Abstract
本発明の課題は、十分な供給量を確保することができると共に製造コストを低く抑えることができるデオキシリボ核酸保存用組成物を提供することである。 本発明に係るデオキシリボ核酸保存用組成物は、多糖体を含有する乳酸菌産生物を有効成分とする。なお、乳酸菌産生物は、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクスOLL1247菌と、ストレプトコッカス・サーモフィラスOLS3078菌との組合せによって生成されることが好ましい。
Description
本発明は、デオキシリボ核酸(以下「DNA」と称する。)を保存するための組成物およびその製造方法に関する。また、本発明は、DNAを保存する方法にも関する。さらに、本発明は、乳酸菌産生物およびその使用方法にも関する。
過去に「松樹皮抽出物を含有するDNA保護剤および健康食品」が提案されている(例えば、特開2003-238427等参照)。
しかし、松は成木になるまでに期間が比較的長く、その供給量に限りがある。また、その抽出物を得るためには伐採等の比較的危険な作業が必要となる共に、抽出後の残りカスの処分も必要となる。このため、「松樹皮抽出物を含有するDNA保護剤および健康食品」の製造コストは必然的に高くなってしまう。したがって、「松樹皮抽出物を含有するDNA保護剤および健康食品」は、高価な商品となることが想定される。
本発明の課題は、十分な供給量を確保することができると共に製造コストを低く抑えることができるDNA保存用組成物を提供することである。
本発明の第1局面に係るDNA保存用組成物は、多糖体を含有する乳酸菌産生物(以下「多糖体含有乳酸菌産生物」と称する。)を有効成分とする。すなわち、多糖体含有乳酸菌産生物をDNA保存用組成物としてまたはその一成分として使用する。なお、ここにいう「組成物」には、医薬品,サプリメントおよび食品添加剤等の製剤、飲食品(動植物そのものを除く。)ならびに飲食品組成物(加工された飲食品を含む。)等の動物(ヒトを含む)が摂取し得る物が含まれる。また、ここにいう「保存」とは、DNAの損傷を抑制したり、DNAの損傷修復を促進したりすることを意味する。
本願発明者らの鋭意検討の結果、本発明の第1局面に係るDNA保存用組成物がDNAの損傷を抑制したり、DNAの損傷修復を促進したりすることができることが明らかとされた。すなわち、このDNA保存用組成物は、良好にDNAを保存することができる。また、このDNA保存用組成物の有効成分は、上述の通り乳酸菌によって産生される。このため、このDNA保存用組成物の有効成分は、比較的安全な作業で廃棄物を出すことなく短時間で産生することができる。このため、このDNA保存用組成物は、十分な供給量を確保することができると共に製造コストを低く抑えることができる。
ところで、上述のDNA保存用組成物において、多糖体含有乳酸菌産生物は、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus)とストレプトコッカス・サーモフィラス(Streptococcus thermophilus)との組合せによって生成されることが好ましい。なお、ここで、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクスは具体的にはラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクスOLL1247菌(受託番号:NITE BP-01814)であることが好ましく、ストレプトコッカス・サーモフィラスは具体的にはストレプトコッカス・サーモフィラスOLS3078菌(受託番号:NITE BP-01697)であることが好ましい。
本発明の第2局面に係る方法は、多糖体含有乳酸菌産生物をDNA保存用組成物として使用する方法である。すなわち、DNA保存用組成物としての使用のための多糖体含有乳酸菌産生物を提供することである。
