본 발명은 신규한 가돌리늄계 화합물, 이의 제조 방법, 및 이를 함유하는 MRI 조영제에 관한 것이다. 구체적으로, 본 발명은 연결기를 통해 가돌리늄 착물과 로즈 벵갈(Rose Bengal)이 결합된 구조를 갖는 신규한 가돌리늄계 화합물, 이의 제조 방법, 및 이를 함유하는 MRI 조영제에 관한 것이다.
오늘날 인구의 고령화로 인하여 퇴행성 뇌질환 환자가 증가하고 있으며, 이에 따라, 해당 질병에 대한 조기 발견의 필요성이 대두되고 있다. 퇴행성 뇌질환으로는 파킨슨병, 혈관성 치매, 알츠하이머병 등이 있고, 해당 질환의 발생 원인 중 하나로 글루타메이트(glutamate)의 과발현으로 인한 신경 독성이 고려되고 있다.
인간의 뇌에서 흥분성 신호전달의 70% 이상을 담당하는 글루타메이트는 학습, 기억, 운동능력, 및 감정을 조절하는 중요 아미노산이다. 시냅스에서 글루타메이트에 의한 흥분성 신호 전달은 여러 글루타메이트 수송체 및 수용체에 의해 엄격하게 조절되고 있다. 소포성 글루타메이트 수송체(vesicular glutamate transporter)(VGLUT)는 뉴런 세포에서 글루타메이트의 저장과 농도 조절에 핵심적인 역할을 한다고 알려져 있다. 평상시 시냅스 간극에서의 글루타메이트의 농도는 1-3 μM이지만 소포 내에 저장되어 있던 글루타메이트가 방출되면 농도는 수백~수천 μM 이상으로 올라간다. 따라서 VGLUT에 이상이 발생하면 소포에 저장되어 있던 글루타메이트가 비정상적으로 과량 분비되고, 최근 연구에 따르면, 이와 같이 글루타메이트 농도가 증가되면 신경아교세포에 의한 재흡수가 감소하여 글루타메이트가 신경 연접 부위에 축적되어 독성을 유발하고, 활성 산소종의 생성 및 산화적 스트레스를 유도하여 신경 세포의 손상을 초래한다고 한다.
또한, 최근에는, 알츠하이머병이 발병되는 초기에 뇌이랑(gyrus)이나 뇌척수액(cerebrospinal fluid)에서 글루타메이트 농도가 증가하였다가 알츠하이머가 진행되면서 신경세포가 손상을 입어 그 수가 감소한다는 연구 결과가 나온바 있다. 따라서, 글루타메이트의 농도 변화를 감지하는 것은 퇴행성 뇌질환의 조기 진단을 가능하게 할 것으로 예상하였다.
한편, 자기공명영상(Magnetic Resonance Image, 이하, MRI)은 체내 조직간 수소 원자의 분포가 다르고 자기장 안에서 수소 원자가 이완되는 현상을 이용하여 신체의 해부학적, 생리학적, 생화학적 정보 영상을 얻는 방법이다. MRI 는 CT 나 PET 과는 다르게 인체에 유해한 방사선을 사용하지 않고, 강한 자기장 하에서 자기장의 기울기 및 라디오파를 사용하여 신체 내부의 이미지를 생성하므로 비침습적이고 해상도가 높으며 연부 조직 검사에 뛰어나다.
이러한 MRI 장비를 좀 더 정밀하게 활용하기 위해서, 조영제(contrast agent)를 대상체에 주입하여 MRI 영상을 얻는다. MRI 이미지 상에서의 조직들 사이의 대조도(contast)는, 조직 내의 물분자 핵스핀이 평형상태로 돌아가는 이완(relaxation) 작용이 조직별로 다르기 때문에 생기는 현상이다. 조영제는 상자성을 띄거나 초상자성을 띄는 물질을 이용하여 상기 이완 작용에 영향을 끼쳐 조직 간의 이완도 차이를 벌리고 MRI 신호의 변화를 유발하여 조직 간의 대조를 보다 선명하게 하는 역할을 한다.
