Abstract
本発明は、がんなどを特異的にX線造影することができ、また、高感度な蛍光標識としても機能することによって高価なPET造影を用いなくてもリアルタイムでの検出を行うことを可能とするバイモーダル造影剤であるX線吸収蛍光ナノ粒子を提供することを課題とする。本発明は、X線吸収ナノ粒子と蛍光ナノ粒子が結合してなる、体積平均粒子径が10nm以上1000nm以下のX線吸収蛍光複合ナノ粒子、および該X線吸収蛍光複合ナノ粒子を含むX線造影剤を提供する。
Description
本発明は、X線造影剤としての機能と、蛍光標識剤としての機能を合わせ持つバイモーダル造影剤として使用できるX線吸収蛍光ナノ粒子に関する。
ナノメートルオーダーの粒子(以下、「ナノ粒子」)は、対応するバルク体とは異なる物性を示すことから近年注目を集めており、造影剤の分野においても、ナノ粒子を用いた造影剤に関する種々の研究がなされている。
X線造影剤への適用に向けた研究として、例えば、特許文献1には、金微粒子と3-メルカプトプロピオン酸とからなる平均粒径2.5nmの金属微粒子複合体を含む水溶液が、高いX線造影効果を示すことが記載されている。
また、非特許文献1には、平均粒径が28±7nm及び38±8nmの金粒子にそれぞれポリエチレングリコール系リンカーを介して抗マウスCD4モノクロナール抗体を結合して得られる金ナノ粒子-抗CD4抗体複合体を調製し、この金ナノ粒子-抗CD4抗体複合体を投与したマウスにおけるX線CTイメージを取得する試みが開示されている。
ところで、近年、光学分野からバイオ分野にわたる様々な分野において、近年量子ドットが注目されている。量子ドットは、半導体材料からなるナノ粒子であり、多くの場合、第1の半導体からなるコアと、このコアを取り囲む第2の半導体からなるシェルとからなる粒子である。そのため、量子ドットは「半導体ナノ粒子」と呼ばれる場合もある。この量子ドットは、量子サイズ効果(粒子が小さくなるにつれて材料中の電子の状態が変わって、より短い波長の光を吸収したり放出したりする現象)により、バルクの半導体とは異なる波長の光を吸収・発光する性質を有しており、従来の有機蛍光剤と比べて発光量が大きく、発光安定性にも優れ、さらに、物理サイズによって発光波長を変えることができるなどの利点がある。
量子ドットのこのような性質に注目して、量子ドットを用いた蛍光標識剤についての研究開発が種々されている。例えば、特許文献2には、平均粒子径1~5nmのCdSeを蛍光性半導体量子ドットにレクチンが結合してなる細胞標識用蛍光プローブが開示されている。
また、蛍光造影剤への適用に向けた研究として、非特許文献2には、コアシェルセレン化カドミウム-硫化亜鉛量子ドットにポリエチレングリコールを結合させてなる複合体を投与したマウスにおける蛍光画像イメージを取得する試みが開示されている。
しかしながら、蛍光性能を付与したX線造影剤については、今までに報告はない。

医療現場において、X線診断で得られた画像は、手術を行う際に広く活用されている。特にがんの治療においては、空間分解能に優れるというメリットを活用して、切除すべき組織範囲の決定や治療効果の判定のためにX線CTスキャンによる画像診断が多用されている。近年では、手術現場において、X線CTスキャンやMRIによる診断をリアルタイムで行いながら手術を進行させる手法が広まりつつあり、さらに、MRXO(Magnetic resonance X-ray Operation suite)などの特殊な手術システムを用いて、X線CT装置やMRI装置など複数の検査装置を組み合わせて診断を行いながら手術を進行させる手法も開発されている。
ただ、X線を用いた画像診断には、X線被爆による副作用や後遺症のおそれがあるという欠点もあることから、X線撮影の頻度には自ずと限界が生じてしまう。したがって、X線CT画像に対応するデータを、手術の現場においてX線撮影を行うことなく取得可能な手法が切望されている。
がんの診断方法として、陽電子が通常の電子とが結合して消滅する際に放出される消滅放射線を検出し、画像化する手法であるポジトロン断層撮影(PET)に基づく検査方法も用いられている。これは、がんが正常細胞の3~8倍もの糖代謝を行っていることに着目し、2-デオキシ-18F-フルオロ-D-グルコース(FDG)など糖代謝を反映するPET薬剤を投与し、その集積度によってがんの所在を評価しようとする方法である。このPETには、X線CTスキャンと比べると被爆のリスクが少ないという利点がある一方で、FDGなどのPET薬剤が高価であることに起因するコスト上の制約もある。
そこで、本発明は、このような問題に対し、がんなどを特異的にX線造影することができ、また、高感度な蛍光標識としても機能することによって高価なPET造影を用いなくてもリアルタイムでの検出を行うことを可能とするバイモーダル造影剤であるX線吸収蛍光ナノ粒子を提供することを課題とする。
本発明者は、X線造影剤において、X線造影能を有するナノ粒子に、蛍光標識剤からなるナノ粒子を結合させることにより、上記の課題を解決しうることを見出し、本発明に至った。
すなわち、本発明は、下記[1]~[9]に示されるX線吸収蛍光複合ナノ粒子およびX線造影剤を提供する。

