Description
本発明は、キチン結合ポリペプチド、ならびに、当該キチン結合ポリペプチドを用いたキチンの検出方法および検出キットに関する。
キチン(Chitin)はポリ-β-1,4-N-アセチルグルコサミンにより構成される窒素含有多糖であり、キノコおよびカビなどの真菌細胞壁、節足動物および甲殻類の外骨格、ならびに、イカなどの軟体動物など、多くの生物に含まれる。国内外で、キチンを利用した食品、化粧品、医薬品およびバイオプラスチックなどへの応用開発が活発に進められている。
キチンは、多様な可能性を持つ有益な天然素材である一方、例えばダニ、ゴキブリ、寄生虫および真菌などに含まれ、ヒトの免疫系に干渉してアレルギーを誘導する分子でもある。キチンの検出に当たり、ダニ自体または真菌自体を検出する技術があるが、実際にヒトの疾患に大きく関与しているのは、これらの死骸および破砕後の微粒子であるため、目視で判別することが難しい。よって、目視での判別が難しい、このようなアレルギー誘導分子を検出する別の技術が求められている。
キチンの構造解析は、複数の工程を経て精製したキチンに対して、例えば、NMR(nuclear magnetic resonance)、HPLC(High performance liquid chromatography)およびGC(Gas chromatograph)などの分析方法を用いて行われる。キチンの定量法としては、プロスキー法(酵素-重量法)のほか、化学的な処理によりキチンを分解し、生成するグルコサミンを定量する方法などが用いられる(特許文献1)。しかしながら、これらの方法では、アミノ酸および糖類などの夾雑物存在下では、測定感度および特異性が低下するため、多くの場合、試験検体の精製プロセスが要求され、多検体の解析には適さないという問題があった。
上記の問題を解決するため、キチンを特異的に認識する分子を応用してキチンを検出する試みがされている。キチンへの結合活性を示す分子としては、例えば、Calcofluor-white(CFW)などの蛍光色素およびWheat germ agglutinin(WGA)などのレクチンが挙げられ、これらを応用したキチン検出方法が複数開発され、現在も多くの研究で使用されている。しかしながら、CFWについては、植物のセルロースなどにも反応性を示すこと、WGAについては、末端にN-アセチルグルコサミンを有するペプチドグリカンならびにシアル酸とも結合することから、これらの検出方法の反応特異性は十分でなかった。
キチンへの反応特異性を向上するために、キチン分解酵素の一部を用いたキチン検出方法も開発されている。キチン分解酵素であるキチナーゼ(Chitinase)は、キチンへの結合活性を示すキチン結合ドメイン(Chitin-binding domain:CBD)と、キチン分解活性を持つ触媒ドメイン(Catalytic domain:CatD)とから構成されている。キチナーゼのこれらのドメインのうち、CBDを応用したキチン検出システムは早期に開発された。例えば、Bacillus coagulans由来chitinase A1のCBDを用いたキチン検出への応用例が提案されている(特許文献2)。また、ヒトキチナーゼのCBD(392~445位のアミノ酸から構成される)は、キチンの検出および真菌感染症の治療に用いるための分子候補である(特許文献3)。
キチン検出のために応用されるCBDは、例えば、ヒトキチナーゼの場合、最小で49アミノ酸から構成される領域がキチン結合活性を示し、分子量が比較的小さいことが利点である。その一方で、CBDは、不溶性キチンに対してはある程度結合するが、可溶性キチンオリゴ糖との結合力は、mMレベルであり、非常に弱い。また、CBDは植物セルロース、細菌のペプチドグリカンなどとの結合性はないが、ヒアルロン酸との結合活性が認められていた(非特許文献1)。
夾雑物存在下において、不溶性および可溶性のキチンを、簡便に且つ高感度で、検出および/または定量するためには、従来用いられてきたキチン認識タンパク質と比べて、キチンへの結合特異性および結合活性の改善された、新たな候補分子が求められていた。

本発明は、キチン結合ポリペプチド、ならびに、当該キチン結合ポリペプチドを用いたキチンの検出方法および検出キットを提供することを目的とする。
本発明者らは、従来はキチン結合分子として用いられていなかった、キチナーゼの触媒領域であるCatDに着目し、その機能改変を試みたところ、驚くべきことに、強力なキチン結合力を持つ分子が得られることを見出した。また、当該分子を活用し、キチンの精製純度および可溶性/不溶性といった性状に関わらず、洗浄工程等を必要とすることなく、効率よく、キチンを検出および/または定量することが可能であることをさらに見出した。さらに、本発明者らは、得られたキチン結合分子のアミノ酸配列に基づき、配列同一性の高いタンパク質を特定し、これらのキチン結合能について検討を行った。本発明は、これらの知見に基づき成されたものである。
即ち、本発明は以下のとおりである:

本発明によれば、キチン結合ポリペプチド、ならびに、当該キチン結合ポリペプチドを用いたキチンの検出方法および検出キットを提供することができる。
以下、本発明を実施するための形態について具体的に説明する。なお、本発明は、以下の実施形態に限定されるものではなく、その要旨の範囲内で種々変形して実施することができる。また、本明細書において特に断りのない限り、ヌクレオチド配列は5’から3’方向に向けて記載される。
本発明の第1の側面は、
(a)配列番号1、配列番号2、配列番号3、配列番号4、配列番号5または配列番号6に示すアミノ酸配列と配列同一性80%以上のアミノ酸配列Aからなり、
(b)キチン結合活性を有するが、キチン分解活性を示さない、ポリペプチドに関する。

キチナーゼは、キチンへの結合活性を示すキチン結合ドメイン(Chitin-binding domain:CBD)と、キチン分解活性を持つ触媒ドメイン(Catalytic domain:CatD)とを含む。配列番号1~6に示すアミノ酸配列は、キチナーゼの触媒ドメイン(以下、「CatD」とも称する。)の野生型アミノ酸配列である。
従来、糖質への結合は、糖質分解酵素内に存在する糖結合ドメインに依存するとの認識が一般的であったところ、キチンの検出にはCBDが利用されることが多かった。例えば、蛍光分子で標識した蛍光標識CBDは、実用化され、市販されているものの、CBDは、植物レクチン類似の構造を有しており、結合におけるキチン構造特異性が低いという点で、特性を改善したツールが求められていた。上述したとおり、本発明者らは、従来、糖質結合分子として認識されていたCBDではなく、キチナーゼの触媒領域であるCatDに着目し、その機能改変を試み、強力なキチン結合力を持つ分子が得られることを新たに見出した。したがって、本発明にかかるポリペプチドは、キチナーゼの触媒領域であるCatDの野生型アミノ酸配列に対して一定割合以上の配列同一性を有するアミノ酸配列(「アミノ酸配列A」に相当する)からなる。
一態様において、キチナーゼの触媒領域CatDを有するポリペプチドは、GH18ファミリーの特徴とされる触媒領域(CatD)を有するキチナーゼからなる群より選択される。当該ポリペプチドとしては、例えば、
