[0003] 正常組織の刺激を受けて 、な 、抗原提示細胞は、自己抗原を提示して!/、たとして も副刺激分子の発現が制御されているため、 T細胞はァナジ一に陥っており活性ィ匕 されず自己寛容が維持されている。しかし、免疫異常状態においては過剰あるいは 継続する副刺激分子の発現異常により自己反応性 T細胞の活性ィヒが生じた結果、 自己免疫疾患が生じるのではないかという可能性が示唆されている。特に CD28/CD 152と CD80/CD86間で生じるシグナルは T細胞活性化のコントロールに重要かつュ ニークな働きをしており、このシグナルを調節することによる免疫治療がマウス実験モ デルにぉ 、て試行され、実際ヒトに対する治験も行なわれ始めて!/、る。
[0004] このような抗原ペプチドを認識する機構を基本とする所謂獲得免疫の機構が力なり 解明されているのに対して、外来の糖脂質や蛋白質、 DNA、 RNAを認識する機構を 基本とするいわゆる自然免疫は現在分子レベルで明らかにされつつある段階にある 。また自然免疫から獲得免疫を繋ぐ分子機構にっ 、ても不明な点が多!ヽ。
[0005] 現在の哺乳類における自然免疫に対する理解はハエや植物の発生学'遺伝学に
ぉ ヽて解析された感染防御機構に始まる。すなわちショウジヨウバエの病原体認識分 子 Tollの哺乳類のホモログ TolHike Receptor (TLR)が発見され、自然免疫における 病原体認識を司ることが明らかとなった(Science、 284卷、 1313-1318頁(1999年))。 この事実は自然免疫がハエ力もヒトまで共通する病原体認識機構であるという概念が 成立することを意味する。獲得免疫が個体レベルで完結するシステムである(抗体や T細胞レセプターの特異性は個々に遺伝子を再構成することで獲得され、子孫に伝 達されない)のに対して、自然免疫はゲノムに共有している進化の記録である。つまり 獲得免疫の多様な特異性の中で、どの特異性に対して反応すればよいのかという方 向付けをするのが自然免疫であり、最も重要な分子の一群が TLRであると考えられる 。自然免疫における TLRの役割はアジュヴアントの認識およびシグナル伝達による副 刺激分子の発現やサイト力イン産生である。獲得免疫において対象となるタンパク質 自身は免疫原性が低 、のに対し、病原体由来糖脂質 (リポポリサッカライド (LPS)な ど)は免疫系を強く活性化する。例えば、抗体作製時に抗原のみでなくアジュヴアント が必要なのはよい例である。つまり獲得免疫発動の制御は TLRによる認識が入口に なって 、ると!/、つても過言ではな!/、。
[0006] 獲得免疫発動の決定,制御の主役は榭状細胞 (DC)であり、 Thl/Th2 skewingと細 胞傷害性 T細胞 (CTL)や活性化マクロファージの誘導 (細胞性免疫)、抗体産生 (体 液性免疫)に関わる。この誘導される免疫反応は病原体の種類によって異なる。細菌 感染の場合には細胞性免疫優位であるが、ある種の寄生虫やウィルスの場合には体 液性免疫優位の場合もある。またアジュヴアントの場合にも水酸ィ匕アルミニウム (Alum )のように抗体産生優位になったりする。これには TLRによる認識と DCによる獲得免 疫の質的ある 、は量的な変化が制御されて 、る可能性が考えられる。
[0007] TLRは細胞外に LRRと呼ばれる蛋白質相互作用に関係する leucine_rich repeatを 有する I型膜タンパク質で、細胞内には TIR domain (Toll/IL- 1R homology domain)と 呼ばれるシグナル伝達に関係するドメインを有する(Trends Cell Biol.、 11卷、 304-31 1頁(2001年))。これまでにヒトにおいては 10種類の TLR分子が報告されており(TLR1 -TLR10)、同時にリガンドの同定も進んでいる。例えば TLR2は細菌由来ペプチドダリ カン(PG)ゃリポ蛋白質を(J. Biol. Chem.、 274卷、 17406-17409頁(1999年);及び J.
