Abstract
【課題】短時間で製造することができ、PET法に好適に用いることができ、シクロオキシゲナーゼ2のイメージングが可能な、標識NSAIDs化合物及びその製造方法を提供する。 【解決手段】 本発明のアイソトープ標識2-アリールプロピオン酸類は、下記一般式(1)(式中Arは置換基を有することがあるアリール基を示し、R1は11CH3、CH2
18F及びCF2
18Fのいずれかであり、R2は水素又は分枝を有することがあるアルキル基を示す。ただし、Arがベンゼン環でありR1が11CH3でありR2が水素である化合物は除く。)又はその薬学上許容される塩、水和物、溶媒和物若しくはプロドラッグからなる。
Description
本発明はアイソトープ標識2-アリールプロピオン酸類及びその製造方法並びに陽電子放射断層画像撮像(以下「PET」という)用プローブ及びそれを用いたシクロオキシゲナーゼのイメージング方法に関する。本発明のアイソトープ標識2-アリールプロピオン酸類及びその製造方法は、陽電子放射断層撮像法(以下「PET法」という)に用いる分子プローブ及びその製造方法として、好適に用いることができる。また、本発明分子プローブを用いることにより、シクロオキシゲナーゼのイメージングを行なうことができる。
解熱・鎮痛や抗炎症剤として広く利用されている非ステロイド性抗炎症薬(以下「NSAIDs」という)の多くは、主にシクロオキシゲナーゼと呼ばれる酵素を阻害することにより薬理作用を示す。シクロオキシゲナーゼは、痛み、発熱、慢性および急性炎症、関節炎、大腸がん、血管新生、喘息、動脈硬化、アルツハイマー病、パーキンソン病など、非常に多くの病態と関与している。このため、NSAIDsの生体内での挙動を調査することで、多くの病態の解明や診断が可能になると考えられている。
以上の理由によりNSAIDsの体内での挙動をPET法によって調べようという試みが、近年、活発になされている。PET法とは、11Cや18Fなどのポジトロンを放出する短寿命放射性核種で標識されたトレーサーを生体内に投与し、トレーサーにより発生するγ線をPETカメラ(ガンマ線シンチレーターと光電子増倍管からなる検出器)によって計測して、その体内分布をコンピューターにより画像化する方法である。PET法は、小動物からヒトまで含めた生体での薬剤の薬物挙動や標的部位への到達度を、非侵襲的かつ定量的に追跡することができる。イブプロフェンやナプロキセンなどのNSAIDsは、体内でシクロオキシゲナーゼという酵素の働きを阻害して抗炎症作用を示すが、シクロオキシゲナーゼは炎症のみならず、腫瘍やアルツハイマー病などに関与していることが分かっている。このため、NSAIDsをPET法へ適用すべく、数々のNSAIDsが11Cや18Fにより標識化されている(非特許文献1~15)。NSAIDsについてのPET画像を解析することにより、シクロオキシゲナーゼの非侵襲的イメージングが可能となり、ひいては生物学、医薬品開発、医療などの各分野において極めて有用な情報を得ることができるからである。

上述のように、NSAIDsの体内での挙動をPET法によって調べようという試みが世界中でなされている。しかしながら、これまでのところ、いくつかの化合物が炎症部位や腫瘍部位に集積することは確認されているものの、シクロオキシゲナーゼに対する特異的な結合であることを実証できていない。
このように、これまでNSAIDsの体内での挙動についての満足のいくPET画像が得られていない原因として、時間的な制約が挙げられる。すなわち、PET法で使用される短寿命放射性核種としては11Cや18Fなどが用いられ、これらの放射性核種で標識された化合物がトレーサーとして用いられる。11Cは有機化合物中に存在している炭素原子を利用しているため適用範囲が極めて広く、理想的な放射性核種ともいえるが、半減期が20分と短く、合成からPET法での測定までを極めて短時間で行なわなければならない。また、18Fの半減期は110分であり、11Cよりも長いとはいうものの、やはり合成からPET法での測定までを短時間で行なわなければならない。こうした時間的な制約という困難さから、合成方法や、合成される標識化合物の数や種類に制限が加わり、現在に至るまで、世界中で多くの試みが行なわれたにもかかわらず、NSAIDsについての満足できるPET画像が得られていないのである。
本発明は、上記従来の実情に鑑みてなされたものであり、短時間で製造することができ、PET法に好適に用いることができ、シクロオキシゲナーゼのイメージングが可能な、標識NSAIDs化合物及びその製造方法を提供することを目的とする。
本発明者らは、上記従来の問題点を解決すべく、標識NSAIDs化合物の中でも2-アリールプロピオン酸系に分類されているイブプロフェン、ナプロキセンなどの2-アリールプロピオン酸およびその誘導体に注目し、その前駆体としてベンジル位に水素を有するアリール酢酸を用い、[11C]メチル基を極めて短時間で導入し、精製することに成功した。そして、さらには、このアイソトープ標識2-アリールプロピオン酸類をトレーサーとしてPET法を行ったところ、標識された薬剤がシクロオキシゲナーゼの発現部位に集積し、特異的な結合を示すことについて、世界で初めて成功した。
すなわち、本発明のアイソトープ標識2-アリールプロピオン酸類は、下記一般式(1)(式中Arは置換基を有することがあるアリール基を示し、R1は11CH3、CH2
18F及びCF2
18Fのいずれかであり、R2は水素又は分枝を有することがあるアルキル基を示す。ただし、Arがベンゼン環でありR1が11CH3でありR2が水素である化合物は除く。)又はその薬学上許容される塩、水和物、溶媒和物若しくはプロドラッグからなることを特徴とする。
本発明のアイソトープ標識2-アリールプロピオン酸類は、共に電子吸引基となるカルボニル基とアリール基に挟まれた炭素にアイソトープ標識されたR1が導入されている。このため上記一般式(1)におけるR1が水素とされた前駆体から、カルボアニオンを経由して容易にアイソトープ標識されたR1を導入することができる。この反応は、瞬時に進行する上、エステル加水分解は1分以内に終了するため、二段階合成であるものの短時間に合成を完了することができる。このため、合成されるまでの時間によってアイソトープの放射能が減衰される率が少なく、PET法に有効な分子プローブとして用いることができる。
なお、R2としては、炭素数が1~6のアルキル基が好ましい。

