Abstract
粒子径が小さく、粒度分布が狭いゲル粒子を含む3D印刷用支持材料が提供される。ゲル粒子と、ゲル粒子安定化剤と、を含み、前記ゲル粒子の個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50が1μm以上500μm以下であり、前記粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)が7以下である3D印刷用支持材料である。
Description
本発明は、3D印刷用支持材料及びその製造方法に関する。
3次元(3D)印刷は、複雑な構造を構築するための強力なプラットフォームとして登場した革新的な技術であり、多彩な用途に適用可能である。従来の3D印刷は通常、材料を層状に付加することによって基板上に3D構造を構築する。しかしながら、この方法を直接適用して、軟質で含水性の生体材料を印刷することは困難である。これは重力により、印刷プロセス中に印刷構造が崩壊する可能性があるためである。この欠点を克服するために、支持媒体で構成されるバスで支持される3D印刷が開発されている。さまざまなバスシステムの中で、ゲル粒子バスは、水分含有量が高く、流体状態と固体状態の間の移行がスムーズであるため、バイオプリンティングで頻繁に使用される支持材料の1つである。
上記に関連して、例えば特表2021-512742号公報及びScience,365,482-487(2019)には、支持材料としてゼラチン微粒子スラリーを含むハイドロゲルが記載されている。
ゲル粒子バスを支持材料とする3D印刷の解像度は、ゲル粒子の粒子径とその粒度分布に依存する。従来技術では、粒子径が小さく、粒度分布が狭いゲル粒子を、簡便かつ大量に調製することは困難であった。
本発明の一態様は、粒子径が小さく、粒度分布が狭いゲル粒子を含む3D印刷用支持材料及びその製造方法を提供することを目的とする。
前記課題を解決するための具体的手段は以下の通りであり、本発明は以下の態様を包含する。第1態様は、ゲル粒子と、ゲル粒子安定化剤と、を含み、前記ゲル粒子の個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50が1μm以上500μm以下であり、前記粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)が7以下である3D印刷用支持材料である。
前記ゲル粒子は、ジェランガム、アルギン酸、ゼラチン、コラーゲン、アラビアガム、ジャンタンガム、セルロース、ヒアルロン酸、ラミニン、マトリゲル(商品名)、ポリアクリル酸、ポリ(N-イソプロピルアクリルアミド)、ポリメタクリル酸、ポリアクリルアミド、ポリスチレンスルホン酸、ポリビニルアルコール、ポリエチレングリコールゲル、デンプン及びフィブリンからなる群から選択される少なくとも1種を含んでいてよい。また、前記ゲル粒子安定化剤は、カルボン酸化合物、水溶性有機溶剤及び塩からなる群から選択される少なくとも1種を含んでいてよい。
第2態様は、ゲル及びゲル粒子安定化剤溶液を含む混合物を準備することと、前記混合物に剪断力を付与して、ゲル粒子分散物を得ることと、を含む3D印刷用支持材料の製造方法である。
前記ゲルは、ジェランガム、アルギン酸、ゼラチン、コラーゲン、アラビアガム、ジャンタンガム、セルロース、ヒアルロン酸、ラミニン、マトリゲル(商品名)、ポリアクリル酸、ポリ(N-イソプロピルアクリルアミド)、ポリメタクリル酸、ポリアクリルアミド、ポリスチレンスルホン酸、ポリビニルアルコール、ポリエチレングリコール、デンプン及びフィブリンからなる群から選択される少なくとも1種を含んでいてよい。また、前記ゲル粒子安定化剤は、カルボン酸化合物、水溶性有機溶剤及び塩からなる群から選択される少なくとも1種を含んでいてよい。また、前記ゲル粒子は、個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50が1μm以上500μm以下であり、前記粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)が7以下であってよい。
