[0005] しかしながら、リボソームなどの微粒子キャリアーの表面をポリアルキレングリコール で修飾した場合には、血中滞留性は改善するものの、標的細胞の内部に微粒子キヤ リア一が取り込まれに《なるという新たな問題が生じることが知られている。この問題 は、特に、薬剤封入リボソームや遺伝子封入 MENDを用いて標的細胞に特異的に抗 腫瘍剤や遺伝子を送達するターゲッティング治療にぉ ヽて、期待したほどの治療効 果を達成できず、また標的細胞に送達されなかった抗腫瘍剤や遺伝子による副作用 も十分に回避できないという深刻な問題を引き起こしている。
[0006] 一方、腫瘍細胞は増殖や浸潤 '転移の過程で細胞外マトリックス (ECM)を分解-再 構築するが、マトリックスメタ口プロテアーゼ (MMP)が重要な役割を果たしていること が知られている(Cell. 91, pp.439- 442, 1997; APMIS, 107, pp.137- 143, 1999)。 MM Pには 20以上のファミリーが同定されており、分泌型と細胞膜上に存在する膜型に分 類されている。 MMPはコラーゲンなどの ECMを分解する機能を有している力 最近の 研究で MMPにより特異的に分解 '切断されるペプチド配列が明らかとなつて!/、る (Nat ure Biotechnology, 19, pp.661- 667, 2001)。
[0007] また、リボソームの構成脂質として利用可能なペプチド結合リン脂質が知られている (特表 2003-513009号公報)。上記公報には、このリン脂質のペプチドは PEG (ポリェチ レンダリコール)などで修飾されて 、てもよ 、との説明がある(上記公報 [0016]段落)。 このリン脂質のペプチドがぺプチダーゼにより開裂されると、この脂質を構成脂質とし て含むリボソームが不安定ィ匕して含有物がその部位で放出されるので、ぺプチダー ゼ分泌細胞に対して特異的に含有物を送達することができる。また、上記べプチダ ーゼとしてはマトリックスメタ口プロテアーゼを利用でき、腫瘍細胞をターゲットとしてリ ポソームを利用できることが教示されて!、る(上記公報 [0021]段落及び [0042]段落)。 もっとも、このリン脂質を用いたリボソームによる薬剤又は遺伝子の送達効率は十分と は言えない。

発明が解決しょうとする課題
[0008] 本発明の課題は、表面をポリアルキレングリコールなどで修飾したリボソームや MEN Dなどの脂質膜構造体を微粒子キャリア一として用いて、マトリックスメタ口プロテア一 ゼの関わる疾患 (悪性腫瘍、肺気腫、肺線維症、慢性関節リウマチ、変形性関節炎、 歯周炎、角膜潰瘍、ブドウ膜炎、糖尿病性網膜症、増殖性硝子体、加齢黄斑変性症 、水泡性疾患、慢性皮膚潰瘍、多発性硬化症、糸球体腎炎、ループス腎炎、慢性肝 炎、肝硬変、心不全など)の予防'治療に関する手段を提供することにある。より具体 的には、マトリックスメタ口プロテアーゼの関わる悪性腫瘍などの疾患部位や新生血 管の近傍に抗腫瘍剤などの薬物や悪性腫瘍等の遺伝子治療のための遺伝子を送 達し、引いては標的細胞内に送達するにあたり、標的細胞の内部に抗腫瘍剤などの 薬物や遺伝子を効率的に取り込ませる手段を提供することにある。
課題を解決するための手段
[0009] 本発明者らは上記の課題を解決すべく鋭意研究を行なった結果、ポリアルキレング リコールなどで修飾した特定のリン脂質にぉ 、て、ポリアルキレングリコールなどの修 飾部分とリン脂質部分との間にマトリックスメタ口プロテアーゼにより加水分解可能な オリゴペプチドを介在させ、その脂質を用いてリボソームや MENDなどの脂質膜構造 体を調製して抗腫瘍剤や遺伝子などの微粒子キャリア一として用いると、血中にお!ヽ てはポリアルキレングリコールなどの修飾部位の存在により該リボソームや MENDの 高 ヽ血中滞留性が保たれる一方で、標的組織に到達した該リボソームや MENDでは 、マトリックスメタ口プロテアーゼにより該オリゴペプチド部分が加水分解されてポリア ルキレングリコールなどの修飾部位が脱離することになる。その結果、一つは、ポリア ルキレングリコールなどの修飾部位が脱離したリボソームや MENDの安定性が低下し てその構造を保つことができずに、脂質膜構造体に保持された抗腫瘍剤等を標的細 胞の外部にて放出する方法によって、またもう一つは、ポリアルキレングリコールなど による修飾が脱離したリボソームや MENDに変化してこれらの脂質膜構造体がそのま ま標的細胞に効率的に取り込まれる方法によって、あるいはこれら二つの方法が合 わさること〖こよって、高い導入率で抗腫瘍剤や遺伝子を標的細胞内に送達できること を見出した。また、このようにして効率的に抗腫瘍剤や遺伝子を標的細胞に送達する ことにより、極めて高い抗腫瘍効果を達成できることを見出した (特願 2006— 049567 号明細書)。
[0010] 本発明者らは、さらに研究を重ねるうち、ポリアルキレングリコールなどの修飾部分 とリン脂質部分との間にマトリックスメタ口プロテアーゼにより加水分解可能なオリゴぺ プチドを介在させた脂質、及び該オリゴペプチドの介在のな 、ポリアルギレングリコー ルなどの修飾リン脂質とを組み合わせて MENDなどの脂質膜構造体を調製して抗腫 瘍剤ゃ遺伝子を標的細胞として悪性腫瘍細胞に送達すると、極めて高!ヽ導入率で 送達できることを見出した。本発明は上記の知見を基にして完成されたものである。
[0011] すなわち、本発明により、少なくとも下記の 2種類のリン脂質誘導体:
(1)アルコールィ匕合物の残基とリン脂質類の残基とを含み、該アルコールィ匕合物の残 基とリン脂質の残基との間にペプチドを含むリン脂質誘導体であって、

