Es wurden je 1 Mio. Melanozyten in 75cm2-Zellkulturflaschen ausgesät und in Medium (Cascade Biologics 254 von Gibco) kultiviert. In den nächsten 3 Wochen wurde 2-Mal die Wo- che ein Medienwechsel durchgeführt. Flasche 1 beinhaltete Medium mit 0,1% DMSO als Lö- semittelkontrolle und Flasche 2 beinhaltete Medium mit 5mM des Alkylamidothiazols N-(4-(2,4- dihydroxyphenyl)thiazol-2-yl)isobutyramide (= Isobutylamido-thiazolyl-resorcinol = Thiamidol, gelöst in DMSO). Nach dem letzten Medienwechsel wurden die Melanozyten weitere 2 Wochen kultiviert (ohne Mediumwechsel, damit das sezernierte Melanin akkumuliert).
Nach den zwei Wochen wurden 10 ml des jeweiligen Überstandes, in welches die Melanozy- ten Melanin sezerniert haben, in jeweils ein Well einer 6-Loch-Platte gegeben. Auf einem „Colour Laser Transparency Film“ (3M; CG3700) wurde ein Motiv mit verschiedenen farbigen Rechtecken ausgedruckt. Dieses Motiv wurde durch die mit den Überständen gefüllten Plas- tikschalen auf einem Leuchttisch (Kaiser Fototechnik, eVision ini.tial:hf) fotografiert (Canon EOS-1 Mark IV mit Canon Compact-Macro Lens EF 50 mm; ISO 100, f 9.0, 1/80, WB Leuchtstoff). Mit Hilfe der Software „imagej“ wurden die Bilder analysiert und die RGB-Kanäle der jeweili- gen Flächen wurden ausgewertet. Die RGB-Werte wurden in Lab-Werte umgerechnet (http://colormine.org/convert/rgb-to-lab). Aus den Lab-Werten wurden die Farbunterschiede ΔE (ΔE = (ΔL2 + Δa2 + Δb2)1/2) innerhalb eines Bildes berechnet. Der Vergleich der entspre- chenden ΔE-Werte zwischen den Bildern (Thiamidol vs. unbehandelt) zeigt, dass die Far- bunterschiede bei der Aufnahme durch den Überstand der mit Thiamidol behandelten Mela- nozyten stets größer sind als bei der entsprechenden Aufnahme der unbehandelten Mela- nozyten (Fig.1 und 2). Verwendung nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass X aus der Gruppe der substituierten Phenylgruppen gewählt wird, insbesondere ge-