・キトトリオシダーゼ(Chitotriosidase(Chitinase-1))、および
・酸性哺乳類キチナーゼ(Acidic mammalian chitinase(AMCase))
が挙げられる。
・キトトリオシダーゼ(Chitotriosidase(Chitinase-1))、および
・酸性哺乳類キチナーゼ(Acidic mammalian chitinase(AMCase))
が挙げられる。
CatDの野生型配列としては、例えば、ヒトキトトリオシダーゼCatDの野生型アミノ酸配列(配列番号1)、ヒト酸性哺乳類キチナーゼCatDの野生型アミノ酸配列(配列番号2)、マウスキトトリオシダーゼCatDの野生型アミノ酸配列(配列番号3)、マウス酸性哺乳類キチナーゼCatDの野生型アミノ酸配列(配列番号4)、ネコキトトリオシダーゼCatDの野生型アミノ酸配列(配列番号5)およびイヌ酸性哺乳類キチナーゼCatDの野生型アミノ酸配列(配列番号6)が挙げられる(図7A~7F)。
アミノ酸配列Aは、典型的には、配列番号1、2、3、4、5または6に示すアミノ酸配列と配列同一性が80%以上であるが、例えば、別の態様において、配列番号1、2、3、4、5または6に示すアミノ酸配列と配列同一性が60%以上、65%以上、70%以上、75%以上、80%以上、85%以上、88%以上、90%以上、93%以上、95%以上、97%以上、98%以上または99%以上であってもよい。アミノ酸配列Aには、例えば、Chitinase-3-like protein1(YKL-40)、Chitinase-3-like protein2(YKL-39)、Chitinase-like protein3(Ym1)およびChitinase-like protein4(Ym2)のアミノ酸配列が包含されていてもよい。一態様において、アミノ酸配列Aは、配列番号9(実施例において後述する)に示すアミノ酸配列である。
アミノ酸配列Aからなる本発明のポリペプチドの由来は、例えば、哺乳類であり、より具体的には、例えば、ヒト、チンパンジー、ゴリラ、マウス、ラット、イヌ、またはネコであるが、これらに限定されない。
アミノ酸配列Aは、配列番号1、2、3、4、5または6に対して、少なくとも1つのアミノ酸置換が行われた配列である。一態様では、アミノ酸配列Aにおいて、配列番号1、2、3、4、5または6に示すアミノ酸配列を基準として、119位に対応する位置のアミノ酸残基が置換されている。
本発明において、「対応する位置のアミノ酸残基」は、BLAST(https://blast.ncbi.nlm.nih.gov/Blast.cgi)配列解析ソフトウェアを使用し、基準とするアミノ酸配列(具体的には、配列番号1~6に示すアミノ酸配列。以下「基準配列」とも称する。)と、対比するアミノ酸配列(具体的には、アミノ酸配列A)とを整列させたとき、基準配列における特定位置と相対する位置のアミノ酸残基のことを指す。例えば、配列番号1に示すアミノ酸配列(ヒトChitinase-1-CatD野生型)と、ブタ由来のChitinase-1-CatDのアミノ酸配列(配列番号8のアミノ酸配列(図8A)。配列番号1に示すアミノ酸配列と80.9%の配列同一性を有する)とを、BLAST配列解析ソフトウェアを使用して整列させたときのアラインメント結果を図8Bに示す。図8Bに示すとおり、ブタ由来のChitinase-1-CatDのアミノ酸配列において、配列番号1に示すアミノ酸配列の119位(配列番号1ではグルタミン酸残基)に対応する位置のアミノ酸残基は、119位のグルタミン酸残基である。
好ましい態様において、本発明にかかるポリペプチドは、アミノ酸配列Aにおいて、配列番号1、2、3、4、5または6に示すアミノ酸配列を基準として、119位に対応する位置のアミノ酸残基が、適宜異なるアミノ酸で置換されている。
アミノ酸配列Aは、キチン分解活性に関わる保存された触媒「DXDXE」モチーフ(配列番号7)を含む。一態様において、アミノ酸配列Aは、当該触媒「DXDXE」モチーフにおけるグルタミン酸(E)が異なるアミノ酸に置換されたアミノ酸配列を含む。すなわち、アミノ酸配列Aは、配列番号7に示すアミノ酸配列(DXDXE)におけるグルタミン酸(E)が変異した配列を含む。例えば、アミノ酸配列Aは、触媒「DXDXE」モチーフにおけるグルタミン酸(E)が、アラニン(A)、アルギニン(R)、アスパラギン(N)、アスパラギン酸(D)、システイン(C)、グルタミン(Q)、グリシン(G)、ヒスチジン(H)、イソロイシン(I)、ロイシン(L)、リシン(K)、メチオニン(M)、フェニルアラニン(F)、プロリン(P)、セリン(S)、トレオニン(T)、トリプトファン(W)、チロシン(Y)またはバリン(V)に置換されたアミノ酸配列を含む。好ましくは、アミノ酸配列Aは、触媒「DXDXE」モチーフにおけるグルタミン酸(E)が、グルタミン(Q)に置換されたアミノ酸配列を含む。一態様において、配列番号7に示すアミノ酸配列(DXDXE)におけるEは、アミノ酸配列Aにおいて119位の残基である。
上記のようなアミノ酸配列を有することにより、本発明にかかるポリペプチドは、キチン分解活性を示さない一方で、キチン結合活性を有する。キチン分解活性の非存在は、天然のアミノ酸配列に対する改変に起因するものであっても、アミノ酸配列の改変によらないものであってもよい。すなわち、本発明にかかるポリペプチドは、野生型のアミノ酸配列にて本来、キチン分解活性を有さないものであってもよい。以下、本発明にかかるポリペプチドを「キチン結合ポリペプチド」または「キチン結合タンパク質」とも称する。
本発明にかかるポリペプチドは、キチン中の、4以上のβ-1,4-N-アセチルグルコサミンを含む領域に特異的に結合する。
本発明にかかるポリペプチドは、例えば、Bio-layer interferometry(BLI)法でエチレングリコールキチンに対する親和性を測定した時に、1.0×10-6M以下、1.0×10-7M以下、1.0×10-8M以下または1.0×10-9M以下の解離定数(KD)でエチレングリコールキチンに結合する。
好ましい態様において、キチンは、4以上のβ-1,4-N-アセチルグルコサミンから構成されるポリ-β-1,4-N-アセチルグルコサミンである。キチンは、より好ましくは5以上のβ-1,4-N-アセチルグルコサミン、さらにより好ましくは6以上のβ-1,4-N-アセチルグルコサミンから構成される。
本発明の別の側面は、先述のポリペプチドをコードする塩基配列が導入された組換え微生物または細胞である。ここで、細胞とは、細胞工学的手法において広く一般に使用される細胞株を示し、例えば、CHO細胞、HEK293T細胞および昆虫細胞などである。
本発明の範囲には、本発明のキチン結合ポリペプチドを利用したキチンの検出も包含される。すなわち、本発明の第2の側面は、キチンの検出方法であって、以下の工程:
(i)キチンを含む試験検体と、試薬とを接触させて、前記試験検体と前記試薬とによって形成された生成物を得る工程、および
(ii)前記工程(i)で形成された前記生成物を検出する工程
を含み、
前記試薬は、本発明のキチン結合ポリペプチドとレポーター分子とを含む検出方法に関する。
(i)キチンを含む試験検体と、試薬とを接触させて、前記試験検体と前記試薬とによって形成された生成物を得る工程、および
