Abstract
본 발명에 따른 융합 단백질-siRNA 복합체는 암세포에 특이적으로 결합하고, 세포 내로 효과적으로 함입되어, 리소좀에 의해 분해되면서 항암활성을 나타내는 것으로, 상기 융합 단백질-siRNA 복합체는 암세포의 면역작용을 억제함으로써 대식세포의 식세포 작용을 높임으로써, 자가면역에 의해 암이 제거될 수 있도록 하는 극대화된 항암활성을 갖는다.
Description
본 발명은 항암활성을 갖는 면역조절 단백질-siRNA 복합체에 관한 것으로, 보다 구체적으로 암세포의 면역 회피 기전을 억제함으로써 대식세포의 식작용을 증대시켜 암세포의 사멸을 유도하는 것을 특징으로 하는 면역조절 단백질-siRNA 복합체에 관한 것이다.
암은 지난 수년간 사망원인 1위를 차지하고 있다. 이러한 암을 치료하기 위하여, 다양한 기술들이 개발되고 왔다. 일예로 독소루비신이나 파클리탁셀과 같은 항암제들이 있는데, 암세포의 성장을 억제하거나 사멸시키는 효과를 갖는다. 그러나 이러한 항암제는 암세포와 정상세포 모두에 대해 독성을 나타내기 때문에, 항암효과에 비해 부작용이 심각하여 치료제로써 사용이 어렵다는 문제가 있다.
상술한 문제점을 해결하기 위하여 siRNA를 활용한 항암제 개발이 활발히 이뤄지고 있다. siRNA는 특정 표적 유전자의 발현양을 억제시킴으로서 관련 유전자의 활동을 간섭하는 역할을 하는 것으로 알려져 있다. 따라서 siRNA의 기능을 적극 활용한다면 부작용은 최소화하면서 항암효능은 우수한 치료제가 개발될 것으로 기대하였다. 이에, 종양 유전자(oncogene), 면역세포, 종양미세환경 조절 인자 등과 같은 다양한 유전자를 표적화하는 siRNA를 개발하였으나, siRNA 자체적으로 생체 내에서 안정적으로 존재하지 못하고, 세포 내로 전달되는 효율이 현저히 낮다는 단점이 있었다.
이를 해결하기 위해 siRNA를 전달하기 위한 전달체로 리포좀(liposome), 키토산나노파티클(chitosan nanoparticle), PLGA(poly lactic-co-glycolic acid) 등 다양한 물질들을 활용한 나노입자가 연구되었다. 상기 나노입자는 siRNA만을 사용하는 것에 비해 안정성과 전달성을 높여줄 수 있으나, 나노입자 자체가 외부물질이기 때문에 체내에서 독성을 유발하게 되는 단점이 존재하며, 세포 내로 물질이 들어가더라도 세포 내 엔도좀/리소좀에서 분해되어 유전자 도입(transfection) 효율이 상당히 낮아지는 단점이 있다. 따라서 최근에는 생체 내에 존재하는 다양한 물질 (단백질, 엑소좀(exosome), 항체 등)을 전달체로 이용함으로써 생체 내 독성을 줄이되, siRNA를 효과적으로 암세포 내로 전달할 수 있는 소재에 대한 연구가 주를 이루고 있다. 이러한 많은 노력에도 불구하고, 물질의 전달 효용성 및 기전에 관한 문제들로 인해 아직까지 상용화되지 못하고 있다. 예를 들어 엑소좀(exosome)은 체내에서 쉽게 분해 및 붕괴되어 약물을 효과적으로 전달하는데 한계가 있고, 항체는 크기가 상대적으로 커서 siRNA를 세포 내에 효과적으로 전달하지 못하는 문제가 있다.

본 발명은 상기와 같은 문제점을 감안하여 안출된 것으로, 본 발명의 목적은 암세포 수용체를 통해 암세포에 특이적으로 전달되어 이중으로 암세포의 면역 회피 기전을 억제시킴으로써 대식세포의 식작용을 향상시키는 우수한 항암활성을 갖는 융합 단백질-siRNA 복합체을 제공하는 것이다.
본 발명은 상기와 같은 문제점을 감안하여 안출된 것으로, 본 발명의 목적은 융합 단백질-siRNA 복합체를 유효성분으로 하는 암 치료 또는 예방용 조성물을 제공하는 것이다.
본 발명은 상기 목적을 이루기 위하여, 면역조절 융합단백질(A); 및 siRNA(B);를 포함하되, 상기 면역조절 융합단백질(A)은 SIPRα 단백질과 리소좀 분해효소로 절단되는 링커 펩티드가 결합된 것을 특징으로 하는 융합 단백질-siRNA 복합체를 제공한다.
상기 SIPR 단백질은 서열번호 3, 5, 7, 9, 11 중에서 선택되는 어느 하나로 표시되며, 상기 링커 펩티드는 서열번호 13으로 표시되는 것일 수 있다.
상기 면역조절 융합단백질(A)은 서열번호 1로 표시되는 것일 수 있다.
상기 siRNA는 CD47 siRNA일 수 있다.
상기 siRNA는 서열번호 17 또는 18로 표시되는 것일 수 있다.
상기 융합 단백질-siRNA 복합체의 평균 분자량은 20 kDa 내지 60 kDa인 것일 수 있다.
본 발명은 상기 다른 목적을 이루기 위하여, 상기 융합 단백질-siRNA 복합체를 유효성분으로 포함하는 암 치료 또는 예방용 약학 조성물을 제공한다.
상기 암은 뇌종양, 척수종양, 망막 세포종, 구강암, 비강암, 부비동암, 인두암, 후두암, 경부암, 두경부암, 흑색종, 피부암, 유방암, 갑상선암, 악성부신종양, 내분비암, 폐암, 흉막종양, 호흡기암, 식도암, 위암, 소장암, 대장암, 항문암, 간암, 담도암, 췌장암, 신장암, 방광암, 전립선암, 고환암, 음경암, 자궁경부암, 자궁체부암, 융모상피암, 난소암, 급성, 만성 백혈병, 악성림프종, 다발성 골수증을 포함하는 혈액암, 골종양, 연부종양, 소아 백혈병 및 소아암으로 이루어진 군으로부터 선택된 어느 하나 이상일 수 있다.
