また、bCDはその内部にコレステロールを取り込む性質が広く知られている(非特許文献3)。一方、コレステロールは原形質膜において、外から入ってくる多くのシグナルを細胞内に伝達する重要なはたらきを有しており、また血中コレステロール値が高い患者群は癌になる率が低いという報告が1980年頃から盛んに行われたことから(非特許文献4)、コレステロールがターゲットとなるbCDの癌治療への試みはこれまでないか、あってもごくわずかと考えられた。

β-シクロデキストリンまたはその誘導体を含む抗腫瘍剤。
ここでβ-シクロデキストリンまたはその誘導体はPI3KとAKTとの間のシグナル伝達を阻害する阻害剤としてはたらく。
1つまたは2つ以上の他の抗腫瘍剤と併用されることを特徴とする、β-シクロデキストリンまたはその誘導体。
他の抗腫瘍剤が2-デオキシグルコースを含む項2のβ-シクロデキストリンまたはその誘導体。

また、上記項1~15において、β-シクロデキストリンを2-ヒドロキシプロピル-γ-シクロデキストリン(HPGCD)に置換した各態様も本発明に包含される。
一方、本発明の複数剤の併用療法においては、それぞれ単独薬剤では健康な細胞への影響が少ないと考えられる程度の作用であるが、複数剤が同じ細胞内で作用することでその作用が相乗的に飛躍する特徴を有する。
一方、bCDは血液脳関門を通過できないので、体内では脳以外の組織に存在することになる。したがって、両剤が同時に存在できる細胞はほぼ腫瘍細胞に限定することができ、bCDと2DGとの併用療法においては、健康な細胞への影響が少ない、腫瘍細胞への効果的なアポトーシス誘導作用が期待できる。
bCDの投与量は患者に対して大きな悪影響を及ぼさず、PI3KとAKTとの間のシグナル伝達の阻害を発揮できる治療上の有効量である限り特に制限はないが、1回の治療あたり2~5000mg、好ましくは2~100mgの用量で投与される。
更に小胞体とミトコンドリアの間で生じるシグナルを基にして2DG-bCDと併用することでアポトーシスを誘導できると想定できる薬剤、具体的にはHSP90の阻害剤(gamitrinibs、PU24FCl、PU-H58、PU-H71、shepherdinなど)、小胞体ストレス剤、thapsigarginとその誘導体(G-202)等が挙げられる。
2DGの投与量は患者に対して大きな悪影響を及ぼさない限り特に制限はないが、1回の治療あたり100~5000mg、好ましくは500~2000mgの用量で投与される。
その他のアポトーシス誘導作用を有する抗腫瘍剤については、経口または注射や点滴などの非経口で投与されるが、それぞれの薬剤で定められた投与経路に従って投与されるのが望ましい。
投与量についても、それぞれの薬剤で定められた用量に従って投与されるのが望ましく、またアポトーシス誘導作用を有する抗腫瘍剤自身の副作用を抑える観点から、適宜減量してもよい。
また2DGを投与の際は、グルコースと合わせて用いてもよく、その量は2DGと同量付近が好ましい。
β-シクロデキストリン(bCD)は、インビトロ試験ではメチル-βシクロデキストリン(MBCD)を用い、インビボ試験では(2-ヒドロキシプロピル)-βシクロデキストリン(HPBCD)を用いた。
抗p-PI3K抗体はSanta Cruz(sc-12929)から、オリゴクローナル抗PI3K抗体(6HCLC)はPierceから、抗PI3KクラスII抗体(D3Q5B)はCSTから、抗シトクロムc抗体はBD Pharmingen(Cat. 556433 for blots and Cat. 556432 for microscopy)から、その他すべての一次抗体はCell Signalingから購入した。
HRPと結合する二次抗体はGE Healthcareから購入し、Alexa Fluor結合二次抗体はLife Technologiesから入手した。
IGF1、EGF、インスリン、ヨウ化プロピジウムおよびβシクロデキストリンはWakoから購入した。
ABT-263はChemietekから購入した。
2-デオキシ-D-グルコース、MBCDおよびHPBCDはSigmaから購入した。
汎カスパーゼ阻害剤z-VADはPromegaから購入した。