Immunol.、 163卷、 2382-2386頁(1999年))、 TLR4はグラム陰性細菌の LPSやグラム 陽性細菌のリポティコ酸(LTA)を(Science, 282卷、 2085-2088頁(1998年)) TLR5は 細菌鞭毛構成タンパク質を(Nature、 410卷、 1099- 1103頁(2001年))、 TLR2/6はマ ィコプラズマ由来リポペプチドや酵母由来ザィモザンを(Int. Immunol, 13卷、 933-94 0頁(2001年))、 TLR7と TLR8は細菌由来一本鎖 RNAを(Science, 303卷、 1529- 31頁 (2004年) )、 TLR9は細菌由来 CpG DNAを(Nature、 408卷、 740-745頁(2000年) )、 それぞれ認識することが報告されて 、る。
[0008] TLRからのシグナル伝達につ!、てはマクロファージあるいは榭状細胞にぉ 、て TLR 2 (MALP- 2刺激など)、 TLR4 (LPS刺激など)、 TLR9 (CpG DNA刺激など)を中心に 解析されてきている。まず TIR domainと会合する分子として IL-1R familyにおけるシグ ナル伝達において必須のアダプター分子である MyD88が TLR4のシグナル伝達にお いても必須の分子であることが明ら力となり、 LPSにより誘導されるマクロファージから のサイト力イン産生、 B細胞ブラスト化、ショック症状の惹起等が全く欠如していた (Im munity、 11卷、 115- 122頁(1999年))。し力し、 MyD88欠損マウス由来マクロファージ にお!/、て LPS刺激に対して NF-kappaBの活性化が見られることから、 MyD88に依存し な 、NFkappaB経路の存在が示唆された。この経路に対する説明として MyD88欠損 マウスにおける DCの解析結果がひとつの答えである。すなわち CDllc陽性の DCに おける LPS刺激による IL-12産生は MyD88欠損マウスにおいては全く見られなくなる 力 CD40, CD80, CD86といった副刺激分子の発現上昇に関しては影響を受けない (J. Immunol, 166卷、 5688-5694頁(2001年))。 TLR4新規アダプター分子として報告 された TIRAP/MALは TLR4による DCの成熟化を阻害することから、 DCにおける畐师 IJ 激分子発現に必要な経路に関与する可能性が示唆される (Nat. Immunol.、 2卷、 835 -841頁(2001年);及び Nature、 413卷、 78-83頁(2001年))。またこの報告とは別に M yD88欠損マウス由来マクロファージおよび DCにおいて IP-10や ISREといった IFN誘導 遺伝子群の発現に必要な IRF3が活性化することが明らかとなった (J. Immunol, 167 卷、 5887- 5894頁(2001年))。
[0009] 一方 TLR2および TLR9のシグナル伝達に関しては MyD88欠損マウスにおける解析 結果ではサイト力イン産生、 NF-kappaB活性化ともに MyD88は必須であった (J. Immu
nol.、 164卷、 554-557頁(2000年))。これとは別に PI3K familyに属し、 SCIDマウスの 原因遺伝子としても知られて 、る DNA-PKcs欠損マウスが CpG DNAに不応答性であ るとの報告もある(Cell、 103卷、 909-918頁(2000年))。 TLR9下流に DNA-PKcsが存 在する、あるいは細胞の活性化には TLR9、 DNA-PKcsの両者が必要である可能性 が示唆される。シグナル伝達に関しては、 TLR欠損マウス及び MyD88欠損マウスを用 V、た解析が中心である。ヒトにつ 、ては MoDCにお!/、て TLR2および TLR4刺激により 異なるサイト力イン、ケモカインが誘導されるという報告もあり(J. Biol. Chem.、 276卷、 37692-37699頁(2001年))、ヒト DCにおける TLR発現特異性を考えると、ヒトでの解析 結果に興味が持たれる。
