Abstract

Procédés d'amélioration de l'expression d'anticorps par réingénierie d'immunoglobulines qui consiste à sélectionner une série de séquences cadres assurant une expression élevée et une liaison spécifique élevée de l'anticorps cible. Les procédés consistent à fournir un ensemble de séquences d'acides aminés correspondant aux segments cadres de zones variables de chaîne légère ou lourde dérivés d'au moins deux chaînes lourdes ou légères d'immunoglobuline, à remplacer les segments cadres de chaîne lourde ou légère d'immunoglobuline parente par des séquences de l'ensemble de séquences d'acides aminés pour obtenir des immunoglobulines à cadres modifiés par réingénierie. Dans certaines variantes, les séquences cadres sélectionnées pour être utilisées dans la la réingénierie d'immunoglobuline ne sont pas toutes dérivées de la même immunoglobuline. Les procédés décrits offrent une nouvelle approche de l'humanisation des anticorps.
