Description
본 출원은 친수성 아미노산, 소수성 아미노산, 및 지방산을 단량체로 포함하는, pH 감응성 고분자, 이를 포함하는 코팅 조성물 및 상기 코팅 조성물로 코팅된 반추위 내 분해 보호 코팅된 사료 첨가제에 관한 것이다.
모든 동물, 특히 가축은 자라는 데 적당한 영양소를 필요로 한다. 아미노산은 단백질을 구성하는 유기 화합물로서, 아미노산과 단백질은 생명체를 이루는 빌딩블록이다. 단백질은 소화되거나 분해되어 아미노산을 생성한다. 생명체는 음식물의 분해, 성장, 체조직의 복구 및 많은 다른 기능을 수행하기 위해 아미노산을 소모하여 단백질을 생성한다. 또한 아미노산은 에너지원으로도 사용될 수 있다. 이러한 아미노산은 필수아미노산(essential amino acids), 비필수아미노산(nonessential amino acids) 및 제한아미노산(conditional amino acids)으로 구분된다. 이중 필수아미노산은 총 9종으로 '히스티딘(histidine)', '라이신(lysine)', '트립토판(tryptophan)', '메치오닌(methionine)', '트레오닌(threonine)', '발린(valine)', '루신(leucine)', '이소루신(isoleucine)', '페닐알라닌(phenylalanine)'을 포함한다. 이러한 필수아미노산은 체내에서 합성되지 않아 음식물을 통해 섭취해야만 한다. 따라서, 이들 필수아미노산을 발효 등의 방법으로 체외 합성하여 사료나 식품의 첨가제의 형태로 제공하기도 한다.

본 출원인은 아미노산 등의 유효성분이 반추위 내에서 분해되는 것을 방지하기 위한 코팅 조성물을 발굴하기 위하여 예의 연구 노력한 결과, 친수성 아미노산, 소수성 아미노산, 지방산 및 선택적으로 사슬연장제를 소정의 몰비율로 포함하는 아미노산 고분자가 중성의 pH에서는 용해되지 않고, pH 2 근처의 산성 용액에서 선택적으로 용해되는 pH 감응성을 나타내므로 이를 포함하는 코팅 조성물로 유효성분 입자를 코팅함으로써 반추위 내 분해로부터 보호된 사료 첨가제를 제공할 수 있음을 확인하고, 본 출원을 완성하였다.
본 출원의 하나의 목적은 친수성 아미노산, 소수성 아미노산, 및 지방산을 단량체로 포함하는, pH 감응성 고분자를 제공하는 것이다.
본 출원의 다른 하나의 목적은 상기 pH 감응성 고분자, 지방산, 및 셀룰로오스 또는 이의 유도체를 포함하는, 반추위 내 분해 보호용 코팅 조성물을 제공하는 것이다.

본 출원의 pH 감응성 아미노산 고분자는 친수성 아미노산, 소수성 아미노산 및 지방산을 소정의 비율로 포함함으로써 중성 조건에서는 거의 용해되지 않고 pH 2 정도의 강산 조건에서 용해도가 급격히 증가하여 선택적으로 분해되므로 반추동물용 사료 첨가제의 반추위 보호 코팅용 소재로 유용하게 사용될 수 있다.