本発明のX線吸収蛍光複合ナノ粒子は、がん等の病巣を特異的に造影するX線CT造影剤として機能するとともに、蛍光標識剤としても機能するX線造影剤を提供する。
そして、本発明のX線吸収蛍光複合ナノ粒子を含むX線造影剤によって、X線CTなどを用いて体外から観察可能なX線と、感度の高い蛍光との2つのモードでの造影が可能となることから、手術等の処置を行う際に高価なPET造影を用いなくてもリアルタイムに病巣の位置・大きさ・範囲を特定することが可能となり、安価に診断および処置を行うことが可能となる。
そして、本発明のX線吸収蛍光複合ナノ粒子を含むX線造影剤によって、X線CTなどを用いて体外から観察可能なX線と、感度の高い蛍光との2つのモードでの造影が可能となることから、手術等の処置を行う際に高価なPET造影を用いなくてもリアルタイムに病巣の位置・大きさ・範囲を特定することが可能となり、安価に診断および処置を行うことが可能となる。
〔X線吸収蛍光複合ナノ粒子〕
本発明に係るX線吸収蛍光複合ナノ粒子は、X線吸収ナノ粒子と蛍光ナノ粒子が結合してなる粒子である。
本発明に係るX線吸収蛍光複合ナノ粒子は、X線吸収ナノ粒子と蛍光ナノ粒子が結合してなる粒子である。
本発明のX線吸収蛍光複合ナノ粒子のサイズは、体積平均粒子径として10nm以上1000nm以下、好ましくは40nm以上200nm以下、より好ましくは50nm以上200nm以下、さらに好ましくは40nm以上100nm以下である。10nmよりも小さいと腎臓より速やかに排出されてしまうため、血中滞留時間が短く、がんへの選択吸着が行われないおそれがある。一方、1000nmよりも大きいと、毛細血管への詰まりが発生することがあり、投与時に生体毒性を持つ場合があるため、好ましくない。ここで体積平均粒子径は、動的光散乱法により測定することができる。
本発明のX線吸収蛍光複合ナノ粒子は、蛍光ナノ粒子からの蛍光発光が効率的にX線吸収蛍光複合ナノ粒子外に放出できるよう、X線吸収ナノ粒子をコアとして、このコアを蛍光ナノ粒子がシェルとして取り囲むコア-シェル構造を有していると好ましい。したがって、この蛍光ナノ粒子とX線吸収ナノ粒子との粒子個数比率は、1:1~10000:1であることが好ましく、10:1~1000:1であることがより好ましい。
ここで、X線吸収ナノ粒子と蛍光ナノ粒子との粒子個数比率は、下記式(1)で表される比として求めることができる。
上記式(1)中、MaおよびMbは、本発明のX線吸収蛍光複合ナノ粒子における、X線吸収ナノ粒子を構成する化合物(以下、「X線吸収ナノ粒子構成化合物」ともいう。)および蛍光ナノ粒子を構成する化合物(以下、「蛍光ナノ粒子構成化合物」ともいう。)の含有量をそれぞれ表し、ρaおよびρbは、X線吸収ナノ粒子構成化合物および蛍光ナノ粒子構成化合物の密度をそれぞれ表し、daおよびdbは、動的散乱法により測定したX線吸収ナノ粒子および蛍光ナノ粒子構成化合物の平均粒子径をそれぞれ表す。
X線吸収ナノ粒子
本発明のX線吸収蛍光複合ナノ粒子を構成するX線吸収ナノ粒子は、X線CT造影剤としての機能を生じさせるものである。
このX線吸収ナノ粒子は、X線を吸収することができるものであれば特にその構成元素に限りはないものの、X線の吸収性能が高い元素で構成されていることが好ましい。一般に原子番号が大きい元素であるほどX線の吸収性能が高い傾向にあることから、原子番号が大きい元素で構成されていることが好ましい。X線吸収ナノ粒子を構成する元素として、具体的には、周期律表の第4周期以上に属する元素が挙げられる。X線吸収性能が高く、X線造影を実用的に行うことが可能であることから、周期律表の第5周期以上、すなわち、原子番号でおおよそ40以上の元素が、X線吸収ナノ粒子を構成する元素として好適に用いられる。このような元素の好適な具体例として、Ag,I,Ba,Au,Bi,Gdなどが挙げられる。この中で、Au、I、Gdが、原子のX線減弱係数からみて、X線撮影に使用される20KeV~80KeVの領域におけるX線吸収性能が高いことから、好ましい。
ここで、X線吸収ナノ粒子と蛍光ナノ粒子との粒子個数比率は、下記式(1)で表される比として求めることができる。
X線吸収ナノ粒子
本発明のX線吸収蛍光複合ナノ粒子を構成するX線吸収ナノ粒子は、X線CT造影剤としての機能を生じさせるものである。
このX線吸収ナノ粒子は、X線を吸収することができるものであれば特にその構成元素に限りはないものの、X線の吸収性能が高い元素で構成されていることが好ましい。一般に原子番号が大きい元素であるほどX線の吸収性能が高い傾向にあることから、原子番号が大きい元素で構成されていることが好ましい。X線吸収ナノ粒子を構成する元素として、具体的には、周期律表の第4周期以上に属する元素が挙げられる。X線吸収性能が高く、X線造影を実用的に行うことが可能であることから、周期律表の第5周期以上、すなわち、原子番号でおおよそ40以上の元素が、X線吸収ナノ粒子を構成する元素として好適に用いられる。このような元素の好適な具体例として、Ag,I,Ba,Au,Bi,Gdなどが挙げられる。この中で、Au、I、Gdが、原子のX線減弱係数からみて、X線撮影に使用される20KeV~80KeVの領域におけるX線吸収性能が高いことから、好ましい。
また、これらの元素は、生体内での安定であり且つ毒性をほとんどまたは全く示さない限りにおいて、単体として存在していてもよく、対応する対イオンとの塩として存在していてもよく、あるいは、有機化合物の構成元素として存在していてもよい。これらの元素の単体、ならびに、これらの元素から構成される塩および有機化合物は、1種単独で、あるいは2種以上を組み合わせて用いることができる。また、これらの元素の単体は、2種以上の元素からなる合金であってもよい。
また、後述する蛍光ナノ粒子と結合させて得られるX線吸収蛍光複合ナノ粒子が上記の体積平均粒径を有するよう、本発明で用いられるX線吸収ナノ粒子は、体積平均粒径が2nm以上1000nm以下であることが好ましく、20nm以上200nm以下であることがより好ましい。ここで、後述する蛍光ナノ粒子と結合するために用いられる反応性官能基を有する適当なリンカーを結合させたX線吸収ナノ粒子について測定した体積平均粒径を、ここにいうX線吸収ナノ粒子の粒径として採用してもよい。
本発明で用いられるX線吸収ナノ粒子の製造方法は、特に限定されるものではないが、粒径分布の制御が可能である点で、対応する前駆物質から液相反応を通じて得る方法が好適な例として挙げられる。
貴金属からなるX線吸収ナノ粒子は、水溶性貴金属化合物を適当な還元剤によって還元することにより得ることができる。ここで、還元反応の際、所要により適当なpH調整剤を添加してもよい。
例えば、金からなるX線吸収ナノ粒子は、塩化金酸あるいはその塩を、クエン酸、アスコルビン酸、水素、ホルムアルデヒド、エタノール、タンニン酸、ジボラン、水素化ホウ素塩などの適当な還元剤を用いて還元することにより得ることができる。ここで、還元剤としてクエン酸を用いると、得られる貴金属の粒子を好適に分散させることもできるので好ましい。
また、酸化物からなるX線吸収ナノ粒子は、対応する水溶性塩を直接酸化物に変化させることにより、あるいは、対応する水溶性塩を一旦水酸化物、炭酸塩などの中間体に変換してから加熱して酸化物に導くことにより得ることができる。例えば、酸化ガドリニウムなどの希土類酸化物からなるX線吸収ナノ粒子は、硝酸ガドリニウムなどの水溶性希土類塩の水溶液に尿素を入れて加熱し、尿素の加水分解によって生成する炭酸イオンと水酸化物イオンによって希土類元素を炭酸塩と水酸化物の混合物として沈殿させ、その後、この混合物を焼成することにより得ることができる。
