Abstract
Description
本出願は、参照によりここに援用されるところの日本出願特願2020-199792号優先権を請求する。
一方、シリコーンハイドロゲルレンズは含水率が低く疎水性が高いため、涙液をはじきやすい、脂質汚れが付着しやすいという欠点があった。また近年、瞬目時の眼瞼結膜縁と眼表面との摩擦が原因となって、眼瞼結膜表層の上皮細胞の脱落と変性が起こることが報告されている(非特許文献1、非特許文献2)。これにより、シリコーンハイドロゲルレンズに親水性とともに潤滑性を付与することが求められている。そのため、シリコーンハイドロゲルレンズの表面の親水性、潤滑性を改善するために、様々な方案が検討されている。
例えば、コンタクトレンズの生産工程において、レンズのプラズマ処理工程を含めることが広く実施されている。プラズマ処理は比較的高い親水性とその耐久性を付与できるという利点があるが、潤滑性が付与できず、また大規模な設備投資を必要とするという欠点がある。
大規模な設備投資を必要としない方法として、N-ビニルピロリドン等の親水性単量体や、ポリビニルアルコール等の湿潤成分をレンズモノマー組成物に添加する方法が知られている。この方法の利点は、成型前のレンズに湿潤成分を添加することで、レンズを湿潤化するための後処理工程を省略できる点にある。しかしながら、親水性の高い湿潤成分を、疎水性の高いシリコーン成分を含むモノマー組成物と相溶させるためには、レンズモノマー組成に制約が伴う。従って、より幅広いレンズ組成に応用可能な技術の開発が求められている。
湿潤成分によるレンズ表面のコーティングする方法が知られている。コーティングのための湿潤成分としては一般に親水性のポリマーが用いられており、特に両性イオン性官能基である2-メタクリロイルオキシエチルホスホリルコリン(以下「MPC」と略記する)を用いたポリマーが、親水性と潤滑性を付与する効果に優れていることが知られている。
以上により、より幅広いレンズ組成に応用可能な技術の開発が求められていた。
また、該眼科デバイス用表面処理剤を用いて表面を処理した表面処理済シリコーンハイドロゲル、及び該表面処理済シリコーンハイドロゲルを用いた眼科デバイスを提供することにある。
すなわち、本発明は以下の通りである。
2.前記熱反応性単量体もしくは光反応性単量体が、光反応性単量体である、前項1に記載の眼科デバイス用表面処理剤。
3.前記式(1)で表される親水性単量体aに基づく構成単位が2-メタクリロイルオキシエチルホスホリルコリンであり、前記式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位がポリジメチルシロキサンモノメタクリレート、トリス(トリメチルシロキシ)シリルプロピルメタクリレート、4-(2-ヒドロキシエチル)-2-メチレン-1-(トリス(トリメチルシロキシ)シリルプロピル)ブタン-1,4-ジオエート若しくは3-(3-(メチルビス(トリメチルシロキシ)シリル)プロピル)グリセロール-1-メタクリレート、並びに、熱反応性単量体もしくは光反応性単量体cに基づく構成単位がメタクリロイルオキシベンゾフェノン、4-(4-アジドベンゾイルオキシメチル)ビニルベンゼン若しくはグリシジルメタクリレートである、前項1に記載の眼科デバイス用表面処理剤。
4.前記式(1)で表される親水性単量体aに基づく構成単位、前記式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位、及び/又は、熱反応性単量体もしくは光反応性単量体cに基づく構成単位の組み合わせは、以下のいずれか1から選択される、前項1に記載の眼科デバイス用表面処理剤。
1)2-メタクリロイルオキシエチルホスホリルコリン、ポリジメチルシロキサンモノメタクリレート及びメタクリロイルオキシベンゾフェノン

9.共重合体でシリコーンハイドロゲルの表面を処理する工程を含む、シリコーンハイドロゲルの表面処理方法、
ここで、該共重合体は、下記の式(1)で表される親水性単量体aに基づく構成単位、下記の式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位並びに熱反応性単量体もしくは光反応性単量体cに基づく構成単位を含有する共重合体であって、60℃の熱水に0.5%(w/v)溶解するが、20℃の水に1%(w/v)溶解しない共重合体である、表面処理方法。
10.前記式(1)で表される親水性単量体aに基づく構成単位が2-メタクリロイルオキシエチルホスホリルコリンであり、前記式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位がポリジメチルシロキサンモノメタクリレート、トリス(トリメチルシロキシ)シリルプロピルメタクリレート、4-(2-ヒドロキシエチル)-2-メチレン-1-(トリス(トリメチルシロキシ)シリルプロピル)ブタン-1,4-ジオエート若しくは3-(3-(メチルビス(トリメチルシロキシ)シリル)プロピル)グリセロール-1-メタクリレート、並びに、熱反応性単量体もしくは光反応性単量体cに基づく構成単位がメタクリロイルオキシベンゾフェノン、4-(4-アジドベンゾイルオキシメチル)ビニルベンゼン若しくはグリシジルメタクリレートである、前項9に記載のシリコーンハイドロゲルの表面処理方法。