本発明の第3局面に係るDNAを保存する方法は、上述のDNA保存用組成物を経口摂取する方法である。ただし、ヒトを治療する行為を除かれる。なお、摂取期間は、1週間以上であることが好ましく、2週間以上であることがより好ましく、3週間以上であることがさらに好ましく、4週間以上であることが特に好ましい。
本発明の第4局面に係るDNA保存用組成物の製造方法では、多糖体を産生する乳酸菌に乳原料が供給されることによってDNA保存用組成物が製造される。すなわち、ここでは、多糖体含有乳酸菌産生物が、DNAを保存するための組成物を製造するために使用される。
本発明の実施の形態に係るDNA保存用組成物は、DNAの損傷を抑制したり、DNAの損傷修復を促進したりするための経口摂取組成物であって、多糖体を含有する乳酸菌産生物(以下「多糖体含有乳酸菌産生物」と称する。)を有効成分とする。以下、このDNA保存用組成物の有効成分について詳述した後、DNA保存用組成物の形態について説明を行い、各形態における任意成分について詳述する。また、その後、さらに、DNA保存用組成物の効率的な摂取方法やその効果についても詳述する。
(1)多糖体含有乳酸菌産生物
本発明の実施の形態において、多糖体含有乳酸菌産生物には、多糖体を含有する乳酸菌発酵物(以下「多糖体含有乳酸菌発酵物」という。)、多糖体を含有する乳酸菌培養物(以下「多糖体含有乳酸菌培養物」という。)、多糖体を含有する乳酸菌代謝物等が含まれる。
本発明の実施の形態において、多糖体含有乳酸菌産生物には、多糖体を含有する乳酸菌発酵物(以下「多糖体含有乳酸菌発酵物」という。)、多糖体を含有する乳酸菌培養物(以下「多糖体含有乳酸菌培養物」という。)、多糖体を含有する乳酸菌代謝物等が含まれる。
なお、ここにいう「多糖体」とは、ガラクトース、グルコース、ラムノース、マンノース、N-アセチルグルコサミンなどの糖類から構成される糖鎖高分子である。この多糖体には、中性多糖体の他、リン酸基が結合した酸性多糖体が含まれてもよい。また、その分子量は、通常、5000から50万の範囲内である。
また、ここにいう「乳酸菌」とは、ブドウ糖を資化して対糖収率で50%以上の乳酸を生産する微生物の総称で、生理学的性質としてグラム陽性菌の球菌または桿菌で、運動性なし、胞子形成能なし、カタラーゼ陰性などの特徴を有しているものである。乳酸菌は古来、発酵乳等を介して世界各地で食されており、極めて安全性の高い微生物と言える。乳酸菌は現在までに、ラクトコッカス(Lactococcus)属、ラクトバチルス(Lactobacillus)属、リューコノストック(Leuconostoc)属、ペディオコッカス(Pediococcus)属、ストレプトコッカス(Streptococcus)属、ワイセラ(Wissella)属、テトラジェノコッカス(Tetragenococcus)属、オエノコッカス(Oenococcus)属、エンテロコッカス(Enterococcus)属、バゴコッカス(Vagococcus)属、カルノバクテリウム(Carnobacterium)属の11属に分類されている。本発明の実施の形態ではこれら全ての乳酸菌を用いることができるが、これらの中でも、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus)とストレプトコッカス・サーモフィラス(Streptococcus thermophilus)とを混合して用いることが特に好ましい。なお、ここにいうラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクスはラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクスOLL1247菌(受託番号:NITE BP-01814)であることが好ましく、ストレプトコッカス・サーモフィラスはストレプトコッカス・サーモフィラス(Streptococcus thermophilus)OLS3078菌(受託番号:NITE BP-01697)であることが好ましい。