Description
しかし、従来の質量分析方法の多く方法では、分析対象試料に高温、真空、高電圧、レーザー照射等の特殊なイオン化の場(閉鎖環境)が必要であった。即ち、原理上、密閉したイオン化室内に試料を封入してイオン化を行うことが必要であり、試料分析を行う上での制約が大きい方法であった(例えば、非特許文献1、2参照)。
ここで、DART法(Direct Analysis in Real Time、リアルタイム直接分析法)は、励起ガスを大気環境下の試料にあてることにより、大気中の分子(特に水分子)と試料との相互作用を誘起して、試料のイオン化を行う方法である。当該DART方法は、試料を開放系にてイオン源にかざすだけで、直接試料のイオン化を可能とする優れた方法である。
また、DESI法(Desorption Electrospray Ionization、脱離エレクトロスプレーイオン化法)は、電圧を印加したキャピラリーを用いて試料表面に対してイオン化用の溶媒をスプレーすることにより、溶媒の帯電微小液滴を試料表面に付着させ、その際に試料表面から脱離したイオン化試料について、質量分析を行う方法である。
これらの方法では、その装置構造上、開放系でイオン化を行う方法であるため、分析対象が揮発性物質である場合には、拡散等でイオン化試料が離散してしまい、検出感度が著しく低下してしまうという原理的な課題が内在していた。
また、従来技術における揮発性物質の検出感度を高める手法としては、ある時間内に捕集瓶やサンプリングバッグ等に採取した気体試料等を分析するしか手段がなく、「大気圧下でのリアルタイム」にて揮発性物質を高感度分析する手法の開発が期待されていた。また、当該分析を簡便な操作により実現可能とすることも要望されていた。
(1)大気圧下にてリアルタイムでの試料分析が可能な質量分析装置において、イオン源の励起ガス噴出口と質量分析計のイオン化試料ガス回収口との間に、励起ガス導入口、試料ガス導入口、及びイオン化試料ガス放出口を有し、且つ、励起ガスと試料ガスを混合のための空間を有する構造、を有する連結デバイスを接続することにより、大気圧下リアルタイム質量分析における揮発性物質の検出感度を著しく増強できることを見出した。
(2)本発明者らは、上記(1)の知見を受けて、前記連結デバイスを接続した質量分析装置を用いることにより、揮発性物質の大気圧下リアルタイム質量分析を、極めて高感度で行うことが可能となることを見出した。
(3)さらに、本発明者らは、当該連結デバイスに外気導入口を形成させ、そこから通じる流路を、連結デバイス内の主流路と連通させることにより、検出感度をさらに増強できることを見出した。
また、当該連結デバイスを用いた大気圧下リアルタイム質量分析法は、試料の前処理が原則的には不要であり、リアルタイムでの揮発性物質の検出が可能な技術である。

しかしながら、当該インターフェイス部材は、「無機元素分析」を行うためのプラズマ質量分析装置(ICP)専用の部材である。ここで、図6に示すように、プラズマ質量分析装置(ICP)は、溶媒試料(符号64)を前処理によって霧化させ、これをプラズマ場(符号63)にてプラズマの作用によって、試料を励起元素化(元素レベルでイオン化)する原理を有する装置である。
また、特許文献1に記載のインターフェイス部材(符号61)は、ICPトーチ(符号62)におけるプラズマ場(符号63)の下流に接続して設置する部材であり、当該励起化元素(イオン化した試料)にさらに電位をかけることにより電子移動度を遅くして、測定感度を向上させることを目的とした部材である。
当該原理が示すように、特許文献1のインターフェイス部材に導入されるものは、既にイオン化した試料(励起元素化した試料:符号65)であり、本発明に係る連結デバイスが有するような「試料ガスと励起ガスが混合する空間」は存在しない。
さらに、プラズマ質量分析装置(ICP)は、溶媒試料(符号64)をサンプルとして供与する必要があるため、原理的に大気圧下でのリアルタイム質量分析を実現することができない。
これらの点から明らかなように、特許文献1のインターフェイス部材と、本発明に係る連結デバイスとでは、その構造及び作用機能が全く異なる部材である。
本発明は、揮発性物質の大気圧下リアルタイム質量分析を高感度にて可能とする質量分析装置用連結デバイスに関するものである。
また、本発明は、揮発性物質の大気圧下リアルタイム質量分析を高感度で行う方法に関するものである。
本発明に係る連結デバイス(符号1)は、大気圧下リアルタイム質量分析装置(符号21)に接続するインターフェイス部材である。イオン源の励起ガス噴出口(符号32)と、質量分析計のイオン化試料ガス回収口(符号42)と、の間に接続して用いる連結デバイスである。
当該連結デバイスを大気圧下リアルタイム質量分析装置接続することにより、質量分析の感度(マスクロマトグラムのピーク値)を飛躍的に増強することが可能となる。即ち、本発明に係る当該連結デバイスは、大気圧下リアルタイム質量分析装置用の感度増強用連結デバイスである。
本発明に係る連結デバイス(符号1)は、励起ガス導入口(符号2)、試料ガス導入口(符号3)、及びイオン化試料ガス放出口(符号7)、を有する部材である。
なお、本明細書において「直管形状」とは、管が屈曲することなく実質的に真っ直ぐな形状を指すものである。横断面の形状が円状や環状の管だけでなく、多角状や多角環状のものも含まれる。
当該連結デバイス(符号1)は、試料ガス導入口(符号3)から通じる試料ガス導入用流路(符号6)が、連結デバイス内主流路(符号10)に連通する構造を有する部材である。当該構造により、連結デバイス内主流路(符号10)の一部の領域には励起ガス-試料ガス混合空間(符号4)が形成される。
励起ガス-試料ガス混合空間(符号4)は、連結デバイス内主流路(符号10)の直管形状の流路部分に形成されてなることが好適である。
当該連結デバイス(符号1)の全体の外形形状としては、上記主要構造を充足するような構造体であれば、如何なる外形形状を有するものを採用することができる。
当該柱状又は略柱状としては、円柱状、樽型形状、角柱状(三角柱状、四角柱状、六角柱状等)、角環柱状、エンタシス状(中央部分が膨らんだ柱状)、逆エンタシス形状(中央部分が窪んだ柱状)、断頭円錐状、断頭角錐状(断頭三角錐状、断頭四角錐状、断頭六角錐状等)、台形柱状、などを横倒しした形状、を挙げることができる。これらの形状には、柱長(横軸の長さ)が断面幅(縦軸の長さ)よりも短い形状も含まれる。即ち、キューブ状、切り株状、なども当該形状に含まれる。
また、これらに準ずる略形状を挙げることも可能である。また、当該列挙した各形状を組み合わせてなる形状についても、当該連結デバイス(符号1)の外形形状として採用することもできる。
なお、本明細書において「管状」及び「管形状」とは、横断面の形状が円状や環状の管だけでなく、多角状や多角環状のものも含まれる。
外形長の下限としては、5mm以上、好ましくは10mm以上、より好ましくは15mm以上、さらに好ましくは20mm以上、特に好ましくは25mm以上、とすることが好適である。