本発明のアイソトープ標識2-アリールプロピオン酸類には、カルボン酸の他、塩、水和物、溶媒和物及びプロドラッグが含まれる。ここで、プロドラッグとは、生体内で加水分解されて本発明のアイソトープ標識2-アリールプロピオン酸を生成する化合物をいう。本発明のプロドラッグには、当業者に知られたプロドラッグ化のすべての手法で製造される化合物が含まれる。例えば、カルボキシル基またはアミノ基等を有する場合、それらの基を生体内で容易に加水分解されうるエステル基またはアミド基等に誘導した化合物が、プロドラッグに相当する。具体的にはアイソトープ標識2-アリールプロピオン酸に存在するカルボキシル基を、メチル、エチル等のアルキル、メチルオキシメチル、エチルオキシメチル、2-メチルオキシエチル、2-メチルオキシエチルオキシメチル等のアルキルオキシアルキル、ピバロイルオキシメチル、アセチルオキシメチル、シクロヘキシルアセチルオキシメチル、1-メチルシクロヘキシルカルボニルオキシメチル等のアシルオキシメチル、エチルオキシカルボニルオキシ-1-エチル等のアルコキシカルボニルアルキル、シクロヘキシルオキシカルボニルオキシ-1-エチル等のシクロアルキルオキシカルボニルアルキル等に誘導した化合物が挙げられる。また、アリール基がアミノ基を有している場合は、そのアミノ基をアセトアミド等に誘導した化合物が挙げられる。
また、薬学上許容される塩としては、塩基付加塩および酸付加塩が挙げられる。塩基付加塩としては、ナトリウム塩、カルシウム塩等の無機塩基塩、メグルミン塩、トリスヒドロキシメチルアミノメタン塩等の有機塩基塩が挙げられる。酸付加塩としては、例えば塩酸塩、臭化水素酸塩、リン酸塩、硫酸塩、硝酸塩等の無機酸塩、ギ酸塩、酢酸塩、プロピオン酸塩、マレイン酸塩、フマル酸塩、コハク酸塩、乳酸塩、リンゴ酸塩、酒石酸塩、クエン酸塩、アスコルビン酸塩、マロン酸塩、シュウ酸塩、グリコール酸塩、フタル酸塩、ベンゼンスルホン酸塩等の有機酸塩が挙げられ、また、上記の塩を組み合わせて用いることもできる。
R1は11CH3とすることができる。
本発明のアイソトープ標識2-アリールプロピオン酸類は、次の方法により、迅速かつ簡単に合成することができる。すなわち、本発明のアイソトープ標識2-アリールプロピオン酸類の製造方法は、下記一般式(2)で示される2-アリール酢酸エステル(式中Arは置換基を有することがあるアリール基を示し、R2は水素又は分枝を有することがあるアルキル基を示し、R3は水素、置換基を有することがあるアルキル基、又は置換基を有することがあるアリール基を示す)に塩基を加えて混合するカルボアニオン生成工程と、当該カルボアニオン生成工程後の反応液に11CH3X、CH2
18FX及びCF2
18FXのいずれか(ここでXはI、Br及びトリフレート基のいずれかを示す)を添加するアイソトープ標識導入工程と、を備えることを特徴とする。
本発明のアイソトープ標識2-アリールプロピオン酸類の製造方法では、カルボアニオン生成工程において、上記一般式(2)で示される2-アリール酢酸エステルに塩基が加えられ、混合される。R3の置換基としてはハロゲン原子、カルボキシル基、エステル基、水酸基、チオール基及びアルコキシ基等が挙げられる。また、R3がアルキル基の場合、好ましいのは炭素数が6~10のアルキル基である。さらに、R3がアリール基の場合、好ましいのは炭素数が6~10のアリール基である。また、溶媒としては、無水N,N-ジメチルホルムアミド(DMF)、無水テトラヒドロフランなどを用いることができる。塩基としては、水素化ナトリウム、水素化カリウム、リチウム ジイソプロピルアミド、ナトリウム ビス(トリメチルシリル)アミド、リチウム ビス(トリメチルシリル)アミドなどを用いることができる。これにより、アリール基及びカルボニル基に挟まれた炭素に結合している活性な水素が脱離し、カルボアニオンが生成する。そして、さらにアイソトープ標識導入工程において、11CH3X、CH2
18FX及びCF2
18FXのいずれかが添加される。これらの標識化合物は、サイクロトロンで発生させた核種から化学変換することにより容易に合成することができる。これにより、ベンジル位の水素に代わって11CH3、CH2
18F及びCF2
18Fのいずれかが導入される。