현재 임상적으로 가장 일반적으로 사용되고 있는 조영제는 가돌리늄(Gd) 킬레이트에 기반을 둔 조영제이다. 현재는 Gd-DTPA (Magnevist®), Gd-DOTA (Dotaram®), Gd(DTPA-BMA) (Omniscan®), Gd(DO3A-HP) (ProHance®), Gd(BOPTA) (MultiHance®) 등이 사용되고 있다. 그러나, 상용화되어 있는 조영제의 대부분이 세포 외공간(extracellular fluide, ECF)으로 분포되는 비특이적 조영제이다. 특이적 조영제로는 특별하게 간 특이적 조영제가 사용되고 있을 따름이다. 최근의 연구는 특정 표적성을 가지거나 생리적 활성 (pH 변화, 효소 활성) 에 의해 신호 증강을 나타낼 수 있는 조영제의 개발이 추진되고 있으나, 현재까지는, 특정 표적성을 가지는 MRI 조영제, 특히, 퇴행성 뇌질환에 대한 특이적 MRI 조영제에 대해 충분한 결과가 얻어지지 못하고 있다.
본 발명의 일 목적은 MRI 조영 물질로 사용될 수 있고, 특히, 퇴행성 뇌질환에 특이성이 있는 가돌리늄계 화합물을 제공하는 것이다.
본 발명의 다른 목적은 상기 화합물을 함유하는 MRI 조영제를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 화합물의 제조 방법을 제공하는 것이다.

본 발명의 상기 화학식 1의 가돌리늄계 화합물은 후술하는 실시예에서 보는 바와 같이, 포유동물의 소포성 글루타메이트 수송체(vesicular glutamate transporter)(VGLUT)에 특이적으로 결합할 수 있다.
또한, 본 발명의 상기 화합물은 수용성을 가지고, 적어도 하나 이상의 물 분자와 배위 결합하여 자기 이완 특성을 가지므로, 인체 내에서 적어도 하나 이상의 물 분자와 수소 원자 이완율을 증가시킴으로써 영상 대조를 향상시킬 수 있고, 이에, MRI 조영 물질로 사용될 수 있다. 본 발명의 화합물은 예를 들어, 3T 자기공명 영상에서 5 mM

이하에서는, 상기 제조한 본 발명의 대표 화합물을 사용하여, MRI 조영 물질로서 사용할 수 있고, 이와 동시에, 글루타메이트 수송체의 하나인 VGLUT1(vesicular glutamate transporter 1)에 표적능력이 있는지 여부를 알아보기 위한 특성 평가를 실시하였다.
2. 본 발명에 따른 화합물의 특성 평가 방법 및 결과
1) 이완율(relaxivity)에 대한 평가
상기 제조예에서 수득한 화합물
Gd-RB 을 10 mM 농도로 디메틸술폭시드(DMSO)/물(Water)(5:5) 에 용해시킨 후, 물로 희석하여 0에서 1 mM 사이의 다섯개 농도 샘플을 3개 세트로 만들었고, 이들을 MRI 영상을 얻는데 사용하였다. 이완율은 3.0 T 와 9.4 T 의 MRI를 이용하여 각각 측정하였고, T1 값은 반전회복법(Inversion recovery method) 으로 수행하였다. 구체적으로 설명하면, 3.0 T 에서 FSE-IR(fast spin echo-inversion recovery) sequence 로 50에서 1800 msec 범위의, 9.4 T 에서는 IR-RARE (inversion recovery-rapid imaging with refocused echoes) sequence 로 85에서 7000 msec 범위의 10개 이상의 서로 다른 반전시간(inversion time, TI)을 이용하였다. T2 측정은 CPMG (Carr-Purcell-Meiboon-Gill) 펄스 서열을 다중 스핀-에코(multiple spin-echo) 측정에 적용시켜 서로 다른 에코 시간(TE) 값을 이용하여 수행하였다. 구체적으로 설명하면, 상기 T2는 3.0 T 에서 T2 MAP sequence 로 8.5 내지 135 msec 범위의, 9.4 T에서 MSME(multi slice multi echo) sequence 로 10에서 700 msec 범위의 10 개 이상의 서로 다른 TE를 이용하여 측정되었다. T1, T2 이완시간은 각 에코 시간의 다중 스핀-에코 측정에 대한 평균 픽셀 값(mean pixel values)의 비-선형 최소제곱법으로부터 구하였다. 그런 다음, 이완성(R1 및 R2)은 mM 당 이완시간의 역으로 계산하였다. 계산된 이완시간(T1 및 T2) 및 이완성(R1 및 R2)으로 최종적으로 이미지-작업하여 각각 이완시간 지도 및 이완율 지도를 만들었고 이완율(r1 및 r2)을 얻었다. 그 결과를 하기 [표 1]에 나타낸다.

