Abstract
EBOVに対する新たなウイルス複製阻害剤を提供することである。 (a)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~337位からなるポリペプチド、(b)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質の1位~500位において20位~337位を含むポリペプチド、(c)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおいて配列番号1の20位~337位に相当する位置からなるポリペプチド、(d)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおける、配列番号1の1位~500位に相当する位置において、配列番号1の20位~337位に相当する位置を含むポリペプチドからなる群から選択されるいずれかのポリペプチド、又は(e)前記(a)~(d)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドからなるフィロウイルス科(Filoviridae)ウイルス複製阻害剤を提供する。
Description
本発明は、フィロウイルス科ウイルス複製阻害剤、フィロウイルス科ウイルス感染症治療用医薬組成物、フィロウイルス科ウイルスの複製阻害方法、及びフィロウイルス科ウイルスの複製活性の評価方法に関する。
エボラウイルス病(エボラ出血熱)は、エボラウイルス(Ebolavirus: EBOV)の感染による致死率の極めて高い急性熱性感染症である。1976年にスーダンにおいて最初の感染例(感染者数284、死亡者数151、致死率51%)が報告されて以来、主に中央アフリカを中心として、20回以上に及ぶアウトブレークを繰り返している。特に、過去最悪の感染事例となった2014年の西アフリカにおけるアウトブレークでは、感染者数27,550人、死亡者数11,235人(致死率41%)に達しただけでなく、アメリカ、スペイン、イギリス、イタリア等の欧米諸国にも感染が飛び火し、その猛威を目の当たりにしたことは記憶に新しい。
近年、アウトブレークの頻度が高くなる傾向が見られることから、世界規模での流行が危惧されている。また、その著しく高い致死率から、これがバイオテロ等に悪用される可能性を懸念する声も聞かれる。各方面での努力にもかかわらず、エボラ出血熱の決定的な予防及び治療法は、現段階では確立されておらず、対症療法に頼らざるを得ないのが現状である(非特許文献1)。
エボラ出血熱の新たな予防及び治療法を確立するために、EBOVに対する新たなウイルス複製阻害剤が必要とされている。

本発明の目的は、EBOVに対する新たなウイルス複製阻害剤を提供することである。
上記課題を解決するために、本発明者らは、EBOVの複製を阻害する新たな阻害剤の開発を試みた。
EBOVは、全長約18~19kbのマイナス鎖RNAをゲノムとするウイルスであり、ゲノム上には、NP(Nucleoprotein)、VP35(Viral protein 35)、VP40(Viral protein 40)、GP(Glycoprotein)、VP30(Viral protein 30)、VP24(Viral protein 24)、及びL(Large protein;RNA Polymerase)の7種類のORFが存在する(図1)。
EBOVは、全長約18~19kbのマイナス鎖RNAをゲノムとするウイルスであり、ゲノム上には、NP(Nucleoprotein)、VP35(Viral protein 35)、VP40(Viral protein 40)、GP(Glycoprotein)、VP30(Viral protein 30)、VP24(Viral protein 24)、及びL(Large protein;RNA Polymerase)の7種類のORFが存在する(図1)。
EBOVウイルス粒子内のゲノムRNAはNP、VP35、VP30、及びLと結合してリボヌクレオプロテイン(RNP)複合体として存在する(図2)。このうちNPはヌクレオカプシドの主成分であり、多量体を形成する。VP30は複製に必須の働きをする多機能タンパク質であり、六量体を形成する。各々の多量体がゲノムRNAやVP35及びLと相互作用することによりRNPが形成される。RNPは、ウイルスタンパク質の発現とゲノムの複製の双方に重要であり、したがってNP及びVP30の多量体形成及び相互作用は、ウイルス複製の重要な初期のステップと言える。
本発明者らは、EBOVのNP及びVP30の創薬標的としての可能性に着目し、鋭意研究を行った。その結果、EBOVの複製を阻害し得る欠失変異型NP及び欠失変異型VP30を見出し、本発明を完成させるに至った。本発明は、上記知見に基づくものであって以下を提供する。
(1)以下の(a)~(e)からなる群から選択されるいずれかのポリペプチドからなるフィロウイルス科(Filoviridae)ウイルス複製阻害剤。

1.フィロウイルス科ウイルス複製阻害剤
1-1.概要
本発明の第1の態様は、フィロウイルス科ウイルス複製阻害剤である。本発明のフィロウイルス科ウイルス複製阻害剤は、フィロウイルス科ウイルスのNPタンパク質若しくはVP30タンパク質に由来するペプチド断片、そのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、又は細胞内でそのいずれかを発現可能な発現ベクターからなる。本発明のフィロウイルス科ウイルス複製阻害剤は、細胞内においてフィロウイルス科ウイルスに対する増殖抑制効果の高いフィロウイルス科ウイルス感染症治療用医薬組成物の有効成分となり得る。
1-1.概要
本発明の第1の態様は、フィロウイルス科ウイルス複製阻害剤である。本発明のフィロウイルス科ウイルス複製阻害剤は、フィロウイルス科ウイルスのNPタンパク質若しくはVP30タンパク質に由来するペプチド断片、そのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、又は細胞内でそのいずれかを発現可能な発現ベクターからなる。本発明のフィロウイルス科ウイルス複製阻害剤は、細胞内においてフィロウイルス科ウイルスに対する増殖抑制効果の高いフィロウイルス科ウイルス感染症治療用医薬組成物の有効成分となり得る。
1-2.用語の定義
本明細書で使用する用語について、以下で定義する。
「フィロウイルス科(Filoviridae)」は、モノネガウイルス目(Mononegavirales)に属し、マイナス鎖RNAゲノムを有するウイルスである。フィロウイルス科ウイルスには、エボラウイルス属(Ebolavirus)、マールブルグウイルス属(Marburgvirus)、クウェバウイルス属(Cuevavirus)が知られている。フィロウイルス科ウイルスの宿主には、ヒト、ゴリラ、チンパンジー、サル、シカ、及びコウモリ等の哺乳動物が含まれる。
本明細書で使用する用語について、以下で定義する。