본 출원의 용어, "셀룰로오스"는 자연계에 존재하는 가장 풍부한 다당류(polysaccharide)로서, 일명 글리코시드 결합(glycosidic bond)이라 불리는 에테르기에 의해 서로 공유결합적으로 연결된 6원 에테르 고리(D-글루코스 또는 덱스트로스)로 구성된 선형 고분자이다. 보통 수천개의 글루코스 반복 단위로 구성되며, 가수분해에 의해 글루코스 분자를 생성하므로 셀룰로오스 및 이의 유도체는 축합 고분자(condensation polymers)로 간주될 수 있다. 셀룰로오스의 유도체는 상기 셀룰로오스를 구성하는 단량체인 글루코스에 존재하는 히드록실기의 일부 또는 전부에 수소를 대신하여 치환기가 도입된 물질을 의미하는 것으로, 상기 치환기가 각각 알킬카보닐, 알킬, 및 니트로인 에스테르 셀룰로오스, 에테르 셀룰로오스, 니트로셀룰로오스(셀룰로오스 니트레이트, 셀룰로이드) 등의 셀룰로오스 유도체가 상용되고 있다. 예컨대, 본 출원의 코팅 조성물에 포함되는 셀룰로오스 또는 이의 유도체는 셀룰로오스 자체와 셀룰로오스의 에테르 유도체인 에틸셀룰로오스 및 메틸셀룰로오스로 구성된 군으로부터 선택되는 1종 이상일 수 있으나, 이에 제한되지 않는다.
본 출원의 용어, "반추위(ruminant stomach)"는 포유류 소목의 동물이 가진 위로, 되새김위라고도 한다. 초식동물은 주로 거친 사료를 섭취하므로 소화기관의 일부가 특히 발달되어 있다. 예컨대, 말이나 토끼는 맹장과 결장이 발달되어 있으나, 소와 염소는 위가 특히 발달하여 반추위가 형성되고 이는 전 소화기관의 약 70%를 차지한다. 그 부피도 커서 단위동물인 돼지나 말의 위 부피가 6 내지 15 L인데 비해 소는 200L에 달한다. 상기 반추위는 제1위인 혹위, 제2위인 벌집위, 제3위인 겹주름위 및 제4위인 주름위 등의 네 부분으로 나누어져 있는데, 혹위와 벌집위는 식도가 변형된 것이고, 겹주름위와 주름위가 본래의 위에 해당한다. 상기 혹위는 특히 커서 전체 위의 80%를 차지하며, 안쪽 면에 무수한 혹이 있다. 한편, 벌집위의 점막은 마치 벌집처럼 주름져 있으며, 겹주름위에는 많은 혹과 주름이 있고, 주름위에는 큰 주름이 있어, 염소는 13 내지 15개, 소는 16개 이다. 이들 네 부분은 서로 통해 있어, 반추위의 소화과정은 거칠게 씹힌 먹이가 혹위를 거쳐 벌집위로 보내지면 여기서 먹이가 뭉쳐져 다시 입으로 보내지고, 이는 되씹기 과정을 거친 후 겹주름위로 들어가 잘게 부숴지고 주름위에서 소화된다. 이때, 소화액을 분비하는 위는 제4위인 주름위뿐이나, 혹위에서는 미생물의 활동으로 거친 섬유질을 발효시킨다.
본 출원의 또 다른 하나의 양태는 유효성분을 포함하는 코어 입자; 및 상기 코팅 조성물을 이용하여 상기 코어 입자 상에 형성된 코팅층을 포함하는, 반추위 내 분해 보호 코팅된 사료 첨가제를 제공한다.
본 출원의 사료 첨가제에 사용되는 유효성분은 아미노산, 효소, 프로바이오틱스, 프리바이오틱스, 박테리오파지, 비타민, 유기산 또는 이들의 혼합물일 수 있으나, 이에 제한되지 않는다.
나아가, 본 출원의 사료 첨가제는 반추동물일 수 있으나, 이에 제한되지 않는다.
본 출원의 용어, "반추동물(ruminants)"은 반추위가 있고 반추작용을 하는 동물을 일컫는 말로, 되새김 동물이라고도 하는 초식성 포유류이다. 포유류 소목 중 낙타과, 애기사슴과, 사슴과, 기린과 및 소과의 동물이 이에 속한다. 이들 반추동물의 소화작용은 반추작용을 한다는 점과 미생물에 의한 먹이 분해가 이루어 진다는 2가지 특징을 가진다. 단위(單胃)동물은 먹이를 섭취할 때 침과 섞어 잘게 부수어 삼키지만 반추동물은 앞니가 없어 혀와 입술로 거칠게 씹어 삼킨 후 되새김질하여 소화시킨다. 이러한 반추작용은 주로 밤에 행해지며, 이는 반추동물이 육식동물의 먹이가 되는 경우가 많으므로 주어진 시간 안에 많은 양의 먹이를 먹어 저장한 후 안전한 장소에서 천천히 소화시키기 위한 것으로 여겨진다. 전술한 바와 같이, 반추동물의 제1위, 즉 혹위에서는 여러 종류의 미생물이 있어 먹이를 발효시키고 이들 미생물은 먹이와 함께 제4위에서 분해된 후 소화 흡수되어 반추동물에 필요한 영양물질이 된다.
본 출원의 사료 첨가제에 있어서, 상기 코팅층은 pH 6 내지 8의 반추위에서는 분해되지 않고, pH 3 이하의 제4위에서 선택적으로 분해되어 유효성분을 방출할 수 있는 것이 특징이다.
다른 동물에서와 마찬가지로 반추동물의 경우에도 성장을 촉진하거나 기능성을 부여하기 위하여 사료 이외에 필수 아미노산과 같은 영양소를 추가로 공급할 필요가 있는데, 특수하게 처리되지 않은 성분 자체로서 공급하는 경우 혹위에서 미생물에 의해 분해되는 등 일련의 반추작용을 거치면서 상당량이 분해되어 소화 효소가 작용하는 제4위 또는 영양분을 흡수하는 소장까지 전달되지 못하고 소실되기 쉽다. 따라서 혹위에서와 같은 중성 조건에서는 분해되지 않고 제4위와 유사한 강산성 조건에서 선택적으로 분해되어 유효성분을 용출시킬 수 있는 본 출원에 따른 pH 감응성 고분자를 포함하는 코팅 조성물로 유효성분을 코팅한 사료 첨가제는 반추동물에 아미노산을 공급하기 위한 제형으로 적합할 수 있다.
이하, 하기 실시예에 의하여 본 출원을 더욱 상세하게 설명하고자 한다. 단, 하기 실시예는 본 출원을 예시하기 위한 것일 뿐 본 출원의 범위가 이들만으로 한정되는 것은 아니다.
실험예 1: pH 감응성 아미노산 고분자의 제조
실험예 1-1: pH 감응성 아미노산 고분자의 제조