본 발명에 따른 융합 단백질-siRNA 복합체는 암세포에서 특이적으로 과발현되는 수용체에 의해 세포 내로 흡수되어, 암세포 면역 방어 기능을 이중으로 제어함으로서 극대화된 항암활성을 갖는다. 게다가 세포 독성이 적고, 전달효율이 높기 때문에 부작용이 거의 없다는 장점이 있다.
또한 암세포의 사멸을 직접적으로 유도하지 않고, 암세포의 면역을 억제함으로써 환자의 자가면역(대식세포 식작용)을 통해 제거되도록 함으로써 몸에 부담이 적다.
도 1은 융합 단백질-siRNA 복합체 구조를 개략적으로 도시한 도면이다.
도 2는 융합 단백질-siRNA 복합체 구조의 작동기전을 개략적으로 도시한 도면이다. 면역조절 단백질과 siRNA 항암제가 암세포의 같은 수용체를 1) 단백질 수준과 2) mRNA수준에서 이중으로 제어함으로서 암면역치료 효율을 극대화한다.
도 3은 실시예 1에서 제조된 선별된 형질전환체로부터 콜로니를 얻은 후, PCR을 수행하여 젤 이미지 시스템(Gel documentation)으로 분석한 결과이다.

이하에서, 본 발명의 여러 측면 및 다양한 구현 예에 대해 더욱 구체적으로 살펴보도록 한다.
본 발명자들은 일반적인 상태에서는 세포 독성이나 활성을 전혀 나타내지 않다가, 특정 세포(암세포)의 내부로 전달된 후에는 독성이나 활성을 나타내는 복합체를 제조하고자 하였다. 이를 위해 가장 고려했던 부분은 세포 내 전달된 후, 세포 내에 존재하는 리소좀에 의해 분해되어 활성을 나타내도록 하는 것이었다. 따라서 이러한 문제를 해결하면서 우수한 암세포 표적능력, 세포내 전달능력을 가지면서 세포 독성을 거의 나타내지 않는 소재를 위해 수많은 실험을 수행하였고, 그 결과 본 발명에 따른 융합 단백질-siRNA 복합체가 암세포에 특이적으로 전달되면서, 세포 내 전달 효율도 우수할 뿐만 아니라 세포내 리소좀에 의해 분해되어 암세포에 대한 대식세포 식작용을 증대시키는 현저한 효과를 가지고 있음을 확인함으로써 본 발명을 완성하게 되었다.
본 발명의 일 측면은 면역조절 융합단백질(A); 및 siRNA(B);를 포함하되, 상기 면역조절 융합단백질(A)은 SIPRα 단백질과 리소좀 분해효소로 절단되는 링커 펩티드가 결합된 것을 특징으로 하는 융합 단백질-siRNA 복합체에 관한 것으로, 구체적인 구조는 도 1에 개략적으로 도시하였다.
본 발명에 따른 융합 단백질-siRNA 복합체는 암세포에 특이적으로 반응하여 세포 내로 들어가며, 세포 내에서 리소좀에 의해 자발적으로 절단되어 활성화될 수 있다.
상기 SIPRα 단백질은 서열번호 3, 5, 7, 9, 11 중에서 선택되는 어느 하나로 표시되는 아미노산 서열을 포함할 수 있다.
상기 융합 단백질은 단백질과 siRNA 복합체를 용이하게 구성하기 위해 면역 조절 융합 단백질의 C 말단기에 시스테인(cystein) 아미노산을 삽입하여 단백질의 thiol기와 siRNA의 결합을 1 대 1 결합이 되도록 설계하였다. 이는 종래의 단백질과 siRNA의 단순한 무작위 결합으로 생길 수 있는 입자간 이질성, 분석의 어려움, 대량생산 및 품질 관리가 어려움 등의 문제를 해결하여, 잘 정의된 균일하고 단순한 구조체(융합 단백질-siRNA 복합체)를 얻게 한다. 따라서 무작위적인 융합 단백질과 siRNA 복합체들에 비해 본 발명의 융합 단백질-siRNA 복합체는 상술한 구조적 특징으로 인하여 향후 임상적용에 유리한 효과를 나타낼 수 있다.
상기 링커 펩티드는 리소좀 내에 존재하는 효소에 의해 쉽게 절단가능한 서열이라면 특별히 이에 제한되지 않고, 바람직하게는 5 내지 15개, 보다 바람직하게는 7 내지 12개의 아미노산으로 구성된 것일 수 있고, 가장 바람직하게는 서열번호 13으로 표시되는 아미노산 서열을 포함하는 것일 수 있다.
상기 리소좀 분해효소로 절단되는 링커 펩티드는 서열번호 13(GFLG)으로 표시되는 아미노산 서열을 포함할 수 있고, 이는 세포 내 리소좀에 존재하는 카텝신 B(Cathepsin B) 효소에 의해 절단되며, 이는 상기 융합 단백질의 수용체 매게 흡수(receptor mediated endocytosis)로 세포 내 흡수된 후 엔도좀 탈출(endosomal escape)을 촉진시키는 기능을 할 수 있다. 엔도좀 내 분해는 siRNA 전달체의 큰 한계점으로 이를 극복하는 것은 세포 내로 유전자를 도입(transfection)하는 효율을 향상시키는 효과를 갖게 한다.
또한 면역원성 세포 사멸(Immunogenic cell death, ICD)은 세포 사멸 시 손상 가능 패턴(Damage-associated molecular pattern, DAMP)을 유도함으로써 면역 반응을 증폭시키는 역할을 하는데 이 때, 면역 조절 융합 단백질 및 siRNA는 같은 표적을 이중으로 제어함으로써 종래의 각각의 단백질과 siRNA가 유발할 수 있는 면역 반응보다 배로 증폭된 효과를 유도할 수 있을 것이다.
본 발명에서 'siRNA(short interfering RNA)'란 유전자의 활성을 억제하는 RNAi를 유발시킬 수 있는 이중가닥 RNA를 의미한다. 본 발명에서 siRNA는 CD47의 활성을 억제할 수 있는 siRNA를 의미하는데, 상기 siRNA는 RNAi를 유발시키기만 하면 어떠한 형태의 것도 가능하고, 예를 들면, 화학합성 또는 생화학적 합성 또는 생체내 합성에 의해 수득되는 siRNA, 혹은 약 40개 염기 이상의 이중가닥 RNA가 체내에서 분해된 10개 염기대 이상의 이중가닥 RNA 등을 사용할 수 있다.
