La présente invention concerne un principe actif comprenant des oligo-glucanes, obtenu à partir de la fraction cytosolique d'une levure de l'espèce Saccharomyces cerevisiae, une composition cosmétique le comprenant et ses utilisations cosmétiques, en particulier pour améliorer la régénération de la peau et lutter contre son vieillissement.

[0002] L'invention concerne un principe actif comprenant des oligo-glucanes, obtenu à partir de la fraction cytosolique d'une levure de l'espèce Saccharomyces cerevisiae, une composition cosmétique le comprenant et ses utilisations cosmétiques, en particulier pour améliorer la régénération de la peau et lutter contre le vieillissement cutané.

[0004] Le processus de régénération, par exemple cutané, est un processus dynamique et complexe mettant en œuvre de nombreux acteurs cellulaires, au cours duquel un nouveau tissu se développe pour restaurer des zones préalablement endommagées et ainsi assurer sa cicatrisation.
[0005] Au niveau du tissu cutané, trois systèmes sont impliqués dans le processus de régénération tissulaire, le système cutané, vasculaire et immunitaire.
[0006] Le tissu cutané est aujourd'hui reconnu comme un organe complexe comprenant notamment des fibroblastes qui jouent le rôle pivot entre ces trois systèmes pour agir sur le renouvellement épidermique, le remodelage matriciel, mais également sur la vascularisation, permettant ainsi la régénération du tissu endommagé.
[0007] Au niveau du système cutané, le renouvellement de l'épiderme est sous la dépendance des kératinocytes qui arrêtent d'adhérer à la membrane basale pour migrer, proliférer et se différencier en cornéocytes fonctionnels. Dans le derme, le remodelage matriciel est réalisé par les fibroblastes, après qu'ils aient migré et se soient différenciés en myofibroblastes.
[0008] Le système vasculaire permet entre autres le transport des nutriments, des médiateurs et des cellules, indispensables à la régénération cutanée. Pour ce faire, les cellules endothéliales migrent pour se regrouper de manière cohésive, afin de former un réseau vasculaire organisé et régulier.
[0009] Enfin, au niveau du système immunitaire, il existe une population de macrophages dite pro-régénérante capable d'interagir avec les fibroblastes permettant de stimuler le remodelage matriciel et de favoriser la vascularisation.
[0010] Au cours du processus de régénération, les fibroblastes sécrètent des facteurs de croissance qui vont agir sur ces systèmes en activant les voies de prolifération, de migration et de différenciation cellulaire. Ces facteurs agissent de manière paracrine sur le métabolisme des fibroblastes environnants mais également sur le métabolisme des autres types cellulaires présents dans le tissu cutané.
[0011] Cependant, au cours du vieillissement, le métabolisme cellulaire ralentit. La capacité des fibroblastes à sécréter des facteurs de croissance pour interagir avec leur environnement diminue. Le processus endogène de régénération ralentit alors et les dommages successifs que notre peau subit ne sont plus correctement réparés. La qualité du renouvellement épidermique diminue, la matrice dermique s'affaisse et le réseau vasculaire se désorganise.
[0012] Les premiers signes du vieillissement cutané apparaissent alors. Le teint devient terne, perd en uniformité, la peau est plus sujette aux rougeurs et les rides s'accentuent. Or, l'éclat du teint mais encore les rides traduisent la santé de la peau. Un teint terne donne un air fatigué et peut accentuer les signes de l'âge, ce qui peut avoir un impact important sur l'estime de soi et l'image que l'on renvoie.
[0013] De nombreux produits cosmétiques visent à lutter contre les signes du vieillissement cutané, par exemple pour lutter contre les rides, améliorer l'hydratation de la peau, ou améliorer l'éclat du teint. Or, ces ingrédients cosmétiques agissent sur l'un des systèmes et ne visent pas une action globale sur l'ensemble du processus de régénération.
[0014] L'apport exogène de facteurs de croissance dans le cadre de la régénération tissulaire a certes démontré son efficacité. Cependant, cette solution repose sur des techniques de médecine régénérative qui sont jugées très invasives. Elle ne peut donc pas s'inscrire dans une routine de beauté.
