Description

ウェルプレート及びこれを用いた対象物の操作方法
 本発明は、固形物や液体からなる対象物を保持するウェルプレート、及びこれを用いた対象物の操作方法に関する。
 細胞乃至は細胞凝集塊等の対象物の観察、選別、検査若しくは培養等のために、ウェルプレートが用いられることがある。一般にウェルプレートは、平板状のプレート本体と、このプレート本体に独立的に設けられた複数のウェルとからなる。通常、細胞の吸引及び吐出が可能なチップにより、各ウェルに細胞及びその培養液が投入される。さらに、細胞を収容したウェルに対し、反応試験や検査等の目的で、前記チップによって試薬が追加投入される場合がある。
 細胞の収容目的のウェルは、その開口サイズが小さい。このため、チップにて当該ウェルに対象物を投入する際に前記チップ先端の位置合わせが困難となったり、ウェルの開口縁部に前記チップ先端が衝突したりする場合がある。この問題の解決のため、特許文献1には、ウェルの内径を底面側から開口部側に向けて拡径してなるウェルを備えたウェルプレートが開示されている。
 特許文献1のウェルプレートによれば、ウェルに対するチップのアプローチ性は良好となる。しかし、対象物によっては、ウェルに収容された後に当該対象物が可及的に位置ズレや型崩れを起こさないことが求められる。また、試薬は非常に高価なものもあるため、その使用量をなるべく少量とすることができる工夫が求められる。これらの点を十分に満たすウェルプレートは未だ提案されていない。

 本発明の目的は、利便性が向上されたウェルプレート、及びこれを用いた対象物の操作方法を提供することにある。
 本発明の一局面に係るウェルプレートは、固形物及び液体の対象物の吸引及び吐出を行うチップによって、前記対象物の投入及び取り出しが行われるウェルプレートであって、第1面と、該第1面と対向する第2面とを備え、前記第1面を上方に向けて用いられるプレート本体と、前記第1面に設けられ前記チップの進入を許容する開口部と、前記プレート本体の内部に位置する底部と、前記開口部から前記底部に向けて下方に延びる筒状の壁面とを含み、前記対象物を保持可能な1又は複数のウェルと、を備え、前記筒状の壁面は、前記底部から上方の所定領域であって、前記固形物の対象物を収容可能な第1の水平断面サイズを有する収容部と、前記収容部の上方から前記開口部に至る領域であって、前記第1の水平断面サイズよりも大きい第2の水平断面サイズを有する受入部と、を含む。
 本発明の他の局面に係る対象物の操作方法は、先端開口を備え、該先端開口から固形物及び/又は液体の対象物を吸引して保持し、且つ貯留した前記対象物を前記先端開口から吐出可能なチップと、請求項5又は6のウェルプレートとを準備するステップと、前記固形物を保持している前記チップの前記先端開口を、前記ウェルの前記収容部の直上に位置させるステップと、前記チップの前記先端開口から前記固形物を吐出させるステップと、前記液体を保持している前記チップの前記先端開口を、前記ウェルの前記受け部の直上に位置させるステップと、前記チップの前記先端開口から前記液体を吐出させるステップと、を含む。

 以下、本発明の実施形態を、図面に基づいて詳細に説明する。図1は、本発明の第1実施形態に係るウェルプレート1の上面図、図2は、図1のII-II線断面図である。ウェルプレート1は、固形物、液体若しくはこれらの混合物からなる対象物を担持することが可能なプレートである。当該対象物の操作には、先端開口を備え、該先端開口を通して前記対象物を吸引しこれを保持すること、並びに、保持している前記対象物を前記先端開口から吐出することが可能なチップ6(図3他参照)が用いられる。このような吸引及び吐出を行うチップ6によって、ウェルプレート1に対し前記対象物が投入され、又は投入された前記対象物がウェルプレート1から取り出される。
 本発明において、対象物について特に制限はなく、各種の固形物、液体をウェルプレート1の担持対象とすることができる。本実施形態では、固形の対象物として生体由来の細胞、特に細胞凝集塊を例示する。また液体の対象物として、細胞凝集塊の培地(細胞培養液)、及び細胞凝集塊に対する反応試験や検査等の目的で使用される試薬液を例示する。この場合、ウェルプレート1は、細胞乃至は細胞凝集塊の観察、選別、検査若しくは培養等のために用いられる。この他、対象物は、小型の電子部品や機械部品、有機又は無機の破砕片や粒子、ペレット等の固形物、水、油、生体液、溶媒、試薬、各種の溶液等の液体であっても良い。
 ウェルプレート1は、プレート本体2と、該プレート本体2に形成される複数のウェル3とを備えている。図1に例示する通り、ここでは4個×6個=24個のウェル3がマトリクス状に配列されているウェルプレート1を示している。ウェル3は、少なくとも1個、プレート本体2に備えられていれば良い。複数個のウェル3をプレート本体2に具備させる場合、その配列態様は、均等配列又は不均等配列のマトリクス状、ハニカム状、或いはランダム配列とすることができる。
 プレート本体2は、所定の厚みを有する平板状の部材からなり、上面21(第1面)と下面22(第1面と対向する第2面)とを備える。ウェルプレート1は、通常使用時には、上面21を上方に向けて用いられる。プレート本体2の材質については特に制限は無く、孔開け加工や金型成型でウェル3を形成可能な材料であって、担持する対象物に化学的、生態的に影響を与えない材料であれば良い。例えば、樹脂成型品からなるプレート本体2は、好適な例の一つである。とりわけ、透明樹脂で形成されたプレート本体2は、ウェル3に保持された細胞凝集塊を下面22側から観察(撮像)することが可能となるので好ましい。また、熱伝導性に優れた樹脂で形成されたプレート本体2は、ウェル3に保持された細胞凝集塊に対する熱環境のコントロールを容易とする観点からは好ましい。
