Abstract

COMBINACIÓN DE BIOPELÍCULA BACTERIANA Y CRISTAL LÍQUIDO PARA LA PREPARACIÓN DE UN DISPOSITIVO ELECTROÓPTJCO
La presente invención se refiere a un material que comprende al menos una biopelícula bacteriana y una sustancia electro-óptica, su procedimiento de obtención y su uso para la fabricación de dispositivos electro-ópticos.
ESTADO DE LA TÉCNICA ANTERIOR
Para la fabricación de dispositivos electro-ópticos basados en cristal líquido disperso en matrices sólidas porosas, el cristal líquido (CL) o sustancia electro- óptica se encapsula en una matriz polimérica o vitrea. Estas matrices encapsulantes deben cumplir con los siguientes requisitos para poder ser utilizadas como medio de confinamiento del CL en un dispositivo de transmisión vanable de luz: por una parte debe tener una macroporosídad, con poros de tamaño del orden de micrómetros para poder adoptar una disposición y volumen específico en términos de tamaño y forma de los microdominios de CL que condicionará su orientación y sus propiedades ópticas. Por otra parte la matriz debe tener un índice de refracción igual o similar al del CL, cuando está orientado por el campo eléctrico, para permitir la obtención de un material transparente. El CL es una sustancia que presenta un orden molecular quasi- crístalino en el estado líquido de la materia, estas extraordinarias características confieren a los materiales especiales propiedades ópticas, eléctricas y reológicas que son explotadas en muchos dispositivos, como pantallas y ventanas ópticas (J.W. Doane, Liq. Cryst. 33 (2006) 1313), vidrios GDLC:

Por otro lado es sabido que las biopelículas son matrices de depósitos microbianos formados por sustancias poliméricas extraceluiares (SPE) que se adhieren tanto a superficies biológicas como no biológicas. El enfoque más tradicional en la investigación con las biopelículas se dirige a evitar su formación, ya que frecuentemente recubren a los microorganismos patógenos e impiden su destrucción (Anderson et Al. Curr. Top. Microbiol. Immunol. 2008) 322, 85-105), y a prevenir la suciedad (incrustaciones de depósitos) en los sistemas industriales y marinos. Otros temas de investigación están orientados a aplicaciones medioambientales (bioremediacion), biocatálisis, (Rosche et Al. Trends in Biotechnoloav, 2009, 27 (1 1 ), pp. 636-643) y hace muy poco, las superficies de las biopelículas han comenzado a ser utilizadas en "materiales", como plantillas para la producción de materiales micro y nano-estructurados (Wang et Al. Adv. Mater. 2009, 21 , 2815-2818). En la solicitud de patente na 9101325, y en la patente ES 2006/00674.
DESCRIPCIÓN DE LA INVENCIÓN
La presente invención proporciona un material que comprende al menos una biopelícula bacteriana y una sustancia electro-óptica (CL), el procedimiento de dicho material y su uso para la fabricación de dispositivos electro-ópticos.
La obtención de un material electro-óptico fabricado mediante la combinación de una sustancia electro-óptica, como el cristal líquido y una matriz de biopelícula cultivada biotecnologicamente en el que la estructura de las biopelículas establece la disposición tridimensional necesaria para obtener la actividad electro-óptica. El cristal líquido se introduce en la estructura macroporosa de la biopelícula crecida en sustratos de vidrio transparente conductor. Este material compuesto, descrito en la presente invención, se utilizó para la fabricación de un dispositivo con transmítancia conmutable eléctricamente.