なお、非特許文献13中の図2には、アセト酢酸エステル合成の手法を用いた11CH3の導入のアイデアが示されているが、脱炭酸の工程を必要とするなど工程数が多く、この方法によって本発明のアイソトープ標識2-アリールプロピオン酸類を製造するのは、操作が煩雑なだけでなく時間を要するため、PET法に適用するのは困難である。
さらに、非特許文献3中に、[18F]フルルビプロフェンが合成されているが、本発明法は数多くある2-アリールプロピオン酸類の共通部分を同一の方法で標識できるため、適応範囲の広い方法と言える。

本発明のアイソトープ標識2-アリールプロピオン酸類は、PET用分子プローブとして好適に用いることができる。本発明者らは、本発明のアイソトープ標識2-アリールプロピオン酸類をPET法に適用することにより、シクロオキシゲナーゼのイメージングや、アルツハイマー病において神経変性に関与する脳内炎症のイメージングが可能であることを確認している。
本発明のアイソトープ標識2-アリールプロピオン酸類と類似の構造を有するNSAIDsとして、例えば下記構造式及び名称の化合物が挙げられる。
これらの化合物のベンジル位のメチル基を11CH3、CH2
18F及びCF2
18Fのいずれかで置換した化合物は、本発明のアイソトープ標識2-アリールプロピオン酸類に含まれる。こうしたNSAIDsを模した本発明のアイソトープ標識2-アリールプロピオン酸類のPET画像を撮影することにより、上記NSAIDsの生体内での経時的な挙動を調べることができる。
また、アイソトープ標識2-アリールプロピオン酸のプロドラッグとしてエステル体を用いることもできる。エステル体にすることで、実施例において後述するように、カルボン酸の形で投与するよりも脳内移行性を向上させることができ、脳の炎症箇所のイメージングを行なうことができる。
すなわち、ベンジル位に水素を有する2-アリール酢酸エステルを前駆体とし、ベンジル位の水素を塩基で引き抜いてカルボアニオンとする。そして、さらにR1X(ここでR1は11CH3、CH2
18F及びCF2
18Fのいずれかであり、XはI、Br及びトリフレート基のいずれかを示す)と反応させてベンジル位にR1を導入し、さらにこれをアルカリ等で加水分解することによって本発明のアイソトープ標識2-アリールプロピオン酸が得られる。アリール基にアミノ基や水酸基などの活性水素をもつ官能基がある場合は、Protective Groups in Organic Synthesis(T.W.Greenら著)に記された保護・脱保護の工程を利用することができる。所望のアイソトープ標識2-アリールプロピオン酸類としてエステル型の場合には、最終段階の加水分解を行なわなければよい。最終的に得られたアイソトープ標識2-アリールプロピオン酸類の投与液は、生理食塩水にエチレングリコール、エタノール、Tween80、アスコルビン酸、シクロデキストリンなどを加えた溶媒に溶解させることにより調製できる。
以下、本発明を具体化した実施例について詳細に説明する。
[試薬・溶媒]
すべての化学試薬およびNSAIDsは特に記さない限り、市販品をそのまま用いた。NMRスペクトルの測定溶媒としては、重クロロホルム、重ジメチルスルホキシドを使用した。抽出およびクロマトグラフィーの溶出用溶媒としては、酢酸エチル、ヘキサン、ジクロロメタン、メタノールの市販品をそのまま用いた。
[標識化反応]
11Cは、住友重機械工業社製CYPRIS HM-12S Cyclotronを使用し、14N(p,α)11Cの核反応により製造した。[11C]ヨウ化メチル([11C]CH3I)の調製、反応溶液の加熱、希釈、高速液体クロマトグラフィー(HPLC)装置への注入、分取、濃縮、殺菌の一連の操作は、独自に開発した自動合成装置により行った。分取用HPLCは日本分光社製LC-2000を使用した。分析用HPLCは、島津社製Prominenceを使用した。放出される放射能はAloka社製RLC-700放射線測定装置を用いて測定した。HPLC用カラムには、ナカライテスク社製COSMOSIL、またキラルカラムには、ダイセル社製のものを使用した。
[分光計]
1H NMRスペクトルは、内部標準物質としてテトラメチルシラン(TMS)を用い、日本電子株式会社製JEOL JNM-AL-400(400MHz)FT-NMRを用いて測定した。化学シフトδ値は、ppmで表示した。化学結合定数(J)は、Hzで示し、シグナルの分裂様式は、一重線をs、二重線をd、三重線をt、四重線をq、多重線をm、幅広線をbrと略記した。
[試薬・溶媒]

