Abstract
일반적으로 노화의 증상 중 하나인 세포 증식에 관해서 HTS screening을 진행했다. 그 결과, 세포 증식에 가장 효과적이였던 화합물 KB1541을 얻었다. 본 발명에서는 이 약물이 어떠한 기전을 통해서 세포증식, 노화를 제어하는지에 대해서 연구했다. KB154에 비오틴을 연결한 약물을 처리해 streptavidin-magnetic beads를 이용하여 해당 약물과 상호작용하는 단백질이 무엇이 있는지를 알아보았고, 그 결과로 미토콘드리아 관련 단백질들이 상호작용하는 것을 알아냈다. ATP assay, 전자 현미경, IP 실험을 통해 해당 약물이 미토콘드리아 관련 단백질들을 조절하며, 결과적으로 ATP 생산량의 증가 및 노화 회복에 도움이 된다는 것을 확인하였다.

본 발명은 세포노화 관련 질병의 예방 또는 치료용 약학적 조성물에 관한 것으로, 구체적으로 하기 [화학식 1]의 화합물을 유효 성분으로 포함하는 세포노화 관련 질병의 예방 또는 치료용 약학적 조성물에 관한 것이다,

노화의 미토콘드리아 이론(Mitochondria theory of aging)은 자유라디칼 가설에 근간을 두고 산화 스트레스에 의해 초래되는 미토콘드리아의 손상과 기능부전이 노화 및 노화 관련질환의 근본 원인이 된다고 주장한다. 즉 미토콘드리아에서 생성되는 활성 산소에 의해 DNA의 체세포 돌연변이가 나타나고 이로 말미암아 미토콘드리아 DNA의 전사 장애가 나타나 단백질 합성이 저하되고 이에 따라 더욱 많은 양의 활성 산소가 발생하는 악순환(vicious cycle)이 반복되어 신체의 노화와 노화 관련 질환들이 나타난다는 것이다. 최근 들어 미토콘드리아의 기능이 동맥경화 및 혈관기능, 심부전, 암, 퇴행성 뇌질환(파킨슨병, 알츠하이머병 등), 당뇨병 등이 노화와 관련이 있음을 시사하는 구체적인 증거들이 밝혀지면서 건강증진과 질병예방을 위해 미토콘드리아의 기능 유지 및 회복이 매우 중요한 과제로 인식되고 있다.
미토콘드리아는 세포의 에너지 생산에 탁월한 영향을 미치는 것으로 알려져 있다. 미토콘드리아 기능 장애가 발생하면 ROS 증가, 전자 수송 시스템에서 ATP 생성 감소, 해당 과정에 대한 의존도 증가, 효율적인 에너지 생성 감소로 인한 세포주기 정지를 유발한다.
미토콘드리아에는 이중 막이 존재한다. 이중 막은 내막(inner membrane)과 함께 외막(outer membrane)으로 구성된다. 세포 호흡과 관련된 단백질은 IM8의 크리스테(cristae)에 집중되어 있다.
*미토콘드리아 IM의 여러 단백질은 에너지 생산에 중요한 기능을 한다. MICOS 복합체, COX 1-4 및 ATP 합성효소(synthase) 단백질은 미토콘드리아 IM를 구성하는 단백질로 잘 알려져 있다. ATP 합성효소의 F1 도메인은 ATP 합성효소 서브유닛 알파 및 ATP 합성효소 서브유닛 베타의 어셈블리로서 ADP 결합 도메인을 형성한다. 이렇게 형성된 구조에서 ADP와 인산염을 ATP으로 변환하여 에너지를 생성한다. 14-3-3 제타/델타 단백질은 여러 단백질을 매개하는 어댑터 단백질로 알려져 있다. 그러나 자세한 규제 과정은 확인되지 않았으며 이러한 단백질 간의 상호 작용 또는 연관성은 정확하게 확인되지 않았다.

