Description
本発明は、ウェルプレート密閉用の蓋、ウェルプレートセット、及び、測定方法に関する。
細胞培養液、薬品、及び、体液等の液体試料についてハイスループット測定を行うために、ウェルプレート(マイクロプレート、マイクロタイタープレート)を用いた測定が行われている。ウェルプレートは一般に6~9600ウェルまでの様々なサイズのものが使用されている。なかでも一般的なのは、各ウェルの容量が200μL程度の96穴の成形プラスチックウェルを有するウェルプレートである。
ウェルプレートを用いた液体試料の測定では、ウェルプレートの開口からの液体媒体の蒸発による液体試料の濃縮や夾雑物質の混入を防ぐために、プラスチック製の密閉シート(プレートシール)が用いられることがある。
また、ウェルプレートの蓋に、密閉機能を持たせたものも知られており、特許文献1には、「密閉器であって:マイクロプレートと共に使用すべく設計されており、外側面と内側面を有する蓋;及び頂面側と底面側を有するマイクロプレートマットであって、該底面側は、そこに、マイクロプレートの開口の配列に対応し、且つそれらを密閉するように形成された多数の突起部を有しており、該マイクロプレートマットの該頂面側が該蓋の該内側面に取り付けられるマイクロプレートマット;からなる密閉器。」が記載されている。
ウェルプレートを用いて、微生物が行う嫌気呼吸、及び、発酵等による電子伝達をハイスループットで電気化学的に測定しようとする試みがある。嫌気環境下における微生物の活動が疾病等の発生とも関連があることがわかってきており、血液、及び、だ液等の生体由来の検体を底面に電極が配置されたウェルプレートに収容して微生物由来の電流生成等を検出することでその活動を明らかにしようとする試みである。
従来、このような目的のためには、測定装置(ウェルプレートリーダー)を嫌気グローブボックス内に配置して、嫌気環境下で測定が実施されてきた。
しかし、嫌気グローブボックスは大掛かりな装置であり、疾病の体外診断等に適用しようという場合、検体の採取場所である医療機関等にこのような装置を配置する余裕はなく、オンサイトでの測定が難しいという問題があった。

これに対して、従来の密閉シートや蓋を用いれば、ウェル内に供給される空気(酸素)を制限することができるものの、依然としてウェル内にはヘッドスペースに一定量の空気(酸素)が存在し、嫌気測定を実施することは難しかった。
反対に、ヘッドスペースを小さくするために液体試料の量を増やすと、蓋をした際に試料の一部がウェル外に漏れて、ウェル間のコンタミネーションを引き起こしたり、ウェル外にあふれ出た液体試料によって測定装置が故障してしまうことがあった。

そこで、本発明は、嫌気グローブボックスを用いなくてもハイスループットな嫌気測定を行うことができるウェルプレートの密閉用の蓋を提供することを課題とする。また、本発明は、ウェルプレートセット、及び、測定方法を提供することも課題とする。
本発明者らは、上記課題を達成すべく鋭意検討した結果、以下の構成により上記課題を達成することができることを見出した。
[1] 上側に開口を有する複数のウェルが配列して形成されたウェルプレートの密閉用の蓋であって、上記ウェルの開口と嵌合し、先端が上記ウェルに進入して、上記ウェルに収容される液体試料の液面と接触するよう設けられた突起を有し、上記突起は、その外周面に、径方向内側に向けて凹入するようにして、上記先端から上側に向けて延びる凹溝を有する、ウェルプレートの密閉用の蓋。

本発明によれば、嫌気グローブボックスを用いなくてもハイスループットな嫌気測定を行うことができるウェルプレートの密閉用の蓋が提供できる。また、本発明は、ウェルプレートセット、及び、測定方法を提供することもできる。
以下、本発明について詳細に説明する。
以下に記載する構成要件の説明は、本発明の代表的な実施形態に基づいてなされることがあるが、本発明はそのような実施形態に制限されるものではない。

