Abstract

The invention relates to a composition containing in a soluble form and in an aqueous phase: - proteins: 0.40% to 0.45%; - calcium: 0.50% to 0.51%; -the following mineral salts: sodium, potassium, magnesium, phosphorus, sulphur; - the following trace elements: selenium, manganese, iron, zinc, copper; - acetic acid: 4.70%. The invention also relates to a method for obtaining the composition that comprises placing raw poultry eggs with their shells in a tank containing diluted acetic acid and sodium chloride for one month so that the composition contains the solubilised forms of the compounds naturally occurring in the shell of a poultry egg. The composition of the invention can be used for preparing a drug or a cosmetic product for treating skin or scalp alopecia.

Description

COMPOSITION FOR THE TREATMENT OF SCALP

AND SKIN

The present invention belongs to the field of formulations intended for topical application for therapeutic or non-therapeutic purposes, prepared by extraction of useful compounds from naturally occurring materials.

It relates to a composition based on proteins, calcium, minerals and oligomers in solution in dilute acetic acid, and a process for preparing from ingredients of exclusively natural origin. A product comprising said composition, intended for the therapeutic or cosmetic treatment of the skin and the scalp, is also claimed.

Protein, calcium, mineral salts and trace elements are naturally present in our body and are essential for its proper functioning. Calcium is a key player. It is found involved in multiple metabolic actions involved in particular in the construction of the structure of the skin.

Currently many theories attempt to account for all the complex phenomena involved in the processes that lead to aging and deterioration of the functioning of the cells of vital organs, pushing them under certain conditions unfavorable to cell suicide (apoptosis).

According to scientists, nutritional deficiencies or hyperproduction of free radicals would be responsible for a large number of cell damage. We talk about replicative and metabolic senescence. Whether brought by the environment (pollution, cigarette smoke, radiation ...) or generated by the cells during their metabolic reactions, free radicals can disrupt DNA replication during cell divisions and reduce their ability to regenerate. They can also destroy cell membranes, deform cellular proteins responsible for calcium transport which is unable to play its role of intercellular messenger. The cells do not communicate anymore, lose their cohesion, disorganize seriously injuring the tissues. At the level of the skin, all these upheavals lead to the acceleration of cutaneous aging and under certain conditions to the deterioration of the tissues and to the appearance of diseases, dermatoses, eczema, psoriasis, pruritus, alopecia.

It is now established that a correlation exists between cellular aging and alopecia. The reproductive cells of the hair follicles are normally programmed to regenerate and give birth to new hair in an uninterrupted way thanks to cycles of growth set between 25 to 30 cycles, which have a duration of 2 to 5 years, are sufficient in number to renew the hair for a lifetime. With age, the hormonal receptors of these target organs become particularly sensitive to the harmful action of dihydrotestosterone resulting from testosterone under the effect of 5-α-reductase. Slowing down the rate of cell divisions and the deterioration of the functioning of the hair follicles results in a decrease in growth cycles that quickly run out, from a few years to a few months. The hair clears and, if we do not stop the fall, we see the final loss of hair.

In addition, the study of the composition of the eggshell shows that it contains on average 3.3% of proteins constituting its frame. These proteins play a major role in the processes of calcification of the shell by crystallizing about 95.1% of minerals, mainly calcium carbonate crystals in the form of calcite. The proteins and calcium absorbed within the organic matrix are therefore very closely involved in the construction of the crystalline microstructure of the shell, which contributes to its strength.

Unexpectedly, it has been demonstrated that the combination of the soluble compounds of the eggshell with mineral salts and trace elements brought by natural ingredients in acidic medium, allowed to successfully treat certain conditions of the leather. hairy and skin. Without a systematic study having been carried out, it can be formulated that the composition according to the present invention makes it possible to reproduce, at the level of the structures of the skin, the mechanisms of natural solidity resulting from the affinity of proteins and calcium in the constitution of the shell. In addition, by its formula, the composition according to the invention brings to the heart damaged or broken cells, all the nutrients necessary for the restoration of functioning. cellular. By restoring the calcium metabolism, the claimed composition reinforces the cohesion of the cutaneous tissues and helps to fight against skin aging.

More specifically, the subject of the present invention is a composition intended for the preparation of a product for therapeutic or cosmetic use, containing, in soluble form in aqueous phase:

proteins: 0.40% to 0.45%;

calcium: 0.50% to 0.51%;

- the following minerals: sodium, potassium, magnesium, phosphorus, sulfur; - the following trace elements: selenium, manganese, iron, zinc, copper;

acetic acid: 4.70%.

