Abstract
L'invention concerne un système et un procédé de conduite d'une amplification en chaîne par polymérase (PCR) en temps réel. Des molécules de capture d'une conception particulière sont immobilisées sur un support solide, et mises en contact avec des amplicons obtenus en un ou plusieurs cycles de PCR. La détection des amplicons peut avoir lieu pendant ou entre les cycles de PCR, tandis que le support solide est en contact fluidique avec la solution de PCR. Dans un mode de réalisation, la détection des amplicons a lieu lorsque le support solide n'est pas en contact fluidique avec la solution de PCR. Dans un autre mode de réalisation, des amplicons marqués sont détectés à l'aide d'un champ évanescent tout en étant hybridés pour capturer des sondes immobilisées sur un support solide dans la chambre de réaction de PCR en temps réel. Le procédé est approprié pour la détection et la quantification simultanées de molécules cibles étroitement homologues.
