Abstract

  【課題】 シグナルの有無を指標として標的特異的リガンドを簡便かつ正確に検出することのできる単一分子型のプローブを提供する。 【解決手段】 生細胞における標的特異的リガンドを検出する一分子型生物発光可視化プローブであり、リガンドを結合した場合にその立体構造を変化させるリガンド結合分子の両端に、発光酵素(LE)を分割したN末端側ポリペプチド(N-LE)とC末端側ポリペプチド(C-LE)とがそれぞれ連結近接され、リガンド結合分子にリガンドが結合した場合のみ、N-LEとC-LEとが可逆的に自己相補して発光シグナルを発する。リガンド結合分子は、例えば、リガンド結合ドメイン(LBD)と、リガンドの結合によってLBDと相互作用するドメイン(LBD相互作用ドメイン)あるいはペプチドとが連結した融合分子である。
