Abstract

L'invention porte sur des procédés d'évaluation de l'aberration du nombre de copies dans une population clonale de cellules. Le procédé comprend les opérations consistant à purifier des noyaux provenant de cellules d'intérêt à l'aide d'un tri de cellules activées par fluorescence, amplifier l'ADN provenant des noyaux avec une ADN polymérase hautement active telle que phi29, traiter l'ADN avec une enzyme qui catalyse des ruptures dans l'ADN telle que l'ADNase ou une endonucléase de restriction, marquer l'ADN par un marqueur fluorescent et hybrider l'ADN à une puce à ADN capable d'effectuer une hybridation génomique comparative sur microréseau d'ADN. Les cellules d'intérêt comprennent des cellules tumorales et peuvent être issues de tissu noyé dans la paraffine fixé par du formaldéhyde. Les paramètres pour isoler les noyaux comprennent la teneur en ADN et l'état de cycle cellulaire et les noyaux peuvent être isolés par centrifugation ou autre dissociation mécanique en présence ou en l'absence de marqueur fluorescent. Un ADN témoin peut être inclus et marqué par un marqueur fluorescent autre que celui de la molécule d'ADN provenant de la cellule d'intérêt.