また、外形長の上限としては、イオン化した試料ガス(符号12)のイオン化状態が保持されている間(長くても1秒以内、好ましくは500m秒以内)に、質量分析計のイオン化試料ガス回収口(符号42)に到達できる長さであれば、特に制限はない。例えば、120mm以下、好ましくは100mm以下、より好ましくは80mm以下、さらに好ましくは60mm以下、特に好ましくは50mm以下、一層好ましくは45mm以下、より一層好ましくは40mm以下、さらに一層好ましくは35mm以下を挙げることができる。
当該外形幅の上限としては、5mm以上、好ましくは6mm以上、より好ましくは8mm以上、さらに好ましくは10mm以上、特に好ましくは12mm以上を挙げることができる。
また、外形幅の下限としては、80mm以下、好ましくは50mm以下、より好ましくは40mm以下、さらに好ましくは30mm以下、特に好ましくは25mm以下、一層好ましくは20mm以下を挙げることができる。
当該連結デバイス(符号1)の支持体を構成する材質としては、十分な強度を有する材質であれば特に制限はなく、如何なる材質のものも用いることが可能である。例えば、樹脂、セラミック、金属、鉱石、ガラス、などを挙げることができる。好ましくは、絶縁製の材質からなる部材が好適である。
当該材質として具体的には、フッ化炭素樹脂(PTFE、PFA、FEPなど)、ポリプロピレン樹脂(PP)、ポリエーテルエーテルケトン樹脂(PEEK)、ポリイミド樹脂、セラミック(アルミナ、窒化アルミ等)、などを挙げることができる。また、これらの材質を組み合わせて成形したものであっても良い。特には、PTFE(ポリテトラフルオロエチレン)、アルミナ、窒化アルミ、等を用いることが好適である。
当該連結デバイス(符号1)は、「励起ガス導入口」(符号2)を有する部材である。当該励起ガス導入口は、イオン源の励起ガス噴出口(符号32)から噴出される励起ガスを、当該デバイス内に導入するために必要な孔である。
当該口幅の下限としては、0.5mm以上、好ましくは0.75mm以上、より好ましくは1mm以上、さらに好ましくは1.5mm以上、特に好ましくは2mm以上、一層好ましくは2.5mm以上を挙げることができる。
また、当該口幅の上限としては、30mm以下、好ましくは20mm以下、より好ましくは10mm以下、さらに好ましくは8mm以下、特に好ましくは6mm以下、一層好ましくは5mm以下、より一層好ましくは4mm以下、さらに一層好ましくは3.5mm以下を挙げることができる。
例えば、当該連結デバイスの支持体部分を内側にくり抜き、励起ガス導入口(符号2)が当該くり抜き部分の底部に形成される形状とすることができる。当該くり抜き形状とすることで、連結デバイスの当該くり抜き部分に、イオン源の励起ガス噴出口(符号32)を差し込んで接続することが容易となる。
なお、当該くり抜き幅(円状又は環状の場合は内径)は、当該励起ガス噴出口(符号32)の外形(円状又は環状の場合は外径)に合わせて、成形することが可能である(図3B参照)。
当該連結デバイス(符号1)は、「イオン化試料ガス放出口」(符号7)を有することを特徴とする部材である。当該イオン化試料ガス放出口(符号7)は、当該連結デバイス内でイオン化した試料ガス(符号12)を、当該連結デバイスから放出して、質量分析計のイオン化試料ガス回収口(符号42)に、受け渡すために必要な孔である。
当該口幅の下限としては、0.5mm以上、好ましくは0.6mm以上、より好ましくは0.8mm以上、さらに好ましくは1mm以上、特に好ましくは1.2mm以上、一層好ましくは1.4mm以上、より一層好ましくは1.5mm以上を挙げることができる。
また、当該口幅の上限としては、30mm以下、好ましくは20mm以下、より好ましくは10mm以下、さらに好ましくは8mm以下、特に好ましくは6mm以下、一層好ましくは5mm以下、より一層好ましくは4mm以下、さらに一層好ましくは3mm以下、さらにより一層好ましくは2.5mm以下を挙げることができる。
当該くり抜き形状とすることで、連結デバイスの当該くり抜き部分に、質量分析計のイオン化試料ガス回収口(符号42)を差し込んで接続することが可能となる。
なお、当該くり抜き幅(円状又は環状の場合は内径)は、当該イオン化試料ガス回収口(符号42)の外形(円状又は環状の場合は外径)に合わせて、成形することが可能である(図3C参照)。
当該連結デバイス(符号1)は、「試料ガス導入口」(符号3)を有することを特徴とする部材である。試料ガス導入口(符号3)は、試料ガスである揮発性物質ガス(符号11)を、当該デバイス内に導入するために必要な孔である。
当該口幅の下限としては、0.05mm以上、好ましくは0.08mm以上、より好ましくは0.1mm以上、さらに好ましくは0.2mm以上、特に好ましくは0.4mm以上、一層好ましくは0.5mm以上、より一層好ましくは0.6mm以上、さらに一層好ましくは0.8mm以上、特に一層好ましくは1.0mm以上、よりさらに一層好ましくは1.2mm以上、特にさらに一層好ましくは1.4mm以上、最も好ましくは1.5mm以上、を挙げることができる。
また、当該口幅の上限としては、30mm以下、好ましくは20mm以下、より好ましくは10mm以下、さらに好ましくは8mm以下、特に好ましくは6mm以下、一層好ましくは5mm以下、より一層好ましくは4mm以下、さらに一層好ましくは3mm以下、特に一層好ましくは2.5mm以下を挙げることができる。
なお、当該試料ガス導入口(符号3)は、当該連結デバイス(符号1)の上下の制限なく配置することができる。
当該試料ガス導入口(符号3)の位置を、励起ガス導入口(符号2)に近い位置に形成させるほど、試料ガス(符号11)のイオン化効率が向上することができて好適である。
また、当該試料ガス導入口(符号3)の位置としては、当該連結デバイス(符号1)の上下の制限なく配置することができる。
当該連結デバイス(符号1)は、励起ガス導入口(符号2)とイオン化試料ガス放出口(符号7)とが連通してなる流路が形成されてなる構造体である。当該流路は、「連結デバイス内主流路」(符号10)となる。
当該連結デバイス内主流路(符号10)の一部は、励起ガス-試料ガス混合空間(符号4)とイオン化試料ガス用流路(符号5)を構成する。
さらに好ましくは、当該連結デバイス内主流路(符号10)としては、直管形状のみの直線流路にして、励起ガス導入口(符号2)とイオン化試料ガス放出口(符号7)とを最短距離で結んで連通させた流路とすることが最適である。当該態様によりガスの流体抵抗を減少させることができる。
当該連結デバイス(符号1)は、試料ガス導入口(符号3)から通じる「試料ガス導入用流路」(符号6)を有する部材である。
試料ガス導入用流路(符号6)は、試料ガス(揮発性物質ガス:符号11)を、励起ガス-試料ガス混合空間(符号4)に導くために必要な流路である。
当該流路幅の下限としては、0.05mm以上、好ましくは0.08mm以上、より好ましくは0.1mm以上、さらに好ましくは0.2mm以上、特に好ましくは0.4mm以上、一層好ましくは0.5mm以上、より一層好ましくは0.6mm以上、さらに一層好ましくは0.8mm以上、特に一層好ましくは1.0mm以上、よりさらに一層好ましくは1.2mm以上、特にさらに一層好ましくは1.4mm以上、最も好ましくは1.5mm以上、を挙げることができる。
