Abstract
本発明は、細胞内に2種類以上の外来性の機能性RNAを保持し、ウイルスなどの病原体に対する抵抗性を有するトランスジェニック動物である。1種類の外来性の機能性RNAを保持する動物と比較し、複数の外来性の機能性RNAを保持する動物の方が、顕著に病原体の増殖を抑制することができる。
Description
本発明は、細胞内に2種類以上の外来性の機能性RNAを保持し、病原体に対する抵抗性を有することを特徴とするトランスジェニック動物に関する。
家畜は人間の生活に重要な産業動物であるが、家畜で伝播する伝染病の中には、家畜や人間に甚大な被害をもたらすものもある。
哺乳類においては、口蹄疫や、狂犬病、豚コレラなどは感染力が強く、ひとたび発生すると、同居群や接触の恐れのある家畜を全殺し、焼却処分し、周囲を消毒することで封じ込めることしか対策はない。
また家禽においては、鳥類間で伝播する病原体が多く知られている。中でもインフルエンザウイルスなどは、鳥類だけでなくヒトや家畜にも感染し、重篤な感染症を引き起こす病原体である。人類は、このようなインフルエンザウイルスに対して、抗インフルエンザ薬やワクチンを開発して対抗してきた。ところが、進化を続ける未知の新型インフルエンザウイルスに対する予防策は未だ充分ではない。インフルエンザウイルスは、鳥類間、または鳥類と他の動物種間、または鳥類とヒトとの間で感染を繰返すことにより、変異する事が知られており、治療薬やワクチンが効果を発揮できない事がある。
病原体に対して抵抗性を示す動物を得る事は極めて重要であるが、その動物の作製方法として、育種による方法と遺伝子組換えによる方法が考えられる。
育種による方法の例として、遺伝的に病原体に抵抗性を持ったニワトリの系統同士の交配を繰返すことによって病原体に対する抵抗性が強化されたニワトリ種を作製する方法が知られている。具体的には、インフルエンザ抵抗性ニワトリやマレック病抵抗性ニワトリの作製が試みられたが、この方法で抵抗性ニワトリを作製するには、極めて長い年月を要し、また、作製された抵抗性ニワトリは、抵抗性を示すがゆえに病原体のキャリアとなって、他の動物に伝播するという問題も指摘されている。加えて、病原体が変異した場合に、必ずしも充分な抵抗性を発揮させることができない。
遺伝子組換えによる方法の例としては、各種ベクターを用いた遺伝子組換え技術とRNA干渉(RNA interference、RNAi)技術(非特許文献1)を融合して、特定の病原体に対して抵抗性を示す(病原体の増殖を抑制する)動物を作製する方法が知られている(特許文献1および特許文献2)。この方法を応用した例として、トリインフルエンザウイルスの増殖を抑制するshRNA(short hairpin RNA)を導入したニワトリの作製が検討された(特許文献3)が、この場合も、病原体が変異した場合の抵抗性の発揮という問題がある。

病原体が変異したとしても充分な抵抗性を有するトランスジェニック動物を作製することを課題とした。
発明者らは、鋭意検討を重ねた結果、細胞内に2種類以上の外来性の機能性RNAを保持するトランスジェニック動物を作製することにより、上記課題を解決した。具体的には、家禽を例として、複数の外来性の機能性RNAを発現したニワトリを作製し、本発明に至った。
すなわち、本発明は以下のとおりである。
(1)細胞内に2種類以上の外来性の機能性RNAを発現し、病原体に対する抵抗性を有することを特徴とするトランスジェニック動物。
(2)機能性RNAが、shRNA、miRNA、リボザイム、siRNAおよびアプタマーから選ばれる1以上のポリヌクレオチド(RNA)であることを特徴とする(1)に記載のトランスジェニック動物。
(3)機能性RNAが、病原体のゲノムの部分塩基配列の相補鎖と50%以上の配列同一性を有する塩基配列からなるRNAであることを特徴とする、(1)または(2)に記載のトランスジェニック動物。
(4)病原体に対する抵抗性が、病原体の増殖を50%以下に抑制することを特徴とする、(1)から(3)のいずれかに記載のトランスジェニック動物。
(5)病原体が、ウイルスであることを特徴とする、(1)から(4)のいずれかに記載のトランスジェニック動物。
(6)ウイルスが、インフルエンザウイルスであることを特徴とする、(5)に記載のトランスジェニック動物。
(7)トランスジェニック動物が、鳥類であることを特徴とする(1)から(6)のいずれかに記載のトランスジェニック動物。
(8)鳥類が家禽であることを特徴とする、(7)に記載のトランスジェニック動物。
(9)家禽が、ニワトリであることを特徴とする、(8)に記載のトランスジェニック動物。
(1)細胞内に2種類以上の外来性の機能性RNAを発現し、病原体に対する抵抗性を有することを特徴とするトランスジェニック動物。