본 발명의 KB1541는 14-3-3 제타/델타(zeta/delta) 단백질을 활성화하고, 본 발명의 KB1541에 의하여 활성화된 14-3-3 제타/델타 단백질을 매개로 하여 ATP 합성효소의 수를 증가시키고 활성화시키는 것을 확인하였다. 또한 KB1541의 처리에 의하여 미토콘드리아의 크리스테(cristae) 수 및 길이가 증가하는 것을 확인하였다. 이는 ATP 합성효소 조립 메커니즘의 초기 발견과 동시에 노화 회복을 위한 효과적인 치료 후보를 확인한 것으로, 본 발명의 KB1541는 노화 회복뿐만 아니라 노화 질환에도 매우 효과적인 약물로서 잠재적 가치가 있다고 할 수 있다.
도 1은 노화 개선을 위한 잠재적인 표적 스크리닝 결과이다. 각 약물은 1,000개의 노화 세포(4μM)가 접종된 96 웰 플레이트에서 처리되었다. 12일 동안 배양한 후 세포를 Gel green 용액(D.W.에서 1:1,000)으로 염색했다. 형광 마이크로 플레이트 리더(여기/방출 485nm/535nm)를 사용하여 형광을 측정하여 세포 수를 결정했다.

구체적으로, 상기 세포노화는 미토콘드리아 기능 저하로 인해 유발되는 것을 특징으로 한다. KB1541은 14-3-3 제타/델타 단백질을 표적으로 하며, 14-3-3 제타/델타 단백질이 KB1541의 처리에 의해 활성화되고, 또한 크리스테(cristae)의 수 및 길이 증가하였다.
KB1541는 14-3-3 제타/델타 단백질을 상향 조절(upregulated), 즉 활성화함으로써 ATP 합성효소의 알파 및 베타 서브 유닛을 수집하고, ATP 합성효소의 개수를 증가시키고, ATP 합성효소를 활성화시켜 효율적인 에너지 생산을 가능하게 한다.
본 발명자는 KB1541를 처리하는 경우 미토콘드리아의 크리스테의 수가 증가하고 길이가 증가하는 것을 확인하였으며, ATP 합성효소가 증가하고 또한 활성화되는 것을 확인하였다. ATP 합성효소 F1 도메인은 3개의 ATP 합성효소 서브유닛 A와 3개의 ATP 서브유닛 베타로 구성되어 있으며, ATP 합성효소는 각 서브유닛의 어셈블리로 형성된다.
14-3-3 제타/델타 단백질은 ATP 합성효소 서브유닛 알파 및 베타와 상호 작용하는 것으로 알려져 있으며, 본 발명자는 14-3-3 제타/델타 단백질이 ATP 합성효소 서브유닛 알파 및 베타를 끌어당겨 조립을 일으킨다는 것을 확인하였다. 구체적으로 14-3-3 제타/델타 단백질 58번째 세린의 인산화는 알파와 베타 서브유닛을 끌어 당겼고, ATP 합성효소 어셈블리는 미토콘드리아 결정에 존재하는 ATP 합성효소를 증가시키기 위해 유도되었다.
ATP 어셈블리가 증가하면서 ATP 생산 효율이 크게 향상되었다. 세포의 핵심 기능인 미토콘드리아 기능은 에너지 생산 효율에 크게 의존한다. 에너지 생산 효율의 증가는 기능 장애 미토콘드리아의 정상화를 의미한다. 차례로 미토콘드리아 기능이 회복된다.
또한 상기 화합물은 세포의 자가 포식(autophagy)을 활성화하며, 상기 화합물은 세로 내 활성산소(ROS)를 감소시키는 것을 특징으로 한다.

상기 피부 질환 또는 장애의 예는 건선, 습진, 주름, 탄력감소, 소양증, 감각장애, 구진설장애, 홍색피부증, 편평태선, 태선모양피부병, 모반, 발진, 두드러기, 아토피성피부염, 호산구피부병, 반응성호중구피부병, 천포창, 유천포창, 면역수포성피부병, 피부섬유조직구증식, 피부림프종, 피부루푸스, 과색소침착, 흉터, 켈로이드, 딸기코, 백반증, 비늘증, 피부근염, 검버섯, 잡티, 주근깨, 기미, 광선각화증 등을 포함한다.