[分光計]

アルゴン雰囲気下、メチル 2-(4-イソブチルフェニル)アセテート(EP347939)(1 mg)の無水DMF (200 μL)溶液に、水素化ナトリウム(1 mg)を添加し攪拌した。そして、[11C]CH3Iを室温で捕獲後すぐに、2 Mの水酸化ナトリウム(300 μL)を加えた。得られた反応溶液を50℃で1分間加熱し、10%ギ酸のアセトニトリル溶液(300 μL)で反応を停止した後、混合液をアセトニトリル(150 μL)、水(150 μL)、および25%アスコルビン酸(50 μL)からなる溶液で希釈した。こうして得られた反応混合物を分取用HPLC (移動相; アセトニトリル:10 mMリン酸ナトリウム緩衝液(pH7.4) = 34:66, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 195 nm)で分離した。その結果、図1に示すように、[11C]イブプロフェン(3)が保持時間19-20分で流出した。目的のフラクションをエバポレーターを用いて減圧下濃縮し、生理食塩水(4.0 mL)、プロピレングリコール(0.3 mL)、Tween80 (0.05 mL)、25%アスコルビン酸(0.2 mL)からなる希釈溶液を添加して投与用溶液とした。
放射能は反応終了時に4.95 GBqであり、比放射能は34 GBq/μmolであった。[11C]CH3Iに基づく崩壊補正後の単離収率は69%であった。また、[11C]イブプロフェン(3)の合成、分離、及び静脈注射液調製の合計時間は約37分であった。[11C]イブプロフェン(3)の同定は、イブプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:1%リン酸水溶液 = 60:40, カラム; COSMOSIL, 5C18-MS-II, 内径 4.6 mm×長さ 150 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 210 nm)。その結果、保持時間は約6分で、純度は99%以上であった。

アルゴン雰囲気下、メチル 2-(4-イソブチルフェニル)アセテート(1 mg)を無水DMF (200 μL)に溶解し、水素化ナトリウム(1 mg)を添加し攪拌した。そして、[11C]CH3Iを室温で捕獲後すぐに、25%アスコルビン酸(50 μL)、アセトニトリル(400 μL)、水(400 μL)からなる溶液で希釈した。こうして得られた反応混合物を分取用HPLC (移動相; アセトニトリル:水 = 75:25, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 195 nm)で分離したところ、図2に示すように、[11C]イブプロフェン メチルエステル(4)が保持時間15-16分で流出した。
放射能は反応終了時に4.00 GBqであり、比放射能は23 GBq/μmolであった。また、[11C]イブプロフェン メチルエステル(4)の合成、分離、及び静脈注射液調製の合計時間は約30分であった。[11C]イブプロフェン メチルエステル(4)の同定は、イブプロフェン メチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:水 = 60:40, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径5 μm, 流速; 1 mL/min, UV検出器; 210 nm)。その結果、保持時間は約6分で、純度は99%以上であった。
放射能は反応終了時に4.00 GBqであり、比放射能は23 GBq/μmolであった。また、[11C]イブプロフェン メチルエステル(4)の合成、分離、及び静脈注射液調製の合計時間は約30分であった。[11C]イブプロフェン メチルエステル(4)の同定は、イブプロフェン メチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:水 = 60:40, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径5 μm, 流速; 1 mL/min, UV検出器; 210 nm)。その結果、保持時間は約6分で、純度は99%以上であった。
実施例1と同様の手順によりメチル2-(6-メトキシナフタレン-2-イル)アセテート(USP3896157)から、[11C]ナプロキセン(5)の合成反応を行った。こうして得られた反応混合物を分取用HPLC(移動相; アセトニトリル:10 mMリン酸ナトリウム緩衝液(pH7.4) = 22:78, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 200 nm)で分離したところ、図3に示すように[11C]ナプロキセン(5)が保持時間19-20分で流出した。
放射能は反応終了時に3.30 GBqであり、比放射能は29 GBq/μmolであった。[11C]ナプロキセン(5)の合成、分離、及び静脈注射液調製の合計時間は、約29分であった。[11C]ナプロキセン(5)の同定は、ナプロキセンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:1%リン酸水溶液 = 50:50, カラム; COSMOSIL, 5C18-MS-II, 内径 4.6 mm×長さ 150 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は5.2分で、純度は99%以上であった。
放射能は反応終了時に3.30 GBqであり、比放射能は29 GBq/μmolであった。[11C]ナプロキセン(5)の合成、分離、及び静脈注射液調製の合計時間は、約29分であった。[11C]ナプロキセン(5)の同定は、ナプロキセンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:1%リン酸水溶液 = 50:50, カラム; COSMOSIL, 5C18-MS-II, 内径 4.6 mm×長さ 150 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は5.2分で、純度は99%以上であった。
実施例2と同様の手順により、メチル-2-(6-メトキシナフタレン-2-イル)アセテートから、[11C]ナプロキセン メチルエステル(6)を合成した。こうして得られた反応混合物をHPLC (移動相; アセトニトリル:水 = 65:35, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 230 nm)で分離したところ、図4に示すように[11C]ナプロキセン メチルエステル(6)が保持時間16.2分で流出した。
放射能は反応終了時に3.30 GBqであり、比放射能は29 GBq/μmolであった。[11C]ナプロキセンメチルエステル(6)の合成、分離、及び静脈注射液調製の合計時間は、約32分であった。[11C]ナプロキセン メチルエステル(6)の同定は、ナプロキセンメチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:水 = 65:35, カラム; COSMOSIL, 5C18-MS-II, 内径 4.6 mm×長さ 150 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 230 nm)。その結果、保持時間は5.5分で、純度は99%以上であった。