本発明によれば、粒子径が小さく、粒度分布が狭いゲル粒子を含む3D印刷用支持材料及びその製造方法を提供することができる。
本明細書において「工程」との語は、独立した工程だけではなく、他の工程と明確に区別できない場合であってもその工程の所期の目的が達成されれば、本用語に含まれる。また組成物中の各成分の含有量は、組成物中に各成分に該当する物質が複数存在する場合、特に断らない限り、組成物中に存在する当該複数の物質の合計量を意味する。さらに本明細書に記載される数値範囲の上限及び下限は、数値範囲として例示された数値をそれぞれ任意に選択して組み合わせることが可能である。以下、本発明の実施形態を詳細に説明する。ただし、以下に示す実施形態は、本発明の技術思想を具体化するための、3D印刷用支持材料及びその製造方法を例示するものであって、本発明は、以下に示す3D印刷用支持材料及びその製造方法に限定されない。
3D印刷用支持材料
3D印刷用支持材料は、ゲル粒子と、ゲル粒子安定化剤とを含んで構成される。3D印刷用支持材料を構成するゲル粒子の個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50は、例えば1μm以上500μm以下であってよい。また粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)は、例えば7以下であってよい。
3D印刷用支持材料は、ゲル粒子と、ゲル粒子安定化剤とを含んで構成される。3D印刷用支持材料を構成するゲル粒子の個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50は、例えば1μm以上500μm以下であってよい。また粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)は、例えば7以下であってよい。
3D印刷用支持材料は、ゲル粒子に加えてゲル粒子安定化剤を含むことで、所定の個数平均径D50と、所定のD90/D10を実現することができる。これにより、3D印刷において、優れた解像度で所望の構造体を製造することができる。また、3D印刷用支持材料は、後述する製造方法で効率的に製造することができる。
ゲル粒子の個数平均径D50は、好ましくは0.1μm以上、10μm以上、15μm以上、又は20μm以上であってよい。またゲル粒子の個数平均径D50は、好ましくは200μm以下、50μm以下、40μm以下、又は30μm以下であってよい。また、ゲル粒子のD90/D10は、例えば10以下、7以下、6以下、5以下、4以下、又は3以下であってよい。ゲル粒子のD90/D10の下限は、例えば1以上、又は2以上であってよい。
ゲル粒子の粒度分布における個数累積10%に対応する粒径であるD10は、例えば5μm以上50μm以下であってよい。D10は、好ましくは8μm以上、10μm以上、又は12μm以上であってよく、また好ましくは40μm以下、30μm以下、20μm以下、又は16μm以下であってよい。ゲル粒子の粒度分布における個数累積90%に対応する粒径であるD90は、例えば20μm以上100μm以下であってよい。D90は、好ましくは25μm以上、30μm以上、又は35μm以上であってよく、また好ましくは80μm以下、60μm以下、40μm以下、又は38μm以下であってよい。
ゲル粒子の個数基準の粒度分布は、例えば、3D印刷用支持材料を共焦点レーザー走査顕微鏡で観察して得られる顕微鏡像において、画像解析ソフトウエアを用いて個々のゲル粒子の粒径を測定することで得ることができる。
3D印刷用支持材料に含まれるゲル粒子を構成するゲル材料としては、3D印刷で形成される構造体の形成に悪影響を及ぼさない限り、イオンの添加、温度変化等によってゲル化できる任意の物質を用いることができる。そのようなゲル材料としては、ジェランガム(GG)等のガム類、アルギン酸、ポリアクリル酸、ポリグルタミン酸、ポリアスパラギン酸、ゼラチン、コラーゲン、アラビアガム、ジャンタンガム、セルロース、ヒアルロン酸、ラミニン、マトリゲル(商品名)、ポリアクリル酸、ポリ(N-イソプロピルアクリルアミド)、ポリメタクリル酸、ポリアクリルアミド、ポリスチレンスルホン酸、ポリビニルアルコール、ポリエチレングリコール、デンプン及びフィブリン等が挙げられる。ゲル材料は、好ましくはジェランガム、アルギン酸、ゼラチン、コラーゲン、アラビアガム、ジャンタンガム、セルロース、ヒアルロン酸、ラミニン、マトリゲル(商品名)、ポリアクリル酸、ポリ(N-イソプロピルアクリルアミド)、ポリメタクリル酸、ポリアクリルアミド、ポリスチレンスルホン酸、ポリビニルアルコール、ポリエチレングリコール、デンプン及びフィブリンからなる群から選択される少なくとも1種を含んでいてよい。