「エボラウイルス属(Ebolavirus; EBOV)」は、全長約18~19kbのマイナス鎖RNAをゲノムとするウイルスであり、エボラ出血熱の病原体として知られる。EBOVの感染機序には不明な点が多いが、動物体内ではマクロファージや樹状細胞等の貪食性免疫細胞に感染すると考えられている。EBOVゲノム上には、NP(Nucleoprotein)、VP35(Viral protein 35)、VP40(Viral protein 40)、GP(Glycoprotein)、VP30(Viral protein 30)、VP24(Viral protein 24)、及びL(Large protein;RNA Polymerase)の7種類のORFが存在する。EBOVゲノムの5'及び3'末端領域には、非翻訳領域であるTrailer及びLeaderが存在する。エボラウイルス属に属するウイルスには、ザイールエボラウイルス(Zaire ebolavirus;ZEBOV)、スーダンエボラウイルス(Sudan ebolavirus;SUDV;SEBOV)、レストンエボラウイルス(Reston ebolavirus;RESTV)、タイフォレストウイルス(Tai Forest ebolavirus;TAFV)、ブンディブギョエボラウイルス(Bundibugyo ebolavirus;BUBV)等が知られている。
「マールブルグウイルス属(Marburgvirus)」は、全長約19kbのマイナスRNAをゲノムとするウイルスであり、マールブルグ出血熱の病原体として知られる。マールブルグウイルス属ウイルスのゲノム上には、エボラウイルス属と同様にNP、VP35、VP40、GP、VP30、VP24、及びLの7種類のORFが存在する。マールブルグウイルス属に属するウイルスには、マールブルグウイルス(Marburg marburgvirus;MARV)が知られている。
「クウェバウイルス属(Cuevavirus)」に属するウイルスには、コウモリから見つかったLloviu cuevavirus(LLOV)が知られている。クウェバウイルス属ウイルスのゲノム上には、エボラウイルス属と同様にNP、VP35、VP40、GP、VP30、VP24、及びLの7種類のORFが存在する。
「NP(Nucleoprotein)」は、フィロウイルス科ウイルスのヌクレオカプシドの主成分であり、多量体を形成する(図2)。NPは、N末端のコアドメイン、C末端のCテール、及びその間の非保存領域を含む(図5A及び図5B)。NPのコアドメインはNローブとCローブを含み(図5A)、NローブとCローブの間にはRNA結合グローブが形成される(図5B)。NPのコアドメインには13個のαヘリックスが含まれる。本明細書において、フィロウイルス科ウイルスのNPは、配列番号1で示すアミノ酸配列からなるZEBOVのNPタンパク質、及びそのフィロウイルス科オルソログである。NPの例として、配列番号1で示すアミノ酸配列からなるZEBOVのNP、配列番号6で示すアミノ酸配列からなるSUDVのNP、配列番号7で示すアミノ酸配列からなるRESTVのNP、配列番号8で示すアミノ酸配列からなるTAFVのNP、配列番号9で示すアミノ酸配列からなるBUBVのNP、配列番号10で示すアミノ酸配列からなるMARVのNP、配列番号11で示すアミノ酸配列からなるLLOVのNPが挙げられる。NPタンパク質のアミノ酸配列のアラインメントを図18-1~図18-3に示す。またNPをコードする塩基配列の例として、ZEBOVのNP遺伝子(配列番号2)及びそのコドン最適化配列(配列番号18)が挙げられる。
「VP30(Viral protein 30)」は、フィロウイルス科ウイルスの複製に必須の働きをする多機能タンパク質であり、六量体を形成する(図2)。VP30は、N末端ドメイン(NTD)及びC末端ドメイン(CTD)の2つのドメイン構造を有する(図10)。NTDには、RNA結合領域、Znフィンガーモチーフ、及び六量体化モチーフが存在する。CTDには7個のαヘリックスが含まれており、二量体化及びNP結合領域が含まれる。本明細書において、フィロウイルス科ウイルスのVP30は、配列番号3で示すアミノ酸配列からなるZEBOVのVP30タンパク質、及びそのフィロウイルス科オルソログである。VP30の例として、配列番号3で示すアミノ酸配列からなるZEBOVのVP30、配列番号12で示すアミノ酸配列からなるSUDVのVP30、配列番号13で示すアミノ酸配列からなるRESTVのVP30、配列番号14で示すアミノ酸配列からなるTAFVのVP30、配列番号15で示すアミノ酸配列からなるBUBVのVP30、配列番号16で示すアミノ酸配列からなるMARVのVP30、配列番号17で示すアミノ酸配列からなるLLOVのVP30が挙げられる。VP30タンパク質のアミノ酸配列のアラインメントを図19-1~図19-2に示す。またVP30をコードする塩基配列の例として、ZEBOVのVP30遺伝子の塩基配列(配列番号4)及びそのコドン最適化塩基配列(配列番号19)が挙げられる。
本明細書において「発現ベクター」とは、遺伝子や遺伝子断片(以下「遺伝子等」と表記する)を発現可能な状態で含み、その遺伝子等の発現を制御できる発現単位を包含するベクターをいう。本明細書において「発現可能な状態」とは、プロモーターの制御下にあるプロモーター下流域に、発現すべき遺伝子等を配置していることをいう。ベクターには、プラスミドベクター、ウイルスベクター等が知られるが、いずれのベクターも利用することができる。通常は、遺伝子組換え操作の容易なプラスミドベクターでよい。発現ベクターは、市販の哺乳動物細胞用発現ベクターを利用してもよい。例えば、Promega社のpCIベクターやpSIベクター等が挙げられる。また、発現ベクターは、哺乳動物細胞と大腸菌等の細菌間とで複製可能なシャトルベクターであってもよい。
本明細書において「プロモーター」とは、発現ベクターを導入した細胞において、下流(3’末端側)に配置された遺伝子等の発現を制御することのできる遺伝子発現調節領域である。プロモーターは、発現制御下にある遺伝子等を発現させる場所に基づいて、ユビキタスプロモーター(全身性プロモーター)と部位特異的プロモーターに分類することができる。ユビキタスプロモーターは、全細胞、すなわち宿主個体全体で対象とする遺伝子等(対象遺伝子等)の発現を制御するプロモーターである。また、部位特異的プロモーターは、特定の細胞又は組織でのみ対象遺伝子等の発現を制御するプロモーターである。
また、プロモーターは、発現の時期に基づいて構成的活性型プロモーター、発現誘導型プロモーター又は時期特異的活性型プロモーターに分類される。構成的活性型プロモーターは、細胞内で対象遺伝子等を恒常的に発現させることができる。発現誘導型プロモーターは、細胞内で対象遺伝子等の発現を任意の時期に誘導することができる。また、時期特異的活性型プロモーターは、細胞内で対象遺伝子等を発生段階の特定の時期にのみに発現誘導することができる。いずれのプロモーターも、宿主細胞内で対象遺伝子の過剰な発現をもたらし得ることから過剰発現型プロモーターと解することができる。
本明細書において「治療」とは、疾患の罹患に伴う症状の緩和又は除去、及び/又は疾患の進行の阻止又は抑制、並びに疾患の治癒をいう。本明細書において「疾患」とは、断りのない限り、フィロウイルス科ウイルス感染症を意味する。
本明細書において「フィロウイルス科ウイルス感染症」とは、フィルウイルス科ウイルスによる感染症をいう。例えば、エボラ出血熱やマールブルグ出血熱が挙げられる。
1-3.構成
本発明のフィロウイルス科ウイルス複製阻害剤は、ペプチド断片(ポリペプチド)又は発現ベクターからなる。それぞれの構成を以下で具体的に説明する。

1-3-1.ペプチド断片(ポリペプチド)
本発明のフィロウイルス科ウイルス複製阻害剤を構成するペプチド断片(ポリペプチド)は、フィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片、若しくはそのN末端及び/若しくはC末端に任意のアミノ酸配列が付加されたポリペプチド、又はフィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片、若しくはそのN末端及び/若しくはC末端に任意のアミノ酸配列が付加されたポリペプチドである。
本発明のフィロウイルス科ウイルス複製阻害剤を構成するペプチド断片(ポリペプチド)は、フィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片、若しくはそのN末端及び/若しくはC末端に任意のアミノ酸配列が付加されたポリペプチド、又はフィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片、若しくはそのN末端及び/若しくはC末端に任意のアミノ酸配列が付加されたポリペプチドである。
(1)NPタンパク質に由来するペプチド断片、又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド
一態様において、本発明のフィロウイルス科ウイルス複製阻害剤は、フィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片、又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドからなる。フィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片は、ザイールエボラウイルスのNPタンパク質、又はザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログのいずれかに由来するペプチド断片であって、細胞内においてフィロウイルス科ウイルスの複製を阻害することができるペプチド断片である。
一態様において、本発明のフィロウイルス科ウイルス複製阻害剤は、フィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片、又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドからなる。フィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片は、ザイールエボラウイルスのNPタンパク質、又はザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログのいずれかに由来するペプチド断片であって、細胞内においてフィロウイルス科ウイルスの複製を阻害することができるペプチド断片である。
本発明のフィロウイルス科ウイルス複製阻害剤を構成するフィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片の具体例としては、(a)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~337位からなるポリペプチド、(b)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質の1位~500位において20位~337位を含むポリペプチド、(c)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおいて配列番号1の20位~337位に相当する位置からなるポリペプチド、又は(d)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおける、配列番号1の1位~500位に相当する位置において、配列番号1の20位~337位に相当する位置を含むポリペプチドが挙げられる。(a)~(d)について以下でより具体的に説明する。
「(a)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~337位からなるポリペプチド」は、NPタンパク質のコアドメインに含まれる13個のαヘリックスのうち、N末端側に位置する12個のαへリックスを含むポリペプチドであり、本明細書の実施例においてEBOV複製を阻害する活性を有することが見出されたΔNP(20-337)に対応する。
「(b)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質の1位~500位において20位~337位を含むポリペプチド」とは、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質の1位~500位に含まれるペプチド断片であって、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質の20位~337位を含むペプチド断片である。すなわち、当該ペプチド断片のN末端に位置するアミノ酸残基は、配列番号1で示すアミノ酸配列における1~20位のいずれかに位置し、当該ペプチド断片のC末端に位置するアミノ酸残基は、配列番号1で示すアミノ酸配列における337~500位のいずれかに位置する。上記(b)のポリペプチドの具体例としては、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~337位、15位~337位、10位~337位、5位~337位、1位~337位、20位~350位、20位~375位、20位~400位、20位~425位、20位~450位、20位~475位、又は20位~500位からなるポリペプチド、例えば、本明細書の実施例においてEBOV複製を阻害する活性を有することが見出された、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~405位からなるポリペプチドが挙げられる。
一実施形態において、上記(b)のポリペプチドは、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において1位~337位を含む。そのようなポリペプチドの例としては、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において1位~337位、1位~350位、1位~360位、1位~370位、1位~380位、1位~390位、1位~400位、1位~410位、1位~420位、1位~430位、1位~440位、1位~450位、1位~460位、1位~470位、1位~480位、1位~490位、又は1位~500位からなるポリペプチド、例えば、本明細書の実施例においてEBOV複製を阻害する活性を有することが見出された、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において1位~405位からなるポリペプチドが挙げられる。
一実施形態において、上記(b)のポリペプチドは、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~500位を含む。そのようなポリペプチドの例としては、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~500位、18位~500位、16位~500位、14位~500位、12位~500位、10位~500位、8位~500位、6位~500位、4位~500位、2位~500位、又は1位~500位からなるポリペプチドが挙げられる。

「そのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド」とは、前記(a)~(d)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドである。任意のアミノ酸配列として、前記(a)~(d)のいずれかに記載のポリペプチドとは異なる任意のアミノ酸配列を用いることができる。特に限定しないが、例えば、ユビキチン化配列、核移行シグナル、タグ配列や、後述の標識タンパク質等が挙げられる。付加されるアミノ酸数は限定しないが、例えば、それぞれ、20個、19個、18個、17個、16個、15個、14個、13個、12個、11個、10個、9個、8個、7個、6個、5個、4個、3個、2個、又は1個であってもよい。一実施形態では、前記(a)~(d)のいずれかに記載のポリペプチドのN末端にMet残基を付加することができる。
一実施形態において、上記(d)のポリペプチドは、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおいて配列番号1の1位~337位に相当する位置を含む。
一実施形態において、上記(d)のポリペプチドは、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおいて配列番号1の20位~500位に相当する位置を含む。
一実施形態において、フィロウイルス科オルソログは、エボラウイルス属オルソログであってもよい。
(2)VP30タンパク質に由来するペプチド断片、又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド
一態様において、本発明のフィロウイルス科ウイルス複製阻害剤は、フィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片、又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドからなる。フィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片は、ザイールエボラウイルスのVP30タンパク質、又はザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログのいずれかに由来するペプチド断片であって、細胞内においてフィロウイルス科ウイルスの複製を阻害することができるペプチド断片である。
一態様において、本発明のフィロウイルス科ウイルス複製阻害剤は、フィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片、又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドからなる。フィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片は、ザイールエボラウイルスのVP30タンパク質、又はザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログのいずれかに由来するペプチド断片であって、細胞内においてフィロウイルス科ウイルスの複製を阻害することができるペプチド断片である。
本発明のフィロウイルス科ウイルス複製阻害剤を構成するフィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片の具体例としては、(f)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~254位からなるポリペプチド、(g)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質の47位~288位において91位~254位を含むポリペプチド、(h)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の91位~254位に相当する位置からなるポリペプチド、又は(i)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおける、配列番号3の47位~288位に相当する位置において、配列番号3の91位~254位に相当する位置を含むポリペプチドが挙げられる。
「(f)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~254位からなるポリペプチド」は、VP30タンパク質の六量体化モチーフ、並びにC末端ドメイン(CTD)に含まれる7個のαヘリックスのうち、N末端側に位置する6個のαへリックスを含むポリペプチドであり、本明細書の実施例においてEBOV複製を阻害する活性を有することが見出されたΔVP30(91-254)に対応する。
「(g)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質の47位~288位において91位~254位を含むポリペプチド」とは、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質の47位~288位に含まれるペプチド断片であって、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質の91位~254位を含むペプチド断片である。すなわち、当該ペプチド断片のN末端に位置するアミノ酸残基は、配列番号3で示すアミノ酸配列における47~91位のいずれかに位置し、当該ペプチド断片のC末端に位置するアミノ酸残基は、配列番号3で示すアミノ酸配列における254~288位のいずれかに位置する。上記(g)のポリペプチドの具体例としては、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~254位、80位~254位、70位~254位、60位~254位、50位~254位、47位~254位、91位~260位、91位~265位、91位~270位、91位~275位、91位~280位、91位~285位、又は91位~288位からなるポリペプチド、例えば、本明細書の実施例においてEBOV複製を阻害する活性を有することが見出された、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~254位からなるポリペプチドが挙げられる。
一実施形態において、上記(g)のポリペプチドは、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において47位~254位を含む。そのようなポリペプチドの例として、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において47位~254位、47位~260位、47位~265位、47位~270位、47位~275位、47位~280位、47位~285位、又は47位~288位からなるポリペプチド、例えば、本明細書の実施例においてEBOV複製を阻害する活性を有することが見出された、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において、91位~263位からなるポリペプチド、91位~272位からなるポリペプチド、及び91位~288位からなるポリペプチドが挙げられる。
一実施形態において、上記(g)のポリペプチドは、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~288位を含む。例えば、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~288位、90位~288位、85位~288位、80位~288位、75位~288位、70位~288位、65位~288位、60位~288位、55位~288位、50位~288位、又は47位~288位からなるポリペプチド、例えば、本明細書の実施例においてEBOV複製を阻害する活性を有することが見出された、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において、47位~288位からなるポリペプチド、及び91位~288位からなるポリペプチドが挙げられる。
「(h)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の91位~254位に相当する位置からなるポリペプチド」とは、フィロウイルス科に属する任意のウイルスにおけるVP30オルソログのアミノ酸配列を配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のアミノ酸配列に対して整列(アラインメント)し、必要に応じてギャップを導入して、両者のアミノ酸一致度が最も高くなるようにしたときに、配列番号3の91位~254位に相当する位置からなるポリペプチドである。当業者であればそのようなアラインメントは容易に実施することが可能であり、フィロウイルス科に属する任意のウイルスにおけるVP30オルソログのアミノ酸配列において、配列番号3の91位~254位に相当する位置を容易に決定することができる。配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の91位~254位に相当する位置からなるポリペプチドの例としては、図19-1~図19-2に示すように、配列番号12で示すアミノ酸配列からなるSUDVのVP30タンパク質の91位~254位からなるポリペプチド;配列番号13で示すアミノ酸配列からなるRESTVのVP30タンパク質の91位~254位からなるポリペプチド;配列番号14で示すアミノ酸配列からなるTAFVのVP30タンパク質の91位~254位からなるポリペプチド;配列番号15で示すアミノ酸配列からなるBUBVのVP30タンパク質の91位~254位からなるポリペプチド;配列番号16で示すアミノ酸配列からなるMARVのVP30タンパク質の97位~261位からなるポリペプチド;及び配列番号17で示すアミノ酸配列からなるLLOVのVP30タンパク質の133位~296位からなるポリペプチドが挙げられる。

「そのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド」とは、前記(f)~(i)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドである。ここで任意のアミノ酸配列には、前記(f)~(i)のいずれかに記載のポリペプチドとは異なる任意のアミノ酸配列を用いることができる。特に限定しないが、例えば、ユビキチン化配列、核移行シグナル、タグ配列や、後述の標識タンパク質等が挙げられる。付加されるアミノ酸数は限定しないが、例えば、それぞれ、20個、19個、18個、17個、16個、15個、14個、13個、12個、11個、10個、9個、8個、7個、6個、5個、4個、3個、2個、又は1個であってもよい。一実施形態では、前記(f)~(i)のいずれかに記載のポリペプチドのN末端にMet残基を付加することができる。
一実施形態において、上記(i)に記載のポリペプチドは、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の47位~254位に相当する位置を含む。
一実施形態において、上記(i)に記載のポリペプチドは、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の91位~288位に相当する位置を含む。
一実施形態において、フィロウイルス科オルソログは、エボラウイルス属オルソログであってもよい。
1-3-2.発現ベクター
本発明のフィロウイルス科ウイルス複製阻害剤を構成する「発現ベクター」は、フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片又はそのいずれかのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドをコードする核酸、及びプロモーターを含み、細胞内で前記ペプチド断片又は前記ポリペプチドを発現可能な発現ベクターである。発現ベクターは、前記構成要素である核酸及びプロモーターに加えて、必要に応じて、標識遺伝子(選抜マーカー)、エンハンサー、ターミネーター、複製起点、及びポリAシグナル等の構成要素を含んでいてもよい。発現ベクターは、プラスミドベクターやウイルスベクターであってもよい。以下、各構成要素について説明をする。
本発明のフィロウイルス科ウイルス複製阻害剤を構成する「発現ベクター」は、フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片又はそのいずれかのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドをコードする核酸、及びプロモーターを含み、細胞内で前記ペプチド断片又は前記ポリペプチドを発現可能な発現ベクターである。発現ベクターは、前記構成要素である核酸及びプロモーターに加えて、必要に応じて、標識遺伝子(選抜マーカー)、エンハンサー、ターミネーター、複製起点、及びポリAシグナル等の構成要素を含んでいてもよい。発現ベクターは、プラスミドベクターやウイルスベクターであってもよい。以下、各構成要素について説明をする。
(核酸)
「フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片又はそのいずれかのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドをコードする核酸」は、「1-3-1.ペプチド断片(ポリペプチド)」に記載のいずれかのペプチド断片又はポリペプチドをコードする核酸であればよい。そのような核酸の塩基配列は、限定はしない。フィロウイルス科ウイルスのNPタンパク質に由来するペプチド断片又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドをコードする核酸の例としては、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~337位からなるポリペプチドをコードする核酸(例えば、配列番号2で示す塩基配列からなるザイールエボラウイルスのNP遺伝子において58位~1011位からなる核酸)、配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~405位からなるポリペプチドをコードする核酸(例えば、配列番号2で示す塩基配列からなるザイールエボラウイルスのNP遺伝子において58位~1215位からなる核酸)、及び配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において1位~405位からなるポリペプチドをコードする核酸(例えば、配列番号2で示す塩基配列からなるザイールエボラウイルスのNP遺伝子において1位~1215位からなる核酸)が挙げられ、そのいずれかの5’末端側に開始コドン(ATG)が付加された塩基配列もまた例示される。フィロウイルス科ウイルスのVP30タンパク質に由来するペプチド断片又はそのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチドをコードする核酸の例としては、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~254位からなるポリペプチドをコードする核酸(例えば、配列番号4で示す塩基配列からなるザイールエボラウイルスのVP30遺伝子において271位~762位からなる核酸)、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~263位からなるポリペプチドをコードする核酸(例えば、配列番号4で示す塩基配列からなるザイールエボラウイルスのVP30遺伝子において271位~789位からなる核酸)、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~272位からなるポリペプチドをコードする核酸(例えば、配列番号4で示す塩基配列からなるザイールエボラウイルスのVP30遺伝子において271位~816位からなる核酸)、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~288位からなるポリペプチドをコードする核酸(例えば、配列番号4で示す塩基配列からなるザイールエボラウイルスのVP30遺伝子において271位~864位からなる核酸)、及び配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において47位~288位からなるポリペプチドをコードする核酸(例えば、配列番号4で示す塩基配列からなるザイールエボラウイルスのVP30遺伝子において139位~864位からなる核酸)が挙げられ、そのいずれかの5’末端側に開始コドン(ATG)が付加された塩基配列もまた例示される。

