Abstract

본 발명은 TNP(N2-(m-Trifluorobenzyl), N6-(p-nitrobenzyl)purine)를 유효성분으로 함유하는 아세트아미노펜 (Acetaminophen, AP) 유래 간 독성 예방 및 치료용 조성물에 관한 것으로, 5-이노시톨 파이로인산 (inositol pyrophosphate)의 저해제로 알려진 TNP가 인간 배아 줄기세포 유래 간세포, 생쥐 유래 간세포 및 인간 간암 세포주에서 아세트아미노펜에 의한 세포 사멸을 억제하고, 간세포 내 환원된 글루타치온 (glutathione, GSH)의 농도를 증가시켰으며, 아세트아미노펜에 의해 증가하는 스트레스 반응인 인산화된 JNK를 억제하는 것을 확인하고, 동물 모델에서 상기와 동일한 아세트아미노펜-유발성 간세포독성의 보호효과를 갖는 것을 확인함으로써, 상기 TNP를 아세트아미노펜에 의한 간 독성 예방 및 치료용 조성물의 유효성분으로 유용하게 사용할 수 있다.

【기술분야】
본 발명은 TNP(N2-(m-Tr i f luorobenzy 1) , N6- (p-n it robenzyl) purine)또는 이의 약학작으로 허용가능한 염을 유효성분으로 함유하는 간 독성 예방 및 치료용 약학적 조성물, 및 건강기능식품에 관한 것이다.
【배경기술】
TNP ( N2- ( m-Tr i ί 1 iaor obenzy 1 ), N6-()-ni:tr obenzy 1 -) ur ie)는 상업적으로 판매가 가능한 화학물질로서 현재까지 알려진 바로는 이노시를 펜타키포스페이트 키'나아제 (inositol pentakisphosphate kinase) 특이적 저해약물이다 (J Biol Chem. Apr 17, 2009; 284(16): 1057110582) . TNP는 5ᅳ이노시를 파이로인산 (inositol pyrophosphate)의 생합성을 억제함으로써, 세포 내 5-이노시를 다인산 (polyphosphate)의 수준을 낮출 뿐 아니라, 간세포의 인슐린 신호전달반웅을 증가시킨다는 효과가 보고되어 있다 (Cell. 2010; 143(6): 897910) . 또한, 골수 유래 중간엽 줄기세포에 처뫼할 경우, 세포의 노화진행반웅을 감소시키는 효과도 알려져있다 (St em Cell Res Ther.2014 Mar 26 ;5(2) :33). 그러나, TNP 약물에 대한 급성 간 독성을 예방 또는 치료하는 효과는 아직 보고된 바 없다.
1950년대 미국에서 개발되어 타이레놀로 잘 알려진 아세트아미노펜 (Acetaminophen, AP)은 진통 및 해열제로 세계에서 가장 보편적으로 사용되는 약물의 일종 (타이레놀의 국내 1년 매출액 250억원)으로 그 화학명칭은 N-아세틸 -ρ-아미노페놀 (N-acetyl_p-aminophenol)이고, 파라세타몰 (paracetamol)이라고도 불린다 (Dargan PI et al. , Crit Care. 6(2) :108-10. 2002) . 아세트아미노펜은 체중 1 kg당 하루 150 mg까지, 성인의 경우 하루에 최고 4 g까지 경구 투여 가능하다고 알려져 있어, 의사의 처방이 없어도 구입할 수 있는 안전한 일반의약품으로 분류되어 있다. 그러나 아세트아미노펜은 과량복용하면 급성 간부전 (fulminant hepatic failure), 간 괴사 (liver necrosis), 신경독소 (nephrotoxicity), 간'경화 ( 1 iver cirrhosis)를 초래해 심할 경우 사망에 이르기도 하는 양면상을 가진 물질이다.
