Abstract
本発明は、光により心筋細胞の自発的な拍動を変化させることを目的とする。本発明の心筋細胞は、光活性化アデニル酸シクラーゼ遺伝子を有する。該心筋細胞に光を照射すると、cAMPが細胞内で産生され、心筋細胞の拍動はcAMP濃度に応じて変化する。
Description
本発明は、心筋細胞と、心筋細胞の拍動を変化させる方法とに関する。
光遺伝学(オプトジェネティクス)は、脳神経分野において世界的な発展を遂げる一方、脳神経以外の分野での成果は乏しい。脳神経以外の分野での研究例として、心筋の拍動を光でコントロールした研究が知られる(非特許文献1)。この研究では、光活性化非選択的陽イオンチャネルロドプシン2(ChR2)を導入した心筋細胞を用いる。ChR2のイオンチャネルは光照射によって開かれるため、ChR2に光を照射することにより心筋細胞内へのイオン輸送を直接的に引き起こし、心筋を収縮させることができる。1回の光の照射で1回の収縮が起るため、この技術では、光パルスの周期によって人為的な拍動ペースを作り出す。

非特許文献1に記載の方法は、光パルスにより心筋細胞に人為的に拍動を起こさせており、非特許文献1に記載の方法により心筋細胞の自発的な拍動を変化させることは困難であった。そこで、本発明は、心筋細胞の自発的な拍動を光によりcAMPシグナル伝達を介して変化させることを目的とする。
本発明の心筋細胞は、光活性化アデニル酸シクラーゼ遺伝子を有する。
本発明の一形態に係る、心筋細胞の拍動を変化させる方法は、心筋細胞に光活性化アデニル酸シクラーゼ遺伝子を導入する工程と、光活性化アデニル酸シクラーゼ遺伝子が導入された心筋細胞に光を照射して、cAMPを細胞内で産生させる工程と、を備える。ここで、心筋細胞の拍動は、心筋細胞内のcAMP濃度に応じて変化する。
本発明の別の一形態に係る、心筋細胞の拍動を変化させる方法は、光活性化アデニル酸シクラーゼ遺伝子を有する改変細胞を心筋細胞に接触させる工程と、心筋細胞と接触している改変細胞に光を照射して、cAMPを改変細胞内で産生させる工程と、を備える。ここで、心筋細胞の拍動は、改変細胞内のcAMP濃度に応じて変化する。改変細胞は、光活性化アデニル酸シクラーゼ遺伝子を導入した、ヒト胎児腎臓細胞又はヒト新生児皮膚線維芽細胞を含んでもよい。
上記いずれの方法においても、光は、350nm~500nmの波長を有する光を含んでよい。
本発明の一形態に係る心筋細胞を成熟させる方法は、心筋細胞に光活性化アデニル酸シクラーゼ遺伝子を導入する工程と、光活性化アデニル酸シクラーゼ遺伝子が導入された心筋細胞を、該心筋細胞に光を照射しながら培養する工程と、を備える。
本発明の別の一形態に係る心筋細胞を成熟させる方法は、光活性化アデニル酸シクラーゼ遺伝子を有する改変細胞を心筋細胞に接触させる工程と、心筋細胞を、該心筋細胞と接触している改変細胞に光を照射しながら培養する工程と、を備える。
心筋細胞を成熟させる上記いずれの方法においても、光は、350nm~500nmの波長を有する光を含んでよい。
心筋細胞を成熟させる上記いずれの方法においても、光の照射は、心筋細胞の拍動周期が0.5Hz~5Hzに維持されるように行われてよい。
心筋細胞を成熟させる上記いずれの方法においても、心筋細胞は幹細胞に由来してよい。
本発明によれば、光により心筋細胞の自発的な拍動を変化させることができる。
また、本発明によれば、未熟な心筋細胞を成熟させることができる。従来、未熟な心筋細胞を成熟させる方法として、例えば、心筋細胞に周期的な電気的刺激を与える方法、及び、心筋細胞を封入したコラーゲンゲルに周期的な機械的伸展刺激を与える方法が知られている。しかしながら、これらの方法は大量生産に向かず、また、特殊な装置を必要とした。本発明者らの新たな知見によれば、心筋細胞は、本来の拍動周期よりも高い拍動周期で拍動させることにより成熟させることができるとものと考えられる。したがって、光を用いることにより心筋細胞の拍動速度を高めることができる本発明によれば、これらの問題を解決しつつ、未熟な心筋細胞を成熟させることができる。さらに、本発明によれば、生体と同様の作用機序により、すなわちcAMPの細胞内濃度を変化させることにより、拍動周期を増加させることができる。そのため、細胞に損傷を与える、電気刺激を用いた方法とは対照的に、細胞に優しい、自然な成熟を促すことができる。本発明の方法により成熟させた心筋細胞は、移植用心筋組織の材料として、又は、心毒性試験の対象としての利用が期待できる。
<PAC遺伝子を有する心筋細胞>
本発明の心筋細胞は、光活性化アデニル酸シクラーゼ(PAC)遺伝子を有する。本明細書において、PAC遺伝子を有する心筋細胞を、PAC発現心筋細胞ともいう。PAC発現心筋細胞の拍動は、後述の方法により変化させることができる。

心筋細胞が由来する動物は特に限定されず、心筋細胞は、例えば、ヒト、サル、ブタ、ウシ、ヒツジ、ウサギ、ラット、爬虫類、両生類、魚類、鳥類又はマウス由来であってよい。また、心筋細胞は、iPS細胞、ES細胞等の幹細胞に由来する心筋細胞であってよく、具体的には、例えば、ヒトiPS細胞由来の心筋細胞であってもよい。
PAC遺伝子は、PACとして機能するタンパク質をコードする核酸であれば限定されず、そのような核酸にはcDNAも包含される。PACは、光により活性化されてcAMPを産生する天然又は人工のタンパク質であり、BLUF(sensor of Blue Light Using FAD)ドメイン及びシクラーゼ触媒ドメインを有する。BLUFドメインは、FADが結合するドメインであり、FADを利用した青色光の感知に関与するドメインである。シクラーゼ触媒ドメインは、ATPをcAMPに変換するドメインである。PAC遺伝子は様々な生物において見つかっており、PACにおける保存アミノ酸は公知である(非特許文献2)。