[0015] Une autre technique de rajeunissement appelée PRP (platelet-rich plasma), consiste en une injection intradermique d'un concentré autologue de facteurs de croissance présents dans les PRP, préalablement obtenu à partir d'un prélèvement sanguin du patient. Là encore, cette technique est controversée de par son caractère invasif et son efficacité qui est limitée par la concentration initiale du patient en PRP.
[0016] Faciliter les processus de régénération de la peau tout en minimisant l'impact pour le patient reste un enjeu majeur pour les industries du secteur dermatologique et cosmétique.
[0017] L'inventeur a ainsi étudié le métabolisme des fibroblastes âgés, mais également le métabolisme des kératinocytes, des cellules endothéliales et des macrophages en présence
du sécrétome de fibroblastes âgés et a observé :
- un ralentissement de la migration des fibroblastes et de leur différenciation en myofibroblastes, conduisant à une réduction de la synthèse du réseau de collagène I ;
- une diminution de la migration kératinocytaire, étape clé du renouvellement de l'épiderme ;
- une diminution de la population immunitaire pro-régénérante, impliquée dans le remodelage matriciel ;
- une réduction du métabolisme des cellules endothéliales, entraînant un défaut de formation du réseau vasculaire.
[0018] Le métabolisme du système cutané, immunitaire et vasculaire est donc réduit ne permettant pas une régénération optimale.
[0019] A cet effet, il a été identifié par l'inventeur un actif spécifique obtenu à partir d'une fraction cytosolique de la levure de Saccharomyces cerevisiae permettant d'agir sur l'ensemble du processus de la régénération cutanée.
[0020] Certes, on connaît déjà des principes actifs cosmétiques obtenus à partir de Saccharomyces cerevisiae. On peut notamment citer une solution anti-âge contenant un lysat de culture de S. cerevisiae permettant de stimuler la biosynthèse des cellules de la peau (US5019391).
[0021] D'autres solutions cosmétiques anti-âge et hydratante reposent sur des hydrolysats de levure, notamment des hydrolysats de levure de S. cerevisiae. On peut ainsi citer une composition cosmétique comprenant des mannoprotéines issues de la paroi des cellules des levures telles que Saccharomyces décrite dans EP2567688. Ledit extrait est obtenu à partir d'une hydrolyse chimique, enzymatique ou physicochimique de la paroi des levures.
[0022] La demande WO 2009/101503 divulgue également l'utilisation d'hydrolysat de levure de S. cerevisiae constitué de protéines issues de la paroi des levures hydrolysées dont le poids moléculaire est compris entre 1 et 5 kDa.
[0023] Ainsi, les solutions déjà connues visent particulièrement un hydrolysat de S. cerevisiae et plus particulièrement un hydrolysat obtenu à partir de la paroi de la levure de S. cerevisiae. Cet hydrolysat est riche en protéines, connues pour lutter contre le vieillissement cutané.
[0024] Il existe donc un besoin pour une solution alternative cosmétique non-invasive, naturelle et permettant également d'agir sur le système cutané, vasculaire et immunitaire
simultanément afin de stimuler les capacités endogènes de la peau pour améliorer ses capacités de régénération.
[0025] Résumé de l'invention
[0026] C'est l'objectif de la présente invention qui, à cet effet, propose un nouveau principe actif obtenu à partir de la fraction cytosolique de la levure de Saccharomyces cerevisiae.
[0027] Une levure est un micro-organisme eucaryote, composée d'une paroi cellulaire entourant la membrane plasmique. Cette paroi est constituée d'une couche externe de mannoprotéines associées à des glucanes et une couche interne de glucanes associés à des chitines. La membrane cytoplasmique est quant à elle composée principalement de phospholipides et de complexes protéiques. Le contenu de la cellule dont le noyau et les organites cytoplasmiques, est compris dans une phase liquide appelée cytosol.
[0028] La levure de Saccharomyces cerevisiae est un eucaryote unicellulaire bien connue de l'homme de l'art, qui se présente sous forme de cellules isolées, ovoïdes à arrondies. Elle est utilisée depuis l'antiquité pour la fabrication de boissons fermentées. Toutefois, elle a été isolée et identifiée qu'au milieu du XIXe siècle pour remplacer le levain traditionnel des boulangers à travers son utilisation dans un procédé de fermentation plus fiable et plus rapide que le levain.