実施例1と同様の手順により、メチル 2-(3-ベンゾイルフェニル)アセテート(Org. Lett. 2002, 4, 3083-3085)から、[11C]ケトプロフェン(7)を合成した。こうして得られた反応混合物をHPLC (移動相; アセトニトリル:10 mMリン酸ナトリウム緩衝液(pH7.4) = 30:70, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図5に示すように[11C]ケトプロフェン(7)が保持時間14分で流出した。
放射能は反応終了時に3.40 GBqであり、比放射能は40 GBq/μmolであった。[11C]ケトプロフェン(7)の合成、分離、及び静脈注射液調製の合計時間は、約28分であった。[11C]ケトプロフェン(7)の同定は、ケトプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:1%リン酸水溶液 = 40:60, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は6.8分で、純度は99%以上であった。
放射能は反応終了時に3.40 GBqであり、比放射能は40 GBq/μmolであった。[11C]ケトプロフェン(7)の合成、分離、及び静脈注射液調製の合計時間は、約28分であった。[11C]ケトプロフェン(7)の同定は、ケトプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:1%リン酸水溶液 = 40:60, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は6.8分で、純度は99%以上であった。
実施例2と同様の手順により、メチル 2-(3-ベンゾイルフェニル)アセテートから、[11C]ケトプロフェン メチルエステル(8)を合成した。こうして得られた反応混合物をHPLC (移動相; アセトニトリル:水 = 70:30, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図6に示すように[11C]ケトプロフェン メチルエステル(8)が保持時間12分で流出した。
放射能は反応終了時に5.4 GBqであり、比放射能は46 GBq/μmolであった。[11C]ケトプロフェンメチルエステル(8)の合成、分離、及び静脈注射液調製の合計時間は、約27分であった。[11C]ケトプロフェン メチルエステル(8)の同定は、ケトプロフェン メチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:水 = 55:45, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は6.0分で、純度は99%以上であった。

実施例1と同様の手順により、エチル 2-(3-フェノキシフェニル)アセテート(J. Med. Chem. 2005, 48, 995-1018)から、[11C]フェノプロフェン(9)を合成した。こうして得られた反応混合物をHPLC (移動相; アセトニトリル:10 mMリン酸ナトリウム緩衝液(pH7.4) = 20:80, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 206 nm)で分離したところ、図7に示すように[11C]フェノプロフェン(9)が保持時間16分で流出した。
放射能は反応終了時に5.99 GBqであり、比放射能25 GBq/μmolであった。[11C]フェノプロフェン(9)の合成、分離、及び静脈注射液調製の合計時間は、約40分であった。[11C]フェノプロフェン(9)の同定は、フェノプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:1%リン酸水溶液 = 45:55, カラム; COSMOSIL, 5C18-MS-II, 内径 4.6 mm×長さ 100 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 220 nm)。その結果、保持時間は8.4分で、純度は99%以上であった。