친수성 아미노산, 및 소수성 아미노산을 단량체로 사용하여 열 축합 중합에 의해 pH 감응성 아미노산 고분자를 제조하였다. 구체적으로, 교반기와 콘덴서를 구비한 반응기에 친수성 아미노산 및 소수성 아미노산을 첨가한 후, 185℃까지 승온시키며 교반하여 반응시켰다. 이때 생성되는 응축수는 콘덴서를 통해 제거하였다. 응측수가 더 이상 발생하지 않는 시점에서 감압하여 진공 상태에서 2 내지 3시간 동안 교반하여 반응시키고, 반응 종료 후 내용물을 토출하여 표제 고분자를 수득하였다.
실험예 1-2: 지방산을 도입한 pH 감응성 아미노산 고분자의 제조
친수성 아미노산, 소수성 아미노산, 및 지방산을 단량체로 사용하여 열 축합 중합에 의해 지방산을 도입한 pH 감응성 아미노산 고분자를 제조하였다. 구체적으로, 상기 실험예 1-1에서와 유사하게, 친수성 아미노산 및 소수성 아미노산을 반응시키면서 생성되는 응축수는 콘덴서를 통해 제거하고, 응측수가 더 이상 발생하지 않는 시점에서 상기 반응기에 지방산을 추가한 후 감압하여 진공 상태에서 2 내지 3시간 동안 교반하여 반응시켰다. 반응 종료 후 내용물을 토출하여 표제 고분자를 수득하였다.
실험예 2: 반추위 내 분해 보호 코팅된 메치오닌 제형의 제조
실험예 2-1: 메치오닌 코어의 제조
유효성분인 메치오닌과 결합제인 폴리라이신을 물과 배합하고, 이를 이중 스크류가 장착된 압출성형기(L/D=15, 스크류 지름 100 mm)로 압출하였다. 압출기의 온도는 상온으로 설정하였으며, 0.1 ton/d의 속도로 유지하였다. 압출물은 통상의 구형화기(Spheronizer)를 사용하여 2.3 내지 2.8 mm의 입경을 갖는 구형 또는 원통형으로 메치오닌 코어를 제조하였다.
실험예 2-2: 반추위 내 분해 보호 코팅
상기 실험예 2-1 또는 2-2에 따라 준비한 지방산을 비도입 또는 도입한 pH 감응성 아미노산 고분자, 에틸셀룰로오스 및 스테아르산을 65℃ 에탄올 수용액에 용해시켜 코팅 조성물을 준비하였다. 상기 실험예 2-1에 따라 준비한 메치오닌 코어를 원통형의 팬 코팅기에 넣고 4 mL/min의 속도로 미리 준비한 코팅 조성물을 분사하였다. 코팅이 완료된 제형은 상온에서 건조시켰다.
실험예 3: 반추위 내 분해 보호 코팅된 히스티딘 제형의 제조
실험예 3-1: 히스티딘 코어의 제조
유효성분으로서 메치오닌 대신에 염산염 또는 프리폼 형태의 히스티딘을 사용하고, 부형제로서 탈지대두분과 스테아르산을 추가로 포함하는 것을 제외하고는 상기 실험예 2-1과 동일한 방법으로 구형 또는 원통형의 히스티딘 코어를 제조하였다.
실험예 3-2: 반추위 내 분해 보호 코팅
메치오닌 코어 대신에 상기 실험예 3-1에 따라 준비한 히스티딘 코어를 사용하는 것을 제외하고는 상기 실험예 2-2와 동일한 방법으로 pH 감응성 아미노산 고분자를 코팅하였다.
실험예 4: 생체이용률 평가
실험예 4-1: in situ 반추위 bypass 평가
상기 실험예 2 및 3에 따라 준비한 보호 코팅된 제형을 나일론백(Ankom사 R510)에 담아 밀봉하여 준비하고, 이를 반추위 캐뉼라 장착 홀스테인 거세우(체중 630 내지 650 kg 내외) 3두의 반추위에서 배양하였다. 실험 종료 후 나일론백을 물로 세척하여 건조하고, DM 및 유효성분 함량을 측정하여 상대적인 잔존량을 비교하였다.
실험예 4-2: in vitro 소화분해 평가
Phosphate buffer 조건에서 펩신, 판크레아틴 및 리파아제를 순차적으로 처리하여 반추위와 소장을 모사하였다. 구체적으로, 0.1 M KH2PO4 buffer에 1 M HCl을 첨가하여 pH 2로 조정한 후 25 mg/mL 펩신 용액을 첨가하였다. 반추위 bypass 평가를 마친 시료를 담아 밀봉한 나일론백(Ankom사 F57)을 넣어 혼합한 후 밀폐하여 39℃ 인큐베이터(Ankom사 DaisyII)에서 배양하였다. 2시간 후 0.2 M KH2PO4 0.5 L를 첨가한 후, 1 M NaOH로 pH 7.8로 조정한 후, 10 mg/mL 판크레아틴 용액을 첨가하여 다시 39℃에서 배양하였다. 실험 종료 후 나일론백을 물로 세척하여 건조하고, DM 및 유효성분 함량을 측정하였다.
실험예 4-3: 생체이용률 계산
반추위 bypass율은 실험 전 시료 대비 반추위 bypass 평가 후 남아있는 잔존 시료의 비율로 계산하였으며, 소화분해율은 소화분해 평가 전 대비 손실된 유효성분의 비율로 계산하였다. 이들 수치로부터 생체이용률을 다음의 방정식으로 계산하였다:
실시예 1: 사슬연장제를 추가로 포함하는 pH 감응성 아미노산 고분자의 물성 분석
상기 실험예 1-1에 따라 준비하되, 사슬연장제를 추가로 포함하여 제조한 pH 감응성 아미노산 고분자의 물성을 측정하여 하기 표 1에 나타내었다. 이때, 친수성 아미노산으로는 라이신을, 소수성 아미노산으로는 페닐알라닌과 트립토판을, 사슬연장제로는 글루탐산을 하기 표 1에 개시한 비율로 사용하여 5종의 고분자를 합성하였다(고분자 A 내지 E).
구체적으로, RID(refractive index detector)를 구비한 겔투과 크로마토그래피(gel permeation chromatography; GPC, Waters)를 이용하여 중량평균분자량과 분자량분포를 분석하였다. 이때, 표준물질로는 폴리스티렌을 사용하였다. 한편, 시차주사열량계(differential scanning calorimetry; DSC, TA Instruments Q20)를 사용하여 질소 분위기 하에 -20℃로부터 280℃까지 온도에 따른 열용량을 측정하여, 용융온도(Tm), 결정화온도(Tc) 및 유리전이온도(Tg)를 산출하였다. 나아가, pH 2 및 pH 7 조건의 0.1 M phosphate buffer(KH2PO4)에 20 g/L 농도로 침지시켜 경시 변화를 관찰하여 pH 감응성을 확인하였다. 각 결과는 하게 표 1에 나타내었으며, 육안으로 확인한 pH에 따른 경시 변화는 도 1에 나타내었다.