Préférentiellement, l'effet cosmétique sur la peau est un effet anti-âge, en particulier un effet régénérant de la peau.

[0262] Le remodelage du réseau vasculaire cutané chez l'adulte en réponse à un stimulus (hypoxie, blessure) se fait via un mécanisme d'angiogenèse. Ce processus se définit comme la formation de vaisseaux à partir de vaisseaux préexistants. Il est le résultat de spécification, de migration et de prolifération des cellules endothéliales. L'angiogenèse physiologique permet à la peau de cicatriser correctement. Avec l'âge, la formation de capillaires par angiogenèse est altérée et empêche la bonne cicatrisation et homéostasie des tissus.

[0278] Le panel caucasien est composé de 19 volontaires sains, de sexe féminin, âgés de 70 à 78 ans, présentant des rides de la patte d'oie et un teint terne.
[0279] Le panel asiatique était constitué de 33 personnes âgées de 60 à 69 ans et le groupe placebo de 32 personnes âgées de 60 à 68 ans.
[0280] Effet du principe actif selon l'invention sur le renouvellement cellulaire.
[0281] Dans un premier temps, l'effet du principe actif selon l'invention a été évalué au niveau des avant-bras sur le panel caucasien en étudiant sa faculté à accélérer le renouvellement cellulaire après 14 jours d'application biquotidienne.
[0282] Le renouvellement cellulaire est mesuré grâce à la capacité de la peau à éliminer la DHA (dihydroxyacetone) qui est un marqueur colorant les protéines. Le renouvellement des cellules entraîne la disparition de la coloration induite par la DHA.
[0283] Un gel contenant 8% de DHA est appliqué à la surface de la peau des volontaires. Le suivi de la couleur obtenue après application du gel de DHA démarre 72 heures après coloration sur des zones non traitées, traitées biquotidiennement avec une formule placebo ou traitées biquotidiennement avec une formule contenant 5% du principe actif selon
l'invention. L'évaluation est réalisée à partir de photographies de la peau prise à intervalles réguliers pendant 14 jours, à l'aide d'un dermatoscope.

Cette étude a été réalisée sur les panels caucasien et asiatique.

[0298] L'objectif de cette étude est d'évaluer in vivo les bénéfices cosmétiques du principe actif selon l'invention formulé à 2,5% en émulsion, en comparaison à une formule placebo, chez des volontaires caucasiens et asiatiques. L'effet du principe actif selon l'invention a été évalué après 21 et 42 jours d'application biquotidienne.

sur la zone de la patte d'oie de volontaires caucasiens par scorage visuel sur photographies numériques en comparaison au placebo sont présentés dans le tableau 16.
[0309] Dès 21 jours de traitement, en comparaison au placebo, le principe actif selon l'invention formulé à 2,5% en émulsion présente un effet anti-rides, en diminuant significativement de 10% le stade de rides de la patte d'oie. Cet effet est observé chez 74% des volontaires.

Le paramètre b* représente la gamme des bleus aux jaunes lors des mesures colorimétriques, il est caractéristique de la pigmentation jaune mélanique cutanée. Plus ce paramètre diminue plus la peau s'éclaircit.
[0321] Cet effet est visible chez 68% des volontaires et 79% d'entre eux ont perçu le produit comme un anti-cernes contre seulement 53% pour le traitement placebo.

[0325] Dès 21 jours d'application biquotidienne et en comparaison au placebo, le principe actif selon l'invention formulé à 2,5% en émulsion : o donne un teint plus lumineux et plus frais en augmentant significativement le rayonnement de la peau de 9% et la couleur rose, de 15%. Effet observé chez respectivement 74% et 84% des volontaires. o diminue significativement la couleur olive de 7% et l'état de fatigue des yeux de 6%. Effet observé chez respectivement 79% et 89% des volontaires
[0326] Cet effet est conforté par l'évaluation des volontaires : 84% à J21 ont trouvé leur peau plus éclatante avec le principe actif selon l'invention contre 63% pour le traitement placebo.