(ii)前記工程(i)で形成された前記生成物を検出する工程
を含み、
前記試薬は、本発明のキチン結合ポリペプチドとレポーター分子とを含む検出方法に関する。
本明細書において、「検出」の語は、「定量」の概念をも包含するものとする。具体的には、キチンの検出方法は、試験検体中に含まれるキチンの量を測定する方法であってもよい。
本明細書における「定量」では、様々な量のキチンを含む標準品と、本発明によるキチン結合ポリペプチドとレポーター分子とを含む検出方法におけるレポーター分子に依存して生じた指標の度合いから、未知検体に含まれるキチン量を標準品重量に換算することによって求めることが出来る。その他、あらかじめキチン量とレポーター分子に依存して生じた指標の度合いの相関を決定し、未知検体から誘導されるレポーター分子に依存して生じた指標の度合いから未知検体に含まれるキチン量を測定することもできる。
工程(i)では、試験検体と試薬とを接触させる。試験検体は、未精製の、精製された、または粗精製されたキチンを含むものであってよい。キチンおよびキチンを含む試験検体は、水に対して可溶性であっても、不溶性であってもよい。
試験検体に含まれるキチンの由来は特に限定されず、例えば、キノコなど担子菌類、細菌類、酵母、藻類、植物などの細胞壁、抽出物、菌体外に分泌されたキチンであってよい。また、試験検体に含まれるのは、動物の構成物もしくは抽出物としてのキチン、または動物の体外に分泌されたキチンであってよい。さらには、室内、土壌、河川、海水、大気中、宇宙空間等環境中に存在するキチンであってもよい。
前記動物には、例えば、節足動物、軟体動物、刺胞動物、棘皮動物、扁形動物および脊椎動物などが含まれる。節足動物には、例えば、六脚類、甲殻類、鋏角類および多足類が含まれる。節足動物の具体例としては、ダニ、ゴキブリ、エビ、カニ、サソリ、ムカデ、ハエ、ハチおよびチョウなどが挙げられる。節足動物以外の動物の具体例としては、貝類、クラゲ、イカ、寄生虫(サナダムシ、アニサキスなど)およびプランクトンが挙げられる。ここで、キチンの由来動物として挙げた例は、例示に過ぎず、これらに限定することを意図するものではない。
試薬は、本発明のキチン結合ポリペプチドとレポーター分子とを含む。
レポーター分子は、検出するための指標を呈する機能を有する。ここで、レポーター分子の機能とは、視覚的に観察される、電気的に検出される、またはその他の手段によって記録される、検出可能なシグナルを、直接的または間接的にもたらすことを示し、具体的に、例えば、発光、放射線、発色、凝集、磁気および電気的特性の変化が挙げられる。発光としては、蛍光およびりん光が挙げられるが、代表的には蛍光発光が検出に利用される。レポーター分子が発光を呈する場合、当該分子としては、例えば、蛍光色素、発光タンパク質(例えば、ルシフェラーゼおよび蛍光タンパク質など)および分子の発光反応を触媒するタンパク質が挙げられる。レポーター分子が放射線を呈する場合、当該分子としては、例えば、放射性同位体が挙げられる。レポーター分子が発色を呈する場合、当該分子としては、例えば、発色染料、あるいは発色によりシグナルを増幅することができる酵素類(例えば、ペルオキシダーゼ、アルカリホスファターゼ、ガラクトシダーゼなど)が挙げられる。その他、着色された高分子量コロイド物質、例えばラテックスビーズなどの微粒子物質、磁気性、常磁性のビーズ類も利用することができる。さらに、これらはバイオセンサーとの相互作用によってシグナルを増幅させる方法との組み合わせに用いることもできる。
好ましい一態様において、レポーター分子は、第1のスプリットレポーター分子と第2のスプリットレポーター分子とを含む。第1のスプリットレポーター分子と第2のスプリットレポーター分子とは、一対となることによって活性型レポーター分子を形成可能である。ここで、「一対となる」とは、第1のスプリットレポーター分子と第2のスプリットレポーター分子とが、互いに近接または結合して、対を形成することを意味する。
具体的には、第1および第2のスプリットレポーター分子は、それぞれが離れて存在する状態では、レポーター分子としての機能を示さないが、これらのスプリットレポーター分子が互いに近接または結合すると、レポーター分子としての機能を示すようになる。スプリットレポーター分子の構造は具体的には限定されず、第1および第2のスプリットレポーター分子が、互いに近接または結合した場合に、形成された活性型レポーター分子を識別可能であればよい。
一態様において、スプリットレポーター分子は、第1および第2のスプリットレポーター分子が近接または結合した場合に、構造的相補性が促進されて活性型レポーター分子が形成されるように構成されていてもよい。
好ましい一態様において、レポーター分子は、レポータータンパク質である。
スプリットレポーター分子がスプリットレポータータンパク質である場合の構成について以下に説明する。例えば、スプリットレポータータンパク質が第1のスプリットレポータータンパク質および第2のスプリットレポータータンパク質を含む場合、第1のスプリットレポータータンパク質と第2のスプリットレポータータンパク質とが近接または結合した場合に、生物発光共鳴エネルギー移動(Bioluminescence resonance energy transfer;BRET)または蛍光共鳴エネルギー移動(Fluorescence resonance energy transfer;FRET)が生じるように構成されていてもよい。この場合、活性型レポータータンパク質と、第1および第2のスプリットレポータータンパク質が近接または結合してBRETまたはFRETを生じる状態になったものをいう。レポータータンパク質がBRETまたはFRETを生じるものである場合、スプリットレポータータンパク質の一方または他方に蛍光物質を結合させる構成としてもよい。
さらに、スプリットレポータータンパク質は、第1および第2のスプリットレポータータンパク質が近接または結合した場合に、プロテインスプライシングにより活性型のレポータータンパク質が形成されるように構成されていてもよい。これは、第1および第2のスプリットレポータータンパク質のそれぞれに、プロテインスプライシングドメインを融合させておくことにより実現することができる。プロテインスプライシングドメインとしては、例えば、シネコシスティスsp.のDnaE遺伝子由来のプロテインスプライシングドメインである、DnaEnおよびDnaEcとの組み合わせなどを例示することができる。
レポータータンパク質の具体例としては、例えば、酵素および蛍光タンパク質が挙げられる。酵素としては、例えば、ルシフェラーゼ、アルカリホスファターゼ、セイヨウワサビペルオキシダーゼ、インベルターゼ、β-ガラクトシダーゼおよびβ-グルクロニダーゼ、ならびに、これらの酵素の改変体を例示することができる。蛍光タンパク質としては、例えば、緑色蛍光タンパク質(Green fluoroescent protein;GFP)およびGFPの改変体を例示することができる。
好ましい一態様において、試薬は、
(1)本発明のキチン結合ポリペプチドから選択される第1のポリペプチドと第1のスプリットレポーター分子とを含む第1の融合タンパク質、および