PAC遺伝子の由来は特に限定されず、PAC遺伝子は、真核生物に由来しても、原核生物に由来してもよい。真核生物として、例えば、ミドリムシ(ユーグレナ・グラシリス(Euglena gracilis))、ユートレプティエラ(Eutreptiella)、ユートレプティア(Eutreptia)、コラキウム(Colacium)、カウキネア(Khawkinea)等のユーグレナ目藻類;及び鞭毛性アメーバのネグレリア・グルベリ(Neagleria gruberi)等のネグレリア属微生物が挙げられる。原核生物として、例えば、ベギアトア(Beggiatoa sp)等のガンマプロテオバクテリア;オスキラトリア(Oscillatoria)、シュードアナベナ(Pseudanabaena)等のシアノバクテリア;及びトルネリエラ(Turneriella)等のスピロヘータが挙げられる。
PACは、例えば、配列番号1、配列番号2、若しくは配列番号3で示されるアミノ酸配列を有するPAC、又は、配列番号1、配列番号2、若しくは配列番号3で示されるアミノ酸配列と、30%以上、50%以上、80%以上、85%以上、90%以上、又は95%以上の配列同一性を有するアミノ酸配列を有するPACであってよい。配列番号1、配列番号2、及び配列番号3で示されるアミノ酸配列を有するPACは、それぞれ、ベギアトアのPAC(BsPAC)、ミドリムシのPACのαサブユニット(PACa)、及びミドリムシのPACのβサブユニット(PACb)に対応する。
PAC遺伝子のコドンは、心筋細胞の種類に応じて最適化してもよい。例えば、心筋細胞の由来がヒトである場合、コドンを哺乳類に最適化するようにPAC遺伝子の配列を改変することが好ましい。コドンの最適化方法は当業者にとって周知であり、コドンの最適化のために種々のソフトウェアを利用することができる。
PAC発現心筋細胞において、PAC遺伝子は一過性であってもよく、安定的であってもよい。一過性のPAC遺伝子は、例えば、非エピソーム型のプラスミドとして保持されていてもよい。安定的なPAC遺伝子は、ゲノムに組み込まれていてもよく、エピソーム型のプラスミドとして保持されていてもよい。
PAC発現心筋細胞は、心筋細胞に上記PAC遺伝子を導入することにより得ることができる。心筋細胞にPAC遺伝子を導入する方法は特に限定されず、公知の一過性トランスフェクション又は安定的トランスフェクションの方法を利用することができる。例えば、PAC遺伝子を任意の一過性又は安定的発現ベクターに組み込み、公知の化学的、物理的、又は生物学的方法により心筋細胞内に導入することができる。物理的方法は、例えば、直接注入法、粒子衝撃法、エレクトロポレーション、レーザー照射法、ソノポレーション、又はマグネトフェクションである。化学的方法は、例えば、カチオン性ポリマー、リン酸カルシウム、又はカチオン性脂質を利用する方法である。生物学的方法は、例えば、ウイルスを利用する方法である。好ましい具体例として、PAC遺伝子は、アデノ随伴ウイルス(AAV)ベクターを用いて、ウイルス感染により心筋細胞に導入することができる。別の具体例として、PAC遺伝子は、エピソーム型ベクターを用いて、エレクトロポレーションにより心筋細胞に導入することができる。エピソーム型ベクターとしては、例えば、エプスタイン・バール・ウイルス(EBV)由来の複製起点及びEBV抗原1(EBNA1)遺伝子を含むエピソーム型ベクターが挙げられ、pEBMultiベクター(富士フイルム和光純薬株式会社製)等の市販品を利用することができる。
PAC発現心筋細胞には、PAC遺伝子とともに、発現マーカーとしてはたらくタンパク質をコードする遺伝子など、他の遺伝子を導入してもよい。発現マーカーとしては、例えば、公知の蛍光タンパク質が挙げられる。蛍光タンパク質は、その蛍光がPACを活性化しない限り限定されず、例えば、Rudolph RFPであってよい。
PAC遺伝子と上記他の遺伝子とは、適切なリンカーで結合されていてもよい。リンカーは、好ましくは細胞内で切断可能であり、このようなリンカーの例として、2Aペプチドが挙げられる。
<心筋細胞の拍動を変化させる方法1>
本発明の一形態に係る、心筋細胞の拍動を変化させる方法は、心筋細胞にPAC遺伝子を導入する工程と、PAC遺伝子が導入された心筋細胞、すなわち、PAC発現心筋細胞に光を照射して、cAMPを細胞内で産生させる工程と、を備える。この方法はイン・ビトロの方法であってよい。
本発明の一形態に係る、心筋細胞の拍動を変化させる方法は、心筋細胞にPAC遺伝子を導入する工程と、PAC遺伝子が導入された心筋細胞、すなわち、PAC発現心筋細胞に光を照射して、cAMPを細胞内で産生させる工程と、を備える。この方法はイン・ビトロの方法であってよい。
心筋細胞が由来する動物は特に限定されず、心筋細胞は、例えば、ヒト、サル、ブタ、ウシ、ヒツジ、ウサギ、ラット、爬虫類、両生類、魚類、鳥類又はマウス由来であってよい。また、心筋細胞は、iPS細胞、ES細胞等の幹細胞に由来する心筋細胞であってよく、具体的には、例えば、ヒトiPS細胞由来の心筋細胞であってもよい。心筋細胞は、単一の心筋細胞であっても、複数の心筋細胞の集団であって、心筋細胞が互いに接着している集団であってもよい。心筋細胞が集団で存在する場合、PAC遺伝子は、集団中の少なくとも一つの心筋細胞に導入すればよい。PAC遺伝子の詳細及び心筋細胞にPAC遺伝子を導入する方法は、上述したとおりである。