El principio de funcionamiento en estos materiales compuestos se relaciona con la orientación del material electro-óptico encapsulado en la macroporosidad de la matriz o biopelícula, lo que permite la conmutación eléctrica de su opacidad. Cuando no se aplica ningún campo eléctrico, el material electro- óptico muestra una orientación aleatoria de los microdominios (las zonas microscópicas donde las moléculas de CL muestran un patrón orientacional común), ya que los microdominios muestran diferentes índices de refracción {debido a la birrefringencia del CL). Esta diversidad de índices de refracción, distintos al de la matriz producen la dispersión de la luz (estado opaco). Cuando un campo eléctrico es aplicado a través del material compuesto, todos los microdominios de CL se re-orientan en la dirección del campo eléctrico. En esta situación, el índice de refracción que presenta el CL en la dirección del campo eléctrico es el mismo para todos los microdominios e igual o similar al de la matriz, obteniéndose un estado transparente.
Un primer aspecto de la presente invención se refiere a un material que comprende al menos una biopelícula bacteriana y una sustancia electro-óptica. Por "sustancia electro-óptica" en la presente invención se entiende por aquella sustancia que posee la capacidad de variar sus propiedades ópticas mediante la aplicación de un campo eléctrico.
En una realización preferida la sustancia electro-óptica se selecciona de entre una sustancia electrocrómica, partículas en suspensión (SPD) o un cristal líquido (CL). Siendo en una realización más preferida la sustancia electro- óptica cristal líquido.
Por "cristal líquido (CL)" en la presente invención se entiende aquella sustancia que se encuentra en un estado de agregación de la materia caractenzado por poseer propiedades tanto de un líquido como de un sólido.
Se entiende por "partículas en suspensión (SPD)" en la presente invención, aquellas que varían su índice de refracción al aplicar corriente eléctrica, ya que se ordenan dichas partículas aumentando la tansmitancia.
Por "sustancia electrocrómica" se entiende en la presente invención aquellas sustancias que cambian de color al aplicarles una corriente eléctrica.
En una realización aún más preferida la composición del cristal líquido seleccionada de entre: 4-n-pentíl-4'-cíano-bifenilo (5CB), 4-n-heptíl-4'-ciano- bifenilo (7CB), 4-n-octil-4'-ciano-bifenilo (8CB) y mezclas nemáticas, siendo estas mezclas nemáticas seleccionadas de entre E7 o ZLI-1840 de Merck o ferroeléctricas como la SCE13 de Merck y otras mezclas comerciales.
Por "mezcla nemática" se entiende en la presente invención por aquella fase de cristal líquido que posee la capacidad de ordenarse posicionalmente sus moléculas al aplicarse un campo externo.
Preferiblemente la bíopelícula bacteriana comprende un soporte de óxido conductor transparente (OCT) y una capa de matriz bacteriana. Más preferiblemente el óxido conductor transparente se selecciona de entre óxido de indio y estaño (ITO), óxido de indio y cinc (IZO), óxido de flúor o estaño (FTO), siendo aún más preferiblemente ITO, cualquiera de estos depositados sobre un vidrio o polímero transparente, el cual le sirve de soporte.
Por otra parte la capa de matriz bacteriana se selecciona preferiblemente de entre las producidas por Pseudomona putida, Escherichia coli, Pseudomona aeruginosa, Pseudomona fluorescens y Bacillus subtilis, y siendo más preferiblemente la Pseudomona putida.
La biopelícula bacteriana tiene más preferiblemente un espesor de entre 0,5 y 50 μηι.
Las biopelículas producidas por el P. putida mt-2 cumplen los requisitos de macroporosídad (poros del orden de mícrómetros) y un índice de refracción similar o igual al del cristal líquido, cuando está orientado por el campo eléctrico. Otras bacterias como E. coli y P. aeruginosa también tienen la capacidad para formar biopelículas que cumplen todos los requisitos. La robustez ambiental, la inocuidad y la versatilidad metabólica de P. putida hacen de este organismo la mejor opción para aplicaciones biotecnológicas e industriales.
En los ejemplos descritos en la presente invención, se ha utilizado la glicerina (medio mínimo M9 + glícerol 0,2%) como fuente de carbono para el crecimiento y mantenimiento de las bacterias, sin embargo pueden utilizarse otros como, medio mínimo M9 + fructosa 0,2%, medio mínimo M9 + glucosa 0,2%, medio mínimo M9 + citrato 0,2%.