[0012] この発明の好ましい態様によれば、マトリックスメタ口プロテアーゼの関わる疾患に 有効な薬物や遺伝子治療のための遺伝子を保持した上記脂質膜構造体;及び、リポ ソームである上記脂質膜構造体が提供される。
さらに好ましい態様によれば、リン脂質誘導体 (1)及び (2)において、該アルコール化 合物がポリアルキレングリコール類であり、該リン脂質がホスファチジルエタノールアミ ン類であり、リン脂質誘導体 (1)において、該ペプチドが Va卜 Pro- Leu- Ser- Leu- Tyr- S er-Glyを含むペプチドである上記の脂質膜構造体;リン脂質誘導体 (1)及び (2)にお ヽ て、該アルコール化合物がポリエチレングリコールであり、該リン脂質がジォレイルホ スファチジルエタノールァミン又はジステアロイルホスファチジルエタノールァミンであ り、リン脂質誘導体 (1)において、該ペプチドが Gly-Gly-Glyをリンカ一として含む上記 の脂質膜構造体;及び、リン脂質誘導体 (1)において、該ペプチドが Gly- Gly- Gly- Val - Pro- Leu- Ser- Leu- Tyr- Ser- Gly- Gly- Gly- Glyである上記の脂質膜構造体が提供さ れる。特に好ましい態様によれば、リン脂質誘導体 (1)とリン脂質誘導体 (2)の脂質膜 構造体の全脂質量に対する割合が約 20モル %である上記の脂質膜構造体が提供さ れる。
[0013] 別の観点からは、マトリックスメタ口プロテアーゼの関わる疾患に有効な薬物や遺伝 子治療のための遺伝子を保持した上記脂質膜構造体を含むマトリックスメタ口プロテ ァーゼの関わる疾患の予防及び z又は治療のための医薬組成物が提供される。 これらにカ卩えて、マトリックスメタ口プロテアーゼの関わる疾患の治療方法であって、 マトリックスメタ口プロテアーゼの関わる疾患に有効な薬物や遺伝子治療のための遺 伝子を保持した上記脂質膜構造体をヒトを含む哺乳類動物に投与する工程を含む 方法が本発明により提供される。
発明の効果
[0014] 本発明の脂質膜構造体は、マトリックスメタ口プロテアーゼにより上記のリン脂質誘 導体 (1)のペプチド部分が切断されてポリアルキレングリコールなどの修飾部位を脱 離する一方、リン脂質誘導体 (2)に由来するポリアルキレングリコールが脂質膜構造 体表面に残留する特性を有している。本発明の脂質膜構造体は、血中においては 該修飾部位の存在により安定であり、マトリックスメタ口プロテアーゼを分泌する悪性 腫瘍細胞や新生血管などの近傍では、該修飾部位が脱離することにより脂質膜構造 体の安定性が低下して、その構造を保つことができずに、脂質膜構造体に保持され た抗腫瘍剤や遺伝子等を標的細胞の外部で放出し、あるいは脂質膜構造体が標的 細胞中に効率的に取り込まれることによって、標的細胞中に薬剤や遺伝子を効率的 に導入することができる。さらに、本発明の脂質膜構造体ではポリアルキレングリコー ルなどの修飾部位の一部が悪性腫瘍細胞や新生血管などの近傍においても残留し 、十分な血中滞留性を維持することができるという特徴がある。

[0016] 本発明の脂質膜構造体の製造に用いられるリン脂質誘導体のうち、少なくとも 1種 のリン脂質誘導体は、アルコールィ匕合物の残基とリン脂質類の残基とを含み、該アル コールィ匕合物の残基とリン脂質の残基との間にペプチドを含むリン脂質誘導体であ つて、
(a)該アルコール化合物が、ポリアルキレングリコール類、グリセリン類、及びポリグリセ リン類カゝらなる群カゝら選ばれるアルコールィ匕合物であり、