上記合成反応で得られた反応混合物を分取用HPLC (移動相; 0.05% TFA ヘキサン:0.05% TFA エタノール = 95 : 5,カラム; CHIRALPAK AD-H, 内径 10 mm×長さ 250 mm, 粒径 5 μm, 流速; 3 mL/min, UV検出器; 254 nm)で分離したところ、図13に示すように、(R)- [11C]フルルビプロフェン(13R)が保持時間10分で流出した。(R)- [11C]フルルビプロフェン(13R)の同定は、(R)-フルルビプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
上記合成反応で得られた反応混合物を分取用HPLC (移動相; 0.05% TFA ヘキサン:0.05% TFA エタノール = 95 : 5,カラム; CHIRALPAK AD-H, 内径 10 mm×長さ 250 mm, 粒径 5 μm, 流速; 3 mL/min, UV検出器; 254 nm)で分離したところ、図13に示すように、(R)- [11C]フルルビプロフェン(13R)が保持時間10分で流出した。(R)- [11C]フルルビプロフェン(13R)の同定は、(R)-フルルビプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
(S)-[11C]フルルビプロフェン(13S)の同定
一方、上記合成反応で得られた反応混合物を分取用HPLC (移動相; 0.05% TFA ヘキサン:0.05% TFA エタノール = 95 : 5,カラム; CHIRALPAK AD-H, 内径 10 mm×長さ 250 mm, 粒径 5 μm, 流速; 3 mL/min, UV検出器; 254 nm)で分離したところ、図13に示すように、(S)-[11C]フルルビプロフェン(13S)が保持時間13分で流出した。(S)- [11C]フルルビプロフェン(13S)の同定は、(S)-フルルビプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
一方、上記合成反応で得られた反応混合物を分取用HPLC (移動相; 0.05% TFA ヘキサン:0.05% TFA エタノール = 95 : 5,カラム; CHIRALPAK AD-H, 内径 10 mm×長さ 250 mm, 粒径 5 μm, 流速; 3 mL/min, UV検出器; 254 nm)で分離したところ、図13に示すように、(S)-[11C]フルルビプロフェン(13S)が保持時間13分で流出した。(S)- [11C]フルルビプロフェン(13S)の同定は、(S)-フルルビプロフェンの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
エチル 2-(2-フルオロビフェニルー4-イル)アセテート(US3755427)を出発原料とし、実施例2の方法と同様の方法により、[11C]フルルビプロフェン エチルエステル(16)を合成した。こうして得られた反応混合物を分取用HPLC (移動相; アセトニトリル:水 = 80:20, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図14に示すように、[11C]フルルビプロフェンエチルエステル(16)が保持時間16分で流出した。

Bull. 1984, 32, 99-105)の標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:水 = 65:35, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は7.1分で、純度は99%以上であった。

Bull. 1984, 32, 99-105)の標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した(移動相; アセトニトリル:水 = 65:35, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は7.1分で、純度は99%以上であった。
n-プロピル 2-(2-フルオロビフェニルー4-イル)酢酸(150 mg, 0.65 mmol)をジクロロメタン(3 mL)に溶解し、撹拌しながら、塩化オキサリル(67 μL, 99 mg, 0.78 mmol)と、DMF (10 μL)を、室温で加えた。5分間撹拌した後、プロパノール(97 mL, 78 mg, 1.3 mmol)を加えた。更に5分間撹拌した後、水(0.2 mL)で反応を停止し、得られた反応混合物を、シリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=20:1)によって分離したところ、目的のn-プロピル 2-(2-フルオロビフェニルー4-イル)アセテート(18)が透明の油状物として得られた(収率84%)。このものの1H NMRスペクトルを以下に示す。

実施例18のステップ1より合成した2-メチルプロピル 2-(2-フルオロビフェニル-4-イル)アセテート(24)から、実施例2と同様の手順により、[11C]フルルビプロフェン 2-メチルプロピルエステル(23)を合成した。こうして得られた反応混合物を分取用HPLC(移動相; アセトニトリル:水 = 75:25, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図18に示すように、[11C]フルルビプロフェン 2-メチルプロピルエステル(23)が保持時間15.5分で流出した。

フルルビプロフェン メチルエステルを出発原料とし、実施例1の方法と同様の方法により、[11C]メチルフルルビプロフェン(25)を合成した。こうして得られた反応混合物を分取用HPLC (移動相; アセトニトリル:10 mMリン酸ナトリウム緩衝液(pH7.4) = 80:20, カラム; COSMOSIL, 5C18-MS-II, 内径 20 mm×長さ 250 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図19に示すように、[11C]メチルフルルビプロフェン(25)が保持時間14分で流出した。
放射能は反応終了時に3.68 GBqであり、比放射能は42 GBq/μmolであった。また、[11C]メチルフルルビプロフェン(25)の合成、分離、及び静脈注射液調製の合計時間は、約32分であった。[11C]メチルフルルビプロフェン(25)の同定は、メチルフルルビプロフェン(Bioorg. Med. Chem. Lett. 2006, 16, 2219-222)の標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した (移動相; アセトニトリル:0.1%リン酸 = 50:50, カラム; COSMOSIL, 5C18-AR-II, 内径 4.6 mm×長さ 100 mm, 粒径 5 μm, 流速; 1 mL/min, UV検出器; 254 nm)。その結果、保持時間は7.6分で、純度は99%以上であった。