[0010] 上記のように TLRからのシグナルが自然免疫の発動に重要であることは言を待たな いが、 TLRの自然免疫シグナルとは別に自然免疫の発動を受けて獲得免疫発動に 必要な分子の存在があることが予想される。自然免疫反応を受けて獲得免疫反応を より高める分子として TREMdriggering Receptor Expressed by Myeloid Cells)- 1が知 られている(J. Immunol.、 164卷、 4991-4995頁(2000年))。 TREM-1は CD14強陽性 の単球及び好中球に発現する細胞外に 1つのィムノグロブリンドメインを有する I型細 胞膜蛋白質で、アダプター分子 DAP12と会合してシグナルを伝えることが知られてい る。細菌や真菌あるいはその構成成分により刺激を受けた CD14強陽性の単球及び 好中球において TREM-1の発現は増強される。好中球に抗 TREM-1ァゴ-スティック 抗体を作用させると大量の IL-8やミエ口ペルォキシダーゼが産生され、単球からは IL -lbetaや TNF-alphaあるいは MCP-1が産生されることが知られている。同時に ICAM- 1、 CDllb、 CD49e、 CD29といった接着分子や CD40、 CD86といった副刺激分子の発 現も増強される。また、これらの炎症性サイトカインゃケモカインの発現は LPS刺激と 協調性をもって増強され、敗血症ショックを引き起こす。敗血症ショックのモデルにお いては TREM-1が重要な機能を果たしていることが明らかにされている(Nature、 410 卷、 1103- 1107頁(2001年))。
[0011] 榭状細胞 (DC)は、生体内では骨髄に存在する CD34陽性細胞を前駆細胞として 分化 '成熟し、抗原提示細胞 (APC)として、免疫応答の誘導、維持、拡大、調節にお いて重要な役割を担っていることが知られている。プロフェッショナル APCである DC
は、自然免疫反応による刺激を受けて活性化し、抗原特異的獲得免疫反応を惹起 するばかりでなぐ自ら様々なサイト力インを産生することによって感染時の自然免疫 反応においても重要な役割を果たす。 DCにおいては各 TLRの発現は、 MoDC、末梢 血中の myeloid DC (MDC)、 plasmacytoid DC (PDC)間で明らかに異なり、リガンドを 作用させた場合に産生されるサイトカインゃケモカインレセプターの発現も異なること が明ら力となっている(J. Exp. Med., 194卷、 863-869頁(2001年);及び Eur. J. Immu nol.、 31卷、 3388- 3393頁(2001年))。また PDCにおいて CpG+CD40Lで大量の IL- 12 p70を産生するという報告もあり(Eur. J. Immunol.、 31卷、 3026-3037頁(2001年))、 実際の生体内において、どの DCサブセットがどのような自然免疫反応を受けて、 auto crine/paracrineに作用するサイト力インの影響でどのような phenotypeを有する DCへと 成熟化していくのか、興味力もたれるところである。
[0012] 1990年代に入って、 DCを前駆細胞力もサイト力インで分化'誘導できるようになり、 大量の DCを扱えるようになって分子 ·細胞 ·生体レベルにぉ 、て DCの免疫応答にお ける役割の重要性が明らかになってくると共に、免疫制御の標的として注目されるよう になった。
[0013] これまでの生物学的研究成果から、ヒト単球 (Monocyte ; Moともいう)を GM-CSF及び IL- 4存在下で培養することにより DC (Monocyte- derived DC ;MoDCともいう)を誘導 することが可能であることが明ら力となっている(J. Leucocyte Biol.、 59卷、 208-218頁 (1996年))。この in vitro分ィ匕誘導系は人工的な部分があるものの、実際に DCとして の機能を有することが明らかとなってきた。 DCの重要な機能は、自然免疫反応を受 けるとともに抗原の細胞内への取り込み作用(食作用)及びその抗原の情報を T細胞 に伝えて T細胞を刺激活性ィ匕することである。また、 DCは分ィ匕の段階によって未成熟 DC (Immature DC)と成熟 DC (Mature DC)の二つに分別することができる。未成熟 D C内に取りこまれた抗原は細胞内でプロセシングを受け、抗原由来のペプチドとして DC表面の MHCクラス II分子上に提示される。 CD4陽性の抗原特異的ヘルパー T細 胞は、抗原受容体で抗原由来のペプチドと MHCクラス II分子の複合体を認識し、副 刺激分子からの刺激なども加わり、感作、活性化される。また、 DCにより MHCクラス I 分子を介して CD8陽性の細胞傷害性 T細胞 (CTL)も刺激、活性化される。食作用は