 ウェル3は、プレート本体2に設けられた有底の孔であり、固形物及び/又は液体の対象物を保持する。ウェル3は、開口部31、底部32及び筒状の壁面33を含む。開口部31は、円形の開口であり、上面21に設けられ前記チップの進入を、余裕を持って許容する内径を有する。底部32は、プレート本体2の内部であって、下面22の近くに位置している。筒状の壁面33は、開口部31から底部32に向けて鉛直下方に延びる壁面である。筒状の壁面33は、底部32から上方にウェル3の上下全長の半分程度の領域(所定領域)を占める収容部34と、収容部34の上方から開口部31に至る領域を占める受入部35とを含む。収容部34は、固形物の対象物を収容可能な第1の水平断面サイズを有する。受入部35は、前記第1の水平断面サイズよりも大きい第2の水平断面サイズを有する。ウェル3の形状については、図5に基づき、後記で詳述する。
 図3及び図4は、ウェルプレート1の使用例を示す図である。ここでは、細胞凝集塊Cの試験システムを例示している。前記試験システムは、本実施形態のウェルプレート1、細胞凝集塊Cの選別容器4、試薬液の容器5及びチップ6を備えたヘッドユニット61を備えている。
 選別容器4は、細胞培養液41を貯留する容器であり、細胞選別用の選別プレート42を細胞培養液41に浸漬される状態で保持している。選別プレート42は、細胞凝集塊Cを個別に担持可能な保持凹部を複数備える。保持凹部の底面には、所定サイズの細胞凝集塊Cは通過できないが、微小な夾雑物等は通過可能な逃がし孔が設けられている。多数の細胞凝集塊Cを含む細胞懸濁液が、選別プレート42の上方から投入される。検査対象となる、所定サイズを有する細胞凝集塊Cは、前記保持凹部にトラップされる一方で、夾雑物等は前記逃がし孔から選別容器4の底面に落下する。これにより、選別プレート42の上面に検査対象の細胞凝集塊Cが担持された状態が形成される。図3は、この状態を示している。なお、ウェルプレート1の底面に、下面22へ至る小径の貫通孔を設ければ、これを選別プレート42として用いることもできる。
 試薬容器5は、試薬液51を貯留する容器である。試薬液51としては、例えば、蛍光試薬や染色試薬を例示することができる。これら試薬は、培地では非蛍光性を示す細胞凝集塊Cに蛍光性を具備させる、または死滅した細胞凝集塊Cを選択的に染色することによって、当該細胞凝集塊Cの観察を容易にするための試薬である。このような試薬としては、Life Technologies社の「LIVE/DEAD Cell Imaging Kit(488/570)」、Roche社 の「Cell Proliferation Kit I(MIT asay)」、Photochemical internalization社の「Live/Dead Cell Straining Kit II」などを例示することができる。この他、試薬液51として、Promega社の「CellTiter-Glo Assay」のような細胞増殖/毒性試験用試薬、ソニーバイオテクノロジー社の「Alexa Flour」や「Brilliant Violet」のような抗体蛍光試薬等を用いることができる。
 チップ6は、先端開口を備えたチューブ状の部材であり、細胞凝集塊C、細胞培養液41及び試薬液51の吸引及び吐出を行う。具体的にはチップ6、選別容器4から、細胞凝集塊Cを細胞培養液41と共に吸引し、これらをウェルプレート1のウェル3へ吐出する。また、チップ6は、試薬容器5から試薬液51を吸引し、これを細胞凝集塊Cの担持したウェル3内へ吐出する。チップ6は、ヘッドユニット61に備えられたロッド6Rの下端に装着されている。ロッド6Rにはピストン機構が搭載されており、該ピストン機構の動作によってチップ6の先端開口に吸引力及び吐出力が与えられる。また、ロッド6Rは上下方向に移動可能である。ヘッドユニット61は、前記ピストン機構の動力部等を備え、ガイドレール62に沿って水平方向に移動可能である。ヘッドユニット61の移動範囲内に、選別容器4、ウェルプレート1及び試薬容器5が順に配列されている。ヘッドユニット61の移動機構、前記ピストン機構の動力部及びロッド6Rの上下動機構等は、図略の制御部によって制御される。
 図3は、細胞凝集塊Cが選別容器4からウェルプレート1へ搬送される工程を示している。先ず、ヘッドユニット61が選別容器4の上空へ移動される。チップ6が選別プレート42の1つの保持凹部に位置決めされた後、ロッド6Rが下降され、チップ6の先端開口が前記保持凹部に担持されている細胞凝集塊Cに接近する。続いて、前記ピストン機構が作動し、周囲の細胞培養液41と共に細胞凝集塊Cがチップ6内へ吸引される。そして、ロッド6Rが元の高さ位置まで上昇される。図3において、実線で示すヘッドユニット61は、前記上昇を終えた状態を示している。
 しかる後、ヘッドユニット61がガイドレール62に沿って矢印A1方向に移動され、図3において点線で示すように、ウェルプレート1の上空で停止される。この際、細胞凝集塊Cを保持しているチップ6と、その細胞凝集塊Cの投入ターゲットとなる1のウェル3とが位置合わせされる。続いて、ロッド6Rが下降され、チップ6の先端開口がウェル3に入り込む。そして、細胞凝集塊Cを含む細胞培養液41が、チップ6の先端開口からウェル3内へ吐出される。