[0018] アルコール化合物としては、ポリエチレングリコール又はポリプロピレングリコールな どのポリアルキレングリコール類、グリセリン又はグリセリンエステルなどのグリセリン類 、あるいはジグリセリン、トリグリセリン、テトラグリセリン、ペンタグリセリン、へキサグリセ リン、ヘプタグリセリン、又はオタタグリセリンなどのポリグリセリン類を用いることができ る。これらのうち、ポリアルキレングリコールが好ましぐ特に好ましいのはポリエチレン グリコールである。ポリエチレングリコールを用いる場合、分子量は特に限定されず、 脂質膜構造体に血中滞留性などの所望の特性を付与するために当業者が適宜選 択可能である。
[0019] アルコール化合物の残基とは、アルコール化合物又は化学修飾されたアルコール 化合物から水素原子などの適宜の原子又は水酸基やハロゲン原子などの適宜の官 能基を除いて得られる基を意味しており、好ましくは 1価の基を意味している。例えば
、アルコールィ匕合物の水酸基力 水素原子を除いて得られる残基、アルコール化合 物の炭素原子に結合する水素原子を除 、て得られる残基、ある 、はアルコール化合 物の末端にカルボキシル基を含む官能基を導入し、そのカルボキシル基カゝら水酸基 又は水素原子を除いて得られる残基などを例示できるが、これらに限定されることは ない。
[0020] リン脂質としては、ホスファチジルエタノールアミン類、ホスファリジルコリン類、ホス ファチジルセリン類、ホスファチジルイノシトール類、ホスファチジルグリセロール類、 カルジォリピン類、スフインゴミエリン類、セラミドホスホリルエタノールアミン類、セラミ ドホスホリルグリセロール類、セラミドホスホリルグリセロールホスファート類、 1,2-ジミリ ストィル- 1,2-デォキシホスファチジルコリン類、プラスマロゲン類、またはホスファチジ ン酸類を用いることができるが、これらのリン脂質における脂肪酸残基は特に限定さ れない。例えば、炭素数 12〜20個程度の飽和又は不飽和の脂肪酸残基を 1個又は 2個有するリン脂質を用いることができ、具体的には、ラウリン酸、ミリスチン酸、パルミ チン酸、ステアリン酸、ォレイン酸、リノール酸等の脂肪酸由来のァシル基を 1個又は 2個有するリン脂質を用いることができる。これらのうち、ホスファチジルエタノールアミ ン類が好ましぐ具体的にはジミリストイルホスファチジルエタノールァミン、ジパルミト ィルホスファチジルエタノールァミン、ジステアロイルホスファチジルエタノールアミン( DSPE)、ジォレイルホスファチジルエタノールァミン (DOPE)等を挙げることができ、中 でもジステアロイルホスファチジルエタノールァミン (DSPE)、ジォレイルホスファチジル エタノールァミン (DOPE)が好まし!/、。
[0021] リン脂質の残基とは、上記のリン脂質又は化学修飾されたリン脂質力も水素原子な どの適宜の原子又は水酸基やハロゲン原子などの適宜の官能基を除いて得られる 基を意味しており、好ましくは 1価の基を意味している。例えば、リン脂質のリン酸部 分の水酸基力も水素原子を除いて得られる残基、リン脂質のリン酸部分の水酸基を 除いて得られる残基、リン脂質の炭素原子に結合する水素原子を除いて得られる残 基、あるいはリン脂質のリン酸部分にカルボキシル基を含む官能基を導入し、その力 ルポキシル基力 水酸基又は水素原子を除いて得られる残基などを例示できるが、 これらに限定されることはない。
[0022] アルコールィ匕合物の残基とリン脂質の残基との間に含まれるペプチドは、マトリック スメタ口プロテア一ゼの基質となりうる基質ペプチドを少なくとも 1個含んでいる。マトリ ックスメタ口プロテアーゼ (MMP)には、例えば、 MMP-1 (間質型コラーゲナーゼ)、 MM P- 2 (ゼラチナーゼ A)、 MMP-3、 MMP-7, MMP_9 (ゼラチナーゼ B)などの存在が知ら れており、これらのうちの 1種又は 2種以上のマトリックスメタ口プロテア一ゼの基質と なりうる基質ペプチドを用いることができる。マトリックスメタ口プロテアーゼについては 、例えば、鶴尾隆編「癌転移の分子機構」、 pp.92-107、メジカルビユー社、 1993年発 行などを参照することができる。
[0023] マトリックスメタ口プロテア一ゼの基質となりうる基質ペプチドについては、例えば、 上記非特許文献 1 (Nature Biotechnology, 19, pp.661- 667, 2001)に特定の種類の マトリックスメタ口プロテアーゼと、それに特異的に認識される基質ペプチドが説明さ れているので、この刊行物を参照することにより、特定の種類のマトリックスメタ口プロ テアーゼにより特異的に切断される基質ペプチドを選択することが可能である。例え ば、 MMP- 9につ!/、て Va卜 Pro- Leu- Ser- Leu- Tyr- Ser- Glyが特異的基質として知られ ており、 MMP-9の基質となりうる基質ペプチドとして上記のォクタペプチドを用いるこ とが好ましい。上記非特許文献 1の開示の全てを参照により本明細書の開示として含 める。

[0024] アルコールィ匕合物の残基とリン脂質の残基との間に含まれるペプチドは、基質ぺプ チドのほか、アルコール化合物の残基及び Z又はリン脂質の残基との結合に関与す るリンカ一として、 1個のアミノ酸力 なるリンカ一、又は 2ないし 8個のアミノ酸を含むォ リゴペプチドからなるリンカ一を含んでいてもよい。リンカ一である該アミノ酸又は該ォ リゴペプチドは、該基質ペプチドの両端に結合していてもよいが、片端のみに結合し ていてもよい。該基質ペプチドの両端に該リンカ一が結合している場合には、 2つのリ ンカ一は同一でも異なっていてもよい。リンカ一を介さずに、基質ペプチドが直接ァ ルコール化合物の残基及び Z又はリン脂質の残基と結合して 、てもよ ヽ。上記のリン 脂質誘導体ではリンカ一が存在することが好ましぐ該基質ペプチドの両端に同一又 は異なるリンカ一が存在していることがさらに好ましぐ該 2個のリンカ一が 2ないし 8個 のアミノ酸を含むオリゴペプチドからなる同一又は異なるリンカ一であることがより好ま しい。

本発明により提供される脂質膜構造体の種類は特に限定されな!ヽが、脂質膜構造 体が水系溶媒に分散した形態としては、例えば、一枚膜リボソーム、多重層リポソ一 ム、 0/W型エマルシヨン、 W/0/W型エマルシヨン、球状ミセル、ひも状ミセル、不定 型の層状構造物などを挙げることができる。これらのうちリボソームが好ましい。分散 した状態の脂質膜構造体の大きさは特に限定されないが、例えば、リボソームやエマ ルシヨンの場合には粒子径が 50 nm力ら 5 μ mであり、球状ミセルの場合、粒子径が 5 nmから 100 nmである。ひも状ミセルや不定型の層状構造物の場合は、その 1層あたり の厚みが 5 nmから 10 nmであり、これらが層を形成しているものが好ましい。