未成熟 MoDCで強ぐ成熟 MoDCでは弱くなる。 T細胞への抗原提示能は、それに関 与する CD40、 CD80、 CD86、 MHCクラス I分子、 MHCクラス II分子の発現の程度と一 致して、未成熟 MoDCでは弱ぐ成熟 MoDCになると強くなる。マウスに関しては骨髄 細胞を GM-CSF存在下培養することによりミエロイド系の榭状細胞を (J. Immunol.、 16 1卷、 3096- 3102頁(1998年))、 Fit- 3リガンド存在下培養することによりミエロイド系の 榭状細胞およびプラズマサイトイド榭状細胞を (J. Exp. Med., 195卷、 953-958頁(20 02年))誘導することが可能であることが明らかとなっている。ミエロイド系の榭状細胞 に関しては、上記ヒト MoDCとほぼ相同の機能を有することが知られている。
[0014] この DCにおける抗原取込能から抗原提示能という機能変化は、前述の自然免疫の 反応を受けて獲得免疫を発動する部分を担って 、ると言っても過言ではな 、。この 機能変化に伴って細胞表面上に提示される膜分子の変化が起こることは想像に難く ないが、この成熟化(maturation)の過程でどのような事象が起こっているかを考えた 場合に、細胞膜表面上に新たに発現した分子の発現はすべて mRNAの発現から起 こるとは限らな 、と 、う事実が存在する。例えば未熟な (Immature)段階から発現が見 られる HLA-DRが成熟化に伴って mRNA及びタンパク質の発現量の変化をほとんど 伴わずに細胞内力 細胞膜表面上に移動 (translocation)するということが知られて いる。また、実際の mRNAの発現量とタンパク質の発現量を比較した場合にほとんど 相関がな 、場合や、 mRNAは発現して 、るのにタンパク質として翻訳されて ヽな ヽ場 合も存在する(Biochem. Biophys. Res. Commun.ゝ 231卷、 1-6頁(1997年))。これま で遺伝子側からは増幅可能なこと、大量に解析することが可能なこと、ハンドリングが 容易なこと、数々の手法が編み出せることなど力 精力的に解析がなされてきたが、 実際に発現しているタンパク質の変化を追うことができれば、そのような変化は細胞 内の事象をよりよく反映しており現実に近 、ものと!/、うことができると!、うのも事実であ る。
[0015] DCに関しては、上述の知見に加えて、 DCにいくつかのサブセットが存在することも 明らかになっているが、そのようなサブセットは成熟、未成熟で機能が異なることが知 られている。しかしサブセット間でのそのような機能の違いが何に起因するの力、その 答えは得られていない。少なくとも細胞内シグナリングに関与すると考えられる膜分
子、特に DCの成熟化に伴って発現する膜分子、の解明が待たれるところである。
[0016] 前述のように、生体内に存在する強力な APCである DCは活性ィ匕刺激に伴い成熟 化すると、 B7ファミリーに属する CD80/CD86を高発現することが知られている。 自己 免疫疾患との関わりについては、例えば慢性関節リウマチの関節液や乾癬の病巣部 組織内、アレルギー性接触性皮膚炎の皮膚組織に存在する DCは CD80/CD86が異 常発現になって ヽることが知られて!/ヽる。