この場合、第1の融合タンパク質および第2の融合タンパク質において、キチン結合ポリペプチドとスプリットレポータータンパク質とを融合させる際に、立体障害を回避するために適切に長さを調節したリンカーペプチドを挿入することが好ましい。リンカーペプチドとしては、1残基~約20残基のペプチドからなり、そのアミノ酸配列としては、融合タンパク質の作製の際に使用される一般的なリンカーのアミノ酸配列と同様のものを例示することができる。具体的には、例えば、Gly-Serを含む繰り返し配列からなるGSリンカー、Asp-Asp-Ala-Lys-Lysの繰り返し配列からなるDDAKKリンカーおよびGlu-Ala-Ala-Ala-Lysの繰り返し配列からなるEAAAKリンカーが挙げられる。また、第1および第2の融合タンパク質において、スプリットレポータータンパク質を融合する領域は、N末端側またはC末端側のどちらか一方を限定するものではなく、キチン結合ポリペプチドの特性に合わせて融合する領域を決定することが好ましい。また、N末端側およびC末端側の両方に融合した融合タンパク質で
あってもよい。

本明細書において、スプリットレポーター分子および本発明のキチン結合ポリペプチドに関し、「融合した」とは、スプリットレポーター分子が、本発明のキチン結合ポリペプチドに直接的に連結する態様に加え、スプリットレポーター分子が、別の分子を介して、本発明のキチン結合ポリペプチドに関節的に連結する態様も包含するものとする。
さらに、本発明のキチン結合ポリペプチドを検出する抗体(例えばHisタグ抗体)にスプリットレポーター分子を結合させて融合タンパク質を用いることもできる。この場合、スプリットレポーター分子としては、例えばルシフェラーゼが挙げられる。
一態様において、
・第1の融合タンパク質は、第1のポリペプチドのN末端側またはC末端側に第1のスプリットレポーター分子が融合した融合タンパク質であり、

工程(i)では、キチンと、第1の融合タンパク質および第2の融合タンパク質とが結合することで、第1のスプリットレポーター分子および第2のスプリットレポーター分子により活性型レポーター分子が形成され、後続の工程(ii)では、工程(i)で形成された前記活性型レポーター分子を検出する。
工程(ii)では、工程(i)で形成された生成物を検出する。レポーター分子がレポータータンパク質である場合は、形成された活性型レポータータンパク質の活性を検出する。例えば、形成された活性型レポータータンパク質の活性がキチンの非存在下と比較して高い場合は、試験検体中にキチンが存在していることを示す。レポーター分子が蛍光物質である場合は、形成された活性型レポーター分子を励起光照射し、放出される蛍光を検出する。例えば、蛍光物質から放出される蛍光がキチンの非存在下と比較して高い場合は、試験検体中にキチンが存在していることを示す。
実施例において後述するように、対象となる構造を持つキチンの非存在下において観測される非活性化レポータータンパク質の活性(バックグラウンド)とキチンの存在下で再構成されるレポータータンパク質の活性の差は約300倍と高感度を示す。
工程(ii)における検出には、検出手段は、特に限定されず、レポーター分子の種類および構造に応じて、公知の手法または装置を適宜選択することができる。検出手法および検出装置としては、例えば、発光光度計、分光高度計、蛍光光度計、フローサイトメーター、マルチプレックス、クロマトグラフィーおよびCCDカメラ等により検出した画像をコンピューターで定量解析する方法および物理的な接触をセンサーチップで識別して定量解析する方法が挙げられる。
工程(i)および(ii)では、第1の融合タンパク質および第2の融合タンパク質を含む試薬を、試験検体を含む試験管内に直接供給して実施することもできるし、これらの融合タンパク質を発現した細胞等を用いて実施することもできる。この場合、第1の融合タンパク質および第2の融合タンパク質の発現は一過性の発現であってもよいし、恒常性の発現であってもよい。すなわち、第1および第2の融合タンパク質の安定発現細胞株を用いることができる。
具体的には、第1および第2の融合タンパク質は、例えば、大腸菌、酵母、植物、動物細胞および無細胞発現系で産生されたものを例示することができ、特に大腸菌で大量に産生されるものが好ましい。また、第1および第2の融合タンパク質を発現する細胞の破砕液など粗精製の溶液を用いても良いが、一般的な低分子量のペプチドタグを融合し、カラム精製などによって高純度に精製された融合タンパク質を用いることが好ましい。この場合、ペプチドタグとしては、GSTタグ、Protein Aタグ、抗体Fc領域、ポリヒスチジンタグ、V5タグ、Mycタグ、SBPタグ、Haloタグ、Strepタグ等を例示することができる。また、ここで使用される細胞としては、動物細胞、昆虫細胞および植物細胞等を例示することができ、樹立された細胞株が使いやすく好ましい。

本発明に従うペプチドは、キチンに対する改善した結合特異性および結合活性を有するため、キチンを効率よく、かつ、簡便に検出することができる。また、当該ペプチドは、キチンの精製純度および可溶性/不溶性といった性状に関わらず、キチンを検出することが可能である。
本発明に従うキチンの検出方法によれば、煩雑な洗浄操作等を要することなく、ハイスループットで試験検体中に含まれるキチンを検出および/または定量することができる。本発明のキチン検出方法を応用することにより、様々な生物種および精製方法に由来し、かつ、可溶性/不溶性の溶解性、ゲル状および粒子状等の様々な形態を示すキチンをスクリーニングできることが期待される。また、本発明の検出方法は、試験検体の洗浄工程等が不要であり、効率よくキチンを検出および/または定量することができる。
また、本発明に従うキチンの検出方法またはキチンの検出キットによれば、生活環境におけるアレルギー誘発リスクを可視化することが可能である。すなわち、生活環境中のキチンを検出および/または定量することにより、キチンによるアレルギーの発症リスクを予測することができる。
さらには、本発明に従うペプチドをELISA等の分析手法に応用することにより、キチンを簡便に検出するツールを提供することができる。また、本発明に従うキチンの検出方法またはキチンの検出キットによって、被験体の血液、分泌液、臓器、器官、臓器もしくは器官を洗浄して得られる洗浄液(例えば、肺胞洗浄液、口内洗浄液および腹腔内洗浄液)または尿に含まれるキチンの濃度を定量することで、真菌症、細菌症および寄生虫症などの感染症発症の可能性およびアレルギーの発症の可能性を予測すること、ならびに、感染症およびアレルギーの発症前、発症時および寛解後の体内の状態を観察することが可能になると期待される。
以下、実施例を挙げて本発明をより具体的に説明するが、本発明はこれら実施例によって何ら限定されるものではない。
<1>材料および方法
(1)材料
