Abstract
Se describe un dispositivo tipo MIR para la realización de determinación de medidas y análisis de partículas mediante medios ópticos, especialmente de partículas de tamaño suprananométrico, que se encuentran en una suspensión inyectada en dicho dispositivo. Asimismo se describe un sistema de análisis de partículas que hace uso de dicho dispositivo conectado a una fuente de luz y a una unidad lectora.

DISPOSITIVO Y SISTEMA DE CONTABILIZACIÓN Y ANÁLISIS DE PARTÍCULAS Y USO DE DICHO SISTEMA
D E S C R I P C I Ó N
OBJETO DE LA INVENCIÓN
La presente invención se refiere al campo técnico de la detección, análisis y contabilización de partículas mediante un sistema optofluídico desechable al que se acopla una fuente de luz y una unidad lectora.
ANTECEDENTES DE LA INVENCIÓN
Hoy en día existen diversas soluciones para el problema de detectar y analizar partículas de tamaño muy pequeño, más concretamente de tamaño suprananométrico. Por ejemplo la celda de Neubauer se utiliza comúnmente para la contabilización de partículas (principalmente células). Sin embargo, éste adolece de imprecisiones debidas a la subjetividad del usuario, así mismo, una única medida no es suficiente para confirmar un recuento, ya que es posible tener una distribución no uniforme en la celda que de un numero erróneo. Así, es necesaria la repetición aleatoria a fin de compensar dicho problema.
La citometría de flujo es un sistema de recuento en serie (medida consecutiva) que permite una determinación más exacta que la celda de Neubauer. Sin embargo, requiere unos tiempos de medida largos, así
como un equipamiento complejo y caro. Además, ninguno de los dos permite el análisis simultáneo de las partículas.
Si se supone un fluido con dos tipos de partículas suprananométricas: absorbentes y no absorbentes a algunas determinadas longitudes de onda. Los espectrofotómetros podrían determinar la densidad óptica de dichas partículas sólo si su longitud de onda de trabajo coincide con las bandas de absorción de las partículas, en caso contrario, son difícilmente detectadas. Los sistemas LUCAS detectan ambos tipos de partículas, pero no permiten discernirlas entre ellas (cuando las dimensiones entre ambas son comparables) ni determinar sus propiedades. Los citómetros de flujo si permiten su diferenciación, pero la medida es en serie y consecutiva, necesitando así largos tiempos totales de interrogación de un volumen definido de muestra
DESCRIPCIÓN DE LA INVENCIÓN
El sistema objeto de la invención permite tanto el análisis como la detección de partículas inyectadas en su interior, como pueden ser las células, independientemente de su posible mareaje ya que dispone de diferentes métodos de análisis que le permite optimizar la medida en función de las propiedades de las partículas a ser medidas. Además, el sistema presentado permite el análisis en continuo, pudiéndose determinar diferenciaciones o variaciones en las partículas suprananométricas- Así mismo, los métodos de análisis permiten tanto medidas uniparamétricas (una única magnitud) como multiparamétricas (diferentes magnitudes) en tiempos de análisis de aproximadamente 30 milisegundos.
El objeto principal de la invención es la utilización de un sistema de
análisis y detección de partículas que dispone de un dispositivo de reflexión interna múltiple (MIR por sus siglas en inglés), en adelante dispositivo MIR, para la medida y la detección multiparamétrica casi simultánea de partículas, dispositivo que además es desechable o reutilizable. Dicho sistema consiste en una celda fluídica en la cual se pueden introducir fluidos mediante unas entradas fluídicas para ser analizados mediante un análisis en el que se acopla una fuente de luz, como una fibra óptica, a dicha celda, permitiendo determinar las propiedades ópticas del fluido inyectado, y por extensión, de las partículas que hubiese dispersadas en el mismo.
Una de las características diferenciadoras del sistema objeto de la invención reside en el uso de una fuente de amplio espectro en la inyección y un sistema de medida espectral en la recogida, de ésta manera, es posible la detección multiparamética en una única medida. Estas dos propiedades permiten solventar los problemas inherentes a los espectrofotómetros actuales (los cuales miden la densidad óptica a una longitud de onda fijada), los citómetros de flujo (con una medida en serie) y a las configuraciones basadas en la contabilización por reconocimiento de sombras (LUCAS en inglés, que permiten su contabilización pero no su análisis).
El sistema objeto de la invención consiste en la utilización de un dispositivo de reflexión interna múltiple para la detección, análisis y/o conteo de partículas suspendidas en un líquido. Dicho dispositivo MIR se encuentra definido en un chip y comprende unos espejos de aire definidos por unas estructuras huecas en forma de coliso curvado en las cercanías de la región de análisis, correspondiente a la denominada zona de interrogación que es la zona donde interacciona la luz con el líquido a ser analizado por el sistema. Los espejos de aire hacen que la luz se propague en una trayectoria de zig-zag, permitiendo la elongación del camino óptico manteniendo las dimensiones del sistema en unos márgenes razonables. El dispositivo citado comprende varios elementos adicionales tales como
canales de autoalineamiento o alineamiento automático, los anteriormente citados espejos de aire y unas microlentes para rectificación del haz de luz preferiblemente alojadas en dichos canales de autoalineamiento; el dispositivo objeto de la invención se encuentra definido por una sola máscara fotolitográfica, que puede ser de aplicación en materiales de bajo coste como materiales poliméricos, tal y como podría ser PDMS.