 図4は、図3の工程の後に、試薬液51が試薬容器5からウェルプレート1へ搬送される工程を示している。当該工程は、例えば、ウェル3内で所定期間だけ細胞凝集塊Cを培養させた後に実行される。この工程では、ヘッドユニット61が試薬容器5の上空へ移動される。ロッド6Rが下降され、チップ6の先端開口が試薬液51内へ浸漬される。続いて、前記ピストン機構が作動し、所定量の試薬液51がチップ6内に吸引される。そして、ロッド6Rが元の位置まで上昇される。図4において、実線で示すヘッドユニット61は、試薬液51の吸引後の上昇を終えた状態を示している。
 しかる後、ヘッドユニット61がガイドレール62に沿って矢印A2方向に移動され、図4において点線で示すように、ウェルプレート1の上空で停止される。この際、試薬液51を保持しているチップ6と、その試薬液51の投入ターゲットとなる、細胞凝集塊Cを担持した1のウェル3とが位置合わせされる。続いて、ロッド6Rが下降され、チップ6の先端開口がウェル3に入り込む。そして、試薬液51が、チップ6の先端開口からウェル3内へ吐出される。
 続いて、第1実施形態のウェルプレート1が備えるウェル3の詳細構造について、図5を参照して説明する。図5は、ウェルプレート1の拡大断面図である。上述の通り、ウェル3は、開口部31、底部32及び筒状の壁面33を含み、筒状の壁面33は、収容部34と受入部35とを含んでいる。模式的な図示である図2の垂直断面図においては、底部32は平坦な底面として描かれているが、詳細を示す図5の垂直断面図では底部32は凹曲面として描かれている。
 開口部31及び底部32は、図1に示している通り、上面視において円の形状を備え、両者は同心円の位置関係にある。開口部31は、筒状の壁面33(受入部35)の上端に連なり、底部32は、筒状の壁面33(収容部34)の下端に連なっている。開口部31の内径は、チップ6の先端を受容するのに十分な径に設定されている。例えば、チップ6の先端開口6T(図6)の部分の外径が300μm程度である場合、開口部31の内径は、360~600μm程度に設定することができる。筒状の壁面33は、いずれの高さにおいても水平断面の形状が円である。このようなウェル3であれば、デッドスペースを減らすことができ、またウェル3の内容積を小さくすることができる。
 収容部34は、全高さに亘り所定の第1の内径を有する壁面であり、円柱状の空間を区画している。つまり、収容部34の水平断面サイズは、上下方向において実質的に変化していない。前記第1の内径は、収容対象とする細胞凝集塊C(固形物の対象物)のサイズに応じて適宜設定される。例えば、ターゲットとする細胞凝集塊Cの大きさが40μm~400μm程度である場合、前記第1の内径は、120~1000μm程度に設定することが好ましい。これにより、水平断面サイズが一定である収容部34に入り込んだ細胞凝集塊Cを水平方向に揺動させ難くし、位置ズレ等を起こすことなく良好に保持することができる。このことは、ウェルプレート1に担持された細胞凝集塊Cの撮像、観察等において有利となる。
 受入部35は、大略的には、前記第1の内径よりも大きい第2の内径を有する壁面である。本実施形態では、受入部35の水平断面サイズが上下方向において連続的に変化するテーパ形状の壁面を例示している。受入部35の内径(水平断面サイズ)は、開口部31に連なる上端部分の内径が最大であり、収容部34との境界部となる下端部分の内径が最小であって、これらの間において内径が連続的に変化している。つまり、受入部35は、開口部31付近に位置する最大サイズ部分と、この最大サイズ部分から下方に向かうに連れてサイズが連続的に小さくなるように変化する変化部分とを含んでいる。このような受入部35によって、円錐台状の空間が区画されている。
 このような形状特徴を有するウェル3によれば、チップ6がウェル3に進入する際のアプローチ性を良好に維持しつつ、当該ウェル3の内容積を少なくすることができる。図5において、点線で示す矩形形状Rは、開口部31の内径と同一径で底部32の位置までウェルが延伸しているとした場合の、当該ウェルの垂直断面形状である。この矩形形状Rに比べて、本実施形態のウェル3は、水平断面サイズが抑制されている。一般に、矩形形状Rの如き形状が、ウェルプレートを生産する際に用いられる金型の形状をシンプルにすることができる形状と言える。しかし、本実施形態のウェル3は、あえて水平断面サイズが細胞凝集塊Cのサイズに応じて抑制された収容部34と、最大サイズ部分が開口部31である円錐台形状の受入部35とによって、筒状の壁面33を構成している。これにより、ウェル3の内容積が抑制され、試薬液51をウェル3へ投入する必要がある場合において、当該試薬液51の使用量を節約することが可能となり、ひいては試験や検査のコストダウンに貢献する。
 ウェル3の内容積が抑制されていても、開口部31付近が最大サイズを有しているので、チップ6のウェル3へのアプローチ性は損なわれない。つまり、開口部31は矩形形状Rの場合と同サイズであり、チップ6が少々芯ズレしてウェル3へ進入しても、これを受け入れることができる。また、受入部35は、下方に向かうに連れて径小となるテーパ形状を有するので、細胞凝集塊C、細胞培養液或いは試薬液などの対象物が受入部35のどの部位に投入された場合でも、重力を利用して当該対象物を下方の収容部34へスムースに導くことができる。そして、上述の通り収容部34に入り込んだ細胞凝集塊Cは良好に保持されるものである。