PAC発現心筋細胞に照射する光は、PACを活性化することができる光であれば限定されない。PACは青色光で活性化されるため、光は、350nm~500nmの領域の波長を含む光である。光のピーク波長は、好ましくは、400nm~500nmであり、より好ましくは430nm~470nmであり、さらに好ましくは430nm~460nmである。光の光源としては、例えば、青色発光ダイオードを用いることができる。
光の強度は、PAC遺伝子の種類、PAC遺伝子の発現量、及び所望の拍動速度に応じて調節することができる。一実施形態において、光は、例えば、0.001μmol/m2/s~100000μmol/m2/sの光量子束密度を有していてもよいが、光量子束密度はこれに限定されない。例えば、心筋細胞の拍動速度をより高めるためには、光量子束密度は、0.1μmol/m2/s以上、1μmol/m2/s以上、又は5μmol/m2/s以上であってもよい。効率良く拍動を変化させる観点から、光量子束密度は、100μmol/m2/s以下、又は50μmol/m2/s以下であってよい。上記光量子束密度は、好ましくは、光の一部であって上記ピーク波長に対応する部分の、光量子束密度である。光の強度は公知の方法で調節することができ、例えば、減光フィルタ(NDフィルタ)を用いて調節することができる。
光の照射時間及び照射回数は、PAC遺伝子の種類、PAC遺伝子の発現量、及び所望の拍動パターンに応じて調節することができる。一実施形態において、光の照射時間は、例えば、0.1秒以上、1秒以上、又は20秒以上であってよい。光の照射は、1度のみ行ってもよく、断続的に複数回行ってもよい。一実施形態において、光の照射を複数回行う場合、光の照射は、例えば、15分以下、10分以下、7分以下、5分以下、又は3分以下の間隔をおいて行うことができる。本実施形態に係る、心筋細胞の拍動を変化させる方法によれば、光照射の間隔が15分以下であっても、拍動速度が高まった状態を維持することができる。一方、別の観点から、本実施形態に係る、心筋細胞の拍動を変化させる方法によれば、1回の光照射から15分以内、10分以内、7分以内、5分以内、又は3分以内で拍動速度が光照射前と同程度に戻るため、心筋細胞の拍動速度を高精度に制御することができるともいえる。
PACを有する又は有しない複数の心筋細胞が互いに接着して集団として存在する場合、PACを有する少なくとも一つの心筋細胞に光を照射すればよい。
PAC発現心筋細胞に光を照射することで、PACにより心筋細胞内でcAMPが産生される。細胞内のcAMP濃度の上昇は、細胞内へのカルシウムイオン(Ca2+)の流入を亢進させる。心筋細胞の収縮、すなわち心筋細胞の拍動は、カルシウムイオン濃度の上昇により起こるため、PAC発現心筋細胞に光を照射することで、PAC発現心筋細胞の拍動速度を高めることができる。
心筋細胞の収縮及び弛緩は、それぞれ細胞内のカルシウムイオン濃度の上昇及び低下により起こるため、心筋細胞の拍動の変化は、カルシウムイオン濃度の変化を観察することにより捉えることができる。細胞内のカルシウムイオン濃度の変化は、カルシウム指示薬を用いることにより観察できる。カルシウム指示薬は、その励起光及び蛍光がPACを活性化しない限り限定されず、公知のカルシウム指示薬を用いることができる。このようなカルシウム指示薬の例は、573nmの励起波長及び588nmの蛍光波長を有するCalbryte(商標)590 AM(AATバイオクエスト社製)である。カルシウム指示薬の他の例は、Calbryte(商標)630 AM、Fluo-3 AM、Fluo-4 AM、及びRhod-2 AM、Rhod-5N AM、Rhod-FF AM、Calcium Orange(商標),AM(サーモフィッシャーサイエンティフィック製)である。
<心筋細胞の拍動を変化させる方法2>
本発明の別の一形態に係る、心筋細胞の拍動を変化させる方法は、PAC遺伝子を有する改変細胞を心筋細胞に接触させる工程と、心筋細胞と接触している改変細胞に光を照射して、cAMPを改変細胞内で産生させる工程と、を備える。この方法はイン・ビトロの方法であってよい。本明細書において、PAC遺伝子を有する改変細胞を、PAC発現細胞という場合もある。
本発明の別の一形態に係る、心筋細胞の拍動を変化させる方法は、PAC遺伝子を有する改変細胞を心筋細胞に接触させる工程と、心筋細胞と接触している改変細胞に光を照射して、cAMPを改変細胞内で産生させる工程と、を備える。この方法はイン・ビトロの方法であってよい。本明細書において、PAC遺伝子を有する改変細胞を、PAC発現細胞という場合もある。
心筋細胞の詳細は、上述のとおりである。心筋細胞はPAC遺伝子を有していてもよいが、本方法の有用性の観点から、心筋細胞は、好ましくはPAC遺伝子を有さない。
PAC発現細胞は、任意の細胞にPAC遺伝子を導入することにより得ることができる。かかる細胞は、特に限定されず、例えば、ヒト胎児腎臓細胞(HEK細胞)、ヒト新生児皮膚線維芽細胞(NHDF)、ヒト脂肪組織由来間葉系幹細胞(hMSC-AT)、及びヒト臍帯静脈内皮細胞(HUVEC)からなる群より選ばれる1種以上の細胞であってよい。改変細胞には、上記心筋細胞とは別の心筋細胞が含まれてもよいが、本方法の有用性の観点から、改変細胞は、好ましくは心筋細胞以外の細胞である。
任意の細胞にPAC遺伝子を導入する方法は、上述した、心筋細胞にPAC遺伝子を導入する方法に準ずる。ただし、PAC遺伝子を導入する細胞が心筋細胞以外の細胞である場合、PAC遺伝子は、ウイルスを利用しない方法で導入することが好ましい。