ここで、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus)OLL1247菌は、2014年3月6日付(受託日)で、独立行政法人製品評価技術基盤機構特許微生物寄託センター(日本国千葉県木更津市かずさ鎌足2-5-8 122号室)に、受託番号NITE BP-01814として、ブタペスト条約に基づき国際寄託されている。また、ストレプトコッカス・サーモフィラス(Streptococcus thermophilus)OLS3078菌は、2013年8月23日付(受託日)で、独立行政法人製品評価技術基盤機構特許微生物寄託センター(日本国千葉県木更津市かずさ鎌足2-5-8 122号室)に、受託番号NITE BP-01697として、ブタペスト条約に基づき国際寄託されている。
ここにいう「乳酸菌発酵物」とは、乳酸菌による発酵によって得られた培養物自体を意味する。そして、この乳酸菌発酵物には、乳酸菌の発酵物およびその処理物、例えば、培養物(乳酸菌発酵物)をろ過・遠心分離もしくは膜分離等で除菌して得られた培養濾液や培養上清液、培養濾液・培養上清液や乳酸菌発酵物等をエバポレーター等により濃縮した濃縮物、ペースト化物、希釈物又は(凍結、加熱、減圧など)乾燥物が含まれる。なお、処理物の調製の際は、ろ過、遠心分離、膜分離等の除菌処理、沈殿、濃縮、ペースト化、希釈、乾燥などの前述の処理工程の1つ又は複数を組み合わせて実施することができる。また、培養物用の培地としては、例えば、酵母エキスを添加した脱脂粉乳培地、MRS培地等が挙げられる。
乳酸菌産生物は乳酸菌の乳発酵物や、乳培養物、乳代謝物であるのが特に好ましい。乳発酵物や、乳培養物、乳代謝物としては、例えば、発酵乳(ヨーグルト)や多糖体が挙げられる。発酵乳(ヨーグルト)は、乳に乳酸菌や酵母を混ぜて発酵することによって調製される発酵食品であり、その美味しさと、美容や健康面から幅広く食されている。発酵乳(ヨーグルト)は、好ましくはその上清とすることができる。この発酵乳には、脱脂粉乳や、ホエイ分解物等の培養液の他、ペクチン、グアーガム、キサンタンガム、カラギーナン、加工でんぶん等の増粘剤やゲル化剤が添加されていてもよい。
乳としては、例えば、牛乳等の獣乳や、その加工品(例えば、脱脂乳、全脂粉乳、脱脂粉乳、れん乳、カゼイン、乳清、生クリーム、コンパウンドクリーム、バター、バターミルクパウダー、チーズ等)、大豆由来の豆乳等の植物性乳等が挙げられる。なお、乳は、殺菌処理されていてもよいし、殺菌処理されていなくてもよい。
ところで、発酵乳の原料として、例えば、発酵乳原料ミックスと呼ばれるものが挙げられる。発酵乳原料ミックスとは、原料乳および他の成分を含む混合物である。この発酵乳原料ミックスは、例えば、原料乳、水、他の任意成分(例えば、砂糖、糖類、甘味料、酸味料、ミネラル、ビタミン、香料等)等の発酵乳の製造に常用される原料を加温して溶解し、混合することによって得られる。原料乳には、水、生乳、殺菌乳、脱脂乳、全脂粉乳、脱脂粉乳、全脂濃縮乳、脱脂濃縮乳、バターミルク、バター、クリーム、チーズ等が含まれてもよい。また、原料乳には、ホエイタンパク質濃縮物(WPC)、ホエイタンパク質単離物(WPI)、α-ラクトアルブミン(α-La)、β-ラクトグロブリン(β-Lg)等が含まれてもよい。
発酵乳は、従来の方法と同様に、原料ミックスの調合工程、原料ミックスの(加熱)殺菌工程、原料ミックスの冷却工程、スターターの添加工程、発酵工程、発酵乳の冷却工程等の工程を経て製造される。原料ミックスの調合工程では、原材料が混合(調合)される。なお、上述の工程では、発酵乳を製造する際に用いられる通常の条件を適宜採用すればよい。また、原料ミックスの(加熱)殺菌工程、原料ミックスの冷却工程、スターターの添加工程、発酵工程および発酵乳の冷却工程は、この順番で実施されることが好ましい。
乳酸菌を培養するための培地としては、通常用いられる培地を使用することができる。すなわち、主炭素源のほか窒素源、無機物その他の栄養素を程良く含有する培地であれば、いずれの培地も使用することができる。炭素源としては、使用菌の資化性に応じて、ラクトース、グルコース、スクロース、フラクトース、澱粉加水分解物、廃糖蜜等を使用することができる。窒素源としては、カゼインの加水分解物、ホエイタンパク質加水分解物、α-ラクトアルブミン、β-ラクトグロブリン、グリコマクロペプチド、大豆タンパク質加水分解物等の有機窒素含有物を使用することができる。ほかに増殖促進剤としては、肉エキス、魚肉エキス、酵母エキス等を使用することができる。