이러한 간 독성의 데카니즘 (mechanism)은 아세트아미노펜이 일부 간 산화효소의 작용을 받아 세포를 파괴하는 반웅성 물질로 변한다는 것으로 보통 이러한 물질이 생성되더라도 해독되가 때문에 문제되지 않지만 과량을 복용할 경우 체내 해독물질이 교갈되어 결과적으로 간세포를 파괴하게 된다. 구체적으로, 낮은 농도의 아세트아미노펜을 투약하면, 체내에서 생변이를 통해 비독성 글루코논산 (gfeicononic acid)과 황산염 (sul f ate)과 결합하여 독성을 제거해 담즙 또는 혈장으로 배출되고, 5- 10%는 P450(CYP), 특히 CYP2E1에 의해서 대사 된다 (Ray SDet. al. , J Pharmacol Exp Ther . 1996 ;279: 1470—83) . 즉 아세트아미노펜은 간에서 N-아세틸 -p-벤조퀴논이민 (N-acetyl-p-benzoquinoneimine, NAPQI)으로 전환되는데, 전환된 대사산물은 글루타치온과 결합하여 독성을 나타내지 않는다 (Hazai E, et al. , Biochem Biophys Res Commun. 291(4): 1089-1094. 2002). 그러나 과량의 아세트아미노펜이 투약되면, 간에서 글루콘산과 황산염이 결합하여 비독성을 나타내는 능력이 상실되어 높은 반웅성 NAPQI가 축적되면서 세포막을 치명적으로 손상시켜 간세포의 사멸을 유도하고 회복할 수 없게 하여 간 독성을 유발하게 되고, 결국 생명체를 사망에 이르게 한다 (Webster PA et . al. , J Clin Pharmacol. 1996; 36: 397-402 ; Albano E et. al. , Mol Pharmacology. 1985;28:306-11 ; Kyle ME et . al. , Biochem Biophys Res Commun. 1987;149:889-94 ; Mahadevan SB et . al . , Arch Dis Child. 2006:91:598-603). 아세트아미노펜 투여에 의한 간 손상 모델로는 마우스와 햄스터가 잘 알려져 있고, 인체에서도 비슷한 현상이 보고되고 있다 (Tee LB et a., Toxicol Appl Pharmacol. 83(2) :294-314. 1986). 게다가 알코올을 함께 흡수할 경우 간 독성의 피해가 더욱 심각하게 나타나 미국식품의약청 (FDA)은 1998년에 매일 세 잔 이상 정기적으로 술을 마시는 사람은 간 독성이 유발될 수 있으니 이 약을 복용해야 할 경우 반드시 의사와 상의해야 한다는 사실을 의무적으로 알리도록 하였다. 최근에 이러한 아세트아미노펜의 간 독성을 제거 및 완화시킬 수 있는 약물을 발굴하기 위한 연구가 활발히 진행되고 있다. 간은 인체의 혈액데사를 주관하고 해독기능을 담슝하는 주요한 장기로 간이 손상되면, 염증반응과 동시에 인체에 각종 질환을 야기하기 때문에 간 손상을보호할 수 있는 약물의 개발은 대단히 중요하다. 이에, 본 발명자들은 간 손상의 예방 및 치료에 효과가 있는 물질을 찾고자 노력한 결과, 5-여노사를 파이로인산의 저해제로 알려진 TNP가 인간 배아 줄기세포 유래 간세포, 생쥐 유래 간세포 및 인간 간암 세포주에서 아세트아미노펜에 와한 세포 사멸을 억제하고, 간세포 내 환원된 글루타치온의 농도를 증가시켰으며, 아세트아미노펜에 의해 증가하는 스트레스 반웅인 인산화된 JNK를 억제하는 것을 확인하고, 동물 모델에서 상기와 동일한 아세트아미노펜-유발성 간세포독성의 보호효과를 갖는 것을 확인함으로써, 상기 TNP 또는 이의 약학적으로 허용가능한 염을 간 독성 예방 및 치료용 조성물의 유효성분으로 유용하게 사용할 수 있음을 밝힘으로써 본 발명을 완성하였다.
【발명의 상세한 설명】

본 발명의 목적은 TNP(N2-(m-Tri f luorobenzyl ) , N6-(p-ni trobenzyl )pur ine) 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 간 독성 예방 및 치료용 약학적 조성물을 제공하는 것이다. 【기술적 해결방법】

【발명의 실시를 위한 최선의 형태】
이하, 본 발명을 상세히 설명한다. 본 발명은 TNP(N2-(m-Tr i f luorobenzyl ) , N6—(p-ni t robenzyl ) pur ine)또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 간 독성 예방 및 치료용 약학적 조성물을 제공한다.

