1-{6-[4-シクロプロピル-1-(テトラヒドロピラン-2-イル)-1H-インダゾール-5-イル]ピリジン-3-イル}-1-エチルプロピルカルバミン酸 tert-ブチルエステル(実施例化合物1、2.1g、4.2mmol)に、エタノール(20ml)および48重量%臭化水素溶液(10ml)を加え、40℃で4時間加熱撹拌した。溶媒を減圧留去し、50℃で残渣にエタノール(10ml)および水(2ml)を加えた。氷水冷却後、析出した固体を濾取し、エタノールで洗浄することにより白色固体(883mg)を得た。
融点:>221-223℃(分解)。
マススペクトル(CI,m/z):321([M+H]+)。
1H-NMRスペクトル(CD3OD):δ 0.45-0.50(m,2H),0.97-1.04(m,8H),2.17-2.53(m,5H),7.60(d,J=8.8Hz,1H),7.65-7.69(m,1H),8.39-8.42(m,2H),8.72(dd,J=8.8,2.4Hz,1H),9.02(dd,J=2.4,0.5Hz,1H)。
1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン 塩酸塩(実施例化合物2、30.0g、76.3mmol)に、n―ブタノール(300ml)および4M水酸化ナトリウム水溶液(370ml)を加え、室温で1.5時間撹拌した。有機層を分離し、水(150ml)で洗浄後、有機層を濃縮し、固体(22.6g)を得た。
マススペクトル(CI,m/z):321([M+H]+)。
1H-NMRスペクトル(CD3OD):δ 0.41-0.47(m,2H),0.79-0.86(m,8H),1.75-2.04(m,4H),2.28-2.37(m,1H),7.42-7.49(m,2H),7.62(dd,J=8.3,0.7Hz,1H),7.93(dd,J=8.3,2.4Hz,1H),8.24(d,J=0.7Hz,1H),8.67(dd,J=2.4,0.7Hz,1H)。
1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン(実施例化合物4、4.0g、12mmol)のエタノール(160ml)溶液に、L-酒石酸(2.8g、19mmol)のエタノール(85ml)溶液を室温で0.5時間かけて滴下し、同温度で0.67時間撹拌した。反応溶液を10℃に冷却し、析出した固体を濾取した。エタノール(40ml)で洗浄後、40℃で1時間減圧乾燥し、さらに60℃に昇温して11時間減圧乾燥させることにより、1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン L-酒石酸塩(実施例化合物5)(5.4g)を白色固体として得た(収率92%)。
融点:215-216℃。
マススペクトル(CI,m/z):321([M+H]+)。
1H-NMRスペクトル(CD3OD):δ 0.41-0.46(m,2H),0.83-0.89(m,2H),0.95(t,J=7.4Hz,6H),2.10(dq,J=14.8,7.4Hz,2H),2.26(dq,J=14.8,7.4Hz,2H),2.34-2.42(m,1H),4.40(s,2H),7.46-7.52(m,2H),7.81(dd,J=8.3,0.5Hz,1H),7.97(dd,J=8.3,2.7Hz,1H),8.27(d,J=0.7Hz,1H),8.66-8.67(m,1H)。
マレイン酸(11mg、0.095mmol)のテトラヒドロフラン(110μl)溶液に、1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン(実施例化合物4、10mg、0.031mmol)のテトラヒドロフラン(800μl)溶液を加え、室温で6日間静置した。析出した固体を濾取し、50℃で減圧乾燥することにより1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン マレイン酸塩(実施例化合物6)(11mg)を白色固体として得た(収率71%)。
融点:174-178℃。
1H-NMRスペクトル(CD3OD):δ 0.41-0.46(m,2H),0.83-0.90(m,2H),0.96(t,J=7.5Hz,6H),2.10(dq,J=14.8,7.5Hz,2H),2.21-2.42(m,3H),6.27(s,3H),7.46-7.53(m,2H),7.82(dd,J=8.5,0.6Hz,1H),7.94(dd,J=8.5,2.7Hz,1H),8.27(s,1H),8.65-8.66(m,1H)。
フマル酸(11mg、0.095mmol)のテトラヒドロフラン(275μl)溶液に、1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン(実施例化合物4、10mg、0.031mmol)のテトラヒドロフラン(667μl)溶液を加え、室温で3日間静置した。析出した固体を濾取し、50℃で減圧乾燥することにより1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン フマル酸塩(実施例化合物7)(8.8mg)を白色固体として得た(収率75%)。
融点:257℃。
1H-NMRスペクトル(CD3OD):δ 0.41-0.46(m,2H),0.82-0.94(m,8H),1.94-2.07(m,2H),2.18(dq,J=14.8,7.3Hz,2H),2.31-2.41(m,1H),6.68(s,1H),7.45-7.51(m,2H),7.76(dd,J=8.3,0.7Hz,1H),7.95(dd,J=8.3,2.4Hz,1H),8.26(d,J=0.7Hz,1H),8.66(dd,J=2.4,0.7Hz,1H)。
コハク酸(11mg、0.093mmol)のテトラヒドロフラン(275μl)溶液に、1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン(実施例化合物4、10mg、0.031mmol)のテトラヒドロフラン(667μl)溶液を加え、室温で6日間静置した。析出した固体を濾取し、50℃で減圧乾燥することにより1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン コハク酸塩(実施例化合物8)(6.9mg)を白色固体として得た(収率58%)。
融点:218℃。
1H-NMRスペクトル(CD3OD):δ 0.41-0.46(m,2H),0.82-0.90(m,8H),1.94(dq,J=14.6,7.3Hz,2H),2.11(dq,J=14.6,7.3Hz,2H),2.30-2.40(m,1H),2.51(s,2H),7.44-7.51(m,2H),7.72(d,J=8.3Hz,1H),7.94(dd,J=8.3,2.4Hz,1H),8.25(d,J=0.7Hz,1H),8.66(d,J=2.4Hz,1H)。
1.ラット肝ミクロソームでの抗酸化作用試験
ミクロソーム脂質過酸化測定系を用いて、被験化合物の抗酸化作用を評価した。この試験はミクロソームにニコチンアミドアデニンジヌクレオチドリン酸および硫酸第一鉄を添加することで惹起される脂質過酸化反応によって生成する、チオバルビツール酸反応物質(TBARS)を定量する。この系に被験化合物を添加し、被験化合物によるTBARS生成抑制作用を抗酸化作用として評価する。この方法は、化合物などの抗酸化作用を測定するための一般的な方法である(参考文献;薬学雑誌119,93-99(1999), Biochimica et Biophysics Acta, 1046 (1990) 207-213,化学と生物 実験ライン2 脂質過酸化実験法(廣川書店))。
(試薬の調製)
1)緩衝溶液の調製
25mM トリスヒドロキシメチルアミノメタン(Tris)(pH7.4)、150mM 塩化カリウム(KCl)となるよう混和して緩衝溶液とした。
硫酸第一鉄(FeSO4・7H2O)を蒸留水に溶解し5mM 溶液を調製した。
ニコチンアミドアデニンジヌクレオチドリン酸(NADPH)を蒸留水に溶解し50mM 溶液を調製した。
エチレンジアミン四酢酸(EDTA)を蒸留水に溶解し200mM 溶液を調製した。
市販のラット肝臓ミクロソーム[XENOTECH, Catalog No.R1000(20mg protein/ml)]を5μl使用した。
被験化合物として、本発明化合物である化合物A[1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-エチルプロピルアミン 塩酸塩(実施例化合物2)]と、それに類似した化合構造である化合物B[1-[6-(4-シクロプロピル-1H-インダゾール-5-イル)ピリジン-3-イル]-1-メチルエチルアミン 塩酸塩(欧州特許出願公開第1679308号明細書記載の方法により合成)]を使用した。化合物Aは、1.3mgをジメチルスルホキシド(DMSO)0.315mlに溶解し、化合物Bは、2.15mgをジメチルスルホキシド(DMSO)0.554mlに溶解し、10mM溶液として被験化合物溶液を調製した。
1)反応チューブに被験化合物溶液(5μl)を入れた。
3)次いで、各反応チューブにラット肝臓ミクロソーム5μlを加えて混和し、インキュベーター(37℃)に入れて5分間プレインキュベーションさせた。
TBARS生成抑制率(%)=[1-(Tx-T0)/(Tn-T0)]×100
T0:非脂質過酸化反応群のTBARS生成値(μM)
Tn:対照溶液添加群のTBARS生成値(μM)
Tx:被験化合物添加群のTBARS生成値(μM)
(化合物添加方法)
・被験化合物添加群:ジメチルスルホキシド(DMSO)溶液に溶解した、化合物溶液を添加した。
以上の結果から、化合物Aは、強力な抗酸化作用を示した。一方、化合物Bは、化合物Aと類似した化学構造を有するにもかかわらず、強力な抗酸化作用は示さなかった。
製剤例を挙げて本発明の薬剤をさらに具体的に説明するが、本発明はこれらの製剤例にのみ限定されるものではない。