상기 siRNA는 CD47의 핵산서열의 일부에 대하여 약 70% 이상, 바람직하게는 75% 이상, 보다 바람직하게는 80% 이상, 보다 더욱 바람직하게는 85% 이상, 더욱 바람직하게는 90% 이상, 특히 바람직하게는 95% 이상, 가장 바람직하게는 100%의 상동성을 가지는 서열로 구성될 수 있고, 이중가닥 부분을 포함하는 RNA 또는 그의 개변체를 사용할 수도 있다. 상동성을 가지는 서열부분은, 통상적으로 적어도 20 뉴클레오티드 이상이고, 바람직하게는 약 23~30 뉴클레오티드이다. 아울러, 상기 siRNA는 CD47의 염기서열 중에서 다른 RNA와 상동성이 가장 낮은 영역에 상보적으로 결합할 수 있는 염기서열로 구성됨이 바람직하다.
본 발명에 따른 융합 단백질-siRNA 복합체는 siRNA를 직접 투여하는 것에 비해 현저히 세포 내 전달 효율이 우수하며, 항암 활성도 현저히 높다는 큰 장점을 갖는다.
또한, 상기 융합 단백질-siRNA 복합체는 생리학적 용액에 쉽게 용해되므로, 흡수가 용이하여 생체 이용률이 뛰어나다.
상기 siRNA의 염기서열은 CD47 유전자를 침묵시킬 수 있는 것, CD47 유전자의 발현을 억제할 수 있는 것이라면 특별히 이에 제한되지 않으나, sliencing 효과면에서 서열번호 17 또는 18을 사용함이 바람직하고, 상기 siRNA의 3' 말단에는 말레이미드가 결합되어 있는 것일 수 있다.
본 발명에 따른 융합 단백질-siRNA 복합체는 암세포에서 특이적으로 발현되는 수용체에 대해 결합하므로, 암세포 특이적 전달효율이 높다. 또한, 융합 단백질-siRNA 복합체는 CD47 수용체와 결합하고 암세포 내에서 분해되어 CD47 유전자를 침묵시키는 등 암세포 내에서 암세포의 면역작용을 억제함으로써 대식세포가 식작용을 통해 암세포를 공격할 수 있게 한다. 즉 본 발명에 따른 융합 단백질-siRNA 복합체 종래 항암제나 전달체와 달리 대식세포가 암세포를 제거할 수 있도록 암세포의 면역작용을 저해시키는 것이므로, 직접적으로 암세포를 사멸시키는 것이 아닌 자가면역으로 항암효과를 낼 수 있게 된다. 이러한 융합 단백질-siRNA 복합체의 작동기전을 도 2에 개략적으로 도시하였다.
이에 본 발명의 융합 단백질-siRNA 복합체는 암이 발생하였을 때, 상기 암세포에 특이적으로 결합하여, 세포 내로 전달되며, 암세포의 면역을 억제하여 대식세포의 식작용을 증대하여 암세포가 제거되도록 할 수 있음을 다양한 실험을 통해 확인하였다.
본 발명의 다른 측면은 상기 융합 단백질-siRNA 복합체을 유효성분으로 하는 암 치료 또는 예방용 약학 조성물에 관한 것이다.
또한 본 발명은 상기 융합 단백질-siRNA 복합체을 유효성분으로 포함하는 암 치료 또는 예방용 동물용 약학 조성물에 관한 것이다.
또한 본 발명은 인간, 또는 인간을 제외한 동물에게 상기 조성물을 투여하여 암을 치료하기 위한 방법을 제공한다.
또한 본 발명은 암 치료 또는 예방용 의약, 또는 동물용 의약 제조를 위한 상기 융합 단백질-siRNA 복합체의 신규 용도를 제공한다.
본 발명에 따른 융합 단백질-siRNA 복합체는 다양한 경로를 통해 투여가 가능하고, 이들 각각을 단독으로 투여하는 것보다 암세포에 대한 약리효과가 현저히(1.5~2 배 이상) 우수함을 확인하였다.
구체적으로 본 발명의 복합체는 siRNA 또는 SIPRα 단백질을 단독으로 사용하는 경우에 비해서, 암세포 특이적 세포내 전달이 용이하며, 잠재적 안정성이 매우 우수하다. 게다가 통제되지 않는 복제의 위험성이 없고, siRNA 전달을 위한 바이러스 혹은 비 바이러스 벡터를 이용하지 않아도 되기 때문에 형질주입에 따른 간섭과 잠재적인 부작용에 의한 염려를하지 않아도 되어 매우 안정적이라 할 수 있다. 게다가, 벡터와 같은 전달체를 이용하는 것에 비해 합성이 매우 저렴하고, 전달효율도 매우 우수하여(낮은 농도의 siRNA와 SIPRα 단백질을 투여하여 최대의 예방 또는 치료효과를 달성함) 경제적 측면에서 큰 이점을 갖는다.
암세포를 사멸시키는 경우에는 특정 암세포에 국소적인 치료(항암제 등)만으로는 그 효과를 달성하기 어렵고 몸에 부담이 많이 된다. 그러나 본 발명의 복합체와 같이 암세포 자체의 면역을 억제시킴으로써, 환자 본인에 의해 자가면역에 의해 암이 제거되므로 몸에 부담이 적고, 부작용이 거의 없는데도 우수한 항암효과를 제공할 수 있다는 점에서 엄청난 이점을 제공한다.
상기 암은 통상의 암을 의미하며, 상기 암은 뇌종양, 척수종양, 망막 세포종, 구강암, 비강암, 부비동암, 인두암, 후두암, 경부암을 포함하는 두경부암, 흑색종을 포함하는 피부암, 유방암, 갑상선암, 악성부신종양을 포함하는 유방암, 내분비암, 폐암, 흉막종양, 악성부신종양을 포함하는 호흡기암, 식도암, 위암, 소장의 악성종양, 대장암, 항문암, 간암, 담도암 또는 췌장암을 포함하는 소화기암, 신장암, 방광암, 전립선암, 고환암 또는 음경암을 포함하는 비뇨기암, 자궁경부암, 자궁체부암, 융모상피암 또는 난소암을 포함하는 부인암, 급성 또는 만성 백혈병, 악성림프종 또는 다발성 골수증을 포함하는 혈액암, 골종양 또는 연부종양을 포함하는 골 또는 연부 종양, 및 소아 백혈병 또는 소아 고형종양을 포함하는 소아암 등일 수 있고, 이들의 재발암도 포함하며, 재발암은 통상의 암 치료 후, 재발생한 암을 의미한다.