상기 TNP는 아세트아미노펜 유래 세포 사멸을 억제하고, 간세포 내. 글루타치은 (glutathione , GSH)을 증가시키며, 아세트아미노펜에 의한 스트레스 반응 증가를 억제하는 것이 바람직하나 이에 한정되지 않는다. 본 발명의 구체적인 실시예에서, 본 발명자들은 인간 배아 줄기세포 유래 간세포, 생쥐 유래 간세포 및 인간 간암 세포주 (HepG2)에서 아세토아미노펜에 의해 유도되는 세포 사멸의 TNP 처리에 따른 변화를 확인한 결과, 아세트아미노펜에 의해 유도되는 간세포 사멸이 TNP 처리에 의해 유의적으로 저해되는 것을 확인하였고 (도 2 내지 도 5 참조), TNP 처리에 따른 아세트아미노펜에 의한 간세포 대사능을 확인한 결과, 아세트아미노펜에 의한 간세포 내에 환원된 글루타치온의 감소효과가 TNP 처리에 의해 저해됨을 확인하였으며 (도 6 참조), 생쥐 유래 간세포 및 인간 간암 세포주 (HepG2)에서 TNP 처리에 따른 아세트아미노펜 유래 간세포 스트레스 반웅조절 효과를 확인한 결과, 아세트아미노펜 처리에 의한 스트레스 반웅의 증가로 인산화된 JNK 신호의 증가를 유발하였으나, TNP 처리에 의하여 이러한 인산화된 JNK 증가가 유의하게 억제됨을 확인하였다 (도 7 내지 도 8 참조) . 또한, 생쥐모델에 아세트아미노펜을 투여하고, TNP를 주사한 결과, 개체생존율이 증가하며 (도 9참조), 생쥐에서 적출한 간의 괴사 반웅이 감소하고 (도 10참조), 간독성의 혈청지표인 AST, ALT수준이 현저히 감소하며 (도 11 참조), 인산화된 JNK 증가가 유의하게 억제됨을 확인하였다 (도 12 참조) .
따라서, 본 발명의 5—이노시를 파이로인산 ( inos i tol pyrophosphate)의 저해제로 알려진 TNP가 인간 배아 줄기세포 유래 간세포ᅳ 생쥐 유래 간세포 및 인간 간암 세포주에서 아세트아미노펜에 의한 세포 사멸을 억제하고, 간세포 내 환원된 글루타치온 (glutathione , GSH)의 농도를 증가시켰으며, 아세트아미노펜에 의해 증가하는 스트레스 반웅인 인산화된 JNK를 억제하는 것을 확인하고, 동물 모델에서 상기와 동밀한 아세트아미노펜-유발성 간세포독성의 보호효과를 갖는 것을 확인함으로써, 상기 TNP 또는 이의 약학적으로 허용가능한 염을 간 독성 예방 및 치료를 위한 약학적 조성물로 유용하게 사용할 수 있다.. 본 발명은 화학식 1로 표시되는 TNP 뿐받 아니라, 이의 약학적으로 허용되는 염, 이로부터 제조될 수 있는 가능한 용매화물, 수화물, 라세미체, 또는 입체이성질체를 모두 포함한다. 본 발명의 화학식 1로 표시되는 TNP는 약학적으로 허용 가능한 염의 형태로 사용할 수 있으며, 염으로는 약학적으로 허용 가능한 유뫼산 ( free acid)에 의해 형성된 산 부가염이 유용하다. 산 부가염은 염산, 질산, 인산, 황산, 브롬화수소산, 요드화수소산, 아질산 또는 아인산과 같은 무기산류와 지방족 모노 및 디카르복실레이트, 페닐-치환된 알카노에이트, 하이드록시 알카노에이트 및 알칸디오에이트, 방 족 산류, 자방족 및 방향족 설폰산류와 같은 무독성 유기산으로부터 얻는다. 이러한 약학적으로 무독한 염류로는 설페이트, 피로설페이트, 바이설페이트, 설파이트, 바이설파이트, 니트레이트 포스페이트, 모노하이드로겐 포스페이트, 디하이드로겐 포스페이트 메타포스페이트, 피로포스페이트 클로라이드, 브로마이드, 아이오다이드 플루오라이드, 아세테이트, 프로피오네이트, 데카노에이트, 카프릴레이트 아크릴레이트, 포메이트, 이소부티레이트, 카프레이트, 헵타노에이트 프로피올레이트,옥살레이트, 말로네이트, 석시네이트, 수베레이트, 세바케이트 푸마레이트, 말리에이트, 부틴 -1 , 4-디오에이트, 핵산 -1,6-디오에이트, 벤조에이트, 클로로벤조에이트, 메틸벤조에이트, 디니트로 벤조에이트, 하이드록시벤조에이트, 메특시벤조에이트, 프탈레이트, 테레프탈레이트, 벤젠설포네이트, 를루엔설포네이트, 클로로벤젠설포네이트ᅳ 크실렌설포네이트, 페닐아세테이트, 페닐프로피오네미트, 페닐부티레이트, 시트레이트, 락테이트, 하이드록시부티레이트, 글리콜레이트, 말레이트, 타트레이트, 메탄설포네이트, 프로판설포네이트, 나프탈헨 -1-설포데이.트, 나프탈 ¾ 2-설포네이트 또는 만델레이트를 포함한다. 본 발명에 따른 산 부자염은 통상의 방법, 예를 들면, 화학식 1로 표시되는 TNP를 과량의 산 수용액 중에 용해시키고, 이 염을 수흔화성 유기 용매, 예를 들면 메탄을, 혜한올, 아세톤 또는 아세토니트릴을 사용하여 침전시켜서 제조할 수 있다. 