水系溶媒に既に分散したリボソーム、エマルシヨン、ミセル、又は層状構造物などの 脂質膜構造体に、さらにマトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及 び Z又は遺伝子治療のための遺伝子を含む水系溶媒を添加して製造する方法であ る。対象となるマトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び Z又は 遺伝子治療のための遺伝子としては、水溶性のものを利用できる。既にでき上がって いる脂質膜構造体に外部力もマトリックスメタ口プロテアーゼの関わる疾患に有効な 薬物及び Z又は遺伝子治療のための遺伝子を添加する方法であることから、マトリツ タスメタ口プロテアーゼの関わる疾患に有効な薬物及び z又は遺伝子治療のための 遺伝子が高分子の場合には、マトリックスメタ口プロテアーゼの関わる疾患に有効な 薬物及び Z又は遺伝子治療のための遺伝子は脂質膜構造体の内部には入り込め ず、脂質膜構造体の表面に存在 (結合)した存在様式をとる可能性がある。脂質膜構 造体としてリボソームを用いた場合、この製造方法 3を用いると、マトリックスメタ口プロ テアーゼの関わる疾患に有効な薬物及び z又は遺伝子治療のための遺伝子がリポ ソーム粒子同士の間に挟まったサンドイッチ構造 (一般的には複合体あるいはコンプ レックスと呼ばれている。)を形成することが知られている。この製造方法では、脂質 膜構造体単独の水分散液をあら力じめ製造するため、乳化時におけるマトリックスメ タロプロテアーゼの関わる疾患に有効な薬物及び z又は遺伝子治療のための遺伝 子の分解等を考慮する必要がなぐ大きさ (粒子径)の制御もし易い。したがって、製 造方法 1や製造方法 2に比べて比較的容易に製造することができる。

水系溶媒に分散した脂質膜構造体を製造して乾燥することにより得られた乾燥物 に、さらにマトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び/又は遺伝 子治療のための遺伝子を含む水系溶媒を添加して製造する方法である。製造方法 3 と同様に対象となるマトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び Z 又は遺伝子治療のための遺伝子としては、水溶性のものを利用できる。製造方法 3と の相違点は、脂質膜構造体とマトリックスメタ口プロテアーゼの関わる疾患に有効な 薬物及び Z又は遺伝子治療のための遺伝子との存在様式にあり、この製造方法 4で は、水系溶媒に分散した脂質膜構造体を一旦製造した上でさらに乾燥させた乾燥物 を製造することから、この段階で脂質膜構造体は脂質膜の断片として固体状態で存 在する。この脂質膜の断片を固体状態に存在させるためには、前記したように水系溶 媒に、さらに糖 (水溶液)、好ましくはショ糖 (水溶液)や乳糖 (水溶液)を添加した溶 媒を用いることが好ましい。ここで、マトリックスメタ口プロテアーゼの関わる疾患に有 効な薬物及び Z又は遺伝子治療のための遺伝子を含む水系溶媒を添加すると、固 体状態で存在していた脂質膜の断片は水の侵入とともに速やかに水和し始め、脂質 膜構造体を再構築することができる。この時、マトリックスメタ口プロテアーゼの関わる 疾患に有効な薬物及び Z又は遺伝子治療のための遺伝子が脂質膜構造体内部に 保持された形態の構造体が製造できる。
製造方法 3では、マトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び Z 又は遺伝子治療のための遺伝子が高分子の場合には、マトリックスメタ口プロテア一 ゼの関わる疾患に有効な薬物及び Z又は遺伝子治療のための遺伝子は脂質膜構 造体内部には入り込めず、脂質膜構造体の表面に結合した存在様式をとるが、製造 方法 4はこの点で大きく異なる。すなわち、マトリックスメタ口プロテアーゼの関わる疾 患に有効な薬物及び Z又は遺伝子治療のための遺伝子の一部又は全部は脂質膜 構造体内部に取り込まれる。この製造方法 4は、脂質膜構造体単独の分散液をあら 力じめ製造するため、乳化時のマトリックスメタ口プロテアーゼの関わる疾患に有効な 薬物及び Z又は遺伝子治療のための遺伝子の分解を考慮する必要がなぐ大きさ( 粒子径)の制御もし易い。従って、製造方法 1や製造方法 2に比べて比較的製造が 容易である。また、この他に、一旦凍結乾燥又は噴霧乾燥を行うため、製剤(医薬組 成物)としての保存安定性を保証し易ぐ乾燥製剤をマトリックスメタ口プロテアーゼの 関わる疾患に有効な薬物及び Z又は遺伝子治療のための遺伝子の水溶液で再水 和しても大きさ (粒子径)を元に戻せること、高分子の抗腫瘍剤及び Z又は遺伝子で あっても、脂質膜構造体内部にマトリックスメタ口プロテアーゼの関わる疾患に有効な 薬物及び z又は遺伝子治療のための遺伝子を保持させ易いことなどの長所がある。