11.前記式(1)で表される親水性単量体aに基づく構成単位、前記式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位、及び/又は、熱反応性単量体もしくは光反応性単量体cに基づく構成単位の組み合わせは、以下のいずれか1から選択される、前項9に記載のシリコーンハイドロゲルの表面処理方法。

なお、本明細書において、「(メタ)アクリル酸」とは、「アクリル酸又はメタクリル酸」を意味し、他の類似用語についても同様である。
また、本明細書において、好ましい数値範囲(例えば、含有量や重量平均分子量の範囲)を段階的に記載した場合、各下限値及び上限値は、それぞれ独立して組み合わせることができる。例えば、「好ましくは10~100、より好ましくは20~90」という記載において、「好ましい下限値:10」と「より好ましい上限値:90」とを組み合わせて「10~90」とすることができる。
また、本発明の眼科デバイスは、コンタクトレンズ、眼内レンズ、人口角膜等を例示することができるが、特に限定されない。
式(1)で表される親水性単量体aのR1は水素原子、またはメチル基を表すが、原料入手性の観点からメチル基が好ましい。
またL1は炭素数2~6の有機基であって、ここで該有機基は任意に1個のエーテル結合および/または1個のヒドロキシ基を含んでもよいが、原料入手性の観点から-C2H4-、―C2H4-O-C2H4-、-C2H5-が好ましく、-C2H5-が特に好ましい。
ここで、有機基とは、C、Si、N、P、O、Sといった元素により構成される基であり、繰り返し単位を有する重合体であってもよい。また、その構造中に、ケトン基、エステル基、エーテル基、ヒドロキシル基、アミド基、チオエーテル基、イソシアヌレート基等の基を含んでもよい。
またR2~R4はそれぞれ独立して炭素数1~3の炭化水素基を表すが、原料入手性の観点から全てメチル基とすることが好ましい。
従って、親水性単量体aの具体例としては、2-メタクリロイルオキシエチルホスホリルコリン等を好ましく挙げることができる。
本発明で用いる単量体aとしては、単一の単量体を用いてもよいし、複数の単量体を組合わせて用いてもよい。
式(2)で表される場合のシリコーン単量体bのX1は、(メタ)アクリロイルオキシ基、3-(2-ヒドロキシエチルオキシカルボニル)-2-メチレンプロパノイルオキシ基、3-(2-ヒドロキシエチルオキシカルボニル)-3-ブテノイルオキシ基または3-(2-ヒドロキシエチルオキシカルボニル)-2-プロペノイルオキシ基のいずれかの重合性官能基を表す。このうち、原料入手性の観点からはメタクリロイルオキシ基が好ましく選択され、また親水性単量体との相溶性の観点からは3-(2-ヒドロキシエチルオキシカルボニル)-2-メチレンプロパノイルオキシ基が好ましく選択される。
またL2は炭素数2~6の有機基を表し、ここで該有機基は任意に1個のエーテル結合および/または1個のヒドロキシ基を含んでもよいが、原料入手性の観点から―n-C3H6-が好ましく選択される。
またR5~R13はそれぞれ独立してメチル基またはエチル基を表すが、原料入手性の観点から全てメチル基とすることが好ましい。
またn1は0または1を表すが、シリコーンハイドロゲル表面への固定化の観点から、1とすることが好ましい。
従って、式(2)で表される単量体bの具体例としては、トリス(トリメチルシロキシ)シリルプロピルメタクリレート、4-(2-ヒドロキシエチル)-2-メチレン-1-(トリス(トリメチルシロキシ)シリルプロピル)ブタン-1,4-ジオエート、3-(3-(メチルビス(トリメチルシロキシ)シリル)プロピル)グリセロール-1-メタクリレート等を好ましく挙げることができる。
またL3は炭素数2~6の有機基を表し、ここで該有機基は任意に1個のエーテル結合および/または1個のヒドロキシ基を含んでもよい。
またn2は4~20の整数を表す。また通常n2は分布を有し、その場合その数平均値が4~20の範囲内であることが好ましい。
また、R15~R17はそれぞれ独立して炭素数1~8のアルキル基を表す。
従って、式(3)で表される単量体bの具体例としては、ポリジメチルシロキサンモノメタクリレート、片末端モノメタクリロイルオキシプロピル変性ポリジメチルシロキサン等を好ましく挙げることができる。
シリコーンハイドロゲル表面の固定化の観点では、式(2)で表される単量体を単独で用いることが好ましい。