Más preferiblemente la fuente de C para el cultivo de las bacterias es la glicerina, ya que es un subproducto común de las industrias de aceite vegetal (bíodiesel, jabones), representando por tanto una oportunidad para un reciclado de este material de desecho industrial de cara a aumentar la sostenibilidad medioambiental.
En una realización preferida los vidrios conductores recubiertos con las biopelículas se encuentran unidos por la cara en la que se encuentra la biopelícula bacteriana formando un sándwich en el cual está incorporado el CL. Este cristal líquido, o cualquier otra sustancia electro-óptica queda confinada dentro de la estructura micrométrica de la matriz bacteriana.
Un segundo aspecto de la presente invención se refiere a un procedimiento de obtención del material según se ha descrito anteriormente, que comprende la adición de una sustancia electro-óptica en la biopelícula bacteriana, donde el método de adición se selecciona de entre capilarídad, espray, inmersión,
centrifugación o impregnación; siendo preferiblemente el método de adición por capílaridad.
Preferiblemente además se realiza un tratamiento térmico a temperaturas entre 50 y 120QC, este tratamiento térmico se realiza más preferiblemente en una atmosfera de un gas que se selecciona entre aire, nitrógeno, argón helio o cualquiera de sus combinaciones.
La biopelícula bacteriana empleada para la fabricación del dispositivo electro- óptico descrito en la presente invención, se obtiene preferiblemente por inmersión del vidrio recubierto con el óxido conductor transparente en un medio de cultivo bacteriano. Posteriormente la biopelícula bacteriana formada se fija preferiblemente mediante lavado con un agente entrecruzante que se selecciona de la lista que comprende: AICI3, AI(N03)3, CaCI2, Ca(N03)2 y FeCI3, siendo más preferiblemente dicho agente entrecruzante el AICI3.
Una vez fijada la película bacteriana sobre el óxido semiconductor transparente en una realización preferida se seca a temperaturas entre 80 y 1509C durante 5 a 30 min.
La presente invención describe un material electro-óptico cuyo objetivo es la fabricación de un dispositivo electro-óptico conjugando las propiedades de las biopelículas y las de los cristales líquidos, por lo que en la presente invención se obtiene preferiblemente un dispositivo de cristal líquido disperso en películas bacterianas (BF-DLC de "Bacterial Flim Dispersed Liquid Cristal").
En un tercer aspecto, la presente invención se refiere al uso del material según se ha descrito anteriormente, para la fabricación de un dispositivo electro- óptico.
Preferiblemente el dispositivo electro-óptico es un objeto de transparencia controlable por campo eléctrico. Estos dispositivos electro ópticos pueden ser
ventanas de transparencia controlable, pantalla o panel de separación interior en edificios, un conmutador óptico, un dispositivo de protección o regulación de luminosidad solar y/o artificial.
A lo largo de la descripción y las reivindicaciones la palabra "comprende" y sus variantes no pretenden excluir otras características técnicas, aditivos, componentes o pasos. Para los expertos en la materia, otros objetos, ventajas y características de la invención se desprenderán en parte de la descripción y en parte de la práctica de la invención. Los siguientes ejemplos y dibujos se proporcionan a modo de ilustración, y no se pretende que sean limitativos de la presente invención.

EJEMPLOS
A continuación se ilustrará la invención mediante unos ensayos realizados por los inventores, que pone de manifiesto la especificidad y efectividad del
material con propiedades electro-ópticas descritos en la presente invención, su procedimiento de obtención y sus usos como dispositivos electro-ópticos.