Preferably, the composition according to the invention comprises the listed mineral salts with the following respective contents: sodium: 1, 70% to 2.50%

- potassium: 0.11%

- magnesium: 0.05%

- phosphorus: 0.055% to 0.060% - sulfur: 0.10%

Also preferably, the composition according to the invention comprises the trace elements listed with the respective contents as follows:

selenium: 29.0 ppm

- manganese: 4.5 ppm - iron: 4.4 ppm

- zinc: 2.0 ppm

- copper: 0.4 ppm

According to an advantageous characteristic of the composition according to the invention, the protein / calcium ratio is in the range from 0.8 to 0.9.

According to a particularly advantageous characteristic, in the composition which is the subject of the invention, the proteins and calcium comprise the solubilized forms of compounds naturally occurring in the poultry eggshell. Indeed, the composition is characterized in that it contains from 0.40% to 0.45% of proteins originating from essentially eggshell and 0.50% to 0.51% calcium from eggshell calcite dissolved in water with 4.70% acetic acid. This state of affairs makes it possible to further underline the natural origin of the composition claimed.

According to an advantageous embodiment, the composition which is the subject of the invention further comprises an aqueous extract of at least one plant.

According to the invention, the components of the claimed composition are preferably of exclusively natural origin. They provide, in a balanced mixture, proteins and calcium mainly from the shell, to which are added the minerals: sodium, potassium, magnesium, phosphorus, sulfur and trace elements: selenium, manganese, iron, zinc, copper from essentially vinegar and sea salt.

According to an advantageous characteristic of the invention, the composition is in the form of a solution of pH 5.5.

Obtaining the composition as described may result from different procedures, but it is particularly recommended to comply with the preparation process which will be described below, insofar as it makes it possible to obtain all the useful components of the process. composition according to the invention in specific proportions.

Thus is claimed a method for obtaining a composition as previously described, essentially comprising the steps of:

- introduce into a tank diluted acetic acid;

- add sodium chloride and stir until completely dissolved;

- introduce raw poultry eggs with their shells without breaking them;

- let rest a month; - remove the eggs, taking care not to perforate the shell membranes;

- filter the solution thus obtained.

According to a preferred characteristic of the claimed process, acetic acid is supplied in the form of white wine vinegar, red wine vinegar or cider vinegar. According to another preferred feature of the claimed process, the sodium is provided as a gray salt.

In a particularly advantageous embodiment of the method according to the invention, the proteins and calcium are provided essentially by the shell of poultry eggs.

Eggs are commonly poultry eggs, and may be chosen for example from chicken eggs and guinea fowl eggs.

According to a preferred embodiment of the process according to the invention, the following ingredients are used:

- 7 ° white wine vinegar: 1 liter

- marine gray salt: 80 grams

- chicken eggs: 3 units

Advantageously, the method according to the invention may comprise a step prior to mixing, in which at least one plant is macerated in dilute acetic acid.

Another object of the present invention is a product for therapeutic or cosmetic use containing the composition as described above.

For the purposes of the formulation, the products may contain ingredients such as a perfume or thickener, or other formulation additive known to those skilled in the art.

The composition according to the invention may also be associated or not with one or more active components intended to potentiate its effects, such as essential oils and plant extracts. The products obtained, which contain said composition associated with another active ingredient, can therefore be proposed according to the specific need to be treated, in one or more of their indications.

Therefore, optionally, the product according to the invention may further comprise one or more ingredients selected from: an essential oil, a plant extract, a perfume, a thickener. The composition according to the invention can be used for the preparation of a medicament for the treatment of alopecia, skin diseases or scalp diseases. It is indicated in particular in the treatment of dermatoses such as seborrheic eczema, seborrheic parakeratosis, psoriasis of the scalp, seborrheic dermatitis, as well as in the treatment of diffuse or circumscribed alopecia accompanied by pityriasis, pruritus and / or seborrhea.

The composition according to the invention can also be used for the preparation of a product for cosmetic use intended for treating the skin or the scalp.

According to the invention, the composition is specially designed to be applied to the scalp and skin. The active ingredients, namely proteins, calcium, minerals, trace elements and acetic acid, are in the form of a pH 5.5 solution favoring their dermal absorption and their bioavailability. It should further be noted that the necessary dose of oligomers being small and still moderate, the composition has the particularity of involving small amounts of elements already naturally present in the body, which pleads in favor of good tolerance.

The invention is illustrated without being limited in any way by the following examples which describe the formulation of a hair fall arrest composition together with a report of clinical experiments and the formulation of an antipruritic composition used in the treatment of eczema.

EXAMPLE 1 Preparation of a composition

The preparation of a composition based on proteins and calcium of the eggshell, minerals, trace elements and acetic acid is carried out as follows. Sodium is added as sea salt; acetic acid is added as vinegar.