本発明により、病原体に対して優れた抵抗性を有するトランスジェニック動物を作製することができ、家畜で伝播する伝染病による家畜や人間への甚大な被害を防止することができる。例えば、ニワトリに代表される家禽の場合には、以下の2つの効果が挙げられる。1つ目は、トリインフルエンザに代表される人獣共通感染症の鳥類間または鳥類からヒトへの感染を予防でき、新型インフルエンザに代表される人類にとって未知の恐るべきウイルスのパンデミックを予防できるという点である。2つ目は、家禽に病原体に対する抵抗性を持たせることによって、鳥類間での病原性ウイルスの蔓延を抑止し、養鶏場などでの甚大な経済損失を低減させることが可能となる。
以下、本発明を実施する好ましい形態について述べるが、本発明の本質は記載の内容によって限定されるものではない。
本発明は、以下の工程によって病原体に抵抗性を有するトランスジェニック動物を作製することを特徴とする。
(1)病原体の増殖を抑制する機能性RNAを選択する。
(2)動物細胞で(1)の機能性RNAの複数発現を可能とするベクターを構築する。
(3)(2)のベクターを用いてトランスジェニック動物を作製する。

(3)(2)のベクターを用いてトランスジェニック動物を作製する。
それぞれの工程について以下に詳述する。
(1)病原体(例えば病原性ウイルス)の増殖を抑制するための機能性RNAを選択する。病原性ウイルスにはRNAウイルスとDNAウイルスが含まれる。また、病原性ウイルスとしては、家畜等で伝播する病原性ウイルスが挙げられる。例えば、口蹄疫、狂犬病、豚コレラなどが例示できる。家禽または野鳥で伝播する病原性ウイルスまたはその変異株、特にニワトリで伝播する病原性ウイルスまたはその変異株を対象とすることも好ましい。
そのような病原性ウイルスとしては、インフルエンザウイルス、中でもトリインフルエンザウイルス、特にH5N1型トリインフルエンザウイルスまたはその変異株が例示できる。これらインフルエンザウイルスは、オルトミクソウイルス科(Orthomyxoviridae)に分類されるRNAウイルスである。インフルエンザウイルスのほかにも、ニューカッスル病ウイルス、鶏伝染性気管支炎ウイルス、鶏白血病ウイルス、鶏脳脊髄炎ウイルス、鶏腎炎ウイルス、鶏伝染性咽頭気管炎ウイルス、細網内皮症ウイルス、マレック病ウイルス、伝染性ファブリキウス嚢病ウイルス、トリレオウイルス、トリアデノウイルス、EDS-76ウイルス、鶏貧血ウイルス、七面鳥鼻気管炎ウイルス、トリパラミクソウイルス、鶏痘ウイルスといった家禽で伝播するウイルスまたはその変異株のほか、水鳥、野鳥を含むあらゆる鳥類で伝播可能なウイルスまたはその変異株が挙げられる。
なかでも、家禽で広範に拡大し、被害が甚大である点からトリインフルエンザウイルスやマレック病ウイルス、ニューカッスル病ウイルスなどがより重視され、ヒトへの感染の恐れからは、トリインフルエンザが特に重視される。
本発明における動物とは、人間によって飼養されうる哺乳類および鳥類が含まれる。例えば、牛、馬、羊、山羊、豚、犬、猫等の哺乳類、ニワトリ、ウズラ、七面鳥、ガチョウ、カモ、アヒル、ダチョウ等の鳥類(家禽)を挙げることができる。
本発明における病原体の増殖を抑制するための機能性RNAとは、標的となる病原体のゲノムの部分塩基配列と相補的な塩基配列を有するRNAを含む機能性RNAを指す。ゲノムの部分塩基配列は、5塩基以上、好ましくは10塩基以上、さらに好ましくは15塩基以上、特に好ましくは20塩基以上である。これらの部分塩基配列と相補的な塩基配列からなるRNAを含む機能性RNAを本発明に好適に使用できる。また、この機能性RNAは標的となる病原体のゲノムの部分塩基配列と相補的な塩基配列と50%以上、好ましくは60%以上、より好ましくは70%以上、さらに好ましくは80%以上、とりわけ90%以上、最も好ましくは100%の配列同一性を示す塩基配列からなるRNAを含む機能性RNAを本発明に好適に使用できる。
標的となる病原体のゲノムの部分塩基配列の領域としては、変異頻度の低い領域が好ましい。ここでいう変異頻度の低い領域とは、ある病原体の複数の異なる株間において、そのDNAゲノムまたはRNAゲノムの塩基配列が80%以上、好ましくは85%以上、さらに好ましくは90%以上、最も好ましくは100%同一である領域のことをいい、病原体の複製や構造維持に機能している領域などが標的となり得る。例えばA型インフルエンザウイルスでは8つの遺伝子領域が存在するが、そのうちPA、PB1、PB2、NPと呼ばれるウイルスのRNAゲノムを複製する領域や、Mと呼ばれるウイルスの構造維持に機能する遺伝子領域は好適な標的となり得る。
動物細胞において、発現させた機能性RNAは標的となる病原体の増殖を抑制する機能を有する限り、その長さや構造において限定されることはない。ここでいう動物細胞とは、鳥類、哺乳類等の動物由来の細胞のことをいう。この細胞の集合体を動物という。