また、当該流路幅の上限としては、30mm以下、好ましくは20mm以下、より好ましくは10mm以下、さらに好ましくは8mm以下、特に好ましくは6mm以下、一層好ましくは5mm以下、より一層好ましくは4mm以下、さらに一層好ましくは3mm以下、特に一層好ましくは2.5mm以下を挙げることができる。
当該流路長の下限としては、2mm以上、好ましくは3mm以上、より好ましくは4mm以上、さらに好ましくは5mm以上を挙げることができる。
また、当該流路長の上限としては、50mm以下、より好ましくは30mm以下、さらに好ましくは25mm以下、特に好ましくは20mm以下、一層好ましくは15mm以下、より一層好ましくは10mm以下を挙げることができる。
連結デバイス内主流路(符号10)における当該連通部分は、好ましくは直管形状であることが好適である。直管形状の流路部分でないところで連通させた場合、試料ガス導入用流路(符号6)へのガスの逆流が発生する可能性があり望ましくない。
当該連通位置を、励起ガス導入口(符号2)に近い位置に形成させるほど、試料ガス(符号11)のイオン化効率を向上することができて好適である。
当該連通(接続)角度の下限としては、鋭角である場合には特に制限はないが、具体的には10°以上、好ましくは20°以上、より好ましくは30°以上の角度を挙げることができる。
当該連結デバイス(符号1)は、連結デバイス内主流路(符号10)の一部の領域に、励起ガス-試料ガス混合空間(符号4)を有する部材である。励起ガス-試料ガス混合空間(符号4)は、励起ガスと試料ガスを混合するための空間であり、連結デバイス内主流路(符号10)に、試料ガス導入用流路(符号6)が連通することにより形成される空間である。
励起ガス-試料ガス混合空間(符号4)では、イオン化前の試料ガス(揮発性物質ガス:符号11)が集中するため、当該空間において励起ガスと試料ガスの混合がおこり、当該試料ガスのイオン化を高効率に誘起することが可能となる。
励起ガス-試料ガス混合空間(符号4)は、連結デバイス内主流路(符号10)の直管形状の流路部分に形成された空間であることが望ましい。
当該流路幅(空間の断面幅)の下限としては、0.5mm以上、好ましくは0.75mm以上、より好ましくは1mm以上、さらに好ましくは1.5mm以上、特に好ましくは2mm以上、一層好ましくは2.5mm以上を挙げることができる。
また、当該流路幅(空間の断面幅)の上限としては、30mm以下、好ましくは20mm以下、より好ましくは10mm以下、さらに好ましくは8mm以下、特に好ましくは6mm以下、一層好ましくは5mm以下、より一層好ましくは4mm以下、さらに一層好ましくは3.5mm以下を挙げることができる。
当該流路長(空間長)の下限としては、2mm以上、好ましくは3mm以上、より好ましくは4mm以上、さらに好ましくは4.5mm以上を挙げることができる。
また、当該流路長(空間長)の上限としては、40mm以下、より好ましくは30mm以下、さらに好ましくは20mm以下、特に好ましくは15mm以下、一層好ましくは12mm以下、より一層好ましくは10mm以下、さらに一層好ましくは8mm以下、特に一層好ましくは6mm以下、よりさらに一層好ましくは5.5mm以下、を挙げることができる。
好ましくは、励起ガス導入口(符号2)から、連結デバイス内主流路(符号10)の流路長50mm以内、好ましくは30mm以内、より好ましくは25mm以内、さらに好ましくは20mm以内、特に好ましくは15mm以内、一層好ましくは10mm以内、より一層好ましくは8mm以内、さらに一層好ましくは6mm以内、特に一層好ましくは5mm以内、よりさらに一層好ましくは4mm以内、に形成された空間であることが好適である。
なお、当該形成位置としては、試料ガス導入用流路(符号6)の連通(接続)位置や角度を調整することにより、所望の位置に形成させることが可能である。
なお、試料ガス導入口(符号3)及び試料ガス導入用流路(符号6)を2以上有する連結デバイス(符号1)、又は、試料ガス導入口(符号3)は1つであるが途中で分岐した形状の試料ガス導入用流路(符号6)を有する連結デバイス(符号1)では、試料ガス導入用流路(符号6)と連結デバイス内主流路(符号10)との連通部位が2以上となるため、励起ガス-試料ガス混合空間(符号4)を2以上有する形態とすることも可能である。
当該連結デバイスでは、励起ガス-試料ガス混合空間(符号4)を、「励起ガス-試料ガス混合室」(部屋様の空間)とすることによって、さらに試料ガス(符号11)の集中を効率良く行うことが可能となる。これにより、試料ガス(符号11)のイオン化を飛躍的に効率化することが可能となる。
励起ガス-試料ガス混合室の形成は、具体的には、連結デバイス内主流路(符号10)の励起ガス導入口(符号2)側に、イオン化試料ガス放出口(符号7)側よりも相対的に横断面積の広い空間を形成することにより実現できる。また、反対に、イオン化試料ガス放出口(符号7)側に、励起ガス導入口(符号2)側よりも相対的に横断面積の狭い空間を形成することによっても実現できる。
当該値の下限としては、0.1mm以上、好ましくは0.2mm以上、より好ましくは0.3mm以上、さらに好ましくは0.4mm以上、特に好ましくは0.5mm以上、一層好ましくは0.6mm以上、より一層好ましくは0.7mm以上、さらに一層好ましくは0.8mm以上、を挙げることができる。
また、当該値の上限としては、20mm以下、好ましくは10mm以下、より好ましくは5mm以下、さらに好ましくは4mm以下、特に好ましくは3mm以下、一層好ましくは2mm以下、より一層好ましくは1.5mm以下、を挙げることができる。
当該狭める横断面積の差が大きすぎる場合、段差部分に圧力が大きくなりすぎて好ましくない。また、狭める横断面積の差が小さすぎる場合、試料ガス(符号11)の集中化がおこりにくくなり、好ましくない。
なお、当該部屋の下流側の流路を、徐々に狭める形状(略円錐曲面状など)とした場合、試料ガス(符号11)の集中化を確保しつつ流体抵抗を低減できるため好適である。
前記連結デバイス内主流路(符号10)のうち、励起ガス-試料ガス混合空間(符号4)より下流側(イオン化試料ガス放出口側)の流路は、「イオン化試料ガス用流路」(符号5)に相当する流路となる。イオン化試料ガス用流路(符号5)は、イオン化した試料ガス(符号12)を、イオン化試料ガス放出口(符号7)に導くために必要な流路である。
当該流路幅の下限としては、0.5mm以上、好ましくは0.6mm以上、より好ましくは0.8mm以上、さらに好ましくは1mm以上、特に好ましくは1.2mm以上、一層好ましくは1.4mm以上、より一層好ましくは1.5mm以上を挙げることができる。

当該連結デバイス(符号1)は、必要に応じて、質量分析装置や固定用アダプターへの固定手段を備えた構造を付加した形態とすることができる。例えば、固定用の穴(符号14)等を穿った構造とすることもできる。