[0328] Les résultats correspondant à l'effet anti-rides du principe actif selon l'invention étudié par scorage visuel par experts sur des volontaires asiatiques est présenté dans le tableau 20. Un jury composé de 3 personnes a évalué le stade de rides de la patte d'oie de chaque photo sur une échelle de score préalablement définie.
[0330] Dès 21 jours de traitement, en comparaison au placebo, le principe actif selon l'invention formulé à 2,5% en émulsion présente un effet anti-rides chez des volontaires asiatiques, en diminuant significativement de 6% le stade de rides de la patte d'oie. Cet effet est observé chez 58% des volontaires. Cet effet se prolonge après 42 jours de traitement.

[0332] Le principe actif selon l'invention formulé à 2,5% en émulsion, apporte des bénéfices cosmétiques dès 21 jours de traitement qui se poursuivent après 42 jours de traitement.
[0333] En effet, le principe actif selon l'invention permet : une amélioration des propriétés biomécaniques de la peau une réduction des rides chez des volontaires caucasiens et asiatiques une amélioration de la vascularisation cutanée, en réduisant les microvaisseaux dystrophiques ainsi que les cernes une amélioration de l'éclat du teint.
Claims
[Revendication 1] Principe actif comprenant au moins un extrait de Saccharomyces cerevisiae, caractérisé en ce que ledit extrait correspond à la fraction cytosolique comprenant des oligo-glucanes de masse molaire compris entre 2 et 5 kDa.
[Revendication 2] Principe actif cosmétique selon la revendication précédente, caractérisé en ce que les oligo-glucanes représentent entre 10% et 35% en poids de matières sèches de l'extrait.
[Revendication 3] Principe actif cosmétique selon la revendication précédente, caractérisé en ce que l'extrait comprend un ratio glucose/mannose supérieur à 60%.
[Revendication 4] Principe actif cosmétique selon l'une des revendications précédentes, caractérisé en ce que l'extrait est un lysat mécanique de Saccharomyces cerevisiae.
[Revendication 5] Principe actif cosmétique selon l'une des précédentes revendications, caractérisé en ce que ledit extrait est susceptible d'être obtenu par un procédé comprenant les étapes suivantes : a. solubilisation d'au moins 50 g/L de Saccharomyces cerevisiae dans l'eau par homogénéisation mécanique, b. séparation des phases soluble et insoluble c. ultrafiltration et récupération du filtrat soluble d. tri moléculaire.
[Revendication 6] Composition cosmétique pour une application topique comprenant au moins un principe actif selon l'une des revendications précédentes et un milieu physiologiquement acceptable.
[Revendication 7] Composition cosmétique selon la revendication précédente, dans laquelle le principe actif sous forme liquide représente au moins 0,1% en poids du poids total de la composition.
[Revendication 8] Utilisation cosmétique d'un principe actif selon l'une des revendications 1 à 5 ou d'une composition cosmétique selon l'une des revendications 6 ou 7, pour améliorer la régénération cutanée liés au vieillissement cutané normal.
[Revendication 9] Utilisation cosmétique selon la revendication précédente, caractérisée en ce que l'extrait ou la composition améliore l'inter-connectivité des systèmes cutané, vasculaire et immunitaire.
[Revendication 10] Utilisation cosmétique selon la revendication précédente, caractérisé en ce que l'extrait ou la composition augmente la synthèse et la sécrétion de
KGF, EGF, IGF, FGF, PDGF, TGF-|3 et VEGF.
[Revendication 11] Utilisation cosmétique selon l'une des revendications 8 à 10 pour améliorer l'éclat du teint.
[Revendication 12] Utilisation cosmétique selon l'une des revendications 8 à 11 pour prévenir et/ou réduire les cernes.