本発明における、病原体の増殖を抑制する機能とは、機能性RNAが導入された細胞もしくは動物が、非操作対照の細胞もしくは動物との比較において、病原体の増殖速度が50%以下、好ましくは40%以下、さらに好ましくは30%以下に抑制されることを言う。ウイルスの増殖速度の測定方法は、血清凝集試験、赤血球凝集抑制試験、中和反応、寒天ゲル内沈降反応、抗原抗体反応、蛍光抗体法、定量的PCR法等のあらゆる公知の手法で測定できる。
本発明における機能性RNAは、ループやヘアピン等の2次元的あるいは3次元的な構造をとりうる機能性RNAを好適に用いることができる。このような機能性RNAとしては、たとえばshRNA、siRNA、miRNA、リボザイムまたはアプタマーを例示することができる。また、shRNAは細胞内においてプロセッシングを受け、短い二本鎖のRNAダイマー(siRNA)として存在することもあり、タンパク質や脂質などの生体分子と会合し、複合体を形成して存在しうる。shRNAやsiRNAなどは、RNA干渉効果を誘導する機能を持つことが多く、顕著に標的病原体ゲノムの分解、抑制効果があり、本発明で好ましく用いることができる。
RNA干渉効果とは、標的とする病原体のゲノムの塩基配列とある程度相補的な配列を有する1本鎖または2本鎖のRNA配列が、細胞内でRNA-induced silencing complex(RISC)と呼ばれるタンパク質-RNA複合体を形成し、標的とする病原体のゲノムを切断する現象である。
特定の病原体に対して増殖抑制効果の高いRNA配列は、標的とする病原体または、その病原体ゲノムを用いて、培養細胞を用いるなどしたin vitro試験または動物感染実験などのin vivo試験、あるいは、機能性RNAを発現させた動物から分離した細胞を用いたex vivo試験など、公知のあらゆる試験法または、文献情報に基づき、選択することができる。
動物で発現させる機能性RNAは、動物細胞内で2種類以上を発現させることが好ましい。変異の速い病原体に対しては複数種の機能性RNAを発現させることで、病原体に対する抵抗性を持続させることが可能になる。
(2)上記に記載の各種病原体の増殖を抑制するための、2種以上の機能性RNA遺伝子を動物細胞に導入可能なベクターを作製する。ベクターとしては、対象とする動物細胞内に導入できれば、プラスミド型DNAでも良いし、2本鎖DNA断片でもよい。本発明におけるプラスミド型DNAは、環状構造をした2本鎖DNAを指す。上記ベクターは、リン酸カルシウム法、リポフェクション法、電気導入法など公知の遺伝子導入法で動物細胞内に導入される。この際、導入されたベクターは、動物細胞の染色体に組込まれることが好ましい。本発明における染色体とは、細胞分裂時に複製され、娘細胞に遺伝するゲノムDNAのことをいう。
染色体に効率的に機能性RNA遺伝子を組込むベクターとしては、公知のものが利用できる。例えば、ウイルスベクター(レトロウイルスベクターなど)を使用することができる。レトロウイルスベクターとしては特に限定されないが、モロニーマウス白血病ウイルス、モロニーマウス肉腫ウイルス、トリ白血病ウイルス(ALV)、マウス肝細胞ウイルス(MSCV)、マウス胚性肝細胞ウイルス(MESV)等を由来とするレトロウイルスベクターの他、ヒト免疫不全ウイルス(HIV)等を由来とするレンチウイルスベクターが挙げられる。
病原体の増殖を抑制する機能性RNAを動物細胞で発現させるために、機能性RNAを発現カセットに組込んだベクターを構築する。ベクターは、特に限定されないが、機能性RNA配列のほかにプロモーター、エンハンサー、調節因子等、細胞または細胞の集合体である動物で該機能性RNAを発現させるために、適切にデザインされたものを用いることが好ましい。
プロモーターとは遺伝子の転写開始部位を決定し、またその頻度を直接的に調節する機能を有するDNA配列のことである。プロモーターとしてはRNAの発現に有効に機能するプロモーターであれば限定されないが、一般的にRNAの発現によく用いられているU6プロモーターやH1プロモーターのようなpolIII系のプロモーターは、短いRNAの発現に好適である。その他、polII系のプロモーターとして、EF1αプロモーター、チミジンキナーゼプロモーター、シミアンウイルス40(SV40)プロモーター、ミューリン・フォスフォグリセロキナーゼ(PGK)プロモーター、サイトメガロウイルス(CMV)プロモーター、ラウスザルコーマウイルス(RSV)プロモーター等のウイルスプロモーター、ニワトリベータアクチンプロモーター等の動物由来のプロモーター、テトラサイクリン誘導型プロモーターのような誘導型プロモーターも用いることができる。また、組織特異的なプロモーターを用いて、特定の組織・細胞で発現させることも可能である。特に粘膜や消化器官で機能するようなプロモーターが好ましい。
エンハンサーとは、プロモーターからの転写を促進する配列である。プロモーターとの組み合わせについては特に限定されない。エンハンサーとしては、特に限定されないが、SV40、CMV、チミジンキナーゼエンハンサー、ステロイド応答エレメントやリゾチームエンハンサー等が挙げられる。
