Abstract
アセナピン又はその薬学的に許容される塩の分解が抑制された貼付剤を開示する。上記貼付剤は、支持体上に粘着剤層を備え、上記粘着剤層は、粘着基剤と、アセナピン又はその薬学的に許容される塩と、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩と、を含有する。
Description
本発明は、アセナピン又はその薬学的に許容される塩を含有する貼付剤に関する。
アセナピンは、中枢神経系疾患、特に統合失調症の治療薬として知られる化合物である。日本では、アセナピンを含有する医薬品として、アセナピンマレイン酸塩の舌下錠(商品名:シクレスト(登録商標)舌下錠)が流通している。舌下投与は一般的に、初回通過効果を受けにくい投与経路として知られている。そのため、代謝安定性が比較的低い化合物であっても、舌下投与することにより、十分な薬理効果を発揮することが期待される。近年では、新たな製剤として、アセナピン含有貼付剤の開発が検討されている。例えば、特許文献1には、粘着剤層に低分子アミンを含有し、粘着剤層が吸湿した場合においても粘着剤層の粘着力の低下が抑制された貼付剤が開示されている。
貼付剤の製造時及び/又は保管時において、アセナピンが分解され、アセナピンのN-オキシド体及びテトラデヒドロ体が生成し得ることを本発明者らは見出した。本発明の目的は、アセナピン又はその薬学的に許容される塩の分解が抑制された貼付剤を提供することにある。また、本発明の目的は、貼付剤中のアセナピン又はその薬学的に許容される塩の分解を抑制する方法を提供することにある。
本発明者らは、鋭意検討の結果、粘着剤層に塩基性アミノ酸又はその薬学的に許容される塩を配合することで、アセナピンの分解を抑制することができることを見出し、本発明を完成するに至った。
すなわち、本発明の貼付剤は、支持体上に粘着剤層を備え、前記粘着剤層が粘着基剤と、アセナピン又はその薬学的に許容される塩と、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩とを含有する。
また、本発明は、支持体上に粘着剤層を備える貼付剤中のアセナピン又はその薬学的に許容される塩の分解を抑制する方法であり、上記粘着剤層は粘着基剤及びアセナピン又はその薬学的に許容される塩を含有しており、上記粘着剤層中に、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩を含有させることにより、アセナピン又はその薬学的に許容される塩の分解を抑制する。
また、本発明は、支持体上に粘着剤層を備える貼付剤中のアセナピン又はその薬学的に許容される塩の分解を抑制する方法であり、上記粘着剤層は粘着基剤及びアセナピン又はその薬学的に許容される塩を含有しており、上記粘着剤層中に、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩を含有させることにより、アセナピン又はその薬学的に許容される塩の分解を抑制する。
本発明によれば、安定性に優れた、アセナピン又はその薬学的に許容される塩を含有する貼付剤を提供することができる。
以下に、本発明の実施形態を示して、本発明を詳細に説明する。
本発明の一実施形態に係る貼付剤は、支持体上に粘着剤層を備え、前記粘着剤層が粘着基剤と、アセナピン又はその薬学的に許容される塩と、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩とを含有する。
本発明の一実施形態に係る方法は、支持体上に粘着剤層を備える貼付剤中のアセナピン又はその薬学的に許容される塩の分解を抑制する方法であり、上記粘着剤層は粘着基剤及びアセナピン又はその薬学的に許容される塩を含有しており、上記粘着剤層中に、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩を含有させることにより、アセナピン又はその薬学的に許容される塩の分解を抑制する。