乳酸菌は、嫌気状態で培養されることが好ましいが、通常、液体静置培養等で用いられる微好気状態で培養されることが好ましい。なお、嫌気状態下での培養方法には、炭素ガス気相下で培養する方法などの公知の手法を採用することができるが、他の方法が採用されてもかまわない。培養温度は一般に30℃以上47℃以下の範囲内であることが好ましく、35℃以上46℃以下の範囲内であることがより好ましく、37℃以上45℃以下の範囲内であることがさらに好ましい。乳酸菌培養中の培地のpHは、6以上7以下の範囲内に維持されることが好ましいが、菌が生育するpHであれば他のpH範囲でもよい。乳酸菌等の培養時間としては、通常、1時間以上48時間以下の範囲内が好ましく、1.5時間以上36時間以下の範囲内であることがより好ましく、2時間以上24時間以下の範囲内であることがさらに好ましい。
発酵乳(ヨーグルト)は、典型的には、無脂乳固形分が8重量%以上であり、乳酸菌数又は酵母数が106個/ml以上1011個/ml以下の範囲内である。
(2)DNA保存用組成物の形態および任意成分
本発明の実施の形態に係るDNA保存用組成物は、医薬品,サプリメントおよび食品添加剤等の製剤、飲食品(動植物そのものを除く。)ならびに飲食品組成物(加工された飲食品を含む。)等の形態を採り得る。

本発明の実施の形態において、製剤とは、製剤化のために許容されうる添加剤を併用して、常法に従い、経口製剤として調製したものである。この製剤は、錠剤、散剤、細粒剤、顆粒剤、カプセル剤、丸剤、徐放剤などの固形製剤、溶液、懸濁液、乳濁液などの液状製剤の形態を採り得る。製剤化のために許容され得る添加剤としては、例えば、賦形剤、安定剤、防腐剤、湿潤剤、乳化剤、滑沢剤、甘味料、着色料、香料、緩衝剤、酸化防止剤、pH調整剤などが挙げられる。なお、食品添加剤としては、具体的には加工調味料、風味調味料、調理ミックス等の調味料等が挙げられる。
また、本発明の実施の形態において、飲食品および飲食品組成物とは、ヒトや動物の飲食のために加工されたものであって、溶液、懸濁液、乳濁液、粉末、固体成形物等の経口摂取可能な形態であればよく特に限定されない。飲食品および飲食品組成物の例としては、具体的には、乳飲料(加工乳を含む)、ヨーグルト類、乳酸菌飲料、発酵乳、アイスクリーム類、クリーム類、チーズ類などの乳製品;清涼飲料、果汁飲料、野菜飲料、豆乳飲料、コーヒー飲料、茶飲料、ゼリー飲料、ココア、スムージーなどの粉末飲料やスポーツ粉末飲料、栄養強化の粉末飲料、美容用の粉末食品、粉末スープ、蒸しパンのもと、濃縮飲料、アルコール飲料などの飲料類;パン、パスタ、麺、ケーキミックス、唐揚げ粉、パン粉などの小麦粉製品;チョコレート、ガム、飴、クッキー、グミ、スナック、和菓子、ゼリー、プリンなどのデザート菓子などの菓子類;カレー、バスタソース、ポトフ、シチュー、和風食品のレトルト食品;加工油脂、バター、マーガリン、スプレッド、マヨネーズなどの油脂類;フリーズドライ食品などの即席食品類;農産缶詰、ジャム・マーマレード類、漬け物、煮豆、シリアル、雑炊などの農産加工品;水産加工品;畜産加工品;ピッツア、ドリア、グラタン、惣菜、フライなど冷凍食品;流動食、さらには動物の飼料、タブレット、口腔内に使用する化粧品などが挙げられる。
なお、本発明の実施の形態において、飲食品および飲食品組成物には、機能性食品、健康栄養食品、健康食品、特定保健用食品、機能性表示食品、栄養機能食品、病者用食品、乳幼児用調製粉乳、妊産婦もしくは授乳婦用粉乳、または疾病リスク低減表示を付した飲食品のような分類のものも包含される。ここで、疾病リスク低減の表示とは、疾病リスクを低減する可能性のある飲食品の表示であって、FAO/WHO合同食品規格委員会(コーデックス委員会)の定める規格に基づいて、またはその規格を参考にして、定められた表示または認められた表示である。