また、ヨウ素など電気陰性な元素から構成されるX線吸収ナノ粒子としては、AgI等からなるX線吸収ナノ粒子が挙げられる。ここで、そのようなX線吸収ナノ粒子を得る方法は特に限定されるものではないが、例えば、均質な沈殿を得やすく粒径制御性にも優れる観点から均一沈殿法等の方法を用いて得ることができる。
蛍光ナノ粒子
本発明のX線吸収蛍光複合ナノ粒子を構成する蛍光ナノ粒子は、蛍光標識剤としての機能を生じさせるものである。本発明のX線吸収蛍光複合ナノ粒子がX線造影剤として用いて得られたX線画像に基づき手術を行う際、この蛍光ナノ粒子は、生体内に取り込まれたX線造影剤の分布を、蛍光、具体的には近紫外領域から近赤外領域にかけての範囲内で発光される蛍光の形で特定する役割を果たす。
本発明のX線吸収蛍光複合ナノ粒子を構成する蛍光ナノ粒子は、蛍光標識剤としての機能を生じさせるものである。本発明のX線吸収蛍光複合ナノ粒子がX線造影剤として用いて得られたX線画像に基づき手術を行う際、この蛍光ナノ粒子は、生体内に取り込まれたX線造影剤の分布を、蛍光、具体的には近紫外領域から近赤外領域にかけての範囲内で発光される蛍光の形で特定する役割を果たす。
この蛍光ナノ粒子は、本発明のX線吸収蛍光複合ナノ粒子が生体内に取り込まれた状態において、X線吸収蛍光複合ナノ粒子が取り込まれていない領域との識別が可能な態様で、蛍光発光を生じうるものである限り、構成材料として用いられる蛍光材料に特に限りはなく、例えば、シリカナノ粒子などの適当な担体に有機蛍光色素を担持して得られる粒子を蛍光ナノ粒子として用いることを妨げるものではない。ただ、本発明において、蛍光ナノ粒子は、発光強度高く、耐久性高い半導体ナノ粒子からなるものが好ましい。蛍光ナノ粒子として好適に用いうる半導体ナノ粒子を構成する半導体として、II-VI族半導体およびIII-V族半導体、並びに、珪素またはゲルマニウムからなる半導体が挙げられる。ここで、II族元素として、マグネシウム,亜鉛,カドミウム,水銀が、III族元素として、アルミニウム,ガリウム,インジウムが、V族元素として、窒素,リン,ヒ素,アンチモンが、VI族元素として、酸素,硫黄,セレン,テルルがそれぞれ挙げられる。上記「II-VI族半導体」の例としては、ZnS,ZnSe,ZnTe,CdS,CdSe,CdTeなどが挙げられる。また、「III-V族半導体」の例としては、AlN,AlP,GaN,GaP,InN,InPなどが挙げられる。さらに、II-VI族半導体およびIII-V族半導体として、3つ以上の元素からなるものを用いてもよい。本発明で用いられる半導体ナノ粒子において、これらの半導体を、1種単独で用いてもよく、あるいは、2種以上を組み合わせて用いてもよい。
なお、用語「X-Y族半導体」は、X族元素とY族元素とを組み合わせてなる半導体を指すものであり、例えば、「II-VI族半導体」は、II族元素とVI族元素とを組み合わせてなる半導体を意味する。
また、本発明で蛍光ナノ粒子として用いうる半導体ナノ粒子は、単成分の第1の半導体からなるナノ粒子をコアとし、その表面に、第2の半導体からなるシェルをコートした構造を有するコアシェル型半導体ナノ粒子であることが好ましい。このような半導体ナノ粒子を用いると、発光効率が向上するとともに、化学的安定性や耐光性が高くなることから有利である。ここで、本発明においては、コアシェル型半導体ナノ粒子を構成する第1の半導体は、上記「II-VI族半導体」または「III-V族半導体」であることが好ましい。また、シェルを構成する第2の半導体については、コアを構成している半導体とは異なる半導体を用いることができる。ただ、本発明で用いられるコアシェル型半導体ナノ粒子は、第2の半導体からなるシェルに代えて、あるいは、第2の半導体からなるシェルとともに、絶縁体からなるシェルを有していてもよい。ここで、シェルとして用いることができる絶縁体として、半導体よりもバンドギャップが広い物質、具体的にはSiO2、ZnOなどが挙げられる。
すなわち、コアシェル型半導体ナノ粒子の例として、上記「II-VI族半導体」および上記「III-V族半導体」から選択される第1の半導体からなるナノ粒子をコアとして有し、その表面に、上記「II-VI族半導体」および上記「III-V族半導体」から選択される第2の半導体からなるシェル及び/または絶縁体からなるシェルが形成されているものが挙げられる。
また、上記X線吸収ナノ粒子と結合させて得られるX線吸収蛍光複合ナノ粒子が上記の体積平均粒径を有するよう、本発明で用いられる蛍光ナノ粒子は、体積平均粒径が1nm以上50nm以下であることが好ましく、2nm以上20nm以下であることがより好ましい。ここで、前述のX線吸収ナノ粒子と結合するために用いられる反応性官能基を有する適当なリンカーを結合させた蛍光ナノ粒子について測定した体積平均粒径を、ここにいう蛍光ナノ粒子の粒径として採用してもよい。
本発明で用いられる蛍光ナノ粒子について、励起波長・発光波長は、いずれも、生体にダメージを与えない350nm以上が好ましい。さらに、術中の観察を考えると、自家蛍光の少ない500nm以上が好ましい。これらの点から、蛍光ナノ粒子として、350nm以上1200nm以下、好ましくは、500nm以上950nm以下の励起波長を有するとともに、400nm以上1250nm以下、好ましくは650nm以上1150nm以下の発光波長を有するものが好適に用いられる。
これらの条件を満たす半導体ナノ粒子組成としては、たとえばCdSe/ZnS、CdTe/ZnS、InP/ZnSがあげられる。この中で、InP/ZnSは元素毒性少なく造影剤として用いるのに好ましい組成である。
本発明で用いられる蛍光ナノ粒子は、従来公知の方法により製造することができるし、市販のものを用いることもできる。
例えば、CdSe/ZnSは、Se粉末をCdOと反応させてCdSを得た後、ヘキサメチルジシリルチアン等の硫黄化剤と、酢酸亜鉛等とを加えて反応させることにより得ることができる。このとき、反応原料の溶解性向上および/または生成した粒子の凝集防止を目的として、トリアルキルホスフィン、トリアルキルホスフィンオキサイドなどを溶媒として用いることができる。また、InP/ZnSは、例えば、適当な高級脂肪酸存在下、高級カルボン酸インジウム、トリメチルシリルホスフィン等のリン化剤、ウンデシレン酸亜鉛等の高級カルボン酸亜鉛、アルカンチオールを適当な高沸点溶媒中で加熱することにより得ることができる。
例えば、CdSe/ZnSは、Se粉末をCdOと反応させてCdSを得た後、ヘキサメチルジシリルチアン等の硫黄化剤と、酢酸亜鉛等とを加えて反応させることにより得ることができる。このとき、反応原料の溶解性向上および/または生成した粒子の凝集防止を目的として、トリアルキルホスフィン、トリアルキルホスフィンオキサイドなどを溶媒として用いることができる。また、InP/ZnSは、例えば、適当な高級脂肪酸存在下、高級カルボン酸インジウム、トリメチルシリルホスフィン等のリン化剤、ウンデシレン酸亜鉛等の高級カルボン酸亜鉛、アルカンチオールを適当な高沸点溶媒中で加熱することにより得ることができる。
分子認識物質
本発明のX線吸収蛍光複合ナノ粒子は、X線造影剤として用いられるとともに、蛍光標識剤として手術の際に病巣の位置・大きさ・範囲を特定する目的にも用いられる。