支持体は、貼付剤、特に粘着剤層の形状を維持し得るものであればよい。支持体の材質としては、例えば、ポリエチレン、ポリプロピレン、ポリブタジエン、エチレン-塩化ビニル共重合体、ポリ塩化ビニル、ナイロン等のポリアミド、ポリエステル、セルロース誘導体、ポリウレタンなどの合成樹脂が挙げられる。支持体の性状は、例えば、フィルム、シート、シート状多孔質体、シート状発泡体、織布、編布、不織布等の布、及びこれらの積層体などである。支持体の厚さは、特に制限されないが、通常、2μm~3000μm程度であることが好ましい。
粘着剤層は、粘着基剤と、アセナピン又はその薬学的に許容可能な塩と、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩と、後述する任意成分と、を混和して得られる粘着剤組成物から形成される。粘着剤層の単位面積当たりの質量は、特に制限されず、30g/m2~400g/m2とすることができ、40g/m2~300g/m2、50g/m2~200g/m2又は70g/m2~120g/m2であってもよい。粘着剤層の単位面積当たりの質量が400g/m2を超えると、被服の着脱時等に貼付剤が脱落しやすくなる。
アセナピンは、(3aRS,12bRS)-5-クロロ-2-メチル-2,3,3a,12b-テトラヒドロ-1H-ジベンゾ[2,3:6,7]オキセピノ[4,5-c]ピロールとも呼ばれる化合物であり、以下の式(1)で表される。
アセナピンの薬学的に許容可能な塩は、アセナピンの酸付加塩のうち、医薬として利用可能なものを意味する。酸としては、例えば、塩酸、臭化水素酸、ヨウ化水素酸、リン酸、酢酸、プロピオン酸、グリコール酸、マレイン酸、マロン酸、コハク酸、酒石酸、クエン酸、アスコルビン酸、サリチル酸、安息香酸等が挙げられる。例えば、アセナピンマレイン酸塩は、統合失調症の治療薬として市販されている。
アセナピンは、複数の光学異性体を包含しており、いずれの光学異性体であってもよく、ラセミ体等の光学異性体の混合物であってもよい。アセナピンに付加される酸としては、薬学的に許容可能な酸であれば、特に制限されない。アセナピンの酸付加塩は、無水物であってもよく、水和物であってもよい。
アセナピン又はその薬学的に許容可能な塩の含有量は、粘着剤層の総質量を基準として、1質量%~30質量%とすることができ、5質量%~25質量%、7質量%~22質量%又は10質量%~20質量%であってもよい。
粘着基剤は、粘着剤層に粘着性を付与する成分であり、例えば、ゴム系粘着基剤、アクリル系粘着基剤、シリコーン系粘着基剤等が挙げられる。粘着基剤は、ゴム系粘着基剤、アクリル系粘着基剤及びシリコーン系粘着基剤からなる群から選択される1種以上であることが好ましい。粘着基剤は、水を含まないこと(非水系粘着基剤)が好ましい。粘着基剤は、ゴム系粘着基剤、アクリル系粘着基剤及びシリコーン系粘着基剤のいずれであってもよく、これらの組合せであってもよい。粘着基剤の総含有量は、粘着剤層の総質量を基準として、10質量%~90質量%とすることができ、20質量%~90質量%、20質量%~60質量%、20質量%~40質量%であってもよい。
ゴム系粘着基剤としては、例えば、天然ゴム、ポリイソブチレン、アルキルビニルエーテル(共)重合体、ポリイソプレン、ポリブタジエン、スチレン-ブタジエン共重合体、スチレン-イソプレン共重合体、スチレン-イソプレン-スチレンブロック共重合体(SIS)等が挙げられる。ゴム系粘着基剤は、これらのうち1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。中でも、本実施形態に係るゴム系粘着基剤としては、粘着剤層のより十分な粘着力を発揮することができる傾向にあるという観点から、スチレン-イソプレン-スチレンブロック共重合体及びポリイソブチレンからなる群から選ばれる少なくとも1種以上であることが好ましい。