エンプティベクターRCC4またはVHLをコードするベクターで安定にトランスフェクトされた腎細胞癌細胞株は、Harada Laboratory(Kyoto University Hospital, Dept. of Anesthesia)から提供を受け、細胞株UOK121およびVHL発現ベクターで安定にトランスフェクトされたUOK121+VHLは、Dr. Marston Linehan(Center for Cancer Research, Urologic Oncology Branch, NCI)から提供を受けた。
Panc-1膵臓癌細胞およびA431類表皮癌細胞は、10%血清で補充された高グルコースDMEM中で培養した。
Panc-1細胞はDr. Koji Yamada(Dept. of Bioscience and Biochemistry, Faculty of Agriculture, Kyusyu University, Japan)から提供を受け、A431細胞はDr. Masaya Imoto(Dept. of Bioscience and Informatics, Faculty of Science and Technology, Keio University, Japan)から提供を受けた。
これらの細胞およびHeLa細胞は10%FBSで補充された高グルコースDMEM(4.5g/ml)中培養した。
血清はGE Health, Cosmo Bisocience等いくつかの異なる起源から入手した。
レーンあたり20μgのタンパク質でウェスタンブロットを実行した。ウェスタンブロットで検出されるタンパク質のサイズに応じて、8、10、12.5、および15%ゲルで実行した。2つのタンパク質が同じようなサイズの場合は、それぞれにゲルを流し、それぞれをウェスタンブロット分析する。免疫沈澱は可溶化された細胞から200μgのタンパク質と、免疫沈澱抗体でプレコンジュゲートされたProtein-GセファロースまたはProtein-Aセファロース(Sigma P3296/P9242)を、免疫沈澱用バッファーに投入し、それを入れたチューブを終夜4℃でゆっくり回転させた。免疫沈殿バッファーは20mM Tris Ph7.5、1% Triton-X100、150mM NaCl、ホスファターゼ阻害剤混合物(Cell Signaling(#58709S))、および10%グリセロールを含む。チューブ内の内容物を翌朝遠心で隔離し、沈殿物を同じバッファーで2回洗浄した後、SDSバッファーに溶解し、15% SDS-PAGE、もしくは12.5% SDS-PAGEを用いてタンパク質を分離し、ウェスタンブロット法で供沈物を確認した。
アポトーシスが化学的に誘導された後、細胞をPBS中洗浄し、通常の培地中再インキュベートし、終夜インキュベートした。細胞死アッセイを、ヨウ化プロピジウム取り込みアッセイを用いて翌朝実施した。細胞をBD FACS Canto IIまたはFACS Calibur IIで分析した。結果を、FlowJoを用いて解析した。これらの実験は3回行い、エラーバーは標準偏差を示す。
生死細胞はトリパンブルー色素排除試験でカウントした。これらの実験は3回行い、エラーバーは標準偏差を示す。
カバーガラス上で成長、処置されたRCC4細胞を、最初3.7%ホルムアルデヒド/PBSで10分間固定化した。次いで、カバーガラスを-20℃冷凍庫中保存された100%メタノールに素早く曝した。カバーガラスを10%血清/PBSでインキュベートし、次いでマウス抗シトクロムc抗体で終夜染色した。翌朝、カバーガラスを洗浄し、10%血清/PBSで30分間遮断した。次いで、カバーガラスをAF488と結合した二次抗マウス抗体に30分間曝した。カバーガラスを再度洗浄し、ヨウ化プロピジウム(1μg/mL PBS中)に15分間曝し核内のDNAを染色し、次いで洗浄し、スライドガラスに取り付けた。観察には顕微鏡(Keyence BZ9000)で100倍の対物レンズを用いて行った。