調節因子は、転写調節や、転写後のRNAの安定化に寄与するDNA配列である。調節因子としては特に限定されないが、ウッドチャック肝炎ウイルス由来調節エレメント(woodchuck post-transcriptional regulatory element:WPRE、米国特許6136597号明細書)等が挙げられる。
このようにして作製されるベクターは、プロモーターおよび/またはエンハンサーを含む発現ユニットを2以上含むことにより、動物細胞中で複数種の機能性RNAを発現させることが可能であるが、1つの発現ユニットにより転写されたRNAが細胞内でプロセッシングされることで、結果的に2種類以上の機能性RNAを発現させても同様の効果が得られることは知られている。もしくは、単独では1種類の機能性RNAを動物細胞で発現可能なベクターを2種類以上動物細胞に導入することにより、該細胞内で複数の機能性RNAを同時発現させることも可能である。発現ユニットを複数含むベクターの場合、発現ユニットは同一方向にタンデムに連結しても良いし、双方向に配置しても良い。
ベクターとしては、特に制限されないが、ウイルスベクターなどを用いることができる。ウイルスベクターを用いる場合、その力価(タイター)は高力価であるほど好ましく、例えば、NIH3T3細胞またはHela細胞を用いて測定した場合、1×106cfu/ml以上であり、好ましくは1×107cfu/ml以上であり、より好ましくは1×108cfu/ml以上であり、さらに好ましくは1×109cfu/ml以上であり、とりわけ1×1010cfu/ml以上である。
(3)上記ベクターを用いて、病原体に抵抗性を示す細胞または動物を作製する手法について詳述する。病原体に抵抗性を示す細胞を作製する場合、上記ベクターを公知の手法で細胞内に導入すればよい。病原体に抵抗性を示す動物を作製する場合は、例えば、哺乳類においては、受精卵または初期胚にベクターを導入後、ホスト動物の子宮へ移植し、個体まで発生させる公知手法を用いることができる。また鳥類においても、有精卵を用いて、初期胚にベクターを導入し、孵化させる公知手法を用いることができる。これにより、細胞内に2種類以上の機能性RNAを発現する細胞又は動物を得ることが可能である。
このように作製した細胞内に2種類以上の機能性RNA(shRNAなど)を発現する細胞又は動物への遺伝子導入コピー数(平均)は、普通0.01以上であり、好ましくは0.1以上、より好ましくは1以上、さらに好ましくは2以上である。
このように作製した細胞内に2種類以上の機能性RNA(shRNAなど)を発現する細胞または動物の評価法としては、公知のあらゆる手法を用いることが可能である。In vitro試験として発現させた機能性RNA配列を検出する方法とIn vivoでのウイルス耐性試験などにより評価ができる。
In vitro試験としては、作製した細胞内に2種類以上の機能性RNAを発現した動物細胞からRNAを抽出し、ノーザンブロット法や定量的もしく半定量的リアルタイムPCRで目的のRNAを検出することが可能である。
またin vivo試験としては、作製した細胞内に2種類以上のshRNAを発現する動物に、病原性ウイルスを感染させ、生存率や抗体産生の有無を調べることにより、病原性ウイルスへの抵抗性を評価することができる。
あるいは、病原性ウイルスを用いることなく、機能性RNAのウイルス増殖抑制効果を実験的に確認することも可能であり、その方法としては、例えば、レポーター遺伝子の発現抑制の程度を測定すればよい。レポーター遺伝子は、機能性RNAによって発現が抑制されるようデザインしておく。機能性RNAが発現した細胞内でレポーター遺伝子の発現抑制をリアルタイムPCR等の公知の手法で定量する。あるいは、レポーター遺伝子が発現した細胞を作製し、この細胞に機能性RNAを発現する遺伝子を導入した後に、レポーター遺伝子の発現抑制の程度をリアルタイムPCR等の公知の手法で測定してもよい。
言うまでもなく、作製した細胞内に2種類以上の機能性RNAを発現する動物は、その動物同士で、あるいは、野生型の動物と交配することによって、病原体に対して優れた抵抗性を示す子孫を得ることができる。
以下、実施例により本発明を詳述するが、本発明はこれら実施例により限定されるものではない。遺伝子操作手法等について特に記述のないものについては代表的な方法に従った(J.Sambrook,E.F.Fritsch,t.Maniatis;Molecular Cloning,A Laboratory Manual,2nd Ed,Cold Spring Harbor Laboratory)。細胞培養については特に記述のないものについては代表的な手法に従った(小山秀機編集「細胞培養ラボマニュアル」Springer-Verlag Tokyo 1999)。商品名もしくはメーカーを記載しているものに関しては、特に記載のない限り添付の説明書の指示に従った。ここで文言として用いる“ベクター”または“ベクターコンストラクト”とは、環状2本鎖DNA(いわゆるプラスミド型ベクター)のことを言う。“レトロウイルスベクター”としては、遺伝子導入のための複製能のないウイルスを用いた。