ゴム系粘着基剤の具体例としては、Quintac(登録商標)3570C(商品名、日本ゼオン株式会社製)、SIS5002(商品名、JSR株式会社製)、Oppanol(登録商標)N50、N80、N100、N150、B11、B12、B50、B80、B100、B120、B150、B220(商品名、BASF社製)、JSRブチル065、268、365(商品名、JSR株式会社製)、SIBSTAR(登録商標)T102(商品名、カネカ株式会社製)等が挙げられる。
ゴム系粘着基剤の含有量は、粘着剤層の総質量を基準として、10質量%~90質量%とすることができ、20質量%~90質量%、20質量%~60質量%、20質量%~40質量%であってもよい。
アクリル系粘着基剤は、粘着剤層に粘着性を付与する成分であり、例えば、1種又は2種以上の(メタ)アクリル酸アルキルエステルの(共)重合体である。(メタ)アクリル酸アルキルエステルとしては、例えば、(メタ)アクリル酸ブチル、(メタ)アクリル酸イソブチル、(メタ)アクリル酸ヘキシル、(メタ)アクリル酸オクチル、(メタ)アクリル酸2-エチルヘキシル、(メタ)アクリル酸デシルなどが挙げられる。なお、本明細書において、「(メタ)アクリル酸」との用語は、アクリル酸及びメタクリル酸のいずれか一方又は両方を意味し、類似の表現についても同様に定義される。
アクリル系粘着基剤は、(メタ)アクリル酸アルキルエステル(主モノマー)とコモノマーから形成される共重合体であってもよい。主モノマーとしては、例えば、(メタ)アクリル酸メチル、(メタ)アクリル酸エチル、(メタ)アクリル酸ブチル、(メタ)アクリル酸ヘキシル、(メタ)アクリル酸へプチル、(メタ)アクリル酸オクチル、(メタ)アクリル酸2-エチルヘキシル等が挙げられ、これらのうちの1種を単独で用いても2種以上を組み合わせて用いてもよい。コモノマーは、(メタ)アクリル酸アルキルエステルと共重合できる成分であればよい。コモノマーとしては、例えば、(メタ)アクリル酸ヒドロキシアルキルエステル、エチレン、プロピレン、スチレン、酢酸ビニル、N-ビニルピロリドン、(メタ)アクリル酸、(メタ)アクリル酸アミドなどが挙げられる。コモノマーは、1種を単独で又は2種以上を組み合わせたものであってもよい。

シリコーン系粘着基剤の含有量は、粘着剤層の総質量を基準として、10質量%~90質量%とすることができ、20質量%~90質量%、20質量%~60質量%、20質量%~40質量%であってもよい。
粘着剤層中にリシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩を含有させることで、貼付剤の製造時及び/又は保管時のアセナピンのN-オキシド体及びテトラデヒドロ体の生成が抑制される。塩基性アミノ酸の薬学的に許容される塩は、塩基性アミノ酸の酸付加塩のうち、医薬として利用可能なものを意味する。酸としては、例えば、塩酸、臭化水素酸、ヨウ化水素酸、リン酸、酢酸、プロピオン酸、グリコール酸、マレイン酸、マロン酸、コハク酸、酒石酸、クエン酸、アスコルビン酸、サリチル酸、安息香酸等が挙げられる。好ましい酸付加塩は塩酸塩(リシン塩酸塩、アルギニン塩酸塩及びヒスチジン塩酸塩)である。
塩基性アミノ酸又はその薬学的に許容される塩の含有量は、粘着剤層の総質量を基準として、0.01質量%~7質量%とすることができ、0.05質量%~7質量%、0.01質量%~3質量%、0.01質量%~1質量%、0.05%質量%~1質量%又は0.1質量%~0.5質量%であってもよい。
粘着剤層は、任意に、その他の添加剤をさらに含有してもよい。その他の添加剤としては、例えば、粘着付与樹脂、可塑剤、吸収促進剤、溶解剤、他の補助的な安定化剤、充填剤、香料などが挙げられる。