本発明の実施の形態において、飲食品および飲食品組成物には、必要に応じて、任意の成分を加えることができる。このような任意の成分としては、特段の制限はないが、通常、飲食品に配合される成分である甘味料、酸味料、野菜や果物や種実の汁やそのエキス、ビタミン、ミネラル、アミノ酸などの栄養素、乳酸菌(本発明の実施の形態に係る必須の乳酸菌を除く。)、ビフィズス菌、プロピオン酸菌などの有用な微生物やその発酵物、オリゴ糖などの機能性をもつ糖類、ローヤルゼリー、グルコサミン、アスタキサンチン、ポリフェノールなどの既存の機能性素材、香料、pH調整剤、賦形剤、酸味料、着色料、乳化剤、保存料等が挙げられる。
<DNA保存用組成物の効率的な摂取方法およびその効果>
本発明の実施の形態に係るDNA保存用組成物は、多糖体の摂取量が200μg以上/日となるように、少なくとも7日間、経口摂取することが好ましい。このDNA保存用組成物をこのように摂取することによって、DNAの損傷を有効に抑制することができると共に、DNAの損傷修復を有効に促進させることができる。
本発明の実施の形態に係るDNA保存用組成物は、多糖体の摂取量が200μg以上/日となるように、少なくとも7日間、経口摂取することが好ましい。このDNA保存用組成物をこのように摂取することによって、DNAの損傷を有効に抑制することができると共に、DNAの損傷修復を有効に促進させることができる。
なお、多糖体の摂取量は、人体に害を及ぼさない限り特に制限されないが、費用対効果を考慮すると、200μg/日以上60000μg/日以下の範囲内であることが好ましく、300μg/日以上45000μg/日以下の範囲内であることがより好ましく、400μg/日以上30000μg/日以下の範囲内であることがさらに好ましく、500μg/日以上15000μg/日以下の範囲内であることが特に好ましい(なお、「質量/日以上」の表記は「質量以上/日」の表記と同義であり、「質量/日以下」の表記は「質量以下/日」の表記と同義である。)。
なお、上述の各有効成分の必要用量は、人体投与実験に基づくものであるが、動物実験(例えばマウス実験)における必要投与用量から食品安全委員会資料に基づく下式を用いて人体への必要投与用量に換算することもできる。

本実験例では、紫外線の照射下における皮膚DNA損傷および皮膚DNA損傷修復に対する多糖体の影響を検証した。具体的には、ヘアレスマウスに紫外線を照射することによって皮膚のDNAを損傷させ、この状態のヘアレスマウスに対する多糖体の影響を検証した。
本実験例では、紫外線の照射下における皮膚DNA損傷および皮膚DNA損傷修復に対する多糖体の影響を検証した。具体的には、ヘアレスマウスに紫外線を照射することによって皮膚のDNAを損傷させ、この状態のヘアレスマウスに対する多糖体の影響を検証した。
(1)ヨーグルトの調製
10質量%の脱脂粉乳を含む培地にラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus)OLL1247菌およびストレプトコッカス・サーモフィラス(Streptococcus thermophilus)OLS3078菌を接種した後、その培地を43℃に保ちながら3時間発酵させた。このようにして得られたヨーグルトには、多糖体が110μg/g含まれていた。
10質量%の脱脂粉乳を含む培地にラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus)OLL1247菌およびストレプトコッカス・サーモフィラス(Streptococcus thermophilus)OLS3078菌を接種した後、その培地を43℃に保ちながら3時間発酵させた。このようにして得られたヨーグルトには、多糖体が110μg/g含まれていた。
なお、ヨーグルト中の多糖体の含有量は、フェノール硫酸法(Hodgeら,「Methods in carbohydrate chemistry」,第1巻,第338頁(1962年))に従って測定した。具体的には以下の通りである。