2)-(1). 실험 방법
① 쥐 뇌 신경세포 초대 배양
임신 18~9일 된 쥐 (Sprague Dawley rats)는 대한바이오링크에서 구입하였다. 임신 쥐를 안정화 시킨 후, 이소플루레인(isoflurane)을 이용하여 호흡 마취하고, 태아를 모체로부터 분리하였다. 태아의 뇌를 분리하여 막을 제거하고, 해마와 대뇌피질만을 분리 선택하여 잘게 잘라 HBSS 완충제에 넣어 4번 수세하였다 (원심분리 1000rpm, 3분). 마지막 수세 후, HBSS를 제거하고, 트립신이 첨가된 HBSS 완충제를 첨가(세포 부피: 트립신 HBSS = 1:1)하여 항온 수조 (37℃)에서 5분간 반응시켰다. 10% FBS가 든 HBSS 완충제를 첨가((세포부피+trypsin HBSS): 10% FBS HBSS = 1:1)하여 트립신의 효소 반응을 중지시키고, 4겹의 lens paper에 통과시켜 단일 세포로 만들었다. HBSS 완충제로 원심분리하여 4회 수세하고, 단일 세포로 분리된 세포만을 획득하였다.
② 팬텀 연구(Phantom study)
획득한 세포는 E-tube당 7×10
6 개의 세포로 1×N2 supplement, 1×B27, 4 mM L-glutamine, 1% 항생제가 든 neurobasal media에 부유시키고. 각 E-tube에 Gd-RB 또는 Gadovist를 0, 50, 100, 200, 400 μM의 농도로 첨가하여 37℃, shaker에서 24시간 동안 반응시켰다. 24 시간 후, 4000 rpm, 4℃에서 5분 동안 원심분리하여 상층액을 제거하고, PBS 1 mL 을 첨가하여 피펫팅하여 세포를 풀어 준 후, 같은 조건으로 원심분리를 3회 반복하여 세포와 결합하지 않은 물질들을 씻어 내었다. RIPA 용액 (150 uL)를 첨가하여 1시간 동안 4℃, 15분 간격으로 vortex하여 세포를 파쇄 하였다. 파쇄된 세포에 350 uL 의 PBS를 추가하여 총 500 uL 의 샘플을 만들고 이 상태로 MR 촬영하였다. MRI 는 3T에서 촬영되었으며 Fast spin echo sequence로 파라미터는 다음과 같다. 256*256 matrix size; number of acquisition (NEX) = 4; phase field of view (FOV) = 0.8; repetition time (TR) = 400 ms; echo time (TE) = 13.2 ms; echo train length (ETL) = 3; field of view (FOV) = 12 mm; slice thickness = 2.5 mm; spacing = 0. MRI 강도는 ImageJ 프로그램을 이용하여 분석되었다.
2)-(2). 실험 결과
상기 2)-(1)에서 실시한 MR 팬텀(Phantom) 이미지를 도 6a에 나타내었으며, 본 발명에 따른 화합물 GD-RB8 및 상용 조영제 Gadovist 각각의 농도에 따른 팬텀 신호 강도의 변화를 도 6b에서 그래프로 나타내었다. 여기서, 대조군(control)은 세포만 있고 다른 조영제를 섞지 않은 샘플에 대한 것이다. 도 6a 및 6b 에서 보는 바와 같이, Gadovist를 일정 개수의 세포에 처리하였을 경우, 처리한 Gd 농도에 따른 밝기 차이가 거의 없었다. 이에 반해, 상기 제조한 본 발명에 따른 화합물
Gd-RB 을 Gd 농도별로 처리하였을 때, 처리한 Gd 농도가 증가함에 따라 팬텀의 밝기도 밝아지는 것을 확인할 수 있었다. 이로부터, VGLUT1 이 존재할 때 화합물 Gd-RB 은 VGLUT1 과 결합한다는 것을 확인할 수 있었고, 화합물 Gd-RB 의 농도에 비례하여 결합이 더 증가한다는 사실도 또한 확인하였다. 오른쪽은 각 팬텀에서의 신호 강도를 측정하여 처리한 Gd 농도에 따른 신호 강도의 변화를 그래프로 표현하였다.
3) 면역형광 염색법(Immunofluorescence staining)에 의한

3)-(1). 실험 방법
초대 배양한 세포를 4mM L-글루타민을 포함한 Neurobasal™ Medium (cat. No. 21103049, Gibco)를 기반으로 하여 Cell Culture Slide 4well (cat. No. 30104, SPL)에 각 well 당 4*10^5개를 seeding 하였다. 24시간 동안 배양 후 L-글루타민이 포함되지 않은 Neurobasal™ Medium을 기초로 하여 차광된 상태를 유지하며 화합물 Gd-RB 을 각각 0, 100, 200, 400 uM 농도로 10분간 처리하고, DPBS 완충제로 3회 수세하였다.