本発明に係る連結デバイス(符号1)は、「外気導入機構」(符号13)を備えた部材とすることにより、質量分析の感度(マスクロマトグラムのピーク値)をさらに増強することが可能となる。
ここで、当該外気導入機構(符号13)とは、特定構造及び特定位置に形成されてなる外気導入口(符号8)及び外気導入用流路(符号9)からなる機構を指す。
ここで、「外気」としては、通常は大気ガスを指すものであるが、精製した空気、窒素ガス、ヘリウムガス、アルゴンガス等を取り込ませることも可能である。この場合、不純物が少ない分さらなる感度の上昇が期待できる。
また、負圧による自然流入でなく、強制的な加圧によって、外気導入機構(符号13)から外気をバランスガスとして導入することも可能である。
(i)当該連結デバイス及び装置全体での圧力制御が安定する作用が奏される。これにより、流速が安定し、励起ガス-試料ガス混合空間(符号4)での試料ガスのイオン化反応を安定して進行させることが可能となる。
(ii)また、取り込まれた外気(大気ガス)に含まれる水分子は、試料ガスのイオン化を、さらに促進する作用がある。当該作用により、励起ガス-試料ガス混合空間(符号4)を通過した未反応の試料ガスについても、イオン化試料ガス用流路(符号5)内にてイオン化させることが可能となる。当該作用は、特にイオン源としてDARTを用いた時に発揮される作用である。
(iii)さらに、外気導入機構(符号13)から外部標準物質を導入することにより、揮発性物質の定量を容易に行うことが可能となる。これにより、内部標準物質を試料に添加する操作(試料の前処理)を必要とすることなく、高感度の定量が可能となる。
当該外気導入機構(符号13)を有する連結デバイス(符号1)としては、「外気導入口」(符号8)を形成した部材とすることが必要である。
当該口幅の下限としては、0.1mm以上、好ましくは0.5mm以上、より好ましくは1mm以上、さらに好ましくは1.5mm以上、特に好ましくは2mm以上、一層好ましくは3mm以上、を挙げることができる。
また、当該口幅の上限としては、30mm以下、好ましくは20mm以下、より好ましくは10mm以下、さらに好ましくは8mm以下、特に好ましくは6mm以下、一層好ましくは5mm以下、より一層好ましくは4.5mm以下を挙げることができる。
なお、当該連結デバイス(符号1)の外形形状が柱状、略柱状、管状、又は筒状等の形状の場合、上記励起ガス導入口(符号2)及びイオン化試料ガス放出口(符号7)を形成した側面とは異なる外形面に、当該外気導入口(符号8)を形成させることも好適である。
また、当該外気導入口(符号8)の位置としては、当該連結デバイス(符号1)の上下方向等に制限されることなく配置することができる。
当該外気導入口(符号8)の位置を、励起ガス-試料ガス混合空間(符号4)から離れた位置に形成させるほど、試料ガス(符号11)のイオン化効率が向上することができて好適である。
なお、当該距離の上限としては、特に制限はないが、例えば、100mm以下、好ましくは90mm以下、より好ましくは80mm以下、さらに好ましくは70mm以下、特に好ましくは60mm以下、一層好ましくは50mm以下、を挙げることができる。
当該外気導入機構(符号13)を有する連結デバイス(符号1)としては、外気導入口(符号8)から通じる「外気導入用流路」(符号9)を有する部材とすることが必要である。
当該外気導入用流路(符号9)は、イオン化試料ガス用流路(符号5)に、連通(接続)してなる流路である。当該外気導入用流路(符号9)は、外気(大気ガス等)を、イオン化試料ガス用流路(符号5)に導くために必要な流路である。
当該流路幅の下限としては、0.1mm以上、好ましくは0.5mm、より好ましくは1mm以上、さらに好ましくは1.5mm以上、特に好ましくは2mm以上、一層好ましくは3mm以上、より一層好ましくは3.5mm以上、を挙げることができる。
また、当該流路幅の上限としては、30mm以下、好ましくは20mm以下、より好ましくは10mm以下、さらに好ましくは8mm以下、特に好ましくは6mm以下、一層好ましくは5mm以下、より一層好ましくは4.5mm以下を挙げることができる。
当該流路長の下限としては、2mm以上、好ましくは3mm以上、より好ましくは4mm以上、さらに好ましくは5mm以上を挙げることができる。
また、当該流路長の上限としては、50mm以下、好ましくは30mm以下、より好ましくは25mm以下、さらに好ましくは20mm以下、特に好ましくは15mm以下、一層好ましくは10mm以下を挙げることができる。
連結デバイス内主流路(符号10)における当該連通部分は、好ましくは直管形状であることが好適である。直管形状の流路部分でないところで連通させた場合、外気導入用流路(符号9)へのガスの逆流が発生する可能性があり望ましくない。
当該凹状構造としては、ドーム状、円錐曲面状、略円錐曲面状に抉った又は穿った形状であっても良いが、周囲の壁面から彫り込んだ段差構造又は略段差構造であることが好適である。
当該凹状構造としては、イオン化試料ガス用流路(符号5)の壁面から0.1mm以上、好ましくは0.2mm以上、より好ましくは0.3mm以上、さらに好ましくは0.4mm以上、特に好ましくは0.5mm以上、彫り込んだ段差構造又は略段差構造であることが好適である。
また、当該段差の上限としては、5mm以下、好ましくは4mm以下、より好ましくは3mm以下、さらに好ましくは2mm以下の段差であることが好適である。
当該凹状構造の上面視からの(平面的な)形状及び構造幅としては、外気導入用流路(符号9)の流路横断面と同形状及び同サイズであることが好適である。
具体的には、試料ガス導入用流路(符号6)と連結デバイス内主流路(符号10)との連通位置の下流側(イオン化試料ガス放出口側)の外縁を起点として、当該外気導入用流路(符号9)と連結デバイス内主流路(符号10)の連通部分の上流側(励起ガス導入口側)の外縁が、イオン化試料ガス用流路(符号5)の流路長の下流側(イオン化試料ガス放出口側)の方向に2.5mm以上、好ましくは5mm以上、より好ましくは7.5mm以上、さらに好ましくは10mm以上、特に好ましくは15mm以上、一層好ましくは20m以上、より一層好ましくは25mm以上、離れた位置であることが好適である。
当該連通(接続)位置を、励起ガス-試料ガス混合空間(符号4)から離れた位置に形成させるほど、試料ガスのイオン化効率が向上することができて好適である。
なお、当該距離の上限としては、特に制限はないが、例えば、100mm以下、好ましくは90mm以下、より好ましくは80mm以下、さらに好ましくは70mm以下、特に好ましくは60mm以下、一層好ましくは50mm以下、を挙げることができる。
当該連通(接続)角度の下限としては、鋭角である場合には特に制限はないが、具体的には10°以上、好ましくは20°以上、より好ましくは30°以上の角度を挙げることができる。
本発明に係る連結デバイス(符号1)は、大気圧下リアルタイム質量分析装置(符号21)と接続することにより、揮発性物質の大気圧下でのリアルタイム質量分析を、極めて高感度にて行うことが可能となる。