또한, 이 흔합물에서 용매나 과량의 산을 증발시켜서 건조하거나 또는 석출된 염을 흡입 여과시켜 제조할 수도 있다. 또한, 염기를 사용하여 약학적으로 허용 가능한금속염을 만들 수 있다. 알칼리 금속 또는 알칼리 토금속 염은, 예를 들면 화합물을 과량의 알칼리 금속 수산화물 또는 알칼리 토금속 수산화물 용액 중에 용해하고, 비용해 화합물 염을 여과하고, 여액을 증발, 건조시켜 얻는다. 이때, 금속염으로는 나트륨, 칼륨 또는 칼슘염을 제조하는 것이 제약상 적합하다. 또한, 이에 대응하는 은 염은 알칼리 금속 또는 알칼리 토금속 염을 적당한 음염 (예, 질산은)과 반웅시켜 얻는다. 상기 조성물을 제제화할 경우, 보통 사용하는 층진제, 증량제, 결합제, 습윤쎄, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 제조된다. 경구투여를 위한 고형 제제에는 정제, 환자, 산제, 과립제, 캡슐제, 트로키제 등이 포함되며, 이러한 고형 제제는 하나 이상의 본 발명의 화학식 1로 표시되는 TNP에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로스 (sucrose) 또는 락토오스 ( lactose) 또는 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다. 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제 또는 시럽제 등이 해당되는데, 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 었다.
비경구 투여를 위한 체 :체에는 열균된 수용액, 비수성용쎄, 현탁용제, 유제, 동결건조제제, 좌제 등이 포함된다.
비수성용제ᅳ 현탁용제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 을리브 오일과 같은 식물성 기름, 에될올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔 (w epsol ) , 마크로골, 트원 ( tween) 61, 카카오지, 라우린지, 글리세를, 젤라틴 등이 사용될 수 있다. 본 발명에 따른 조성물은 약제학적으로 유효'한 양으로 투여한다. 본 발명에 있어서, "약제학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜 /위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
구체적으로, 본 발명에 따른 화합물의 유효량은 환자의 나이, 성별, 체중에 따라 달라질 수 있으며, 일반적으로는 체중 1 kg당 0. 1 mg내지 100 mg, 바람직하게는 0.5 mg내지 10 mg을 매일 또는 격일 투여하거나 1일 1내지 3회로 나누어 투여할 수 있다. 그러나 투여 경로 비만의 중증도, 성별, 체중, 연령 등에 따라서 증감될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다. 또한, 본 발명은 TNP 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 간 독성 예방 및 개선용 건강기능식품을 제공한다.
상기 TNP는 상기 화학식 1의 구조를 갖는 것이 바람직하나 이에 한정되지 않는다.
상기 TNP는 아세트아미노펜 유래 세포 사멸을 억제하고, 간세포 내 글루타치온 (glutathione , GSH)을 증가시키며, 아세트아미노펜에 의한 스트레스 반웅 증가를 억제하는 것이 바람직하나 어에 한정되지 않는다.

. - 본 발명의 TNP가 상기와 같은 간 독성 예방 및 개선을 위해 이용되기 위해서는, 식품학 또는 약제학적 분야에서 공지된 다양한 방법에 의해 제조될 수 있으며 그 자체 또는 식품학적으로 허용되는 담체, 부형제, 희석제 등과 흔합하여 경구로 섭취할 수 있는 어떤 식품 형태로도 제조될 수 있다. 바람직하게는 음료, 환, 과립, 정제 또는 캅셀 형태이다.
본 발명의 건강기능식품은, 식품 제조 시에 통상적으로 첨가되고 식품학적으로 허용되는 성분을 더 포함할 수 있다. 예컨대, 음료수로 제조되는 경우에는 본 발명의 TNP이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙 등에서 하나 이상의 성분을 추가로 포함시킬 수 있다.