<실험예>
1. KB1541 및 비오틴화된 KB1541 합성
(1) KB1541 합성
KB1541의 합성 방법은 도 2A를 참고하여 설명한다. 화합물 1은 80℃에서 2시간 동안 아세토니트릴 중 tert-부틸 니트릴과 염화구리(II)를 처리하여 시판되는 에틸 2-아미노옥사졸-4-카르복실 레이트로부터 64% 수율로 합성되었다. 화합물 1을 톨루엔에 4-(트리플루오로메틸)페닐보론산, 테트라키스(트리페닐포스핀)팔라듐(0) 및 2M 탄산칼륨 용액을 80℃에서 1시간 동안 반응시켜 64% 수율로 화합물 2를 얻었다. 화합물 3은 2-요오도니트로벤젠, 팔라듐 아세테이트, 트리페닐포스핀, 탄산 세슘을 톨루엔에 넣고 90℃에서 3시간 동안 반응시켜서 44% 수율로 얻었다. 화합물 3의 니트로 그룹은 메탄올 중 활성 탄소 중 10 중량% 팔라듐 촉매량으로 환원되어 화합물 4를 제공한다. 혼합물을 수소 가스(50psi) 하에 실온에서 1시간 동안 98% 수율로 흔들었다. 화합물 4의 분자 내 고리화는 에틸렌글리콜 디메틸에테르(DME) 및 2M 탄산칼륨 용액으로 90℃에서 12시간 동안 수행되어 화합물 5를 72% 수율로 수득하였다. 화합물 6은 톨루엔에서 화합물 5와 옥시 염화인을 120℃에서 4시간 동안 84% 수율로 반응시켜 수득하였다. 화합물 7(KB 1541)은 화합물 6과 피롤리딘을 80℃에서 3시간 동안 반응시켜 68% 수율로 얻었다. 간단히 말해서, 총 7단계의 반응이 상업적 공급원으로부터 구입한 에틸 2-아미노옥사졸-4- 카르복실레이트를 사용하여 수행되었다. 순서대로 샌드마이어 반응, 스즈키 반응, 헥 반응, 수소화, 고리 화, 염소화 및 알킬화이다.
(2) 비오틴화된 KB1541 합성
비오틴화된 KB1541 합성은 도 2B를 참고하여 설명한다. 화합물 7(KB 1541)이 스크리닝된 화합물 중 가장 강력한 분자로 간주되었기 때문에 본 발명자는 비오틴과 스트렙타비딘 사이의 결합 친화성을 사용하여 풀다운 분석을 수행하기로 결정했다. 풀다운 분석을 통해 화합물 7이 결합하는 정확한 단백질을 확인할 수 있다. 화합물 9는 시판되는 화합물 8을 활성 탄소 중 10 중량% 팔라듐 및 메탄올 중 촉매량의 아세트산과 반응시켜 탈벤질화를 통해 수득되었다. 상기 혼합물을 수소 가스(50psi) 하에서 실온에서 8시간 동안 흔들었다. 화합물 6과 테트라하이드로퓨란(THF)에 과량의 트리에틸아민(TEA)을 60℃에서 반응시켜 화합물 10을 얻었다. 화합물 10에서 Boc 그룹의 탈보호는 트리플루오로아세트산(TFA)을 디클로로메탄에서 실온에서 3시간 동안 처리하여 화합물 11을 수득함으로써 달성되었다. 이 반응을 통해 화합물 7에 링커가 결합된 화합물을 얻을 수 있었다. 조 생성물 11을 추가 정제없이 최종 단계에 사용하였다. 화합물 11을 디메틸포름아미드(DMF) 중의 N-숙신이미딜 D-비오티네이트, TEA와 실온에서 반응시켜 화합물 12를 53% 수율로 얻었다.