(実施例1)shRNA発現ベクターコンストラクトの構築
H5N1型トリインフルエンザウイルスの抑制効果のあるshRNAの発現ベクターを構築した。

1-1:shRNAの合成
A型インフルエンザウイルスのNP、M2を標的としたshRNAをコードするDNA配列を合成した。配列番号1、配列番号2をそれぞれ合成(SIGMA Genosys)した。また、コントロールとしてGFPを標的としたshRNAをコードするDNA配列(配列番号3)を合成(SIGMA Genosys)した。
A型インフルエンザウイルスのNP、M2を標的としたshRNAをコードするDNA配列を合成した。配列番号1、配列番号2をそれぞれ合成(SIGMA Genosys)した。また、コントロールとしてGFPを標的としたshRNAをコードするDNA配列(配列番号3)を合成(SIGMA Genosys)した。
1-2:hU6プロモーターの合成
polIII系プロモーターとしてヒト由来U6プロモーター配列を合成した。配列番号4に記載のDNA配列を合成(SIGMA Genosys)した。

配列番号4を制限酵素EcoRIと制限酵素BamHIで消化し、3%アガロースゲルで電気泳動を行い、エチジウムブロマイドで染色後、300bp付近のバンドをカッターナイフで切り出した後、QIAEXII(QIAGEN)を用いてアガロースゲルからDNAを精製し、10μlのTEに溶出し、インサートとした。特許文献(特開2007-89578号公報実施例6)に公開されているpMSCVneobactfEPOwpreベクターをベースとするレトロウイルスベクターコンストラクト1μgを制限酵素EcoRIと制限酵素BamHIで消化し、1%アガロースゲル電気泳動を行い、エチジウムブロマイドで染色後、6000bp付近のバンドをカッターナイフで切り出した後、QIAEXII(QIAGEN)を用いてアガロースゲルからDNAを精製し、10μlのTEに溶出し、ベクターとした。インサートおよびベクターを1μlづつ混合し、ミリQ水2μl、SolutionI(DNA Ligation Kit Ver2.1:TAKARA)4μlを加え、よく混ぜた後、16℃で30分 Ligation反応を行なった。反応液の1μlを大腸菌DH5α(TAKARA)に形質転換(混合後、42℃、45秒間のヒートショック)し、アンピシリンを含むLB寒天培地でコロニー形成させた。翌日、シングルコロニーをピックアップし、LB液体培地で37℃16時間培養し、培養上清を回収後、プラスミドを抽出した(QIAprep Spin Miniprepkit:QIAGEN)。このプラスミドをpMSCVneoU6wpreとした。配列番号1、配列番号2、配列番号3のDNA断片を上記の手法と同様に制限酵素で消化、DNAを精製して、インサートとし、それぞれpMSCVneoU6wpreのhU6プロモーターの下流に組み込み、このベクターをそれぞれpMSCVneoU6NPwpre、pMSCVneoU6M2wpre、pMSCVneoU6GFPwpreとした。

pMSCVneoU6NPwpreを鋳型として、hU6とNPの領域をPCRで増幅した。このときに得られた300bp程度のDNA断片をU6NPとした。
pMSCVneoU6M2wpreをベクターとし、U6NPをインサートとしてライゲーションすることで、2つのshRNAの発現ユニットを組み込んだレトロウイルスベクターコンストラクトであるpMSCVneoU6NPU6M2wpreを構築した。
(実施例2)レトロウイルスベクターの調製
2-1:一過性レトロウイルスの調製
実施例1で作製したベクターコンストラクト(pMSCVneoU6NPwpre)を大腸菌DH5α(TAKARA)に形質転換し、シングルコロニーを100mlのLB培地で37℃16時間振とう培養した後、遠心分離により菌体を回収し、エンドトキシンフリーで精製した(EndoFree-plasmid Maxi Kit:QIAGEN)。
2-1:一過性レトロウイルスの調製
実施例1で作製したベクターコンストラクト(pMSCVneoU6NPwpre)を大腸菌DH5α(TAKARA)に形質転換し、シングルコロニーを100mlのLB培地で37℃16時間振とう培養した後、遠心分離により菌体を回収し、エンドトキシンフリーで精製した(EndoFree-plasmid Maxi Kit:QIAGEN)。