PAC発現細胞を心筋細胞に接触、より具体的には接着させる方法は特に限定されず、例えば、心筋細胞を含む培地にPAC発現細胞を含む培養液を添加することにより、PAC発現細胞を心筋細胞に接触させることができる。これらの細胞を一定時間一緒に培養することで、PAC発現細胞を心筋細胞に接着させることができる。ここで、接着は、好ましくはギャップ結合を有する細胞―細胞接着である。心筋細胞が集団で存在する場合、少なくとも一つの心筋細胞に改変細胞を接触させればよい。
改変細胞に照射する光の詳細及びその照射の仕方は、上述したPAC発現心筋細胞に照射する光の詳細及びその照射の仕方に準ずる。心筋細胞と接触しているPAC発現細胞が複数存在する場合、複数のPAC発現細胞のうちの少なくとも一つのPAC発現細胞に光を照射すればよい。
PAC発現細胞に光を照射することで、PACにより該細胞内でcAMPが産生される。cAMPは互いに接着した細胞の間で輸送される。したがって、心筋細胞自体がPAC遺伝子を有していなくても、PAC発現細胞で産生されたcAMPが該PAC発現細胞と接着する心筋細胞に輸送されて、心筋細胞内のcAMP濃度の上昇をもたらす。したがって、本実施形態に係る方法によれば、心筋細胞自体にはPAC遺伝子を導入する必要なしに、光により心筋細胞の拍動を変化させることができる。一般に、心筋細胞への遺伝子導入は難しく、また、心筋細胞に遺伝子を導入するのに最も効果的であるAAVを使用する方法は、AAVの作製に時間、手間、及び高額な費用を要する。したがって、心筋細胞にPAC遺伝子を導入する必要のない本実施形態に係る方法は、有用である。
<心筋細胞を成熟させる方法1>
本発明の一形態に係る心筋細胞を成熟させる方法は、心筋細胞にPAC遺伝子を導入する工程と、PAC遺伝子が導入された心筋細胞、すなわちPAC発現心筋細胞を、該心筋細胞に光を照射しながら培養する工程と、を備える。この方法はイン・ビトロの方法であってよい。
本発明の一形態に係る心筋細胞を成熟させる方法は、心筋細胞にPAC遺伝子を導入する工程と、PAC遺伝子が導入された心筋細胞、すなわちPAC発現心筋細胞を、該心筋細胞に光を照射しながら培養する工程と、を備える。この方法はイン・ビトロの方法であってよい。
心筋細胞にPAC遺伝子を導入する工程の詳細は、PAC発現心筋細胞を用いて心筋細胞の拍動を変化させる方法において述べたとおりである。
培養方法は特に限定されず、心筋細胞の培養に通常用いられる培地及び培養条件を適用することができる。例えば、培地としては、PSC Cardiomyocyte Maintenance Medium(カタログ番号:A2920801、サーモフィッシャーサイエンティフィッ社)、iCell(登録商標) 心筋細胞 維持用培地(製品コード:M1003、製造元:フジフイルム セルラー ダイナミクス社)等の市販品を用いることができる。培養は、例えば、37℃、5%CO2の条件下で行うことができる。十分に成熟させた心筋細胞を得る観点から、培養期間は、好ましくは6日以上又は1週間以上である。培養期間は、例えば1週間~2週間であってよい。
PAC発現心筋細胞に照射する光の詳細は、上述のとおりである。光照射によりPAC発現心筋細胞の拍動周期を増加させることができ、これにより心筋細胞を成熟させることができる。光の照射の仕方は、心筋細胞の拍動速度が高まりさえすれば、特に制限されない。光の照射は、例えば、心筋細胞の拍動周期が0.5Hz~5Hz、1Hz~5Hz、1.5Hz~4Hz、又は2Hz~4Hzに維持されるように行ってよい。本明細書において、Hzは、1秒あたりの拍動回数を示す。拍動周期は、例えば、0.5Hz、1Hz、1.5Hz、2Hz、3Hz、4Hz、又はそれ以上であってよい。心筋細胞の拍動周期は、通常、成熟が進むにつれて減少するため、拍動周期の下限値は成熟の度合いに依存する。拍動周期は、5Hz、4Hz、3Hz、2Hz、1.5Hz、1Hz又はそれ以下であってよい。心筋細胞の拍動周期は、例えば、光の強度、照射の間隔、照射時間、及び照射回数を調節することにより制御することができる。培養中、光の照射は、ずっと行ってもよく、断続的に行ってもよい。光の照射を断続的に行う場合、より短い期間で心筋細胞を成熟させる観点から、光の照射は、3分以下、5分以下、10分以下、又は15分以下の間隔をおいて行うことが好ましい。光強度を上げれば、照射の間隔が長くても心筋細胞の拍動周期を0.5Hz~5Hzに維持することが可能である。一例として、0.2μmol/m2/sの光を10分の間隔をおいて心筋細胞に照射することで、拍動周期を0.5Hz~5Hzに維持することができる。また、光の照射を断続的に行う場合、一回の照射の時間は、例えば、0.1秒以上、1秒以上、20秒以上、3分以上、5分以上、10分以上、又は15分以上であってよい。
PACを有する又は有しない複数の心筋細胞が互いに接着して集団として存在する場合、PACを有する少なくとも一つの心筋細胞に光を照射すればよい。上述のとおり、光照射によりPAC発現心筋細胞内で産生されたcAMPは、互いに接着した心筋細胞の間で輸送される。したがって、少なくとも一つのPAC発現心筋細胞に光を照射することで、集団中の全ての心筋細胞の拍動を同時に制御でき、すなわち、集団中の全ての心筋細胞を同時に成熟させることができる。また、PAC遺伝子又はPAC発現心筋細胞は、成熟後に取り除くことができるし、万が一これらが残存したとしても、PACの活性化に必要な光は体内に届かないため、本実施形態に係る方法により成熟させた心筋細胞は、移植用途に安全に用いることができる。