ここで、「大気圧下リアルタイム質量分析装置」(符号21)とは、大気圧下においてリアルタイムにて、高感度の質量分析を可能とする質量分析装置を指す。
当該連結デバイス(符号1)と大気圧下リアルタイム質量分析装置(符号21)との接続は、励起ガス導入口(符号2)をイオン源の励起ガス噴出口(符号32)と接続し、イオン化試料ガス放出口(符号7)を質量分析計のイオン化試料ガス回収口(符号42)と接続することにより実現される。
ここで、「接続状態」としては、密閉状態とすることが望ましいが、特に気密性が高い状態とすることまでは要さない。接触により摺動するような緩い接続状態であっても、当該接続態様に含まれる。
また、これらの接続部分には、アダプターやアクセサリー部材を噛ませることも可能である。
また、装置と接続するために装置固定用のアダプターやアクセサリー部材(符号46)に、本発明に係る連結デバイス(符号1)を装着する態様とすることも可能である。
大気圧下リアルタイム質量分析装置(符号21)としては、試料のイオン化を大気圧下において実現可能な「イオン源」(符号31)を用いた装置であれば、如何なる装置を挙げることができる。
当該イオン源(符号31)としては、大気圧下の気相中において、試料ガス(符号11)をイオン化することが可能な原理を利用したイオン源であれば、如何なるものを用いることもできる。具体的には、DART法(リアルタイム直接分析法)の原理を利用したイオン源を、好適に用いることができる。
さらに、DESI法(脱離エレクトロスプレーイオン化法)、ESI法(エレクトロスプレーイオン化法)、API法(大気圧イオン化法)、APPI法(大気圧光イオン化法)、APCI法(大気圧化学イオン化法)、ASAP法(大気圧固体試料プローブ法)、MALDI法(マトリックス支援レーザー脱離イオン化法)、EI法(電子イオン化法)、CI法(化学イオン化法)、FD法(電界脱離法)、FAPA法(流動大気圧残光法)、DBD法(誘導バリア放電法)、ADI法(大気脱離イオン化法)、HPIS法(ヘリウムプラズマイオン化源法)、LTP法(低温プラズマ法)、などの原理を利用したものであっても、試料ガス(符号11)の大気圧下でのイオン化が可能な方法であれば、当該イオン源として応用利用することが可能である。
当該質量分析装置(符号21)に用いる「質量分析計」(符号41)としては、通常の質量分析計の分析部に相当するものであれば、如何なるものを用いることができる。
例えば、飛行時間型(TOF型:Time-of-Flight型)、磁場偏光型(Magnetic Sector型)、四重極型(Q型:Quadrupole型)、イオントラップ型(IT型:Ion Trap型)、フーリエ変換イオンサイクロトロン共鳴型(FT-ICR型:Fourier-Transform Ion Cyclotron Resonance型)、加速器質量分析型(AMS型:Accelerator Mass Spectrometry型)の分析計を挙げることができる。また、これらを複数組み合わせたタンデム型も挙げることができる。
これらのうち、特に飛行時間型(TOF型:Time-of-Flight型)は、原理的に測定可能な質量の制限がなく且つ高感度であることから、好適に用いることができる。
当該排気手段としては、例えば、排気管(符号44)を接続して、真空ポンプ(符号45)を用いてイオン化試料ガス(符号12)以外の気体を外へ積極的に排気する手段、を挙げることができる。
当該連結デバイス(符号1)の試料ガス導入口(符号3)には、試料ガス取り込み管(符号51)を接続して用いることで、当該連結デバイス内に試料ガス(符号11)を効率良く導くことが可能となる。
当該試料ガス取り込み管(符号51)は、試料物質を封入した容器(試料封入容器:符号52)と接続して用いることが好適である。また、当該接続部分には、アダプターやアクセサリー部材(符号55)を噛ませることも可能である。
揮発性の低い成分を検出する場合には、試料封入容器(符号52)に揮発用ガス導入管(符号53)を接続し、揮発用ガス(ヘリウムガス、窒素ガス、大気など)をパージすることで、当該試料に含まれる揮発性成分の揮発を促進させることができる。
本発明では、前記連結デバイス(符号1)を接続した大気圧下リアルタイム質量分析装置(符号21)を用いることにより、揮発性物質のリアルタイムでの質量分析を極めて高感度で行うことが可能となる。
また、当該連結デバイス(符号1)は、市販の如何なる大気圧下リアルタイム質量分析装置とも接続可能に成形できる汎用部材であるため、市販装置への脱着が容易である。そのため、簡便にリアルタイムでの高感度質量分析を実現することが可能となる。
なお、当該質量分析法では、試料の前処理が原則的には不要であり、リアルタイムでの揮発性物質の検出が可能となる。
当該質量分析方法において分析対象となる試料は、揮発性物質である。本発明では、あらゆる揮発性物質の高感度質量分析を可能とする。
ここで、「揮発性物質」とは、大気中において蒸気圧を有する物質の総称である。具体的には、大気中で冷気ガスと接触する条件において、分圧を有する物質と定義することができる。
例えば、飲食品、香料、化粧品等に含まれる香気成分、風味成分、臭気成分、;医薬品に含まれる薬理成分、;病理検体に含まれる微量成分、;塗料、色素等に含まれる色素成分、;など、様々な分野の製品等を構成する物質を挙げることができる。
当該質量分析方法では、連結デバイス(符号1)の試料ガス導入口(符号3)から、励起ガス-試料ガス混合空間(符号4)に、測定対象である試料ガス(揮発性物質ガス:符号11)を導入する。
当該試料ガス(符号11)の導入は、励起ガス流が生み出す負圧を利用して行うこともできるが、揮発用ガスをパージして積極的に試料を揮発させて導入する方が効率的である。
当該質量分析方法では、当該連結デバイス(符号1)の励起ガス導入口(符号2)に、イオン源の励起ガス噴出口(符号32)から噴出される励起ガスを導入する。当該励起ガスは、イオン源からのガス流によって、そのまま励起ガス-試料ガス混合空間(符号4)内に導かれる。
ここで、イオン源(符号31)から導入される励起ガスとしては、具体的には、励起ヘリウムガス、励起窒素ガス、励起ネオン、などを挙げることができる。好ましくは励起ヘリウムガスであることが好適である。
励起ガス-試料ガス混合空間(符号4)内へ試料ガス導入及び励起ガスを導入することにより、両者の混合がおこる。これにより、試料ガス(符号11)のイオン化が促進される。
ここで、「試料ガスのイオン化」とは、試料(揮発性物質)の気体分子(符号11)が、励起ガス及び大気成分との相互作用によって、イオン化した状態(符号12)を指す。
また、外気導入機構(符号13)を備えた連結デバイスの場合、さらに飛躍的に試料ガス(符号11)のイオン化を促進させることが可能となる。
当該連結デバイス(符号1)のイオン化試料ガス放出口(符号7)から、イオン化した試料ガス(符号12)が放出され、質量分析計のイオン化試料ガス回収口(符号42)に、イオン化試料ガス(符号12)が導入される。