[0064] マトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び Z又は遺伝子治療 のための遺伝子を保持した脂質膜構造体を含む本発明の医薬組成物は、マトリック スメタ口プロテアーゼの関わる疾患の予防及び Z又は治療に有用である。本発明の 医薬組成物により予防及び Z又は治療可能な疾患は限定されるべきものではなぐ また疾患が悪性腫瘍の場合、悪性腫瘍としての種類は特に限定されないが、特にマ トリックスメタ口プロテアーゼの発現が多 、悪性腫瘍が好適である。悪性腫瘍細胞とし ては、例えば、線維肉腫、扁平上皮癌、神経芽細胞腫、乳癌、胃癌、肝細胞癌、膀 胱癌、甲状腺腫瘍、尿路上皮癌、グリア芽細胞腫、急性骨髄性白血病、脾管癌及び 前立腺癌等の細胞を挙げることができるが、これらの細胞に限定されることはない。ま た、本発明の医薬組成物をヒト等の動物や実験用細胞に投与すると、腫瘍内部にお ける血管新生先端部位にマトリックスメタ口プロテアーゼの関わる疾患に有効な薬物 としての抗腫瘍剤及び Z又は遺伝子を効率的に送達することができる。腫瘍内部に おける血管新生先端部位としては、 ruffling edgeの内皮細胞(endothelial cells)などを 挙げることができる力 これに限定されることはない。
[0065] いかなる特定の理論に拘泥するわけではないが、本発明の医薬組成物をヒトを含 む哺乳類動物に投与すると、血中においてはポリアルキレングリコールなどの修飾部 位の存在により優れた血中滞留性が得られ、一方、マトリックスメタ口プロテアーゼを 分泌する悪性腫瘍細胞や新生血管の近傍などで、該修飾部位が脱離することにより 、本発明の医薬組成物に含まれるマトリックスメタ口プロテアーゼの関わる疾患に有効 な薬物及び Z又は遺伝子治療のための遺伝子が放出され、あるいは該医薬組成物 が標的細胞により取り込まれ易くなることにより、該細胞に対して薬物及び Z又は遺 伝子の効果を発揮させることができる。また、一部の修飾部位は脱離せずに脂質膜 構造体表面に残留して適度な血中滞留性を与える。
[0066] 本発明の脂質膜構造体を含む医薬組成物の投与方法は特に限定されず、経口投 与又は非経口投与のいずれも選択可能である。経口投与の剤形としては、例えば、 錠剤、散剤、顆粒剤、シロップ剤、カプセル剤、内服液剤等を挙げることができ、非経 口投与の剤形としては、例えば、注射剤、点滴剤、点眼剤、軟膏剤、坐剤、懸濁剤、 パップ剤、ローション剤、エアゾール剤、プラスター剤等を挙げることができる。これら のうち注射剤又は点滴剤が好ましぐ投与方法としては、静脈注射、動脈注射、皮下 注射、皮内注射などのほか、標的とする細胞や臓器に対しての局所注射を挙げるこ とができる。本発明の医薬組成物の投与量及び投与期間などは特に限定されず、有 効成分として作用するマトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び Z又は遺伝子治療のための遺伝子の種類や保持量、脂質膜構造体の種類、対象疾 患の種類や患者の体重、年齢など、種々の条件に応じて適切な投与量及び投与期 間を適宜選択可能である。
なお、上記リン脂質誘導体 (1)を用いた脂質膜構造体は特願 2006— 049567号明細 書に開示されている。上記明細書の全ての開示を参照により本明細書の開示として 含める。
実施例
[0067] 以下、実施例により本発明をさらに具体的に説明するが、本発明の範囲は下記の 実施例に限定されることはない。
例 1
NHSは和光純薬より購入した。 DCCは関東化学より購入した。 GPCは Tosoh8120 ( 東ソー株式会社)、検出器 Tosoh HLC- 8120(RI)、カラム TSK- Gel SuperHZ3000,2500 を使用し、キャリアを THF、流速を 0.35 mL/min,温度を 40°Cに設定した。 'H-NMRは JEOL EX- 400 (400MHz)使用し測定した。 TOF- MSは Bruker MALDI- TOF- MS Refl ex IIを使用し測定した。透析膜 Spectra/Por Membren MWCO: 1,000は Spectrumより 購入した。ペプチドはグライナ一ジャパンより購入した。配列は Gly-Gly-Gly-Va卜 Pro - Leu- Ser- Leu- Tyr-Ser-Gly-Gly-Gly-Glyであり分子量は 1178.9であった。
[0068] (1)ポリエチレングリコール (PEG)の合成 アルゴン下、ナスフラスコにテトラヒドロフラン (THF、 20mL)を添カ卩し、スターラーで撹 拌しながら、重合開始剤として 2-メトキシエタノールを 80 L添加した。続いてカリウム ナフタレ-ド(0.342 mol/L)を 2.9 mL添加した。次にエチレンオキサイド (EO)を 2.3 mL (45 mmol)を添加した。常圧下、常温で撹拌しながら 48時間反応した。 48時間後、反 応溶液の一部 (約 2 mL)を取りゲル浸透クロマトグラフィー(GPC)により分子量を測定 した。数平均分子量 (Mn)は 1,678であり、重量平均分子量 (Mw)は 1,825であった。ま た、 Mw/Mnで求められる多分散度は 1.078と小さぐ GPCで単分散のピークが得られ たことから、 PEGの重合が十分に進んでいると判断した。
[0069] (2) PEG末端のカルボキシルイ匕
上記 (1)で得た反応液のうち半量の 12.5 mLをアルゴン下でナスフラスコに添カロし、 無水コハク酸(0.727 mol/L)を 3.5 mL添加し、常温で 24時間、スターラーで撹拌しな 力 PEG重合の停止反応を行なった。 24時間後、反応液をジェチルエーテル(1 L) で沈殿させた。沈殿物を濾取してビーカーに取り、メタノールで溶解した。再びジェ チルエーテル(1 L)で沈殿させ、沈殿物を濾取してメタノールに溶解し、蒸留水で透 祈した。透析後、ナスフラスコに移して凍結乾燥を行ない、淡黄色の粉末 (0.904 g)を 得た。 GPCにより分子量を測定したところ、 Mnは 1,773、 Mwは 1,903であった。多分散 度は 1.073であり、単分散のピークが得られたことから、副反応なく反応が進行してい ることが確認できた。また、 ^-NMR ^ベクトルを測定したところ、 3.4 ppm付近のメトキ シ基(OMe)由来のピークと 2.6 ppm付近の - CO-CH CH -CO-由来のピークの積分

比はほぼ 1: 1であり、末端にカルボン酸を導入したことが確認された。

[0070] (3) PEG末端のカルボキシル基の活性エステル化(MeO-PEG- NHSの合成)
上記 (2)で得た化合物 0.2 gを 50 mL用スクリュー管に添カ卩した。 NHS 57.6 mg、 DCC 41.26 mg、及びクロ口ホルム 10 mLをスクリュー管に添カ卩し、スターラーで撹拌しなが ら常温で 24時間反応した。反応終了後、固体を除くためにろ過を行ない、ろ液を 300 mLのジェチルエーテルで沈殿させた。沈殿物を濾取して乾燥し、得られた固体をビ 一力一に取り、クロ口ホルムに溶解した。ジェチルエーテルでの沈殿及び濾取を 2度 繰り返し、得られた固体をクロ口ホルムに溶解して 100 mLナスフラスコに入れ、溶媒を 留去した後にベンゼンを添加して凍結乾燥し、淡褐色粉末 (104 mg)を得た。 TOF-M Sによる Mnは 2192.12であり多分散度は 1.02であった。
[0071] TOF-MSスペクトルから各ピークの間隔は 44でありこれは PEGのそれぞれのユニット の分子量であることが確認できた。 TOF-MSスペクトルでの各ピークの測定値と理論 値を比較することにより、末端に目的の官能基が導入できたかを確認することができ る。表 1に各 n数に対する分子量の理論値と測定値を示した。各 nに対する測定誤差 は 2mass以内であり、 目的とする化合物が得られたことを示している。また、 ^-NMR^ ベクトルを測定したところ、 NHS基の導入率 (3.37 ppmのメトキシ基に由来するピーク と 2.84 ppmの NHS基に由来するピークの積分比)は 97.3%であり、ほぼ 100%の割合で NHS基が導入されたことが確認できた。