ゲル材料は、より好ましくはジェランガム、コラーゲン、ジャンタンガム、ヒアルロン酸、ラミニン、マトリゲル(商品名)、ポリアクリル酸、ポリ(N-イソプロピルアクリルアミド)、ポリメタクリル酸、ポリアクリルアミド、ポリスチレンスルホン酸、ポリビニルアルコール、ポリエチレングリコール、デンプン及びフィブリンからなる群から選択される少なくとも1種を含んでいてよい。
ゲル粒子を構成するゲル材料はハイドロゲルであってもよい。ゲル粒子を構成するゲル材料は1種類単独であってもよく、2種以上の組み合わせであってもよい。なお、マトリゲルはBD Biosciences社から発売されている細胞外マトリクスの商品名であり、ラミニン、ニドジェン、コラーゲン、ヘパラン硫酸プロテオグリカン等の混合物である。
ゲル粒子を構成するゲル材料は、好ましくはガム類であってよい。ガム類には、植物由来ガム類、細菌由来ガム類、藻類由来ガム類等が含まれる。植物由来ガム類の具体例としては、グアーガム、ローカストビーンガム、カッシアガム、トラガカントゴム、タラガム、カラヤゴム、アカシアゴム、ガティガム、チェリーガム、カシューガム、アンズガム、タマリンドガム、メスキートガム、カラマツガム、オオバコ、コロハガム等が挙げられる。細菌および藻類に由来するガム類としては、キサンタンガム、海草ガム、ジェランガム、アガーガム、カラゲナン、カードラン等が挙げられる。
ガム類のうち、ジェランガム(GG)は、シュードモナス・エロディア(Pseudomonas elodea)がブドウ糖などを炭素源として細胞外に産生する、直鎖状の天然高分子多糖類である。GGは1価または2価の金属塩の存在下で、透明性、耐熱性、耐酸性のあるゲルを形成する。GGには、高アシル基含有のHAジェランガムと、アシル基が除去されたLAジェランガムが存在するが、本発明ではいずれを用いてもよい。あるいは、両方のGGを組み合わせて用いてもよい。GGは、例えばNanogel(登録商標)-TC、Grovgel、AppliedGel、Phytagel(商標)またはGelriteとして市販されている。
ゲル粒子はゲル材料に加えて、ゲル材料に担持される液体を含んでいてよい。ゲル材料に担持される液体は、ゲル材料の種類等に応じて適宜選択することができる。ゲル材料に担持される液体として具体的には、例えば、水、リン酸緩衝生理食塩水等が挙げられる。
ゲル粒子におけるゲル材料の含有率は、ゲル材料の種類、ゲル材料が担持する液体の種類等に応じて適宜選択することができる。ゲル粒子におけるゲル材料の含有率は、例えば、0.01質量%以上30質量%以下であってよい。ゲル粒子におけるゲル材料の含有率は、好ましくは0.1質量%以上、又は0.3質量%以上であってよく、また好ましくは10質量%以下、5質量%以下、2質量%以下、1質量%以下、又は0.8質量%以下であってよい。
3D印刷用支持材料に含まれるゲル粒子安定剤としては、ゲルからゲルを構成する溶媒の少なくとも一部を除去する性質を示すものであれば特に制限はない。また、ゲル粒子安定剤はゲル粒子を構成するゲル材料に応じて適宜されてもよい。ゲル粒子安定化剤は水溶性を有することが好ましい。ここで水溶性とは100gの純水に対する25℃における溶解度が1g以上であることを意味する。
ゲル粒子安定剤として具体的には、カルボン酸化合物、水溶性有機溶剤、塩等が挙げられる。また、カルボン酸化合物として具体的には、乳酸、グリコール酸等のモノカルボン酸、リンゴ酸等のジカルボン酸、クエン酸等のトリカルボン酸等が挙げられる。ここでカルボン酸化合物は、アルカリ金属、アルカリ土類金属等との塩として用いられてもよい。ゲル粒子安定剤としての水溶性有機溶剤としては、エタノール、プロパノール、イソプロパノール等のアルコール類、アセトニトリル等のニトリル類等が挙げられる。塩としては、塩化カルシウム、塩化ナトリウム、塩化カリウム、塩化マグネシウム、塩化アルミニウム、硫酸ナトリウム、硫酸カルシウム、硫酸マグネシウム、硫酸カリウム、硝酸ナトリウム、硝酸カリウム、硝酸カルシウム、炭酸ナトリウム、炭酸水素ナトリウム、炭酸カリウム、炭酸水素カリウム等のアルカリ金属塩、アルカリ土類金属塩が挙げられる。