20週齢NSGマウス(JAXTM Mice strain NOD.Cg-Prkdcscid l2rgtm1Wjl/SzJ obtained from Charles River, Japan)の左右いずれかの側腹部の皮下に、0.2mlの50%マトリゲル(Falcon 356234)に5×106のUOK121細胞を混ぜ、移植した。腫瘍を有するマウスを4つか5つの処置群に分け、第1の実験および第2の実験はそれぞれ1つの群に3匹のマウスを有した。マウスには、2mgの2DGおよび2mgのグルコース/0.2ml PBS、または2mg HPBCD/0.2ml PBSを経口投与した。ABT-263を最初経口投与した(2mg/kg ABT-263/10%エタノール、30%ポリエチレングリコール400(Wako)、および60% Cremphore EL(Sigma))。最初、2DG/グルコース混合物を投与し、次いで2時間半後HPBCDを投与し、30分後ABTを投与した。いくつかのマウスにはこれらの試薬をすべて投与し、他はその一部を投与し、または全く投与しなかった。第1週は、マウスを2度処置した。続く3週間、マウスを週3度処置した。腫瘍のサイズは電子ノギス(volume = (length×width2)/2)で週3~4回測定した。3匹のマウスの1群は各処置条件で処置され、翌日腫瘍のサイズを記録した。グラフのエラーバーは標準偏差を示す。UOK121異種移植されていないNSGマウスは、3つの薬物の組み合わせで処置され、体重が記録された。尾静脈からの血液を用いて、血液中の白血球、赤血球、血小板の数をHoriba Hematology Analyzer LC-152で測定し、血中グルコースレベルをMediSafe Mini(Terumo, Japan)で測定した。
EGFRおよびIGF1Rの両方を発現するHeLa細胞を用いて、EGF活性化およびIGF1活性化の低下に関してbCDの効果を評価した。なお、bCDとしてはMBCDを用いた。
(方法)
(実験A)HeLa細胞を、0、1.75、3.5、および7.0mMの濃度のbCDでそれぞれ2つずつ、30分間無血清培地中培養した。それぞれの濃度の一方の培地を、20ng/mlのIGF1で20分間刺激し、他方の培地を対照群とした。細胞を収集し、AKTのリン酸化をAKTの活性化の指標とし、ホスホセリンAKTおよびAKTについてウェスタンブロット法で分析した。
(実験Bおよび実験C)HeLa細胞を、10mMのbCDで30分間無血清培地中培養し、無処置の対照群(Untreated)の培地も調製した。これらの細胞を次いで、実験Bでは0、2.5、および5.0分間、100ng/mLのEGFで、実験Cでは0、5、および10分間、10ng/ml IGF1で刺激した。細胞を収集し、EGFR、IGF1R、ERK、PI3K、およびAKT、並びに各リン酸化体についてウェスタンブロット法で分析した。PI3Kの検出については、抗PI3KクラスII抗体を用いた。
(結果)
結果を図1のA~Cに示す。
実験Aにおいては、7mM bCDにより、IGF1がAKTへのシグナルを生成することを完全に遮断した(図1A)。
次に、実験Bおよび実験Cにおいては、bCDで処置されていないHeLa細胞は、どちらも明らかに活性化された。bCDで処置された細胞においては、EGFR、IGF1R、PI3Kのいずれも活性化したが、AKT活性化はかなり減少した(図1Bおよび図1Cの各下段のレーン)。
このように、bCDはPI3KからAKTへのシグナル伝達を妨害できると考えられる。
(考察)
・ほとんどのRTKは2つの異なるシグナル伝達カスケード:RTK-Ras-ERK増殖経路とRTK-PI3K-AKTプロサバイバル経路を活性化する。
・同時に、RTK-Ras-ERKシグナルは影響を受けないようである(図1Bおよび図1Cの2番目のボックス)。
・このように、PI3KとAKTの間のシグナル伝達を破壊し、これらのRTKで生成されるPI3K-AKTプロサバイバルシグナルを減少させ、一方Ras-ERK増殖シグナルを損なわなかった。
VHL欠損腎臓癌細胞(例えば、RCC4細胞)は、IGF1Rを発現するので2DG-ABTへの感受性が低い。ここでbCDと組み合わせた2DG-ABTがその効果を増加させるかどうか確認するにあたっては、2DGが多くの癌細胞株においてAKTリン酸化を刺激することも考慮する必要がある。そこで、bCDを用いた2DGの組み合わせ治療がRCC4細胞におけるAKTリン酸化を増加させるか減少させるか、まず試験した。