 図6は、ウェル3に、チップ6から細胞凝集塊Cが投入される状態を示す垂直断面図である。図3に基づき先述した通り、細胞凝集塊Cがウェル3に投入される際、細胞凝集塊C及び細胞培養液41を保持したチップ6が、上方からウェル3の開口部31に向けて下降される。そして、チップ6の先端開口6Tが、受入部35に所定深さだけ入り込んだ位置で、チップ6の前記下降が停止される。その後、先端開口6Tから細胞凝集塊C及び細胞培養液41が吐出される。図6の左方のウェル3は、前記吐出が行われる直前の状態を示している。なお、前記吐出の前に、予め所定量の細胞培養液をウェル3に注液しておいても良い。一方、図6の中央及び右方のウェル3は、前記吐出が完了した状態を示している。これらのウェル3においては、細胞培養液41は、ウェル3の収容部34の全部と受入部35の下半分程度とを満たし、細胞凝集塊Cは、収容部34で側周囲が拘束される態様で、底部32付近に接面又は浮遊している。細胞培養液41の液量は、適宜調整される。予め細胞培養液をウェル3に注液する実施形態では、細胞凝集塊Cの吸引時にチップに保持される細胞培養液の液量分を考慮して、ウェル3に予め保持させる細胞培養液の液量が決定される。
 なお、先端開口6Tが受入部35に進入しない位置で、チップ6の下降が停止されても良い。すなわち、開口部31はチップ6の先端開口6Tの進入を許容するサイズの開口を有していれば良く、対象物の投入時に、実際に先端開口6Tが開口部31を越えてウェル3内に進入しなくても良い。例えば、開口部31の位置よりも数mm程度上方に先端開口6Tを停止させ、その位置の先端開口6Tから細胞凝集塊C及び細胞培養液41を吐出させても良い。この場合、先端開口6Tが、底部32の直上でなくとも、開口部31の上空に位置していれば、吐出された細胞凝集塊C及び細胞培養液41は、受入部35のテーパ状の壁面に案内されて収容部34へ向かうことができる。
 図7(A)及び図7(B)は、第1実施形態の変形例に係るウェルプレート1A及び1Bを各々示す垂直断面図である。図7(A)に示すウェルプレート1Aが備えるウェル3Aは、開口部31A、底部32A及び筒状の壁面33Aを含み、筒状の壁面33Aは、収容部34Aと受入部35Aとを含んでいる。先に例示したウェル3と相違する点は、受入部35Aの形状である。受入部35Aは、水平断面サイズが上下方向において一定であり、開口部31Aと同じ大きさの内径(第2の内径)を全長に亘って有している。収容部34Aは、細胞凝集塊Cの大きさに応じた所定サイズの内径(第1の内径)を有し、該内径よりも受入部35Aの内径は大きく設定されている。なお、さらなる変形例として、受入部35Aの内径が、下方に向かうに連れて段階的に小さくなるように変化させる態様としても良い。
 図7(B)に示すウェルプレート1Bが備えるウェル3Bは、開口部31B、底部32B及び筒状の壁面33Bを含み、筒状の壁面33Bは、収容部34Bと受入部35Bとを含んでいる。受入部35Bは、水平断面サイズが上下方向において一定な上段部分351Bと、水平断面サイズが下方に向かうに連れて小さくなるよう変化する下段部分352Bとからなっている。収容部34Bは、細胞凝集塊Cの大きさに応じた所定サイズの内径を有する、水平断面サイズが上下方向において一定な部分である。これらのウェルプレート1A及び1Bによっても、先に説明したウェルプレート1と同様な利点を享受することができる。
 図8は、本発明の第2実施形態に係るウェルプレート1Cを示す断面図である。第2実施形態では、受け部36を有するウェル3Cを具備するウェルプレート1Cを例示する。図9は、ウェル3Cの上面視の図である。ウェル3Cは、プレート本体2の上面21に開口した開口部31Cと、プレート本体2の内部であって下面22の近くに配置された底部32Cと、開口部31Cと底部32Cとの間に上下方向に延びる筒状の壁面33Cとを含む。筒状の壁面33Cは、収容部34C及び受入部35Cに加え、受け部36を備えている。受け部36は、専らチップ6の先端開口6Tから投入される対象物を受け止めるために設けられている。
 収容部34Cは、水平断面サイズが上下方向において一定な部分であり、底部32Cから上方にウェル3Cの上下全長の半分程度の領域を占めている。受入部35Cは、水平断面サイズが下方に向かうに連れて小さくなるテーパ形状の部分であり、収容部34Cの上方から開口部31Cに至る領域を占めている。収容部34C及び受入部35Cは、第1実施形態で例示した収容部34及び受入部35と基本的に同じ機能を果たす部分である。但し、図9に示す上面視の通り、受け部36を設けるために、底部32Cは開口部31Cの円中心に対して偏心して配置されている。
 受け部36は、受入部35Cを形成する円錐台状の壁面に形成されている。図8の垂直断面において詳しく言うと、受け部36は、テーパ形状を備える受入部35Cの壁面の、上下方向における中間付近の勾配を、他の部分の勾配に比べて水平に近付けた部分からなる。図9の上面視では、受け部36は、底部32Cに向けて凹の、三日月型の形状を有している。この三日月形状の最大幅の部分は、チップ6から吐出される液体の対象物を十分に受け止めることができるよう、概ねチップ6の先端開口6Tの内径と同等か、やや大きい程度のサイズを有していることが望ましい。
 このウェルプレート1Cによれば、受け部36に向けて、チップ6の先端開口6Tから対象物を投入させることができる。例えば、収容部34Cに既に細胞凝集塊Cのような固形物の対象物が収容されており、かかる状態で細胞培養液41や試薬液51のような液体の対象物をウェル3Cに投入する場合に、一旦液体対象物を受け部36に衝突させてから、収容部34Cへ向かわせることができる。これにより、液体対象物の投入によって生じる液体フローが細胞凝集塊Cに直接的に当たらないようにすることができる。これにより、細胞凝集塊Cのような易破壊性の固形物が収容部34Cに収容されている場合であっても、当該固形物の崩壊や型崩れを防止することができる。さらには、固形物の位置ズレも防止することができ、このことは固形物の画像を所定の時間間隔をおいて撮像する場合に、これらの画像の比較を的確に行える点で有利である。しかも、受け部36は緩いながらも下り勾配を有しているので、受け部36にて受け止めた対象物を、滞留させることなくスムースに収容部34Cへ導くことができる。