当該質量分析方法における分析工程及び検出工程については、特殊な操作等を必要とせず、通常の大気圧下リアルタイム質量分析装置の使用方法に準じて行うことが可能である。
当該質量分析方法においては、外気導入機構(符号13)を備えた連結デバイス(符号1)を用いた場合、外部標準物質を外気導入機構(符号13)から取り込ませることにより、揮発性物質の高精度での定量を行うことが可能となる。
外気導入機構(符号13)により検出感度が飛躍的に向上することにより、従来は困難であった高精度での揮発性物質のリアルタイムでの定量が可能となる。
ここで、定量に用いる外部標準物質としては、検量線を精度良く作成可能な物質であれば、特に制限なく使用可能である。
本発明に係る質量分析法により、試料からの揮発性物質の直接のモニタリングが可能となる。これにより、飲食品、香料、化粧品、医薬品、医療、診断、塗料、溶剤、農薬、法医学、麻薬検査、有機物質合成等の様々な分野での応用が期待される。
特に、本発明に係る質量分析法は、食品等の物性変化試験及び状態変化試験、有機化合物の合成反応過程、及び製造工程等、これまでは揮発性物質のリアルタイムモニタリングが不可能であった分野において、利用させることが期待される。
本発明に係る質量分析装置用連結デバイスを用いて、大気圧下リアルタイム質量分析装置であるDART-MSによる揮発性物質のリアルタイム分析を行った。
本実施例で用いた感度増強用連結デバイス(符号1)は、横倒しした円柱形状(側面の直径15mm、長さ35mm)の外形形状を有する連結部材である(図3A参照)。なお、当該連結デバイスは、耐熱性に優れたPTFE樹脂製(テフロン(登録商標)樹脂製)の素材からなる。
当該連結デバイス(符号1)のイオン源接続側(左側面側、図3B参照)は、イオン源の励起ガス噴出ノズル(符号32、図4参照)との接続に適した形状を有する。当該形状は、励起ガスを当該連結デバイスに導入するのに適した形状である。
具体的には、当該側面端から質量分析計側(右側面側)方向へ4mmの位置まで、円柱の外側厚さ1mmの外縁を残して内側がくり抜かれた構造を有する。また、そこからさらに質量分析計側(右側面側)方向へ3mm(当該側面端から7mm)の位置まで、中心部が略鈍角円錐状にくり抜かれた形状を有する(図3B参照)。当該略鈍角円錐曲面の中心には、内径3mmの励起ガス導入口(符号2、図3B参照)が穿ってある。
当該励起ガス導入口(符号2)からは、質量分析計側(右側面側)方向へ5mmの位置(イオン源接続側の端部から12mmの位置)まで、内径3mmの管が水平に穿ってある。当該管内に形成される空間(内径3mm、長さ5mmの太円柱状空間)は、励起ガス-試料ガス混合室(符号4、図1及び2参照)を形成する。
当該試料混合室のさらに奥には、内径2mmの直管(イオン化試料ガス用流路:符号5、図1及び2参照)が、質量分析計側(右側面側)に向かって水平に穿ってある。
励起ガス導入口(符号2)から質量分析計側(右側面側)方向へ3mmの位置(イオン源接続側の端部から10mm)の位置には、円柱側面下側に内径2mmの試料ガス導入口(符号3、図3D参照)が穿ってある。
当該試料ガス導入口(符号3)は、当該円柱の中心方向(垂直方向)へ、内径2mmの直管(試料ガス導入用流路:符号6)が垂直に穿ってある。当該流路は、上記励起ガス-試料ガス混合室(符号4)に連通している(図1及び2参照)。
当該連結デバイス(符号1)の質量分析計接続側(右側面側、図3C参照)は、質量分析計のイオン化試料ガス回収管(符号42、図4参照)との接続に適した形状を有する。当該形状は、イオン化試料を質量分析計のイオン化試料ガス回収管(符号42)に導入するのに適した形状とするために、当該側面の中心部が段階的にくり抜かれた形状を有する。
具体的には、イオン源側(左側面側)方向へ向かって2mmの位置まで、円柱外側の厚さ4mmの外縁を残して内側が略鈍角円錐状に、そこからさらに同方向へ1mm(当該側面端から3mm)の位置までは垂直方向に、さらにそこから同方向へ1mm(当該側面端から4mm)の位置までは、内側が略鈍角円錐状になるように、中心部が段階的にくり抜かれた形状を有する。
当該略鈍角円錐曲面の中心には、内径2mmのイオン化試料ガス放出口(符号7、図3C参照)が穿ってある。
当該イオン化試料ガス放出口(符号7)は、イオン源側(左側面側)方向へ内径2mmのイオン化試料ガス用流路(符号5、図1及び2参照)が水平に穿ってある。当該流路は、上記励起ガス-試料ガス混合室(符号4)に連通してなる。
当該連結デバイスでは、励起ガス導入口(符号2)から質量分析計(右側面側)方向へ11mm(イオン源側の端部から18mm、試料ガス導入口の中心から質量分析計方向へ8mm)の位置には、円柱側面の上側に内径4mmの外気導入口(符号8、図3E参照)を穿ってある。
当該外気導入口(符号8)は、当該円柱中心方向(垂直方向)へ、内径4mmの外気導入流路(符号9)が直進するように垂直に穿ってある。当該流路は、水平方向に穿ってある上記イオン化試料ガス用流路(符号5、図1及び2参照)に連通してなる。
本実施例で用いた質量分析装置(符号21、図4参照)は、イオン源(符号31)としてDART-SVP(ionSense社製)、質量分析計(符号41)としてmicroOTO-QIII、及び前記連結デバイス(符号1)、を主たる構成要素としてなる質量分析装置である(図4参照)。
ここで、質量分析計のイオン化試料ガス回収管(符号42)は、外径6.2mm、内径4.7mm、及び長さ94mmのセラミックス製チューブであって、当該連結デバイスとの接続側から35mmの領域に、φ0.26mmのニクロム線(抵抗発熱線)を巻きつけてなるものを採用した。また、当該質量分析計の底面には、排気管(符号44)が接続されてなる。当該排気管の先には、真空ポンプ(符号45)が接続されている。
当該連結デバイスの具体的な装着態様は、図4に示した通りである。具体的には、当該連結デバイスの励起ガス導入口(符号2、図1~3参照)を、DART-SVP側の励起ガス噴出ノズル(符号32)の先端に接続してなる。また、当該連結デバイスのイオン化試料ガス放出口(符号7、図1~3参照)を、質量分析計のイオン化試料ガス回収管(符号42)に接続してなる。これらの接続は、パッキン等を噛ませない態様で部材の接続部どうしを噛ませて接続した。
また、当該連結デバイスの試料ガス導入口(符号3)は、試料ガス取り込み管(符号51)を介して試料バイアル瓶(符号52)と接続した。当該バイアル瓶は、樹脂チューブ(符号53)を介して、ヘリウムガスを供給する装置(符号54)と接続した。
本実施例では、以下の揮発性物質を分析に供した。これらの化合物は香気成分として知られる物質である。
下記(式1)で示す構造を有する。粉末1mg/1.5mL容バイアル瓶を分析試験に供した。
下記(式2)で示す構造を有する。粉末5mg/1.5mL容バイアル瓶を分析試験に供した。
下記(式3)で示す構造を有する。上記2つの化合物に比べて、特にイオン化が誘起されにくく検出し難い化合物である。粉末1mg/1.5mL容バイアル瓶を分析試験に供した。
上記(2)に記載の「感度増強用連結デバイスを接続した質量分析装置(本発明:符号2)」を用いて、揮発性物質の検出を行った。