単量体aと単量体bの比率は、単量体a:単量体b=5/1~300/1(質量比)とすることが好ましく、10/1~50/1とすることがより好ましく、20/1~50/1とすることがよりさらに好ましい。該範囲とすることで、シリコーンハイドロゲルに濡れ性、滑り性を付与する効果が高くなる。
また、単量体aが100質量部に対して、単量体bが0.3~20質量部、好ましくは2~10質量部、より好ましくは2~5質量部とすることができる。
また、本発明の共重合体を調製する際に使用する全ての単量体のうちに占める単量体a及びbを合わせた比率は、50~100mol%とすることが好ましく、80~100mol%とすることがさらに好ましい。本発明の効果が好ましく発現するからである。
熱反応性単量体もしくは光反応性単量体cは、単量体cを共重合させた共重合体が、加熱、または光照射されることによって誘導され、共重合体と共存する有機物との間に炭素間共有結合を形成しうるような反応性を指す。
加熱によって反応性が誘導される単量体c(熱反応性単量体もしくは光反応性単量体c)の具体例としては、グリシジル(メタ)アクリレート等が好ましく挙げられる。
光照射によって反応性が誘導される単量体c(熱反応性単量体もしくは光反応性単量体c)の具体例としては、4-(メタ)アクリロイルオキシベンゾフェノン、4-(4-アジドベンゾイルオキシメチル)スチレン等が好ましく挙げられる。
また上記の単量体cの反応性を利用して、共重合体の合成後に高分子反応によって(メタ)アクリル基、ビニル基等の重合性官能基を導入することも、本発明の趣旨から可能である。
単量体cの反応性は、光照射によって誘導される反応性が好ましい。より好ましく、シリコーンハイドロゲルの表面処理が実施できるからである。
単量体cは市販品を用いてもよいし、国際公開WO2018/003821号に開示されている様な方法を用いて調製してもよい。
本発明で用いる熱反応性単量体もしくは光反応性単量体cとしては、単一の単量体を用いてもよいし、複数の単量体を組み合わせて用いてもよい。
本発明の共重合体を調製する際に使用する全ての単量体に占める単量体cの比率は、0~50mol%とすることが好ましく、0~20mol%とすることがさらに好ましい。本発明の効果が好ましく発現するからである。
本発明の共重合体は、本発明の効果を損なわない範囲で、他の単量体dを共重合させたものであってもよい。
単量体dは単量体a、b及びcと共重合可能な単量体から選択することが好ましい。
そのような単量体としては例えば、(メタ)アクリル酸及びその誘導体、(メタ)アクリルアミド、及びその誘導体、ビニル化合物が好ましく挙げられる。
(メタ)アクリル酸の誘導体としては例えば、(メタ)アクリル酸メチル、(メタ)アクリル酸エチル等の(メタ)アクリル酸アルキル、ヒドロキシエチル(メタ)アクリレート、ヒドロキシプロピル(メタ)アクリレート等のヒドロキシアルキル(メタ)アクリレート、等が挙げられる。
(メタ)アクリルアミド誘導体としては例えば、ジメチル(メタ)アクリルアミド、ヒドロキシエチル(メタ)アクリルアミド等が好ましく挙げられる。
ビニル化合物としては例えば、ヒドロキシエチルビニルエーテル、ヒドロキシブチルビニルエーテル等のヒドロキシアルキルビニルエーテル、ジエチレングリコールモノビニルエーテル等のオリゴエチレングリコールモノビニルエーテル、N-ビニルピロリドン等のピロリドン誘導体が好ましく挙げられる。
単量体dは単一の単量体を用いてもよいし、複数の単量体を組み合わせて用いてもよい。
本発明の共重合体を調製する際に使用する全ての単量体に占める単量体dの比率は0~10mol%とすることが好ましい。本発明の効果が好ましく発現するからである。
本発明の共重合体を得るための重合方法としては、溶液重合、塊状重合、乳化重合、懸濁重合等の公知の方法を用いることができ、例えば、単量体a、単量体b、単量体c及び必要に応じて単量体dを溶媒中で重合開始剤の存在下で重合させる、ラジカル重合などの方法を採用することができる。
重合反応に用いる開始剤としては、通常用いられる開始剤ならばいずれを用いてもよく、例えば、ラジカル重合の場合には脂肪族アゾ化合物、有機過酸化物、過硫酸塩等を用いることができる。これらの重合開始剤の例としては、例えば、2,2’-アゾビス(イソブチロニトリル)、2,2’-アゾビス(2,4-ジメチルヴァレロニトリル)、過酸化ベンゾイル、過酸化ラウロイル、ジイソプロピルペルオキシジカーボネート、t-ブチルペルオキシ-2-エチルヘキサノエート、t-ブチルペルオキシピバレート、t-ブチルペルオキシジイソブチレート、過硫酸カリウム、過硫酸アンモニウム等が挙げられる。これらの重合開始剤は2種以上を混合して使用してもよい。また、重合開始剤の使用にはレドックス系のラジカル促進剤を使用してもよい。
重合温度としては、30~80℃が好ましく、40~70℃がより好ましい。また重合時間は2~72時間が好ましい。重合反応が良好に進行するからである。