心筋細胞の成熟は、例えば、心筋細胞の大きさの増大により、複数の核の存在により、又は筋原線維の規則的な配列により、確認することができる。
<心筋細胞を成熟させる方法2>
本発明の別の一形態に係る、心筋細胞を成熟させる方法は、PAC遺伝子を有する改変細胞、すなわちPAC発現細胞を心筋細胞に接触させる工程と、心筋細胞を、該心筋細胞と接触している改変細胞に光を照射しながら培養する工程と、を備える。この方法はイン・ビトロの方法であってよい。
本発明の別の一形態に係る、心筋細胞を成熟させる方法は、PAC遺伝子を有する改変細胞、すなわちPAC発現細胞を心筋細胞に接触させる工程と、心筋細胞を、該心筋細胞と接触している改変細胞に光を照射しながら培養する工程と、を備える。この方法はイン・ビトロの方法であってよい。
PAC発現細胞を心筋細胞に接触させる工程の詳細は、PAC発現細胞を用いて心筋細胞の拍動を変化させる方法において述べたとおりである。ただし、本実施形態に係る方法においては、本実施形態に係る方法により成熟させた心筋細胞を移植用途に用いる観点から、改変細胞が成熟した又は未熟な心筋細胞であることが好ましい。
培養方法は、上記実施形態に係る心筋細胞を成熟させる方法において述べたとおりである。また、改変細胞に照射する光の詳細及びその照射の仕方は、上記実施形態に係る心筋細胞を成熟させる方法において述べた、PAC発現心筋細胞に照射する光の詳細及びその照射の仕方に準ずる。心筋細胞と接触しているPAC発現細胞が複数存在する場合、複数のPAC発現細胞のうちの少なくとも一つのPAC発現細胞に光を照射すればよい。
本実施形態に係る方法によれば、心筋細胞自体にはPAC遺伝子を導入する必要なしに、心筋細胞を成熟させることができる。PAC発現細胞は、成熟後に取り除くことができるため、本実施形態に係る方法により成熟させた心筋細胞は、移植用途に安全に用いることができる。
1.試験例1
<材料及び方法>
(1)細胞
心筋細胞としては、株式会社マイオリッジから購入したヒトiPS細胞由来の心筋細胞を用いた。特に断りがない限り、PAC発現心筋細胞は次のようにして調製した。まず、フィブロネクチンコートを施したφ35mmディッシュに心筋細胞を播種し、株式会社マイオリッジ製の専用培地にて、37℃、5%CO2の条件下で2日間培養した。次いで、AAVベクターを用いて、心筋細胞にPAC遺伝子を導入した。得られたPAC発現心筋細胞は、遺伝子導入後4日~10日の間に使用した。
<材料及び方法>
(1)細胞
心筋細胞としては、株式会社マイオリッジから購入したヒトiPS細胞由来の心筋細胞を用いた。特に断りがない限り、PAC発現心筋細胞は次のようにして調製した。まず、フィブロネクチンコートを施したφ35mmディッシュに心筋細胞を播種し、株式会社マイオリッジ製の専用培地にて、37℃、5%CO2の条件下で2日間培養した。次いで、AAVベクターを用いて、心筋細胞にPAC遺伝子を導入した。得られたPAC発現心筋細胞は、遺伝子導入後4日~10日の間に使用した。
AAVベクターを用いたPAC遺伝子導入の詳細は次のとおりである。セルバイオラボ社から購入したAAVヘルパーフリー発現システム(2型血清)を用いて、pAAV-mBsPAC-2A-RudolphRFP-WPRE発現ベクター、pAAV-RCベクター及びpHelperベクターを宿主細胞に導入し、パッケージングすることで組換えアデノ随伴ウイルス粒子(recombinant AAV)であるrAAV-mBsPAC-2A-RudolphRFP-WPREを構築した。なお、WPRE(ウッドチャック肝炎ウイルス転写後調節エレメント)は、AAVの力価と感染性を向上させる作用を有する配列である。パッケージング及び精製は、シグナジェンラボラトリーズ社に委託した。得られたAAVの力価は1.46×1013VG/mLであった。AAVを1×108VG/mL~1×1011VG/mLの割合で心筋細胞に添加して心筋細胞に感染させ、組換えアデノ随伴ウイルス粒子rAAV-mBsPAC-2A-RudolphRFPを有する心筋細胞を得た。2日後にRFP蛍光により発現を確認した。プラスミド中のmBsPACは、哺乳類に最適化されたコドンを有するベギアトアのPAC遺伝子であり、その塩基配列は配列番号4で示される。
心筋細胞以外の細胞として、HEK細胞及びNHDFを使用した。
HEK細胞としては、国立研究開発法人医薬基盤・健康・栄養研究所JCRB細胞バンクから購入した、HEK293(細胞番号JCRB9068)を使用した。HEK細胞は、ダルベッコ改変イーグル培地(D-MEM培地)+10%ウシ胎児血清(FBS)を用いて、37℃、5%CO2条件下で、培養及び維持した。pEBMulti-Hygベクター(富士フイルム和光純薬株式会社製)を用いて、mBsPAC遺伝子をエレクトロポレーション(DNA10μg/2×107細胞、260V、950μF)によりHEK細胞に導入することにより、PAC発現HEK細胞を調製した。エレクトロポレーションには、ミラスバイオ社製のIngenio(登録商標)Electroporation kit with 0.4cm cuvettesを使用した。
NHDFは、ロンザ株式会社から購入した。NHDFは、同社製の専用培地を用いて、37℃、5%CO2条件下で、培養及び維持した。上記と同様の方法により、pEBMultiベクターを用いてmBsPAC遺伝子をNHDFに導入することにより、PAC発現NHDFを調製した。
(2)カルシウムイオン濃度の観察