一般的に2DGの効果を認識するのに1~2時間掛かり、bCDは実施例1の結果より30分以内に作用するので、血清を含まない培地を用い、RCC4細胞は最初2DGで2時間処理し、最後の30分以内に細胞をbCDでも処理することとする。なお、bCDとしてはMBCDを用いた。
(方法A)
RCC4細胞を無血清培地で10mM 2DGを加えて、または加えずに2時間培養した。終了1時間前に、各培地の細胞の分画に10mM bCDを加えた。次いで0~30ng/ml IGF1を加え、培養を5分間続けた後、細胞を収集し、ウェスタンブロット法で分析した。
(結果A)
結果を図2Aに示す。2DGで前処置した群では、IGF1に対して感受性を示し、2DG処置された細胞におけるAKTのリン酸化は増加した。しかし、bCDとの組み合わせ処置群では、AKTリン酸化が完全に遮断された(図2A)。これらの結果はたとえ2DGがIGF1R活性を向上させても、IGF1RからのシグナルはbCDの存在下AKTに到達しなかったことを示す。
上記の実施例2では特定のRTKにおけるbCDの効果を試験するために、血清を含まない培地中細胞をインキュベートし、次いで特定の成長因子で刺激した。しかしながら、通常、血清は多重の成長因子、並びにこれらの細胞中発現する多重のRTKを活性化できるインスリンを含むので、bCD自身が血清による継続的に刺激を受ける中でPI3K-AKTシグナルを制御できるかどうか確認するため、RCC4細胞をbCDで、2DGで、およびその組み合わせで処置し、AKTのリン酸化状態を試験した。なお、bCDとしてはMBCDを用いた。
(方法B)
RCC4細胞に10%血清の存在下、10mM 2DGを加えて、または加えずに2時間培養した。終了30分前に、各培地の細胞の分画に10mM bCDを加え、次いで細胞を収集し、分析した。
(方法C)
HeLa細胞を10%血清の存在下、10mM bCDで1時間培養し、次いで細胞を洗浄し、示された時間(~120分)10%血清を含む培地中再培養した。
(結果)
方法Bの結果を図2Bに、方法Cの結果を図2Cに示す。AKTのリン酸化は、bCDでの処置および2DGとbCDで共処置において、ほとんど完全に遮断された(図2B)。
しかしながら、bCDがない10%血清を含んだ培地へ細胞を戻した場合、AKT活性を復活するのに時間は掛からなかった(図2C)。
bCD誘導体の1つであるヒドロキシプロピル-β-シクロデキストリン(HPBCD)をマウスに注入して4時間後の、血循環に維持される量は約50%であることは、既にいくつかの研究で示されていた(J Inherit Metab Dis 2013, 36(3): 491-498; Toxicol Pathol 2008, 36(1):30-42)。したがって、癌療法のためにはbCDによる誘導でプロサバイバルシグナルを消滅させる利点を活かすには、アポトーシスを適正に素早く誘導される必要がある。
bCDが動物におけるPI3K-AKT経路に影響を与えるかどうか知るために、以下の実験を行った。なお、bCDとしてはHPBCDを用いた。
(方法D)
5時間絶食のマウスを40μg bCDで30分間前処置し、または前処理しなかった。それらに100ng IGF1を注入した。更に30分後にマウスのしっぽから採血して血中グルコースレベルを測定した。実験は3回行い、誤差は標準偏差を示す。マウスはすべて約20gであった。
(結果)
結果を図2Dに示す。
bCDは明らかにIGF1で誘導される低血糖を弱め、bCDが動物の体内のIGF1RとAKTとの間のシグナル伝達を部分的に遮断したことを暗示した。
bCDを2DG-ABTと組み合わせて、アポトーシス誘導が促進されるかどうか確認した。