본 발명에서 "복합체"란 하기 융합 단백질과 siRNA를 포함하며, 이들의 유전적 융합이나 화학적 결합으로 형성된 공유결합 복합체를 의미한다.
또한, 상기 "유전적 융합"이란 단백질을 코딩하는 DNA 서열의 유전적 발현을 통해서 형성된 선형, 공유결합으로 이루어진 연결을 의미한다.
상기 화학적 결합은 당업계에서 일반적으로 사용되는 것이라면 특별히 이에 제한되지 않으나, 바람직하게는 융합 단백질의 시스테인 잔기와 siRNA를 결합하기 위하여, siRNA의 말단에 말레이미드기를 포함하는 유도체를 링커로 결합시키는 것일 수 있고, 상기 융합 단백질에 존재하는 시스테인 잔기의 티올이 siRNA 말레이미드 기와 반응하여 결합을 형성하는 것을 의미한다.
또한, 상기 융합 단백질-siRNA 복합체에 있어서, 상기 융합 단백질과 siRNA를 혼합하여 제조하였을 때, 상기 혼합 비율이 1 : 1-10인 것이 바람직하며, 1 : 1 미만인 경우, 제대로 복합체를 형성하지 못하여, 효과적으로 암세포에 전달되지 못하게 되는 문제가 있다.
따라서 상기 융합 단백질-siRNA 복합체를 투여함으로써, 종래 siRNA(CD47 siRNA), 혹은 상기 SIPRα 단백질만을 투여할 때 보다 암세포로 전달되는 효율이 증가하였고, 대식세포 식작용 증가효과가 현저히(1.5~3 배 이상) 우수하였음을 확인하였다.
본 발명의 약학 조성물은 투여를 위해 상기 기재한 유효성분 이외에 추가로 약제학적으로 허용가능한 담체를 1종 이상 포함하여 약학 조성물로 제제화한 것일 수 있다.
액상 용액으로 제제화되는 조성물에 있어서 허용 가능한 약제학적 담체로는, 멸균 및 생체에 적합한 것으로서, 식염수, 멸균수, 링거액, 완충 식염수, 알부민 주사용액, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다.
또한, 상기 암 치료 또는 예방용 약학 조성물은 상기 유효성분 이외에 약제학적으로 적합하고 생리학적으로 허용되는 보조제를 사용하여 제조될 수 있으며, 상기 보조제로는 부형제, 붕해제, 감미제, 결합제, 피복제, 팽창제, 윤활제, 활택제 또는 향미제 등의 가용화제를 사용할 수 있다.
또한, 본 발명의 암 치료 또는 예방용 약학 조성물은 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 더 나아가 해당분야의 적절한 방법으로 Remington's Pharmaceutical Science, Mack Publishing Company, Easton PA에 개시되어 있는 방법을 이용하여 각 질환에 따라 또는 성분에 따라 바람직하게 제제화할 수 있다.
또한, 본 발명의 암 치료 또는 예방용 약학 조성물은 암세포의 수용체를 표적으로 하여, 이미 암세포에 대한 특이성을 가지고 있으므로 투여 경로에 특별히 제한되지 않으나, 바람직하게는 정맥내, 동맥내, 복강내, 근육내, 동맥내, 복강내, 흉골내, 경피, 비측내, 비강, 흡입, 국소, 직장, 경구, 안구내 또는 피내 경로를 통해 통상적인 방식으로 투여할 수 있으나, 가장 바람직하게는 정맥 내 주사를 통해서 주입되는 것일 수 있지만 특별히 이에 제한되지 않다. 목적하는 바에 따라, 복강 내 투여, 근육 내 투여, 피하 투여, 경구 투여, 비강 내 투여, 폐내 투여, 직장 내 투여될 수 있다.
상기 '약학 조성물', '의약', '동물용 약학 조성물' 또는 '동물용 의약'은 유효성분으로 융합 단백질-siRNA 복합체 이외에, 약학 조성물 등의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다.
상기 '담체'는 세포 또는 조직 내로의 화합물의 부가를 용이하게 하는 화합물이다. 상기 '희석제'는 대상 화합물의 생물학적 활성 형태를 안정화시킬 뿐만 아니라, 화합물을 용해시키게 되는 물에서 희석되는 화합물이다.
본 발명의 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토오스, 덱스트로오스, 수크로오스, 소르비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘포스페이트, 칼슘 실리케이트, 셀룰로오스, 메틸 셀룰로오스, 미정질 셀룰로오스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
본 발명에서 사용되는 용어 "투여"는 임의의 적절한 방법으로 개체에게 소정의 본 발명의 조성물을 제공하는 것을 의미한다.
상기 약학 조성물, 의약, 동물용 약학 조성물 또는 동물용 의약의 사용량은 환자 또는 치료대상 동물의 나이, 성별, 체중에 따라 달라질 수 있으며, 무엇보다도, 치료대상 개체의 상태, 치료 대상 질환의 특정한 카테고리 또는 종류, 투여 경로, 사용되는 치료제의 속성에 의존적일 것이다.
상기 약학 조성물, 의약, 동물용 약학 조성물 또는 동물용 의약은 체내에서 활성성분의 흡수도, 배설속도, 환자 또는 치료대상 동물의 연령 및 체중, 성별 및 상태, 치료할 질병의 중증정도 등에 따라 적절히 선택되나, 일반적으로 1일 0.0001 내지 1,000mg/kg, 바람직하게는 0.001 내지 500mg/kg, 더욱 바람직하게는 0.001 내지 250mg/kg으로 투여하는 것이 바람직하다. 이렇게 제형화 된 단위 투여형 제제는 필요에 따라 일정시간 간격으로 수회 투여할 수 있다.
상기 약학 조성물, 의약, 동물용 약학 조성물 또는 동물용 의약은 개별적으로 예방제 또는 치료제로서 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다.