本発明のX線吸収蛍光複合ナノ粒子による造影の標的とする病巣(以下、「標的病巣」と呼ぶ。)には、多くの場合特有の生体物質(例えば、腫瘍マーカー)が発現している。ここで、そのような生体物質を認識可能な分子認識物質が、本発明のX線吸収蛍光複合ナノ粒子に結合しているならば、この分子認識物質と、標的とする生体物質との結合反応を通じて、このX線吸収蛍光複合ナノ粒子を目的とする病巣に特異的に誘導することができる。この観点から、本発明のX線吸収蛍光複合ナノ粒子は、分子認識物質が結合していることが好ましい。
ここで、標的病巣に発現している特有の生体物質として、抗原となり得る生体物質、例えば、核酸(一本鎖であっても二本鎖であってもよいDNA、RNA、ポリヌクレオチド、オリゴヌクレオチド、PNA(ペプチド核酸)等、またはヌクレオシド、ヌクレオチドおよびそれらの修飾分子)、タンパク質(ポリペプチド、オリゴペプチド等)、アミノ酸(修飾アミノ酸も含む。)、糖質(オリゴ糖、多糖類、糖鎖等)、脂質、またはこれらの修飾分子、複合体などが挙げられる。このような生体物質の具体例として、腫瘍マーカー、シグナル伝達物質、ホルモンなどが挙げられるが、これらに限定されるものではない。
本発明においては、X線吸収蛍光複合ナノ粒子に結合している分子認識物質が、このような抗原を認識しうる分子認識物質、すなわち抗体であることが好ましい。ここで、「抗体」という用語は、任意の抗体断片または誘導体を含む意味で用いられ、例えば、Fab、Fab'2、CDR、ヒト化抗体、多機能抗体、単鎖抗体(ScFv)などを含む。
製造方法
本発明に係るX線吸収蛍光複合ナノ粒子を製造する方法は、特に限定されないものの、その典型例として、
(a)X線吸収ナノ粒子に、第1の反応性官能基を導入して、第1の反応性官能基を有するX線吸収ナノ粒子を得る工程と、
(b)蛍光ナノ粒子に、前記第1の反応性官能基と反応して結合を形成可能な第2の反応性官能基を導入して、第2の反応性官能基を有する蛍光ナノ粒子を得る工程と、
(c)前記工程(a)で得られた「第1の反応性官能基を有するX線吸収ナノ粒子」と前記工程(b)で得られた「第2の反応性官能基を有する蛍光ナノ粒子」とを反応させて、X線吸収蛍光複合ナノ粒子を得る工程と
を含む製造方法が挙げられる。