 図10は、第2実施形態のウェル3Cに対し、チップ6から液体対象物が投入される状態を示す垂直断面図である。ここでは、ウェル3Cに細胞培養液41及び細胞凝集塊Cが既に投入されている状態を示す。細胞凝集塊Cは、底部32C付近において収容部34Cに収容され、細胞培養液41は、受入部35Cの内容積の80%程度を占める程度に充填されている。受け部36は、細胞培養液41で覆われている状態である。チップ6の内部には、所定量の試薬液51が保持され、該試薬液51が細胞凝集塊Cの試験のためにウェル3C内に投入されようとしている。
 先端開口6Tがウェル3C内の細胞培養液41に浸漬されるまで、チップ6は下降されている。先端開口6Tは、受け部36と対向している。この状態において、先端開口6Tから試薬液51が吐出されると、矢印Bで示す通り、その吐出により生じる液流は受け部36に衝突する。その後、試薬液51はウェル3C内の細胞培養液41に混ざり合うことになる。従って、収容部34C内の細胞培養液41には大きな液流は生じることは無く、細胞凝集塊Cは液流の力の影響を殆ど受けない。よって、細胞凝集塊Cが崩壊したり型崩れしたりする不具合は、あまり発生しない。これに対し、細胞凝集塊Cの直上から試薬液51を吐出させた場合、その吐出により生じる液流の力によって細胞凝集塊Cが舞い上がって変形したり、液流の力によって細胞凝集塊Cが分離乃至は崩壊したりする頻度が多くなる不具合が生じる。さらに、液流の力によって細胞凝集塊Cが移動又は姿勢変更すると、細胞凝集塊Cの画像を定点でモニターしている場合に、モニター位置が変化してしまうことになる。
 図11(A)及び図11(B)は、第2実施形態の変形例に係るウェルプレート1D及び1Eを各々示す垂直断面図である。図11(A)に示すウェルプレート1Dが備えるウェル3Dは、開口部31D、底部32D及び筒状の壁面33Dを含み、筒状の壁面33Dは、収容部34D、受入部35D及び受け部36Dを含んでいる。受入部35Dは、水平断面サイズが上下方向において一定であり、開口部31Dと同じ大きさの内径を全長に亘って有している。収容部34Dも、水平断面サイズが上下方向において一定で、受入部35Dよりも小さい内径を有する領域である。受け部36Dは、収容部34Dと受入部35Dとの内径差に基づき、両者の境界部分に生じる段差部分からなる。受け部36Dは略水平な面からなる。図中に矢印B1で示すように、この受け部36Dに向けて試薬液51が吐出される。
 図11(B)に示すウェルプレート1Eが備えるウェル3Eは、開口部31E、底部32E及び筒状の壁面33Eを含み、筒状の壁面33Eは、収容部34E、受入部35E及び受け部36Eを含んでいる。受入部35Eの上方部分は、水平断面サイズが上下方向において一定であるが、下方部分は、垂直断面視において垂直方向に対して傾きを有する部分からなる受け部36Eが形成されることによって、水平断面サイズが下方に向かうに連れて小さくなっている。収容部34Eは、水平断面サイズが上下方向において一定な部分である。図中に矢印B2で示すように、この受け部36Eに向けて試薬液51が吐出される。これらのウェルプレート1D及び1Eによっても、先に説明した第2実施形態に係るウェルプレート1Cと同様な利点を享受することができる。
 続いて、第2実施形態に係るウェルプレート1Cの好ましい使用例を、図12及び図13に基づき説明する。ここでは、細胞凝集塊C及び細胞培養液41のウェル3Cへの投入操作と、その後のウェル3Cへの試薬液51の投入操作とからなる一連の操作方法(対象物の操作方法)のステップを例示する。
 最初のステップは、受け部36を有するウェル3Cを具備したウェルプレート1Cと、細胞凝集塊C、細胞培養液41及び試薬液51の吸引及び吐出が可能なチップ6とを準備するステップである。図12(A)は、ウェルプレート1Cが備える1つのウェル3Cの垂直断面を示している。
 次のステップは、細胞凝集塊C及び細胞培養液41を保持するチップ6を前記1のウェル3Cの上空において位置決めするステップである。この際、図12(B)に示すように、チップ6が搭載されているヘッドユニット61を移動させ、収容部34Cの筒心に沿った軸Z1上に、チップ6を位置決めさせる。つまり、細胞凝集塊C及び細胞培養液41を保持するチップ6の先端開口6Tを、収容部34Cの直上に位置させる。チップ6は、このステップの前に、先に図3に基づき説明した、選別容器4から細胞凝集塊Cを吸引するステップを実行している。
 続くステップでは、図12(C)に示すように、チップ6が取り付けられているロッド6Rが所定長だけ下降される。これにより、チップ6の先端開口6Tは、ウェル3Cの受入部35Cに進入する。そして、図12(D)に示す通り、先端開口6Tから細胞凝集塊C及び細胞培養液41が吐出される。その後、チップ6は上昇される。この吐出動作によってウェル3Cは概ね細胞培養液41で満たされ、沈降時間が経過すると細胞凝集塊Cは底部32C付近に接面する。収容部34Cの壁面によって取り囲まれているので、少々の振動がウェルプレート1Cに与えられても、細胞凝集塊Cは水平方向には揺動しない。