세포는 4% 파라포름알데히드(PFA) 에 15분간 고정 시킨 뒤, TBS 완충제로 5분간 3회 수세하고, 5% normal gout serum (NGS) 과 5% bovine serum albumin (BSA) 를 포함한 TBS 완충제로 1시간 동안 blocking을 실시하였다. VGLUT1 Polyclonal Antibody (Cat. No. 48-2400, invitrogen)는 5% NGS 와 5% BSA 를 포함한 TBS buffer와 1:250의 비율로 희석하여 4
oC에서 하룻밤 동안 반응시키고, TBS 완충제로 15분간 3회 수세한 뒤, 2차 항체인 Goat anti-Rabbit Alexa Fluor 488 (Cat. No. A-11008, invitrogen)을 5% NGS 와 5% BSA가 포함된 TBS 완충제와 1:500의 비율로 희석하여, 25
oC에서 1시간 30분간 암 반응하였다. TBS 완충제로 10분간 3회 수세한 뒤, VECTASHIELD® Hardset™ Antifade Mounting Medium with DAPI (Cat. No. H-1500 VECTOR) 로 Mounting을 실시하고, lionhart 장비를 사용하여 형광 염색된 세포의 이미지를 획득하였다. 렌즈는 20x 대물렌즈를 사용하고, 형광 강도는 lionhart 소프트웨어를 사용하여 ROI 값을 분석하였다.
화합물 Gd-RB 는 형광현미경에서 595nm (적색) 파장에서 검출이 가능하기 때문에 VGLUT1(도 7a에서 "VGLUT1"이라는 표시 하에 도시한 도면)과 Gd-RB 의 사진 (도 7a에서 "Rose Bengal"이라는 표시 하에 도시한 도면)을 병합(merge)하여 co-localization (도 7a에서 "Merged"이라는 표시 하에 도시한 도면)되는 값을 분석하였다 (도 7a 및 도 7b 참조). 도 7a는 형광 이미지를 나타내고, 도 7b는 화합물 Gd-RB 농도에 따른 각 형광 강도를 그래프로 나타낸 것이다.
3)-(2). 실험 결과
도 7a를 참조하면, 로즈 벵갈의 형광 (도 7a에서 "Rose Bengal"이라는 표시 하에 도시한 도면)을 관측하고 VGLUT1 에 대한 항체의 형광 (도 7a에서 "VGLUT1"이라는 표시 하에 도시한 도면)을 관측하여 두 이미지를 겹쳐 보았을 때 (도 7a에서 "Merged"이라는 표시 하에 도시한 도면), 두 영상이 서로 겹쳐지는 것을 확인하였다. 상기 결과를 통해, 본 발명에 따른 화합물 (Gd-RB)은 VGLUT 를 잘 표적한다는 것을 확인할 수 있으며, 이에 따라 본 발명에 따른 화합물은 VGLUT에 대한 표적성 MRI 조영제로 적합하게 이용될 수 있을 것으로 볼 수 있다.

5XFAD 유전자삽입 AD 마우스 모델 및 대조군으로서 야생-형 C57BL/6 마이스를 대구경북첨단의료산업진흥재단(DGMIF)의 동물실험윤리위원회(Institutional Animal Care and Committee)의 승인에 의해 사용했고 모든
in vivo 실험을 승인된 프로토콜에 따라 수행했다.
in vivo MR 이미징을 위해, 12개월 수컷 5XFAD 마이스 및 C57BL/6 마이스를 이소플루레인(isoflurane)으로 마취했다. 선-이미지 습득 이후, DMSO 내 50 mM Gd-RB를 0.1 mmol/kg 의 투여량으로 뇌실 내 (ICV) 주사했고 3시간 동안 이미지화했다. 9.4 T 스캐너 (Bruker, BioSpin, Germany)를 사용하여 도 8a의
in vivo 동물 실험을 통해 얻은 뇌 MR 영상을 “후-전”처리 이후에 컬러맵핑하여 나타낸 도면을 얻었다. T1-가중치 이미지에 대한 이미징 매개변수는 다음과 같다: TR = 700 ms; TE = 7 ms; 20 mm FOV; 128 x 128 매트릭스 크기; 0.5 mm 슬라이스 두께.