상기 표 1에 나타난 바와 같이, 친수성 아미노산, 소수성 아미노산 및 사슬연장제의 비율을 달리하여 준비한 pH 감응성 아미노산 고분자의 미세구조를 시주차열량계로 측정하여, 비결정 구조를 갖는 것을 확인하였다. 경시 변화를 관찰하여 pH 감응성 아미노산 고분자의 용해도를 비교하였을 때, 친수성 아미노산인 라이신 함량이 57몰% 이상으로 높은 경우, 및/또는 사슬연장제를 포함하는 경우 보다 강한 pH 감응성이 발현되는 것으로 나타났다.
실시예 2: 사슬연장제를 추가로 포함하는 pH 감응성 아미노산 고분자의 물성 분석
상기 실시예 1과 유사하게, 사슬연장제를 추가로 포함하여 제조한 pH 감응성 아미노산 고분자의 물성을 측정하여 하기 표 2에 나타내었다. 이때, 친수성 아미노산으로는 라이신을, 소수성 아미노산으로는 페닐알라닌 또는 트립토판을 단독으로, 사슬연장제로는 글루탐산을 하기 표 2에 개시한 비율로 사용하여 4종의 고분자를 합성하였다(고분자 F 내지 I).

아울러, 상기 고분자로부터 선택된 고분자 G 및 H의 경시 변화를 관찰하여 pH에 따른 용해 정도를 확인하고, 그 결과를 하기 표 3 및 도 2에 나타내었다.