 次のステップでは、図13(A)に示すように、試薬液51を保持するチップ6を、試験対象となる細胞凝集塊Cが投入されているウェル3Cの上空において位置決めするステップである。チップ6は、このステップの前に、先に図4に基づき説明した、試薬容器5から試薬液51を吸引するステップを実行している。この位置決めの際、ヘッドユニット61を移動させ、受け部36の中心位置を上下に貫く軸Z2上に、チップ6を位置決めさせる。つまり、試薬液51を保持しているチップ6の先端開口6Tを、受け部36の直上に位置させる。
 続くステップでは、図13(B)に示すように、チップ6が取り付けられているロッド6Rが所定長だけ下降される。これにより、チップ6の先端開口6Tは、受け部36と対向する位置においてウェル3Cの受入部35Cに進入する。また、先端開口6Tは、細胞培養液41内に浸漬される。そして、図中に矢印を付している通り、先端開口6Tから試薬51が吐出される。この後、先端開口6Tの吸引力及び吐出力を交互に発生させて、試薬51と細胞培養液41とを混合させることが好ましい。より好ましくは、上記の吸引力及び吐出力の発生と、チップ6の上下動とを連動させ、前記混合を促進する。具体的には、先端開口6Tに吸引力を発生させつつチップ6を下降させ、続いて先端開口6Tに吐出力を発生させつつチップ6を上昇させるという動作を、1回又は複数回行う。その後、図13(C)に示す通り、チップ6は上昇される。以上のような操作方法によれば、細胞凝集塊Cの如き易破壊性の固形物が先に収容部34Cに収容されている場合において試薬液51を追加投入する場合であっても、細胞凝集塊Cが試薬液51の液流によって崩壊や型崩れすることを防止できる。
 上記の利点は、細胞培養液41をウェル3Cから吸引させる場合においても享受することができる。図12(D)に示す通り、ウェル3Cに細胞凝集塊C及び細胞培養液41を投入した後、若しくは、さらに図13(C)に示す通り、試薬液51を追加投入した後、細胞培養液41をウェル3Cから吸引させるステップが必要となる場合がある。このステップの実行には、受け部36が浸漬されるまで細胞培養液41がウェル3Cに注液されていることを前提として、図13(D)に示すように、対象物を保持しない空の状態のチップ6の先端開口6Tを、受け部36の直上であって細胞培養液41に浸漬される位置に配置する。
 しかる後、チップ6に吸引力が発生され、図13(D)中に矢印で示す通り、先端開口6Tから細胞培養液41が吸引される。これにより、先端開口6Tが受け部36の直上に配置された状態で細胞培養液41の吸引が行われる。このため、吸引の際に発生する液流の影響が、収容部34Cに収容されている細胞凝集塊Cに及びにくい。従って、細胞凝集塊Cがチップ6によって吸引されてしまわないようにすることができる。
 ウェルのさらなる変形例を説明する。図14(A)は、変形例に係るウェルを示す上面図、図14(B)は図14(A)のG1線の断面図である。ここでは、プレート本体2に、第1ウェル3Fと第2ウェル3Gとがペアで具備される例を示す。このようなウェルのペアが、1組又は複数組、プレート本体2に具備される。
 第1ウェル3Fは、第1開口部31F、第1底部32F及び筒状の壁面33Fを含み、筒状の壁面33Fは、第1収容部34Fと第1受入部35Fとを含んでいる。第1収容部34Fは、水平断面サイズが上下方向において一定である。第1受入部35Fは、水平断面サイズが下方に向かうに連れて小さくなるよう変化している。第1受入部35Fの上面視の形状は半円形であり、垂直断念視の形状は、1辺が第1収容部34Fの壁面と面一の非等脚台形である。第1収容部34Fは、半円形の第1受入部35Fの中心点を通る二等分線(G1線に相当)上であって、外周円に内接する位置に配置されている。同様に第2ウェル3Gも、第2開口部31G、第2底部32G及び筒状の壁面33Gを含み、筒状の壁面33Gは、第2収容部34Gと第2受入部35Gとを含んでいる。
 第1ウェル3Fの第1収容部34Fと、第2ウェル3Gの第2収容部34Gとは、互いに所定距離だけ離間して配置されている。いずれも上面視で半円形の第1受入部35Fと第2受入部35Gとは、それぞれの割線同士が稜線部分37を挟んで隣接するように配置されている。つまり、第1受入部35Fと第2受入部35Gとによって、1つの略円形の受け部が形成されている。もちろん、この受け部の形状は、四角形などの矩形のものとしても良い。
 図14(B)において、第1ウェル3Fには細胞凝集塊C及び細胞培養液41が既に投入され、チップ6の先端開口6Tから試薬液51が矢印G2の通り吐出される状態を示している。第1受入部35Fの斜面は緩く(図8のウェル3Cの受け部36と同じ程度の勾配)、当該斜面に向けて試薬液を吐出させることで、液流の影響が第1収容部34F内の細胞凝集塊Cに及ばないようにすることができる。一方、第2ウェル3Gに対しては、チップ6から細胞凝集塊C及び細胞培養液41が、矢印G3の通り吐出される状態を示している。
 このようなウェル3F、3Gを備えるウェルプレートによれば、チップ6により対象物を第1ウェル3F及び第2ウェル3Gに個別に投入する場合に、稜線部分37を境としてチップ6をいずれかの側に移動させるだけで、第1収容部34F又は第2収容部34Gのいずれかに所望の対象物を投入することができる。従って、チップ6の移動制御の厳密性を緩和させることができる。
 以上説明した通り、本実施形態によれば、ウェル3に対するチップ6のアプローチ性が良好なだけではなく、ウェル3に収容された後に細胞凝集塊Cが位置ズレや型崩れを起こし難いウェルプレート1を提供することができる。また、試薬液のような高価な液体をウェル3に投入する必要がある場合でも、当該試薬液の使用量を少量に抑制することができ、試験や検査等のコストダウンに貢献することができる。