さらに、重合反応を円滑に行うために溶媒を用いてもよく、該溶媒としては、水、メタノール、エタノール、プロパノール等のアルコール、ベンゼン、トルエン、ジメチルホルムアミド、テトラヒドロフラン、ジオキサン、クロロホルム、またはこれらの混合溶媒を挙げることができる。
本発明の共重合体の重量平均分子量は、10,000~2,000,000とすることが好ましく、10,000~500,000とすることがさらに好ましく、100,000~200,000とすることがよりさらに好ましい。重量平均分子量をこれらの範囲とすることで、シリコーンハイドロゲル表面に濡れ性と滑り性を好ましく付与することができる。
なお、本発明の共重合体の重量平均分子量は、例えば、EcoSECシステム(東ソー株式会社製)を用いたGPC(ゲルろ過クロマトグラフィー)測定により、ポリエチレングリコール換算で求められる。
本発明の共重合体は、60℃の熱水に0.5%(w/v)溶解するが、20℃の水に1%(w/v)溶解しない。
20℃の水に1%(w/v)以上溶解する高いレベルの水溶性を有する場合は、疎水性の高いシリコーンハイドロゲル表面との親和性が低く、好適な表面処理ができない可能性がある。また熱水に0.5%(w/v)溶解しない低いレベルの水溶性では、シリコーンハイドロゲルに濡れ性を付与する効果が損なわれる可能性が高い。
(1)密栓可能な耐熱容器に、適量の共重合体を秤量する。そのような容器としては、例えばガラスバキュームバイアルが挙げられる。
(2)所定の濃度に調製すべく純水を該容器に添加する。例えば5.0%(w/v)に調製する場合は、0.5gの共重合体に対し、10mLの純水を添加する。
(3)20℃条件下で攪拌し、目視で溶解性を確認する。
(4)60℃で30分加温しながら攪拌し、室温に戻してから目視で溶解性を確認する。(熱水への溶解性)
なお、ここで言う「溶解」とは、目視下において、投入した共重合体の全てが水と均一に混合し、不溶分や濁り、よどみが視認できなくなる状態を指す。
そこで、水溶性の指標WSI(Water Solubility Index)を下記の式(4)の通り定義できる。
WSIが上記の範囲となるとき、好ましい程度の水溶性を有する共重合体が得やすい。
本発明のもう1つの形態は、本発明の共重合体で表面を処理した表面処理済シリコーンハイドロゲルである。
詳しくは、(工程A)本発明の共重合体を0.5wt%以上含有する溶液にシリコーンハイドロゲルを浸漬させる工程、及び(工程B)該共重合体をシリコーンハイドロゲル表面に固定化する工程、を含むシリコーンハイドロゲルの表面処理方法により得られるシリコーンハイドロゲルである。
前記工程Aで用いる、本発明の共重合体を0.5wt%以上含有する溶液は、本発明の共重合体を適切な溶媒と混合し、適切な温度で攪拌する等の方法で容易に調製することができる。
前記工程Bは、前記工程Aを経たシリコーンハイドロゲルに対し、加熱、または光照射を施すことで実施できる。
前記工程Bが加熱による場合は、例えば前記工程Aを経たシリコーンハイドロゲルをオートクレーブすることで実施できる。
また、前記工程Bが光照射による場合は、前記工程Aを経たシリコーンハイドロゲルに対し、適切な光照射を行うことにより実施できる。
前記工程Bは光照射によることがより好ましい。対象のシリコーンハイドロゲルに好適に濡れ性、滑り性を付与できるからである。
前記工程Bを施すシリコーンハイドロゲルは、工程Aで用いた本発明の共重合体を含む溶液に浸漬されたままでもよい。
前記工程Bは、前記工程A以降の任意の段階で実施できるが、前記工程Aの後(直後)に行うことが好ましい。
本発明で表面処理の対象とするシリコーンハイドロゲルの調製は、例えば国際公開WO2020/054711号等に開示されている様な公知の方法を用いて行うことができる。
また、シリコーンハイドロゲルは、シリコーン単量体に基づく構成単位とハイドロゲル単量体に基づく構成単位を含み。シリコーン単量体としては、以下を例示することができる。
トリメチルシロキシ基を有する(メタ)アクリレート類としては、3-[トリス(トリメチルシロキシ)シリル]プロピル(メタ)アクリレート、3-[ビス(トリメチルシロキシ)メチルシリル]プロピル(メタ)アクリレート、3-[(トリメチルシロキシ)ジメチルシリル]プロピル(メタ)アクリレート等が挙げられる。
トリメチルシロキシ基を有する(メタ)アクリルアミド類としては、3-[トリス(トリメチルシロキシ)シリル]プロピル(メタ)アクリルアミド、3-[ビス(トリメチルシロキシ)メチルシリル]プロピル(メタ)アクリルアミド、3-[(トリメチルシロキシ)ジメチルシリル]プロピル(メタ)アクリルアミド等が挙げられる。
トリメチルシロキシ基を有するメチル(メタ)アクリレート類としては、[トリス(トリメチルシロキシ)シリル]メチル(メタ)アクリレート、[ビス(トリメチルシロキシ)メチルシリル]メチル(メタ)アクリレート、[(トリメチルシロキシ)ジメチルシリル]メチル(メタ)アクリレート等が挙げられる。