표 3 및 도 2에 나타난 바와 같이, 조절된 비율로 친수성 아미노산, 소수성 아미노산 및 사슬연장제를 포함하여 제조한 고분자의 경우, 소수성 아미노산의 종류에 무관하게 모두 강한 pH 감응성을 발현하는 것으로 나타났다.
실시예 3: 지방산을 도입한 pH 감응성 아미노산 고분자의 물성 분석
상기 실험예 1-2에 따라 준비하되, 사슬연장제를 추가로 포함하여 제조한 지방산을 도입한 pH 감응성 아미노산 고분자의 물성을 측정하여 하기 표 4에 나타내었다. 이때, 친수성 아미노산으로는 라이신을, 소수성 아미노산으로는 페닐알라닌 및/또는 트립토판을, 사슬연장제로는 글루탐산을, 지방산으로는 스테아르산을 하기 표 4에 개시한 비율로 사용하여 5종의 고분자를 합성하였다(고분자 J 내지 N).
구체적으로, 상기 실시예 1에서와 마찬가지로 중량평균분자량, 분자량분포, 용융온도(Tm), 및 유리전이온도(Tg)를 산출하였다. 나아가, pH 2 및 pH 7 조건의 용액에서 경시 변화를 관찰하여 pH 감응성을 확인하였다. 각 결과는 하게 표 8에 나타내었다.

표 4에 나타난 바와 같이, 순수 지방산에서 나타나는 용융 온도는 이를 단량체로 포함하는 지방산을 도입한 pH 감응성 아미노산 고분자에서는 관찰되지 않았으며, 상기 지방산이 고분자 사슬 내에 공유결합의 형태로 존재함을 나타내는 것이다.
상기 고분자 J, K, L, 및 N을 코팅 조성물에 사용하여, 코팅과 유효성분의 비율을 변화시키면서, 실험예 2에 따라 준비한 반추위 내 분해 보호 코팅된 메치오닌 제형(이하 제형 a 및 d로 명명)에 대해 실험예 4에 따른 생체이용률 평가를 수행하고, 그 결과를 하기 표 5에 나타내었다. in situ 반추위 bypass 평가는 48시간 동안 배양하여 수행하였으며, 모든 실험 결과는 DM과 유효성분 함량의 곱을 기준으로 계산하였다.