상기 암의 치료방법은 인간, 또는 인간을 제외한 동물, 특히 포유동물에게 상기 조성물을 투여 하는 것으로, 예를 들어 암 질환을 갖는 치료대상 개체에게 상기 조성물을 투여하는 것이다.
상기 암은 통상의 암을 의미하며, 상기 암은 뇌종양, 척수종양, 망막 세포종, 구강암, 비강암, 부비동암, 인두암, 후두암, 경부암을 포함하는 두경부암, 흑색종을 포함하는 피부암, 유방암, 갑상선암, 악성부신종양을 포함하는 유방암, 내분비암, 폐암, 흉막종양, 악성부신종양을 포함하는 호흡기암, 식도암, 위암, 소장의 악성종양, 대장암, 항문암, 간암, 담도암 또는 췌장암을 포함하는 소화기암, 신장암, 방광암, 전립선암, 고환암 또는 음경암을 포함하는 비뇨기암, 자궁경부암, 자궁체부암, 융모상피암 또는 난소암을 포함하는 부인암, 급성 또는 만성 백혈병, 악성림프종 또는 다발성 골수증을 포함하는 혈액암, 골종양 또는 연부종양을 포함하는 골 또는 연부 종양, 및 소아 백혈병 또는 소아 고형종양을 포함하는 소아암 등일 수 있고, 이들의 재발암도 포함하며, 재발암은 통상의 암 치료 후, 재발생한 암을 의미한다.
상기 치료를 위한 투여량, 투여 방법 및 투여 횟수는 상기 약학 조성물, 의약, 동물용 약학 조성물 또는 동물용 의약의 투여량, 투여 방법 및 투여 횟수를 참고할 수 있다.
이하, 바람직한 실시예를 들어 본 발명을 더욱 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 범위가 이에 의하여 제한되지 않는다는 것은 당업계의 통상의 지식을 가진 자에게 자명할 것이다.
실시예 1. SIPRα 단백질과 리소좀 분해효소로 절단되는 링커 펩타이드가 융합된 면역조절 융합 단백질(SIPRα-linker) 제조
먼저, 면역조절 융합단백질 발현을 위한 벡터를 제작하기 위하여, 먼저, 코스모진텍(Seoul, South korea)을 통해 면역조절 융합단백질(서열번호 1)의 암호화하는 유전자(서열번호 2)을 합성하였고, 이를 포함하는 벡터를 제공받았다. 상기 벡터를 주형으로 하여, 하기 표 1에 개시된 프라이머를 이용하여 중합효소 연쇄반응(PCR: polymerase chain reacttion)을 수행하였다. 그 결과 면역조절 단백질(SIRPα 단백질)(서열번호 3)을 암호화하는 유전자(서열번호 4)에 링커 펩티드(서열번호 13)를 암호화하는 유전자(GGT GGC TTT CTG GGT GGC GGT GGC TGC GGT; 서열번호 14)가 융합된 면역조절 융합단백질이 도입된 유전자(서열번호 2)를 얻었다. 상기 증폭된 면역조절 융합단백질 유전자를 pet28a 벡터의 Nde1과 HindIII 제한효소 자리에 삽입하여 발현 벡터를 제조하였다.
상기 서열번호 3으로 표시되는 면역 조절 단백질(A)을 대신하여 서열번호 3으로 표시되는 variant SIRPα 단백질, 서열번호 11로 표시되는 wild type SIRPα 단백질, 서열번호 12로 표시되는 wild type SIRPg 단백질, 서열번호 13으로 표시되는 SIRPg variant 1 단백질 또는 서열번호 14로 표시되는 SIRPg variant 2을 사용하여 융합 단백질을 각각 제조하였으며, 실질적으로 이들 각각을 사용하여 실시예 1과 같이 융합 단백질을 제조하였을 때 융합 단백질이 제조됨을 확인하였다.

상기 His-tag는 추후 상기 유전자가 융합 단백질로 발현될 때, 발현된 융합 단백질의 C-말단에 부착되어 단백질의 정제 및 확인 가능하게 하는 특징이 있다. 상기 서열번호 7, 8을 사용하여 유전자를 얻고, 이를 pet28a 벡터의 Nde1과 HindIII 제한효소 자리에 삽입하여 pET-His 발현 벡터도 제조하였다.
이후, 대장균 BL21(DE3, Novagen 사)에 CaCl2 버퍼를 사용하여 수용(competent) 세포를 만들었다. 이후, 42 ℃에서 2분간 방치하는 열충격 방법을 이용하여 상기 수용 세포를 상기 각각의 발현 벡터로 형질전환하였다. 상기 형질전환체를 증에서 융합 단백질을 발현시키는 형질전환체를 선별하고, 12시간 동안 30℃ 배양기에서 배양하였다. 이후 상기 배양된 형질전환체를 4,000 g에서 10분간 원심분리하였고, 상등액을 제거한 후 침전물 내에서 세포를 회수하였다. 회수한 세포들은 초음파 해체기(sonicatior)를 사용하여 파쇄하였다. 상기 수득된 세포 용해물을 4℃에서 14,000 rpm으로 20분동안 다시 원심분리하여 상층액을 수득하였고, IMAC Kit His tag 흡착 컬럼(Bio-Rad Laboratories, 미국)이 장착된 고속 단백질 액체 크로마토그라피(Fast Protein Liquid Chromatography)(Bio-Rad Laboratories, 미국)를 이용하여 상기와 같이 수득된 상층액으로부터 과발현된 서열번호 1의 아미노산 서열을 갖는 융합 단백질을 분리하였다.
실시예 2. 융합 단백질-siRNA 복합체 제조
5'- GGG AUA UUA AUA CUA CUU CAG UAC A*C *A (*DNA)-3'(서열번호 17)의 서열을 갖는 센스가닥과 5'-UGU ACU GAA GUA UUA AUA UCC CCG-3'(서열번호 18)의 서열을 갖는 안티센스 가닥의 siRNA(CD47 siRNA)를 사용하였고, 이는 엠바이오텍(IDT korea, Gyeonggi, South korea)에 의뢰하여 구입하였다. 상기 서열번호 17에서 26번째 27번째 염기서열은 DNA이다.