RCC4細胞の分画を10%血清の存在下、1つの分画は10mM 2DGで2時間前培養し、別の分画は10mM bCDで30分間前培養し、更に別の分画は両方で処置し、いずれも処置していない対照(Untreated)も作成した。図2E中に示された量のABT-263を加えて1時間後、すべての細胞をPBSで洗浄し、10%血清を含む通常の培地中、終夜再培養した。細胞を収集し、FACSによるPI取り込みを分析した。プロトコールを図3に示す。なお、bCDとしてはMBCDを用いた。
(結果)
結果を図2Eのグラフに示す。
1μM ABT-263を用いて、2DG-bCD-ABTの組み合わせがRCC4細胞の約95%でアポトーシスを誘導した。
1μM ABT濃度での図2Eのデータを用いて、bCDと2DGが独立して作用すると仮定して、72%細胞死として期待される結果を計算したが、実測値は97%であった。その結果を図4に示す。独立性カイ二乗検定では0.0001未満のp値を示した。このようにbCDと2DGは、ABT誘導アポトーシスに相乗的にはたらくことがわかった。
まず、実験Aにて各種癌細胞株上のAKTプロサバイバルシグナルにおけるbCDの効果を試験し、A431類表皮癌細胞においては、実験BにてEGF、ERK1/2、PI3K、並びに各リン酸化体についてウェスタンブロット法で分析した。次に実験Cにて、ABTと組み合わせた場合のアポトーシス効果を確認した。なお、bCDとしてはMBCDを用いた。
(方法)
(実験A)HeLa子宮頸部癌細胞、UOK121腎臓癌細胞、Panc-1膵臓癌細胞、およびA431扁平上皮癌細胞の各細胞を、培地内に25mMグルコースの存在下、無処置、10mM 2DGでの前処置、最後の30分間の10mM bCDで共インキュベート、およびそれらの両方の処置を行い、細胞を集め、ウェスタンブロット分析した。
(実験B)(実験A)でのA431細胞溶菌液の同サンプルを、EGFR活性化、ERK1/2活性化、およびPI3K活性化について分析した。A431細胞におけるPI3K検出に関しては、抗PI3K p85オリゴクローナル抗体(6HCLC)を用いた。
(実験C)(実験A)と同様に処置された無処置の細胞、2DGとbCDで処置された細胞、1μM ABTで2時間インキュベートした細胞、および2DGとbCDの処置およびABTの処置をすべて行った細胞を調製した。細胞を集め、生細胞をトリパンブルー染色排除アッセイでカウントした。エラーバーは3サンプルからの標準偏差を示す。
(結果)
期待どおり、bCDはHeLa子宮頸部癌細胞、UOK121腎臓癌細胞、Panc-1膵臓癌細胞、およびA431扁平上皮癌細胞におけるAKTリン酸化を弱め、すべてにおいて、bCDがAKTプロサバイバルシグナルを弱めた(図5A)。
実験Aで用いた癌細胞の中で、EGFRを過剰発現することが知られたA431扁平上皮癌細胞を用いて、bCDと2DGでの処置の効果を確認した実験Bの結果より、EGFR、ERK1/2、PI3Kは、いずれも活性化された(図5B)。
しかしながら、実験Cの結果では、ABTを含めた3剤の組み合わせにより、すべての細胞株で非常に効果的にアポトーシスを誘導したことから、実験Aと実験Bの結果を考慮すれば、AKTプロサバイバルシグナルを弱めることで、ABTのアポトーシス誘導活性が高められることが示唆された。
なお、実験Cの結果で、Panc-1細胞においては、2DGとbCDの2剤で処置された結果と、ABTを含めた3剤の結果に顕著な差はなかったが、これはPanc-1細胞がABTを細胞内に取り込まない構成となっているためと考えられる。
2DGとbCDに相乗効果は上記の結果から非常に高い確率であるといえるが、これを分子生物学的に確立するため、実際にどの分子上でその相乗効果がみられるかを検索するため次の実験を組み立てた。最初にBakと供沈するタンパク質を集め、ウェスタンブロット法で、どの時点でMcl-1及びBcl-xLがBak結合体から失われるかを同定し、またどの時点でシトクロムcがミトコンドリアから放出されるかを光学顕微鏡で観察した。
(方法)
(AおよびB)RCC4細胞を、実施例6と同様の方法で、2DG、bCD、2DG+bCD、または無処置で処置した。約20μgの全細胞溶菌液(WCL)をウェスタンブロットで分析した。BakおよびBcl-xL結合するタンパク質を約200μgの細胞溶菌液から免疫沈降し、ウェスタンブロットで分析した。