[0073] (4) PEG-ペプチドの合成
ペプチド 24.6 mgにジメチルスルホキシド (DMSO) 1 mL、上記 (3)で得た化合物 41. 17 mg、及びトリェチルァミン 5 Lを添カ卩し、常温で 3時間振とうして反応させた。 HPL Cにより反応終了を確認し、凍結乾燥して白色粉末 (66.76 mg)を得た。 TOF-MSを測 定したところ Mnは 3223.82であり、理論値のである 3255.93とは 32mass違っていたが、 分子量の分布をもっていること力も PEG由来の化合物であり、 PEG-ペプチドであると 推定された。また、多分散度は 1.01で、単分散のピークであることから反応は定量的 に進行していることを確認した。 NMR ^ベクトルを測定したところ、 6.6-7.4 ppmに ペプチド中のチロシンのベンゼン環由来のピークを確認した。 3.38 ppm付近のメトキ シ基に由来するピークとチロシンのベンゼン環由来のピークの積分比がモル比で 1: 1 であることから、 PEG末端へペプチドが 100%導入されて!、ることが確認できた。
[0074] (5) PEG-ペプチド- DOPEの合成
例 (4)で得た PEG-ペプチド 54.7 mgをクロ口ホルム 4 mLに溶解し、 DOPE 24.7 mg、 DCC 6.85 mg、 NHS 3.82 mg、及びトリェチルァミン 4.63 μ 1を添カ卩し、常温で 48時間 振とう反応させた。反応終了後、デシケーターによりクロ口ホルムを除き、再びクロロホ ルム 1.5 mLに溶解した。スターラーで撹拌しながらジェチルエーテル 100 mLに上 記のクロ口ホルム溶液をゆっくり滴下し、 PEG由来化合物を析出させた。常温で個体 が沈殿するまで静置し、上清を除いた。ジェチルエーテル 50 mLを添加しスターラー で撹拌後、沈殿するまで静置して上清を取り除いた。沈殿物を水で溶解し、 HPLCに て分離精製し、分取した画分を透析して凍結乾燥した。 TOF-MSより求められた Mn は 3955.15、多分散度は 1.00であった。ピークが分子量分布をもっていることから PEG 由来のものであり、理論値 3949.86との誤差は 5.3massと近いことから PEG-ペプチド- DOPEであることが示された。 H-NMRスペクトルを測定したところ、 3.35ppmの PEG末 端メトキシ基由来のピークと、 6.6-7. lppmのペプチドのチロシンのベンゼン環由来の ピークとの積分比は 3 :4.3とモル比がほぼ 1: 1であり、 PEG-ペプチドと比較して大きな 変化はなかった。

(1)方法
(a)MENDの調製
PEG修飾脂質 (PEG- DSPE)及び PEG-ペプチド- DOPE(PPD)を用いて PEGのみで修 飾された MEND (PEG-MEND)、 PEG-ペプチドのみで修飾された MEND (PPD- MEN D)、並びに PEG及び PEG-ペプチドで修飾された MEND (PEG/PPD- MEND)をそれ ぞれ調製した。コンパクション体をルシフェラーゼをコードしたプラスミド DNAとプロタミ ンの N/Pを 1として HEPES buffered glucose (10mM HEPES, pH7.4, 5% glucose;HBG) 中で調製した。脂質膜の脂質組成は 1 mMのエタノール溶液 DOTAP、 DOPE, Choles terolをそれぞれ 3 :4: 3の割合でガラス試験管に添カ卩しこれを 100%とした。 PEG及び P EG-ペプチドの修飾を 1: 1の割合で行うために 1 mMのエタノール溶液として PEG- DS PE及び PPDを全修飾濃度が 5、 15、又は 20 mol%となるように半量ずつ添カ卩した。その 後、クロ口ホルムをエタノール溶液と等量添加し、ボルテックスで攪拌混合した後、デ シケーターで溶媒を留去して脂質膜を調製した。 [3H]標識する場合は脂質膜溶液中 に [3H]- Cholesteryl hexadecyl ether (CHE)を添カ卩した。 MENDはコンパクション体を 脂質膜に添加し、ソ-ケーシヨン処理することにより調製した。調製した MENDの粒子 径 ίま dynamic light scattering (DLS)によつ飞孭 U定し 7こ。
(b)インビトロ実験
HT1080 (ヒト繊維芽肉腫由来)を播種し、 24時間後に 24 well-plate (4 X 104 cells/50 0 ul DMEM/well)の各 wellから DMEMを回収し、細胞などを除くため 1000rpm、 4°Cで 5分間遠心し、上清を Conditioned mediumとして実験に用いた。 0.4 μ g DNA MEND /250 L in conditioned medium/wellで MENDを HEK293 (ヒト腎臓内皮様由来)(4 X 10' cells/well)に添カ卩して 3時間インキュベーション後、 Fresh DMEMと conditioned m ediumを 1: 1で 750 μ L/well追加して 45時間インキュベーションし、ルシフェラーゼ活 性を測定した。
MMP- 2を 14 nMの濃度で含む DMEMで MENDを 0.4 μ g DNA /250 μ Lに調整し、 HEK293細胞(4 X 104 cells/well)に対して 0.4 μ g/wellで MENDを添カ卩し、 3時間後に f resh DMEMを 250 μ 1/wellの割合で添カ卩して 45時間インキュベーションした後、ルシ フェラーゼ活性を測定した。
また、他の細胞系 MG-63 (ヒト骨肉種由来)、 HOS (ヒト骨肉種由来)、 PC-3 (ヒト前立 腺がん由来)におけるトランスフエクシヨン実験は、トランスフエクシヨン実験前日に各 細胞を 24wellプレートに 4 X 104cells/wellで培地 0.5 mL中に播種した。 0.4 g DNA相 当に調製したサンプルを無血清培地の場合には FBS'抗生物質非含有 D'MEM (以 下、「D'MEM( -)」と略記する。)で 250 Lとなるように希釈し、血清含有培地の場合は D'MEMで同様に希釈した。各 wellを PBSで洗い、 250 L/wellの割合でサンプルを添 加し、 5%CO下に 37°Cで 3時間インキュベートした後、 D'MEMを 0.75 mL/well添カロし、