心筋細胞内のカルシウムイオン濃度の変化は、カルシウムイオン指示薬Calbryte 590 AM(AATバイオクエスト社製)を用いて捉えた。メーカーのプロトコルに従って、ジメチルスルホキシド(DMSO)に溶解したCalbryte 590 AMを終濃度20μMでHanks and Hepes bufferに混合し、混合液に終濃度0.04%でプルロニック(登録商標)F-127を添加することにより、working solutionを調製した。心筋細胞に1mLの維持培地と1mLのworking solutionとを添加し、心筋細胞をインキュベータ(37℃、5%CO2)内で45分間静置することで、心筋細胞にカルシウム指示薬を導入した。培地及びworking solutionは、蛍光観察を行う前にHanks and Hepes bufferに置換した。
心筋細胞内のカルシウムイオン濃度の変化は、カルシウムイオン指示薬Calbryte 590 AM(AATバイオクエスト社製)を用いて捉えた。メーカーのプロトコルに従って、ジメチルスルホキシド(DMSO)に溶解したCalbryte 590 AMを終濃度20μMでHanks and Hepes bufferに混合し、混合液に終濃度0.04%でプルロニック(登録商標)F-127を添加することにより、working solutionを調製した。心筋細胞に1mLの維持培地と1mLのworking solutionとを添加し、心筋細胞をインキュベータ(37℃、5%CO2)内で45分間静置することで、心筋細胞にカルシウム指示薬を導入した。培地及びworking solutionは、蛍光観察を行う前にHanks and Hepes bufferに置換した。
(3)光照射
照射光の光源として、ロイヤルブルーLED(LXML-PR01-0425、フィリップス・ルミレッズ・ライティング・カンパニー製:ピーク波長447.5nm;半値全幅20nm)を用いた。ロイヤルブルーLEDは顕微鏡の近傍に設置し、NDフィルタを複数枚取り付け可能なフォルダを、光源と顕微鏡の間に固定した。光強度は、1枚又は複数枚のNDフィルタ(ND1、ND3、ND10、又はND30)を用いて、0.01W/m2、0.05W/m2、0.38W/m2、2.33W/m2、又は9.28W/m2(すなわち0.048μmol/m2/s、0.19μmol/m2/s、1.41μmol/m2/s、8.76μmol/m2/s、又は34.9μmol/m2/s)に調節した。1回の光照射の長さは20秒に設定した。
照射光の光源として、ロイヤルブルーLED(LXML-PR01-0425、フィリップス・ルミレッズ・ライティング・カンパニー製:ピーク波長447.5nm;半値全幅20nm)を用いた。ロイヤルブルーLEDは顕微鏡の近傍に設置し、NDフィルタを複数枚取り付け可能なフォルダを、光源と顕微鏡の間に固定した。光強度は、1枚又は複数枚のNDフィルタ(ND1、ND3、ND10、又はND30)を用いて、0.01W/m2、0.05W/m2、0.38W/m2、2.33W/m2、又は9.28W/m2(すなわち0.048μmol/m2/s、0.19μmol/m2/s、1.41μmol/m2/s、8.76μmol/m2/s、又は34.9μmol/m2/s)に調節した。1回の光照射の長さは20秒に設定した。
(4)細胞及び蛍光の観測
細胞及び細胞から発せられる蛍光は、倒立蛍光顕微鏡(DMI6000B、ライカ社製)及び内蔵された蛍光観察フィルタセットM2(BP546/14、DM/580、及びLP590)を用いて、EM-CCDカメラ(ImagEM C9100-23B、浜松ホトニクス株式会社製)により観測した。観測の条件は、以下の通りである。

<実施例1>
上述の方法により調製したPAC発現心筋細胞に強度8.76μmol/m2/sの光を照射し、拍動の変化を観察した。PAC発現心筋細胞を1つ有するコロニーの、(A)光照射前、(B)光照射中、(C)光照射直後、(D)光照射から1分後、(E)光照射から3分後、及び、(F)光照射から5分後のカルシウム指示薬の蛍光強度を、図1に示す。図1に示されるように、拍動速度は、光照射を開始した直後に加速し、その後の20秒間で最も高くなり、その後徐々に元に戻った。
上述の方法により調製したPAC発現心筋細胞に強度8.76μmol/m2/sの光を照射し、拍動の変化を観察した。PAC発現心筋細胞を1つ有するコロニーの、(A)光照射前、(B)光照射中、(C)光照射直後、(D)光照射から1分後、(E)光照射から3分後、及び、(F)光照射から5分後のカルシウム指示薬の蛍光強度を、図1に示す。図1に示されるように、拍動速度は、光照射を開始した直後に加速し、その後の20秒間で最も高くなり、その後徐々に元に戻った。
上記実験において、図2の(A)に示すとおり、同一コロニー中に存在する五つの心筋細胞のそれぞれに関心領域(ROI)を設定し、各ROIにおける蛍光強度の変化を観察した。結果を図2の(B)に示す。蛍光強度、すなわちカルシウムイオン濃度は細胞毎に異なるものの、互いに接着しコロニーを形成した心筋細胞の収縮開始のタイミングは、同期していることが分かった。
さらに、PAC発現心筋細胞に、強度の異なる光を、十分な間隔をあけて照射し、拍動の変化を観察した。観察対象は一つの細胞に固定した。光の照射は、前の照射により増加した拍動速度が元に戻ってから行った。それぞれの強度の光を照射した直後(すなわち、光照射から0秒後~20秒後)の心筋細胞の拍動の波形を図3の(A)に示す。また、光強度と拍動速度の関係を図3の(B)に示す。これらの図に示すとおり、光強度依存的な拍動速度の変化を確認することができた。特に、光強度0.1μmol/m2/sから10μmol/m2/sの間では、拍動速度と光強度の高い相関が認められた(図3の(B))。また、図3の(A)において、波形の振幅の大きさで表されるカルシウムイオン濃度の変化量は、照射光の強度に依存しないことが分かった。