2. 약물 스크리닝
노화 섬유아세포(Senescent fibroblast)는 웰당 1,000개 세포의 밀도로 96 웰 플레이트에서 성장했다. 약물 라이브러리의 구성 요소를 최종 농도 4μM로 희석하고 4일마다 웰에 추가했다. 약물 처리 후 12일에 세포를 인산염 완충 식염수(PBS)로 두 번 세척하고 50μl의 0.2% SDS에 용해시켰다. 플레이트를 37℃에서 1시간 동안 배양하였다. SYBR Green I(150 μl) nucleic acid gel stain(1:1,000 in DW; S-7567; Molecular Probes, Eugene, OR, USA)을 웰에 추가했다. VICTOR Multilabel Plate Reader(2030-0050; PerkinElmer, Waltham, MA, USA)를 사용하여 Plates를 사용하여 형광 강도를 측정하여 세포 수를 측정했다. 6회 반복의 평균 및 표준 편차는 각 실험 그룹에 대해 결정되었다.
3. IM-MS/MS TOF 분석
HDF(Human diploid fibroblasts) 세포를 비오티닐화 KB1541로 14일 동안 처리한 후, 스트렙타비딘 자기 비드를 사용하여 면역 침전을 수행했다. 용출된 단백질은 한국 기초 과학 연구원에서 IM-MS/MS(ST006; 서울 센터) 시퀀싱 및 데이터 분석을 거쳤다.
4. 플라스미드 제조
이 실험에 사용된 플라스미드는 인간 MIC60 서브유닛과 14-3-3 제타/델타 단백질을 암호화하는 HDF cDNA에서 증폭하여(표 1), pCMV-Myc-puromycin 벡터에 삽입하였다. 14-3-3 제타/델타(S58A) 돌연변이 유형은 부위 지정 돌연변이 유발에 의해 복제되었다.
5. 부위 지정(Site-directed) 돌연변이 유발
플라스미드를 주형 벡터로 사용하여 PCR 프라이머를 준비한다. 돌연변이 유발할 서열을 포함하여 2개의 PCR 프라이머를 역방향으로 구축하였다(표 1). 증폭 된 PCR 산물을 Dpn I로 1시간 동안 37℃에서 처리하고 PCR 정제를 통해 정제했다. 정제된 PCR 산물을 HiFi DNA Assembly Master Mix(E2621L; New England Biolabs)를 사용하여 결찰하고 DH5α Chemically Competent E. coli(CP011; Enzynomics, 대전, 한국)로 형질 전환했다.
6. 공동 면역 침전(Co-immunoprecipitation)
pCMV-Myc-14-3-3 zeta/delta(WT), pCMV-Myc-14-3-3 zeta/delta(S58A) 플라스미드는 HEK293T(CRL-11268; ATCC)로 형질 감염되었습니다. 형질 감염 후 5μg/ml 퓨로마이신 농도에서 14일 동안 선별을 수행하고, 선별된 세포주에 8μM 농도의 비오티닐화 KB1541 약물을 5일 동안 처리하였다. 약물 처리된 세포를 0.1 % NP-40 세포 용해 완충액으로 분해하여 세포를 파괴하고, AccuNanoBeadTM Streptavidin Magnetic Nanobeads(TA-1015-1; Bioneer, 대전, 한국)를 사용하여 MIC60 subunit의 IP를 수행했다. 14-3-3 zeta/delta(WT) 및 14-3-3 zeta/delta(S58A)의 IP는 anti-Myc 태그 항체를 사용하여 수행되었다.
7. 자가 포식 플럭스(autophagic flux) 측정
세포는 20 μM 클로로퀸(CQ; C6628, Sigma-Aldrich)이 포함된 배지에서 배양되었다. 배양 24시간 후, 세포를 Cyto-ID 염색 용액과 50 nM LTDR(ENZ-51031-0050; ENZO, Lausen, Switzeland)로 30분 동안 추가로 염색하고 FACS 분석을 위해 준비했다. 배경 자가 형광을 측정하기 위해 세포를 염료없이 배지에서 배양했다. Cyto-ID의 형광은 LTDR의 형광으로 정규화되었다. 자가 포식 플럭스는 다음 방정식을 사용하여 계산되었다; ΔMFI Cyto-ID = MFI Cyto-ID (+CQ)/ MFI Cyto-ID (-CQ).