1.5mL容の試料バイアル瓶(符号52)に、上記(3)に記載のいずれかの揮発性物質(Cumarin、Geraniol、又はVanillin)を、上記(3)に記載の量だけ添加し封入した。
イオン源であるDART-SVPのガスヒーターを400℃に設定し、当該イオン源のノズル(符号32)から励起ヘリウムガスを噴出させ、当該連結デバイス中の励起ガス-試料ガス混合室(符号4、図1~3参照)に励起ヘリウムガスを導入した。
試料バイアル瓶(符号52)に、流量0.5L/min.にてヘリウムガスをパージし、当該連結デバイス中の励起ガス-試料ガス混合室(符号4、図1~3参照)に、揮発させた試料ガスを導入した。
質量分析計(符号41)のイオン化試料ガス回収管(加熱部付のセラミックチューブ:符号42)は、25Vの電圧をかけて加熱することで、当該イオン化試料ガス回収管へのイオン化試料の吸着を防止した。当該質量分析計に取り込まれたイオン化試料ガスは、そのまま直進して当該分析計中のTOF型分析部(符号43)に導入されるが、それ以外の気体(イオン化試料以外の気体)は、当該分析部の底面に接続された排気管(符号44)を通して、真空ポンプ(符号45)で引かれて排気される。
microOTO-QIIIの分析モードをポジティブイオンモードに設定して、マスクロマトグラムを得た。
当該対照装置の構造を図5に示した。具体的には、イオン源であるDART-SVPの励起ガス噴出ノズル(符号32)と、質量分析計であるmicroOTO-QIIIのイオン化試料ガス回収管(符号42)との間に、試料バイアル瓶(符号52)を開封して設置し、そのまま分析を行った(図5参照)。
また、Geraniolを試料バイアル瓶に封入して当該連結デバイスを用いた装置で分析した結果を図8Aに示した(試験1-3)。当該連結デバイスを用いない装置での分析結果を図8Bに示した(試験1-4)。
また、Vanillinを試料バイアル瓶に封入して当該連結デバイスを用いた装置で分析した結果を図9Aに示した(試験1-5)。当該連結デバイスを用いない装置での分析結果を図9Bに示した(試験1-6)。
その結果、DARTイオン源と質量分析計の間に、本発明に係る連結デバイスを接続して質量分析を行うことによって、Cumarinに由来するピーク(プロトン付加を示すピーク m/z=147)の強度は、対照装置(連結デバイス非装着)を用いた場合に比べて、約3.3倍に増加することが示された(図7A(試験1-1)、図7B(試験1-2)、表1)。
また、Geraniolに由来するピーク(脱水しプロトン付加されたピーク m/z=137、プロトン脱離したピーク m/z=153)の強度は、対照装置(連結デバイス非装着)を用いた場合に比べて、m/z=137のピークが約2.8倍に、m/z=153のピークは約7.0倍に、それぞれ増加することが示された(図8A(試験1-3)、図8B(試験1-4)、表1)。
また、Vanillinに由来するピーク(脱水しプロトン付加されたピーク m/z=153)の強度は、対照装置(連結デバイス非装着)を用いた場合に比べて、約9.0倍に増加することが示された(図9A(試験1-5)、図9B(試験1-6)、表1)。
当該感度増強作用は、(i)当該連結デバイスの励起ガス-試料ガス混合室において、イオン化前の揮発性物質ガスを一カ所に集中させて励起ガスと混合し、イオン化を高効率に誘起させること、及び、(ii)当該イオン化された揮発性物質ガスを、拡散させることなく効率良く質量分析計に送り込ませること、によって達成される作用であると推測された。
本発明に係る質量分析装置用連結デバイスにおいて、外気導入機構が奏する作用効果を検証した。
実施例1(1)にて作製した連結デバイスを、外気導入機構(符号13)を備えた連結デバイス(部材2-1)として準備した。
一方、外気導入機構(符号13)を有さない構造であることを除いては、実施例1(1)に記載の連結デバイスと同じ構造の連結デバイス(部材2-2)を作製した。当該連結デバイスは、外気導入口(符号8)及び外気導入用流路(符号9)が存在しないため、イオン化試料ガス用流路(符号5)がそのままイオン化試料ガス放出口(符号7)に直結する構造を有するものである。
実施例1(2)に記載の方法と同様にして、上記(1)で準備した連結デバイス(部材2-1、部材2-2)を、それぞれ質量分析装置に接続した。そして、実施例1(4)に記載の方法と同様にして、揮発性物質の検出を行った。検出対象の揮発性物質としては、特にイオン化が誘起されにくく検出し難いVanillin(C8H8O3、Mw=152、CAS 121-33-5 和光純薬工業製)を検出対象とした。
分析条件は、実施例1(4)と同様にして行い、マスクロマトグラムを得た。部材2-1を用いた装置での分析結果を図10Aに示した(試験2-1)。部材2-2を用いた装置での分析結果を図10Bに示した(試験2-2)。
その結果、外気導入機構を備えた連結デバイス(部材2-1)を接続して質量分析を行った場合、Vanillinに由来するピーク(脱水しプロトン付加されたピーク m/z=153)の強度は、外気導入機構を有さない連結デバイス(部材2-2)を接続した場合と比べて、約2.4倍という高い値を示した(図10A(試験2-1)、図10B(試験2-2)、表2)。
なお、当該感度増強作用は、(i)当該連結デバイス及び装置全体での圧力制御が安定することにより、流速が安定し、励起ガス-試料ガス混合空間(符号4)での試料ガスのイオン化反応が安定して進行すること、及び、(ii)取り込まれた外気(大気ガス)に含まれる水分子が、励起ガス-試料ガス混合空間(符号4)を通過した未反応の試料ガスを、イオン化試料ガス用流路(符号5)内にてさらにイオン化させること、によって発揮される作用であると推測された。
ここで、試験1-5(部材2-1に相当)と試験1-6(従来技術)の差異である「約9.0倍」という値は、実施例2の試験2-1(部材2-1)と試験2-2(部材2-2)の差異である「約2.4倍」よりも顕著に大きな差異であると認められた。
実施例2にて示された外気導入機構の感度増強作用について、抽出イオンクロマトグラム(EIC)による検証を行った。
実施例1(1)にて作製した連結デバイスを、「外気導入機構を備えた連結デバイス」(部材3-1)として準備した。一方、実施例2(1)で作製した連結デバイスを、「外気導入機構のない連結デバイス」(部材3-2)として準備した。
上記(1)で準備した連結デバイスのうち、まず「外気導入機構を有さない連結デバイス」(部材3-2)を質量分析装置に接続し、実施例1(4)に記載の方法と同様にして、Vanillin(C8H8O3、Mw=152、CAS 121-33-5 和光純薬工業製)の検出を行った。30秒の測定の後、連結デバイスを「外気導入機構を備えた連結デバイス」(部材3-1)に付け替えて30秒の測定を行った。
分析条件は、実施例1(4)と同様にして行い、m/z=153に注目したスペクトル変化のマスクロマトグラムを得た。得られたマスクロマトグラムを図11に示した。