ゲル粒子安定剤は、ゲル粒子の分散安定性の観点から、カルボン酸化合物が好ましく、ジカルボン酸、トリカルボン酸等の多価カルボン酸化合物がより好ましい。
3D印刷用支持材料は、ゲル粒子及びゲル粒子安定化剤に加えて、液媒体をさらに含んでいてよい。液媒体は、ゲル粒子安定化剤を可溶であればよく、例えば水を含んでいてよい。液媒体におけるゲル粒子安定化剤の濃度は、ゲル粒子安定化剤の種類等に応じて適宜選択すればよい。例えば、ゲル安定化剤がカルボン酸化合物の場合、液媒体におけるゲル粒子安定化剤の濃度は、0.1mM以上1M以下であってよい。ゲル安定化剤がカルボン酸化合物の場合の液媒体におけるゲル粒子安定化剤の濃度は、好ましくは10mM以上、100mM以上、200mM以上、300mM以上、又は400mM以上であってよく、また好ましくは0.7M以下、0.6M以下、又は0.5M以下であってよい。例えば、ゲル安定化剤が水溶性有機溶剤の場合、液媒体におけるゲル粒子安定化剤の濃度は、1体積%以上99体積%以下であってよい。ゲル安定化剤が水溶性有機溶剤の場合の液媒体におけるゲル粒子安定化剤の濃度は、好ましくは5体積%以上、10体積%以上、20体積%以上、25体積%以上、又は30体積%以上であってよく、また好ましくは60体積%以下、50体積%以下、40体積%以下、又は35体積%以下であってよい。
3D印刷用支持材料が液媒体を含む場合、3D印刷用支持材料におけるゲル粒子の濃度は、例えば、1体積%以上99体積%以下であってよい。液媒体を含む3D印刷用支持材料におけるゲル粒子の濃度は、好ましくは10体積%以上、又は30体積%以上であってよく、また好ましくは80体積%以下、又は70体積%以下であってよい。
3D印刷用支持材料は、必要に応じて界面活性剤を含んでいてもよい。界面活性剤は、ノニオン性、カチオン性、アニオン性、両性などのいずれであってもよく、好ましくはノニオン性であってよい。ノニオン性界面活性剤としては、例えばポリオキシアルキレンアルキルエーテル、ポリオキシエチレンポリオキシプロピレンアルキルエーテル(エチレンオキサイドとプロピレンオキサイドの付加形態は、ランダム状、ブロック状の何れでもよい)、ポリエチレングリコールプロピレンオキサイド付加物、ポリプロピレングリコールエチレンオキサイド付加物、グリセリン脂肪酸エステルまたはそのエチレンオキサイド付加物等が挙げられる。3D印刷用支持材料のおける界面活性剤の含有量は、例えば0質量%を超えて5質量%以下であってよい。界面活性剤の含有量は、好ましくは0.01質量%以上、又は0.1質量%以上であってよい。また界面活性剤の含有量は、好ましくは2質量%以下、1質量%以下、0.5質量%以下、0.1質量%以下、又は0.01質量%以下であってよい。
3D印刷用支持材料は、3D印刷に用いることができる。例えば3D印刷は、3D印刷用支持材料中に3D印刷用インクを所望の形状に吐出することで行うことができる。3D印刷用インクとしては、例えばコラーゲン、フィブリノーゲン、ゼラチン、アルギン酸等の有機物、シリカ、ナノクレイ等の無機物を含むものであってよい。
3D印刷用支持材料の製造方法
3D印刷用支持材料の製造方法は、ゲル及びゲル粒子安定化剤を含む混合物を準備する準備工程と、準備された混合物に剪断力を付与して、ゲル粒子を得る分散工程と、を含んでいてよい。分散工程で得られるゲル粒子の分散物は、3D印刷用支持材料を構成してよい。
3D印刷用支持材料の製造方法は、ゲル及びゲル粒子安定化剤を含む混合物を準備する準備工程と、準備された混合物に剪断力を付与して、ゲル粒子を得る分散工程と、を含んでいてよい。分散工程で得られるゲル粒子の分散物は、3D印刷用支持材料を構成してよい。