粘着付与樹脂は、粘着剤層の粘着性を調整する成分である。粘着付与樹脂としては、例えば、脂環族飽和炭化水素樹脂;ロジン、ロジンのグリセリンエステル、水添ロジン、水添ロジンのグリセリンエステル、ロジンのペンタエリスリトールエステル、マレイン化ロジン等のロジン誘導体;テルペン系粘着付与樹脂;石油系粘着付与樹脂等が挙げられる。粘着付与樹脂は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。粘着剤層が粘着付与樹脂を含有する場合、粘着付与樹脂の含有量は、粘着剤層の総質量を基準として、20質量%~80質量%とすることができ、30質量%~70質量%であってもよい。
可塑剤として、例えば、パラフィンオイル(流動パラフィン等)、スクワラン、スクワレン、植物油類(オリーブ油、ツバキ油、ひまし油、トール油、ラッカセイ油、スペアミント油、ユーカリ油、ホホバ油、樟脳白油、ヒマワリ油、オレンジ油等)、油脂類(ジブチルフタレート、ジオクチルフタレート等)、及び液状ゴム(液状ポリブテン、液状イソプレンゴム等)が挙げられる。好ましい可塑剤は、流動パラフィン又は液状ポリブテンである。粘着剤層が可塑剤を含有する場合、可塑剤の含有量は、粘着剤層の総質量を基準として、例えば、3質量%~50質量%、5質量%~30質量%、又は7質量%~20質量%である。
吸収促進剤は、従来、経皮吸収促進作用を有することが知られている化合物であればよい。吸収促進剤としては、例えば、有機酸及びその塩(例えば、炭素原子数6~20の脂肪族カルボン酸(以下、「脂肪酸」ともいう。)及びその塩、ケイ皮酸及びその塩)、有機酸エステル(例えば、脂肪酸エステル、ケイ皮酸エステル)、有機酸アミド(例えば、脂肪酸アミド)、脂肪アルコール、多価アルコール、エーテル(例えば、脂肪エーテル、ポリオキシエチレンアルキルエーテル)などが挙げられる。これらの吸収促進剤は、不飽和結合を有していてもよく、環状、直鎖状又は分枝状の化学構造であってもよい。また、吸収促進剤は、モノテルペン系化合物、セスキテルペン系化合物、及び植物油(例えば、オリーブ油)であってもよい。これらの吸収促進剤は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。
かかる有機酸としては、脂肪族(モノ、ジ又はトリ)カルボン酸(例えば、酢酸、プロピオン酸、クエン酸(無水クエン酸を含む)、イソ酪酸、カプロン酸、カプリル酸、脂肪酸、乳酸、マレイン酸、ピルビン酸、シュウ酸、コハク酸、酒石酸等)、芳香族カルボン酸(例えば、フタル酸、サリチル酸、安息香酸、アセチルサリチル酸等)、ケイ皮酸、アルカンスルホン酸(例えば、メタンスルホン酸、エタンスルホン酸、プロパンスルホン酸、ブタンスルホン酸)、アルキルスルホン酸誘導体(例えば、ポリオキシエチレンアルキルエーテルスルホン酸、N-2-ヒドロキシエチルピペラジン-N’-2-エタンスルホン酸)、コール酸誘導体(例えば、デヒドロコール酸等)が挙げられる。これらの有機酸は、ナトリウム塩等のアルカリ金属塩であってもよい。中でも、脂肪族カルボン酸、芳香族カルボン酸又はこれらの塩が好ましく、酢酸、酢酸ナトリウム又はクエン酸が特に好ましい。脂肪酸としては、例えば、ラウリン酸、ミリスチン酸、パルミチン酸、ステアリン酸、イソステアリン酸、オレイン酸、リノ-ル酸、リノレン酸が挙げられる。