(C)20μM 汎カスパーゼ阻害剤z-VADを含むまたは含まない3μMのABTを、2時間2DGおよびbCDの組み合わせで前処置した細胞に加え、カスパーゼ9についてウェスタンブロットで分析した。切断したカスパーゼ9をcC9で表示した。
(D)最後の2つのサンプルから200μgを用いてBak結合タンパク質を沈殿し、BcL-xLおよびBakについてブロットした。
(E)2DG-bCDで処置されたRCC4細胞(左のパネル)、2DG-bCD-ABTで処置されたRCC4細胞(中央のパネル)、および20μM 汎カスパーゼ阻害剤z-VADの存在下2DG-bCD-ABTで処置されたRCC4細胞(右のパネル)を、抗シトクロムc抗体およびGFP抱合抗マウス抗体を用いて免疫染色した。核をヨウ化プロピジウムで赤染色した。
2DGおよびbCDの両方で処置された細胞に現れる点状の緑のスポット(左のパネル)は、ミトコンドリアに局在するシトクロムcを表し、中央と右のパネルの両方の除かれた染色は、ミトコンドリアから放出されたシトクロムcを示す。
Note: シトクロムcはABTが加えられた後のみ生じる。本件および他の実験のプロトコールの図式的な詳細は図3にある。
(結果)
結果を図6のA~Eに示す。
Mcl-1-Bakの関係は消失した(図6A)。
それとは対照的に、ABTはBcl-xLと直接結合して、Bak-Bcl-xL複合体の解離の原因となった(図6D)。その場合だけ、カスパーゼ9が活性化された(図6C)。
このように、シトクロムcの放出は、ABTの添加後のみ起こり、カスパーゼ阻害剤の存在下、すなわち、カスパーゼ活性化がない状況でも起こった(図6DおよびE)。シトクロムc放出に続いて、カスパーゼ9が活性化された。
フルのシトクロムcの放出と、フルのカスパーゼ9活性化の両方がABT添加の2時間以内に観察され、最初に2DGを投入してから4時間以内でアポトーシスの最終段階に入ったことを我々は注目する。
これらの結果から、アポトーシス抑制因子の一つ、Mcl-1がBakから離れ、細胞が細胞死に感作されるには、2DGとbCDの両方が必要なことがわかった。つまり2DGとbCDの相乗作用でMcl-1がBak結合体から解き放された。さらにABTを加えると、ABTはBcl-xLに結合しBak結合体から失われ、すべての抑制因子から解き放されたBakは活性化し、シトクロムcをミトコンドリアから放出し、細胞死が始まることもわかった(図3C参照)。
(方法)
(A)ヒト由来癌細胞であるUOK121細胞をマウスに移植し、7日目に図3のインビボのプロトコールに従い治療を開始した。治療群は、無処置、2DG-ABT、HPBCD、および2DG-HPBCD-ABTで行った。処置は更に2度行い、腫瘍の大きさを記録した。エラーバーは標準偏差を示す。なお、2DG-HPBCD-ABTで治療されたマウスに限り、50日目に4度目の治療をした。
(B)(A)の高次評価として、(A)と同様の試験について、マウスを無処置、2DG-ABT、HPBCD、HPBCD-ABT、および2DG-HPBCD-ABTの5つの治療群に分け、10日目から30日目の間に8回処置した。
(結果)
3剤の組み合わせで処置された群だけで、腫瘍退縮が観察された(図7A)。他のすべてのマウスは、腫瘍が600mm3以上に成長したので、殺処分した。最初の2週間3剤の組み合わせで処置された群では、腫瘍は小さいままだった(60mm3未満)。マウスはその後の数週間処置せずに放置したところ、その間腫瘍はゆっくり成長し、遂には50日で400mm3に達した。
更に高次評価として行った結果では、図7Aの評価と同じく、3剤の組み合わせで処置されたマウスだけが治療に応答があり、腫瘍が小さいままだった(図7B)。他の治療群すべてのすべてのマウスにおける腫瘍が成長し、腫瘍は6週目までに600-1200mm3の範囲のサイズだった。これとは対照的に、3剤の組み合わせ群におけるマウスでは腫瘍はゆっくりと成長した。