<実施例2>
心筋細胞に108VG/mLのAAVを添加することにより、心筋細胞にPAC遺伝子を導入した。得られたPAC発現心筋細胞に強度1.41μmol/m2/sの光を、20秒間隔で8回断続的に照射し、拍動の変化を観察した。カルシウム指示薬の蛍光強度の変化を図4に示す。図4は、光照射開始1分前、光照射直前(光照射開始20秒前)、n回目の光照射中(図中、nBで示す)、n回目のインターバル(図中、nDで示す)、並びに8回目の光照射終了から3分、6分、9分、及び12分後の拍動速度を示す(nは1~8である)。これら各段階において、拍動速度は、拍動数を20秒間計測することにより求めた。図4に示すとおり、心筋細胞に繰り返し光を照射することにより、高い拍動速度を持続的に維持することに成功した。
心筋細胞に108VG/mLのAAVを添加することにより、心筋細胞にPAC遺伝子を導入した。得られたPAC発現心筋細胞に強度1.41μmol/m2/sの光を、20秒間隔で8回断続的に照射し、拍動の変化を観察した。カルシウム指示薬の蛍光強度の変化を図4に示す。図4は、光照射開始1分前、光照射直前(光照射開始20秒前)、n回目の光照射中(図中、nBで示す)、n回目のインターバル(図中、nDで示す)、並びに8回目の光照射終了から3分、6分、9分、及び12分後の拍動速度を示す(nは1~8である)。これら各段階において、拍動速度は、拍動数を20秒間計測することにより求めた。図4に示すとおり、心筋細胞に繰り返し光を照射することにより、高い拍動速度を持続的に維持することに成功した。
<実施例3>
PAC非発現心筋細胞を含むディッシュにPAC発現HEK細胞を添加した。インキュベータ内に2時間静置後、HEK細胞の生着を確認した。HEK細胞と心筋細胞は、細胞の大きさ及び形状が異なるため、明視野観察で判別が可能であった。明視野像中、HEK細胞はひし形であり、心筋細胞に比べて小さく見えた。両細胞が互いに接着していることを確認し、これらの細胞に強度266μmol/m2/sの光を照射した。PAC非発現心筋細胞の光照射前後のカルシウム指示薬の蛍光強度を図5に示す。拍動速度は光照射に伴い高まっていた。この結果から、心筋細胞がPACを発現していなくても、PACを発現する他種細胞の添加により、心筋細胞の拍動の光制御が可能であることが明らかとなった。なお、図5中、光照射中に蛍光強度のバックグラウンドが高くなっているのは、カルシウム指示薬の蛍光とともに光源からの光を直接検出したためである。
PAC非発現心筋細胞を含むディッシュにPAC発現HEK細胞を添加した。インキュベータ内に2時間静置後、HEK細胞の生着を確認した。HEK細胞と心筋細胞は、細胞の大きさ及び形状が異なるため、明視野観察で判別が可能であった。明視野像中、HEK細胞はひし形であり、心筋細胞に比べて小さく見えた。両細胞が互いに接着していることを確認し、これらの細胞に強度266μmol/m2/sの光を照射した。PAC非発現心筋細胞の光照射前後のカルシウム指示薬の蛍光強度を図5に示す。拍動速度は光照射に伴い高まっていた。この結果から、心筋細胞がPACを発現していなくても、PACを発現する他種細胞の添加により、心筋細胞の拍動の光制御が可能であることが明らかとなった。なお、図5中、光照射中に蛍光強度のバックグラウンドが高くなっているのは、カルシウム指示薬の蛍光とともに光源からの光を直接検出したためである。
<実施例4>
PAC非発現心筋細胞を含むディッシュに、1×105個又は5×105個のPAC発現HEK細胞を添加し、実施例3と同様にしてPAC非発現心筋細胞の拍動の変化を調べた。1×105個のPAC発現HEK細胞を添加した場合、光照射前は33ビート/分であった拍動速度が光照射直後には36ビート/分に上昇し、光照射から5分以内に再び33ビート/分に戻った。5×105個のPAC発現HEK細胞を添加した場合、光照射前は63ビート/分であった拍動速度が光照射直後には69ビート/分に上昇し、光照射から7分以内に再び63ビート/分に戻った。これらの結果から、添加するPAC発現細胞の量を変えることによって、心筋細胞の拍動を分単位で制御することができることが示唆された。

<実施例5>
PAC非発現心筋細胞を含むディッシュに、1×105個のPAC発現NHDFを添加し、実施例3と同様にしてPAC非発現心筋細胞の拍動の変化を調べた。光照射前は42ビート/分であった拍動速度が光照射直後には45ビート/分に上昇し、光照射から3分以内に再び42ビート/分に戻った。
PAC非発現心筋細胞を含むディッシュに、1×105個のPAC発現NHDFを添加し、実施例3と同様にしてPAC非発現心筋細胞の拍動の変化を調べた。光照射前は42ビート/分であった拍動速度が光照射直後には45ビート/分に上昇し、光照射から3分以内に再び42ビート/分に戻った。
実施例1~5の結果より、心筋細胞にPAC遺伝子を導入することにより、心筋細胞の拍動を光によって高精度で制御できることが分かった。また、同期して拍動するコロニー中の心筋細胞のうち少なくとも一つの心筋細胞がPACを発現していれば、コロニー全体の拍動を制御できることも分かった。さらに、細胞同士が接着していれば、PACを発現した他種細胞の極少量の添加によっても、心筋細胞の拍動を制御できることも示された。
2.試験例2