[ウェルプレートセット]
本発明の実施形態に係るウェルプレートセットの構成について、図1~4を用いて説明する。まず、図1は、ウェルプレートセットの斜視図である。

ウェルプレートセット10は、ウェルプレート15と、密閉用の蓋11とにより構成されている。ウェルプレート15は、ウェルプレート本体18を有しており、これには、液体試料を収容するためのウェル16が複数配置されている。本実施形態においては、ウェル16がマトリクス状に96穴(8×12)形成された透明なマイクロプレートとなっている。但し、ウェル16の数は、本実施形態のものに限定されない。容器本体は、例えば、ポリスチレン、及び、ポリオレフィン等の樹脂、又は、ガラス等から形成でき、透明でなくてもよい。
また、ウェル16は底がある円筒状の内周面を有している。ウェル16の内周面はいずれも同一の素材で形成されているが、本発明のウェルプレートセットにおけるウェルプレート本体としては上記に限定されない。例えば、ウェル16の底面だけがガラス製、又は、透明プラスチック製で、他は、有色のプラスチック製であってもよい。また、底に電極が配置されていてもよい。電極が配置される場合、例えば、作用極、対極、及び、参照極が配置され、ウェルプレート本体18の底面に各電極から引き出された端子が配置され、測定装置と接続可能に構成されることが好ましい。
ウェル16の容積は例えば、300μL程度であり、一般に、50~200μL程度の液体試料が収容されて、測定に供される。各ウェルの開口面と一致する容器本体の上面17は平滑な平面状とされている。
密閉用の蓋11は平板状の蓋本体12と、蓋本体12から突出する複数の突起13とを備えている。密閉用の蓋11についても、ウェルプレート本体18と同様の材料から形成できる。
突起13は、各ウェル16の開口面に対応する位置に配置されていて、各ウェル16と嵌合して、これを密閉する。図1では、突起は8×12の96個、配置されているが、ウェルプレート本体18が有するウェル16の数に応じて変更される。

密閉用の蓋11をウェルプレート本体18上に載置すると、ウェル16に突起13が嵌め合わされて進入し、ウェル16内の空気がウェル16外へと押し出され、ウェル16が密閉される。このとき、ウェル16に収容される液体試料の液面と突起13の先端とが接触することで、ウェル16の内周面と突起の先端とによって形成される空間はより空気(特に酸素)が少ない状態となり、従来、大規模な嫌気ボックス内で行われてた嫌気環境下での測定がより簡単な装置で実施できる。
図2は、ウェルプレート本体18上に、密閉用の蓋11が載置された状態のウェルプレートセットのA-A′断面図であり、図3は、B-B′断面図である。
各ウェル16には、突起13が嵌め合わされ、ウェル16が密閉されている。ウェル16の内周面と、突起13の先端21とによって画される空間Cには液体試料が収容される。

突起13は、その外周面に、径方向内側に向けて凹入するようにして、先端21から上側(Z方向)に向けて延びる凹溝14を有している。図中、凹溝14は、先端から、蓋本体12まで伸びているが、上記形態に限定されず、先端21を起点に、蓋本体12に至る突起13の外周面のいずれかの位置を終点としてもよい。
また、図中凹溝14は、先端21から蓋本体12方向へと延びる突起13の高さ方向に平行な直線状であるが、凹溝は、上記形態に限定されず、例えば、突起13の先端21を起点とし、突起13の外周面をらせん状に延びる曲線等であってもよいし、その途中、又は、終点に液だまりが設けられていてもよい。
また、凹溝は、各突起に少なくとも1つ設けられていればよく、複数設けられていてもよい。
また、図中凹溝14は、先端21から蓋本体12方向へと延びる突起13の高さ方向に平行な直線状であるが、凹溝は、上記形態に限定されず、例えば、突起13の先端21を起点とし、突起13の外周面をらせん状に延びる曲線等であってもよいし、その途中、又は、終点に液だまりが設けられていてもよい。

次に、ウェルプレートセット10の使用方法について説明する。
まず、ウェルプレート本体18の各ウェル16に液体試料が収容される。このとき、96穴の全てに液体試料が収容されてもよいし、収容されないウェル16があってもよい。測定を実施するウェル16については、いずれも同様の量の液体試料が収容されることが好ましい。

収容される液体試料の量は、突起13の蓋本体12からの高さに応じて決定される。すなわち、空間C(図2)を満たす量の液体試料が各ウェル16に収容される。そのようにすることで、ウェル16内の空気が十分にウェル外に押し出されるために、酸素がより少ない状態で測定ができる。
最適な液体試料の量は、ウェル16の底面積、及び、突起13の高さから適宜計算できる。

