Abstract
Description
さらに、以上の結果から、精子ゲノムDNAにおいて、上記母性インプリント遺伝子のアレル特異的メチル化領域が高度にメチル化されている場合には、本来機能すべき父方アレルの遺伝子が機能できないので、該精子由来の子供はインプリント異常症を発症する可能性が考えられる。そこで、本願発明者は、上記母性インプリント遺伝子のアレル特異的メチル化領域のメチル化異常と、インプリント異常症との関連を調べるために、シルバーラッセル症候群(SRS)患者及びベックウィズ・ヴィーデマン症候群(BWS)患者由来の血液からゲノムDNAを抽出して、DIRAS3、NAP1L5、FAM50B、GRB10、INPP5Fv2、RB1、ZNF597、ZNF331、PSIMCT-1、NNAT、L3MBTL、NESPAS、及びGNAS1Aののアレル特異的メチル化領域におけるメチル化状態を解析した。その結果、SRS患者では、GRB10、ZNF597、INPP5Fv2、ZNF331、及びFAM50Bのメチル化異常が認められること、また、BWS患者では、ZNF331のメチル化異常が認められることが明らかとなった。これらの結果から、上記母性インプリント遺伝子のアレル特異的メチル化領域におけるメチル化異常がインプリント異常症の発症に関与することが強く示唆された。
以上のように、本願発明者は、DIRAS3、NAP1L5、FAM50B、GRB10、INPP5Fv2、RB1、ZNF597、ZNF331、PSIMCT-1、NNAT、L3MBTL、NESPAS、及びGNAS1Aが母性インプリント遺伝子であること、さらに、これらの遺伝子のアレル特異的メチル化領域におけるメチル化異常がインプリント異常症の発症に関与することをはじめて見い出し、本発明を完成するに至った。