ウシ血清アルブミン(BSA)、4-ニトロフェニルN,N’-ジアセチル-β-D-キトビオシド[4-NP-(GlcNAc)2]およびSaccharomyces cerevisiae(S.cerevisiae)由来細胞壁多糖であるザイモサンA、マンナン(α-1,6-/α-1,2-マンナン、α-1,3-マンナン)は、Sigma-Aldrich社から購入した。キチン、エチレングリコールキチン(EGC)、セルロース粉末(β-1,4-D-グルカン)、カードラン(β-1,3-D-グルカン)、デキストラン(α-1,4-/α-1,6-グルカン)、ヒアルロナン、ヘパリンおよびダルベッコリン酸緩衝生理食塩液(PBS)は、富士フイルム和光純薬株式会社から入手した。プスツラン(β-1,6-D-グルカン)は、InvivoGen社から購入した。GlcNAc単量体とGlcNAc2量体、3量体、4量体、5量体、6量体およびエンド-β-1,3-グルカナーゼ(zymolyase 100T)は、生化学工業株式会社より購入した。N-アセチル-D-グルコサミン(>98.0%)およびトウモロコシ芯由来キシランは、東京化学工業株式会社から購入した。Hisタグ組換えヒトデクチン-1は、R&D Systemsから購入した。高脱アセチル化キトサンDAC-100[脱アセチル化度(DAC):各ロットにつき、89.9%および97.4%。コロイド滴定法を用いて決定]は、甲陽ケミカル株式会社から提供された。
(1)材料

(2)プラスミド構築
ヒトキトトリオシダーゼ-1(Chit1、Uniport:Q13231、EC:3.2.1.14)の最小キチン結合領域(CBD49)(参考文献1)としてのC末端49アミノ酸(Thr418~Asn466)をコードするDNAおよびGlu140(配列番号1に示すアミノ酸配列の119位に相当)上の点突然変異を有するCatD(Ala22~Pro388)をコードするDNA(CatD-E140Q)を、pCold I DNAベクター(Takara Bio社)にクローニングした。Oplophorus gracilirostris由来NanoLuciferase(NanoLuc)のN末端サブユニット(LgBiT、18kDa)またはNanoLucのC末端領域の11アミノ酸残基(LgBiTに低親和結合性を有するSmBiTタグ、またはLgBiTに高親和結合性を有するその改変HiBiTタグ)をコードするDNA断片を、CBD49およびCatD-E140QのC末端またはN末端に、GSリンカーペプチド(Gly-Gly-Ser-Gly-Gly-Gly-Ser-Gly-Gly-Ser-Gly配列)と共に挿入した。また、8残基ペプチド配列(Trp-Ser-His-Pro-Gln-Phe-Glu-Lys)、Ser-Ala残基から成るstrep-tag II(ST-II)(参考文献2)のDNA配列をGSリンカー配列と共にCatD-E140QのN末端に挿入したものを作製した。CBD49誘導体およびCatD-E140Q誘導体に用いられたDNA断片の大部分は、大腸菌用にコドン最適化して合成した。マウス酸性哺乳類キチナーゼ(Murine acidic mammalian chitinase;AMCase)をコードする遺伝子(Chia、Tyr22~Pro473)も、pCold Iベクターにクローニングした。Neurospora crassa由来のエンドβ-1,6-グルカナーゼ(Neg1)の発現ベクターも作成し(参考文献3)、本試験のためにHiBiT融合Neg1-E321Qを設計した。すべてのタンパク質発現プラスミドベクターを大腸菌DH5αコンピテント細胞に形質転換し、増殖させプラスミドベクターを精製した。

(3)タンパク質の調製
発現ベクターは、タンパク質発現用大腸菌に形質転換し、細胞をアンピシリン添加LB培地中、37℃で培養した。前培養後、15℃に冷却して24~48時間培養することにより、組換えタンパク質の発現を誘導した。洗浄後、プロテアーゼ阻害剤を添加した細胞懸濁液を氷上で超音波処理した。遠心分離により集めた可溶性画分を、Hisタグタンパク質精製用のコバルトカラム(タカラバイオ社)に結合させ、イミダゾール(15mM)を含むPBSで洗浄した。カラムに結合したタンパク質を、イミダゾール(150mM)を含むPBSで溶出し、透析してイミダゾールを除去した。タンパク質濃度は市販のキット(Bradford法)を用いて測定した。組換えタンパク質の発現は、市販のキットを用いてドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動(SDS-PAGE)を行った。分離したタンパク質は銀染色IIキット(富士フイルム和光純薬株式会社)またはCBB色素(クイックブルー染色液;バイオダイナミクス研究所)を用いて染色した。CatD-E140Qのアミノ酸配列を配列番号9に示す(図9)。

(4)キチン染色および蛍光顕微鏡
キチンまたはカードラン(1mg/ml、10μL)を、Hisタグ融合グルカン結合タンパク質0.5μg(CBD49、CatD-E140Q-HiBiT及びヒトdectin-1)と混合し、4℃で30分間インキュベートした。非結合タンパク質を洗浄し、さらにAlexa Fluor 647標識抗Hisタグモノクローナル抗体(MBL社)を加えて4℃でインキュベートした。洗浄後、EVOS FL Cell Imaging System(Thermo Fisher Scientific社)を用いて撮像した。
キチンまたはカードラン(1mg/ml、10μL)を、Hisタグ融合グルカン結合タンパク質0.5μg(CBD49、CatD-E140Q-HiBiT及びヒトdectin-1)と混合し、4℃で30分間インキュベートした。非結合タンパク質を洗浄し、さらにAlexa Fluor 647標識抗Hisタグモノクローナル抗体(MBL社)を加えて4℃でインキュベートした。洗浄後、EVOS FL Cell Imaging System(Thermo Fisher Scientific社)を用いて撮像した。
(5)ルシフェラーゼ断片相補性アッセイ
NanoLuc断片融合キチン結合プローブ(200nM)の混合溶液(5μL)を、96ウェル白色プレート中の試験試料(10μL)に加え、振盪条件下で30分間インキュベートした。CBD49-LgBiTとCBD49-SmBiTとの組み合わせ、または、CatD-E140Q-LgBiTとCatD-E140Q-SmBiTとの組合せを用いて、キチンの構造を解析した。振盪後、フリマジン基質溶液(Promega社)15μLをウェルに添加し、GloMaxルミノメーター(Promega社)を用い、再構成NanoLucからの発光シグナルを測定した。
NanoLuc断片融合キチン結合プローブ(200nM)の混合溶液(5μL)を、96ウェル白色プレート中の試験試料(10μL)に加え、振盪条件下で30分間インキュベートした。CBD49-LgBiTとCBD49-SmBiTとの組み合わせ、または、CatD-E140Q-LgBiTとCatD-E140Q-SmBiTとの組合せを用いて、キチンの構造を解析した。振盪後、フリマジン基質溶液(Promega社)15μLをウェルに添加し、GloMaxルミノメーター(Promega社)を用い、再構成NanoLucからの発光シグナルを測定した。
(6)キチン分解活性の測定
キチン分解活性は、合成発色基質、4-NP-(GlcNAc)2を用いて測定した。pH5.0のMcIlvain緩衝剤中の基質(200μM)は、キトトリオシダーゼ-CatD-E140Q-HiBiTまたは大腸菌で発現させたマウスキトトリオシダーゼ(mChit1)と混合し(各160ng/mL)、37℃で30分~60分間インキュベートした。放出された4-NPを、405nmにてマイクロプレートリーダー(MTP450;コロナ社)を用いて検出した。