4)-(2). 이미지 분석
MATLAB 소프트웨어 (R2015a, MathWorks Inc., Natick, MA, USA)를 사용하여
in vivo 이미지 감산 및 컬러 맵핑을 수행했다.
컬러 맵핑된 이미지를 식 (1)을 통해 신호 강도 값으로 표시했다.

4)-(3). 사후 뇌 절편의 면역조직화학
in vivo MR 이미지 습득 다음, 주사로부터 3 시간 이후 마우스 뇌를 적출했다. OCT 화합물 (Tissue-Tek, Sakura Finetek, USA) 내에서 조직을 완전히 동결시켰을 때, 동결 마이크로톰 (Leica Biosystems, Wetzlar, Germany)을 사용하여 약 20 μm 두께의 조직 절편으로 몇몇의 부분 동결-절편으로 조직을 박절했고 현미경 유리 슬라이드 상에 마운트했다. 면역조직화학적 염색을 위해 뇌 당 해마를 포함하는 평균 6-8의 절편을 사용했고 연구자가 블라인드로 분석했다. 해마를 포함하는 모든 절편을 0.1% 소듐 아지드를 함유하는 DPBS 용액 내 수집했고, 4°C에서 저장했다. 뇌 절편을 0.1% Triton X-100을 포함하는 TBS 및 3% 염소 혈청 (Gibco Co., Grand Island, NY, USA)으로 블로킹했고 25°C에서 2시간 동안 1차 항체 (항-VGLUT1)로 배양했다. 그 다음 뇌 절편을 TBS로 세척했고 Alexa Fluor (Cell Signaling Technology Inc., Beverly, MA, USA)와 함께 IgG-라벨링된 2차 항체의 존재 하에서 1 시간 동안 배양했다. DAPI 용액 내 절편을 10 분 동안 마운트했고 그 다음 형광 현미경 (ECLIPSE Ti, Nikon, NY, USA)으로 관찰했다. NIS-Elements Basic Research 이미징 소프트웨어 (버전 4.50)를 사용하여 이미지를 분석했다.
4)-(4). 실험 결과
12개월의 후기 치매 모델과 같은 개월수의 정상 쥐에 Gd-RB를 뇌실 내 주사하여 2시간 30분 후 얻은 MR 영상을 매트랩 프로그램을 이용하여 “후 - 전” 이미지를 얻고 컬러 맵핑을 하였다. 도 8a를 통해 치매 쥐에 비해 정상 쥐에서 더 많은 영역에서 신호 강도 변화가 있는 것을 볼 수 있고, 3시간 후 뇌 적출 후 면역형광염색을 시행한 후 얻은 결과인 도 8b에서도 같은 결과 (즉, 정상 쥐에서 더 많은 영역에 신호 강도 변화가 있음; 예를 들어 도 8b에서 "Merge"라는 표시 하에 도시한 도면에서 강도 차이 나는 부분을 흰색 화살표로 표시하였고 정상 쥐에서 흰색 화살표가 더 많음)를 확인할 수 있었다. 즉, 도 8b에서 또한 면역형광염색 결과에서 VGLUT1 항체(도 8b에서 "VGLUT1 antibody"라는 표시 하에 도시한 도면)와 Gd-RB (도 8b에서 "Gd-RB"라는 표시 하에 도시한 도면)의 신호가 겹쳐지는 것을 통해
in vivo 실험에서도 Gd-RB 의 VGLUT1 표적성을 확인할 수 있었다.
5) 세포 생존도(viability) 테스트
5)-(1). 세포 배양 및 세포 독성 실험
인간 유래 신경모세포종 세포(SH-SY5Y)를 96 well 플레이트의 1 well 당 1×10
4 개수로 10% FBS(Fetal bovine serum), 4mM 글루타민, 1% 항생제가 들어있는 MEM(Minimum Essential Medium)에 배양하였다. 세포를 24시간 동안 부착 및 안정화 시킨 후, 각 well당 성장 배지 100 μl에 Gd-RB과 Gadovist를 0, 50, 75, 100, 150, 200, 400 μM의 농도로 희석하여 처리하고 22시간 동안 배양하였다. 이후 각 well에 CCK-8 (Cell Counting Kit-8, Dojindo Laboratories, Kumamoto, Japan) 용액을 10 μl씩 첨가하여 2시간 추가 배양하였다. 배양이 완료된 플레이트는 microplate reader기를 이용하여 450nm에서 흡광도를 측정하였다.