1) The ingredients are used in the following proportions:

- chicken eggs 3 units

- marine gray salt 80 grams

- white wine vinegar 7 ° 1 liter 2) One operates as follows: fill a vat with white wine vinegar, add the salt, dissolve by stirring. Add the raw eggs with their shells, immersing them one by one so as not to break them. Leave to rest for a month without shaking. At this stage we observe the complete dissolution of the shells. 3) Elemental analysis of the composition

Protein and calcium are here in a protein / calcium ratio of 0.8.

EXAMPLE 2: Anti-hair loss hair lotion

a) - Macerate for one month in a liter of white wine vinegar at 7 °, 5g of nettle, 5g of nettles, 5g of burdock, 5g of thyme flowers, 5g of nasturtium seeds. A vinegar solution is obtained containing plant compounds from plants having macerated, called "vinegar plants". b) - Proceed to the mixing of the ingredients, as described in Example 1 using as vinegar, the plant vinegar above. The composition C1 is obtained. c) - Proceed with the preparation of the lotion according to the formulation: - Composition C1: 100.00 g - essential oil of lavender: 0.40 g

alkylphenol polyglycol ether: 5.00 g

- perfume composition: 1.00 g

We obtain a hair lotion used in prevention or treatment against hair loss. The lotion may also be provided in the treatment of seborrhea, pruritus and pityriasis, whether these indications are associated or isolated.

EXAMPLE 3 Ointment used in the treatment of pruritus and eczema

a) - Prepare a vinegar of plants by maceration for one month in a liter of white wine vinegar 7 °: 5g of wild pansy flowers, 5g of sarsaparilla roots, 5g of elm pyramidal barks, 5g of juniper berries, 5g d sweet woodruff. b) - Proceed to the mixing of the ingredients, as described in Example 1 using as vinegar, the plant vinegar above. The composition C2 is obtained. c) - Proceed with the preparation of the lotion according to the formulation:

A cream for topical application is obtained in the treatment of pruritus and eczema. The cream can be proposed in these indications that they are associated or isolated.

EXAMPLE 4: Clinical Trials

The demonstration of the effectiveness of composition according to the invention is based on clinical experiments conducted at the Saint Louis Hospital in Paris in the department of diseases of the scalp. 1) Protocol:

A hair lotion was prepared according to Example 2 above and tested on 40 people (22 males, 18 females), aged 13 to 65 years. Patients, male or female, were selected based on a moderate to severe tendency to hair loss. For men, androgenetic alopecia is observed in all cases. For women, the majority have Ludwig's Type I female alopecia. Of these, four are menopausal or already menopausal.

The patient applies each evening a third dropper pipette (ie 2 ml) the lotion. It is reviewed on the 2 ^nd and 3 ^rd month (according to need 5 ^th , 6 ^th , 7 ^th or 10 ^th month) after the first consultation. Seborrhea, pruritus, pityriasis and assessment of hair loss are graded at each consultation on a scale of 0 to 4. A sample for a trichogram is taken at the first consultation and after three months of treatment.

The trichograms allowing the study of the impact on the hair formula, are made by taking a lock of hair with the forceps in the frontal zone of the hair and, if possible, in one of the temporal zones. All samples of the same patient are made by the same doctor, who also performs trichograms. This one also carries out the anamnesis and the clinical examination.

2) Results

- Seborrhea

Seborrhea decreased in 68.3% of cases, or "very little" in 58% of cases, or a good 10% of cases. This anti-seborrhoeic effect, however, has little effect on the frequency of shampoos.

- Pityriasis capitis

Pellicular state is reduced in 82% of cases, of which 46.4% of cases are "well diminished" and 35.7% of cases are "very diminished". This effect is observed early, ie from the end of the first month, and is maintained. - Pruritus

For 79% of cases, pruritus is reduced, with one third of patients considering it as suppressed and 45.8% of cases reporting it "decreased". This antipruritic effect is obtained at the end of the first month. - Anti fall effect

A reduction in hair loss is reported by 59.5% of patients. It is considered "diminished" by 47.5% of patients and as "very diminished" by 12.5% of patients. This effect is observed as early as the second month. It stays in the 5 ^th and 6 ^th months.

Overall the clinical effects are defined as fast by the patients. They call their hair "brighter, softer, more supple, more toned, thicker and giving the hair a volume look".

3) Conclusions

At the end of the clinical study and the anamnesis, the results obtained are remarkable.

Antipruritic and anti-dandruff effects are most satisfactory. Given the improvements noted, the lotion tested can be proposed in local treatment in these indications, whether associated or isolated.

The anti-hair loss effect is found in trichograms for one-third of the patients in the frontal area and for half in the temporal areas. Whatever the age and sex of the patients, the efficacy of the lotion tested is even better than the trend of alopecia is recent, with the persistence of a good capital hair. The five-month course provides the most satisfactory results.

Women also perform well when they are menopausal or already menopausal. The formula is therefore used in men as well as women, and at any age.