8. 활성 산소 종(ROS), 미토콘드리아 질량 측정
미토콘드리아 ROS의 정량화를 위해 세포를 5μM MitoSOX(M36008; Life Technologies, Carlsbad, CA, USA)를 포함하는 배지에서 30분 동안 37℃에서 배양했다. 미토콘드리아 질량의 정량화를 위해, 세포를 37℃에서 30분 동안 50 nM MitoTracker green(M22426; Life Technologies)을 포함하는 배지에서 배양했다. 미토콘드리아 막 전위 측정을 위해 염색 후, FACS 분석을 위해 세포를 준비하였다.
9. 14-3-3 제타/델타(WT) 및 14-3-3 제타/델타(S58A)의 과발현
14-3-3 zeta/delta(WT) 및 14-3-3 zeta/delta(S58A)의 PCR 단편을 pCMV-Myc-puromycin에 별도로 클로닝했다. HEK293T(CRL-11268; ATCC)는 LipofectamineTM 2000 Transfection Reagent(11668-019; Thermo Scientific)로 형질 감염되었으며, 선택은 퓨로마이신 3μg/ml로 수행되었다.
10. 해마 분석(Seahorse analysis)
XFe24 플럭스 분석기(Seahorse Bioscience XFe24 Instrument)는 제조업체의 프로토콜에 따라 사용되었다. 간단히 말해서, XF24 FluxPak(100850-001; Seahorse Bioscience, North Billerica, MA, USA)에서 XFe24 세포 배양 플레이트의 각 웰에 5 Х 104개 세포를 배포한 다음 5% CO2 인큐베이터에서 16시간 동안 37℃ 온도에서 배양했다. 다음으로, 배지를 포도당이 부족한 XF Assay 배지(102365-100; Seahorse Bioscience)로 교체한 다음 동일한 배양기에서 세포를 1시간 더 배양했다. 세포 외 산성화 속도(ECAR, extracellular acidification rate)은 XF Glycolysis Stress Test 키트(102194-100; Seahorse Bioscience)를 사용하여 측정되었다. ECAR은 mpH/min으로 보고되었다. 대사 전환제로 아코니타제 억제제인 데페리프론(379409-5G; Sigma-Aldrich)이 사용되었다.
11. 세포 ATP 수준 측정
세포를 20 μM 올리고마이신이 있고/없는 배지에서 24시간 동안 배양한 다음 용해 완충액으로 용해시켰다. ATP 함량은 제조업체의 지침에 따라 ViaLight Plus Kit(LT07-221, Lonza, Basel, Switzerland)를 사용하여 측정되었다. DNA 함량은 AccuBlue broad range dsDNA quantitation kit(31007; Biotium, Fremont, CA, USA)를 사용하여 측정되었다. 상대 ATP 함량 측정을 위해 각 샘플의 발광을 DNA 함량으로 정규화했다.
<실험 결과>
1. 노화를 완화하는 약물 스크리닝
세포 노화는 세포주기 정지가 특징이고, 본 발명자는 이러한 사실을 바탕으로 세포주기 정지를 회복시키는 약물을 검색하고 추가적인 노화 방지 효과가 있는 약물을 찾았다. 스크리닝 전략은 세포 노화를 회복시키는 약물을 찾는 데 사용되었다. HTS 실험을 통해 4μM 농도의 27개 약물 각각을 웰당 1,000개씩 접종한 노화 HDF 세포를 처리한지 12일 후에 측정을 수행했다(도 1). 상대적 세포 증식 효과는 노화 HDF를 기준으로 측정되었다. 스크리닝 결과 세포 증식 효과가 가장 큰 약물이 KB1541임을 확인하였다(도 2A). 이 약물은 노화 회복에 효과적인 것으로 생각되어 후속 실험에 사용되었다.
