Both tocopherols and tocotrienols include a chroman ring and a 16-carbon side chain in their structure.   • High glucose Dulbecco's Modified Eagle Medium (DMEM Glutamax, Fisher, Spain) was used for keratinocyte culture and DMEM (Gibco, Spain) for fibroblast culture; both were supplemented with 2% penicillin/streptomycin and 50 mL of fetal bovine serum (FBS) in a humidified incubator (Hucoa-Erlbss S.A., Spain) at 5% CO2 and 37 °C.

Images were captured at an 8-bit greyscale and processed with LOS software (version 2.5.1347a, Leica, Germany) containing multicolor, macro and 3D components. Control tissue that follows the same incubation time with MilliQ water was also analyzed to establish the autofluorescence. Dll intensity was quantified five times per sample using Image J software.

  • MicroVue SC5b-9 Plus an immunoassay kit MicroVue SC5b-9 Plus. Sera of three healthy individuals was used for further testing. The concentration employed 0.2 mg/mL VitE equivalents. Hepes Buffered Saline (HBS) was used as a negative control and Zymosan (0.25 mg/mL) as a positive control. All samples were incubated by mixing 1 part polymer suspension with 4 parts human serum for 1 h at 37 °C under constant agitation. Following the incubation samples were diluted 100-fold (700-fold for Zymosan) with manufacturers dilution buffer and the assay was conducted as described in the manufacturers protocol. Table 13 shows the SC5b-9 amounts and standard error mean (SEM):
  • iPrPSar(20)-succ-VitE shows 10x less immunogenic response than the positive control zymosan. And comparable results to negative control HBS. Therefore, iPrPSar(20)-succ-VitE has no immunogenic response in these conditions.