(R)- [11C]フルルビプロフェン メチルエステル(27R)の同定
上記合成反応で得られた反応混合物を分取用HPLC (移動相; メタノール:水 = 90 : 10,カラム; CHIRALCEL OJ-RH, 内径 20 mm×長さ 150 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図21に示すように、(R)- [11C]フルルビプロフェン メチルエステル(27R)が保持時間17分で流出した。(R)- [11C]フルルビプロフェン メチルエステル(27R)の同定は、(R)-フルルビプロフェン メチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
上記合成反応で得られた反応混合物を分取用HPLC (移動相; メタノール:水 = 90 : 10,カラム; CHIRALCEL OJ-RH, 内径 20 mm×長さ 150 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図21に示すように、(R)- [11C]フルルビプロフェン メチルエステル(27R)が保持時間17分で流出した。(R)- [11C]フルルビプロフェン メチルエステル(27R)の同定は、(R)-フルルビプロフェン メチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
(S)-[11C]フルルビプロフェン メチルエステル(27S)の同定
一方、上記合成反応で得られた反応混合物を分取用HPLC (移動相; メタノール:水 = 90 : 10,カラム; CHIRALCEL OJ-RH, 内径 20 mm×長さ 150 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図21に示すように、(S)- [11C]フルルビプロフェン メチルエステル(27S)が保持時間22分で流出した。(S)- [11C]フルルビプロフェン メチルエステル(27S)の同定は、(S)-フルルビプロフェン メチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
一方、上記合成反応で得られた反応混合物を分取用HPLC (移動相; メタノール:水 = 90 : 10,カラム; CHIRALCEL OJ-RH, 内径 20 mm×長さ 150 mm, 粒径 5 μm, 流速; 10 mL/min, UV検出器; 254 nm)で分離したところ、図21に示すように、(S)- [11C]フルルビプロフェン メチルエステル(27S)が保持時間22分で流出した。(S)- [11C]フルルビプロフェン メチルエステル(27S)の同定は、(S)-フルルビプロフェン メチルエステルの標品を混合して、分析用HPLCの結果が単一ピークであることにより決定した。
<脳内炎症モデルラットを用いたPET撮影>
実施例6で合成した[11C]ケトプロフェンメチルエステル(8)について、脳内炎症モデルラットを用いたPET撮影を行った。実験では、ラットの左側線条体にグラム陰性菌細胞壁外膜の構成成分であるリポポリサッカライド(LPS) 0.5μgを微量注入することにより、脳内炎症を誘発させた(Hunter RL, Dragicevic N, Seifert K, Choi DY, Liu M, Kim HC, Cass WA, Sullivan PG, Bing G. Inflammation induces mitochondrial dysfunction and dopaminergic neurodegeneration in the nigrostriatal system. J Neurochem. 2007 100:5:1375-86.)。LPS注入から1日後のラットに、 [11C]ケトプロフェンメチルエステル(8)を尾静脈より投与し、小動物用PET装置(microPET、SIMENS社製)を用いて45分間の撮像を行った。[11C]ケトプロフェンメチルエステル(放射能量72-74 MBq)注射液は、生理食塩水で1.0 CCの容積に希釈して調製した。さらに、非RI標識体による結合阻害効果を検討するために、ケトプロフェンメチルエステル 10mg/kg を加えた注射液(溶媒の最終濃度は5 % DMSO, 5% tween20)を調製した。また、PET撮像終了後、脳切片を作製し、Ex vivoオートラジオグラム(ARG)実験を実施した。これは、ラットを生理食塩水で灌流し、取り出した脳から2 mmの厚さで作製した切片をイメージングプレートBAS SR-2040(富士フィルム社製)の上に1時間露光させ、フルオロイメージアナライザー(FLA7000)を用いて読み取ることで行った。さらに、シクロオキシゲナーゼ2の発現部位を同定するために、特異的抗体を用いて脳切片の免疫組織化学染色を行った。すなわち、前述のオートラジオグラムに使用した脳切片から20μmの凍結切片を作製し、スライドグラスの上に貼り付けたものを、一次抗体として200倍の濃度でPBS-Tに希釈した抗シクロオキシゲナーゼ2(ウサギIgG、ポリクローナル抗体、Cayman社製) で、4℃下にオーバーナイトでインキュベーションする。次に、二次抗体として200倍の濃度でPBS-Tに希釈したビオチン標識抗ウサギIgG抗体(VECTOR社製)と室温で1時間の反応を行い、アビジン-ビオチン標識ペルオキシダーゼ複合体(VECTASTAIN Elite ABC Kit、VECTOR社製)と結合させる。発色方法には、ジアミノベンジンと反応させるDAB法を用いた。