상기 siRNA과 융합 단백질(실시예 1)과 복합체가 형성되기 위해, 상기 수득한 siRNA 센스 가닥의 3' 말단에 말레이미드를 도입하였고, 구체적으로 상기 siRNA와 maleimide-NHS를 pH 8.3 0.1M HEPES 완충액 하에서 4시간동안 반응시킨 후, NAP 컬럼으로 siRNA만을 정제한 후 동결건조하였다.
HEPES 완충액(pH 7.4) 존재하에서, 상기 실시예 1로부터 제조된 융합 단백질에 상기 말레이미드가 도입된 siRNA를 1 : 1의 몰비로 첨가한 후, 반응이 완료되면 30k 아미콘튜브를 사용해 정제하여 융합 단백질-siRNA 복합체를 제조하였다.
상기 siRNA에 도입된 말레이미드(maleimide)와 상기 융합 단백질의 말단에 존재하는 시스테인의 티올(thiol)기가 1 : 1로 결합하여 융합 단백질-siRNA 복합체를 형성한다. 상기 융합 단백질-siRNA 복합체는 NAP-10 컬럼을 이용하여 정제하고, 동결 건조하였다.
시험예 1. 융합 단백질(SIPRα-linker)의 발현 확인
실시예 1에서 제조된 선별된 형질전환체로부터 콜로니를 추출하여 상기 표 1의 프라이머를 사용하여 PCR을 수행하였다. 이로부터 수득한 PCR 산물에 대한 젤 이미지 시스템(Geldoc-it 300, UVP, CA, USA)으로 측정하였다. 확인된 밴드는 약 400bp로 넣어준 유전자 서열의 밴드와 일치하는 것을 확인하였다.
또한 실시예 1로부터 분리·정제한 융합 단백질을 SDS-PAGE용 버퍼(0.5M Tris-[0113] HCl (pH 6.8), 10% glycerol, 5% SDS, 5% β-mercaptoethanol, 0.25% bromophenol blue)에 희석한 후 100ㅀC에서 15분간 끊여 변형시켰으며, 이 후 15% SDS-PAGE 젤에서 전기영동하였다.
도 3은 실시예 1에서 제조된 선별된 형질전환체로부터 DNA를 분리하고, PCR을 수행하여 젤 이미지 시스템(Gel documentation)으로 분석한 결과로, 이에 따르면 형질전환체에 정상적으로 융합 단백질을 암호화하는 유전자가 삽입된 것을 확인하였다.
도 4는 실시예 1로부터 분리·정제한 융합 단백질의 SDS-PAGE로 확인한 결과로, 1 레인(lane)은 발현 벡터를 넣지 않은 대조군(control)에 대한 결과이고, 2 레인은 융합 단백질(SIPRα-linker)에 대한 결과를 나타낸 것이다. 이에 따르면, 융합 단백질이 발현 벡터에 의해 정상적으로 발현됨을 확인하였다.
시험예 2. 융합 단백질-siRNA 복합체의 합성 확인
융합 단백질-siRNA 복합체가 실시예 2로부터 정상적으로 합성되었지를 확인하고자 하였다.
도 5는 실시예 2로부터 분리·정제한 융합 단백질-siRNA 복합체의 결합을 6X DNA gel loading dye(Thermo Scientific, MA, USA)를 통해 염색한 후 1% 아가로스 젤에 로딩하여 확인하였다. 1 레인(lane)은 CD47 siRNA를 대조군(control)으로 한 결과이고, 2 레인은 융합 단백질-siRNA 복합체(SIRPα-Cd47sirna)에 대한 결과를 나타낸 것이다. 이에 따르면, 융합 단백질-siRNA 복합체가 정상적으로 형성되었음을 확인하였다.
시험예 3. 융합 단백질-siRNA 복합체의 효소 특이성 분석
실시예 2로부터 제조된 융합 단백질-siRNA 복합체가, 의도한 바와 같이 리소좀 내 분해효소에 의해 절단되는지를 확인하고자 하였다. 우선 실시예 2로부터 제조된 융합 단백질-siRNA 복합체에 파파인(papain) 효소를 처리하고, 상온에서 10분간 효소 반응시킨 후, HPLC(고성능 액체 크로마토그래피; high-performance liquid chromatography)를 수행하였다.
HPLC용 시료는 실시예 2로부터 제조된 융합 단백질-siRNA 복합체를 30K 아미콘 튜브(Merck Millipore, MS, USA)로 정제 및 농축하여 수득하였다, 상기 시료를 c-18 컬럼(Xbridge C18, Waters, MS, USA)에 가하여 흡착시키고, 0.1% TFA/ACN 용액 5% 내지 95% 농도구배 용액으로 30분 동안 용출시켜 HPLC(Agilent Technologies, CA, USA)로 분석하였다. 대조군으로 파파인 효소를 처리하지 않은 실시예 2로부터 제조된 융합 단백질-siRNA 복합체를 사용하였다.
도 6은 실시예 2로부터 분리·정제한 융합 단백질-siRNA 복합체를 HPLC로 분석한 결과이고, 도 7은 실시예 2로부터 분리·정제한 융합 단백질-siRNA 복합체를 파파인 효소로 처리한 후, HPLC로 분석한 결과이다.
도 6 및 도 7에 나타난 바와 같이 실시예 2로부터 분리·정제한 융합 단백질-siRNA 복합체일 때는 검출되지 않다가, 파파인 효소 처리한 후 SIRPα 단백질이 검출되었다. 이를 통해 파파인 효소에 의해 실시예 2로부터 분리·정제한 융합 단백질-siRNA 복합체 내에 존재하는 링커 펩티드가 분해되어 SIRPα 단백질이 분리되어 나옴을 알 수 있다.
시험예 4. 융합 단백질-siRNA 복합체의 세포 내 전달능 검증
생체 외에서(In vitro), 실시예 2로부터 제조된 융합 단백질-siRNA 복합체가 세포 내로 효과적으로 섭취되는지 확인하고자 하였다. 우선, 인간유래 대장암 세포인 HT-29 세포를 35 ㎜ 커버글라스 바텀디쉬에 2 x 105 개씩 분주하였다. 여기에 SIRPα 단백질(SIRPα), CD47 siRNA, 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIPRα-Cd47 siRNA)를 각각 투여하여 37 ℃에서 하루동안 배양하였다. 상기 세포에 toto3 염색약을 사용하여 CD47 SiRNA을 적색으로 염색하였다. 다음으로 상기 세포에 실시예 2로부터 분리ㅇ정제한 융합 단백질-siRNA 복합체를 100pmol/ml 농도로 처리하고, 4시간동안 이산화탄소 배양기에서 37 ℃로 배양하였다. 이후, 상기 세포에 고정액을 처리하고, FITC가 붙은 His-tag 항체를 사용하여 SIRPα 단백질을 녹색으로 염색하였다. 여기에 4,6-디아미디노-2-페닐인돌(4′,6-diamidino-2-phenylindole, DAPI) 용액을 10 분간 처리하여 세포의 핵을 염색하였고, 이들 각각의 형광을 공초점 현미경으로 측정하였다.