有機酸エステルとしては、例えば、酢酸エチル、酢酸プロピル、乳酸セチル、乳酸ラウリル、サリチル酸メチル、サリチル酸エチレングリコール、ケイ皮酸メチル、脂肪酸エステルが挙げられる。脂肪酸エステルとしては、例えば、ラウリン酸メチル、ラウリン酸ヘキシル、ミリスチン酸イソプロピル、ミリスチン酸ミリスチル、ミリスチン酸オクチルドデシル、パルミチン酸イソプロピル、パルミチン酸セチルが挙げられる。脂肪酸エステルは、グリセリン脂肪酸エステル、プロピレングリコール脂肪酸エステル、ソルビタン脂肪酸エステル、ポリエチレングリコールソルビタン脂肪酸エステル、ポリエチレングリコール脂肪酸エステル、ショ糖脂肪酸エステル、又はポリオキシエチレン硬化ヒマシ油であってもよい。脂肪酸エステルの具体例としては、グリセリンモノカプリレート、グリセリンモノカプレート、グリセリンモノラウレート、グリセリンモノオレエー卜、ソルビタンモノラウレート、ショ糖モノラウレート、ポリソルベート20、プロピレングリコールモノラウレート、ポリエチレングリコールモノラウレート、ポリエチレングリコールモノステアレート、Span40、Span60、Span80、Span120(商品名、クローダジャパン株式会社製)、Tween20、Tween21、Tween40、Tween60、Tween80、NIKKOL HCO-60(商品名、日光ケミカルズ株式会社製)が挙げられる。
有機酸アミドとしては、例えば、脂肪酸アミド(例えば、ラウリン酸ジエタノールアミド)、ヘキサヒドロ-1-ドデシル-2H-アゼピン-2-オン(Azoneともいう。)及びその誘導体、ピロチオデカンが挙げられる。
脂肪アルコールとは、炭素原子数6~20の脂肪族アルコールを意味する。脂肪アルコールとしては、例えば、ラウリルアルコール、ミリスチルアルコール、オレイルアルコール、イソステアリルアルコール、セチルアルコールが挙げられる。多価アルコールとしては、例えば、プロピレングリコールが挙げられる。
脂肪エーテルとは、炭素原子数6~20の脂肪族基(例えば、アルキル基、アルケニル基)を有するエーテルを意味する。ポリオキシエチレンアルキルエーテルとしては、例えば、ポリオキシエチレンラウリルエーテルが挙げられる。
モノテルペン系化合物としては、例えば、ゲラニオール、チモール、テルピネオール、l-メントール、ボルネオール、d-リモネン、イソボルネオール、ネロール、dl-カンフルが挙げられる。モノテルペン系化合物として、ハッカ油を使用してもよい。
吸収促進剤としては、脂肪酸(特にオレイン酸)、パルミチン酸イソプロピル、オレイルアルコール、ラウリルアルコール、イソステアリルアルコール、ラウリン酸ジエタノールアミド、グリセリンモノカプリレート、グリセリンモノカプレート、グリセリンモノオレエー卜、ソルビタンモノラウレート、プロピレングリコールモノラウレート、ポリオキシエチレンラウリルエーテル又はピロチオデカンがより好ましい。
粘着剤層が吸収促進剤を含有する場合、吸収促進剤の含有量は、粘着剤層の総質量を基準として、2質量%~40質量%とすることができる。
溶解剤は、アセナピン又はその薬学的に許容可能な塩を粘着剤組成物中に溶解させやすくする成分である。溶解剤としては、例えば、脂肪酸(例えば、カプリン酸、オレイン酸、リノール酸)、脂肪酸アルキルエステル(例えば、ミリスチン酸イソプロピル、パルミチン酸イソプロピル)、脂肪酸多価アルコールエステル(例えば、モノラウリン酸プロピレングリコール、モノラウリン酸グリセリン、モノオレイン酸グリセリン、モノラウリン酸ソルビタン)、脂肪酸アミド(例えば、ラウリン酸ジエタノールアミド)、脂肪族アルコール(例えば、オクチルドデカノール、イソステアリルアルコール、オレイルアルコール)、多価アルコール(例えば、プロピレングリコール、ジプロピレングリコール、ポリエチレングリコール)、ピロリドン誘導体(例えば、N-メチル-2-ピロリドン)、有機酸又はその塩(例えば、酢酸、乳酸、酢酸ナトリウム、乳酸ナトリウム)、水酸化ナトリウムが挙げられる。粘着剤層が溶解剤を含有する場合、溶解剤の含有量は、粘着剤層の総質量を基準として、2質量%~40質量%とすることができる。