El material extracelular formado por las bacterias que se utiliza para unir las células en las biopelículas, está compuesto principalmente por polísacáridos, con un índice de refracción de aproximadamente 1 ,54. La morfología de la biopelícula incluye los huecos internos y canales que permiten el flujo de nutrientes, enzimas, metabolitos, productos de desecho y otros solutos, a través del sistema, proporcionando una estructura macroporosa a las biopelículas, como muestra la figura 1. La singularidad del dispositivo, descrito de la presente invención, se encuentra en el uso de la biotecnología para producir una matriz con la macroporosidad necesaria y un índice de refracción que se corresponde muy de cerca el índice de refracción ordinario (nord) del CL nematico utilizado (5CB), con un nord de 1.53-1.54 (a λ = 589 nm) en todo el rango de temperaturas del cristal líquido.
El procedimiento seguido para la fabricación del material electro-óptico de biopelícula-CL está representado en la figura 2. Las biopelículas han sido crecidas en la superficie conductora de vidrios recubiertos con ITO (óxido de indio y estaño). Se encontró que el tratamiento previo de estas superficies conductoras con KOH y el crecimiento de una capa muy delgada del óxido de titanio (Tí02), son muy eficaces para mejorar la adherencia y la homogeneidad de las biopelículas. Las biopelículas crecen en el medio de cultivo bacteriano durante 5-7 días y luego se fija mediante el lavado con una solución acuosa de AICI3 que entrecruza la matriz. Los dispositivos fueron construidos haciendo un sándwich entre dos biopelículas, que se apretan fuertemente. De este modo, se obtiene una matriz microporosa de aproximadamente 1 ,0 a 1 ,5 μιη de espesor entre los vidrios conductores. El CL fue incorporado en la matriz de la biopelícula mediante llenado capilar, seguido de un tratamiento térmico con aire caliente para asegurar el humedecimiento completo de la matriz de la biopelícula con las moléculas de CL.
El funcionamiento de los dispositivos construidos fue probado mediante la aplicación de tensiones alternas (frecuencia = 1 04 Hz) a los vidrios conductores (para generar un campo eléctrico), la operación del dispositivo se muestra en la figura 3. Como puede verse en la fotografía, cuando no se aplica voltaje se obtiene un estado opaco debido a la dispersión de la luz, mediante la aplicación del campo eléctrico se logra una transparencia virtualmente completa. Antes de aplicar el campo eléctrico los dispositivos muestran un estado opaco (4-7% de transmisión). Un estado muy transparente (70-80% de transmisión) se puede lograr con bajos voltajes efectivos (<1 0 V). Los tiempos de conmutación, (respuesta, ÍON) y (relajación, ÍOFF), son del orden de 0, 1 -3 ms para ÍO Y 1 0-30 ms para ÍOFF, en todo el rango de voltajes aplicados.
El dispositivo biopelícula-LC de la presente invención, muestra unas características de funcionamiento muy buenas: alto contraste y una buena estabilidad.
Métodos
Sustratos. Se han utilizado vidrios recubiertos con óxido de indio y estaño (ITO) como sustratos, cepas, medios de crecimiento, condiciones de crecimiento y la formación de biopelículas. Para la formación de biopelículas, las células fueron cultivadas a 30 9C en cajas de estériles de 12 x 1 0 x 3 cm con 35 mi de medio mineral sintético M9 (Miller, 1972) con 0.2% de glícerol como fuente de carbono. Los vidrios fueron introducidos en el medio de cultivo en horizontal, con la superficie conductora hacia arriba. El medio de cultivo fue inoculado a una densidad óptica de 0,1 y el crecimiento de las células se midió a 600 nm (OD 600). Para su uso posterior en el dispositivo óptico, las biopelículas crecidas durante 6 días se lavaron con AICI3 0, 1 M y se secaron a 1 00 SC durante 15 minutos. La formación de biopelículas fue cuantificada usando el método de cristal violeta.
Montaje de dispositivos. Dos vidrios cubiertos con biopeitculas se unieron y pegaron formando un sándwich con las caras de biopelículas hacia adentro. El CL se introdujo en el sándwich de la matriz de la biopelícula mediante llenado capilar, a continuación, se aplicó un tratamiento térmico con aire caliente a 50eC durante 20 minutos para asegurar la impregnación completa de la matriz de biopelícula con el CL.