(c)前記工程(a)で得られた「第1の反応性官能基を有するX線吸収ナノ粒子」と前記工程(b)で得られた「第2の反応性官能基を有する蛍光ナノ粒子」とを反応させて、X線吸収蛍光複合ナノ粒子を得る工程と
を含む製造方法が挙げられる。
ここで、上記第1の反応性官能基および第2の反応性官能基として、カルボキシル基、アミノ基、アルデヒド基、チオール基、マレイミド基などの化学官能基、並びに、ビオチン、ストレプトアビジン、アビジンなどアフィニティ相互作用に基づく結合を形成しやすい分子が挙げられる。
X線吸収ナノ粒子への第1の反応性官能基の導入方法は、特に限定されるものではないが、例えば、X線吸収ナノ粒子と結合性のある官能基と、第1の反応性官能基とを有する適当なリンカー、例えば、アミノ基を有するシランカップリング剤やアミノ基を有するチオール化合物等をX線吸収ナノ粒子と反応させる方法が挙げられる。いうまでもないが、このようなシランカップリング剤やチオール化合物等は、アミノ基以外の反応性官能基を第1の反応性官能基として有していてもよい。
蛍光ナノ粒子への第2の反応性官能基の導入方法についても、特に限定されるものではないが、例えば、蛍光ナノ粒子と結合性のある官能基と、第2の反応性官能基とを有する適当なリンカー、例えば、カルボキシル基を有するシランカップリング剤やカルボキシル基を有するチオール化合物等を蛍光ナノ粒子と反応させる方法が挙げられる。このようなリンカーが、カルボキシル基以外の反応性官能基を第2の反応性官能基として有していてもよいことはいうまでもない。
また、上記第1の反応性官能基と上記第2の反応性官能基との反応に際しては、これらの反応性官能基のうちいずれか一方、あるいはその両方について、一旦より反応性の高い官能基に変換してから反応を行ってもよい。例えば、アミノ基とカルボキシル基とを反応させる際に、カルボキシル基を一旦対応する活性エステルに変換してからアミノ基と反応させてもよい。
さらに、本発明では、上記分子認識物質を導入するために、上記製造方法には、
(d)前記工程(c)で得られたX線吸収蛍光複合ナノ粒子に、上記分子認識物質を結合させて、分子認識物質が結合しているX線吸収蛍光複合ナノ粒子を得る工程
をさらに含んでいてもよい。