キチン分解活性は、合成発色基質、4-NP-(GlcNAc)2を用いて測定した。pH5.0のMcIlvain緩衝剤中の基質(200μM)は、キトトリオシダーゼ-CatD-E140Q-HiBiTまたは大腸菌で発現させたマウスキトトリオシダーゼ(mChit1)と混合し(各160ng/mL)、37℃で30分~60分間インキュベートした。放出された4-NPを、405nmにてマイクロプレートリーダー(MTP450;コロナ社)を用いて検出した。
(7)キチンビーズ結合アッセイ
磁気キチンビーズ(New England Biolabs社)を洗浄し、BSAとTween20を含むPBS(BPBST)に懸濁した。磁気キチンビーズとHiBiT-tag融合CatD-E140QまたはNeg1-E321Q(0~200nM、25μL)を96ウェル白色プレート(Greiner Bio-one社)内で混合し、5分間インキュベートした。プレートは、磁気プレートキャリア付き洗浄器(TECAN社)を用いて洗浄した。各タンパク質の結合能を評価するために、キチンビーズ結合タンパク質からのHiBiTタグ由来シグナルを、25μLのLgBiTと基質(Nano-Glo HiBiT LyticDetection System; Promega社)混合液を添加することによって検出し、マイクロプレートリーダー(GloMax)を用いて再構成されたルシフェラーゼ活性を測定した。
磁気キチンビーズ(New England Biolabs社)を洗浄し、BSAとTween20を含むPBS(BPBST)に懸濁した。磁気キチンビーズとHiBiT-tag融合CatD-E140QまたはNeg1-E321Q(0~200nM、25μL)を96ウェル白色プレート(Greiner Bio-one社)内で混合し、5分間インキュベートした。プレートは、磁気プレートキャリア付き洗浄器(TECAN社)を用いて洗浄した。各タンパク質の結合能を評価するために、キチンビーズ結合タンパク質からのHiBiTタグ由来シグナルを、25μLのLgBiTと基質(Nano-Glo HiBiT LyticDetection System; Promega社)混合液を添加することによって検出し、マイクロプレートリーダー(GloMax)を用いて再構成されたルシフェラーゼ活性を測定した。
(8)真菌試料および節足動物試料の調製
Candida albicans NBRC 1385(NITE生物資源センター、日本)をYPD寒天平板上に播種し、酵母コロニーを107細胞/mLの濃度でPBS中に懸濁した。懸濁液の半分を90℃で10分間加熱し、熱殺菌したカンジダ・アルビカンス(C.albicans)(HKCA)として用いた。10μLの生細胞(LiveCA)とHKCA懸濁液を、キトトリオシダーゼ-CatD-E140Qを用いたルシフェラーゼ断片相補性アッセイに利用した。節足動物試験試料について、コナヒョウヒダニ(Dermatophagoides farinae)およびヤケヒョウヒダニ(D.pteronyssinus)の凍結体、または、チャバネゴキブリ(Blattella germanica)(Biostir社)の凍結乾燥粉末をPBSに懸濁し(4mg/mL)、金属ビーズを加えてビーズ破砕機(FastPrep-24;MP Biomedicals社)で20秒間処理した。コナヒョウヒダニ(D.farinae)、ヤケヒョウヒダニ(D.pteronyssinus)およびチャバネゴキブリ(B.germanica)破砕溶液を10分間煮沸し、キトトリオシダーゼ-CatD-E140Qを用いたルシフェラーゼ断片相補性アッセイに利用した。

(9)キトトリオシダーゼ-CatD-E140Qを用いたELISA様試験
EGC固相化マイクロプレート(50ng/ml)をBPBSTでブロッキングし、GlcNAcオリゴマー(モノマーからヘキサマー、200μm)またはEGC(200μg/ml)の存在または非存在下、CatD-E140Q-HiBit(50ng/ml)にてインキュベートした。EGC結合CatD-E140QからのHiBitタグ由来のシグナルを検出するため、遊離LgBiTおよび基質(Nano-Glo HiT Lytic Detection System)を洗浄プレートに添加し(20~25μL/ウェル)、マイクロプレートリーダ(GloMax)を用いてルシフェラーゼ活性を測定した。可溶性キチンを高感度に検出・定量する目的でサンドイッチELISA様試験を構築した。マイクロプレートをCatD-E140Q-ST-II(2μg/ml)でコーティングし、BPBSTによりブロッキングした。洗浄後、3倍連続希釈したEGCをプレートに添加し、1時間インキュベートした。プレートを洗浄し、CatD-E140Q-HiBiT(1μg/ml)を添加して1時間インキュベートした。十分な洗浄後、上記と同様にHiBiTタグ由来のシグナルを測定した。
EGC固相化マイクロプレート(50ng/ml)をBPBSTでブロッキングし、GlcNAcオリゴマー(モノマーからヘキサマー、200μm)またはEGC(200μg/ml)の存在または非存在下、CatD-E140Q-HiBit(50ng/ml)にてインキュベートした。EGC結合CatD-E140QからのHiBitタグ由来のシグナルを検出するため、遊離LgBiTおよび基質(Nano-Glo HiT Lytic Detection System)を洗浄プレートに添加し(20~25μL/ウェル)、マイクロプレートリーダ(GloMax)を用いてルシフェラーゼ活性を測定した。可溶性キチンを高感度に検出・定量する目的でサンドイッチELISA様試験を構築した。マイクロプレートをCatD-E140Q-ST-II(2μg/ml)でコーティングし、BPBSTによりブロッキングした。洗浄後、3倍連続希釈したEGCをプレートに添加し、1時間インキュベートした。プレートを洗浄し、CatD-E140Q-HiBiT(1μg/ml)を添加して1時間インキュベートした。十分な洗浄後、上記と同様にHiBiTタグ由来のシグナルを測定した。
(10)Bio-layer Interferometry(BLI)による結合親和性測定
固定化EGCに対するキトトリオシダーゼ-CatD-E140Qの結合親和性を、BLIバイオセンサー(BLItzシステム;Pall ForteBio社)を用いて計算した。ビオチン化EGC(50μg/mL)をセンサーチップと120秒間反応させ、結合させた。2倍連続希釈したCatD-E140Q-HiBiT(45kDa)とEGC結合センサーチップを反応させて、結合速度定数(ka)、解離速度定数(kd)および平衡解離定数(KD)を算出した。データはBLItz Proソフトウェア(Pall ForteBio社)を用いて解析した。
固定化EGCに対するキトトリオシダーゼ-CatD-E140Qの結合親和性を、BLIバイオセンサー(BLItzシステム;Pall ForteBio社)を用いて計算した。ビオチン化EGC(50μg/mL)をセンサーチップと120秒間反応させ、結合させた。2倍連続希釈したCatD-E140Q-HiBiT(45kDa)とEGC結合センサーチップを反応させて、結合速度定数(ka)、解離速度定数(kd)および平衡解離定数(KD)を算出した。データはBLItz Proソフトウェア(Pall ForteBio社)を用いて解析した。