トリメチルシロキシ基を有するメチル(メタ)アクリルアミド類としては、[トリス(トリメチルシロキシ)シリル]メチル(メタ)アクリルアミド、[ビス(トリメチルシロキシ)メチルシリル]メチル(メタ)アクリルアミド、[(トリメチルシロキシ)ジメチルシリル]メチル(メタ)アクリルアミド等が挙げられる。
トリメチルシロキシ基を有するスチレン類としては、[トリス(トリメチルシロキシ)シリル]スチレン、[ビス(トリメチルシロキシ)メチルシリル]スチレン、[(トリメチルシロキシ)ジメチルシリル]スチレン等が挙げられる。
トリメチルシロキシ基を有するカルバミン酸ビニル類としては、N-[3-[トリス(トリメチルシロキシ)シリル]プロピル]カルバミン酸ビニル、N-[3-[ビス(トリメチルシロキシ)メチルシリル]プロピル]カルバミン酸ビニル、N-[3-[(トリメチルシロキシ)ジメチルシリル]プロピル]カルバミン酸ビニル等が挙げられる。
本発明は、以下のシリコーンハイドロゲルの表面処理方法も含む。
本発明の共重合体でシリコーンハイドロゲルの表面を処理する工程を含む、シリコーンハイドロゲルの表面処理方法。

下記の式(1)で表される親水性単量体aに基づく構成単位、下記の式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位並びに熱反応性単量体もしくは光反応性単量体cに基づく構成単位を含有する共重合体であって、60℃の熱水に0.5%(w/v)溶解するが、20℃の水に1%(w/v)溶解しない共重合体。
また、当業者は、表1の記載の各構成単位の構成を適宜変更することにより、様々な構成単位を含む共重合体を得ることができる。
実施例に用いる共重合体の合成のため、表1に示す9種の共重合体、すなわち本発明の範囲内である共重合体1-1~2-3を合成した。
2-メタクリロイルオキシエチルホスホリルコリン(以下、MPCと記載)19.3g、ポリジメチルシロキサンモノメタクリレート(信越化学工業株式会社製X-22-174AS-X、以下、PDMSMAと記載)1.0g、4-メタクリロイルオキシベンゾフェノン(以下、MBPと記載)2.0g(モノマー組成モル比:MPC/PDMSMA/MBP=89/1/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるn-プロパノール(以下、NPAと記載)88.9gを加えて溶解させた。反応容器内を十分に窒素置換した後、60℃に昇温した。重合開始剤として2,2’-アゾビス(2,4-ジメチルバレロニトリル)(以下、ADVNと記載)55mgを加え、そのまま一晩60℃に保つことで重合反応を行った。得られた反応液を大過剰のアセトンに滴下することで重合体を沈殿させた。沈殿を濾別し、アセトンで洗浄した後、真空乾燥して白色固体の共重合体1-1を得た。
得られた共重合体1-1の重量平均分子量は、ゲル濾過クロマトグラフィー(以下、GPCと記載)測定により、ポリエチレングリコール(以下、PEGと記載)換算で170,000と決定された。
MPC 21.0g、PDMSMA 0.4g、MBP 2.1g(モノマー組成モル比:MPC/PDMSMA/MBP=89.5/0.5/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 94.1gを加えて溶解させた。反応容器内を十分に窒素置換した後、60℃に昇温した。重合開始剤としてADVN 58mgを加え、その後は合成例1-1と同様にして重合と回収を行い、白色固体の共重合体1-2を得た。
得られた共重合体1-2の重量平均分子量は、GPC測定により、PEG換算で105,000と決定された。
MPC 21.0g、PDMSMA 0.4g、MBP 2.1g(モノマー組成モル比:MPC/PDMSMA/MBP=89.5/0.5/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 35.3gを加えて溶解させた。反応容器内を十分に窒素置換した後、重合開始剤としてADVN 6mgを加え、ゆっくりと60℃に昇温した。そのまま一晩60℃に保つことで重合反応を行った。その後は合成例1-1と同様に回収を行い、白色固体の共重合体1-3を得た。
得られた共重合体1-3の重量平均分子量は、GPC測定により、PEG換算で951,000と決定された。
MPC 18.4g、トリス(トリメチルシロキシ)シリルプロピルメタクリレート(以下、TRISと記載) 1.9g、MBP 2.0g(モノマー組成モル比:MPC/TRIS/MBP=84/6/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 94.1gを加えて溶解させた。反応容器内を十分に窒素置換した後、60℃に昇温した。重合開始剤としてADVN 58mgを加え、その後は合成例1-1と同様に重合と回収を行い、白色固体の共重合体1-4を得た。
得られた共重合体1-4の重量平均分子量は、GPC測定により、PEG換算で72,000と決定された。