をさらに含んでいてもよい。
ここで、上記工程(d)に際しては、公知の活性エステル法などの常法を用いて結合反応を行うことができる。この工程(d)は、工程(c)との一連の工程として行ってもよいし、あるいは、工程(c)の後、一旦後処理を行った後に独立した工程として行ってもよい。
〔X線造影剤〕
上記記載のX線吸収蛍光複合ナノ粒子は、X線造影剤として好適に用いられるものである。そこで、本発明では、上記記載のX線吸収蛍光複合ナノ粒子を含むX線造影剤も提供される。

本発明のX線造影剤は、上述したX線吸収蛍光複合ナノ粒子のほかに、所要により、安定化剤などの補助剤、製薬上許容される充填剤など、X線造影剤に一般に配合しうる副成分を含むことができ、その濃度や配合についても、用途に応じて適宜変えることができる。
1.ナノ粒子合成処方
[調製例1-1] Auナノ粒子の合成
X線吸収ナノ粒子として、以下の方法によりアミノプロピル修飾Auナノ粒子を合成した。
まず、以下の要領で、α液とβ液をそれぞれ調製した:
α液の調整:1%塩化金酸 3mlを純水に混合し80mlとする。
β液の調整:クエン酸を純水に溶解し、1%クエン酸溶液4mlとする。
[調製例1-1] Auナノ粒子の合成
X線吸収ナノ粒子として、以下の方法によりアミノプロピル修飾Auナノ粒子を合成した。
まず、以下の要領で、α液とβ液をそれぞれ調製した:
α液の調整:1%塩化金酸 3mlを純水に混合し80mlとする。
β液の調整:クエン酸を純水に溶解し、1%クエン酸溶液4mlとする。
次に、このα液・β液をホットプレートで60℃に加熱したのち、α液をマグネチックスターラーで激しく撹拌している中にβ液を添加した。添加したのち、溶液の温度を100℃とし、溶液が赤色に着色してから30分間、100℃で反応を行った。反応終了後、室温にてアミノプロピルトリエトキシシランを300μL添加し70℃で24時間反応後、遠心分離にて、副生成物を除去後、純水に再分散し、Au基準で0.3mol/lに調整してアミノプロピル修飾Auナノ粒子の溶液(溶液Xa)を得た。
得られた溶液Xa中の粒子を透過型電子顕微鏡で観察し、平均粒径40nmのAu粒子が形成していることを確認した。

[調製例1-2] Gd2O3 ナノ粒子の合成
X線吸収ナノ粒子として、以下の方法によりアミノプロピル修飾Gd2O3ナノ粒子を合成した。
まず、以下の要領で、α液とβ液をそれぞれ調製した:
α液の調整:尿素を純水に溶解し、0.8mol/l 500ccとした
β液の調整:硝酸ガドリニウムを純水に溶解し、0.05mol/l 100ccとした。
X線吸収ナノ粒子として、以下の方法によりアミノプロピル修飾Gd2O3ナノ粒子を合成した。
まず、以下の要領で、α液とβ液をそれぞれ調製した:
α液の調整:尿素を純水に溶解し、0.8mol/l 500ccとした
β液の調整:硝酸ガドリニウムを純水に溶解し、0.05mol/l 100ccとした。
次に、このα液とβ液を混合し、激しく撹拌しながら、100℃で30min反応を行った。反応終了後、遠心分離をし、純水で洗浄したのち、600℃1時間焼成し、Gd2O3ナノ粒子を得た。その後、純水5cc中にGd2O3 350mgを分散し、アミノプロピルトリエトキシシランを300μL添加し70℃で24時間反応後、遠心分離にて、副生成物を除去後、純水に再分散してアミノプロピル修飾Gd2O3ナノ粒子の溶液(溶液Xb)を得た。
得られた溶液Xb中の粒子を透過型電子顕微鏡で観察し、平均粒径60nmのGd2O3粒子が形成していることを確認した。