(プロモーター)
本明細書においてプロモーターは、ペプチド断片をコードする核酸を細胞内で発現誘導できるプロモーターである。本発明のフィロウイルス科ウイルス複製阻害剤を適用する、すなわち発現ベクターを導入する標的細胞は、原則として哺乳動物細胞、特にヒト又はチンパンジー由来の細胞であることから、それらの細胞内で下流の遺伝子を発現できるプロモーターであればよい。例えば、CMVプロモーター(CMV-IEプロモーター)、SV40初期プロモーター、RSVプロモーター、EF1αプロモーター、Ubプロモーター等が挙げられる。

(標識遺伝子)
本明細書において「標識遺伝子」は、選抜マーカー又はレポータータンパク質とも呼ばれる標識タンパク質をコードする遺伝子である。「標識タンパク質」とは、その活性に基づいて標識遺伝子の発現の有無を判別することのできるペプチドをいう。活性の検出は、標識タンパク質の活性そのものを直接的に検出するものであってもよいし、色素のような標識タンパク質の活性によって発生する代謝物を介して間接的に検出するものであってもよい。検出は、生物学的検出(抗体、アプタマー等のペプチドや核酸の結合による検出を含む)、化学的検出(酵素反応的検出を含む)、物理的検出(行動分析的検出を含む)、又は検出者の感覚的検出(視覚、触覚、嗅覚、聴覚、味覚による検出を含む)のいずれであってもよい。

標識遺伝子がコードする標識タンパク質の種類は、当該分野で公知の方法によりその活性を検出可能な限り、特に限定はしない。検出に際して形質転換体に対する侵襲性の低い標識タンパク質は好ましい。例えば、タグペプチド、薬剤耐性タンパク質、色素タンパク質、蛍光タンパク質、発光タンパク質等が挙げられる。
「タグペプチド」は、タンパク質を標識化することのできる十数アミノ酸~数十アミノ酸からなる短ペプチドであって、タンパク質の検出用、精製用として用いられる。通常は、標識すべきタンパク質をコードする遺伝子の5’末端側又は3’末端側にタグペプチドをコードする塩基配列を連結し、タグペプチドとの融合タンパク質として発現させることで標識化する。タグペプチドは、当該分野で様々な種類が開発されているが、いずれのタグペプチドを使用してもよい。タグペプチドの具体例として、FLAG、HA、His、PA、及びmyc等が挙げられる。
「薬剤耐性タンパク質」は、培地等に添加された抗生物質等の薬剤に対する抵抗性を細胞に付与するタンパク質であり、多くは酵素である。例えば、アンピシリンに対して抵抗性を付与するβラクタマーゼ、カナマイシンに対して抵抗性を付与するアミノグリコシド3’ホスホトランスフェラーゼ、テトラサイクリンに対して抵抗性を付与するテトラサイクリン排出トランスポーター、クロラムフェニコールに対して抵抗性を付与するCAT(クロラムフェニコールアセチルトランスフェラーゼ)等が挙げられる。
「色素タンパク質」は、色素の生合成に関与するタンパク質、又は基質の付与により色素による形質転換体の化学的検出を可能にするタンパク質であり、通常は酵素である。ここでいう「色素」とは、形質転換体に色素を付与することができる低分子化合物又はペプチドで、その種類は問わない。例えば、β-ガラクトシダーゼ(LacZ)、β-グルクロニターゼ(GUS)、メラニン系色素合成タンパク質、オモクローム系色素、又はプテリジン系色素が挙げられる。
「蛍光タンパク質」は、特定波長の励起光を照射したときに特定波長の蛍光を発するタンパク質をいう。天然型及び非天然型のいずれであってもよい。また、励起波長、蛍光波長も特に限定はしない。具体的には、例えば、CFP、RFP、DsRed(3xP3-DsRedのような派生物を含む)、YFP、PE、PerCP、APC、GFP(EGFP、3xP3-EGFP等の派生物を含む)等が挙げられる。
「発光タンパク質」とは、励起光を必要とすることなく発光することのできる基質タンパク質又はその基質タンパク質の発光を触媒する酵素をいう。例えば、基質タンパク質としてのルシフェリン又はイクオリン、酵素としてのルシフェラーゼが挙げられる。
(エンハンサー)
本明細書において「エンハンサー」は、ベクター内の遺伝子又はその断片の発現効率を増強できるものであれば特に限定はされない。

本発明における発現ベクターは、フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片等を哺乳動物細胞内で一過的に発現することができればよい。したがって、哺乳動物細胞用の複製起点は不要である。しかし、シャトルベクターとして、例えば、大腸菌等の細菌内で発現させる場合には、その複製起点が必須となる。複製起点は、公知の配列を利用することができる。例えば、大腸菌用の複製起点であればf1 origin等を利用すればよい。

本発明の第2の態様は、フィロウイルス科ウイルス感染症治療用医薬組成物である。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、第1態様のフィロウイルス科ウイルス複製阻害剤を有効成分とし、フィロウイルス科ウイルスの複製を阻害することでフィロウイルス科ウイルスの増殖を抑制し、フィロウイルス科ウイルス感染症を治療することができる。
2-1.概要
本発明の第2の態様は、フィロウイルス科ウイルス感染症治療用医薬組成物である。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、第1態様のフィロウイルス科ウイルス複製阻害剤を有効成分とし、フィロウイルス科ウイルスの複製を阻害することでフィロウイルス科ウイルスの増殖を抑制し、フィロウイルス科ウイルス感染症を治療することができる。
2-2.構成
2-2-1.構成成分
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物の構成成分について説明をする。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、必須の構成成分として一以上の有効成分、及び溶媒及び/又は担体、さらに薬剤送達系(DDS;Drug Delivery System)粒子を含む。以下、各構成成分について具体的に説明をする。
2-2-1.構成成分
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物の構成成分について説明をする。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、必須の構成成分として一以上の有効成分、及び溶媒及び/又は担体、さらに薬剤送達系(DDS;Drug Delivery System)粒子を含む。以下、各構成成分について具体的に説明をする。
(1)有効成分
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、必須の有効成分として第1態様に記載のフィロウイルス科ウイルス複製阻害剤を包含する。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、1種又は複数種のフィロウイルス科ウイルス複製阻害剤を含むことができる。
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、必須の有効成分として第1態様に記載のフィロウイルス科ウイルス複製阻害剤を包含する。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、1種又は複数種のフィロウイルス科ウイルス複製阻害剤を含むことができる。
一実施形態において、本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、第1態様に記載のNPタンパク質に由来するペプチド断片、若しくはそのN末端及び/若しくはC末端に任意のアミノ酸配列が付加されたポリペプチド、並びに/又はVP30タンパク質に由来するペプチド断片、若しくはそのN末端及び/若しくはC末端に任意のアミノ酸配列が付加されたポリペプチドを含んでもよい。
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物に含まれる有効成分の含有量は、特に限定はしない。一般に含有量は、有効成分の種類、剤形、並びに後述する他の構成成分である溶媒や担体の種類によって異なる。したがって、それぞれの条件を勘案して適宜定めればよい。単回適用量のフィロウイルス科ウイルス感染症治療用医薬組成物に有効量の有効成分が含有されていればよい。ただし、有効成分の薬理効果を得る上で被験体にフィロウイルス科ウイルス感染症治療用医薬組成物を大量に投与する必要がある場合には、被験体の負担軽減のために数回に分割して投与することもできる。この場合、有効成分の量は、総合量で有効量を含んでいればよい。「有効量」とは、有効成分としての機能を発揮する上で必要な量であって、かつそれを適用する被験体に対して有害な副作用をほとんど又は全く付与しない量をいう。この有効量は、被験体の情報、適用経路、及び適用回数等の様々な条件によって変わり得る。したがって、フィロウイルス科ウイルス感染症治療用医薬組成物を医薬として使用する場合、有効成分の含有量は、最終的には、医師又は薬剤師等の判断によって決定される。
本明細書において「被験体」とは、第1態様に記載のフィロウイルス科ウイルス複製阻害剤、又は本態様のフィロウイルス科ウイルス感染症治療用医薬組成物の適用対象物をいう。例えば、細胞(培養細胞を含む)、組織、器官、又は個体である。個体の場合、好ましくはヒト個体であり、この場合、特に「被験者」と表記する。フィロウイルス科ウイルスに感染した被験者、例えばエボラウイルス感染者は特に好ましい。
本明細書において、「被験体の情報」とは、被験体の特徴や状態に関する様々な情報である。例えば、被験体がヒト個体の場合には、年齢、体重、性別、全身の健康状態、疾患の有無、疾患の進行度や重症度、薬剤感受性、併用薬物の有無及び治療に対する耐性等が挙げられる。
(2)溶媒
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、必要に応じて薬学的に許容可能な溶媒を含むことができる。「薬学的に許容可能な溶媒」とは、製剤技術分野において通常使用する溶媒をいう。例えば、水若しくは水溶液、又は有機溶剤が挙げられる。水溶液には、例えば、生理食塩水、ブドウ糖又はその他の補助剤を含む等張液、リン酸塩緩衝液、酢酸ナトリウム緩衝液が挙げられる。補助剤には、例えば、D-ソルビトール、D-マンノース、D-マンニトール、塩化ナトリウム、その他にも低濃度の非イオン性界面活性剤、ポリオキシエチレンソルビタン脂肪酸エステル類等が挙げられる。有機溶剤には、エタノールが挙げられる。