표 5에 나타난 바와 같이, 유사한 조성의 pH 감응성 아미노산 고분자에서 지방산을 포함하지 않는 것에 비해 지방산을 도입한 고분자로 코팅한 경우 bypass율이 현저히 향상되어 소화분해율이 다소 낮더라도 개선된 생체이용률을 기대할 수 있었다.
실시예 4: 지방산의 종류에 따른 pH 민감성 평가
상기 실험예 1-2에 따라 준비하되, 사슬연장제인 글루탐산을 추가로 포함하고, 각각 2종의 상이한 지방산을 사용하여 중합시켜 지방산을 도입한 pH 감응성 아미노산 고분자를 제조하였다. 이때, 각 단량체의 비율은 고분자 K와 동일하게 조절하였으며, 상기 지방산으로는 C18의 스테아르산과 C12의 라우르산을 사용하였다. 상기와 같이 준비한 고분자들의 용해도를 상이한 pH 조건에서 측정하여 비교함으로써 pH 감응성을 확인하였다. 구체적으로, 0.1 M KH2PO4 용액에 염산 및 수산화나트륨으로 각각 pH를 2와 7이 되도록 보정한 buffer에 일정한 농도로 고분자를 용해시키고 시간에 따라 상등액을 취하여 1000배 희석하고, 200 nm 파장에서의 흡광도를 UV/Vis 분광계로 측정하였다. 상기 각각의 pH 조건에서 시간 경과에 따라 측정한 흡광도를 하기 표 6에 나타내었다.

상기 표 6에 나타난 바와 같이, 상대적으로 탄소수가 높은 스테아르산을 포함하는 고분자에 비해 라우르산을 포함하는 고분자에서 pH 2에서의 흡광도는 더 높고 pH 7에서의 흡광도는 더 낮았다. 그러나, 정도의 차이는 있으나, 지방산의 종류와 무관하게 상기 2종 고분자 모두 pH 7의 중성 조건에서는 거의 용해되지 않고, pH 2의 산성 조건에서는 현저히 높은 용해도를 나타내는, pH 감응성을 나타내었다.
실시예 5: 대량생산된 지방산을 도입한 pH 감응성 아미노산 고분자의 물성 분석
본 출원에 따른 지방산을 도입한 pH 감응성 아미노산 고분자를 50 L 반응기에서 대량생산하였다. 단량체로는 친수성 아미노산인 라이신, 소수성 아미노산인 트립토판, 사슬연장제인 글루탐산, 및 지방산인 스테아르산을 사용하여 중합시켜 3종의 고분자(이하, 고분자 Q, R, 및 S)를 합성하고, 이들 고분자의 물성을 측정하여 하기 표 7에 나타내었다. 이때, 고분자 R 및 S는 연속 배치 중합하였다. 중량평균분자량, 및 유리전이온도는 실시예 1과 동일한 방법으로 측정하였다. 점도는 회전형 유변물성 측정기(Rheometer, Anton Paar)를 사용하여 상대점도를 측정하되, 1.0 mm 갭을 가진 축(shaft)과 평형판(parallel plate) 사이에 고분자 물질을 배치하고, 70 내지 190℃ 온도 영역에서 2.5℃/min의 속도로 승온시키면서 일정한 전단율(shear rate)로 축을 회전시켰다. 이때, 고분자 물질의 거동으로부터 150℃에서의 상대점도를 측정하였다. 용융지수는 용융테스터(Meltflow indexer, Toyoseiki)를 사용하여 국제규격 ISO E1133, ASTM D1238에 따라 150℃, 2160 g 하중에서 측정하였다.

상기 고분자 Q, R, 및 S를 코팅 조성물에 사용하여, 실험예 2에 따라 준비한 반추위 내 분해 보호 코팅된 메치오닌 제형(이하 제형 e, f, 및 g로 명명)에 대해 실험예 4에 따른 생체이용률 평가를 수행하고, 그 결과를 하기 표 8에 나타내었다. in situ 반추위 bypass 평가는 48시간 동안 배양하여 수행하였으며, 모든 실험 결과는 DM과 유효성분 함량의 곱을 기준으로 계산하였다.