5%CO下に 37°Cで 45時間インキュベートした。 45時間後、各 wellの培地を除去して PB

Sで洗浄した後、 Reporter Lysis Buffer (75 μ L/well)を添カ卩して- 80°Cで凍結した。 30 分後、凍結したプレートを室温で融解させ、氷上にて CELL SCRAPERを用いて細胞 を剥がしとり、 Lysis溶液を回収した後、遠心 (4°C、 15,000rpm、 5分)して上清を回収し た。上清 20 /z Lあたりのルシフェラーゼ活性 (RLU)を測定し、上清を DDWで 5倍希釈 して BCA法によりタンパク定量を行い、 RLU/mg proteinを算出した。
メディウム中の MMP- 2発現量の測定から、 MG- 63 (68.7士 2.85 ng/ml)、 HOS (28. 7士 4.52 ng/ml)を MMP- 2高発現細胞、 PC- 3 (7.09士 3.06 ng/ml)を MMP- 2低発 現細胞とした。メディウム中の MMP- 2発現量に関しては、 Matrix Metalloproteinase- 2 (MMP- 2), Human, Biotrak ELISA System (Amersham Bioscience)を用いて測定した 。サンプルの調製はキットのプロトコルに従って行なった。
[0077] (c)動物実験
血中濃度の経時変化の測定は DDYマウス (雄性: 5週齢)を用いて行なった。腫瘍 移行量の測定及び腫瘍における発現量の測定には、 HT1080細胞(1 X 106 cells)を B ALB/cヌードマウス (雄性: 5週齢)の背部皮下に移植し腫瘍サイズ (長径)が 12-18 m mとなった担癌マウスを用いた。
MENDの動態実験では [3H]標識 MENDを(7.5 μ g pDNA/125 L in HBG、 0.5 m M lipid)を尾静脈より投与し、タイムコースにおける血液や腫瘍組織を回収し、それぞ れ 3Hカウントを測定し、血中濃度および腫瘍移行量を %ID/mlおよび %ID/g tumorとし て算出した。
遺伝子発現実験では MEND (25 μ g pDNA/420 μ L HBG、 0.5 mM lipid)を尾静 脈より投与し、 48時間後に腫瘍組織を採取し重量を測定した。腫瘍組織を Lysis buffe r(0.1% Triton X- 100, 2mM EDTA, 0.1M Tris- HC1, pH7.8)中でホモジュナイズし、 4 。C、 15,000 rpmで 10分間遠心し、上清についてルシフェラーゼ活性を測定し、遺伝 子発現量を RLU/g tumorとして算出した。
[0078] (2)結果
(a)インビトロ実験
•PPD- MENDの HEK293細胞における遺伝子発現活性への MMP-2添カ卩の影響
MMP-2低発現細胞である HEK293において、 5% PPD- MENDの発現活性は 5% PEG -MENDと比較して 4〜5倍高い程度であった(図 1、白色バー)。一方、 Conditioned m ediumを用いた際の 5% PPD- MENDの発現活性は 5% PEG- MENDと比較して 14.3倍で あり(灰色バー)、 MMP- 2を含むメディウムを用いた際は 24.8倍 (黒色バー)の発現活 性を示し、いずれの場合も、通常の場合と比べて優位に高い値を示していた。これら の条件下において、 PEG未修飾群での発現活性には変化が認められな力つた。以 上の結果より、 5% PPD- MENDの発現活性は、 MMPを含むメディウム中で PEGの切断 を受けて上昇したものと考えられる。
•他の細胞系における PPD-MENDの遺伝子発現活性
MG-63、 HOS、及び PC-3細胞において、 5% PPD- MENDの発現活性は 5% PEG-M ENDと比較してそれぞれ 20.9倍、 12.7倍、 5.21倍上昇していた。これらの発現活性の 上昇は MMP-2発現量に依存しており、 PPD- MENDの発現活性上昇は MMPによる PE Gの切断の結果であることが示唆された(図 2)。
[0079] (b)インビボ実験 (1)
•血中濃度推移
PPD- MENDの血中濃度推移は PEG- MENDと比較して劣るものの、 MENDと比較し て有意に高ぐ PPDの修飾によって血中滞留性がある程度得られることが示唆された (図 3(a))。
'腫瘍移行量
血中濃度依存的な腫瘍への移行が認められた (図 3(b))。
•腫瘍遺伝子発現 (c,d)
腫瘍における発現量は MENDおよび 5% PPD- MENDでは見られなかった。また、同 等の腫瘍移行量を示した 5% PEG- MENDと 15% PPD-MENDを比較すると遺伝子発現 活性は 15% PPD- MENDにおいて 55倍上昇していた。この結果は PPD- MENDの腫瘍 到達後に MMPにより PEGが切断され、その結果、発現活性が上昇したものと考えられ る。また 15% PEG- MENDと 15% PPD- MENDは同等の発現活性を示していた(図 3(c)) 。発現量(RLU/g tumor)を移行量 (%ID/g tumor)で除した specific activity of particles を算出すると、 PPD- MENDは PEG- MENDと比較して約 3倍高かった。以上の結果は 、腫瘍到達後に MMPによる PEGが切断され、 PPD- MENDが PEG- MENDと比較して 標的細胞への移行や発現活性が促進されて ヽることを示唆して ヽる(図 3(d))。
[0080] (c)インビボ実験 (2) 以上の結果から、 PPDを用いた PEG-ペプチドによる修飾は血中滞留性及び腫瘍 移行量を上昇させることが明らかになった。 PEG-ペプチドによる修飾を利用してさら に高 ヽ血中滞留性及び腫瘍移行量を達成すべく、 PEG-ペプチドと PEGとを組み合 わせた修飾の効果を判定した。 PEG-ペプチドと PEGとの組み合わせの比率は 1: 1と して試験を行なった。表 2は各濃度における PEG/PPD-MENDの粒子径を示す(単位 は nm