本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、必要に応じて薬学的に許容可能な担体を含むことができる。「薬学的に許容可能な担体」とは、製剤技術分野において通常使用する添加剤をいう。例えば、賦形剤、結合剤、崩壊剤、充填剤、乳化剤、流動添加調節剤、滑沢剤、ヒト血清アルブミン等が挙げられる。
賦形剤には、例えば、単糖、二糖類、シクロデキストリン及び多糖類のような糖、金属塩、クエン酸、酒石酸、グリシン、ポリエチレングリコール、プルロニック、カオリン、ケイ酸、又はそれらの組み合わせが挙げられる。
結合剤には、例えば、植物デンプンを用いたデンプン糊、ペクチン、キサンタンガム、単シロップ、グルコース液、ゼラチン、トラガカント、メチルセルロース、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロースナトリウム、セラック、パラフィン、ポリビニルピロリドン又はそれらの組み合わせが挙げられる。
崩壊剤としては、例えば、前記デンプンや、乳糖、カルボキシメチルデンプン、架橋ポリビニルピロリドン、アガー、ラミナラン末、炭酸水素ナトリウム、炭酸カルシウム、アルギン酸若しくはアルギン酸ナトリウム、ポリオキシエチレンソルビタン脂肪酸エステル、ラウリル硫酸ナトリウム、ステアリン酸モノグリセリド又はそれらの塩が挙げられる。
充填剤としては、ワセリン、前記糖及び/又はリン酸カルシウムが例として挙げられる。
乳化剤としては、ソルビタン脂肪酸エステル、グリセリン脂肪酸エステル、ショ糖脂肪酸エステル、プロピレングリコール脂肪酸エステルが例として挙げられる。
流動添加調節剤及び滑沢剤としては、ケイ酸塩、タルク、ステアリン酸塩又はポリエチレングリコールが例として挙げられる。
上記の他にも、必要であれば医薬組成物等において通常用いられる可溶化剤、懸濁剤、希釈剤、分散剤、界面活性剤、無痛化剤、安定剤、吸収促進剤、増量剤、付湿剤、保湿剤、湿潤剤、吸着剤、矯味矯臭剤、崩壊抑制剤、コーティング剤、着色剤、保存剤、防腐剤、抗酸化剤、香料、風味剤、甘味剤、緩衝剤、等張化剤等を適宜含むこともできる。
担体は、被験体内で酵素等による前記有効成分の分解を回避又は抑制する他、製剤化や投与方法を容易にし、剤形及び薬効を維持するために用いられるものであり、必要に応じて適宜使用すればよい。
(4)薬剤送達系粒子(DDS粒子)
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物は、必要に応じてDDS粒子を含むことができる。DDS粒子は、その内部等に有効成分や他の担体等を包含して、標的部位にまで内容物、特に有効成分を分解させることなく送達し、また生体内での薬物分布を時間的に、量的に制御し得る粒子をいう。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物の有効成分はペプチド又は核酸であることから、投与後に生体内でプロテアーゼやヌクレアーゼによる分解から保護するためにも、DDS粒子の使用は好適である。DDS粒子の種類は問わない。例えば、リポソーム、高分子ミセル、ウイルス粒子等が挙げられる。

2-2-2.剤形
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物の剤形は、特に限定しない。被験体の体内で有効成分を失活させることなく目的の部位にまで送達される形態であればよい。

具体的な剤形は、後述する適用方法によって異なる。適用方法は、非経口投与と経口投与に大別することができるので、それぞれの投与法に適した剤形にすればよい。
例えば、投与方法が非経口投与であれば、好ましい剤形は、対象部位への直接投与又は循環系を介した全身投与が可能な液剤である。液剤の好例としては、注射剤が挙げられる。注射剤は、溶媒の他、前記賦形剤、乳化剤、懸濁剤、界面活性剤、安定剤、pH調節剤等と適宜組み合わせて、一般に認められた製薬実施に要求される単位用量形態で混和することによって製剤化することができる。
投与方法が経口投与であれば、好ましい剤形は、固形剤(錠剤、カプセル剤、ドロップ剤、トローチ剤を含む)、顆粒剤、粉剤、散剤、液剤(内用水剤、乳剤、シロップ剤を含む)が挙げられる。固形剤であれば、必要に応じて、当該技術分野で公知の剤皮を施した剤形、例えば、糖衣錠、ゼラチン被包錠、腸溶錠、フィルムコーティング錠、二重錠、多層錠にすることができる。
なお、上記各剤形の具体的な形状、大きさについては、いずれもそれぞれの剤形において当該分野で公知の剤形の範囲内にあればよく、特に限定はしない。本発明のB型肝炎治療用医薬組成物の製造方法については、当該技術分野の常法に従って製剤化すればよい。
2-3.適用方法
本発明のフィロウイルス科ウイルス感染症治療用医薬組成物の適用方法は、経口投与でも、非経口投与でもよい。一般に経口投与法は全身投与となるが、非経口投与法は、さらに全身投与と局所投与に細分できる。局所投与には、例えば、筋肉内投与、皮下投与、組織投与、及び器官投与が該当し、非経口投与法の全身投与には、循環器内投与、例えば、静脈内投与(静注)、動脈内投与及びリンパ管内投与が挙げられる。本発明のフィロウイルス科ウイルス感染症治療用医薬組成物を局所投与する場合には、注射等で局所感染部位に直接投与すればよい。また、全身投与する場合には、静注等により循環器内に投与すればよい。投与量は、有効成分が奏効する上で有効な量であればよい。有効量は、前述のように被験体情報に応じて適宜選択される。

EBOV等のフィルウイルス科ウイルスは、マクロファージや樹状細胞等の貪食性免疫細胞に感染すると考えられ、感染後の時間経過とともに、創傷部位、呼吸器、又は消化器粘膜等の局所感染から全身感染へと移行するものと考えられる。それ故、全身感染に至っていない感染初期のステージでは局所感染部位や筋肉に投与し、全身感染のステージでは静注等により循環器内に投与することも可能である。
3.フィロウイルス科ウイルスの複製阻害方法
3-1.概要
本発明の第3の態様は、フィロウイルス科ウイルスの複製阻害方法である。本発明のフィロウイルス科ウイルス複製阻害方法は、フィロウイルス科ウイルス感染細胞若しくはその恐れのある細胞等の宿主内に、フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片、そのいずれかのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、又は前記ペプチド断片若しくは前記ポリペプチドをコードする核酸を含み、細胞内で前記ペプチド断片又は前記ポリペプチドを発現可能な発現ベクターを導入することで宿主内におけるフィロウイルス科ウイルスの複製を阻害し、それによってフィロウイルス科ウイルスの複増殖を抑制することができる。また、この方法をフィロウイルス科ウイルス感染者若しくはその感染の恐れがある者に投与すれば、フィロウイルス科ウイルス感染症の治療方法となり得る。