ゲル粒子安定化剤の存在下で、ゲルに剪断力を付与することで、粒子径が小さく、粒度分布が狭いゲル粒子を含む3D印刷用支持材料を、優れた生産性で効率的に製造することができる。なお、ゲル粒子安定化剤の不存在下で、ゲルに剪断力を付与することでゲル粒子を調製する場合、得られるゲル粒子の粒度分布は、比(D90/D10)が7を超える広い粒度分布を示すことになる。
混合物に含まれるゲルは、既述のゲル材料に既述の液体を担持させて形成したものであればよく、ゲルを構成するゲル材料及び液体は、目的等に応じて適宜選択すればよい。ゲルは例えば、ゲル材料を液体と混合し、加温してゲル材料を液体に溶解した後、降温してゲル化させることで調製することができる。また、ゲルは化学反応で調製してもよい。
混合物を構成するゲルは、粒子状であってもよい。粒子状のゲルは、例えば、調製されたゲルに適当な剪断力を付与することで調製することができる。剪断力の付与方法としては、例えば、ホモジナイザーを用いる方法、超音波処理、フィルターメッシュを用いる方法等を挙げることができる。
混合物に含まれるゲル粒子安定化剤の詳細については既述の通りである。混合物は、ゲル粒子及びゲル粒子安定化剤に加えて、液媒体をさらに含んでいてよい。液媒体は、ゲル粒子安定化剤を可溶であればよく、例えば水を含んでいてよい。液媒体におけるゲル粒子安定化剤の濃度は、ゲル粒子安定化剤の種類等に応じて適宜選択すればよい。例えば、ゲル安定化剤がカルボン酸化合物の場合、液媒体におけるゲル粒子安定化剤の濃度は、0.1mM以上1M以下であってよい。ゲル安定化剤がカルボン酸化合物の場合の液媒体におけるゲル粒子安定化剤の濃度は、好ましくは10mM以上、100mM以上、200mM以上、300mM以上、又は400mM以上であってよく、また好ましくは0.7M以下、0.6M以下、又は0.5M以下であってよい。例えば、ゲル安定化剤が水溶性有機溶剤の場合、液媒体におけるゲル粒子安定化剤の濃度は、1体積%以上99体積%以下であってよい。ゲル粒子安定化剤が水溶性有機溶剤の場合の液媒体におけるゲル粒子安定化剤の濃度は、好ましくは5体積%以上、10体積%以上、20体積%以上、25体積%以上、又は30体積%以上であってよく、また好ましくは60体積%以下、50体積%以下、40体積%以下、又は35体積%以下であってよい。
混合物が液媒体を含む場合、混合物におけるゲルの濃度は、例えば、1体積%以上99体積%以下であってよい。混合物におけるゲルの濃度は、好ましくは10体積%以上、又は30体積%以上であってよく、また好ましくは80体積%以下、又は70体積%以下であってよい。
分散工程では、準備された混合物に剪断力を付与して、ゲル粒子を得る。剪断力の付与方法としては、例えば、ホモジナイザーを用いる方法、超音波処理、フィルターメッシュを用いる方法等を挙げることができる。
分散工程で得られるゲル粒子は、個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50が1μm以上500μm以下であってよい。また、ゲル粒子は粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)が7以下であってよい。分散工程で得られるゲル粒子の詳細は、既述の3D印刷用支持材料と同様であってよい。
以下、本発明を実施例により具体的に説明するが、本発明はこれらの実施例に限定されるものではない。
(実施例1)
200mgのジェランガム(GG)粉末(商品名:KELCOGEL AFT;三晶株式会社製)を40mLのリン酸緩衝生理食塩水(PBS)に加え、100℃で3時間加熱して溶解してGG溶液を得た。溶解後、GG溶液を室温で3時間維持してゲル化させた。次に、GGゲルをホモジナイザーで6分間処理して粉砕してGG粒子を得た。
200mgのジェランガム(GG)粉末(商品名:KELCOGEL AFT;三晶株式会社製)を40mLのリン酸緩衝生理食塩水(PBS)に加え、100℃で3時間加熱して溶解してGG溶液を得た。溶解後、GG溶液を室温で3時間維持してゲル化させた。次に、GGゲルをホモジナイザーで6分間処理して粉砕してGG粒子を得た。
粉砕したGG粒子の10mLに、4.2mLの0.5Mクエン酸三ナトリウム水溶液を加えた後、ホモジナイザーで6分間処理した。次いで2000rpm、3分間で遠心分離して気泡を除去し、3D印刷用支持材料(0.15M TSC)を得た。