[0017] CD28は、ナイーブ T細胞の活性ィ匕を強力に増強させる特徴を有する。この CD28に よるシグナルの存在により IL-2の産生及び IL-2レセプターの発現が増強され、増殖 反応が亢進し、結果的に T細胞の様々なエフェクター機能が増強されることになる。 C D28シグナルは IL- 2だけでなく IL- 4, IL- 5, IL- 13, IFN- gamma, TNF- alpha, GM-CSF など様々なサイト力イン産生も増強させる。また CD40リガンドのような T細胞活性ィ匕抗 原や IL-8, RANTESなどのケモカイン発現にも関与している。 CD80あるいは CD86の CD28への結合が CD4陽性 T細胞の Thlあるいは Th2への分ィ匕誘導に関与して 、るこ とを示す多くの報告がある。し力し、 CD80は Thl、 CD86は Th2といったような一面的な ものではなぐナイーブ CD4陽性 T細胞の分化方向の決定は、 CD28および CD152シ グナル以外にも抗原あるいは APCそのものから規定されてくる様々な因子によって影 響を受けると考えられる。し力しながら、 CD28シグナルが特にナイーブ T細胞の Th2 分化'活性ィ匕において他の分子の機能によって代償されない重要な役割を担ってい ることが、(1) CD28ノックアウトマウスにおける Th2サイト力イン依存性の免疫グロブリ ンである IgGl, IgG2bの選択的産生障害(Science, 261卷、 609-612頁(1993年))や( 2) D011- 10TCRトランスジエニックマウス CD4陽性 T細胞と CD80/CD86ダブルノック アウトマウス由来 APCによる抗原刺激による Th2分ィ匕の選択的阻害 (J. Immunol.、 161 卷、 2762-2771頁(1998年))などの結果から裏付けられている。また、 CD28シグナル のもうひとつの重要な役割として、 be卜 xLのようなサバイバル因子の発現を増強させ、 抗原刺激を受けた後の T細胞のアポトーシスを抑制することが挙げられる。
[0018] CD28が恒常的に T細胞に発現しているにもかかわらず、糸且織細胞において CD80/ CD86の発現が制御されているために、たとえ自己抗原に反応する T細胞が存在して も過剰な免疫反応が起こらな 、ような仕糸且みになって 、る。このトレランス誘導システ
ムが破綻した場合には、自己免疫疾患やアレルギー疾患等の様々な疾患が引き起 こされる可能性を秘めている。脾島 beta細胞に CD80あるいは CD86を強制発現させ ただけでは脾臓への炎症性リンパ球浸潤は認められるが、自己免疫性糖尿病である インスリン依存性糖尿病 (IDDM)の発症には至らず、主要組織適合抗原 (MHC)クラス I I分子、 TNF-alpha、あるいは自己抗原としてのウィルスタンパク質を CD80とともに発 現させてはじめて beta細胞の破綻を伴う IDDMを発症させることができる。このことは C D28-CD80/CD86シグナルに加え、 T細胞レセプター (TCR)シグナルの強化を促すよ うな環境下に置かれてはじめて自己免疫疾患が発症する可能性が示唆されている。
[0019] いくつかのヒト臓器特異的自己免疫疾患において CD80/CD86の発現がプロフェツ ショナル APCである DCにおける発現が報告されて!、る。このように正常組織の APCに お!、ては制御下にある CD80/CD86が、疾患局所にお!、ては異常発現して 、る例が 数多く見つかつてきており、自己免疫疾患との関連が示唆されている。
[0020] 現在、自己免疫疾患の治療は副作用の多い非特異的な免疫抑制剤が中心である 。副刺激 (costimulation)阻害では、抗原特異的な抑制及び投与終了後における持 続効果があり、投与期間短縮による副作用の軽減や感染症防御免疫反応の維持が 期待できる。免疫系細胞の相互作用や機能が明らかになっていくのにしたがってより 効果的な治療が行なわれることが期待される。活性化 T細胞に発現がみられる CD28 ファミリーの分子として ICOS (indudble T cell co- stimulator)や PD-1が見つかつてき ている。これらはそれぞれ B7ファミリーに属する B7-H2、 B7-H1がそのリガンドであるこ とが明ら力となり、複数の B7ファミリーと CD28ファミリーを介した APCによる T細胞 costi mulationの作用の差異の解明が待たれるところである。