使用される液体試料としては特に制限されず、だ液、及び、血液等の体液、並びに、細菌等の培養液等の生体試料、一般的な測定に用いられる液体試料をいずれも利用できる。
なかでも、より簡便に嫌気測定が実施できる観点で、液体試料にはヒドラジン、タンニン、アスコルビン酸及びその塩、グリオキシル酸及びその塩、ヒドラジン、ジメチルヒドラジン、ヒドロキノン、ジメチルアミンボラン、NaBH4、NaBH3CN、KBH4、亜硫酸及びその塩、チオ硫酸及びその塩、ピロ亜硫酸及びその塩、亜リン酸及びその塩、及び、次亜リン酸及びその塩等の還元剤が添加されることが好ましい。
なかでも、より簡便に嫌気測定が実施できる観点で、液体試料にはヒドラジン、タンニン、アスコルビン酸及びその塩、グリオキシル酸及びその塩、ヒドラジン、ジメチルヒドラジン、ヒドロキノン、ジメチルアミンボラン、NaBH4、NaBH3CN、KBH4、亜硫酸及びその塩、チオ硫酸及びその塩、ピロ亜硫酸及びその塩、亜リン酸及びその塩、及び、次亜リン酸及びその塩等の還元剤が添加されることが好ましい。
本ウェルプレートセット10では、ウェル内の空間Cが液体試料の体積と略同程度に調整されるため、もともと空気(酸素)の少ない環境下で測定可能である上に、液体試料に還元剤が添加されている場合、より安定的に嫌気測定が実施できる。
各ウェル16に所定量の液体試料を収容した後、ウェルプレート本体上に蓋11が載置される。図4は、各ウェル16に液体試料が収容されたウェルプレート本体18上に、蓋11が載置された状態のウェルプレートセットのA-A′断面図(図2と同様の断面図)である。
これを測定装置にセットして測定すればよい。測定の方法としては特に制限されないが、ウェル16が電極を有する(底部に電極が印刷されている場合)電気化学測定であることが好ましい。

図4において、各液体試料Lのうち、空間Cに収まらなかった分が、凹溝14に収容されている。一般に、ウェルプレートには、製造上の寸法状のばらつきがあり、かつ、ウェル毎に同量の液体試料を収容したとしても、分注のばらつき等があるため、突起13の先端21を液体試料の液面と接触するよう調整すると、液体試料があふれてしまうウェルが出てくる場合がある。
しかし、本ウェルプレートセット10は、突起13が凹溝14を有するために、液体試料は凹溝14にトラップされて、ウェル16外まであふれることがない。図4においては、トラップされている液体試料の量がh1とh2とで異なっているが、本ウェルプレートセット10では、凹溝14によって、寸法のばらつきや、分注操作のばらつきによる差があっても、凹溝14がバッファーとなり、液体試料Lがウェルプレート外にあふれにくい。突起13が複数の凹溝14を有している場合、この傾向はより顕著である。
なお、突起13に形成されたのと同様の凹溝をウェルプレート本体18に形成してもよい。すなわち、ウェル16の内周面に、径方向外側に向けて凸出するようにして、Z方向に向けて延びる凹溝を設けてもよい。これにより、更に優れた効果が期待できる。なお、ウェルプレート本体18に形成される凹溝の位置は特に制限されないが、一形態として、前記突起13に形成された凹溝14と対向する位置に配置され、かつ、凹溝14と同様の形状であることが好ましい。

微生物の嫌気呼吸や発酵を電気化学的に検出するために、ウェル底面に電気化学測定用の電極が配置されている場合、マイクロプレート底面には、上記電極と外部配線とを接続するための端子が配置されていることがある。この場合、液体試料がウェル外へとあふれてしまうと、これらの接続に悪影響を及ぼしたり、場合によっては、装置を破損してしまう場合がある。
ウェルプレートセット10は、特にこのような電気化学測定を行うためのウェルプレートセットとして適している。

Claims (11)
- 上側に開口を有する複数のウェルが配列して形成されたウェルプレートの密閉用の蓋であって、
前記ウェルの開口と嵌合し、先端が前記ウェルに進入して、前記ウェルに収容される液体試料の液面と接触するよう設けられた突起を有し、
前記突起は、その外周面に、径方向内側に向けて凹入するようにして、前記先端から上側に向けて延びる凹溝を有する、ウェルプレートの密閉用の蓋。 - 前記突起が、複数の前記凹溝を有する、請求項1に記載のウェルプレートの密閉用の蓋。