[調製例2-1] CdSe/ZnS粒子の合成
蛍光ナノ粒子として、以下の方法によりCdSe/ZnS粒子を合成した。
Se粉末(0.7896g)を、トリオクチルホスフィン(TOP、7.4g)へ添加し、混合物を150℃まで加熱して(窒素気流下)、TOP-Seストック溶液を作成した。
蛍光ナノ粒子として、以下の方法によりCdSe/ZnS粒子を合成した。
Se粉末(0.7896g)を、トリオクチルホスフィン(TOP、7.4g)へ添加し、混合物を150℃まで加熱して(窒素気流下)、TOP-Seストック溶液を作成した。
別途、CdO(0.450g)及びステアリン酸(8g)をアルゴン雰囲気下、三口フラスコ中で150℃まで加熱した。CdOが溶解した後、溶液を室温まで冷却した。前記溶液に、トリオクチルホスフィンオキサイド(TOPO、8g)、及び1-ヘプタデシル-オクタデシルアミン(HDA、12g)を添加し、混合物を再び150℃まで加熱し、ここで、TOP-ストック溶液を素早く添加する。そののちチャンバーの温度を220℃まで加熱し、さらに一定の速度で、120分かけて250℃まで上昇させた。その後、温度を100℃まで下げ、酢酸亜鉛二水和物を添加撹拌し溶解させたのち、ヘキサメチルジシリルチアンのトリオクチルフォスフィン溶液を滴下し、数時間撹拌を続けて反応を終了した。室温に下げた後、メルカプトプロピオン酸0.01gを添加して1時間撹拌したのち、洗浄し、純水中に再分散し、発光ピーク波長620nm、Cd基準で濃度3.0 MのCdSe/ZnS 溶液(溶液C)を得た。
得られた溶液Cにおける、動的光散乱法によるCdSe/ZnS粒子の平均粒径は12nmであった。

[調製例2-2] InP/ZnS粒子の合成
蛍光ナノ粒子として、以下の方法によりInP/ZnS粒子を合成した。
ミリスチン酸インジウム0.1mmol、ステアリン酸0.1mmol、トリメチルシリルホスフィン0.1mmmol、ドデカンチオール0.1mmol、ウンデシレン酸亜鉛0.1mmolを、オクタデセン8mlとともに三口フラスコに入れ、窒素雰囲気下で還流を行いながら300℃1時間加熱した。
室温に下げた後、メルカプトプロピオン酸0.01gを添加して1時間撹拌し、発光ピーク波長620nm、InP基準で濃度3.0MのInP/ZnS溶液(溶液D)を得た。
得られた溶液Dにおける、動的光散乱法によるInP/ZnS粒子の平均粒径は16nmであった。
蛍光ナノ粒子として、以下の方法によりInP/ZnS粒子を合成した。
ミリスチン酸インジウム0.1mmol、ステアリン酸0.1mmol、トリメチルシリルホスフィン0.1mmmol、ドデカンチオール0.1mmol、ウンデシレン酸亜鉛0.1mmolを、オクタデセン8mlとともに三口フラスコに入れ、窒素雰囲気下で還流を行いながら300℃1時間加熱した。
室温に下げた後、メルカプトプロピオン酸0.01gを添加して1時間撹拌し、発光ピーク波長620nm、InP基準で濃度3.0MのInP/ZnS溶液(溶液D)を得た。
得られた溶液Dにおける、動的光散乱法によるInP/ZnS粒子の平均粒径は16nmであった。
2.サンプル
[X線吸収ナノ粒子と蛍光ナノ粒子との粒子個数比率について]
下記実施例1および2において、複合ナノ粒子におけるX線吸収ナノ粒子と蛍光ナノ粒子との粒子個数比率は、以下の手順に基づいて算出した。
まず、各実施例で用いたX線吸収ナノ粒子の溶液(すなわち、上記溶液XaまたはXb)を一旦乾燥して固形分を得、この固形分の質量を測定し、その後、この固形分を王水に溶解させて得られる試料についてICPを用いた元素分析を行い、固形分に占めるX線吸収ナノ粒子構成化合物の含有量を測定した。そして、これらの測定値を用いて、複合ナノ粒子におけるX線吸収ナノ粒子構成化合物の含有量Maを算出した。
また、各実施例で用いた蛍光ナノ粒子の溶液(すなわち、上記溶液CまたはD)についても同様の手順により、固形分の質量および固形分に占める蛍光ナノ粒子構成化合物の含有量を測定し、これらの測定値を用いて複合ナノ粒子における蛍光ナノ粒子構成化合物の含有量Mbを算出した。
また、X線吸収ナノ粒子の平均粒子径daおよび蛍光ナノ粒子の平均粒子径dbを、動的散乱法を用いてそれぞれ測定した。
そして、X線吸収ナノ粒子と蛍光ナノ粒子との粒子個数比率を、上記Ma、Mb、daおよびdb、並びに、X線吸収ナノ粒子構成化合物の密度ρaおよび蛍光ナノ粒子構成化合物の密度ρbを用いて、下記式(1)で表す比として算出した。
[実施例1] Au-CdSe/ZnS複合ナノ粒子の合成
調製例1-1で得られたアミノプロピル修飾Auナノ粒子の溶液(溶液Xa)と調製例2-1で得られたCdSe/ZnS粒子の溶液(溶液C)とを、アミノプロピル修飾Auナノ粒子とCdSe/ZnS粒子とが1:100の粒子個数比率となる割合で混合し、EDC(1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド)を添加、撹拌し1時間反応させた後、遠心分離にて、未反応物を除去した。この後、抗HER2抗体200μgを添加、再度EDCを添加、撹拌し、1時間反応させた後、遠心分離にて、未反応物を除去し、PBS溶液1mlに再分散し、0.2Au原子Mとした。