[0021] また、感染症等に対する生体防御の観点から、自然免疫と獲得免疫を繋ぐ分子メ 力-ズムの解明が待たれるところである。すなわち、 LPSや CpGなどは IL-12の大量産 生や CD40ある!/、は CD80/CD86の発現増強を伴って、 DCの maturationを誘導するが 、これに関わる詳細な分子メカニズムは明らかにされて ヽな 、。
発明の開示
発明が解決しょうとする課題
[0022] 本発明者らは、 DCの機能発現に重要となる DCの成熟化に着目し、 DCの成熟によ
つて発現量が上昇する膜分子の同定を中心とした探索研究を、プロテオミクス (Prote omics)による DCの成熟に伴い発現が調節される膜分子の網羅的同定を基に実施し てきた。プロテオミタスとは、タンパク質について、遺伝子のゲノミタス(Genomics)に対 応した言葉であり、タンパク質の大規模研究を行うものである。タンパク質の発現量、 翻訳後の修飾や相互作用などの性質を研究し、例えば正常細胞とがん細胞にぉ 、 て発現タンパク質レベルで何が起こって 、る力、細胞内ネットワークやプロセスなどの 全体的な生物学的情報を得るものである。 DCに特異的に存在する膜分子が同定さ れるならば、該膜分子に対する抗体の作製や、医療分野での該抗体の利用が可能と なるだろう。
[0023] 本発明の目的は、強力な APCである成熟 DC上に発現している本分子を標的として DCの機能制御による感染症防御、自己免疫疾患等の制御である。また、該分子は 成熟 DCなどの APCに発現し、 T細胞活性ィ匕に関わる分子と予想されることから、該分 子を特異的に認識する抗体を使用して成熟 DC等の APCを分離する方法、および検 出する方法を提供することである。
課題を解決するための手段
[0024] 本発明者らは、 DCの成熟化により発現が調節される膜分子を探索研究した結果、 未成熟 DCと成熟 DCとの間で、発現がみられる新規膜分子を同定し、この分子が DC の成熟化に伴 、、そのタンパク質レベルでの発現が顕著に上昇することを今回見出 した。この膜分子は従来知られているマクロファージゃ好中球で発現の見られる TRE M-1にホモロジ一を有して!/、ることが明らかとなつた。また本膜分子は TREMファミリ一 に特徴的な 1つのィムノグロブリン(Ig)ドメインを有することが推定されており、立体構 造の保持に必須の S-S結合に関わる Cys残基の位置が保存されて 、た。 これらの事 実力ゝら本膜分子群は自然免疫と獲得免疫を繋ぐ主役たる DCにおいて発現の見られ る TREMファミリーに属する新規膜分子と考えられる。
[0025] 最近の研究成果から(1) DCは不均一な細胞集団でありいくつかのサブセットが存 在すること [Stem Cells, 15卷、 409- 419頁(1997年)]、 (2) DCサブセットは T細胞活 性ィ匕において異なる機能 (アポトーシス誘導、 T細胞の Thl, Th2への分ィ匕誘導など) を持つこと [Science、 283卷、 1183-1186頁(1999年)]、(3) DCを介して免疫応答を制
御することが可能であること (DC療法、 DC特異的な薬剤、抗体、サイト力インなど)、 が明らかになつてきた。
[0026] ヒト DCサブセットとしては大別して、ミエロイド系 DCとリンパ球系 DCとがある。ミエ口 イド系 DCには 2つの分化経路が存在することが示されており [J. Exp. Med., 184卷、 6 95-706頁(1996年) ]、 CD34陽性の造血幹細胞を GM- CSFと TNF-alphaで培養する 系では CD14陽性 CDla陰性および CD14陰性 CDla陽性の前駆細胞が出現し、前者 からは真皮(dermal) DC、後者からは表皮ランゲルノヽンス細胞に分ィ匕することが知ら れている。本発明者らが用いた MoDCは生体内では前者由来の DCと類似しており、 ミエロイド系 DCに属すると考えられている。一方、ヒトリンパ球系 DCは、形質細胞様の CD4陽性 T細胞が IL-3存在下で未熟な DC (CDllc陰性、 CD14陰性)へと分ィ匕し、 IL -3と CD40リガンドの刺激により DCとして機能的に成熟することが知られている [J. Exp . Med., 185卷、 1101- 1111頁(1997年)]。