GP293細胞(Clontech)を100mmコラーゲンコートした培養ディッシュ(IWAKI)に5×106細胞まき、D-MEM High-Glucose(GIBCO)(10%FBS含)で24時間培養した後、上記のベクターコンストラクトとpVSVGプラスミド(Clontech)それぞれ24μgをLipofectAMINE2000(Invitrogen)でトランスフェクトした。トランスフェクトの条件はLipofectAMINE2000のマニュアルに従った。6時間後に培養ディッシュから培養上清を吸引し、あらたに9mlのD-MEM(10%FBS含)と200μlの1M HEPES Buffer Solution(GIBCO)を加えてさらに24時間培養を継続した。
培養上清を0.45μmのセルロースアセテートフィルター(アドバンテック社製)に通し、ろ液を遠心管に集めた。超遠心機CS100GXL(日立工機社製)を用いて超遠心(50000G、1.5時間、4℃)によって一過性発現させたレトロウイルスを濃縮した。濃縮したレトロウイルスを20μlのTEに懸濁し、一過性レトロウイルス調製液とした。
2-2:パッケージング細胞株の樹立と選抜
前日に96ウェルコラーゲンコートプレート(IWAKI)に1×104細胞/ウェルとなるようGP293細胞を播種しておき、ポリブレンを終濃度8μg/mlで加えた後、2-1で作製した一過性レトロウイルス調製液20μlを加えて感染させた。感染後の上記GP293細胞を96ウェルコラーゲンコートプレート(IWAKI)上で0.5細胞/ウェルとなるようにD-MEM培地で限界希釈した。終濃度1mg/mgとなるようにG418(SIGMA)を加えたD-MEM High-Glucose(GIBCO)(10%FBS含)で薬剤選択を行った。同培地を3日に一度交換し、プレート内でコロニー形成の観察を行い、細胞増殖速度が速いクローンを選抜してパッケージング細胞株(pMSCVneoU6NPwpre/GP293)として選抜した。

実施例1で作製したpMSCVneoU6NPU6M2wpreのベクターコンストラクトに関しても上記と同様にパッケージング細胞株を選抜し、pMSCVneoU6NPU6M2wpre/GP293とした。
2-3:高力価レトロウイルスベクターの調製
2-2で作製したパッケージング細胞株(pMSCVneoU6NPwpre/GP293)を100mmコラーゲンコートディッシュ(IWAKI)に5×106細胞播き、一晩培養した(10%FBSを含むD-MEM培地)。LipofectAMINE2000を用いてpVSVGプラスミド24μgをトランスフェクトし、6時間後に培地を交換(10%FBSを含むD-MEM培地)し、200μlの1M HEPES Buffer Solution(GIBCO)を加えてさらに24時間培養した。
2-2で作製したパッケージング細胞株(pMSCVneoU6NPwpre/GP293)を100mmコラーゲンコートディッシュ(IWAKI)に5×106細胞播き、一晩培養した(10%FBSを含むD-MEM培地)。LipofectAMINE2000を用いてpVSVGプラスミド24μgをトランスフェクトし、6時間後に培地を交換(10%FBSを含むD-MEM培地)し、200μlの1M HEPES Buffer Solution(GIBCO)を加えてさらに24時間培養した。

2-4:レトロウイルスベクターの力価測定
NIH3T3細胞(ATCC)を6ウェルプレート(IWAKI)に1.5×104細胞/ウェルとなるように播き、24時間後に培地(10%FBS(GIBCO)を含むD-MEM、終濃度8μg/mlとなるようにPolybrene(Sigma)を添加)を交換した。各ウェルに2-3で調製したレトロウイルスを段階希釈して加え(103~108)24時間後に培地(10%FBSを含むD-MEMにG418を終濃度1mg/mlとなるように添加)を交換した。以降2日ごとにG418を含む培地で選択培養し、コロニーの形成を確認後、カウントして力価を算出した。pMSCVneoU6NPwpre/GP293、pMSCVneoU6NPU6M2wpre/GP293のどちらのパッケージング細胞株からも、108~109cfu/mlの力価をもつレトロウイルスベクターを調製した。

2-5:トランスジェニックニワトリの作製
特許文献(特開2007-89578、実施例7)の内容と同様の手法にて、ニワトリ受精卵へのレトロウイルスのマイクロインジェクションを行った。

ニワトリ受精卵(城山種鶏場:加古川)を38℃、湿度50%以上の条件下で55時間インキュベートした。その際90度の転卵を1時間おきに行った(昭和フランキ社製孵卵機)。
ダイアモンドカッターを付けたミニルーター(プロクソン社製)を用いて鈍端部を開口し、同様に鈍端部を直径4.5cm切り取り、中身を捨てたニワトリ二黄卵(城山種鶏場:加古川)に移し替えた。胚体が上にくるよう調整し、実体顕微鏡システムSZX12(オリンパス社製)下でフェムトチップII(エッペンドルフ社製)にウイルス液を充填し、マイクロインジェクター(フェムトジェット:エッペンドルフ社製)を用いて2-3で調製したレトロウイルスを2μlずつマイクロインジェクションした。