この溶液を溶液Aとする。
得られた溶液Aにおける、動的光散乱法によるAu-CdSe/ZnS複合ナノ粒子の平均粒径は72nmであった。

[実施例2] Gd2O3-InP/ZnS複合ナノ粒子の合成
調製例1-2で得られたアミノプロピル修飾Gd2O3ナノ粒子の溶液(溶液Xb)と調製例2-2で得られたInP/ZnS粒子の溶液(溶液D)とを、アミノプロピル修飾Gd2O3ナノ粒子とInP/ZnS粒子とが1:100の粒子個数比率となる割合で混合し、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミドを添加、撹拌し1時間反応させた後、遠心分離にて、未反応物を除去した。この後、抗HER2抗体200μgを添加、再度EDCを添加、撹拌し、1時間反応させた後、遠心分離にて、未反応物を除去し、PBS溶液1mlに再分散し、0.2Gd原子Mとした。

この溶液を溶液Bとする。
得られた溶液Bにおける、動的光散乱法によるGd2O3-InP/ZnS複合ナノ粒子の平均粒径は93nmであった。

[比較例1]
既存のX線造影剤として、イオパミドールを主成分とする市販のX線造影剤(バイエル社製イオパミロン(登録商標))をそのまま使用した。ここで、イオパミドールは、ヨウ素を構成原子として含む化合物である。

Gd2O3ナノ粒子を含まないInP/ZnS粒子の溶液として、上記調製例2-2で得られた溶液Dをそのまま用いた。
3.評価:X線造影性能・蛍光性能
上記実施例およぶ比較例に示した各サンプルについて、X線造影性能および蛍光性能の評価を行った。

X線造影性能の評価は、各サンプルについて、サンプルを5mm×5mm×4cmのセルに充填して、Varian社製FPD Paxscan1313Rを用いてX線加速電圧70kV、X線照射量1mRの条件でX線画像を撮影し、撮影時の撮影画像カウント値を測定することにより行った。

Claims (9)
- X線吸収ナノ粒子と蛍光ナノ粒子が結合してなる、体積平均粒子径が10nm以上1000nm以下のX線吸収蛍光複合ナノ粒子。
- 分子認識物質が結合している請求項1に記載のX線吸収蛍光複合ナノ粒子。
- 上記分子認識物質が抗体である請求項2に記載のX線吸収蛍光複合ナノ粒子。
- 上記X線吸収ナノ粒子が、Gd、IおよびAuからなる群より選ばれるいずれかの元素を含む請求項1~3のいずれかに記載のX線吸収蛍光複合ナノ粒子。
- 上記蛍光ナノ粒子の励起波長が350nm以上1200nm以下であり、かつ発光波長が400nm以上1250nm以下である請求項1~4のいずれかに記載のX線吸収蛍光複合ナノ粒子。
- 上記蛍光ナノ粒子の体積平均粒子径が1nm以上50nm以下である、請求項1~5のいずれかに記載のX線吸収蛍光複合ナノ粒子。
- 上記蛍光ナノ粒子が、II-VI族半導体およびIII-V族半導体、並びに、珪素またはゲルマニウムからなる半導体から選択されるいずれか1種以上の半導体を含む半導体ナノ粒子からなることを特徴とする請求項1~6のいずれかに記載のX線吸収蛍光複合ナノ粒子。
- 上記半導体ナノ粒子が、CdSe/ZnS、CdTe/ZnSおよびInP/ZnSからなる群より選ばれるいずれからなる請求項7に記載のX線吸収蛍光複合ナノ粒子。
- 請求項1~8のいずれかに記載のX線吸収蛍光複合ナノ粒子を含むX線造影剤。