MPC 20.2g、4-(2-ヒドロキシエチル)-2-メチレン-1-(トリス(トリメチルシロキシ)シリルプロピル)ブタン-1,4-ジオエート(以下、ETSと記載)2.5g、4-(4-アジドベンゾイルオキシメチル)ビニルベンゼン(以下AzStと記載)2.3g(モノマー組成モル比:MPC/ETS/AzSt=84/6/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 100.0gを加えて溶解させた。反応容器内を十分に窒素置換した後、重合開始剤としてADVN 62mgを加え、その後は合成例1-3と同様に重合と回収を行い、白色固体の共重合体1-5を得た。
得られた共重合体1-5の重量平均分子量は、GPC測定により、PEG換算で70,000と決定された。
MPC 18.4g、3-(3-(メチルビス(トリメチルシロキシ)シリル)プロピル)グリセロール-1-メタクリレート(以下、SiGMAと記載) 1.9g、MBP 2.0g(モノマー組成モル比:MPC/SiGMA/MBP=84/6/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 88.9gを加えて溶解させた。反応容器内を十分に窒素置換した後、重合開始剤としてADVN 55mgを加え、その後は合成例1-3と同様に重合と回収を行い、白色固体の共重合体1-6を得た。
得られた共重合体1-6の重量平均分子量は、GPC測定により、PEG換算で74,000と決定された。
MPC 20.7g、PDMSMA 0.4g、グリシジルメタクリレート(GMA) 1.1g(モノマー組成モル比:MPC/PDMSMA/GMA=89.5/0.5/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 88.9gを加えて溶解させた。反応容器内を十分に窒素置換した後、重合開始剤としてADVN 6mgを加え、その後は合成例1-3と同様に重合と回収を行い、白色固体の共重合体2-1を得た。
得られた共重合体2-1の重量平均分子量は、GPC測定により、PEG換算で371,000と決定された。
MPC 19.0g、TRIS 2.1g、GMA 1.1g(モノマー組成モル比:MPC/TRIS/GMA=84/6/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 88.9gを加えて溶解させた。反応容器内を十分に窒素置換した後、重合開始剤としてADVN 55mgを加え、その後は合成例1-3と同様に重合と回収を行い、白色固体の共重合体2-2を得た。
得られた共重合体2-2の重量平均分子量は、GPC測定により、PEG換算で72,000と決定された。
MPC 20.5g、ETS 2.3g、GMA 1.2g、ヒドロキシエチルアクリルアミド(以下、HEAAと記載)1.1g(モノマー組成モル比:MPC/ETS/GMA/HEAA=76/5/9/10)を重合用ガラス製フラスコに秤量し、重合溶媒であるNPA 100.0gを加えて溶解させた。反応容器内を十分に窒素置換した後、重合開始剤としてADVN 62mgを加え、その後は合成例1-3と同様に重合と回収を行い、白色固体の共重合体2-3を得た。
得られた共重合体2-3の重量平均分子量は、GPC測定により、PEG換算で71,000と決定された。
以上合成例1-1~2-3の各共重合体のGPC測定は以下の条件で実施した。
GPCシステム:EcoSECシステム(東ソー株式会社製)
カラム:Shodex OHpak SB-802.5HQ(昭和電工株式会社製)、及びSB-806HQ(昭和電工株式会社製)を直列に接続
展開溶媒:20mMりん酸ナトリウム緩衝液(pH 7.4)
検出器:示差屈折率検出器
分子量標準:EasiVial PEG/PEO(Agilent Technologies社製)
流速:0.5mL/分
カラム温度:40℃
サンプル:得られた共重合体を終濃度0.1重量%となるよう展開溶媒で希釈
注入量:100μL
前記合成例1-1~2-3に記載の各共重合体の水溶性の判定は以下の手順で実施した。
(1)30ccガラスバイアルに、各共重合体を下記の所定濃度とすべく適量秤量した。
(2)10mLのイオン交換水を添加した。
(3)20℃で攪拌し、目視で室温の水への溶解性を確認した。
(4)60℃の恒温槽で1時間インキュベートした後攪拌し、室温に戻してから目視で熱水への溶解性を確認した。
(5)上記(1)~(4)を、0.1%(w/v)、0.5%(w/v)、1.0%(w/v)、5.0%(w/v)の各濃度について行った。
(6)室温(20℃)の水、熱水(60℃)のそれぞれについて、上記4濃度のうち、溶解することが確認された最大の濃度を記録した。ただし、0.1%(w/v)でも溶解しなかったものについては、「×」と記録した。