インジェクション後、卵白を糊としてサランラップ(旭化成)で開口部を覆い、38℃、一時間に30度転卵の条件で再びインキュベートし、孵化させた(表1)。その際に導入したウイルスタイター(108cfu/ml以上)と、孵化率を図1に示した。一般に、複数の機能性RNA(shRNAなど)を発現した細胞を得ることは困難と言われているが、図1から分かるように、驚くべきことに、2種類のshRNAを発現した個体では、高タイターベクターを用いた場合でも低タイターベクターを用いた場合と同等の孵化率であり、また、高タイターベクターを用いた場合において、1種類のshRNAを発現した個体と比較して、明らかに高い孵化率を示した。
2-6:トランスジェニックニワトリの導入遺伝子の検出と導入遺伝子コピー数測定
2-4で作製したトランスジェニックニワトリより全血を翼下採血により採取した。抗凝固剤としてクエン酸を使用した。小型遠心機にてフラッシュし、上清を20μl分取し、Mag-Extractor Genome(TOYOBO)のキットを用いてゲノムDNAを抽出した。J Virol.2005 Sep;79(17):10864-74.に記載のトランスジェニックニワトリ(1コピーの導入遺伝子を全身に持つ完全トランスジェニックニワトリ)のゲノムをスタンダードとして導入コピー数を算出した。導入コピー数の算出は、リアルタイムPCR装置(LightCycler:Roche)およびキット(LightCycler First Start DNA Masterhybprobe:Roche)、増幅プライマー(配列番号5および配列番号6:SIGMA Genosys)、蛍光標識プローブ(配列番号7および配列番号8:日本遺伝子研究所)を用いてキットのプロトコルに従い測定した。尚、配列番号7は3´側にFITCで標識され、配列番号8は5´側にLCRed640で標識されている。リアルタイムPCRの反応条件は以下で行なった。
熱変性反応 95℃、10分
増幅検出反応 95℃、10秒 → 58℃、15秒 → 72℃、10秒(45サイクル)
冷却反応 40℃。

(実施例3)トランスジェニックニワトリの発現解析
実施例2で作製したトランスジェニックニワトリ(2種類発現:NP・M2)および陰性コントロールとして野生型ニワトリより、それぞれ翼下採血により全血を採取した。抗凝固剤としてクエン酸を使用し、Trizol Reagent(Invitrogen)を用いてTotal RNAを抽出した。これらのTotal RNA 各0.9ngをサンプルとして逆転写反応後、Custom TaqMan Small RNA Assays(applied biosystems)とApplied Biosystems 7300 リアルタイムPCRシステムを用いてsiRNAの発現解析を実施したところ、2種類のsiRNAを検出した。尚、スタンダードとして、濃度既知の配列番号9、配列番号10のsiRNAを合成(SIGMA Genosys)し、これを標準として検量線を作成し、陰性コントロールを基準として、トランスジェニックニワトリで発現しているshRNAの量を数値化(表3)した。

(実施例4)抗インフルエンザウイルス効果の検証
ニワトリ胚線維芽細胞(CEF)を用いて、shRNAの発現による、ターゲット遺伝子の発現抑制効果の検証を行なった。

4-1:レポーター発現ベクターコンストラクトの作製
NPおよびM2のshRNAが認識する合成配列(配列番号11)を合成した。配列番号11をレポーター遺伝子下流につなぎ、レポーター発現ベクターコンストラクトを構築した(図2)。作製したベクターコンストラクトを大腸菌DH5α(TAKARA)に形質転換し、シングルコロニーを100mlのLB培地で37℃16時間振とう培養した後、遠心分離により菌体を回収し、エンドトキシンフリーで精製した(EndoFree-plasmid Maxi Kit:QIAGEN)。
NPおよびM2のshRNAが認識する合成配列(配列番号11)を合成した。配列番号11をレポーター遺伝子下流につなぎ、レポーター発現ベクターコンストラクトを構築した(図2)。作製したベクターコンストラクトを大腸菌DH5α(TAKARA)に形質転換し、シングルコロニーを100mlのLB培地で37℃16時間振とう培養した後、遠心分離により菌体を回収し、エンドトキシンフリーで精製した(EndoFree-plasmid Maxi Kit:QIAGEN)。
4-2:CEFの調製
ニワトリ有精卵を実施例2記載の方法で7日間孵卵した。この孵卵した有精卵から胚を取り出し頭、内臓、四肢を取り除いた後、細断してトリプシン(GIBCO)を加え、37℃20分間インキュベートした。そこにD-MEM High-Glucose(GIBCO)(10%FBS含)を加え遠心分離を行い、上清を除いた後D-MEMを加え3分間放置した。その後上清をシャーレにまきCEFとした。