본 발명에 따른 건강기능식품의 유효성분으로 포함될 수 있는 양은 간 독성 예방 및 개선을 원하는 사람의 연령, 성별, 체중, 상태, 질병의 증상에 따라 적절히 선택될 수 있으며, 바람직하게는 성인기준 1일 0.01 g내지 10.0 g 정도로 포함되는 것아 좋으며, 이러한 함량을 갖는 건강기능식품을 섭취함으로써 간 독성 예방 및 개선 효과를 얻을 수 있다.

단, 하기 실시예 및 실험예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예 및 실험예에 한정되는 것은 아니다.
<실시예 1>인간 배아줄기세포 (hESC) 유래 간세포의 배양
인간 배아 줄기세포 (hESC)로부터 내배엽과 같은 다른 분화적 계통 세포로의 분화 여부를 확인하기 위하여, 인간 배아 줄기세포로부터 내배엽인 간세포 (hepatocytes)로의 분화를 유도하였다 (Cai, J. et . al, (2007) Hepatol ogy 45(5): 1229-1239).
구체적으로, hESC 세포주는 CHA-hESC 세포주로 지지세포가 없는 시스템 (feederrfree system)에서 조절된 wfl ^(conditioned medium)에 가득 차 (confluent)도톡 3일 동안 배양하였다. 배양 후, 상기 hESC세포를 50 ng/m£ 액티빈 MActivin A; Peprotech 사, 미국)를 포함하는 RPMI— 1640 (Hyc lone 사, 미국) 배지에서 5 일간 배양하여 분화를 유도하였다. 상기 분화된 세포는 30 ng/mi 섬유아세포 성장인자 4(fibroblast growth factor4; Peprotech사) 및 20 ng/mi 골 형성 단백질 2 (bone morphogenet ic protein 2, BMP2; Peprotech사)를 포함하는 간세포 배양 배지 (hepatocyte culture medium, HCM; Lonza 사, 미국)에서 5 일간 배양한 후, 20 ng/mi 간세포 성장 인자 (hepatocyte growth factor, HGF; Peprotech 사)를 첨가한 간세포 배양 배지에서 5일 동안 추가로 배양하여 hESC로부터 간세포로 분화를 유도하였다. 상기 분화된 간세포는 10 ng/nrf 온코스태틴 M oncostatin M; R&D Systems 사, 미국) 및 0.1 μΜ 덱사메타손 (dexametasone; 시그마-알드리치 사, 미국)을 첨가한 간세포 배양 배지에서 5 일간 배양하면서 간세포의 성숙 (maturation)을 유도하여 성숙된 간세포를 수득하였다. <실시예 -2> 생쥐 유래 간세포의 배양
생쥐 유래 간세포 배양은 콜라게나아제 (coUagenase) 관류법을 이용하여 다음과 같이 실시하였다.
구체적으로, 10-12 주령의 수컷 C57BL/6J 생취에 에버틴을 복강 내로 주사하여 마취한 후 개복하였으며ᅳ 간문맥에 24 게이지 (gauge) 카테터 (catheter)를 삽관하여 관류 (Perfusion) 용액 (1.42 M Nacl, 0.067 M Kcl, 0.1 M HEPES PH 7.4, 0.05 M EGTA)으로 관류시키고 콜라게나아제 (Worthington, USA)로 구성된 소화용액을 순환 시켰다. 간세포가 소화된 후 간막을 벗겨 간세포가 유리되게 하였으며, 나일론 메시 (nylon mesh)를 사용하여 여과하였다. 여과액은 100 X g에서 3분간 원심분리한 후 상등액을 버리고 침전된 간세포를 HBSSCHanks ' Balanced salt solution, WelGene) 완충용액에 현탁시켜 원심분리하였다. 침전된 간세포를 페르콜 (Percol) 용액 (Autoclaved percolKsigma), 10 X PBS, 1M HEPES PH7.4)에 2400 rpm에서 10분간 원심분리한 후 상등액을 버리고 침전된 간세포를 HBSSCHanks' Balanced salt solution, WelGene) 완충용액에 현탁시켜 100 X g로 다시 원심분리하였다. 침전된 간세포를 배양액에 현탁시켜 세포 현탁액을 얻은 뒤, 간세포 농도가 1 X 10 cell/ml이 되도록 조절하여, 젤라틴 (gelatin)으로 미리 도포된 배양용기에 이식하였다. 배양액은 10% FBS, 10 nM dexa, 10 nM 인슐린 (insulin), 페니실린 /스트렙토마이신 (penicillin/streptomycin lOO g/ml)을 포함하는
DMEM 배지 (bic est)를 사용하였으며 일정한 온도와 습도가 유지되는 370C 배양기에서 C025>의 흔합기체를 공급하여 배양하였다. <실시예 3>인간간암세포주 (HepG2)의 배양
인간 간암 세포인 HepG2 세포는 젤라틴으로 미리 도포된 6-웰 플레이트에 세포농도 1 X 10 cell/n 로 분주한 다음 세포부착을 위해 24시간 배양하였다. 배양액은 10% FBS, 10 nM dexa, 10 nM 인슐린, 페니실린 /스:^ ί마이신 a¾0 irl r 첨가한 DMEM쒜지 (biowest)로 일정한 온도와 습도가 유지되는 370C 배양기에서 C025%의 흔합기체를 공급하여 배양하였다.