本発明の第3の態様は、フィロウイルス科ウイルスの複製阻害方法である。本発明のフィロウイルス科ウイルス複製阻害方法は、フィロウイルス科ウイルス感染細胞若しくはその恐れのある細胞等の宿主内に、フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片、そのいずれかのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、又は前記ペプチド断片若しくは前記ポリペプチドをコードする核酸を含み、細胞内で前記ペプチド断片又は前記ポリペプチドを発現可能な発現ベクターを導入することで宿主内におけるフィロウイルス科ウイルスの複製を阻害し、それによってフィロウイルス科ウイルスの複増殖を抑制することができる。また、この方法をフィロウイルス科ウイルス感染者若しくはその感染の恐れがある者に投与すれば、フィロウイルス科ウイルス感染症の治療方法となり得る。
3-2.方法
本態様におけるフィロウイルス科ウイルスの複製阻害方法は、必須の工程として導入工程を含む。
「導入工程」は、フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片、そのいずれかのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、又は前記ペプチド断片又は前記ポリペプチドをコードする核酸を含み、細胞内で前記ペプチド断片を発現可能な発現ベクター、すなわち、第1態様に記載のフィロウイルス科ウイルス複製阻害剤を宿主内に導入する工程である。
本態様におけるフィロウイルス科ウイルスの複製阻害方法は、必須の工程として導入工程を含む。
「導入工程」は、フィロウイルス科ウイルスのNPタンパク質又はVP30タンパク質に由来するペプチド断片、そのいずれかのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、又は前記ペプチド断片又は前記ポリペプチドをコードする核酸を含み、細胞内で前記ペプチド断片を発現可能な発現ベクター、すなわち、第1態様に記載のフィロウイルス科ウイルス複製阻害剤を宿主内に導入する工程である。
本態様において「宿主」とは、フィロウイルス科ウイルス複製阻害剤を導入可能な、細胞、組織、又は個体をいう。宿主が細胞の場合、細胞は、1又は複数の哺乳動物細胞であればよい。好ましくはフィロウイルス科ウイルスの宿主であるヒト、ゴリラ、チンパンジー、サル、コウモリ、及びシカ由来の細胞である。由来細胞の種類は問わない。フィロウイルス科ウイルスの感染対象細胞である各種器官や組織由来の細胞(例えば、マクロファージや樹状細胞等の貪食性免疫細胞)が対象となり得る。また宿主は、細胞株系又は初代培養細胞系のいずれであってもよい。限定はしないが、宿主はフィロウイルス科ウイルスに感染した細胞やその恐れがある細胞が好ましい。宿主が個体の場合、前述の被験者であればよい。
各発現ベクターの宿主への導入方法は、特に限定はしない。例えば、宿主が細胞や組織であれば、Green & Sambrook、2012、Molecular Cloning: A Laboratory Manual Fourth Ed.、Cold Spring Harbor Laboratory Press、Cold Spring Harbor、New York等に記載された当該分野で公知の遺伝子導入方法(形質転換方法)を用いればよい。例えば、リポフェクチン法(PNAS、1989、86: 6077;PNAS、1987、84: 7413)、エレクトロポレーション法、リン酸カルシウム法(Virology、1973、52: 456-467)、DEAE-Dextran法等が挙げられる。一方、宿主が個体、例えば被験者であれば、第2態様の「2-3.適用方法」に記載の方法で、被験者にフィロウイルス科ウイルス複製阻害剤、又はそれを有効成分として含むフィロウイルス科ウイルス感染症治療用医薬組成物を投与すればよい。それらを被験者に投与することで、本態様のフィロウイルス科ウイルス複製阻害方法は、フィロウイルス科ウイルス感染症治療方法となり得る。
一態様において、本態様のフィロウイルス科ウイルス複製阻害方法は、(a)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~337位からなるポリペプチド、(b)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質の1位~500位において20位~337位を含むポリペプチド、(c)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおいて配列番号1の20位~337位に相当する位置からなるポリペプチド、(d)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおける、配列番号1の1位~500位に相当する位置において、配列番号1の20位~337位に相当する位置を含むポリペプチド、(e)前記(a)~(d)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、(f)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~254位からなるポリペプチド、(g)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質の47位~288位において91位~254位を含むポリペプチド、(h)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の91位~254位に相当する位置からなるポリペプチド、(i)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおける、配列番号3の47位~288位に相当する位置において、配列番号3の91位~254位に相当する位置を含むポリペプチドからなる群から選択されるいずれかのポリペプチド、又は(j)前記(f)~(i)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド、又は前記ポリペプチドをコードする核酸を含み、細胞内で前記ポリペプチドを発現可能な発現ベクターを宿主内に導入する工程を含む。上記(a)~(j)の構成は第1態様の記載に準じる。
4.フィロウイルス科ウイルスの複製活性の評価方法
4-1.概要
本発明の第4の態様は、フィロウイルス科ウイルスの複製活性の評価方法である。本発明の評価方法は、レポーター遺伝子発現ベクターと共に、NP遺伝子発現ベクター、VP30遺伝子発現ベクター、VP35遺伝子発現ベクター、及びL遺伝子発現ベクターを宿主細胞内に導入し、レポーター遺伝子の発現を検出することで、フィロウイルス科ウイルスの複製活性を評価することができる。
4-1.概要
本発明の第4の態様は、フィロウイルス科ウイルスの複製活性の評価方法である。本発明の評価方法は、レポーター遺伝子発現ベクターと共に、NP遺伝子発現ベクター、VP30遺伝子発現ベクター、VP35遺伝子発現ベクター、及びL遺伝子発現ベクターを宿主細胞内に導入し、レポーター遺伝子の発現を検出することで、フィロウイルス科ウイルスの複製活性を評価することができる。
4-2.方法
本態様の評価方法は、必須の工程として導入工程及び検出工程を含む。

本明細書において「レポーター遺伝子発現ベクター」は、RNAポリメラーゼIプロモーター配列、フィロウイルス科ウイルスのトレーラー配列、レポーター遺伝子配列、フィロウイルス科ウイルスのリーダー配列、及びRNAポリメラーゼIターミネーター配列を上記順序で含む。レポーター遺伝子発現ベクターにおいて、レポーター遺伝子配列は、前記プロモーター配列に対して逆向きに含まれている。レポーター遺伝子は、その発現を検出可能な遺伝子であれば限定せず、例えば色素タンパク質、蛍光タンパク質、又は発光タンパク質をコードする遺伝子であってもよい。
NP遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのNP遺伝子が発現可能な状態で配置されている。
VP30遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのVP30遺伝子が発現可能な状態で配置されている。
VP35遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのVP35遺伝子が発現可能な状態で配置されている。
L遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのL遺伝子が発現可能な状態で配置されている。
上記の発現ベクターは、遺伝子配列及びプロモーターに加えて、必要に応じて、標識遺伝子(選抜マーカー)、エンハンサー、ターミネーター、複製起点、及びポリAシグナル等の構成要素を含んでいてもよい。各構成要素の構成は第1態様の発現ベクターの記載に準じる。
一実施形態において、NP遺伝子発現ベクター、VP30遺伝子発現ベクター、VP35遺伝子発現ベクター、及びL遺伝子発現ベクターの導入比率は4:1:1:2である。
「検出工程」は、レポーター遺伝子の発現を検出する工程である。レポーター遺伝子の発現を検出する方法は、レポーター遺伝子の種類に応じて適宜選択すればよい。レポーター遺伝子が蛍光タンパク質遺伝子であれば、特定波長の励起光を照射して蛍光タンパク質の蛍光強度を測定することができる。レポーター遺伝子が発光タンパク質遺伝子であれば、発光基質を培地に添加して発光強度を測定することができる。
一実施形態において、本態様の方法は、選択工程として宿主細胞を被験物質で処置する被験物質処置を含んでもよい。本工程を含む場合、被験物質のフィロウイルス科ウイルスに対する複製活性阻害効果を評価することができる。
<実施例1:EBOVミニゲノムアッセイの開発>
(目的)
EBOV複製効率を測定するためのEBOVミニゲノムアッセイを開発する。
(方法と結果)
(1)プラスミドの構築
EBOVタンパク質(L、NP、VP35、及びVP30)の発現プラスミドは、コドンをヒト細胞に最適化した遺伝子を哺乳動物細胞発現ベクターpCI (Promega)にサブクローニングしたものを使用した。ΔNP及びΔVP30発現プラスミドは、pCI-NP又はpCI-VP30から主に部位特異的変異導入(PrimeSTAR Mutagenesis Basal Kit, TaKaRa)を用いて作製した。一部のコンストラクトには、配列番号5で示すアミノ酸配列(GVAMPGAEDDVV)からなるPAタグを導入した。
(目的)
EBOV複製効率を測定するためのEBOVミニゲノムアッセイを開発する。
(方法と結果)
(1)プラスミドの構築
EBOVタンパク質(L、NP、VP35、及びVP30)の発現プラスミドは、コドンをヒト細胞に最適化した遺伝子を哺乳動物細胞発現ベクターpCI (Promega)にサブクローニングしたものを使用した。ΔNP及びΔVP30発現プラスミドは、pCI-NP又はpCI-VP30から主に部位特異的変異導入(PrimeSTAR Mutagenesis Basal Kit, TaKaRa)を用いて作製した。一部のコンストラクトには、配列番号5で示すアミノ酸配列(GVAMPGAEDDVV)からなるPAタグを導入した。
(2)EBOVミニゲノムアッセイ
本明細書の実施例においてウイルス複製効率の測定に用いたEBOVミニゲノムアッセイの原理を図3に示す。

EBOVゲノムの非翻訳領域であるTrailer (740bp)及びLeader (469bp)(図1)を両端に有するアンチセンス鎖の分泌型ルシフェラーゼ遺伝子(Gluc)をRNA ポリメラーゼI(RNA PolI)プロモーター(PRO)とターミネーター(TER)との間に挿入したレポーター遺伝子(図3、miniG)を作製した。miniG及びEBOVタンパク質(NP、VP35、VP30、及びL)の発現ベクターを電気的穿孔法によりHeLa細胞に導入し、24時間培養後に培養上清中のルシフェラーゼ活性を測定した。
miniGが細胞に導入されると、宿主のRNAポリメラーゼI(PolI)によってEBOVゲノムを模したレポーターRNA(図3、vRNA)が転写される。このレポーターRNAを鋳型として、NP、VP35、VP30、及びLによって、CapとPolyAを付加したGluc mRNA(図3、Gluc mRNA)が産生される。培養上清中に分泌されたルシフェラーゼ活性を測定することにより、EBOV複製活性を評価することができる。
(3)EBOVミニゲノムアッセイにおけるL、NP、VP35、及びVP30の最適比率の検討
EBOVミニゲノムアッセイにおけるL、NP、VP35、及びVP30の最適な導入比率を検討した。miniG及びEBOVタンパク質(L、NP、VP35、及びVP30)の発現プラスミドをHeLa細胞に導入し、24時間培養後に培養上清中のルシフェラーゼ活性を測定した。L、NP、VP35、及びVP30は図4に示す様々な比率で導入し、miniGとEBOVタンパク質との比率は1:1とした。
EBOVミニゲノムアッセイにおけるL、NP、VP35、及びVP30の最適な導入比率を検討した。miniG及びEBOVタンパク質(L、NP、VP35、及びVP30)の発現プラスミドをHeLa細胞に導入し、24時間培養後に培養上清中のルシフェラーゼ活性を測定した。L、NP、VP35、及びVP30は図4に示す様々な比率で導入し、miniGとEBOVタンパク質との比率は1:1とした。
結果を図4に示す。EBOVタンパク質の比率は、L:NP:VP35:VP30=4:1:1:2が最適であった。以降の実施例ではこの比率でEBOVタンパク質をHeLa細胞に導入し、その上で様々な欠失変異型タンパク質を導入することで、EBOV複製阻害効果を評価した。
<実施例2:欠失変異型NPによるEBOV複製阻害効果>
(目的)
様々な欠失変異型NPを作製し、EBOV複製に対する阻害効果を検討する。
(方法)
EBOVの野生型NPは、配列番号1で示すアミノ酸配列からなり、全長721アミノ酸からなるタンパク質である。EBOVの野生型NPには、N末端のコアドメイン(20~405アミノ酸残基)とC末端のCテール(641~721アミノ酸残基)との間に非保存領域(Non-conserved region)が存在する(図5A及び図5B)。NPのコアドメインはNローブ(20~240アミノ酸残基)とCローブ(241~405アミノ酸残基)を含み、NローブとCローブの間にはRNA結合グローブ(RNA binding groove)が形成される。NPの結晶構造解析の結果によれば、コアドメインには13個のαヘリックスが含まれる(Dong S, et al., Protein Cell, 2015, 6(5):351-62)。