그래프 그리기: 계산된 값은 GraphPad Prism 응용 프로그램을 이용하여 그래프를 그렸다. 나온 수치의 통계적 유의성은 일원 분산 분석 (One-way ANOVA with Dunnett’s multiple comparison test)을 통해 확인하였다. * p<0.05 ** p< 0.01, *** p<0.001 vs. 대조군의 유의성을 가짐을 나타낸다. 상기 계산법에 따라 구해진 세포 생존도를, 화합물 Gd-RB와 Gadovist 각각의 농도에 따른 그래프로 하여 도 9에 나타내었다.
5)-(2). 실험 결과
도 9를 참조하면, 화합물 Gd-RB 과 Gadovist를 농도별로 SH-SY5Y 세포에 처리하였을 때, 두 군에서의 세포 생존 정도가 유사하게 측정되었다. 이로써, 본 발명의 화합물 Gd-RB의 세포 독성은 상용 조영제와 유사함을 확인하였다.
상기에서는 본 발명의 바람직한 실시예를 참조하여 설명하였지만, 해당 기술 분야의 숙련된 당업자는 하기의 특허 청구 범위에 기재된 본 발명의 사상 및 영역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경시킬 수 있음을 이해할 수 있을 것이다.

- 하기 화학식 1로 표시되는 것을 특징으로 하는 가돌리늄계 화합물;[화학식 1]상기 화학식 1에서,A는 *-(CH 2) n-A 1-*를 나타내고,n은 0 내지 5 중 임의 정수를 나타내고,A 1 은 *-COO-*, *-CO-*, *-NR 1-*, *-CH 2-*, *-CONH-*, 또는 *-O-*를 나타내고,Linker는 *-L 1-NHCO-L 2-*, *-L 1-O-R 2-O-L 2-*, *-L 1-CH 2-L 2-*, *-L 1-NR 3-L 2-*, 또는 *-L 1-COO-L 2-*를 나타내고,L 1 및 L 2 는 각각 독립적으로, 선형 또는 분지형의 (C1-C30)알킬을 나타내고,R 1 및 R 3은 각각 독립적으로, 수소 또는 선형 또는 분지형의 (C1-C10)알킬을 나타내고,R 2는 선형 또는 분지형의 (C1-C20)알킬을 나타내고,RB는 하기 화학식 2를 나타내고:[화학식 2]* 은 연결 자리를 나타낸다.

- 제1항에 있어서, 포유동물의 소포성 글루타메이트 수송체(vesicular glutamate transporter)(VGLUT)에 특이적으로 결합하는 것을 특징으로 하는 가돌리늄계 화합물.
- 제1항에 따른 가돌리늄계 화합물을 함유하는 것을 특징으로 하는 MRI 조영제.
- 제6항에 있어서, 퇴행성 뇌질환의 진단에 사용되는 것을 특징으로 하는 MRI 조영제.
- 제7항에 있어서, 알츠하이머병의 진단에 사용되는 것을 특징으로 하는 MRI 조영제.
- (a) 로즈 벵갈의 염을 할로겐-치환된 알카노산(halogen-substituted alkanoic acid)와 함께 반응시켜, 하기 화학식 1-1의 화합물을 수득하는 단계;[화학식 1-1]상기 화학식 1-1에서,L 2 는 제1항에서 정의한 바와 같다.(b) 상기 화학식 1-1의 화합물을 하기 화학식 1-2의 화합물과 반응시켜, 하기 화학식 1-3의 화합물을 수득하는 단계:[화학식 1-2][화학식 1-3]상기 화학식 1-2 및 1-3에서,PT는 보호기를 나타내고,L 1, A, Linker, 및 RB는 제1항에서 정의한 바와 같다.(c) 상기 화학식 1-3의 화합물에서 보호기(PT)를 제거하는 단계, 및(d) 상기 (c) 단계에서 수득한 화합물을 가돌리늄 수화물과 반응시켜 제1항에 기재된 화학식 1의 화합물을 수득하는 단계를 포함하는, 제1항에 따른 가돌리늄계 화합물의 제조 방법.
- 제9항에 있어서, 상기 (a) 단계에서, 상기 로즈 벵갈의 염은 로즈 벵갈의 알칼리 금속 염이고, 상기 할로겐-치환된 알카노산은 선형 또는 분지형의 (C1-C30)알칸에서 유래되고 1개의 할로겐에 의해 치환된 알카노산인, 제조 방법.