2. KB1541 및 14-3-3 제타/델타 단백질 상호 작용
본 발명자는 KB1541이 세포 증식에 긍정적인 영향을 미치기 때문에 세포 노화 회복에 효과적인 약물이 될 수 있다고 가정했다(도 1).
본 발명자는 KB1541이 세포에서 어떻게 작동하는지 확인하기 위해 KB1541에 비오틴이 부착된 약물 바이오닐화 KB1541을 설계했다(도 2B). 약물을 4μM 농도의 senescent HDF로 처리한 후 IP를 통해 얻은 샘플을 IM-MS/MS TOF에 적용했다.
IM-MS/MS TOF 실험의 결과, 937개의 단백질과의 상호 작용 결과가 얻어졌다. 수많은 데이터 중에서 선택된 단백질은 노화와 p-값에 초점을 두고 접근했으며 추가 실험이 수행되었다(도 3B).
ATP 합성효소 알파 및 베타 서브유닛은 미토콘드리아 크리스테(cristae)를 구성하는 많은 단백질 중에서 ATP 합성효소의 구성 요소로 알려져 있다. 그 주요 기능은 ADP를 ATP로 변환하는 것이다. 14-3-3 제타/델타 단백질은 어댑터 단백질로 알려져 있다.
본 발명자는 ATP 합성효소 서브유닛 알파(synthase subunit alpha), 베타(beta)와 14-3-3 제타/델타 상호 작용(interaction)을 가정했고 ATP 서브유닛을 조립하기 위해 14-3-3 제타/델타 단백질을 예상했다.
KB1541에 대한 14-3-3 제타/델타 단백질의 도킹 위치는 상당한 결과를 보여주었다(도 3C). 14-3-3 zeta/delta-coding 유전자와 IP를 스트렙타비딘으로 형질 감염시킨 HEK293T에서 비오티닐화 KB1541 약물로 처리한 결과, 비오티닐화 KB1541 약물과 상호 작용한 도 3B의 결과가 재검증되었다(도 3D).
3. 14-3-3 제타/델타 단백질과 관련된 ATP 합성효소를 조절하기 위한 메커니즘
상기 결과는 KB1541이 상위 단백질인 14-3-3 제타/델타 단백질과 상호 작용한다는 것을 보여주었다. 이는 미토콘드리아 크리스테의 구성 요소이며 ATP 합성 효소 서브 유닛 단백질은 14-3-3 제타/델타 단백질의 영향을 받을 것으로 예상된다.
본 발명자는 하위 단백질이며 노화와 직접 관련된 에너지 생성 단백질인 ATP 합성효소에 초점을 맞추었다. ATP 합성효소에는 여러 유형이 있으며 ATP는 미토콘드리아 크리스테에 존재한다. ATP 합성효소가 생성되는 것의 측정은 ATP 서브유닛의 조립으로 볼 수 있다.
따라서 본 발명자는 어댑터 단백질로 작용하는 14-3-3 제타/델타를 통해 ATP 합성효소 알파와 베타의 두 가지 단백질을 조사했다.
정상 단백질은 번역 후 변형(PTM, post translational modification)을 통해 기능을 조절한다. PTM의 인산화는 중요한 단백질 조절 메커니즘 중 하나로 알려져 있다. 본 발명자는 14-3-3 제타/델타 단백질의 5개 인산화 부위 중 하나인 58번째 세린 영역을 ATP 합성효소 알파와 베타를 연결하는 PTM 부위로 간주했다(도 4A).
따라서 그 기능을 조사하기 위해 14-3-3 제타/델타의 58번째 세린 영역은 알라닌으로 변이되었다. Myc-태그 14-3-3 zeta/delta(WT) 및 14-3-3 zeta/delta(S58A)의 두 벡터는 HEK293T로 형질 감염 및 과발현되었으며 내인성 ATP 합성효소 알파 및 베타와의 상호 작용은 IP로 확인되었다(도 4B).