 なお、上述した具体的実施形態には以下の構成を有する発明が主に含まれている。
 本発明の一局面に係るウェルプレートは、固形物及び液体の対象物の吸引及び吐出を行うチップによって、前記対象物の投入及び取り出しが行われるウェルプレートであって、第1面と、該第1面と対向する第2面とを備え、前記第1面を上方に向けて用いられるプレート本体と、前記第1面に設けられ前記チップの進入を許容する開口部と、前記プレート本体の内部に位置する底部と、前記開口部から前記底部に向けて下方に延びる筒状の壁面とを含み、前記対象物を保持可能な1又は複数のウェルと、を備え、前記筒状の壁面は、前記底部から上方の所定領域であって、前記固形物の対象物を収容可能な第1の水平断面サイズを有する収容部と、前記収容部の上方から前記開口部に至る領域であって、前記第1の水平断面サイズよりも大きい第2の水平断面サイズを有する受入部とを含む。
 このウェルプレートによれば、受入部の第2の水平断面サイズは、収容部の第1の水平断面サイズよりも大きく設定されるので、ウェルに対するチップのアプローチ性を良好とすることができる。また、収容部の水平断面サイズを比較的小さく設定することで、収容された対象物の位置ズレが起こり難くすることができる。さらに、ウェルに液体を注液する必要がある場合、収容部のサイズが抑制されている分だけ前記液体の使用量を減らすことが可能となる。
 上記のウェルプレートにおいて、前記筒状の壁面は、水平断面の形状が円であり、前記収容部が第1の内径を有し、前記受入部が前記第1の内径よりも大きい第2の内径を有することが望ましい。このウェルプレートによれば、ウェルのデッドスペースを減らし、ウェルの容積を小さくすることができる。
 上記のウェルプレートにおいて、前記受入部の水平断面サイズが上下方向において変化するものであって、該受入部は、前記開口部付近に位置する最大サイズ部分と、下方に向かうに連れてサイズが連続的又は段階的に小さくなるように変化する変化部分とを含み、前記収容部の水平断面サイズは、上下方向において実質的に変化しないことが望ましい。
 このウェルプレートによれば、ウェルは、開口部付近が最大サイズを有しているので、チップのアプローチ性が良好であると共に、対象物が受入部のどの部位に投入された場合でも、重力を利用して当該対象物を下方の収容部へ導くことができる。また、収容部の水平断面サイズは一定であるので、該収容部に入り込んだ対象物を良好に保持することができる。
 この場合、前記受入部は、その水平断面サイズが、前記開口部の内径が最大であり、前記収容部と前記受入部との境界部の内径が最小であって、これらの間において内径が連続的に変化しているテーパ形状を有していることが望ましい。
 このウェルプレートによれば、前記テーパ形状に沿って、対象物を収容部に向けてよりスムースに誘導することができる。
 上記のウェルプレートにおいて、前記受入部を形成する前記筒状の壁面に形成され、前記チップの先端から投入される前記対象物を受け止める受け部を備えることが望ましい。
 このウェルプレートによれば、前記受け部に向けて、チップの先端から対象物を投入させることができる。例えば、収容部に既に固形物の対象物が収容されており、かかる状態で液体の対象物をウェルに投入する場合に、液体の投入により生じる液流が固形物に直接的に当たらないようにすることができる。これにより、易破壊性の固形物が収容部に収容されている場合であっても、当該固形物の崩壊や型崩れを防止することができる。
 上記のウェルプレートにおいて、前記受入部を形成する前記筒状の壁面に形成され、前記チップの先端から投入される前記対象物を受け止める受け部を備え、前記受け部は、前記テーパ形状を備える壁面の一部の勾配を、水平に近付けた部分からなる構成とすることができる。
 このウェルプレートによれば、受け部にて受け止めた対象物を、滞留させることなくスムースに収容部へ導くことができる。
 上記のウェルプレートにおいて、前記プレート本体に第1ウェルと第2ウェルとが具備される場合において、前記第1ウェルの第1収容部と、前記第2ウェルの第2収容部とは、互いに所定距離だけ離間して配置されている一方で、前記第1ウェルの第1受入部と、前記第2ウェルの第2受入部とは、両者の間に位置する稜線部分を挟んで隣接する部分を含むことが望ましい。
 このウェルプレートによれば、稜線部分を挟んで第1受入部と第2受入部とが並存している。従って、チップにより対象物を第1ウェル及び第2ウェルに個別に投入する場合に、前記稜線部分を境としてチップ6いずれかの側に移動させるだけで、第1収容部又は第2収容部のいずれかに所望の対象物を投入することができる。従って、チップの移動制御の厳密性を緩和させることができる。
 本発明の他の局面に係る対象物の操作方法は、先端開口を備え、該先端開口から固形物及び/又は液体の対象物を吸引して保持し、且つ貯留した前記対象物を前記先端開口から吐出可能なチップと、上記の受け部を備えるウェルプレートとを準備するステップと、前記固形物を保持している前記チップの前記先端開口を、前記ウェルの前記収容部の直上に位置させるステップと、前記チップの前記先端開口から前記固形物を吐出させるステップと、前記液体を保持している前記チップの前記先端開口を、前記ウェルの前記受け部の直上に位置させるステップと、前記チップの前記先端開口から前記液体を吐出させるステップと、を含む。
 この方法によれば、チップから吐出させた固形物を収容部に収容させた後、前記チップから液体をウェル内へ追加投入するに際し、当該液体は受け部に向けて吐出される。従って、チップから液体を投入する際に発生する液流が、収容部に収容された固形物に直接的に当たらない。これにより、易破壊性の固形物(例えば細胞凝集塊)が先に収容部に収容されている場合において液体(例えば試薬)を追加投入する場合であっても、当該固形物が液流によって崩壊や型崩れすることを防止できる。