<실험예 1> 인간 배아 줄기세포 유래 간세포에서 TNP 처리에 따른 아세트아미노펜 유래 세포사멸 억제 확인
상기 <실시예 1>과 동일한 방법으로 배양한 인간 배아 줄기세포 유래 간세포에서 아세트아미노펜에 의해 유도되는 세포 사멸의 TNP 처리에 따른 변화를 확안하였다.
구체적으로, 인간 줄기세포로부터 분화 19일째 간세포를 6-웰 플레이트에 각 웰 당 9OT농도로 분주하였다. 1000 mg/ί DMEM—낮은 포도당 (low glucose) 배양액 (Dulbecco's Modified Eagle's Medium, 제조사 Wei gene No. 001-11)을 사용하여 교체한 후 2시간 동안 배양하였다. 아세트아미노펜 25 mM 단독처리군과 아세트아미노펜 25 mM, TNP 100 μΜ의 농도로 흔합처리한군을
18시간 처리한 후 세포를 현미경으로 관찰하였다. 3회 이상 반복실험을 바탕으로 세포 생존율을 분석하였다. 세포 생존율 분석은 PBS 1 로 2차례에 걸쳐 세포배양액을 제거한 후,트립신 (Trypsin)/EDTA흔합액 (Invitrogen) 1 m£을 1분간 처리한 후, 피펫팅으로 세포를 수거하였다. 트립판 블루 와 수거한 세포를 함유하는 PBS 10 ≠를 흔합한 후 자동 세포 생존율 분석기
Countess( Invi trogen)를 이용하여 세포 생존율 결과를 얻었다. 본 발명의 통계처리는 모든 실험값의 평균 ±표준오차 (mean ± SD)로 표시했으며, 통계분석은 Student ' s t-test로 처리했으며, 유의성 최대 한계는 p<0.05로 정하였다.
그 결과 도 2에 나타낸 바와 같이, 아세트아미노펜에 의해 유발되는 간세포 사멸이 TNP 처리에 의하여 유의적으로 저해되는 것을 확인하였다 (도
2) . <실험예 2> 생쥐 유래 간세포에서 TNP 처리에 따른 아세트아미노펜 유래 세포사멸 억제 확인
상기 <실시예 .2>와 동일한 '방법으로 쎄양한 생쥐 유래 간세포에서 아세트아미노펜에 의해 유도되는 세포 사멸꾀 TNP 처리에 따른 변화를 확인하였다.
구체적으로, 생쥐 유래 간세포는 6-웰 플레이트에 각 웰 당 100% 농도로 분주한 후, 상기 <실험예 1〉과 동일한 방법으로 세포 생존율 결과를 얻었다. 본 발명의 통계처리는 모든 실험값은 평균 ±표준오차 (mean ± SD)로 표시했으며, 통계분석은 Student ' s t-test로 처리했으며, 유의성 최대 한계는 p<0.05로 정하였다.
그 결과 도 3에 나타낸 바와 같이, 아세트아마노펜에 의해 유발되는 간세포 사멸이 TNP 처리에 의하여 유의적으로 저해되는 것을 확인하였다 (도

<실험예 3> 인간 간암 세포주 (HepG2)에서 TNP 처리에 따른 아세트아미노펜 유래 세포사멸 억제 확인
상기 <실시예 3〉과 동일한 방법으로 배양한 인간 간암 세포주 (HepG2)에서 아세트아미노펜에 의해 유도되는 세포 사멸의 TNP 처리에 따른 변화를 확인하였다.
구체적으로, HepG2 인간 간암 세포주는 6—웰 플레이트에 각 웰 당 90% 농도로 분주한 후, 상기 <실험예 1>과 동일한 방법으로 세포 생존율 결과를 얻었다. 본 발명의 통계처리는 모든 실험값은 평균 ±표준오차 (mean ± SD)로 표시했으며, 통계분석은 Student ' s t_test로 처리했으며, 유의성 최대 한계는 p<0.05로 정하였다.