Las pruebas de funcionamiento. La transmitancia del dispositivo se ha estudiado mediante la medición de la intensidad del haz de un láser de He-Ne λ = 638,8 nm, 5 mW) antes y después de pasar por el dispositivo con un fotodetector conectado a un osciloscopio. Los campos eléctricos se generaron mediante la aplicación de tensiones AC (f=104 Hz) al dispositivo, las tensiones eficaces (rms) se midieron con el osciloscopio. La respuesta y los tiempos de relajación se midieron mediante el análisis de las señales en el osciloscopio.
Claims
1. Material que comprende al menos una biopelícula bacteriana y una sustancia electro-óptica.
2. Material según la reivindicación 1 , donde la sustancia electro-óptica es cristal líquido.
3. Material según cualquiera de las reivindicaciones 1 a 2, donde la composición del cristal líquido se selecciona entre: 4-n-pentil-4'-cíano- bifenilo, 4-n-heptil-4'-ciano-bifenilo, 4-n-octil-4'-ciano-bifenilo y mezclas nemáticas.
4. Material según la reivindicación 3, donde el cristal líquido es 4-n-pentil-4'- ciano-bifenilo.
5. Material según cualquiera de las reivindicaciones 1 a 4, donde la biopelícula bacteriana está depositada sobre un soporte de óxido conductor transparente.
6. Material según cualquiera de las reivindicaciones 1 a 5, donde el óxido conductor transparente se selecciona de entre ITO, IZO y FTO.
7. Material según la reivindicación 6, donde el conductor transparente es ITO.
8. Material según cualquiera de las reivindicaciones 1 a 7, donde la capa de biopelícula bacteriana se selecciona de la lista que comprende: Pseudomona putída, Escherichia coli, Pseudomona aeruginosa, Pseudomona fluorescens y Bacillus subtilis
9. Material según la reivindicación 8, donde la bacteria es Pseudomona putida.
10. Material según cualquiera de las reivindicaciones 5 a 9, donde el espesor de la biopelícula es de entre 0.5 a 50 μιη.
11. Material según cualquiera de las reivindicaciones 1 a 10, que comprende dos biopelículas bacterianas, donde dichas películas se encuentran unidas por la biopelícula bacteriana y dicha biopelícula comprende cristal líquido.
12. Procedimiento de obtención del material según cualquiera de las reivindicaciones 1 a 11 , que comprende la adición de sustancia electro- óptica a biopelícula bacteriana, donde el método de adicción se selecciona de entre capilaridad, espray, inmersión, centrifugación o impregnación.
13. Procedimiento según reivindicación 12 donde el método de adición de la sustancia electroóptíca es capilaridad.
14. Procedimiento según cualquiera de las reivindicaciones 12 ó 13, donde además se realiza un tratamiento térmico a temperaturas entre 50 y 120°C.
15. Procedimiento según la reivindicación 14, donde el tratamiento térmico se realiza con un gas que se selecciona entre aire, nitrógeno, argón, helio o cualquiera de sus combinaciones.
16. Procedimiento según cualquiera de las reivindicaciones 12 a 15, donde la biopelícula bacteriana se obtiene por inmersión del óxido conductor transparente en un medio de cultivo bacteriano.
17. Procedimiento según la reivindicación 16, donde la biopelícula bacteriana formada se fija mediante lavado con un agente entrecruzante que se selecciona de la lista que comprende: AlC , AI(N03)3, CaC , Ca(N03)2 y FeCI3.
18. Procedimiento según la reivindicación 17, donde el agente entrecruzante es AICI3.
19. Procedimiento según cualquiera de las reivindicaciones 16 a 18, donde la biopelícula bacteriana se seca a temperaturas entre 80 y 150°C durante 5 a 30 min.
20. Uso del material según cualquiera de las reivindicaciones 1 a 11 , para la fabricación de dispositivos electro-óptico.
21. Uso según la reivindicación 20, donde el dispositivo electro-óptico es una objeto de transparencia controlable.