室温(20℃)、熱水(60℃)のそれぞれについて、各共重合体が溶解した最大の濃度を記載(単位:%(w/v)。ただし、0.1%(w/v)溶解しなかったものについては「×」と記載。
実施例、比較例に用いる各シリコーンハイドロゲルは、以下の工程1~6、及びA、Bの方法で調製した。
各モノマーを表2に示す各配合比で混合し、モノマー組成物を調製した。この際、MC4ではMPC、NPA、ヒドロキシエチルメタクリレート(以下、HEMAと記載)、を所定量秤量して混和させたあと、N-ビニルピロリドン(以下、NVPと記載)、テトラエチレングリコールジメタクリレート(以下、TEGDMAと記載)、トリエチレングリコールジビニルエーテル(以下、TEGDVと記載)、MC2ではメタクリル酸(以下、MAと記載)、MC3ではアミノエチルメタクリレート(以下、AeMAと記載)を所定量添加して再び混和させ、ETSを添加してまた再び混和させ、PDMSMAを添加してさらに再び混和させ、最後に反応開始剤としてAIBNを添加することで調製した。
前記のモノマー組成物をコンタクトレンズのモデルとして円盤状に成形すべく、内寸φ1.1cm×0.1mmのポリプロピレン製の鋳型に注入し、オーブン内に静置した。オーブン内を窒素置換した後、ゆっくりと80℃に昇温し、そのまま80℃で12時間保温することで熱硬化を行い、硬化物を得た。
工程2で調製した硬化物を鋳型から取り出し、硬化物1枚につき40gのiso-プロパノール(以下、IPAと記載)に4時間浸漬することで、不純物等を除去する精製を行った。
前記工程3を経た硬化物を、50gのイオン交換水(又は、工程Aを兼ねる場合はイオン交換水(純水)を溶媒とした処理液)に4時間浸漬して、不純物未反応物等を抽出除去する精製を行った。
前記工程4を経た硬化物を取り出してISO-18369-3に記載の生理食塩水(以下、PBSと記載)(又は、工程Aを兼ねる場合はPBSを溶媒とした処理液)に4時間浸漬して水和、膨潤させた、
前記工程5を経た硬化物を新鮮なPBSに浸漬して密栓し、121℃、20分オートクレーブすることにより滅菌した。

AIBN:2,2’-アゾビス(イソブチロニトリル)
[処理液の調製]
表4に示す組成の各処理液を調製した。
処理液の調製は、本発明の各共重合体1-1~2-3を表4に示した所定の溶媒に所定濃度溶解させることで行った。この際、各共重合体の溶解性に応じて適宜加熱した。
[工程A]
各モノマー組成物を硬化させて得られた硬化物を、調製した処理液に浸漬し、室温で所定時間静置した。
光照射による工程Bは、工程Aを経た硬化物に対し、波長365nm、または405nm、照度1.5mW/cm2の光を20分間照射することで行った。この際、硬化物は処理液に浸漬したままとした。
工程Bがシリコーンハイドロゲルの調製における最後の工程となる場合は、光照射の後、得られた表面処理済み硬化物を10mLの新鮮なPBSで3回リンスした。
加熱による工程Bは、工程Aを経た硬化物を新鮮なPBSに浸漬し、密栓して121℃で20分間オートクレーブすることにより行った。
工程1~6及び工程A、Bの実施順序は、表3に示す各スキームの通りとした。
工程1:モノマー組成物の調製
工程2:モノマー組成物の硬化
工程3:アルコール抽出精製
工程4:水抽出精製
工程5:生理食塩水置換
工程6:高圧蒸気滅菌
工程A:本発明の共重合体の溶液への硬化物の浸漬(比較例の場合、溶媒のみへの硬化物の浸漬)
工程B:硬化物の加熱処理又は光照射処理により本発明の共重合体を硬化物表面に固定化する工程(比較例の場合、硬化物の加熱処理又は光照射処理する工程)
=:並列された工程は、2つの工程を兼ねて1つの工程として実施したことを示す。
以上の手順により得られた表面処理済みの硬化物を、以下の方法で評価した。
調製したシリコーンハイドロゲルの透明性を目視により以下の基準で判定した。
++:透明
+:微濁
×:白濁
調製したシリコーンハイドロゲルの形状について、目視により以下の基準で判定した。
++:歪みを認めない
+:やや歪みを認める
×:歪み、硬化不良を認める
調製したシリコーンハイドロゲルの表面濡れ性について、以下の手順で評価した。
(1)PBSに漬かっている調製したシリコーンハイドロゲルを、ピンセットを用いて空中に取り出した。
(2)シリコーンハイドロゲルの表面を目視で観察し、取り出してから表面の水膜が切れ始めるまでの時間[秒](BUT)を測定した。すなわち、BUTが長いほど、表面濡れ性が高いものとして評価した。
(3)以下の基準に従って判定した。
+++:30秒≦BUT
++:10秒≦BUT<30秒
+:5秒≦BUT<10秒
×:BUT<5秒
調製したシリコーンハイドロゲルの表面滑り性について、以下の手順で評価した。
(1)シリコーンハイドロゲルレンズを親指と中指でつまんだ。
(2)官能評価により、Polymaconの滑り性を2点、Omafilcon Aの滑り性を8点として0~10点の評点を付した。すなわち、評点が高いほど、滑り性が高いことを示す。
(3)以下の基準に従って判定した。
