실험재료인 넓패는 제주도 성산포 연안에서 채집하였다. 채집된 넓패를 수세하여 이물질을 제거한 다음 동결건조한 후 균질하게 분쇄하였다. 분쇄한 넓패 분말 500g 을 80% 에탄올 12 L에 넣어 상온에서 24시간 추출한 후 필터링을 실시하였다. 넓패 에탄올 추출물은 감압농축기 이용하여 유기용매를 제거 후 dimethyl sulfoxide (DMSO)로 녹여 실험에 사용하였으며, DMSO의 최종 농도는 0.01%를 초과하지 않도록 하였다. 생후 3주령인 Wistar 랫트 수컷을 (Japan SLC, Hamamatsu, Janpan) ㈜ 중앙실험동물로부터 구입하여 ethyl ether로 마취 후 경추도살 하였다. 랫트 왼쪽과 오른쪽 mystacial pads를 분리하여 100 units/ml 페니실린-100㎍/㎖ 스트렙토마이신 (Gibco Inc, NY, USA)이 함유된 E/P buffer [Earle's balanced salts solution (EBSS, Sigma MO, USA) + phosphate-buffered saline (PBS, Sigma MO, USA)]에 넣었다. 해부 현미경으로 관찰하며 콧털모낭을 조심스럽게 분리하였다. 모낭이 모두 분리될 때까지 E/P buffer를 넣은 Petri dish에 분리된 모낭을 넣고 37℃, 5% CO2 항온기에서 1시간 정도 배양하였다. 24 well plate의 각 well에 2 mM L-글루타민 (Gibco Inc, NY, USA), 10 ㎍/㎖ 인슐린 (Sigma MO, USA), 50 nM hydrocortisone (Sigma MO, USA)과 100 units/ml 페니실린-100 ㎍/㎖ 스트렙토마이신을 포함하는 500 ㎕의 William E medium (Gibco Inc, NY, USA)을 넣고, 하나의 well에 하나씩의 모낭을 넣어서 37℃, 5% CO2 항온기에서 배양하였다. 한 실험군에 8-12개의 모낭을 이용하였으며, 배양 중에 배지는 3 일마다 교환하였다. 넓패 추출물 (1, 및 10 ㎍/㎖) 및 양성 대조물질인 미녹시딜을 10 μM의 농도로 처리하였다. DHT는 모유두세포와 모낭의 점진적인 축소화를 가져오고 모발 성장을 억제하며, 테스토스테론에서 DHT로의 전환은 5α-환원효소 활성에 의해 일어남이 밝혀져 있다. 이러한 연구 결과로 보아 5α-환원효소 활성 억제를 통한 DHT의 감소가 모발 성장에서 필요함을 말해준다. 따라서 넓패 추출물로부터 분리된 옥타프로레톨 에이(octaphlorethol A)에 모발의 성장에 매우 중요한 역할을 하는 5α-환원효소 억제 효과가 있는지 생후 7~8 주령의 SD rat 전립선의 조효소를 사용하여 조사하였다. TLC plate는 전개용매 (50% 시클로헥산, 50% 에틸 아세테이트)하에서 전개한 후, 건조하였다. UV spectrometer로 254 nm에서 테스토스테론을 확인하고 10% 황산을 뿌려서 DHT를 확인하였다. Radioactivity는 테스토스테론과 DHT 부위를 오려낸 후 10 ml의 Ultima GoldTM cocktail (PerkinElmer, Massachusetts, USA)을 넣어 Liquid scintillation analyzer (Packard Bioscience, Meriden, USA)로 측정하였다. 5α-환원효소 활성에 의한 테스토스테론의 전환율을 [DHT/(테스토스테론+DHT)]×100의 비로 계산하였다.5α-reductase assay was performed as follows.