その結果、「外気導入機構のない連結デバイス」(部材3-2)を接続した際に検出されていたm/z=153のピーク(vanillin由来ピーク)は、「外気導入機構を備えた連結デバイス」(部材3-1)に付け替えることで、約2倍以上に増強して検出されることが示された。当該EICの結果は、実施例2の分析結果を裏付けるものであった。
本発明に係る感度増強用連結デバイスを備えた質量分析装置を用いて、標品試薬でなく、実際の市販食品から揮発する物質の高感度検出を行った。具体的には、2種類の市販食品から揮発する物質の差異を検出した。
実施例1(1)にて作製した連結デバイスを、外気導入機構(符号13)を備えた連結デバイスとして準備した。
本実施例では、2種類のチョコレートを分析対象として試験に供した。
市販ブラックチョコレート30mgを1.5mL容バイアル瓶に量り取り、室温にて分析試験に供した。
市販ミルクチョコレート30mgを1.5mL容バイアル瓶に量り取り、室温にて分析試験に供した。
実施例1(2)に記載の方法と同様にして、上記(1)で準備した連結デバイスを質量分析装置に接続した。そして、実施例1(4)に記載の方法と同様にして、揮発性物質の検出を行った。分析対象としては、上記(2)の各チョコレートを対象とした。
分析条件は、実施例1(4)と同様にして行い、マスクロマトグラムを得た。ブラックチョコレートの分析結果を図12Aに示した(試験4-1)。ミルクチョコレートの分析結果を図12Bに示した(試験4-2)。
その結果、DARTイオン源と質量分析計の間に本発明に係る連結デバイスを接続して質量分析を行うことによって、室温状態で保管されているブラックチョコレートとミルクチョコレートからの揮発性物質の差異を、明確に検出できることが示された。
具体的には、ブラックチョコレートからの揮発成分は、ミルクチョコレートからの揮発成分よりも、m/z=137のピーク(Limonene由来ピーク、及び、2,3,5,6-tetramethylpyrazine由来のピーク)に対して、m/z=153のピーク(Vanillin由来ピーク)が低く検出されることが示された。
食品を摂取した際の口腔内の状態を再現し、本発明に係る感度増強用連結デバイス(連結デバイス)を備えた質量分析装置を用いて放出される揮発性物質の経時的変化を測定した。本実施例では、スペアミントチョコレートを分析対象として試験に供し、口腔内の状態を再現した際に放出される揮発性物質の経時変化を測定した。
実施例1(1)にて作製した連結デバイスを、「外気導入機構を備えた連結デバイス」として準備した。
市販のチョコレートにスペアミントを配合し、スペアミントチョコレートを調製した。当該スペアミントチョコレート約30mgを20mL容バイアル瓶に量り取り、純水0.5mLを添加して、湯煎で溶解させた場合の揮発性物質の挙動をリアルタイムで分析した。
上記(1)で準備した連結デバイスを質量分析装置に接続し、バックグラウンド測定(測定5-1)を行った。その後室温状態での分析試料からの揮発性物質の測定を行い(測定5-2)、次いで湯煎で分析試料を溶解させながらの測定(測定5-3)を行った。なお、一連の測定操作は、リアルタイムにて連続的に行った。
分析条件は、実施例1(4)と同様にして行い、TIC(トータルイオンクロマトグラム)のスペクトル変化、m/z=137(Limonene由来ピーク)に注目したスペクトル変化、及び、m/z=151(Carvone由来ピーク)に注目したスペクトル変化について、それぞれのマスクロマトグラムを得た。結果を図13に示した。
その結果、スペアミントチョコレートを湯煎で溶解し始めた時(測定5-3の始期)と同じタイミングで、m/z=137のピーク(Limonene由来ピーク)とm/z=151のピーク(Carvone由来ピーク)が、急激に上昇することが検出された。ここで、Limonene及びCarvoneはスペアミントに含まれる揮発性化合物である。
食品を摂取した際の口腔内の状態を再現し、本発明に係る感度増強用連結デバイスを備えた質量分析装置を用いて放出される揮発性物質の経時的変化を測定した。本実施例では、オレンジ風味クッキーを分析対象として試験に供し、口腔内の状態を再現した際に放出される揮発性物質の経時変化を測定した。
実施例1(1)にて作製した連結デバイスを、「外気導入機構を備えた連結デバイス」として準備した。
市販オレンジ風味クッキー約0.5gをそのまま20mL容バイアル瓶に入れ、容器内にて粉砕した場合における揮発性物質の挙動をリアルタイムで分析した。
上記(1)で準備した連結デバイスを質量分析装置に接続し、バックグラウンド測定(測定6-1)を行った。その後室温状態での分析試料からの揮発性物質の測定を行い(測定6-2)、次いで容器内にてクッキーを粉砕しながら測定(測定6-3)を行った。なお、一連の測定操作は、リアルタイムにて連続的に行った。
分析条件は、実施例1(4)と同様にして行い、TIC(トータルイオンクロマトグラム)のスペクトル変化、及び、m/z=137(Limonene由来ピーク)に注目したスペクトル変化について、それぞれのマスクロマトグラムを得た。結果を図14に示した。
その結果、オレンジ風味クッキーを容器内で粉砕し始めた時(測定6-3の始期)と同じタイミングで、m/z=137のピーク(Limonene由来ピーク)が急激に上昇することが検出された。ここで、Limoneneはオレンジ風味成分に含まれる揮発性化合物である。
実施例1(1)で作製した連結デバイスとは異なる形状の感度増強用連結デバイスを作製した。これらの形状の外観の写真像を、図21A及び図21Bに示した。
また、質量分析装置に接続するためのアダプター部材に装着した状態の写真像を、図22A及び図22Bに示した。
特に、食品等の物性変化試験及び状態変化試験、有機化合物の合成反応過程、及び製造工程等、これまでは揮発性物質のリアルタイムモニタリングが不可能であった分野において、利用させることが期待される。

Claims (8)
- 大気圧下リアルタイム質量分析装置に接続するインターフェイス部材であって、
(A)励起ガス導入口、試料ガス導入口、及びイオン化試料ガス放出口を有し、
(B)前記励起ガス導入口と前記イオン化試料ガス放出口とが連通してなる流路を有し、
(C)前記試料ガス導入口と前記(B)に記載の流路が連通する構造を有することにより、前記(B)に記載の流路の一部の領域に、励起ガスと試料ガスを混合するための空間が形成されてなる、
ことを特徴とする、質量分析装置用連結デバイス。 - 前記質量分析装置用連結デバイスが、
DART法の原理を利用したイオン源の励起ガス噴出口と、質量分析計のイオン化試料ガス回収口と、の間に接続するためのインターフェイス部材である、
請求項1に記載の質量分析装置用連結デバイス。 - 前記(C)に記載の空間が、
前記(B)に記載の流路の直管形状の流路部分に形成された空間である、

請求項5に記載の質量分析装置用連結デバイス。 - 請求項1~6のいずれかに記載の質量分析装置用連結デバイスを備えた、大気圧下リアルタイム質量分析装置。
- 請求項7に記載の大気圧下リアルタイム質量分析装置を用いることを特徴とする、大気圧下リアルタイムでの揮発性物質の質量分析法。