(11)各種哺乳類キチナーゼ-CatD-改変体を用いたELISA様試験
EGC固相化マイクロプレート(0ng/mL,12.5ng/mL,50ng/mL,200ng/mL(マウスAMCaseの改変体の場合)または0μg/mL,0.25μg/mL,1,4μg/mL(その他の改変体の場合))をBPBSTでブロッキングし、各種哺乳類キチナーゼCatD-改変体-HiBit(2μg/ml)にてインキュベートした。ここで用いた各種哺乳類キチナーゼCatD-改変体-HiBitは、図7に示す各配列において、119番目のGlu(E)をGln(Q)に置換した改変体に、HiBitタグを付加して作製したものである。EGC結合CatD-改変体からのHiBitタグ由来のシグナルを検出するため、遊離LgBiTおよび基質(Nano-Glo HiT Lytic Detection System)を洗浄プレートに添加し(20~25μL/ウェル)、マイクロプレートリーダ(GloMax)を用いてルシフェラーゼ活性を測定した。

<2>結果
(1)キチン結合タンパク質融合ルシフェラーゼ断片相補性アッセイによるキチンの検出(図1)
簡便かつ迅速なキチン検出システムを実証するべく、まずヒトChit1のCBDを用いた。Chit1の最後の49個のC末端残基(CBD49)は、キチン結合タンパク質として証明されている(参考文献1および4)。6×Hisタグ組換えCBD49およびNanoLuc断片融合体は大腸菌で発現させることに成功し、精製したタンパク質のそれぞれについて予想される分子量の単一バンドが検出された(図1AおよびB)。リコンビナントCBD49-Hisは、不溶性のカニ殻キチンに結合活性を示し(図1C)、ルシフェラーゼ断片融合CBD49(CBD49-LgBiTおよびCBD49-SmBiT)は、キチンとともに存在する場合のみ、用量依存的に再活性化されたが、β-グルカン粒子(カードラン)の存在下では再活性化されなかった(図1D)。これらの結果から、キチンの簡便な検出法のコンセプトが実証された。しかし、CBD49を用いたアッセイでは十分なキチン検出感度を提供できなかった。
(1)キチン結合タンパク質融合ルシフェラーゼ断片相補性アッセイによるキチンの検出(図1)
簡便かつ迅速なキチン検出システムを実証するべく、まずヒトChit1のCBDを用いた。Chit1の最後の49個のC末端残基(CBD49)は、キチン結合タンパク質として証明されている(参考文献1および4)。6×Hisタグ組換えCBD49およびNanoLuc断片融合体は大腸菌で発現させることに成功し、精製したタンパク質のそれぞれについて予想される分子量の単一バンドが検出された(図1AおよびB)。リコンビナントCBD49-Hisは、不溶性のカニ殻キチンに結合活性を示し(図1C)、ルシフェラーゼ断片融合CBD49(CBD49-LgBiTおよびCBD49-SmBiT)は、キチンとともに存在する場合のみ、用量依存的に再活性化されたが、β-グルカン粒子(カードラン)の存在下では再活性化されなかった(図1D)。これらの結果から、キチンの簡便な検出法のコンセプトが実証された。しかし、CBD49を用いたアッセイでは十分なキチン検出感度を提供できなかった。
(2)ヒトキトトリオシダーゼCatD改変体の潜在的なキチン結合活性(図2)
ルシフェラーゼ断片相補性アッセイのキチン検出感度を改善するため、GH18キチナーゼ(参考文献5)に保存されたDXDXEモチーフ上のGlu140をGlnに置換し、不活性型キトトリオシダーゼCatD(CatD-E140Q)をコードするプラスミドを作製し、HiBiT融合タンパク質として大腸菌で発現させた(図2Aおよび2B)。CBD領域を除去したCatD(野生型)は、完全長Chit1と同様に、キチン分解活性を示すことが知られているが(参考文献6)、CatD-E140Q変異体は、合成発色性基質に対する糖分解活性を示さなかった(図2C)。
ルシフェラーゼ断片相補性アッセイのキチン検出感度を改善するため、GH18キチナーゼ(参考文献5)に保存されたDXDXEモチーフ上のGlu140をGlnに置換し、不活性型キトトリオシダーゼCatD(CatD-E140Q)をコードするプラスミドを作製し、HiBiT融合タンパク質として大腸菌で発現させた(図2Aおよび2B)。CBD領域を除去したCatD(野生型)は、完全長Chit1と同様に、キチン分解活性を示すことが知られているが(参考文献6)、CatD-E140Q変異体は、合成発色性基質に対する糖分解活性を示さなかった(図2C)。
次いで、CatD-E140Qのキチンに対する結合能が保存されているかどうかを評価した。HiBiT融合CatD-E140QおよびNeg1-E321Q(ネガティブコントロール)を、キチン磁気ビーズと混合したが、CatD-E140Qのみがキチンビーズに対して有意な結合活性を示した(図2D)。
さらに、CatD-E140Qの潜在的なキチン結合能およびその構造特異性も、蛍光顕微鏡観察から次の結果によって証明された:(i)キチン粒子はCatD-E140Qと直接結合するが、ネガティブコントロールとして使用したデクチン-1(主要なβ-1,3-グルカン受容体)とは結合しない、(ii)デクチン-1は、β-1,3-グルカン粒子(カードラン)に強く結合するが、CatD-E140Qはカードランと結合しない(図2E)。以上より、適切に改変されたCatDは、グリコヒドロラーゼ活性を有さない一方でキチン結合活性を保持しており、キチン認識プローブの新たな候補になることが示された。
(3)キチン構造を検出・定量するためのスプリットNanoLuc融合CatD-E140Q(図3)
高感度な無洗浄キチン検出・定量法を確立すべく、ルシフェラーゼ断片相補性アッセイにCatD-E140Qを使用した。C端末側と融合したCatD-E140Q-SmBiTは、HiBiT融合の場合のように、大腸菌で発現させることに成功した(図3Aおよび3B)。一方、LgBiTをCatDのC端末に融合した場合、大腸菌による発現効率は低かった。そこで、LgBiTのCatDのN末端への融合を設計したところ、CatD-E140Q-LgBiTを得ることが出来た(図3Aおよび図3B)。
高感度な無洗浄キチン検出・定量法を確立すべく、ルシフェラーゼ断片相補性アッセイにCatD-E140Qを使用した。C端末側と融合したCatD-E140Q-SmBiTは、HiBiT融合の場合のように、大腸菌で発現させることに成功した(図3Aおよび3B)。一方、LgBiTをCatDのC端末に融合した場合、大腸菌による発現効率は低かった。そこで、LgBiTのCatDのN末端への融合を設計したところ、CatD-E140Q-LgBiTを得ることが出来た(図3Aおよび図3B)。
CatD誘導体を用いたルシフェラーゼ断片相補性アッセイでは、キチンビーズ(リガンド)またはいずれかのタンパク質が存在しない場合には、バックグラウンドが低く、両方のタンパク質、リガンドおよび基質のいずれもが存在する場合にのみ、強いルシフェラーゼシグナルが観察された(図3C)。