他の補助的な安定化剤は、紫外線等の光線、熱又は活性化学種の作用により発生するフリーラジカルの生成及びその連鎖反応の進行を抑制できるものであればよい。安定化剤を任意に含有することにより、貼付剤の製造時におけるアセナピンの安定性をより向上させることができる。安定化剤としては、例えば、トコフェロール及びそのエステル誘導体、アスコルビン酸及びそのエステル誘導体、2,6-ジブチルヒドロキシトルエン(BHT)、2,6-ジブチルヒドロキシアニソール(BHA)、2-メルカプトベンズイミダゾール等が挙げられる。安定化剤は、1種を単独で用いてもよく、2種以上を組み合わせて用いてもよい。粘着剤層が安定化剤を含有する場合、安定化剤の含有量は、粘着剤層の総質量を基準として、0.05質量%~3質量%とすることができ、0.05質量%~1質量%、0.05質量%~0.25質量%又は0.1質量%~0.25質量%であってもよい。安定化剤の含有量が0.05質量%~3質量%であると、貼付剤における各成分の安定性が優れる傾向がある。
充填剤としては、例えば、金属化合物(酸化アルミニウム、水酸化アルミニウム、酸化亜鉛、酸化チタン、炭酸カルシウム等)、セラミクス(タルク、クレー、カオリン、シリカ、ハイドロキシアパタイト、合成ケイ酸アルミニウム、メタケイ酸アルミン酸マグネシウム等)又は有機化合物(セルロース粉末、ステアリン酸塩等)の、粉末又はこれらを含む樹脂の短繊維が挙げられる。粘着剤層が充填剤を含有する場合、充填剤の含有量は、粘着剤層の総質量を基準として、0.1質量%~20質量%とすることができる。
貼付剤は、さらに剥離ライナーを備えていてもよい。剥離ライナーは、粘着剤層に対して、支持体と反対側の面に積層される。剥離ライナーを備えていると、保管時において、粘着剤層へのゴミ等の付着を低減することができる傾向がある。剥離ライナーの粘着剤層と接する面は、シリコーン又はフッ素化ポリオレフィン等により離型処理されていることが好ましい。
剥離ライナーの素材としては、特に限定されず、当業者に一般的に知られているライナーを用いることができる。剥離ライナーとしては、例えば、ポリエチレンテレフタレート、ポリエチレンナフタレート等のポリエステル;ポリエチレン、ポリプロピレン等のポリオレフィン;ポリ塩化ビニル、ポリ塩化ビニリデン、ナイロン、アルミニウム等のフィルムが挙げられる。剥離ライナーは、上質紙とポリオレフィンとのラミネートフィルムであってもよい。剥離ライナーの材質としては、ポリプロピレン又はポリエチレンテレフタレート製のフィルムが好ましい。
貼付剤は、例えば、次の方法により製造することができるが、これに限定されず、公知の方法を使用することができる。まず、粘着剤層を構成する各成分を所定の割合で混合して均一な溶解物(粘着剤組成物)を得る。次に、剥離可能なフィルム(剥離ライナー)上に粘着剤組成物を所定の厚みで展延して粘着剤層を形成する。さらに、粘着剤層が剥離ライナーと支持体とに挟まれるように、粘着剤層に支持体を圧着する。最後に、所望の形状及び寸法に裁断することにより、貼付剤を得ることができる。この場合、剥離ライナーは、貼付剤の適用時に除去される。貼付剤の形状及び寸法は、例えば、短辺が3~14cmかつ長辺が7~20cmの矩形、又は直径が1~10cmの円形であってもよい。
試験例1:塩基性アミノ酸を含有する貼付剤の安定性評価
貼付剤の調製
下記表1~表2に従って、各成分を混合し粘着剤組成物を得た。得られた粘着剤組成物を剥離ライナー(離型処理が施されたポリエチレンテレフタレート製フィルム)上に、単位面積当たりの質量が100g/m2になるように展延し、溶媒を乾燥除去して粘着剤層を形成した。得られた粘着剤層の前記反対の面上に支持体層(ポリエチレンテレフタレート製フィルム)を積層し、支持体層/粘着剤層/剥離ライナーの順に積層された貼付剤を得た。
貼付剤の調製
下記表1~表2に従って、各成分を混合し粘着剤組成物を得た。得られた粘着剤組成物を剥離ライナー(離型処理が施されたポリエチレンテレフタレート製フィルム)上に、単位面積当たりの質量が100g/m2になるように展延し、溶媒を乾燥除去して粘着剤層を形成した。得られた粘着剤層の前記反対の面上に支持体層(ポリエチレンテレフタレート製フィルム)を積層し、支持体層/粘着剤層/剥離ライナーの順に積層された貼付剤を得た。