先ず、10gのヨーグルトに1gのトリクロロ酢酸を加えてよく撹拌した。次に、そのトリクロロ酢酸添加ヨーグルトを10000rpm、10分、4℃の条件下で遠心分離処理した後、その上清を別のチューブに移した。次いで、その上清に2倍容量の99.5%エタノールを加えてから、そのエタノール添加上清を冷凍庫で一晩放置したところ、チューブ中に沈殿物が生じていた。この沈殿物を10000rpm、10分、4℃の条件下で遠心分離処理した後、得られた沈殿物に超純水を3mL加えて多糖体抽出液とした。そして、500μLの多糖体抽出液に500μLのフェノール試薬(5%(w/v))を加えてその混合液を撹拌した後に、さらにその混合液に2.5mLの濃硫酸を加えてその混合液を直ぐに10秒間撹拌した。その後、その混合液を室温で20分以上放置してから、分光光度計でその混合液の490nmの吸光度を測定した。なお、次の通りに対照液を調製してから、上述と同様にしてその対照液の490nmの吸光度を測定した。500μLの標準グルコース溶液に500μLのフェノール試薬(5%(w/v))を加えてその混合液を撹拌した後に、さらにその混合液に2.5mLの濃硫酸を加えてその混合液を直ぐに10秒間撹拌した。その後、その混合液を室温で20分以上放置した。
(2)多糖体抽出物の調製
ヨーグルトの一部を分取し、その上清に3倍量のエタノールを添加して冷凍保管した。その後、その上清を遠心分離処理したところ、沈殿物が得られた。そして、この沈殿物を凍結乾燥して多糖体抽出物を得た。なお、11.3gのヨーグルト中に70mgの多糖体抽出物が含まれていた。
ヨーグルトの一部を分取し、その上清に3倍量のエタノールを添加して冷凍保管した。その後、その上清を遠心分離処理したところ、沈殿物が得られた。そして、この沈殿物を凍結乾燥して多糖体抽出物を得た。なお、11.3gのヨーグルト中に70mgの多糖体抽出物が含まれていた。
(3)紫外線照射によるDNA損傷
72匹のヘアレスマウス(Hos:HR-1,雌,8週齢,日本エスエルシー株式会社)を4日間馴化させた後、それらのヘアレスマウスを8匹ずつの群(紫外線照射前水群、紫外線照射前ヨーグルト群、紫外線照射前多糖抽出物群、紫外線照射後24時間水群、紫外線照射後24時間ヨーグルト群、紫外線照射後24時間多糖体抽出物群、紫外線照射後48時間水群、紫外線照射後48時間ヨーグルト群、紫外線照射後48時間多糖体抽出物群)に分けて、各群のヘアレスマウスに水、ヨーグルトおよび多糖体抽出物(以下これらをまとめて「試料」と称することがある。)をそれぞれ皮膚採取の前日まで経口投与した。なお、ヨーグルトは11.3g/kg体重/日で投与し、多糖体抽出物群は70mg/kg体重/日で投与した。そして、各試料投与開始から7日経過後に紫外線照射前水群、紫外線照射前ヨーグルト群、紫外線照射前多糖抽出物群のヘアレスマウスの背部皮膚をイソフルラン麻酔下採取した。また、各試料投与開始から7日経過後に残りの群のヘアレスマウスの背部に、0.4mW/cm2の照度で紫外線を50秒間照射した(なお、紫外線照射装置として三共電気株式会社製のGL20SE(波長領域:280nmから400nm,ピーク波長:306nm)を用いた。このときの紫外線量は20mJ/cm2(=0.4mW/cm2×50秒)である。)。そして、この紫外線照射から24時間経過時および72時間経過時に各群のヘアレスマウスの背部皮膚をイソフルラン麻酔下採取した。なお、以下、説明の便宜上、水を投与したヘアレスマウス群(比較例に相当)を「コントロール群」と称し、ヨーグルトを11.3g/kg体重/日で経口投与したヘアレスマウス群(実施例に相当)を「ヨーグルト群」と称し、多糖体抽出物を70mg/kg体重/日で経口投与したヘアレスマウス群(実施例に相当)を「多糖体抽出物群」と称する。

(4)評価
(4-1)紫外線によるDNA損傷の抑制効果の検証