我々は2DG-bCDが効率的な癌治療に、FasやTNF関連アポトーシス誘導リガンド(TRAIL)などのような、Bcl-2アンタゴニスト以外のアポトーシス誘導因子と組み合わせてできると仮説を立てた。bCDが単独または2DGと一緒になって、誘導のシトクロムc放出のためにミトコンドリアを明らかに感受性にしながら、bCDがカスパーゼ 8のデスレセプター活性化を妨げないことを暗示する報告がある(Molecular and cellular biology 2002, 22(1):207-220)。したがって、タイプII細胞中、アポトーシスの外因および内因の両経路が活性化され得るよう、TRAILはミトコンドリアを感受性にさせるのに有効であり得る。上記のことを確認するため、以下の実験を行った。
(方法)
(A)Panc-1細胞を標準的な2DG-bCD-TRAILプロトコール(図3)で処置した。生細胞を死細胞とトリパンブルー色素排除アッセイで識別し、生細胞をカウントし、グラフにした。実験は3剤の組み合わせで行い、エラーバーは標準偏差を示す。
(B)Panc-1細胞のウェスタンブロットは、無処置、TRAIL処置、2DG-bCDの組み合わせ処置、および図3に示す2DG-bCD-TRAILの組み合わせ処置で行った。細胞を6時間で収集した。実験は3回繰り返し、同様の結果を得た。
(結果)
結果を図8に示す。TRAILは10ng/mlの濃度で、Panc-1細胞において実質的に効果はなかった。しかしながら、これらの細胞を2DG-bCDで前処理した場合、同じ10ng/mlのTRAILは90%の細胞においてアポトーシスを誘導するのに十分であった。したがって、2DG-bCDはTRAILを介したアポトーシスのために、Panc-1細胞を明確に感受性にし、このことは2DG-bCD-TRAILが膵臓癌の効果的な治療であり得ることを暗示した。
腎がん由来のUOK121細胞をもちいてα-、β-、およびγ-シクロデキストリンが及ぼす、AKT活性化と2DG-ABTのアポプトーシス誘導効果を確認した。
(方法)
(実験A)UOK121細胞に10%血清の存在下、それぞれ5および10mMの濃度のα-、β-、およびγ-シクロデキストリンを加え45分間培養した。シクロデキストリン無添加の対照も同様に培養した。細胞を収集し、AKTおよびERK1/2のリン酸化をAKTおよびERK1/2の活性化の指標とし、ウェスタンブロット法で分析した。
(実験B)UOK121細胞を培地内に25mMグルコースの存在下、10mM 2DGを加えて、1.5時間インキュベートし、10mMのα-、β-、およびγ-シクロデキストリンをそれぞれ加え、30分間インキュベートし、その後、0.3μMのABT-263を加え、さらに2時間インキュベートした。別途、2DGもABT-263も無添加でα-、β-、およびγ-シクロデキストリンだけをそれぞれ加えた対照、並びにすべて無添加の対照を作成した。すべての細胞をシクロデキストリン添加して2.5時間後(ABT-263添加2時間後)、培地で2回洗い、翌日生きている細胞の数をトリパンブルー染色排除アッセイで測定した。
(結果)
結果を図9のAおよびBに示す。
実験Aにおいては、β-シクロデキストリンにより処理された細胞のみ、AKTの活性化が低下した(図9A)。実験Bおいても、β-シクロデキストリンで処置されたUOK121細胞には明らかにDG-ABTとの併用効果が上昇したが、α-およびγ-シクロデキストリンでは併用による改善効果はほとんど観察されなかった(図9B)。
Claims (7)
- β-シクロデキストリンまたはその誘導体を含む抗腫瘍剤。
- 1つまたは2つ以上の他の抗腫瘍剤と併用されることを特徴とする、β-シクロデキストリンまたはその誘導体。
- 他の抗腫瘍剤が2-デオキシグルコースを含む請求項2のβ-シクロデキストリンまたはその誘導体。
- 他の抗腫瘍剤がアポトーシス誘導作用を有する抗腫瘍剤を含む請求項2のβ-シクロデキストリンまたはその誘導体。
- 他の抗腫瘍剤が2-デオキシグルコースおよびアポトーシス誘導作用を有する抗腫瘍剤を含む請求項2のβ-シクロデキストリンまたはその誘導体。
- 他の抗腫瘍剤の投与と同時、または前もしくは後にβシクロデキストリンまたはその誘導体が投与される、請求項2~5のいずれかのβ-シクロデキストリンまたはその誘導体。
- 請求項2~6のいずれか一項のβ-シクロデキストリンまたはその誘導体を含む抗腫瘍剤。