Los sistemas de análisis, detección o conteo de partículas generalmente trabajan en uno de los tres siguientes regímenes: LS ("large scattering" dispersión con ángulos de 15-150°), LS+ABS ("scattering" dispersión + absorción) y ABS (absorción). Por el contrario, El hecho diferencial de ésta patente es que el sistema objeto de la invención puede trabajar simultáneamente los tres regímenes anteriormente mencionados. Al contrario de lo que ocurre con los citómetros, donde las partículas suprananométricas, normalmente células, son contadas de forma secuencial, el sistema objeto de la invención realiza una única medición durante 30 ms en toda la región; en el caso de que las células no se encuentren marcadas o no tengan bandas de absorción, se obtiene el espectro de dispersión; si las células se encuentran marcadas se observa una banda de absorbancia superimpuesta. En ambos regímenes LS y ABS+LS se puede contabilizar el número de partículas presentes.
El sistema puede obtener el espectro relativo solo a la absorción (ABS) mediante la substracción de los dos resultados mencionados en el párrafo anterior. Así mismo el sistema no solo permite contabilizar la población de células, si no que además permite establecer una tasa de células marcadas/sin marcar utilizando dos o más marcadores diferentes. Adicionalmente, si se produce una diferenciación de dichas partículas (como podría ser debido a un crecimiento celular o a una modificación de
las propiedades de las mismas) también sería detectado por el sistema propuesto.
Un factor adicional en el sistema objeto de la invención del cual no dispone ninguno de los sistemas actuales es su portabilidad. El sistema objeto de la invención puede ser fabricado tanto en tecnología microelectrónica como en tecnología de polímero, tal y como se ha descrito más arriba. Una vez definida la geometría, y una vez conocidos los índices de refracción de los materiales a ser utilizados, la fabricación de dichos sistemas requiere una complejidad mínima
DESCRIPCIÓN DE LOS DIBUJOS
Para complementar la descripción que se está realizando y con objeto de ayudar a una mejor comprensión de las características de la invención, de acuerdo con un ejemplo preferente de realización práctica de la misma, se acompaña como parte integrante de dicha descripción, un juego de dibujos en donde con carácter ilustrativo y no limitativo, se ha representado lo siguiente: Figura 1.- Muestra una vista en 3D del sistema objeto de la invención.

REALIZACIÓN PREFERENTE DE LA INVENCIÓN
A la vista de las figuras se describe a continuación un modo de realización preferente del dispositivo (1 ) objeto de esta invención.
Para la realización de este ejemplo de realización se utiliza un dispositivo (1 ) fabricado mediante técnicas litográficas sobre un cuerpo transparente (10) polimérico en el que, tal y como se observa en la figura 1 , se han definido unos canales de autoalineamiento (3) que albergarán unas fibras ópticas de emisión y de recepción y unas microlentes (8,13) que se encuentran ubicadas al final de dichos canales de autoalineamiento (3), a su vez se definen unos espejos de aire (2,12) conformados mediante unos colisos curvados huecos, unas entradas fluídicas (4) de fluido entre las que se encuentra definido un canal (11 ) cuyo recorrido central comprende varios tramos (5,6,7) donde los espejos de aire (2) se encuentran definidos en paralelo a los lados más cortos de un segundo tramo (6) del canal (11 ).
En una realización preferente se usa como fuente de luz una fuente de luz de banda ancha, como puede ser la Ocean Optics HL-2000, acoplada a una fibra óptica de entrada multimodo de un diámetro de 230 μηπ. Se incorpora así mismo una unidad de lectura, un espectrómetro, al cual va conectada una fibra óptica de salida idéntica a la anterior que lleva la señal al espectrómetro, como puede ser un Ocean Optics HR4000 con una resolución espectral de 0.2 nm. Un tiempo de análisis de 30 ms permite obtener los espectros de las partículas inyectadas. El experimento se realiza en una habitación con temperatura controlada.