上述の通りに採取した各群のヘアレスマウスの背部皮膚からquagen QIAamp DNA Mini Kit(キアゲン社製)を用いてDNAを採取して精製した。紫外線によるDNA損傷マーカーであるシクロブタンピリミジンダイマーの存在量をELISAキット(High Sensitivity CPD ELISA kit Ver.2, コスモバイオ社製)を用いて測定した。なお、標準品として、孔牛胸腺のDNAを0、2.5、5.0、7.5、10J/m2の紫外線量(紫外線波長は主に254nmであった。)で照射したDNAサンプル(UVC-irradiated DNA samples,コスモバイオ社製)を用いた。
(4-1)紫外線によるDNA損傷の抑制効果の検証

本実験例の結果は図1に示される通りであり、紫外線照射24時間後のシクロブタンピリミジンダイマーの存在量はコントロール群に比べてヨーグルト群、多糖体抽出物群で有意に低くなった。すなわち、ヨーグルトおよび多糖体抽出物に、紫外線照射による直接的なDNA損傷を抑制する効果が見られた。なお、図1中、「*」の記号はP<0.05でコントロール群に対して有意差あることを示している。
(4-2)活性酸素によるDNA損傷の抑制効果の検証
上述の通りに採取した各群のヘアレスマウスの背部皮膚からqiagen QIAamp DNA Mini Kit(キアゲン社製)を用いてDNAを採取して精製した。そして、DNase I、alkaline phosphatase、phosphodiesterase (8-OHdG Assay Preparation Reagent Set,和光純薬工業社製)を用いてこのDNAを分解した。その後、DNA損傷マーカーである8-オキソ-2’-デオキシグアノシン(8-OHdG)の存在量を定量した。
上述の通りに採取した各群のヘアレスマウスの背部皮膚からqiagen QIAamp DNA Mini Kit(キアゲン社製)を用いてDNAを採取して精製した。そして、DNase I、alkaline phosphatase、phosphodiesterase (8-OHdG Assay Preparation Reagent Set,和光純薬工業社製)を用いてこのDNAを分解した。その後、DNA損傷マーカーである8-オキソ-2’-デオキシグアノシン(8-OHdG)の存在量を定量した。
なお、この定量には高速液体クロマトグラフィ/タンデム質量分析計(4500QTRAP,サイエックス社製)を用いた。カラムとしてACQUITY UPLC BEH C18(1.7μm,2.1mmX100mm,Waters社製)を用いた。このとき、移動相Aとして0.1%ギ酸水溶液を、移動相Bとして0.1%ギ酸メタノール溶液を用いた。そして、開始時の移動相の組成を移動相A:100%とし、その後5分後に移動相の組成が「移動相A:50%,移動相B:50%」になるようにグラジエントしてから、移動相の組成を「移動相A:1%,移動相B:99%」にして1分間で保持し、さらに移動相の組成を移動相A:100%になるように切り替えて2分間保持した。ここで、高速液体クロマトグラフィ/タンデム質量分析計における1試料の測定時間を8分間、移動相の流速を0.3mL/分、カラム温度を40℃に設定し、エレクトロ・スプレー・イオン化を用いて、ポジティブモードで8-OHdGを検出した。高速液体クロマトグラフィ/タンデム質量分析計の分析変数を以下の通りに設定した。

本実験例の結果は図2に示される通りであり、いずれの時点においても、8-OHdGの存在量はコントロール群に比べてヨーグルト群、多糖体抽出物群で有意に低くなった。すなわち、ヨーグルトおよび多糖体抽出物に、活性酸素によるDNA損傷を抑制する効果が見られた。なお、図2中、「*」の記号はP<0.05でコントロール群に対して有意差あることを示している。
(4-3)紫外線による活性酸素生成の抑制効果の検証
上述の通りに採取した各群のヘアレスマウスの背部皮膚からRNeasy Mini Kit(キアゲン社製)を用いてTotal RNAを抽出した。その後、抽出したTotal RNAからRivertAid First Strand cDNA Synthesis Kit(サーモフィッシャー社製)を用いてcDNAを合成した。そして、ABI 7500 Fast Realtime PCR system(アプライドバイオシステムズ社製)を用いて、そのcDNAおよび内在性遺伝子としてのGAPDHからカタラーゼのmRNAの相対量を定量した。