그 결과 도 4 내지 도 5에 나타낸 바와 같이, 아세트아미노펜에 의해 유발되는 간세포 사멸이 TNP 처리에 의하여 유의적으로 저해되는 것을 확인하였다 (도 4 내지 도 5) .
<실험예 .4> 생쥐 유래 간세포 서 rap 처리에 따른 아세트아미노펜 유래 세포 내 글루타치온 (glut hi one, GSE) 소 겨해 확인
TNP 처리에 따른 아세트아미노펜에 의한 간세포 대사능을 확인하였다. 구체적으로, 상기 <실험예 2>와 동일한 방법으로 아세트아미노펜 및 TNP를 처리한 생쥐 유래 간세포를 이용하여 환원된 글루타치은 (reduced glutathione , GSH) 농도를 측정하였다. 환원된 글루타치온 측정은 GSHᅳ Glo Glutathione Assay 키트 (Promega)를 사용하였으며, Tr i star2 LB 942 플레이트 분석기를 활용하였다.
그 결과 도 6에 나타낸 바와 같이, TNP 단독처리에 의해 간세포 내에 환원된 글루타치온의 농도가 증가함을 확인하였고, 또한, 아세트아미노펜에 의한 간세포 내에 환원된 글루타치온의 감소효과가 TNP 처리에 의해 저해됨을 확인하였다 (도 6) .
<실험예 5> 생쥐 유래 간세포에서 TOP 처리에 따른 아세트아미노펜 유래 JNK 인산화증가 억제 확인
TNP 처리에 따른 아세트아미노펜 유래 간세포 스트레스 반웅조절 효과를 확인하였다. ' 구체적으로, 상기 <실험예 2>와 동일한 방법으로 아세트아미노펜 및 TNP를 처리한 생쥐 유래 간세포를 수거한 후, 단백질 분석을 위하여 세포 추출액을 준비한 후 SDSᅳ PAGE 분석 및 웨스턴 블롯팅을 수행하였다. 세포는 단백잘분해효소 칵테일 (protease inhibitor cocktai 1 ) (Complete Mini , Roche , 독일)을 포함하는 10 mi SDS완충용액 (50 mM Tris-HCl, 1% IGEPAL CA-630, 0.25% deoxychol ic acid, 150 mM NaCI , 1 mM ethylene-diamine tetra acetic acid, 1 mM NaF, 0.1% sodium dodecyl sulfate, )1 7.4)으¾ 용해하였고, 20분 동안 얼음에 놓아둔 후, 40C에서 13,000 rpm으로 10분간 원심분리하였다. 원심분리 후 상층액의 단백질 농도를 확인한 뒤 &i:0" ad .Laboratories, 미국), SDS-PAGE( SDS-po 1 yacry 1 am i de gel electrophoresis)를 위해 50 의 단백질을 로딩 (loading)하였다. 전기영동한 단백질을 일렉트로블랏팅 (electroblott ing) 장치를 이용하여 Immobilon-P transfer 멤브레언 (Mill ipore Corporate, 미국)으로 이동시킨 뒤,상기 멤브레인은 0.1%트왼 -20(Tween-20)및 10%분유를 포함하는 TBS(Tris-buffered saline)에 넣어 1시간 동안 블로킹 (blocking)한 후, 인산화된 JNK Cell Signaling사) 및 전체 JNK. Cell Signaling사) 항체를 넣고 밤새 반웅시켰다. 그리고나서, 멤브레인은 0.1% 트원 -20을 포함하는 TBS로 20분씩 3번 세척하고, HRP(horseradish peroxidase)가 결합된 2차 항체를 넣고 1시간 동안 반웅시킨 후, 0.1% 트원 -20을 포함하는 TBS로 20분씩 3번 세척하였다. 단백질 밴드는 웨스턴 블랏팅 루미놀 시약 (luminol reagent) (Santa Cruz Biotechnology)을 이용하여 시각화하였다. 활성도는 Image J 소프트웨어 (version 1.37, NIH, 미국)를 이용하여 분석하몄다.
그 결과 도 7에 나타낸 바와 같이, 아세트아미노펜 처리에 의한 스트레스 반웅의 증가로 인산화된 JNK신호의 증가를 유발하였으나, TNP처리에 의하여 인산화된 JNK 증가가 유의하게 억제됨을 확인하였다 (도 7).