ニワトリ有精卵を実施例2記載の方法で7日間孵卵した。この孵卵した有精卵から胚を取り出し頭、内臓、四肢を取り除いた後、細断してトリプシン(GIBCO)を加え、37℃20分間インキュベートした。そこにD-MEM High-Glucose(GIBCO)(10%FBS含)を加え遠心分離を行い、上清を除いた後D-MEMを加え3分間放置した。その後上清をシャーレにまきCEFとした。
4-3:shRNA及びレポーター遺伝子発現CEFの調製
調製したCEFを1×105細胞になるようにプレートに播種し、24hr培養した。培養したCEFに実施例2の方法で調製したpMSCVneoU6NPU6M2wpreレトロウイルスベクター20μlを感染させ、24hr培養後、最終濃度1mg/mlとなるようにG418(SIGMA)を加えたD-MEMに培地交換した。その後、G418を含む培地で一週間培養した。このようにして得られた細胞を、NM/CEFとした。同様に、pMSCVneoU6NPwpre、pMSCVneoU6M2wpre、pMSCVneoU6GFPwpreの3種の一過性レトロウイルスベクターを用いて、NP/CEF、M2/CEF、GFP/CEFの3種のshRNAを発現するCEF細胞を調製した。
調製したCEFを1×105細胞になるようにプレートに播種し、24hr培養した。培養したCEFに実施例2の方法で調製したpMSCVneoU6NPU6M2wpreレトロウイルスベクター20μlを感染させ、24hr培養後、最終濃度1mg/mlとなるようにG418(SIGMA)を加えたD-MEMに培地交換した。その後、G418を含む培地で一週間培養した。このようにして得られた細胞を、NM/CEFとした。同様に、pMSCVneoU6NPwpre、pMSCVneoU6M2wpre、pMSCVneoU6GFPwpreの3種の一過性レトロウイルスベクターを用いて、NP/CEF、M2/CEF、GFP/CEFの3種のshRNAを発現するCEF細胞を調製した。
上記のようにして得られた4種のCEFを1×105細胞になるように6wellプレートに播種し、24hr培養した。これにLipofectAMINE2000を用いて4-1に記載のレポーター発現ベクターコンストラクト3μgをトランスフェクトし、6時間後に培地を交換(10%FBSを含むD-MEM培地)し、200μlの1M HEPES Buffer Solution(GIBCO)を加えてさらに24時間培養した。
4-4:遺伝子抑制効果の検証
4-3でレポーター発現ベクターコンストラクトを導入した4種のCEFよりTotalRNAを抽出した。レポーター遺伝子の発現を、特異的プライマーを用いてリアルタイムPCR法により確認した(LightCycler RNA Master SYBRGreenI:Roche)。リアルタイムPCR法の条件は以下で行なった。
逆転写反応 61℃、20分
熱変性反応 95℃、30秒
増幅反応 95℃、5秒 → 55℃、10秒 → 72℃、25秒(45サイクル)
冷却 40℃。
4-3でレポーター発現ベクターコンストラクトを導入した4種のCEFよりTotalRNAを抽出した。レポーター遺伝子の発現を、特異的プライマーを用いてリアルタイムPCR法により確認した(LightCycler RNA Master SYBRGreenI:Roche)。リアルタイムPCR法の条件は以下で行なった。
逆転写反応 61℃、20分
熱変性反応 95℃、30秒
増幅反応 95℃、5秒 → 55℃、10秒 → 72℃、25秒(45サイクル)
冷却 40℃。
結果を図3に示す。図3から分かるように、1種類の機能性RNA(shRNA)を発現させた場合と比較して、2種類の機能性RNA(shRNA)を発現させた場合には、顕著にレポーター遺伝子の発現を抑制した。
以上、上記実施例はニワトリを例に説明したが、本発明のコンセプトを用いれば、細胞内に2種類以上の機能性RNAを保持し、病原体に対する充分な抵抗性を有する他の鳥類、さらには哺乳類も作製しうる。

- 細胞内に2種類以上の外来性の機能性RNAを発現し、病原体に対する抵抗性を有することを特徴とするトランスジェニック動物。
- 機能性RNAが、shRNA、miRNA、リボザイム、siRNAおよびアプタマーから選ばれる1以上のポリヌクレオチド(RNA)であることを特徴とする請求項1に記載のトランスジェニック動物。
- 機能性RNAが、病原体のゲノムの部分塩基配列の相補鎖と50%以上の配列同一性を有する塩基配列からなるRNAであることを特徴とする、請求項1または2に記載のトランスジェニック動物。
- 病原体に対する抵抗性が、病原体の増殖を50%以下に抑制することを特徴とする、請求項1から3のいずれかに記載のトランスジェニック動物。
- 病原体が、ウイルスであることを特徴とする、請求項1から4のいずれかに記載のトランスジェニック動物。
- ウイルスが、インフルエンザウイルスであることを特徴とする、請求項5に記載のトランスジェニック動物。
- トランスジェニック動物が、鳥類であることを特徴とする請求項1から6のいずれかに記載のトランスジェニック動物。