含量試験
上記製造方法により製造した貼付剤の製造直後と、60℃にて1か月間包装袋に保存した後の貼付剤について、アセナピンのN-オキシド体及びテトラデヒドロ体の発生量をHPLCにより定量した。
上記製造方法により製造した貼付剤の製造直後と、60℃にて1か月間包装袋に保存した後の貼付剤について、アセナピンのN-オキシド体及びテトラデヒドロ体の発生量をHPLCにより定量した。
より詳細には、貼付剤の粘着剤層を取り出し、テトラヒドロフラン(高速液体クロマトグラフィー用グレード)5mLに浸漬して有機物を抽出し、これに希釈溶液(0.1%リン酸水溶液/メタノール=50/50(v/v))45mLを加えて全量を50mLとし、不溶物をろ過した後、以下の分析条件における高速液体クロマトグラフィー法により、アセナピン、そのN-オキシド体及びそのテトラデヒドロ体のピークが分離したクロマトグラムを得た。N-オキシド体及びテトラデヒドロ体の含有量は、アセナピンの理論量を100として、N-オキシド体及びテトラデヒドロ体に対応するピークの曲線下面積の値から算出した。なお、N-オキシド体のアセナピンに対する相対保持時間(RRT)は0.24であり、テトラデヒドロ体のアセナピンに対するRRTは1.10であった。

塩基性アミノ酸を含有する貼付剤では、アセナピンのN-オキシド体及びテトラデヒドロ体の発生を十分に抑制されていることが確認された。
試験例2:塩基性アミノ酸を含有する貼付剤の安定性評価
下記表3~表4に従って、各成分を混合し粘着剤組成物を得た。試験例1と同様にして、貼付剤を得た。試験例1と同様にして、製造直後の貼付剤及び60℃にて1か月間包装袋に保存した後の貼付剤中の、アセナピンのN-オキシド体及びテトラデヒドロ体の発生量をHPLCにより定量した。
下記表3~表4に従って、各成分を混合し粘着剤組成物を得た。試験例1と同様にして、貼付剤を得た。試験例1と同様にして、製造直後の貼付剤及び60℃にて1か月間包装袋に保存した後の貼付剤中の、アセナピンのN-オキシド体及びテトラデヒドロ体の発生量をHPLCにより定量した。
塩基性アミノ酸を含有する貼付剤では、アセナピンのN-オキシド体及びテトラデヒドロ体の発生を十分に抑制されていることが確認された。
Claims (6)
- 支持体上に粘着剤層を備える貼付剤であって、
前記粘着剤層が、粘着基剤と、アセナピン又はその薬学的に許容される塩と、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩と、を含有する、貼付剤。 - 前記粘着基剤がゴム系粘着基剤、アクリル系粘着基剤及びシリコーン系粘着基剤からなる群から選択される少なくとも1種である、請求項1に記載の貼付剤。
- 前記塩基性アミノ酸又はその薬学的に許容される塩の含有量が粘着剤層の総質量を基準として0.01質量%~7質量%である、請求項1又は2に記載の貼付剤。
- 貼付剤中のアセナピン又はその薬学的に許容される塩の分解を抑制する方法であって、
前記貼付剤は支持体上に粘着剤層を備え、前記粘着剤層は粘着基剤及びアセナピン又はその薬学的に許容される塩を含有する、貼付剤であり、
前記粘着剤層中に、リシン、アルギニン及びヒスチジンからなる群から選択される少なくとも1種の塩基性アミノ酸又はその薬学的に許容される塩を含有させる、方法。 - 前記粘着基剤がゴム系粘着基剤、アクリル系粘着基剤及びシリコーン系粘着基剤からなる群から選択される少なくとも1種である、請求項4に記載の方法。
- 前記塩基性アミノ酸又はその薬学的に許容される塩の含有量が粘着剤層の総質量を基準として0.01質量%~7質量%である、請求項4又は5に記載の方法。