したがって、本発明の評価方法においては、上記工程(b)によって、被検者の精子DNAゲノムにおける、本発明の母性インプリント遺伝子群から選択される1又は2以上のアレル特異的メチル化領域に含まれるCpG配列のシトシン残基のメチル化の程度が高いという測定結果が得られた場合には、該被検者の子孫がインプリント異常症を発症するリスクが存在する、或いは、リスクが高いと評価することができる。一方、上記工程(b)によって、被検者の精子DNAゲノムにおける、本発明の母性インプリント遺伝子群から選択される1又は2以上のアレル特異的メチル化領域に含まれるCpG配列のシトシン残基のメチル化の程度が低いという測定結果が得られた場合には、該被検者の子孫がインプリント異常症を発症するリスクが低いと評価することができる。ここで、「メチル化の程度」とは、上記工程(b)において、測定の対象とされたアレル特異的メチル化領域に含まれる、全CpG配列数に対するメチル化CpG配列数の割合(メチル化率)を意味し、「メチル化の程度が高い」とは、例えば、メチル化率が50%以上、より好ましくは70%以上、さらに好ましくは90%以上であることを意味し、また、「メチル化の程度が低い」とは、例えば、メチル化率が20%以下、より好ましくは10%以上、さらに好ましくは5%以下であることを意味する。
20の正常個体由来の精子及び血液サンプルを用いて、これまでに報告されている22のgDMR(ヒト生殖細胞におけるDNAメチル化差異領域;germline Differentially Methylated Region)、すなわち、ZDBF2、H19、GTL2、ZAC、PEG1、LIT1、SNRPN、LINE-1、Alu、DIRAS3、NAP1L5、FAM50B、GRB10、INPP5Fv2、RB1、ZNF597、ZNF331、PSIMCT-1、NNAT、L3MBTL、NESPAS、及びGNAS1Aにおける一塩基多型(SNP)を解析した。また、繰り返し配列(LINEI及びAlu)についても併せて解析した。さらに、精子及び血液サンプル由来のゲノムDNDにおける、上記22のgDMRのメチル化状態をバイサルファイトシークエンス法によって確認した。表1に、解析した遺伝子又は領域名と、SNP解析及びバイサルファイトシークエンス法に用いたプライマー配列をそれぞれ示す。
ラッセルシルバー症候群(SRS)患者においては、H19の低メチル化が認められることが知られており、さらに、他の様々な領域におけるメチル化がSRSに関与する可能性が示唆されている。本実施例においては、H19のメチル化異常が既に確認されている、15例のSRS患者(5例のARTにより出生したSRS患者、及び10例の自然生殖によって出生したSRS患者)から血液を採取してゲノムDNAを抽出し、3種類の父性DMR(ZDBF2、H19、GTL2)、及び19種類の母性DMA(DIRAS3、NAP1L5、FAM50B、ZAC,GRB10、PEG10、PEG1、INPP5Fv2、LIT1、RB1、SNRPN、ZNF597、ZNF331、PEG3、PSIMCT-1、NNAT、L3MBTL、NESPAS、GNAS1A)のメチル化状態を解析した。
ベックウィズ・ヴィードマン症候群(BWS)患者においては、H19の高メチル化、又はLIT1の低メチル化が認められることが報告されている。本実施例においては、LIT1のメチル化異常が既に確認されている、7例のBWS患者(1例のARTにより出生したBWS患者、及び6例の自然生殖によって出生したBES患者)から血液を採取して、ゲノムDNAを抽出し、3種類の父性DMR(ZDBF2、H19、GTL2)、及び19種類の母性DMA(DIRAS3、NAP1L5、FAM50B、ZAC,GRB10、PEG10、PEG1、INPP5Fv2、LIT1、RB1、SNRPN、ZNF597、ZNF331、PEG3、PSIMCT-1、NNAT、L3MBTL、NESPAS、GNAS1A)のメチル化状態を解析した。
Claims (35)
- 以下の工程(a)及び(b)を含むことを特徴とする、被検者の子孫がインプリント異常症を発症するリスクを評価する方法。
(a)前記被検者より採取された精子からゲノムDNAを抽出する工程;
(b)前記工程(a)により抽出されたゲノムDNAにおける、DIRAS3、NAP1L5、FAM50B、GRB10、INPP5Fv2、RB1、ZNF597、ZNF331、PSIMCT-1、NNAT、L3MBTL、NESPAS、及びGNAS1Aからなる群より選択される1又は2以上の母性インプリント遺伝子のアレル特異的メチル化領域に含まれるCpG配列のシトシン残基のメチル化の程度を測定する工程; - DIRAS3のアレル特異的メチル化領域が、GenBankアクセッション番号AF202543.1(更新日:2001年8月13日)として登録されているヒト第1染色体の塩基配列中の第1849位~第2197位の領域(配列番号1)であることを特徴とする請求項1記載の方法。

iii)配列番号18及び19に示される塩基配列からなる、FAM50Bのアレル特異的メチル化領域を増幅させるためのプライマーセット;
iv)配列番号20及び21に示される塩基配列からなる、GRB10のアレル特異的メチル化領域を増幅させるためのプライマーセット;

xi)配列番号34及び35に示される塩基配列からなる、L3MBTLのアレル特異的メチル化領域を増幅させるためのプライマーセット;
xii)配列番号36及び37に示される塩基配列からなる、NESPASのアレル特異的メチル化領域を増幅させるためのプライマーセット;
xiii)配列番号38及び39に示される塩基配列からなる、GNAS1Aのアレル特異的メチル化領域を増幅させるためのプライマーセット; - 以下のi)~xiii)より選択される1又は2以上のプライマーセットを備えることを特徴とする、被検者の子孫がインプリント異常症を発症するリスクを評価するための検査キット。
i)配列番号14及び15に示される塩基配列からなる、DIRAS3のアレル特異的メチル化領域を増幅させるためのプライマーセット;
ii)配列番号16及び17に示される塩基配列からなる、NAP1L5のアレル特異的メチル化領域を増幅させるためのプライマーセット;
iii)配列番号18及び19に示される塩基配列からなる、FAM50Bのアレル特異的メチル化領域を増幅させるためのプライマーセット;
iv)配列番号20及び21に示される塩基配列からなる、GRB10のアレル特異的メチル化領域を増幅させるためのプライマーセット;
v)配列番号22及び23に示される塩基配列からなる、INPP5Fv2のアレル特異的メチル化領域を増幅させるためのプライマーセット;
vi)配列番号24及び25に示される塩基配列からなる、RB1のアレル特異的メチル化領域を増幅させるためのプライマーセット;