그 결과, KB1541을 4μM 농도로 HEK293T에 처리했을 때 Myc-14-3-3 zeta/delta(WT)와 상호 작용하는 ATP 합성효소 알파 및 베타 단백질이 증가했다(도 5B). 또한 동일한 방식으로 Myc-14-3-3 zeta/delta(S58A) IP와 상호 작용하는 ATP 합성효소 알파 및 베타 단백질이 크게 변하지 않은 것으로 나타났다(도 4B).
도 4B의 결과를 바탕으로 실제로 ATP 서브유닛을 조립할 때 ATP 생산의 변화를 조사했다. oxphos 부분(oxphos portion)은 미토콘드리아 크리스테에서 산화적 인산화의 결과로 생성된 ATP를 확인하기 위해 측정되었다(도 4C). oxphos 부분의 결과에 따르면, KB1541로 처리했을 때 senescent HDF보다 높았다. 또한 해당 과정 동안 ATP의 양을 측정하기 위해 해당 과정 부분(glycolysis portion)을 측정했다(도 4D). KB1541이 처리되었을 때, 해당 과정 부분은 senescent HDF에 비해 감소되었다(도 4D).
4. 전반적인 노화 마커 감소를 통한 노화 회복
자세한 메커니즘을 살펴 보았지만 궁극적으로 이 약물이 노화 회복에 얼마나 효과적인지 알아보기 위해 노화 마커를 조사했다.
먼저 정상적인 미토콘드리아 대사와 항상성을 손상시키는 물질인 활성 산소 종(ROS)을 측정하고 조사했다. 정상적인 노화 세포에서 ROS가 크게 증가하여 노화가 진행 중임을 나타낸다. KB1541은 약물을 처리했을 때 노화 HDF에 비해 약 절반으로 감소하는 것으로 나타났다(도 5A).
노화 세포에서는 기능 장애가 있는 세포 기관을 빠르게 제거하는 능력이 약화된다. 자가 포식 기능의 회복은 기능 장애 미토콘드리아의 축적을 억제하여 노화 회복의 징후를 가져온다. 이러한 기능을 조사하기 위해 미토콘드리아 질량(mitochondrial mass) 및 자가 포식(autophagy)을 측정했다. KB1541 약물로 처리된 HDF에서 미토콘드리아 질량은 감소했다(도 5B). 또한 자가 포식 수준은 증가했다(도 5C).
마지막으로 해당 작용의 의존성은 ECAR을 통해 측정되었다. 이전 ATP 부분과 마찬가지로 KB1541 약물 처리 HDF는 포도당에 관계없이 해당 작용 의존성이 감소한 것으로 나타났다(도 5D 및 E).
본 발명자는 노화 HDF로 처리 후 21일 후에 미토콘드리아 크리스타 변화를 조사하여 실제로 노화 회복이 발생했는지 확인했다. 전자 현미경으로 촬영한 미토콘드리아를 보면 노화 세포는 주름이 훨씬 적다는 것을 알 수 있으며, KB1541이 처리되었을 때 다시 회복되었고 주름이 증가했다(도 6A). 이 사진을 이미지를 통해 정량화하여 그래프로 표현한 경우에도 KB1541 약물을 적용했을 때 크리스테 길이가 증가한 것을 확인할 수 있다(도 6B).
요약하면, 본 발명의 KB1541은 14-3-3 제타/델타 단백질을 상향 조절하고 상향 조절된 14-3-3 제타/델타 단백질은 ATP 합성 효소를 형성한다(도 7). 이때 14-3-3 제타/델타 단백질이 관여하며 58번째 세린이 인산화됨에 따라 알파 및 베타 ATP 서브유닛을 수집한다(도 7). 이렇게 형성된 ATP 합성효소는 효율적인 에너지 생산을 가능하게 하고 미토콘드리아 기능 회복 및 노화 회복에 기여한다. 따라서 세포 수준에서 보았을 때 KB1541은 노화 회복에 효과적인 후보 물질로서 충분한 가치를 가지고 있다고 생각할 수 있다.