Primeramente se llenan con solución PBS las entradas fluídicas (4) y el canal (11 ) que comprende unas trayectorias curvas estrechas en las zonas adyacentes a dichas entradas fluídicas (4) y una trayectoria central en zig-zag definida por la sucesión de los tramos (5,6,7) romboidales; una vez llenado el dispositivo (1 ) se realiza una primera medición emitiendo un haz de luz con la fuente de luz que atraviesa una microlente (8) ubicada en el canal de autoalineamiento (3) que alberga la fibra óptica de emisión y se
toma la medida con la unidad de lectura de salida para establecer una medida de referencia en estas condiciones, que será utilizada como medida de referencia para el resto de las mediciones Para la realización de mediciones en LS y en LS+ABS se inyectan en el dispositivo (1 ) concentraciones en disolución de células vivas (sin marcar) o muertas (marcadas) en concentraciones variables entre 50 y 2000kcélulas/ml. El marcador utilizado con las células muertas es azul tripan ya que es posible su uso a temperatura ambiente con un pico de absorción situado a una longitud de onda de 581 nm. Para cada concentración de células se realizan 10 escaneos consecutivos. Una vez realizados las mediciones con la concentración más alta, se vuelve a inyectar solución PBS para determinar posibles fluctuaciones en la señal de referencia. La realización de las mediciones se realiza mediante la introducción de las fibras ópticas, una de emisión conectada a la fuente de luz y otra de recepción conectada a la unidad lectora, en los canales de autoalineamiento (3) donde la fibra óptica de emisión conectada a la fuente de luz emite un haz de luz que atraviesa una primera microlente (8), que se encuentra ubicada al final del canal de autoalineamiento (3) que alberga la fibra óptica de emisión, para luego entrar en un primer tramo (5) de forma romboidal del canal (11 ), atravesando el fluido que se encuentra en dicho primer tramo (5) que contiene las células anteriormente inyectadas, el haz de luz emitido atraviesa a su vez parte del cuerpo (10) hasta verse reflejado por la acción de un primer espejo de aire (2) ubicado en paralelo al primer tramo (5) cuyo centro de curvatura se encuentra dispuesto en la dirección del eje longitudinal del canal de alineamiento (3) que alberga la fibra óptica de emisión. El haz reflejado en el espejo de aire (2) atraviesa un segundo tramo (6) con forma romboidal del canal (11 ) en cual el haz de luz reflejada define una zona de interrogación (9), correspondiente a la zona donde se realiza el
cruce de los haces de luz y donde se realiza el análisis que se puede apreciar en detalle en la figura 2, antes de reflejarse de nuevo en un segundo espejo de aire (12) para atravesar un tercer tramo (7) también de forma romboidal hasta llegar a una segunda microlente (13) ubicada en el canal de alineamiento (3) que alberga la fibra óptica de recepción conectada al espectrómetro. Dicho espectrómetro recibe el haz de luz que atraviesa el fluido y ha sido reflejado por los espejos de aire (2,12)
Claims
1. Dispositivo (1 ) de contabilización y análisis de partículas caracterizado porque consiste en un cuerpo transparente (10) que
5 comprende:
- unas entradas fluídicas (4) definidas en sus esquinas entre las cuales discurre un canal (11 ) que comprende dos primeros tramos curvos de paredes paralelas conectados a dichas entradas fluídicas (4) y de sección menor a tres tramos romboidales (5,6,7) contiguos definidos 0 entre dichos primeros tramos,
- unos espejos de aire (2,12) definidos por unas estructuras huecas en forma de coliso curvado que se encuentran ubicados a ambos lados de un segundo tramo (6) del canal (11 ) que definen una zona de interrogación (9) en dicho segundo tramo (6),y
5 - unos canales de autoalineamiento (3), definidos en su interior, encargados de albergar unas fibras ópticas.
2. Dispositivo (1 ) según reivindicación 1 caracterizado porque adicionalmente comprende unas microlentes (8,13) que se o encuentran respectivamente ubicadas en los extremos de los canales de autoalineamiento (3).
3. Dispositivo (1 ) según reivindicación 2 caracterizado porque las microlentes son cilindricas.

4. Dispositivo (1 ) según un cualquiera de las reivindicaciones anteriores caracterizado porque el cuerpo (10) es de un material que se selecciona de entre los siguientes: un material polimérico, un material cerámico, un material semiconductor, un material aislante y un 0 material conductor.
5. Dispositivo (1 ) según reivindicación 4 caracterizado porque el material polimérico es PDMS.
6. Dispositivo (1 ) según reivindicación 5 caracterizado porque el PDMS está funcionalizado.
7. Sistema de contabilización y análisis de partículas caracterizado porque comprende el dispositivo (1 ) descrito en una cualquiera de las reivindicaciones anteriores y porque adicionalmente comprende unas fibras ópticas en el interior de los canales de autoalineamiento (3) respectivamente conectadas a una fuente de emisión de luz y a una unidad de lectura.
8. Sistema según reivindicación 7 donde la fuente de emisión de luz es una fuente de luz de banda ancha.
9. Sistema según reivindicación 7 u 8 donde la unidad de lectura es un espectrómetro.
10. Sistema según una cualquiera de las reivindicaciones 7 a 9 caracterizado porque adicionalmente comprende unos medios de inyección de fluido conectados al dispositivo (1 ).
11. Uso del sistema descrito en las reivindicaciones 7 a 10 para la detección de partículas en suspensión en fluidos.
12. Uso del sistema descrito en las reivindicaciones 7 a 10 para la contabilización de partículas en suspensión en fluidos.
13. Uso del sistema descrito en las reivindicaciones 7 a 10 para el análisis de partículas en suspensión en fluidos.
14. Uso del sistema descrito en las reivindicaciones 7 a 10 para el análisis de partículas susceptibles de tener diferenciación o alteración temporal tanto en su número, como morfología o variación de sus propiedades ópticas.
15. Uso del sistema según una cualquiera de las reivindicaciones 11 a 13 caracterizado porque las partículas se encuentran sin marcar.