(実施例2)
1Mクエン酸三ナトリウム水溶液を用いたこと、及びクエン酸三ナトリウムの終濃度を0.3Mに調整したこと以外は実施例1と同様にして、3D印刷用支持材料(0.3M TSC)を得た。

(実施例3)
1.5Mクエン酸三ナトリウム水溶液を用いたこと、及びクエン酸三ナトリウムの終濃度を0.45Mに調整したこと以外は実施例1と同様にして、3D印刷用支持材料(0.45M TSC)を得た。
1.5Mクエン酸三ナトリウム水溶液を用いたこと、及びクエン酸三ナトリウムの終濃度を0.45Mに調整したこと以外は実施例1と同様にして、3D印刷用支持材料(0.45M TSC)を得た。
(実施例4)
2Mクエン酸三ナトリウム水溶液を用いたこと、及びクエン酸三ナトリウムの終濃度を0.6Mに調整したこと以外は実施例1と同様にして、3D印刷用支持材料(0.6M TSC)を得た。

(実施例5)
クエン酸三ナトリウム水溶液の代わりに99%エタノール水溶液を2.5mL用いたこと、エタノールの終濃度を20%に調整したこと以外は実施例1と同様にして、3D印刷用支持材料(20% EtOH)を得た。

クエン酸三ナトリウム水溶液の代わりに99%アセトニトリル水溶液を4.2mL用いたこと、アセトニトリルの終濃度を30%に調整したこと以外は実施例1と同様にして、3D印刷用支持材料(ACN)を得た。

(比較例1)
クエン酸三ナトリウム水溶液の代わりにリン酸緩衝生理食塩水(PBS)を4.2mL用いたこと以外は実施例1と同様にして、3D印刷用支持材料(PBS)を得た。

評価
フルオレセイニルグリシンアミド修飾GG粉末を用いたこと以外は、実施例3と同様にして顕微鏡観察用の3D印刷用支持材料1を得た。また、クエン酸三ナトリウム(TSC)水溶液の代わりにPBSを用いて、同様にして顕微鏡観察用の3D印刷用支持材料2を得た。得られた3D印刷用支持材料を共焦点レーザー走査顕微鏡(CLSM、FV-3000)で観察して顕微鏡画像をそれぞれ得た。結果を図1及び図2に示す。
フルオレセイニルグリシンアミド修飾GG粉末を用いたこと以外は、実施例3と同様にして顕微鏡観察用の3D印刷用支持材料1を得た。また、クエン酸三ナトリウム(TSC)水溶液の代わりにPBSを用いて、同様にして顕微鏡観察用の3D印刷用支持材料2を得た。得られた3D印刷用支持材料を共焦点レーザー走査顕微鏡(CLSM、FV-3000)で観察して顕微鏡画像をそれぞれ得た。結果を図1及び図2に示す。
上記で得られた顕微鏡画像から、画像解析ソフトウエア(Image-J)を用いて、GG粒子の大きさを測定して、個数基準の粒度分布を得た。結果を図3及び図4に示す。
エタノールの終濃度を20%又は30%に調整して得られた3D印刷用支持材料について、同様にして粒度分布を得た。結果を図5及び図6に示す。
上記で得られた粒度分布から、個数平均径D50と、比(D90/D10)を求めた。結果を表1に示す。
コラーゲンインクの調製
コラーゲン粉末(ニッピ株式会社製)を0.02M酢酸に加え、ホモジナイザーで6分間処理した後、4℃で24時間維持して完全に溶解させた。得られたコラーゲン溶液を4000rpm、5分間の遠心分離により気泡除去を行って、コラーゲン濃度が10mg/mLであるコラーゲンインクを調製した。
コラーゲン粉末(ニッピ株式会社製)を0.02M酢酸に加え、ホモジナイザーで6分間処理した後、4℃で24時間維持して完全に溶解させた。得られたコラーゲン溶液を4000rpm、5分間の遠心分離により気泡除去を行って、コラーゲン濃度が10mg/mLであるコラーゲンインクを調製した。
印刷プロセス1
25℃の3D印刷用支持材料を入れた浴中に、20ゲージ(内径600μm)のノズルからコラーゲンインクを直線状に吐出した。ノズルの移動速度は2mm/sとした。浴中の印刷されたコラーゲンファイバーを室温で1時間保持してゲル化させた。ファイバーをピンセットで取り出し、50%エタノール溶液で洗浄してGG粒子を除去した。得られたファイバーを50%エタノール溶液中の0.5v/v%グルタルアルデヒドに37℃で24時間浸漬して架橋させた。架橋後、サンプルをPBSに24時間浸漬して洗浄し、機械的試験に供した。
