Abstract

본 발명은 (a) CSP-B(Cytotoxic Serine Protease-B)의 5'-SAR(Scaffold or Matrix Attachment Region); (b) 동물세포에서 작동하는(operable) 프로모터; 및 (c) 폴리아데닐화 서열을 포함하는 동물세포용 발현 벡터, 그리고 이를 이용하여 재조합 단백질을 제조하는 방법에 관한 것이다. 본 발명의 벡터는 CSP-B 5'-SAR을 포함하는 벡터로, 동물세포에 도입한 외래 유전자의 위치에 따른 유전자 발현 억제 현상을 극복하는 효과를 가지며, 목적 단백질의 발현율을 크게 향상시킨다. 본 발명의 벡터는 의약품용 재조합 단백질 또는 항체를 동물세포에서 효과적으로 발현시킨다. 본 발명의 벡터 및 이를 이용한 재조합 단백질의 제조방법은 의약품의 산업적 대량 생산에 매우 유용하게 사용될 수 있다.
