(G39A) 마우스에 PF1802_M008 후보물질을 주 2회씩 10주 동안 피하투여 하였다. 생후 109일에 면역조직학적 분석을 위해 마우스를 희생하였으며, 신경근 접합부 (Neuromuscular junction, NMJ) 평가를 위해 비복근 근육(gastronemius muscle) 샘플에서 anti-synatotagmin 2 Ab, alpha-bungarotoxin으로 염색한 후 형광현미경으로 이미징 분석을 수행하였다 (도 8).

그 결과, PF1802_M008 후보물질을 투여한 G4, G5 그룹에서 용량의존적으로 완전히 신경화된 신경근 접합부 (Fully innervated NMJ)는 증가하였으며, 탈신경 신경근 접합부 (denervated NMJ)는 감소하는 것을 확인하였다.
이를 통해, PF1802_M008 후보물질을 처리한 경우 근위축성 측상경화증 질환에 대한 예방 및 진행 지연 효과가 있음을 알 수 있었다.
실시예 4. PF1802 후보물질의 최적화
실시예 4-1. PF1802_M008 후보물질의 구조 분석 및 최적화 물질 제조
실시예 2. 및 실시예 3.을 통하여 효능이 확인된 PF1802_M008 후보물질의 최적화를 위하여, 각기 다른 3가지 linker를 도입하여 M023, M024, M025 후보물질을 디자인하였으며, 실시예 1.과 동일한 방법으로 진행하여 물질을 제조하였다. 그 다음, 제조된 M023, M024, M025 후보물질을 비환원 및 환원 조건의 SDS-PAGE gel을 수행한 결과, cleavage form으로 확인되는 M023을 제외하고, M024, M025 물질은 이상 없음을 확인하였다 (도 9).
실시예 4-2. SPR을 이용한 PF1802_M024, M025 후보물질의 결합력 평가
실시예 4-1.에서 제조한 최적화가 완료된 PF1802_M024, M025의 GLP-1R, IGF-1R에 대한 결합력 확인을 위해 SPR을 이용하여 결합에 따른 신호 변화를 측정하였다. 구체적으로, CM5 sensor chip을 BIAcore T200(GE Healthcare)에 장착한 후, 100mM N-hydroxysuccinimide(NHS)와 400mM 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC)를 1:1 비율로 주입하여 flow cell을 활성화 한 다음, anti-His tag 항체를 8000 ~ 10000 RU 수준으로 고정화하였다. 리간드로 사용한 GLP-1R-His, IGF-1R-His 재조합 단백질은 각각 매 사이클 마다 2.5 ug/mL 농도로 주입하여 280 ~ 290 RU 수준으로 capture 하였으며, 이 후 M024, M025 후보물질을 표 9의 조건으로 주입하여 SPR 결합력을 측정 하였다 (도 10).

그 결과, M024 후보물질의 SPR 결합력은 12.2 nM (GLP-1R), 18.1 nM (IGF-1R)이며, M025 후보물질의 SPR 결합력은 10.6 nM (GLP-1R), 20.7 nM(IGF-1R)임을 확인하였다.
이를 통해, M024, M025 후보물질은 모두 GLP-1R, IGF-1R에 결합한다는 점을 알 수 있었으며, 두 물질간의 결합력 차이는 없는 것으로 유의한 차이가 존재하지 않음을 확인하였다. 이는 M024, M025 후보물질은 GLP-1 및 IGF-1 활성이 우수함을 시사하는 바이다.
실시예 5. PF1802_M024 후보물질의 효능 검정
실시예 5-1. ALS 유전자 변형 마우스 모델에서 PF1802_M024 후보물질의 효능 검정
실시예 4.를 통하여 물질 최적화가 완료된 PF1802_M024 후보물질의 효능 확인을 위해, 표 10과 같이 생후 10주된 SOD-1(G39A) 마우스에 PF1802_M024 후보물질을 주 2회씩 8주 동안 피하투여 하였다. 면역조직학적 분석을 위해 생후 115일에 마우스를 희생하였다. 그 후, 신경근 접합부(Neuromuscular junction) 평가를 위해 비복근 근육(gastronemius muscle) 샘플에서 anti-synatotagmin 2 Ab, alpha-bungarotoxin으로 염색하였으며 (도 11), 신경 염증 확인을 위해 척수 (spinal cord) 샘플에서 anti-Iba1 Ab로 염색한 후 (도 12), 형광현미경으로 이미징 분석을 수행하였다.

그 결과, PF1802_M024 후보물질을 투여한 G4, G5 그룹에서 완전히 신경화된 신경근접합부 (Fully innervated NMJ)는 증가하였으며, 탈신경 신경근접합부 (denervated NMJ)는 감소함을 확인하였다. 또한, G4, G5 그룹에서 염증 여부를 확인 할 수 있는 Iba1가 무처리 대비 현저히 감소하고, 양성 대조군과 유사한 활성을 나타냄을 확인하였다.
이를 통해, PF1802_M024 후보물질이 NMJ 개선 및 Iba1 감소 결과를 통해 PF1802 후보물질이 근위축성 측상경화증 질환에 대한 예방 및 지연효과가 있는 것을 알 수 있었으며, 동시에 척수샘플에서 Iba1 감소 결과는 PF1802_M024 후보물질이 BBB를 통과한다는 것을 증명할 수 있는 결과이기도 하다.
실시예 5-2. PF1802_M024 후보물질의 Mouse BBB 투과율 평가
BBB 투과율 확인을 위하여, Alexa fluor 647 antibody labeling kit(PerkinElmer)을 사용하여 형광 표지된 물질을 Cranial imaging window 마우스 모델(C57BL/6N)에 1회 정맥투여 한 후 시간별로 이미징을 확보하였다. 뇌 vessel 부위는 빨간색으로 (도 13a), 시험물질(PF1802 & Human IgG)은 초록색으로 (도 13b)로 표시하였으며, cortex는 보라색으로 이는 뇌 vessel과 시험물질이 merge된 이미지에서 확인할 수 있다. (도 13c)
그 결과, PF1802_M024 후보물질은 Human IgG 대비 BBB 투과율이 우수함을 확인하였다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (7)
- GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질.
- 제1항에 있어서, 상기 면역글로불린 Fc 단편은 IgG, IgA, IgD, IgE, IgM, 이들의 조합(combination) 및 이들의 하이브리드(hybrid)로 이루어진 군으로부터 선택되는 것인, 융합단백질.
- 제1항에 있어서, 상기 면역글로불린 Fc 단편은 IgG1 유래인 것인, 융합단백질.
- GLP-1, 면역글로불린 Fc, 및 IGF-1을 포함하는 융합단백질을 유효성분으로 포함하는 신경계 질환의 예방 또는 치료용 약학적 조성물.
- 제4항에 있어서, 상기 신경계 질환은 파킨슨병, 알츠하이머 치매(노인성 치매), 뇌졸중, 루게릭병, 픽크병, 크로이츠펠트 야콥병, 헌팅톤병, 진행성 핵상마비, 척수소뇌 변성증, 소뇌 위축증, 다발성 경화중, 근위축성 측삭경화증, 말초신경병증, 중증근무력증 및 척수성 근위축증으로 이루어진 군에서 선택되는 어느 하나인, 약학적 조성물.
- 제4항에 있어서, 상기 조성물은 신경근 접합부 (Neuromuscular junction, NMJ)의 기능 회복 또는 개선 효과를 갖는 것인, 약학적 조성물.
- 제4항의 조성물을 개체에 투여하는 단계를 포함하는, 신경계 질환 예방 또는 치료방법.