실시예 6: 유효성분의 종류에 따른 생체이용률 평가
상기 실시예 5에 개시한 고분자 R을 코팅 조성물에 사용하여 실험예 2 및 3에 따라 각각 유효성분으로 메치오닌과 히스티딘을 포함하는 반추위 내 분해 보호 코팅된 메치오닌 제형과 반추위 내 분해 보호 코팅된 히스티딘 제형을 제조하되, 코팅과 유효성분을 하기 표 9에 개시한 비율로 포함하도록 조절하여 3종의 제형(이하, 각각 제형 h, i, 및 j로 명명)을 준비하였다. 이들 제형에 대해 실험예 4에 따른 생체이용률 평가를 수행하고, 그 결과를 하기 표 9에 나타내었다. in situ 반추위 bypass 평가는 24시간 동안 배양하여 수행하였으며, 모든 실험 결과는 DM과 유효성분 함량의 곱을 기준으로 계산하였다.

표 9에 나타난 바와 같이, 코팅층의 비율을 조절함으로써 유효성분의 종류에 무관하게 우수한 생체이용률을 달성할 수 있었다.

Claims (17)
- 친수성 아미노산, 소수성 아미노산, 및 지방산을 단량체로 포함하는, pH 감응성 고분자.
- 제1항에 있어서,상기 친수성 아미노산은 라이신, 아르기닌 또는 둘 모두인 것인, pH 감응성 고분자.
- 제1항에 있어서,상기 소수성 아미노산은 페닐알라닌, 트립토판, 발린, 및 타이로신으로 구성된 군으로부터 선택되는 1종 이상인 것인, pH 감응성 고분자.
- 제1항에 있어서,상기 지방산은 스테아르산, 팔미트산, 및 라우르산으로 구성된 군으로부터 선택되는 1종 이상인 것인, pH 감응성 고분자.
- 제1항에 있어서,사슬연장제를 추가로 포함하는 것인, pH 감응성 고분자.
- 제5항에 있어서,상기 사슬연장제는 글루탐산, 아스파르트산 또는 둘 모두인 것인, pH 감응성 고분자.
- 제1항에 있어서,전체 단량체 중 상기 친수성 아미노산을 50 내지 75몰%로 포함하는 것인, pH 감응성 고분자.
- 제1항에 있어서,전체 단량체 중 상기 소수성 아미노산을 10 내지 35몰%로 포함하는 것인, pH 감응성 고분자.
- 제1항에 있어서,전체 단량체 중 상기 지방산을 5 내지 20몰%로 포함하는 것인, pH 감응성 고분자.
- 제5항에 있어서,전체 단량체 중 상기 사슬연장제를 0 내지 15몰%로 포함하는 것인, pH 감응성 고분자.
- 제1항 내지 제10항 중 어느 한 항의 pH 감응성 고분자, 지방산, 및 셀룰로오스 또는 이의 유도체를 포함하는, 코팅 조성물.
- 제11항에 있어서,상기 코팅 조성물은 반추위 내 분해 보호용인 것인, 코팅 조성물.
- 제11항에 있어서,상기 지방산은 스테아르산, 라우르산 및 팔미트산으로 구성된 군으로부터 선택되는 1종 이상인 것인, 코팅 조성물.
- 제11항에 있어서,상기 셀룰로오스의 유도체는 셀룰로오스, 에틸셀룰로오스 및 메틸셀룰로오스로 구성된 군으로부터 선택되는 1종 이상인 것인, 코팅 조성물.
- 유효성분을 포함하는 코어 입자; 및 제11항의 코팅 조성물을 이용하여 상기 코어 입자 상에 형성된 코팅층을 포함하는, 반추위 내 분해 보호 코팅된 사료 첨가제.
- 제15항에 있어서,상기 사료 첨가제는 반추동물용인 것인, 사료 첨가제.
- 제15항에 있어서,상기 코팅층은 pH 6 내지 8의 반추위에서는 분해되지 않고, pH 3 이하의 제4위에서 선택적으로 분해되어 유효성분을 방출하는 것인, 사료 첨가제.