(方法)

本発明者らは、様々な欠失変異型NP(ΔNP)のEBOV複製に対する阻害効果を検討する目的で、図5C、図6A、及び図7Aに示すΔNPを発現する発現プラスミドを作製した。各々のΔNPについて、以下の方法でEBOVミニゲノムアッセイを行った。miniG及びEBOVタンパク質(NP、VP35、VP30、及びL)発現プラスミドと共に、ΔNP発現プラスミドをHeLa細胞に遺伝子導入した。遺伝子導入の比率は、miniG:NP:VP35:VP30:L:ΔNP=8:4:1:1:2:16とした。野生型NPとΔNPの比率は、およそ1:4となる。24時間培養後に培養上清中のルシフェラーゼ活性を測定した。
(結果)
(1)欠失変異型NPによるEBOV複製阻害効果
図5C及び図6Aに示す9種類のΔNPによるEBOV複製阻害効果を検討した。各々のΔNPについてEBOVミニゲノムアッセイを行った結果を図6Bに示す。
(1)欠失変異型NPによるEBOV複製阻害効果

コアドメイン全長を含み非保存領域以降を欠くΔNP(1-405)又はΔNP(20-405)を導入した細胞では、ルシフェラーゼ活性が著しく阻害され、EBOV複製の抑制が認められた(図6B)。
一方、Nローブのみを含むΔNP(1-240)、及びCローブのみを含むΔNP(241-405)では、阻害効果が減弱した。
この結果から、コアドメイン全長を含み、非保存領域及びCテールを欠くことが複製阻害活性に重要であることが示された(図6B)。
(2)ΔNP(20-405)をさらに改変した欠失変異型NPによるEBOV複製阻害効果
(1)で阻害効果を示したΔNP(20-405)について、N末端側又はC末端側の部分をさらに欠失させたΔNPを作製した(図7A)。野生型NPとΔNPを1:4の比率で導入する条件、及び野生型NPとΔNPを1:1の比率で導入する条件下でEBOV複製阻害効果を測定した。
(1)で阻害効果を示したΔNP(20-405)について、N末端側又はC末端側の部分をさらに欠失させたΔNPを作製した(図7A)。野生型NPとΔNPを1:4の比率で導入する条件、及び野生型NPとΔNPを1:1の比率で導入する条件下でEBOV複製阻害効果を測定した。
ルシフェラーゼ活性を測定した結果、ΔNP(20-405)のN末端からαヘリックスを1つ欠損させた場合(ΔNP(63-405))、及びC末端のαヘリックスを2つ欠損させた場合(ΔNP(20-313))に、複製阻害活性が減弱する傾向が見られた(図7B)。一方、C末端のαヘリックスを1つ欠損させた場合(ΔNP(20-337))には複製阻害活性が減弱しなかった(図7B)。この結果から、コアドメインに含まれる13個のαヘリックスのうちα1~α12が複製阻害活性に重要であることが明らかとなった。
さらに、ΔNP(20-337)は、細胞に導入したプラスミドの量に依存してEBOV複製を阻害することが示された(図8)。
(3)アミノ酸置換変異を導入したΔNP(20-337)によるEBOV複製阻害効果
EBOVの野生型NPが持つNP-NP相互作用(多量体形成)、RNA結合、及びVP35結合活性について、その各々の活性を損なうアミノ酸置換変異が知られている。これらの変異をΔNP(20-337)発現プラスミドに導入し、EBOV複製阻害効果に対するその影響を検討した(図9)。
EBOVの野生型NPが持つNP-NP相互作用(多量体形成)、RNA結合、及びVP35結合活性について、その各々の活性を損なうアミノ酸置換変異が知られている。これらの変異をΔNP(20-337)発現プラスミドに導入し、EBOV複製阻害効果に対するその影響を検討した(図9)。
NP-NP相互作用の活性を損なうY21A/H22A(YHAA)変異を導入したΔNP(20-337)は、野生型ΔNP(20-337)と比較して複製阻害活性が減弱していた(図9、Wt及びYHAA)。RNA結合能を損なうK160A/K171A/R174A(KKR)変異を導入したΔNP(20-337)、及びVP35との結合能を損なうR240A/K248A/D252A(RKD)変異を導入したΔNP(20-337)では、複製阻害活性がほとんど認められなかった(図9、KKR及びRKD)。
これらの結果から、ΔNPのEBOV複製阻害活性には、NP-NP相互作用、RNA結合能、及びVP35結合能のいずれも不可欠であることが示された。
<実施例3:欠失変異型VP30によるEBOV複製阻害効果>
(目的)
VP30の様々な欠失変異体を作製し、EBOV複製に対する阻害効果を検討する。
(方法)
EBOVの野生型VP30は、配列番号3で示すアミノ酸配列からなり、全長288アミノ酸からなるタンパク質である。VP30は、N末端ドメイン(NTD、1~141アミノ酸残基)及びC末端ドメイン(CTD、142~288アミノ酸残基)の2つのドメイン構造を有する(図10)。NTDには、RNA結合領域(26~46アミノ酸残基)、Znフィンガーモチーフ(72~90アミノ酸残基)、及び六量体化モチーフ(100~104アミノ酸残基)が存在する。CTDには二量体化及びNP結合領域(180~197アミノ酸残基)が含まれる。
(目的)
VP30の様々な欠失変異体を作製し、EBOV複製に対する阻害効果を検討する。
(方法)
EBOVの野生型VP30は、配列番号3で示すアミノ酸配列からなり、全長288アミノ酸からなるタンパク質である。VP30は、N末端ドメイン(NTD、1~141アミノ酸残基)及びC末端ドメイン(CTD、142~288アミノ酸残基)の2つのドメイン構造を有する(図10)。NTDには、RNA結合領域(26~46アミノ酸残基)、Znフィンガーモチーフ(72~90アミノ酸残基)、及び六量体化モチーフ(100~104アミノ酸残基)が存在する。CTDには二量体化及びNP結合領域(180~197アミノ酸残基)が含まれる。
本発明者らは、様々な欠失変異型VP30(ΔVP30)のEBOV複製に対する阻害効果を検討する目的で、図11A、図12A、図13A、及び図14Aに示すΔVP30を発現する発現プラスミドを作製した。各々のΔVP30について、以下の方法でEBOVミニゲノムアッセイを行った。miniG及びEBOVタンパク質(NP、VP35、VP30、及びL)発現プラスミドと共にΔVP30発現プラスミドをHeLa細胞に遺伝子導入した。遺伝子導入の比率は、miniG:NP:VP35:VP30:L:ΔVP30=8:4:1:1:2:16とした。野生型VP30とΔVP30の比率は、1:16となる。24時間培養後に培養上清中のルシフェラーゼ活性を測定した。
(結果)
(1)欠失変異型VP30によるEBOV複製阻害効果
図11Aに示す9種類のΔVP30についてEBOV複製阻害効果を検討した。各々のΔVP30についてEBOVミニゲノムアッセイを行った結果を図11Bに示す。

9種類のΔVP30では、N末端のRNA結合領域とZnフィンガーモチーフを欠くΔVP30(91-288)がEBOV複製を強く抑制することが示された(図11B、7)。一方、C末端を欠損したΔVP30(図11B、1~5)や、N末端のRNA結合領域とZnフィンガーモチーフに加えて六量体化モチーフを欠損するΔVP30(図11B、8及び9)では、複製阻害活性が減弱していることが示された。
この結果から、ΔVP30の複製阻害活性には、N末端のRNA結合領域とZnフィンガーモチーフを欠くことと同時に、六量体化モチーフ及びNP結合領域を含むことが重要であることが示唆された。
(2)ΔVP30(91-288)をさらに改変した欠失変異型VP30によるEBOV複製阻害効果
(1)で阻害効果を示したΔVP30(91-288)を、C末端側からさらに欠損させたΔVP30を作製し、そのEBOV複製阻害効果を検討した(図12)。