乳糖 66.4mg
トウモロコシデンプン 20mg
カルボキシメチルセルロースカルシウム 6mg
ヒドロキシプロピルセルロース 6mg
ステアリン酸マグネシウム 0.6mg
化合物A、乳糖を混合機中で混合し、その混合物にカルボキシメチルセルロースカルシウムおよびヒドロキシプロピルセルロースを加えて造粒し、得られた顆粒を乾燥後整粒し、その整粒顆粒にステアリン酸マグネシウムを加えて混合し、打錠機で打錠する。また、化合物Aの添加量を変えることにより、100mg中の含有量が0.1mg、10mgまたは50mgの錠剤を調製できる。

流動パラフィン 10.0g
白色ワセリン 適量
均一に溶融した白色ワセリンおよび流動パラフィンに、化合物Aを加え、これらを十分に混合して後に徐々に冷却することで眼軟膏を調製する。化合物Aの添加量を変えることにより、濃度が0.05%(w/w)、0.1%(w/w)、0.5%(w/w)または1%(w/w)の眼軟膏を調製できる。

ポリソルベート80 適量
滅菌精製水 適量
化合物Aおよび塩化ナトリウムを滅菌精製水に加えて注射剤を調製する。化合物Aの添加量を変えることにより、10ml中の含有量が0.1mg、10mgまたは50mgの注射剤を調製できる。

ポリソルベート80 適量
リン酸水素二ナトリウム 適量
リン酸二水素ナトリウム 適量
滅菌精製水 適量
滅菌精製水に化合物Aおよびそれ以外の上記成分を加え、これらを十分に混合して点眼液を調製する。化合物Aの添加量を変えることにより、濃度が0.05%(w/v)、0.1%(w/v)、0.5%(w/v)または1%(w/v)の点眼剤を調製できる。