비교예로 실시예 2로부터 제조된 융합 단백질-siRNA 복합체와 CD47 항체(Ab)를 혼합한 혼합액을 사용하였다.
도 8은 HT-29 세포에서의 실시예 2로부터 제조된 융합 단백질-siRNA 복합체의 거동을 확인하기 위한 공초점 현미경(confocal microscopy) 분석 결과이다. 도면에서 청색은 세포의 핵이고, 적색은 CD47 siRNA이며, 녹색은 SIRPα 단백질을 나타낸다.
도 8에 나타난 바와 같이, 실시예 2로부터 제조된 융합 단백질-siRNA 복합체는 세포 내로 충분히 흡수되는 것을 확인하였다. 이에 반해 CD47 siRNA가 단독으로 사용될 경우에는 세포 내로 전혀 흡수되지 못함을 확인하였다.
또한, 실시예 2로부터 제조된 융합 단백질-siRNA 복합체를 CD47 항체와 함께 처리할 경우, 세포 내로 흡수되는 양이 현저히 저하되는 것을 알 수 있다. 이를 통해 실시예 2로부터 제조된 융합 단백질-siRNA 복합체가 CD47 수용체에 결합하여 세포 내로 함입됨을 알 수 있다.
즉, 생체 외(in vitro) 실험을 통해 본 발명에 따른 실시예 2로부터 제조된 융합 단백질-siRNA 복합체는 암세포의 표면에 존재하는 수용체에 특이적으로 결합함과 동시에, 효과적으로 암세포 내로 흡수되어 CD47 수용체와 결합하여 암세포의 면역 회피 기전을 억제함을 확인하였다.
시험예 5. HPRT와 CD47 유전자 침묵 효과 확인
실시예 2로부터 제조된 융합 단백질-siRNA 복합체가 HPRT와 CD47 유전자(gene)에 대해 침묵 효과를 갖는지 확인하고자 하였다.
쥐의 대장암 세포인 CT26.CL25를 무혈청 배지에 분주하고, 실시예 2로부터 제조된 융합 단백질-siRNA 복합체를 투여한 후 4시간 동안 배양하였다. 상기 배양액을 제거한 뒤, 혈청 배지로 바꿔주고 2 일동안 더 배양하였다. 배양이 완료된 세포를 회수하여 RNA 추출 키트(Rneasy Plus mini kit, Qiagen, Mettmann, Germany)를 이용해 RNA를 분리하였다. 상기 분리된 RNA를 HPRT 유전자 프라이머(F: GGC TAT AAG TTC TTT GCT GAC CTG C, R: GCT TGC AAC CTT AAC CAT TTT GGG) 과 CD47 유전자 프라이머(F: AGG AGA AAA GCC CGT GAA G, R: TGG CAA TGG TGA AAG AGG TC)를 이용하여 RT-PCR 하였다. 상기 증폭된 PCR로부터 1 ㎍을 취해 GoScript reverse transcription system을 이용하여 cDNA로 합성하였다. 상기 합성된 cDNA는 1% agarose gel에서 분리하였고, gene stain detection 용액을 이용하여 이미지 장비로 관찰하였다.
본 실험에서는 세포 수준에서 HPRT와 CD47 유전자 침묵 효과를 검증하기 위하여 아무것도 처리하지 않은 PBS 군, 단독 siRNA만을 처리한 것, Scramble RNA를 처리한 군, Lipo 단백질과 siRNA의 복합체(비교예 1)를 처리한 군, 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)를 투여한 군(-), 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)과 CD47 항체(Ab)를 함께 투여한 군(+)으로 나누어 실험하였다.
도 9는 암세포(CT26.CL25)에 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)를 비롯한 다양한 시료를 각각 처리한 후, HPRT 유전자 발현 침묵 효과를 측정한 결과이다. 이에 따르면 HPRT 유전자의 발현량에는 변화가 거의 나타나지 않음을 알 수 있다.
도 10은 암세포(CT26.CL25)에 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)를 비롯한 다양한 시료를 각각 처리한 후, CD47 유전자 발현 침묵 효과를 측정한 결과이다. 이에 따르면 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)는 리포펙타민(lipofectamine)-siRNA 복합체와 유사한 수준으로 CD47 유전자 발현을 감소시키는 것을 확인하였다.
이에 반해, CD47 항체가 함께 투여된 군(+)이나, siRNA 단독으로 처리된 군에서는 CD47 유전자 발현이 감소하지 않음을 확인하였다.
즉, 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)는 CD47 유전자의 발현을 감소시키는데 효과적임을 확인하였다.
시험예 6. 세포 내 독성 확인
실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)의 세포 내 독성을 확인하고자 하였다. 이를 위해 쥐의 대장암 세포(CT26.CL25)에 0, 50, 100, 200, 400, 800 nM의 농도로 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)과 비교예 1의 Lipo-CD47 siRNA 복합체를 각각 처리하고, 72시간 동안 배양하였다. 다음 CCK-8(cell counting kit-8) 키트를 이용해 분석하였다.
도 11은 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)과 비교예 1의 Lipo-CD47 siRNA 복합체의 세포독성을 CCK-8을 통해 평가한 결과이다. 이에 따르면 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)를 고농도(800 nM)로 처리하여도 80% 이상의 세포생존율(cell viability,%)를 유지하고 있음을 확인하였다.
이에 비해, 비교예 1의 Lipo-CD47 siRNA 복합체는 200 nM 농도에서 독성을 나타내는 것으로 확인되었다.
시험예 7. 대식 작용 증대 효과 확인
실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)가 대식세포의 식작용을 증대시킴을 검증하고자 하였다.