次いで、市販のキチン粒子試薬に対する反応性を、CBD49-ルシフェラーゼ断片相補性アッセイとCatD-E140Q-ルシフェラーゼ断片相補性アッセイとで比較した。いずれの方法においても、リガンドであるキチンの存在下にて、バックグラウンド(ブランク)と比較して、ルシフェラーゼシグナルの有意な増加が観察されたが、シグナル対ノイズ比は、CBD49-ルシフェラーゼ断片相補性アッセイでは約7.7、CatD-E140Q-ルシフェラーゼ断片相補性アッセイでは約340であった(図3D)。これらの結果は、改変型CatDを使用することにより、感度が大幅に改善したことを示している。
(4)CatD-E140Q-ルシフェラーゼ断片相補性アッセイのリガンド認識における構造特異性(図4)
CatD-E140Q-ルシフェラーゼ断片相補性アッセイのリガンド認識における構造特異性について、市販の試薬を用いて検証した。CatD-E140Qは、不溶性(カニ殻キチン)および可溶性(EGC)の両形態のキチン構造、または、酵母粗精製粒子(ザイモサンA)のキチン構造に反応したが、GlcNAcモノマー、セルロース、カードラン、プスツラン、マンナン、キシラン、デキストラン、ヒアルロナンおよびヘパリンのようなキチン構造を含まないとされる他の試薬には反応しなかった(図4)。この結果より、改変前のCatDの持つ厳密な基質特異性が、CatD-E140Qに継承されていることを証明した。

(5)CatD-E140Q-ルシフェラーゼ断片相補性アッセイを用いた天然物中のキチン構造の検出(図5)
次に、生きたカンジダ・アルビカンス(C.albicans)(LiveCA)または加熱殺菌したC.albicans(HKCA)の細胞表面上のキチンを、直接検出できるかどうか試験した。CatD-E140Q-ルシフェラーゼ断片相補性アッセイは、LiveCAおよびHKCAの両方に対して、細胞濃度依存的にルシフェラーゼ活性を有意に増加させた(図5A)。
次に、生きたカンジダ・アルビカンス(C.albicans)(LiveCA)または加熱殺菌したC.albicans(HKCA)の細胞表面上のキチンを、直接検出できるかどうか試験した。CatD-E140Q-ルシフェラーゼ断片相補性アッセイは、LiveCAおよびHKCAの両方に対して、細胞濃度依存的にルシフェラーゼ活性を有意に増加させた(図5A)。
続いて、CatD-E140Q-ルシフェラーゼ断片相補性アッセイが細胞壁表面上の各多糖構造の位置関係を評価するツールとして利用できるか評価した。HKCA細胞壁上の多糖を糖質分解酵素で分解し、細胞壁に生じる構造変化をCatD-E140Q-ルシフェラーゼ断片相補性アッセイを用いて、リアルタイムに解析した。細胞壁キチン上で再構築されたルシフェラーゼ活性を検出する際、基質にエンド-β-1,4-N-アセチルグルコサミニダーゼ(組換えmurineAMCase)を加えた場合、発光シグナルは、対照(PBS)に比べて時間の経過とともに減少し、表面キチン構造の損失を示した(図5B)。一方、β-1,3-またはβ-1,6-グルカナーゼの存在下では、発光シグナルは時間依存的に増加した。特に、発光シグナルはβ-1,3-グルカナーゼ処理により大きく増加し、キチンが、β-1,3-グルカンによって高度に覆われていることを示した。
また、主要な2種類のダニ(D.farinaeおよびD.pteronyssinus)の虫体ならびにチャバネゴキブリ(B.germanica)粉体に含まれるキチンを直接検出することが可能か検証した。その結果、検体の濃度依存的に発光強度が増加したことから、これらの天然検体中の微量キチンにおいても、CatD-E140Q-ルシフェラーゼ断片相補性アッセイ由来の発光シグナルが検出できることを確認した(図5C)。
(6)CatD-E140Qのキチンへの分子構造依存性結合およびサイズ依存性結合(図6)
CatD-E140Qの結合性をさらに理解するために、(i)類似のキトサン構造との交差反応性の有無、(ii)CatD-E140Qとの結合に必要なGlcNAcユニット数、および、(iii)結合親和性の強さについて調べた。天然物は、キチン構造とキトサン構造との両方を含むことがあるため、CatD-E140Qが、キチンとキトサンとを明確に区別することができるかどうかを評価した。この試験のために、高度に脱アセチル化したキトサン(DAC 89.9%および97.4%、コロイド滴定法)を入手し、CatD-E140Q-ルシフェラーゼ断片相補性アッセイのキチンまたはキトサンに対する反応性を比較した。
CatD-E140Qの結合性をさらに理解するために、(i)類似のキトサン構造との交差反応性の有無、(ii)CatD-E140Qとの結合に必要なGlcNAcユニット数、および、(iii)結合親和性の強さについて調べた。天然物は、キチン構造とキトサン構造との両方を含むことがあるため、CatD-E140Qが、キチンとキトサンとを明確に区別することができるかどうかを評価した。この試験のために、高度に脱アセチル化したキトサン(DAC 89.9%および97.4%、コロイド滴定法)を入手し、CatD-E140Q-ルシフェラーゼ断片相補性アッセイのキチンまたはキトサンに対する反応性を比較した。
図6Aに示すように、キトサン(DAC 89.9%)に対するCatD-E140Qの反応性は、キチンへの反応性と比べ、大幅に低下し、キトサン(DAC 97.4%)ではほとんど消滅した。次に、CatD-E140Qの結合に必要な最小GlcNAc単位を明らかにするため、EGC固相化マイクロプレートを用いた競合ELISAを実施した。固相EGCとCatD-E140Q-HiBiTとの相互作用は、4量体以上のGlcNAcユニットおよび可溶性EGCによって有意に抑制された(図6B)。
続いて、BLIを用いてバイオセンサー上に結合したEGCに対するCatD-E140Q-HiBiTの結合力(KD値)を算出した。計算により得られた親和性(KD:9.9×10-8 M)は、一般的な抗体と同等レベルの値を示した(図6C)。CatD-E140Qが予想よりも強いKD値を示したため、最後に、CatD-E140Qを用いたサンドイッチELISAを構築することで、検出・定量可能な微量キチンの濃度を求めた。
CatD-E140Q-ST-IIコート済みマイクロプレートおよびHiBiT融合CatD-E140Qを用いてサンドイッチ-ELISAを確立し、2.7~2,000pg/mlのEGC濃度に対する反応曲線を得た(図6D)。この反応曲線から算出された定量限界は13.6pg/mLであった。これらの結果から、CatD-E140Qは、キトサン、GlcNAc1量体~3量体に結合しないこと、ならびに、ピコグラムレベルの可溶性キチンが、CatD-E140Qの高い結合親和性により、サンドイッチELISAによって検出できることが実証された。
(7)各種哺乳類キチナーゼ-CatD-改変体のキチン結合活性
上記(11)のELISA様試験から、各種哺乳類キチナーゼ-CatD-改変体のキチン結合活性が示された(図10A~10D)。

- (a)配列番号1、配列番号2、配列番号3、配列番号4、配列番号5または配列番号6に示すアミノ酸配列と配列同一性80%以上のアミノ酸配列Aからなり、
(b)キチン結合活性を有するが、キチン分解活性を示さない、ポリペプチド。 - 前記アミノ酸配列Aにおいて、配列番号1、配列番号2、配列番号3、配列番号4、配列番号5または配列番号6に示すアミノ酸配列を基準として、119位に対応する位置のアミノ酸が置換されている、請求項1に記載のポリペプチド。