ルシフェラーゼ活性を測定した結果、NP結合領域を欠くΔVP30(91-179)及びΔVP30(91-141)では、EBOV複製阻害効果が著しく減弱することが明らかとなった。この結果から、NP結合領域を含むCTDがEBOV複製阻害活性には重要であることが示唆された。
VP30のCTDは7個のαヘリックスによって構成されることが知られている。そこで、EBOV複製阻害効果を示したΔVP30(91-288)からC末端のαヘリックスを欠損させ、EBOV阻害効果に及ぼす影響を検討した(図13)。その結果、7番目のαヘリックス(図13A、α7)を欠損させたΔVP30(91-254)は、7個のαヘリックスを全て含むΔVP30(91-272)と同等のEBOV複製阻害活性を示した(図13B)。一方、6番目以降のαヘリックスを欠くΔVP30(91-231)では、阻害効果が減弱することが明らかとなった(図13B)。
また、ΔVP30(91-254)は、ΔVP30(91-272)と同様に濃度依存的なEBOV複製阻害活性を示した(図14)。
以上の結果から、ΔVP30(91-254)が、顕著なEBOV複製阻害活性を有する最小のΔVP30であることが示された。
<実施例4:カプシド形成に対する阻害効果>
(目的)
ΔNP及びΔVP30によるEBOV複製阻害の作用機序を検討する。
(方法と結果)
(1)PAタグをC末端に付加したNPであるNP-PAの作製
配列番号5で示すアミノ酸配列(GVAMPGAEDDVV)からなるPAタグをC末端に付加したNPであるNP-PAを作製し、上記と同様にEBOVミニゲノムアッセイを行った。その結果、NP-PAは野生型NPと同様にEBOV複製において機能することが示された(図15A)。
(目的)
ΔNP及びΔVP30によるEBOV複製阻害の作用機序を検討する。
(方法と結果)
(1)PAタグをC末端に付加したNPであるNP-PAの作製
配列番号5で示すアミノ酸配列(GVAMPGAEDDVV)からなるPAタグをC末端に付加したNPであるNP-PAを作製し、上記と同様にEBOVミニゲノムアッセイを行った。その結果、NP-PAは野生型NPと同様にEBOV複製において機能することが示された(図15A)。
次に、野生型NPの代わりにNP-PAを用いたEBOV複製が、ΔNP(20-405)又はΔVP30(91-254)の存在下で阻害されるか否かを検討した。その結果、NP-PAによるEBOV複製は、野生型NPによるEBOV複製と同様に、ΔNP(20-405)及びΔVP30(91-254)によって効率的に阻害することが示された(図15B)。
(2)NP-PAによる複合体形成のパーティクルブロッティングによる検出
NP-PAの細胞内における複合体形成をパーティクルブロッティングにより検討した。NP-PA発現プラスミドと共にminiG及びEBOVタンパク質(VP35、VP30、L)発現プラスミドをHeLa細胞に遺伝子導入した。miniG:NP-PA:VP35:VP30:Lの導入比率は8:4:1:1:2とした。24時間培養後に抗PA抗体を用いたパーティクルブロッティングを行った。
NP-PAの細胞内における複合体形成をパーティクルブロッティングにより検討した。NP-PA発現プラスミドと共にminiG及びEBOVタンパク質(VP35、VP30、L)発現プラスミドをHeLa細胞に遺伝子導入した。miniG:NP-PA:VP35:VP30:Lの導入比率は8:4:1:1:2とした。24時間培養後に抗PA抗体を用いたパーティクルブロッティングを行った。
パーティクルブロッティングは以下の方法で行った。遺伝子導入した細胞を24時間培養後にPB lysis buffer (1%NP40, 25mM Tris pH7.4, 150mM NaCl, 1mM EDTA, 50mM NaF)で溶解し、1.2%アガロース(TAE)に泳動し、PVDF膜に転写した。抗PAタグモノクローナル抗体(NZ-1)は富士フィルム和光純薬から購入した。
パーティクルブロッティングの結果、NP-PAが複合体(カプシド)を形成していると思われるバンドが確認された(図16)。
(3)ΔNP及びΔVP30の複合体形成に対する阻害効果の検討
次に、NP-PAを用いたミニゲノムアッセイにΔNP又はΔVP30発現プラスミドを共導入し、NP-PAの複合体形成に対する阻害効果を検討した。ΔNPの導入比率は、miniG:NP-PA:VP35:VP30:L:ΔNP=8:4:1:1:2:16とした。ΔVP30の導入比率は、miniG:NP-PA:VP35:VP30:L:ΔVP30=8:4:1:1:2:16とした。
次に、NP-PAを用いたミニゲノムアッセイにΔNP又はΔVP30発現プラスミドを共導入し、NP-PAの複合体形成に対する阻害効果を検討した。ΔNPの導入比率は、miniG:NP-PA:VP35:VP30:L:ΔNP=8:4:1:1:2:16とした。ΔVP30の導入比率は、miniG:NP-PA:VP35:VP30:L:ΔVP30=8:4:1:1:2:16とした。
その結果、ΔNP(20-405)を導入した細胞においては、NP-PAが形成する複合体のバンドが消失していた(図17)。したがって、ΔNP(20-405)発現はNPの複合体すなわちカプシド形成自体を阻害することにより、EBOV複製を阻害することが示唆された。
一方、ΔVP30(91-254)の発現は、NPによるカプシド形成は阻害しないものの、VP30のカプシドへの結合を阻害することにより、EBOV複製を阻害することが示唆された。

(j)前記(f)~(i)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド - 前記(g)に記載のポリペプチドが、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において47位~254位を含む、又は
前記(i)に記載のポリペプチドが、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の47位~254位に相当する位置を含む、
請求項4に記載のフィロウイルス科ウイルス複製阻害剤。 - 前記(g)に記載のポリペプチドが、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~288位を含む、又は
前記(i)に記載のポリペプチドが、配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の91位~288位に相当する位置を含む、
請求項4又は5に記載のフィロウイルス科ウイルス複製阻害剤。 - 前記フィロウイルス科オルソログがエボラウイルス属オルソログである、請求項1~6のいずれか一項に記載のフィロウイルス科ウイルス複製阻害剤。
- 請求項1に記載の(a)~(e)及び請求項4に記載の(f)~(j)からなる群から選択されるいずれかのポリペプチドをコードする核酸を発現可能な状態で含む発現ベクターからなるフィロウイルス科ウイルス複製阻害剤。
- 有効成分として請求項1~8のいずれか一項に記載のフィロウイルス科ウイルス複製阻害剤を含む、フィロウイルス科ウイルス感染症治療用医薬組成物。
- 以下の(a)~(j)からなる群から選択されるいずれかのポリペプチド、又は
前記ポリペプチドをコードする核酸を含み、細胞内で前記ポリペプチドを発現可能な発現ベクター
を宿主内に導入する工程を含む、フィロウイルス科ウイルスの複製阻害方法。
(a)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質において20位~337位からなるポリペプチド
(b)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質の1位~500位において20位~337位を含むポリペプチド
(c)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおいて配列番号1の20位~337位に相当する位置からなるポリペプチド
(d)配列番号1で示すアミノ酸配列からなるザイールエボラウイルスのNPタンパク質のフィロウイルス科オルソログにおける、配列番号1の1位~500位に相当する位置において、配列番号1の20位~337位に相当する位置を含むポリペプチド
(e)前記(a)~(d)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド
(f)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質において91位~254位からなるポリペプチド
(g)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質の47位~288位において91位~254位を含むポリペプチド
(h)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおいて配列番号3の91位~254位に相当する位置からなるポリペプチド
(i)配列番号3で示すアミノ酸配列からなるザイールエボラウイルスのVP30タンパク質のフィロウイルス科オルソログにおける、配列番号3の47位~288位に相当する位置において、配列番号3の91位~254位に相当する位置を含むポリペプチド
(j)前記(f)~(i)のいずれかに記載のポリペプチドのN末端及び/又はC末端に任意のアミノ酸配列が付加されたポリペプチド - フィロウイルス科ウイルスの複製活性の評価方法であって、
レポーター遺伝子発現ベクター、NP遺伝子発現ベクター、VP30遺伝子発現ベクター、VP35遺伝子発現ベクター、及びL遺伝子発現ベクターを宿主細胞内に導入する導入工程、並びに
前記レポーター遺伝子の発現を検出する検出工程
を含み、
前記レポーター遺伝子発現ベクターは、
RNAポリメラーゼIプロモーター配列、
フィロウイルス科ウイルスのトレーラー配列、
レポーター遺伝子配列、
フィロウイルス科ウイルスのリーダー配列、及び
RNAポリメラーゼIターミネーター配列
を上記順序で含み、
ここで前記レポーター遺伝子配列は、前記プロモーター配列に対して逆向きに含まれており、
前記NP遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのNP遺伝子が発現可能な状態で配置されており、
前記VP30遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのVP30遺伝子が発現可能な状態で配置されており、
前記VP35遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのVP35遺伝子が発現可能な状態で配置されており、
前記L遺伝子発現ベクターは、宿主細胞内で遺伝子発現誘導が可能なプロモーターの下流に、フィロウイルス科ウイルスのL遺伝子が発現可能な状態で配置されており、
NP遺伝子発現ベクター、VP30遺伝子発現ベクター、VP35遺伝子発現ベクター、及びL遺伝子発現ベクターの導入比率が4:1:1:2である、