<結 果>
[11C]ケトプロフェンメチルエステル投与後45分間におけるPET加算平均画像、および放射能量を図22aに、また、その際の各脳部位における放射能の時間経過を図22bに示す。図22aは、[11C]ケトプロフェンメチルエステル(8)が、血液脳関門を透過し、脳の左側のLPS注入部位周辺の炎症領域に集積することを示している。この集積は、非RI標識ケトプロフェンメチルエステル10 mg/kgの投与により、有意に低下することから、[11C]ケトプロフェンメチルエステルの炎症領域における集積は特異的であることが示された。また、図23に示すように、ラット脳切片を用いたオートラジオグラム(ARG)実験においても、PET撮像時同様、[11C]ケトプロフェンメチルエステルは、炎症部位に対して特異的に集積することが分かった。さらに、免疫組織化学染色を行った結果、図24に示すように、[11C]ケトプロフェンメチルエステルの集積部位はシクロオキシゲナーゼ2の免疫染色の陽性部位と一致していた。これらの事実から、実施例6の[11C]ケトプロフェンメチルエステル(8)を用いることで、脳におけるシクロオキシゲナーゼ2のイメージングが可能であることが世界で初めて立証された。
[11C]ケトプロフェンメチルエステル投与後45分間におけるPET加算平均画像、および放射能量を図22aに、また、その際の各脳部位における放射能の時間経過を図22bに示す。図22aは、[11C]ケトプロフェンメチルエステル(8)が、血液脳関門を透過し、脳の左側のLPS注入部位周辺の炎症領域に集積することを示している。この集積は、非RI標識ケトプロフェンメチルエステル10 mg/kgの投与により、有意に低下することから、[11C]ケトプロフェンメチルエステルの炎症領域における集積は特異的であることが示された。また、図23に示すように、ラット脳切片を用いたオートラジオグラム(ARG)実験においても、PET撮像時同様、[11C]ケトプロフェンメチルエステルは、炎症部位に対して特異的に集積することが分かった。さらに、免疫組織化学染色を行った結果、図24に示すように、[11C]ケトプロフェンメチルエステルの集積部位はシクロオキシゲナーゼ2の免疫染色の陽性部位と一致していた。これらの事実から、実施例6の[11C]ケトプロフェンメチルエステル(8)を用いることで、脳におけるシクロオキシゲナーゼ2のイメージングが可能であることが世界で初めて立証された。
<アルツハイマー病モデル動物のPET撮影>
実施例6で合成した[11C]ケトプロフェンメチルエステル(21)について、アルツハイマー病モデルマウスを用いたPET撮像を行った。アルツハイマー病モデルマウスは、家族性アルツハイマー病におけるアミロイド前駆体タンパク質(APP)を過剰発現させたトランスジェニックマウス(APP-Tg)を米国タコニック社より購入して用いた。アルツハイマー病の病理変化としてアミロイドβ蛋白の蓄積が確認された24ヶ月齢において、[11C]ケトプロフェンメチルエステル(21)を尾静脈より投与し、小動物用PET装置(microPET、SIMENS社製)を用いて30分間の撮像を行った。[11C]ケトプロフェンメチルエステル(21)(放射能量45-60 MBq)注射液は、生理食塩水で1.0 CCの容積に希釈して調製した。集積程度の指標として、SUV (Standard Uptake Value)= 関心領域における組織放射能(MBq/g)÷〔投与量(MBq)/体重(g)〕を用い、APP Tgマウス脳への集積量について正常マウスであるWild Typeマウス脳との比較を行った。

<結 果>
[11C]ケトプロフェンメチルエステル(21)投与5分後から25分間のPET加算平均画像および、集積量の定量値(SUV)を図25に示す。[11C]ケトプロフェンメチルエステル(21)はWild Typeマウス脳と比較して、APP-Tgマウス脳に高く集積することが示された。また、定量値から、[11C]ケトプロフェンメチルエステルが高い集積を示す部位は、APP-Tgマウス脳において老人斑の主要構成成分であるアミロイドβの蓄積が報告されている部位 (Am J Pathol. 1998;152 (1):307-17.)と一致することが示された。この事実から、[11C]ケトプロフェンメチルエステル(21)を用いることで、アルツハイマー病において神経変性に関与する脳内炎症のイメージングが可能であることが立証された。
[11C]ケトプロフェンメチルエステル(21)投与5分後から25分間のPET加算平均画像および、集積量の定量値(SUV)を図25に示す。[11C]ケトプロフェンメチルエステル(21)はWild Typeマウス脳と比較して、APP-Tgマウス脳に高く集積することが示された。また、定量値から、[11C]ケトプロフェンメチルエステルが高い集積を示す部位は、APP-Tgマウス脳において老人斑の主要構成成分であるアミロイドβの蓄積が報告されている部位 (Am J Pathol. 1998;152 (1):307-17.)と一致することが示された。この事実から、[11C]ケトプロフェンメチルエステル(21)を用いることで、アルツハイマー病において神経変性に関与する脳内炎症のイメージングが可能であることが立証された。