 上記の対象物の操作方法において、前記液体を吐出させるステップによって、前記受け部が浸漬されるまで前記液体を前記ウェルに注液された場合において、前記対象物を保持しない状態の前記チップの前記先端開口を、前記ウェルの前記受け部の直上であって前記液体に浸漬される位置に配置するステップと、前記チップの前記先端開口から前記液体を吸引させるステップと、をさらに含むことが望ましい。
 この方法によれば、チップの先端開口が受け部の直上に配置された状態で液体の吸引が行われるので、収容部に収容された固形物がチップで吸引され難くすることができる。
 本発明によれば、ウェルに対するチップのアプローチ性が良好なだけではなく、ウェルに収容された後に当該対象物が位置ズレや型崩れを起こし難いウェルプレートを提供することができる。また、高価な液体を対象物としてウェルに投入する必要がある場合でも、当該液体の使用量を少量とすることができる。

Claims (9)

  1.  固形物及び液体の対象物の吸引及び吐出を行うチップによって、前記対象物の投入及び取り出しが行われるウェルプレートであって、
     第1面と、該第1面と対向する第2面とを備え、前記第1面を上方に向けて用いられるプレート本体と、
     前記第1面に設けられ前記チップの進入を許容する開口部と、前記プレート本体の内部に位置する底部と、前記開口部から前記底部に向けて下方に延びる筒状の壁面とを含み、前記対象物を保持可能な1又は複数のウェルと、を備え、
     前記筒状の壁面は、
      前記底部から上方の所定領域であって、前記固形物の対象物を収容可能な第1の水平断面サイズを有する収容部と、
      前記収容部の上方から前記開口部に至る領域であって、前記第1の水平断面サイズよりも大きい第2の水平断面サイズを有する受入部と、を含むウェルプレート。
  2.  請求項1に記載のウェルプレートにおいて、
     前記筒状の壁面は、水平断面の形状が円であり、
     前記収容部が第1の内径を有し、前記受入部が前記第1の内径よりも大きい第2の内径を有する、ウェルプレート。
  3.  請求項1又は2に記載のウェルプレートにおいて、
     前記受入部の水平断面サイズが上下方向において変化するものであって、該受入部は、前記開口部付近に位置する最大サイズ部分と、下方に向かうに連れてサイズが連続的又は段階的に小さくなるように変化する変化部分とを含み、
     前記収容部の水平断面サイズは、上下方向において実質的に変化しない、ウェルプレート。
  4.  請求項1に記載のウェルプレートにおいて、
     前記筒状の壁面は、水平断面の形状が円であり、
     前記収容部が第1の内径を有し、前記受入部が前記第1の内径よりも大きい第2の内径を有し、
     前記受入部は、その水平断面サイズが、前記開口部の内径が最大であり、前記収容部と前記受入部との境界部の内径が最小であって、これらの間において内径が連続的に変化しているテーパ形状を有し、
     前記収容部の水平断面サイズは、上下方向において実質的に変化しない、ウェルプレート。
  5.  請求項1~3のいずれか1項に記載のウェルプレートにおいて、
     前記受入部を形成する前記筒状の壁面に形成され、前記チップの先端から投入される前記対象物を受け止める受け部を備える、ウェルプレート。
  6.  請求項4に記載のウェルプレートにおいて、
     前記受入部を形成する前記筒状の壁面に形成され、前記チップの先端から投入される前記対象物を受け止める受け部を備え、
     前記受け部は、前記テーパ形状を備える壁面の一部の勾配を、水平に近付けた部分からなる、ウェルプレート。
  7.  請求項1~6のいずれか1項に記載のウェルプレートにおいて、
     前記プレート本体に第1ウェルと第2ウェルとが具備される場合において、
     前記第1ウェルの第1収容部と、前記第2ウェルの第2収容部とは、互いに所定距離だけ離間して配置されている一方で、
     前記第1ウェルの第1受入部と、前記第2ウェルの第2受入部とは、両者の間に位置する稜線部分を挟んで隣接する部分を含む、ウェルプレート。
  8.  先端開口を備え、該先端開口から固形物及び/又は液体の対象物を吸引して保持し、且つ貯留した前記対象物を前記先端開口から吐出可能なチップと、請求項5又は6のウェルプレートとを準備するステップと、
     前記固形物を保持している前記チップの前記先端開口を、前記ウェルの前記収容部の直上に位置させるステップと、
     前記チップの前記先端開口から前記固形物を吐出させるステップと、
     前記液体を保持している前記チップの前記先端開口を、前記ウェルの前記受け部の直上に位置させるステップと、
     前記チップの前記先端開口から前記液体を吐出させるステップと、
    を含む、対象物の操作方法。
  9.  請求項8に記載の対象物の操作方法において、
     前記液体を吐出させるステップによって、前記受け部が浸漬されるまで前記液体を前記ウェルに注液された場合において、
     前記対象物を保持しない状態の前記チップの前記先端開口を、前記ウェルの前記受け部の直上であって前記液体に浸漬される位置に配置するステップと、
     前記チップの前記先端開口から前記液体を吸引させるステップと、
    をさらに含む、対象物の操作方法。