25℃の3D印刷用支持材料を入れた浴中に、20ゲージ(内径600μm)のノズルからコラーゲンインクを直線状に吐出した。ノズルの移動速度は2mm/sとした。浴中の印刷されたコラーゲンファイバーを室温で1時間保持してゲル化させた。ファイバーをピンセットで取り出し、50%エタノール溶液で洗浄してGG粒子を除去した。得られたファイバーを50%エタノール溶液中の0.5v/v%グルタルアルデヒドに37℃で24時間浸漬して架橋させた。架橋後、サンプルをPBSに24時間浸漬して洗浄し、機械的試験に供した。
3D印刷用支持材料(0.45M TSC)を用いて得られたファイバーの顕微鏡画像を図7に示す。また、顕微鏡画像から測定されたファイバーの長さ方向における幅変化を図8に示す。なお、得られたファイバーの平均径は、約800μmであった。また、得られた架橋後のファイバーの引張強度は約300kPaであった。
3D印刷用支持材料(30% EtOH)を用いて、同様にして得られた架橋後のファイバーの引張強度は約200kPaであった。なお、3D印刷用支持材料(PBS)を用いて同様にしてファイバーを作製したところ、浴中から取り出すことができなかった。
印刷プロセス1
25℃の3D印刷用支持材料(0.3M TSC)を入れた浴中に、25ゲージ(内径260μm)のノズルを装着した3Dプリンタ(Bio-X)を用いて、プログラムされたルートでコラーゲンインクを吐出して3次元の構造体を形成した。形成された構造体を図9A及び図9Bに示す。
25℃の3D印刷用支持材料(0.3M TSC)を入れた浴中に、25ゲージ(内径260μm)のノズルを装着した3Dプリンタ(Bio-X)を用いて、プログラムされたルートでコラーゲンインクを吐出して3次元の構造体を形成した。形成された構造体を図9A及び図9Bに示す。
以上から、本発明の3D印刷用支持材料を浴として用いることで、高解像度3D印刷を実現できることが分かる。

Claims (7)
- ゲル粒子と、ゲル粒子安定化剤と、を含み、
前記ゲル粒子の個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50が1μm以上500μm以下であり、
前記粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)が7以下である3D印刷用支持材料。 - 前記ゲル粒子は、ジェランガム、アルギン酸、ゼラチン、コラーゲン、アラビアガム、ジャンタンガム、セルロース、ヒアルロン酸、ラミニン、マトリゲル(商品名)、ポリアクリル酸、ポリ(N-イソプロピルアクリルアミド)、ポリメタクリル酸、ポリアクリルアミド、ポリスチレンスルホン酸、ポリビニルアルコール、ポリエチレングリコール、デンプン、及びフィブリンからなる群から選択される少なくとも1種を含む請求項1に記載の支持材料。
- 前記ゲル粒子安定化剤は、カルボン酸化合物、水溶性有機溶剤及び塩からなる群から選択される少なくとも1種を含む請求項1又は2に記載の支持材料。
- ゲル及びゲル粒子安定化剤溶液を含む混合物を準備することと、
前記混合物に剪断力を付与して、ゲル粒子分散物を得ることと、を含む3D印刷用支持材料の製造方法。 - 前記ゲルは、ジェランガム、アルギン酸、ゼラチン、コラーゲン、アラビアガム、ジャンタンガム、セルロース、ヒアルロン酸、ラミニン、マトリゲル(商品名)、ポリアクリル酸、ポリ(N-イソプロピルアクリルアミド)、ポリメタクリル酸、ポリアクリルアミド、ポリスチレンスルホン酸、ポリビニルアルコール、ポリエチレングリコール、デンプン及びフィブリンからなる群から選択される少なくとも1種を含む請求項4に記載の製造方法。
- 前記ゲル粒子安定化剤は、カルボン酸化合物、水溶性有機溶剤及び塩からなる群から選択される少なくとも1種を含む請求項4又は5に記載の製造方法。
- 前記ゲル粒子は、個数基準の粒度分布における個数累積50%に対応する粒径である個数平均径D50が1μm以上500μm以下であり、
前記粒度分布における個数累積10%に対応する粒径であるD10に対する個数累積90%に対応する粒径であるD90の比(D90/D10)が7以下である請求項4から6のいずれか1項に記載の製造方法。