+++:7点~10点
++:5点~6点
+:3~4点
×:0~2点
本発明の共重合体として、構成単量体がMPC、PDMSMA及びMBPである共重合体1-1~1-3を用いて、硬化物の表面処理を行った。ここでは、工程1~6を経た硬化物を処理液に室温で1時間浸漬し(工程A)、直後に波長365nm、照度1.5mW/cm2の光を照射することで表面処理を行った(工程B)。最後に、評価を行った。
用いた本発明の共重合体として、共重合体1-4~1-6を用いて、実施例1-1~1-5と同様の表面処理及び評価を行った。
工程Bにおいて照射する光の波長を405nmとし、共重合体1-4を用いて、表面処理を行った場合について検討した。
工程1~3を経た硬化物について、工程4と工程Aを兼ねて、本発明の共重合体を1重量%添加した純水を用いて水抽出精製を行い、直後に波長365nm、照度1.5mW/cm2の光を照射した後、工程5~6を行い、評価を行った。
工程1~4を経た硬化物について、工程5と工程Aを兼ねて本発明の共重合体を0.5重量%添加したPBSを用いて生理食塩水置換を行い、この硬化物を新鮮なPBSに移して、工程6と工程Bを兼ねて高圧蒸気滅菌した場合について検討した。

本発明の共重合体を用いない対照試験として、表5に示す6種の表面処理済み硬化物を実施例と同様の方法で調製し、実施例と同様の方法で評価した。
本発明の共重合体を用いた表面処理を実施せず、MPCモノマーをモノマー組成物に添加する方法を採用した場合で評価した。
以上、本発明の共重合体を用いた表面処理は、特定の相補性官能基を提示せずとも、シリコーンハイドロゲルの表面に本発明の共重合体を固定化させることにより、該シリコーンハイドロゲルの濡れ性、滑り性を好適に改善できる。
Claims (8)
- 下記の式(1)で表される親水性単量体aに基づく構成単位、下記の式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位並びに熱反応性単量体もしくは光反応性単量体cに基づく構成単位を含有する共重合体であって、60℃の熱水に0.5%(w/v)溶解するが、20℃の水に1%(w/v)溶解しない共重合体を含む眼科デバイス用表面処理剤。
- 前記熱反応性単量体もしくは光反応性単量体が、光反応性単量体である、請求項1に記載の眼科デバイス用表面処理剤。
- 前記式(1)で表される親水性単量体aに基づく構成単位が2-メタクリロイルオキシエチルホスホリルコリンであり、前記式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位がポリジメチルシロキサンモノメタクリレート、トリス(トリメチルシロキシ)シリルプロピルメタクリレート、4-(2-ヒドロキシエチル)-2-メチレン-1-(トリス(トリメチルシロキシ)シリルプロピル)ブタン-1,4-ジオエート若しくは3-(3-(メチルビス(トリメチルシロキシ)シリル)プロピル)グリセロール-1-メタクリレート、並びに、熱反応性単量体もしくは光反応性単量体cに基づく構成単位がメタクリロイルオキシベンゾフェノン、4-(4-アジドベンゾイルオキシメチル)ビニルベンゼン若しくはグリシジルメタクリレートである、請求項1に記載の眼科デバイス用表面処理剤。
- 前記式(1)で表される親水性単量体aに基づく構成単位、前記式(2)または式(3)で表されるシリコーン単量体bに基づく構成単位、及び/又は、熱反応性単量体もしくは光反応性単量体cに基づく構成単位の組み合わせは、以下のいずれか1から選択される、請求項1に記載の眼科デバイス用表面処理剤。
1)2-メタクリロイルオキシエチルホスホリルコリン、ポリジメチルシロキサンモノメタクリレート及びメタクリロイルオキシベンゾフェノン
2)2-メタクリロイルオキシエチルホスホリルコリン、トリス(トリメチルシロキシ)シリルプロピルメタクリレート及びメタクリロイルオキシベンゾフェノン
3)2-メタクリロイルオキシエチルホスホリルコリン、4-(2-ヒドロキシエチル)-2-メチレン-1-(トリス(トリメチルシロキシ)シリルプロピル)ブタン-1,4-ジオエート及び4-(4-アジドベンゾイルオキシメチル)ビニルベンゼン
4)2-メタクリロイルオキシエチルホスホリルコリン、3-(3-(メチルビス(トリメチルシロキシ)シリル)プロピル)グリセロール-1-メタクリレート及びメタクリロイルオキシベンゾフェノン
5)2-メタクリロイルオキシエチルホスホリルコリン、ポリジメチルシロキサンモノメタクリレート及びグリシジルメタクリレート

- 請求項1~4のいずれか1に記載の共重合体で表面を処理した表面処理済シリコーンハイドロゲル。
- 前記共重合体は請求項2に記載の共重体であり、前記表面処理が光照射である、請求項5に記載の表面処理済シリコーンハイドロゲル。
- 請求項5又は6に記載の表面処理済シリコーンハイドロゲルを用いた眼科デバイス。
- ソフトコンタクトレンズである請求項7に記載の眼科デバイス。