<実施例6>
心筋細胞(製品名:iCell心筋細胞2.0-01434、製品コード:CMC-100-012-000.5、フジフイルム セルラー ダイナミクス社)を製造元のプロトコルに従って溶解し、5μg/mLのフィブロネクチンでコートしたディッシュに1.6×105細胞/cm2の密度で播種した。培地として、iCell心筋細胞解凍用培地(製品コード:M1001、製造元:フジフイルム セルラー ダイナミクス社)及びiCell心筋細胞維持用培地(製品コード:M1003、製造元:フジフイルム セルラー ダイナミクス社)を用いた。細胞の播種から2日後にAAVベクター(力価:1×1010VP/mL)を用いてPAC遺伝子及びlifeact-Rudolph RFP遺伝子を、又はlifeact-Rudolph RFP遺伝子のみを心筋細胞に導入した。lifeact-Rudolph RFPは、アクチンフィラメントに結合するペプチドlifeactとRudolph RFPとの融合タンパク質であり、筋原線維の蛍光可視化に用いた。AAVの添加から2日後に、AAVを含まない心筋細胞維持培地で培地を交換し、青色光をディッジュの下から細胞に照射しながら細胞を培養した。光照射の詳細は次のとおりである。
光源:ピーク波長450nmのLED(品番:LXML-PR01-0425、ルクシオン・レベル、フィリップス・ルミレッズ社)
光の強度:1.3μmol/m2/s
照射周期:15分(15分の点灯と15分の消灯の繰り返し)
照射期間:6日間
心筋細胞の拍動を、試験例1と同様に、カルシウム指示薬を用いてカルシウムイオン濃度の変化を捉えることにより観察した。光照射により、PAC発現心筋細胞の拍動周期は、照射開始時は2Hzに、照射期間の最終日は0.5Hzに維持されていた。
<実施例6>
心筋細胞(製品名:iCell心筋細胞2.0-01434、製品コード:CMC-100-012-000.5、フジフイルム セルラー ダイナミクス社)を製造元のプロトコルに従って溶解し、5μg/mLのフィブロネクチンでコートしたディッシュに1.6×105細胞/cm2の密度で播種した。培地として、iCell心筋細胞解凍用培地(製品コード:M1001、製造元:フジフイルム セルラー ダイナミクス社)及びiCell心筋細胞維持用培地(製品コード:M1003、製造元:フジフイルム セルラー ダイナミクス社)を用いた。細胞の播種から2日後にAAVベクター(力価:1×1010VP/mL)を用いてPAC遺伝子及びlifeact-Rudolph RFP遺伝子を、又はlifeact-Rudolph RFP遺伝子のみを心筋細胞に導入した。lifeact-Rudolph RFPは、アクチンフィラメントに結合するペプチドlifeactとRudolph RFPとの融合タンパク質であり、筋原線維の蛍光可視化に用いた。AAVの添加から2日後に、AAVを含まない心筋細胞維持培地で培地を交換し、青色光をディッジュの下から細胞に照射しながら細胞を培養した。光照射の詳細は次のとおりである。

6日間の培養後、心筋細胞を蛍光顕微鏡で観察した。PACを発現しない心筋細胞の蛍光画像を図6の(A)に示し、PAC発現心筋細胞の蛍光画像を図6の(B)に示す。図6中、スケールバーは50μmを示す。図6に示されるように、光を照射しながら培養したPAC発現心筋細胞(B)は、光を照射しながら培養したPACを発現しない心筋細胞(A)よりも10倍以上大きい細胞面積を有していた。また、PAC発現心筋細胞(B)では、筋原線維が心筋細胞の長軸方向(画像の左下から右上)に規則的に整列しており、サルコメア構造がはっきりと現れていた。このことから、光を照射しながら培養したPAC発現心筋細胞は成熟していることが確認できた。
Claims (10)
- 光活性化アデニル酸シクラーゼ遺伝子を有する、心筋細胞。
- 心筋細胞の拍動を変化させる方法であって、
心筋細胞に光活性化アデニル酸シクラーゼ遺伝子を導入する工程と、
光活性化アデニル酸シクラーゼ遺伝子が導入された心筋細胞に光を照射して、cAMPを細胞内で産生させる工程と、備え、
心筋細胞の拍動は、心筋細胞内のcAMP濃度に応じて変化する、方法。 - 心筋細胞の拍動を変化させる方法であって、
光活性化アデニル酸シクラーゼ遺伝子を有する改変細胞を心筋細胞に接触させる工程と、
心筋細胞と接触している改変細胞に光を照射して、cAMPを改変細胞内で産生させる工程と、備え、
心筋細胞の拍動は、改変細胞内のcAMP濃度に応じて変化する、方法。 - 改変細胞が、光活性化アデニル酸シクラーゼ遺伝子を導入した、ヒト胎児腎臓細胞又はヒト新生児皮膚線維芽細胞を含む、請求項3に記載の方法。
- 光が、350nm~500nmの波長を有する光を含む、請求項2~4のいずれか一項に記載の方法。
- 心筋細胞を成熟させる方法であって、
心筋細胞に光活性化アデニル酸シクラーゼ遺伝子を導入する工程と、
光活性化アデニル酸シクラーゼ遺伝子が導入された心筋細胞を、該心筋細胞に光を照射しながら培養する工程と、を備える、方法。 - 心筋細胞を成熟させる方法であって、
光活性化アデニル酸シクラーゼ遺伝子を有する改変細胞を心筋細胞に接触させる工程と、
心筋細胞を、該心筋細胞と接触している改変細胞に光を照射しながら培養する工程と、を備える、方法。 - 光が、350nm~500nmの波長を有する光を含む、請求項6又は7に記載の方法。
- 光の照射が、心筋細胞の拍動周期が0.5Hz~5Hzに維持されるように行われる、請求項6~8のいずれか一項に記載の方法。
- 心筋細胞が幹細胞に由来する、請求項6~9のいずれか一項に記載の方法。