우선, CT26.CL25 세포에 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)를 200 pmol/ml의 농도로 처리하고, 이틀 간 10% FBS와 1% antibiotics antimycotics 을 포함한 RPMI 1640 배지(Welgene, Gyeongsan, South Korea)에 배양하였다. 배양이 완료된 후 CT26.CL25 세포를 pH rodo(pHrodo Red, Invitrogen, CA, USA)로 염색하고, 여기에 실험쥐(C57bl/6, 수컷, 6주령)의 골수에서 추출한 대식세포(Bone marrow-derived macrophage, BMDM)를 투여하여 같이 RPMI 1640 배지에서 3시간동안 배양하였다. 이때 상기 대식세포는 Green cell tracker(Thermo Fisher scientific, MS, USA)로 염색하였으며, 이들 각각의 형광을 형광 현미경(Nikon, Japan)으로 측정하였다.
대조군으로 아무것도 처리하지 않은 SIPRα 단백질만을 처리한 군을 사용하였다.
도 12는 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)의 대식세포 식작용 효과를 분석한 결과이다. 이에 따르면 실시예 2로부터 제조된 융합 단백질-siRNA 복합체(SIRPα-CD47 siRNA)는 siRNA를 단독으로 사용했을 때보다 더 대식세포의 식작용을 향상시키는 것으로 확인되었다.
상술한 결과를 통해서 일반적인 조건에서 본 발명에 따른 융합 단백질-siRNA 복합체는 면역조절 단백질 SIRPα과 siRNA가 세포에 대해 독성을 나타내지 않는다. 종양세포가 존재하면 종양세포 표면에 존재하는 수용체에 특이적으로 반응하여 세포 내로 흡수되게 된다. 세포 내로 흡수된 융합 단백질-siRNA 복합체는 세포 내에 존재하는 분해효소에 의해 면역조절 단백질 SIRPα과 siRNA로 쉽게 분해되어 암 세포의 면역을 억제하고 CD47 유전자 침묵을 유도하며, 암세포에 대한 대식세포의 식작용을 향상시켜 극대화된 항암효과를 유도하는 것을 확인하였다. 또한 siRNA의 세포 내 전달능력이 단독으로 사용될 때 보다 세포 내로 전달되는 siRNA 양이 증대되었으며, CD47 유전자에 대한 침묵 효과 또한 향상됨을 알 수 있다.
종양세포에 과다하게 존재하는 면역 조절 관련 수용체를 이용해 종양세포에 특이적으로 전달할 뿐 만 아니라 관련 기작을 억제할 수 있으며, 또한 여기에 같은 수용체를 표적화하는 치료제를 접목시켜 이중 치료효과를 나타내는 새로운 이중제어형 면역조절 단백질 표적 siRNA 합성체에 관한 것이다.
siRNA를 면역 조절 기능 단백질을 전달체로 이용함으로써, 이를 통해 암세포 수용체를 통해 특이적으로 전달할 뿐 만 아니라 면역 관련 수용체의 유전자를 표적함으로써 이를 이중으로 제어하는 이중제어형 siRNA 기반 항암면역치료기법이다.
면역 조절 단백질에 siRNA를 결합하여 종양세포 내로 들어가 자발적으로 리소좀 내에서 절단되어 활성화되는 단백질-siRNA 복합체를 제공한다. 본 면역조절단백질- siRNA 복합체는 암세포의 전달 능력을 향상시켰을 뿐만 아니라, 단백질 및 siRNA를 통해 면역 방어기전을 이중으로 제어함으로써 항암 치료를 극대화할 수 있다는 점이다.
상기 면역조절단백질-siRNA 복합체는 종양세포에서 과발현하는 수용체를 매개체로 세포 내로 흡수되어 리소좀 (lysosome)에서 분해되어 약효를 작동한다. 이로 정상세포의 무분별한 독성을 줄일 수 있을 뿐만 아니라 세포 내에서 효율적으로 siRNA가 작동할 수 있다.
Claims (8)
- 면역조절 융합단백질(A); 및 siRNA(B);를 포함하되,상기 면역조절 융합단백질(A)은 SIPRα 단백질과 리소좀 분해효소로 절단되는 링커 펩티드가 결합된 것을 특징으로 하는 융합 단백질-siRNA 복합체.
- 제1항에 있어서,상기 SIPRα 단백질은 서열번호 3, 5, 7, 9, 11 중에서 선택되는 어느 하나로 표시되는 아미노산 서열이고, 상기 링커 펩티드는 서열번호 13으로 표시되는 것을 특징으로 하는 융합 단백질-siRNA 복합체.
- 제1항에 있어서,상기 면역조절 융합단백질(A)은 서열번호 1로 표시되는 것을 특징으로 하는 융합 단백질-siRNA 복합체.
- 제1항에 있어서,상기 siRNA는 CD47 siRNA인 것을 특징으로 하는 융합 단백질-siRNA 복합체.
- 제1항에 있어서,상기 siRNA는 서열번호 17 또는 18로 표시되는 것을 특징으로 하는 융합 단백질-siRNA 복합체.
- 제1항에 있어서,상기 융합 단백질-siRNA 복합체의 평균 분자량은 20 kDa 내지 60 kDa인 것을 특징으로 하는 융합 단백질-siRNA 복합체.
- 제1항 내지 제6항 중 어느 한항에 따른 융합 단백질-siRNA 복합체를 유효성분으로 포함하는 암 치료 또는 예방용 약학 조성물.
- 제7항에 있어서,상기 암은 뇌종양, 척수종양, 망막 세포종, 구강암, 비강암, 부비동암, 인두암, 후두암, 경부암, 두경부암, 흑색종, 피부암, 유방암, 갑상선암, 악성부신종양, 내분비암, 폐암, 흉막종양, 호흡기암, 식도암, 위암, 소장암, 대장암, 항문암, 간암, 담도암, 췌장암, 신장암, 방광암, 전립선암, 고환암, 음경암, 자궁경부암, 자궁체부암, 융모상피암, 난소암, 급성, 만성 백혈병, 악성림프종, 다발성 골수증을 포함하는 혈액암, 골종양, 연부종양, 소아 백혈병 및 소아암으로 이루어진 군으로부터 선택된 어느 하나 이상인 것을 특징으로 하는 암 치료 또는 예방용 약학 조성물.