[0081] 血中滞留性は PEG- MENDでは 15%で一番高い値を示し(図 4(a)、白色バー)、腫瘍 移行量も同様の傾向を示した(図 4(b)、白色バー)。一方、 PEG/PPD-MENDでは血 中濃度は Total PEG濃度 20%まで上昇しており(図 4(a)、黒色バー)、腫瘍移行量も同 様の傾向であった(図 4(b)、黒色バー)。 PEG濃度 20%では、 PEG- MENDと PEG/PP D- MENDの腫瘍移行量はほぼ同等であった。 PEG- MEND及び PPD- MENDの腫瘍 における遺伝子発現活性は、腫瘍移行量と同様な上昇'減少傾向を示したが、 20% P EG- MENDと 20% PEG/PPD- MENDにおける発現量には 60倍以上の大きな差が見ら れた(図 4(c))。さらに、発現量を移行量で除した単位移行量あたりの発現活性では 9 0倍以上の上昇が見られた。この結果は、 PEG濃度 20%において PEG : PEG-ペプチド 修飾の比率が 1:1のときに腫瘍への移行性の上昇とペプチド切断による MEND活性 化上昇の組み合わせ効果が相乗的に発揮されたものと考えられる。
産業上の利用可能性
[0082] 本発明の脂質膜構造体は、血中にお!ヽてはポリアルキレングリコールなどの修飾部 位の存在により優れた血中滞留性を示し、一方、マトリックスメタ口プロテア一ゼを分 泌する悪性腫瘍細胞や新生血管の近傍などで、該修飾部位が脱離することにより薬 物などを放出することができ、あるいは標的細胞により取り込まれ易くなることにより、 該細胞に対して薬物などによる効果を発揮させることができる。

請求の範囲 少なくとも下記の 2種類のリン脂質誘導体:
(1)アルコールィ匕合物の残基とリン脂質類の残基とを含み、該アルコールィ匕合物の残 基とリン脂質の残基との間にペプチドを含むリン脂質誘導体であって、
(a)該アルコール化合物が、ポリアルキレングリコール類、グリセリン類、及びポリグリセ リン類カゝらなる群カゝら選ばれるアルコールィ匕合物であり、
(b)該リン脂質力 ホスファチジルエタノールアミン類、ホスファリジルコリン類、ホスファ チジルセリン類、ホスファチジルイノシトール類、ホスファチジルグリセロール類、カル ジォリピン類、スフインゴミエリン類、セラミドホスホリルエタノールアミン類、セラミドホス ホリルグリセロール類、セラミドホスホリルグリセロールホスファート類、 1,2-ジミリストイ ル- 1,2-デォキシホスファチジルコリン類、プラスマロゲン類、及びホスファチジン酸類 力 なる群力 選ばれるリン脂質であり、及び

を構成脂質として含む脂質膜構造体。 [2] マトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び Z又は遺伝子治療の ための遺伝子を保持した請求項 1に記載の脂質膜構造体。
[3] リボソームである請求項 1又は 2に記載の脂質膜構造体。
[4] リン脂質誘導体 (1)及び (2)において、該アルコールィ匕合物がポリアルキレングリコー ル類であり、該リン脂質がホスファチジルエタノールアミン類であり、リン脂質誘導体 (1 )にお!/、て、該ペプチドが Va卜 Pro- Leu- Ser- Leu- Tyr- Ser- Glyを含むペプチドである 請求項 1な!、し 3の 、ずれか 1項に記載の脂質膜構造体。
[5] リン脂質誘導体 (1)及び (2)において、該アルコール化合物がポリエチレングリコール であり、該リン脂質がジォレイルホスファチジルエタノールァミンまたはジステアロイル ホスファチジルエタノールァミンであり、リン脂質誘導体 (1)において、該ペプチドが G1 y-Gly-Glyをリンカ一として含む請求項 1な 、し 3の 、ずれか 1項に記載の脂質膜構 造体。
[6] リン脂質誘導体 (1)において、該ペプチドが
uly-uly-uly-Val-Pro-Leu-ber-Leu-Tyr-Ser-Gly-Gly-Gly-Gly
である請求項 1な 、し 3の 、ずれか 1項に記載の脂質膜構造体。
[7] リン脂質誘導体 (1)とリン脂質誘導体 (2)との比がモル比として、 10:1〜1:10である請求 項 1な 、し 6の 、ずれか 1項に記載の脂質膜構造体。
[8] リン脂質誘導体 (1)及びリン脂質誘導体 (2)の脂質膜構造体の全脂質量に対する割合 力^〜 40モル %である請求項 1な 、し 7の 、ずれか 1項に記載の脂質膜構造体。
[9] マトリックスメタ口プロテアーゼの関わる疾患に有効な薬物及び Z又は遺伝子治療の ための遺伝子を保持した請求項 2な 、し 8の 、ずれか 1項に記載の脂質膜構造体を 含む医薬組成物。