<실험예 6> 인간 간암 세포주 (HepG2)에서 TNP 처리에 따른 아세트아미노펜 유래 JNK 인산화증가 억제 확인 TNP 처리에 따른 아세트아미노펜 유래 간세포 스트레스 반웅조절 효과를 확인하였다.
구체적으로, 상기 <실험예 3>과 동일한 방법으로 아세트아미노펜 및 TNP를 처리한 HepG2인간 간암 세포주를 수거한 후, 단백질 분석을 위하여 세포 추출액을 준비한 후 상기 <실험예 5>와 동일한 방법으로 SDS-PAGE 분석 및 웨스턴 블롯팅을 수행하였다.
그 결과 도 8에 나타낸 바와 같이, 아세트아꾀노펜 처리에 의한 스트레스 반웅의 증가로 인산화된 JNK신호의 증가를 유발하였으나 , TNP처리에 의하여 인산화된 JNK 증가가 유의하게 억제됨을 확인하였다 (도 8) .
<실시예 7 생쥐모델에서의 아세트아미노펜 및 TNP 처리에 따른 개체생존율 분석
TNP에 의한 아세트아미노펜-유발성 간독성 예방 효과를 동물 실험을 통해 확인하였다.
구체적으로, 8 내지 10주령의 수컷 C57BL/6 생쥐를 이용하여 500 nig/ kg 아세트아미노펜 (Sigma)을 물에 녹여 생쥐에 경구 투여한 후 10 mg/kg TNP(Caibiocham)를 올리브오일 (Sigma)에 녹인 후 생쥐 복강 내 주사하여 개체생존율을 분석하였다. 통계분석은 Kaplan-Meier survival analysi s로 분석하였으며, 유의성 최대 한계는 ρθ.05로 정하였다.
그 결과, 도 9에 나타난 바와 같이, TNP 처리에 꾀하여 아세트아미노펜ᅳ유발성 간독성에 대한 현저한 개체생존율의 보호효과를 확인하였다 (도 9) .
<실시예 8> 생쥐모델에서의 아세트아미노펜 및 TNP 처리에 따른 간독성 분석
상기 <실시예 7>의 동물 모델에서 TNP 처리에 의한 아세트아미노펜-유발성 간독성 예방 효과를 조직, 혈액 및 JNK 인산화 활성을 분석하여 확인하였다. 구체적으로, 아세트아미노펜 및 TNP가 주입된 <실시예 7>의 수컷 생쥐에서 TNP 주사 후 6시간 후 간을 적출하여 조직병리학적 분석, 혈액분석 (AST, ALT) 및 스트레스 신호전달 활성 (JNK)을 분석하였다. 혈액분석은 생쥐의 안와채혈로 수득한 혈액 샘플을 13000 rpm에서 10분간 원심분리하여 얻어진 혈청 100 ^으로 AST, ALT를 분석 ( Idexx VetTest8008 chemi stry analyzer)하였으며, 스트레스 신호전달 활성을 분석하기 위해, 적출한 간 조직을 RIPA배지 ( IM tr i s— cl (PH7.4) , IM Nacl , 0.5M EDTA, NP-40 , 10% sodium deoxycholate , 10% SDS)에 넣고, 호모게나이저로 분해한 다음, 원심분리 후 상등액을 수거하여 SDS-PAGE 분석' 및 웨스턴 블롯팅을 수행하였다. 인산화된 JNK 및 전체 JNK 양 측정은 상기 <실시예 5〉와 동일한 방법으로 웨스턴 블롯팅을 수행하였다.
그 결과, 도 10에 나타낸 바와 같이, 아세트아미노펜 처리에 의한 간조직의 조직학적 분석에서 TNP를 함께 처라한 생쥐의 간에서 괴사반움이 매우 경미한 병리학적 표현형을 보였다 (도 10) . 또한, 도 11에 나타낸 바와 같이 간독성의 혈청지표인 AST, ALT수준이 TNP를 함께 처리한 생쥐에서 현저히 감소된 것을 확인하였으며 (도 11), 도 12에 나타낸 바와 같이 , 아세트아미노펜 처리에 의한 스트레스 반웅의 증가는 인산화 JNK 신호의 증가를 유발하였으나, TNP 공동처리에 의하여 인산화 JNK 증가가 유의하게 억제됨을 확인하였다 (도 12) .
이로써, 동물 모델을 활용한 간조직 분석에서도 아세트아미노펜에 의해 유도되는 간독성에 대하여 TNP약물에 의한 간독성 보호효과를 나타냄을 확인하였다.
