도 4에 나타낸 바와 같이, 운동을 시키지 않은 정상군에 비하여 트레드밀 운동 실험을 실시한 음성대조군에서는 혈중 젖산 및 젖산탈수소효소의 수준이 증가하고, 간 내 글리코겐의 양은 감소하였다. 이에 반하여, 오이풀 추출물 투여 시, 혈중 젖산 및 젖산탈수소효소의 수준이 음성 대조군에 비하여 감소한 것을 확인하였으며, 간 내 글리코겐 수준은 음성 대조군에 비하여 증가한 것을 확인하였다. 따라서, 오이풀 추출물이 피로 물질인 젖산과 젖산 탈수소효소는 낮추며, 간 내 글리코겐은 증가시키는 효과를 나타내는 것을 확인하였다. 또한, 양성대조군으로 사용한 타우린과 비교하여, 오이풀 추출물이 현저히 적은 농도에서도 유사한 효과가 나타나는 것을 확인하였다. 미토콘드리아의 수와 활성은 PGC-1α(peroxisome proliferator-activated receptor gamma coactivator 1-alpha)에 의해서 조절되며, AMPK(AMP activated protein kinase)는 세포 내 에너지 항상성 유지 역할을 하는 효소로써 운동이나 스트레스에 의해 ATP가 고갈되어 AMP/ATP 비율이 증가하는 경우 활성도가 증가하게 된다. 오이풀 추출물이 근육 내 세포 에너지 대사에 미치는 영향을 확인하기 위해, 근 조직 내 PGC-1α 및 AMPK 수준을 측정하는 실험을 실시하였다. 구체적으로, 실시예 5에서 채취한 근 조직 50mg 에 RIPA 완충액 300 μL를 넣고 4 ℃에서 12,000 rpm 으로 20 분 간 원심 분리하였다. Bradford 어세이 방법으로 단백질을 정량한 후, 단백질의 SDS-PAGE (Polyacrylamide Gel Electrophoresis)를 이용하여 크기 별로 분리하였다. Semidry transfer system (Bio-Rad, USA)를 이용하여 PVDF 막에 이동시킨 후, 5% 탈지 우유(skim milk)를 함유한 차단 완충액(0.5% 탈지 우유(skim milk), 1×TBS/T 완충액)을 1시간 동안 처리하였다. 1×TBS/T 완충액으로 10 분, 3회 세척 후, 일차 항체인 PGC1α, p-AMPK, AMPK, GAPDH를 1:1000의 비율로 처리한 후, 4 ℃에서 밤새 반응시켜, 1×TBS/T 완충액으로 10분, 3회 세척하였다. 웨스턴 블롯 검출 키트 (Biorad, hercules, USA)을 이용하여 막과 반응시킨 후 Chemi-Doc 장비를 이용하여 발현 정도를 관찰하였으며, 그 결과를 도 5에 나타내었다.The number and activity of mitochondria is regulated by peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α), and AMP-activated protein kinase (AMPK) is an enzyme that maintains energy homeostasis in cells. After washing 3 times for 10 minutes with 1×TBS/T buffer, primary antibodies PGC1α, p-AMPK, AMPK, and GAPDH were treated at a ratio of 1:1000, and then reacted at 4°C overnight, 1×TBS/T Washed 3 times for 10 minutes with buffer. After reacting with the membrane using a Western blot detection kit (Biorad, hercules, USA), the expression level was observed using Chemi-Doc equipment, and the results are shown in FIG. 5 .
