Definitions
- the present invention relates to a library of azoline compounds and azole compounds.
- a peptide library rich in diversity can be constructed in a short time by preparing an mRNA library and translating it in one pot. Further, by combining with the mRNA display method or the like, it is possible to simultaneously obtain information on the peptide obtained by screening and the nucleic acid encoding the peptide, so that the genotype and the phenotype can be easily correlated. As described above, there are many excellent points in the synthesis of peptide libraries by a translation system, but only peptidic compounds can be produced. In screening using a library, it is often necessary to identify a compound that inhibits a target substance having protease activity. However, since the library of peptidic compounds is cleaved by proteases, it is not possible to efficiently screen for compounds that inhibit the activity of the target substance.
- patellamide produced by the ascidian symbiotic alga Prochloron didemni is a low-molecular cyclic peptide considered to have various physiological activities, and is biosynthesized through a unique pathway by the products of the pat gene group consisting of patA to G.
- FIG. 15 shows an outline of the pat gene group and this biosynthetic pathway.
- the precursor in this biosynthesis is the PatE peptide of the patE gene product. Since the patE gene has a hypervariable region (cassette region), its product constructs a natural combinatorial library.
- the PatE peptide has a recognition sequence by a post-translational modification enzyme on both sides of the cassette region.
- PatD introduces an azoline skeleton into Cys, Ser, and Thr in the PatE cassette, and converts Cys into a thiazoline skeleton and Ser and Thr into an oxazoline skeleton.
- PatA cleaves the recognition sequence on the N-terminal side of the cassette region of PatE.
- PatG consists of two domains, and the N-terminal oxidase domain converts the azoline skeleton introduced by PatD into an azole skeleton, ie, a thiazoline skeleton into a thiazole skeleton.
- the C-terminal peptidase domain undergoes macrocyclization while cleaving the C-terminal recognition sequence of the PatE cassette region.
- Non-patent Document 1 describes the sequences shown in the following table.
- the sequence of the natural cassette region includes (i) 7 or 8 residues, (ii) the Ser / Thr that is modified in the second, fourth, sixth, and eighth positions from the N-terminus of the cassette region.
- Residues subject to modification (Ser / Thr / Cys) are rarely lined up
- Residues other than Ser / Thr / Cys include Val, Ala, Ile , Phe, Leu and many other hydrophobic residues. Therefore, it was thought that these common points were necessary to become substrates for the post-translational modification enzymes PatD and PatG. However, it is unclear which of these Ser / Thr / Cys are modified or not, and the substrate specificity of PatD and PatG has not been clarified so far.
- An object of the present invention is to provide a method for efficiently producing a library that can be used for screening a compound that has sufficient diversity and binds to a target substance having protease activity.
- PatE library is expressed by cell-free translation system as a precursor peptide containing PatE leader sequence, recognition sequence 1 by azoline skeleton-introducing enzyme, cassette region, and recognition sequence 2 by azoline skeleton-introducing enzyme This can be efficiently performed in a single pot by modifying it with an azoline skeleton-introducing enzyme and cleaving unnecessary regions, and as a result, it can be used for screening using target substances that are sufficiently diverse and have protease activity.

- the types of primers used for each library construction are shown below. Primer sequences are shown in the following primer list.
- leader sequence In order to examine the necessity of the leader sequence, in the same manner as in [1] to [8] above, when the leader sequence is removed from the substrate peptide, the leader sequence is translated and synthesized as a peptide separate from the substrate peptide. Thus, the experiment was conducted with respect to the case of adding a chemically synthesized leader sequence to final concentrations of 1 uM, 10 uM, and 50 uM.
- the cassette sequence was 8 amino acids (VTACITFC). The results are shown in FIG.
- leader sequence is fused to the substrate peptide, the modification efficiency by the enzyme increases, but even if there is no leader sequence, one molecule is dehydrated, and it is found that the leader sequence is not essential to become a substrate for the enzyme It was. In addition, if the leader sequence was present in the reaction system, dehydration of 4 molecules was observed even if it was not fused with the substrate peptide, confirming that the substrate peptide was sufficiently modified. This suggests that the leader sequence is not necessary for substrate selection, and that the structure of the leader sequence contributes to the activation of the enzyme and affects the reaction efficiency.
- leader sequence (2) In the same manner as in [1] to [8] above, a part of the leader sequence was deleted as shown in FIGS. 8 and 19, and the enzyme activity by PatD was confirmed. As shown in FIGS. 8 and 19, it was found that the full-length leader sequence is not essential for modification by PatD. In addition, it was confirmed that the sequence on the C-terminal side is important for the modification by PatD, and if the C-terminal of the leader sequence is included, for example, even about 6 amino acids are sufficiently modified by PatD.
- leader sequence (3) Examination of leader sequence (3) In the same manner as in [1] to [8] above, the entire leader sequence was replaced with a completely different peptide sequence as shown in FIG. 18, and the enzyme activity by PatD was confirmed. As shown in FIG. 18, the leader sequence that needs to be present in order to be modified by PatD is not limited to the leader sequence of PatE. It was confirmed that the leader sequence may be a shuffled sequence.

- FIG. 11 Thr isomers, substituted Sers, diamino acids, and the like that were not previously considered to be PatD substrates are also modified by PatD and converted to the corresponding azoline ring skeleton. It was confirmed.
- MMP12 MMP12 was expressed in E. coli as a construct with a 6xHis tag and an Avi tag attached to the C-terminus. Purification was performed using a His tag. In addition, since the Avi tag is biotinylated by BirA expressed by birA incorporated in the same plasmid, MMP12 was added for immobilization with streptavidin beads during mRNA display.
- [16-3-8] Identification of selected azoline-containing compound sequence The above-described series of operations is repeated, and TA cloning is performed using the amplified DNA when no change is observed in the DNA recovery rate. The sequence of the azoline-containing compound was identified.
- [16-3-9] Evaluation of binding ability of selected azoline-containing compound According to the mRNA display scheme, a series of operations are performed from mRNA of a selected peptide, not from an mRNA library. Whether MMP12 bound to the bound beads was examined. In addition, the operation was performed under conditions where post-translational modification with PatD was performed and not performed. As a result, it was suggested that the resulting sequence was bound to MMP12 only when it was modified with PatD, and that the azoline skeleton contributed to binding to MMP12 in those sequences.
- FIG. 1 An image of the mRNA display method is shown in FIG. The results of selection are shown in FIG. As a result of the selection, the sequences of the five kinds of azoline-containing compounds A to E selected (data not shown) were analyzed, and the results of evaluating the binding ability to MMP12 are shown in FIG.

Abstract
Description

A-(Xaa0)m-B (I)
Ser:
MNKKNILPQQGQPVIRLTAGQLSSQLAELSEEALGDA(配列番号:1)

A-(Xaa0)m-B (I)

(i) mが2~20(但し、7及び8を除く)。mは、例えば、2、3、4、5、6、9、10、12、14、16、20、30、40である;
(ii) nが1~10(但し、3及び4を除く)。nは、例えば、1、2、5、6、7、8、10、15、20である;

当該方法は、N末端から順に、アゾリン骨格導入酵素による認識配列1、-(Xaa0)m-、及びアゾリン骨格導入酵素による認識配列2を含む前駆体ペプチドをコードするmRNA製造する工程と(ここで、認識配列1及び2は、0~10アミノ酸からなるアゾリン骨格導入酵素による認識配列をいう。);
上記mRNAを用いて、無細胞翻訳系により前駆体ペプチドを発現させる工程と;