上述の通りに採取した各群のヘアレスマウスの背部皮膚からRNeasy Mini Kit(キアゲン社製)を用いてTotal RNAを抽出した。その後、抽出したTotal RNAからRivertAid First Strand cDNA Synthesis Kit(サーモフィッシャー社製)を用いてcDNAを合成した。そして、ABI 7500 Fast Realtime PCR system(アプライドバイオシステムズ社製)を用いて、そのcDNAおよび内在性遺伝子としてのGAPDHからカタラーゼのmRNAの相対量を定量した。
本実験例の結果は図3に示される通りであり、紫外線照射後24時間後、72時間後において抗酸化酵素であるカタラーゼのmRNAの相対量はコントロール群に比べてヨーグルト群および多糖体抽出物群で有意に増加した。すなわち、ヨーグルトおよび多糖体抽出物に、紫外線照射による活性酸素の生成を抑制する効果が見られた。ヨーグルトおよび多糖体抽出物が紫外線照射による活性酸素の生成を抑制した結果、活性酸素により生成するDNA損傷8-OHdGが低下したためであると考えられた。なお、図3中、「*」の記号はP<0.05でコントロール群に対して有意差あることを示している。
(4-4)DNA損傷修復促進効果の検証
上述の通りに採取した各群のヘアレスマウスの背部皮膚からRNeasy Mini Kit(キアゲン社製)を用いてTotal RNAを抽出した。その後、抽出したTotal RNAからRivertAid First Strand cDNA Synthesis Kit(サーモフィッシャー社製)を用いてcDNAを合成した。そいsて、ABI 7500 Fast Realtime PCR system(アプライドバイオシステムズ社製)を用いて、そのcDNAおよび内在性遺伝子としてのGAPDHからxeroderma pigmentosum complementation group A(XPA)のmRNAの相対量を定量した。

本実験例の結果は図4に示される通りであり、紫外線照射後24時間後、72時間後においてDNA修復たんぱく質であるXPAのmRNAの相対量はコントロール群に比べてヨーグルト群および多糖体抽出物群で有意に増加した。すなわち、ヨーグルトおよび多糖体抽出物に、紫外線照射や活性酸素により生じるDNA損傷の修復を促進する効果が見られた。なお、図4中、「*」の記号はP<0.05でコントロール群に対して有意差あることを示している。

Claims (8)
- 多糖体を含有する乳酸菌産生物を有効成分とするデオキシリボ核酸保存用組成物。
- 前記乳酸菌産生物は、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクス(Lactobacillus delbrueckii subsp. bulgaricus)とストレプトコッカス・サーモフィラス(Streptococcus thermophilus)との組合せによって生成される
請求項1に記載のデオキシリボ核酸保存用組成物。 - 前記ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクスは、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリクスOLL1247菌(受託番号:NITE BP-01814)であり、
前記ストレプトコッカス・サーモフィラスは、ストレプトコッカス・サーモフィラスOLS3078菌(受託番号:NITE BP-01697)である

- 請求項1から4のいずれかに記載のデオキシリボ核酸保存用組成物を経口投与することによりデオキシリボ核酸を保存する方法(ただし、人を治療する行為を除く。)。
- デオキシリボ核酸保存用組成物としての使用のための、多糖体を含有する乳酸菌産生物。
- デオキシリボ核酸を保存するための組成物の製造のための、多糖体を含有する乳酸菌産生物の使用。
- 多糖体を産生する乳酸菌に乳原料を供給してデオキシリボ核酸保存用組成物を製造する方法。