また、 MoDCは CD40リガンドあるいはエンド トキシンによる刺激により大量の IL-12を産生し、ナイーブ T細胞を Thlに分化'誘導 する機能を有する DCに分化するという報告がある [Science、 283卷、 1183-1186頁(19 99年)]。このように本発明者らが用いた MoDCは少なくともナイーブ T細胞を Thlタイ プに分ィ匕し得る一群であることがわかる。また、末梢血には少なくとも 2つの DCサブ セットが存在しており、系統マーカー(CD3、 CD19、 CD56、 CD14)陰性、 HLA-DR陽 性、 CDllc陽性と系統マーカー(CD3、 CD19、 CD56、 CD14)陰性、 HLA- DR陽性、 C Dllc陰性、 CD123強陽性の表現型を示す。それら DCは成熟後、前者は大量の IL-1 2を産生する MDC、ナイーブ T細胞を Thlに、後者は大量の type I IFNを産生する PD し/ NIPC (natural type I interferon producing cell)であること力 S知りれて ヽる [science 、 284卷、 1835-1837頁(1999年)]。
[0027] このように、本膜分子は MoDCの成熟化に伴 、発現が顕著に上昇して 、ることなど の知見から、免疫応答を制御するための標的分子としても期待される。

[0038] 本発明のヒトおよびマウス榭状細胞膜分子は、マウス骨髄細胞を GM-CSF存在下 培養することにより誘導したマウス骨髄由来未成熟榭状細胞 (DC)をリポポリサッカラ イドで刺激し、その可否に基いて成熟 DCを得たのち、細胞膜を調製し、その可溶性 タンパク質についてコンカナパリン Aセファロースクロマトグラフィー、小麦胚ァグルチ ニンセファロースクロマトグラフィー、 SDS-ポリアクリルアミドゲル電気泳動などの手法 を用いて膜タンパク質を分画し、 LC/MS (特に Applied Biosystems社製 QTRAP)による 微量分析にかけて同定された (後述の実施例 1〜5参照)。さらに LC/MS法で同定さ れた複数の部分アミノ酸配列に基づくプライマーを合成し、マウス成熟 DC由来 cDNA ライブラリーを铸型にしてポリメラーゼ連鎖反応 (PCR)を行 ヽ本発明の膜分子の遺伝 子断片を増幅し取得し、この遺伝子断片をプローブとしてコロニーハイブリダィゼー シヨンにより本発明の膜分子 (A)の遺伝子を含むクローンを選抜し、その塩基配列( 配列番号 1)およびアミノ酸配列 (配列番号 2)を決定した (後述の実施例 6参照)。

利用することによって該アミノ酸配列力もなるペプチドを合成することができる。目的 のペプチドは、例えば、 R.B. Merrifield [Science, 232卷、 341- 347頁(1986年)]によつ て開発された固相ペプチド合成に基いた市販のペプチド合成機を使用して合成し、 保護基を脱離後、イオン交換クロマトグラフィー、ゲル濾過クロマトグラフィー、逆相ク 口マトグラフィ一等を単独もしくは組合わせた方法により精製する。得られた精製ぺプ チドはキーホールリンペットへモシァ-ン(KLH)やアルブミンなどのキヤリャタンパク 質と結合し免疫原として使用することができる。
[0080] さらに、遺伝子組換え本膜分子を免疫原として本膜分子に対するポリクローナル抗 体あるいはモノクローナル抗体を公知の手法により作製することもできる。この場合、 本膜分子、モノクローナル抗体、ポリクローナル抗体、または他のタンパク質に関して 用いられる「組換え」という用語は、これらのタンパク質が宿主細胞内で組換え DNAに よって生産されたものであることを意味する。宿主細胞としては、原核生物(例えば大 腸菌のような細菌)及び真核生物(例えば酵母、 CHO細胞、昆虫細胞等)のいずれも 使用され得る。
[0081] 本発明の「抗体」はペプチド抗体、ポリクローナル抗体、モノクローナル抗体!/、ずれ でもよい。「抗体」は、マウスまたは他の適した宿主動物を免疫に用いられたタンパク 質に特異的に結合するであろう抗体を産生するか、産生するであろうリンパ球を引き 出すために、皮下、腹腔内、または筋肉内の経路によって、抗原あるいは抗原発現 細胞により免疫化することによって得られる。さらに宿主動物としてはヒト抗体遺伝子 のレパートリーを有するトランスジエニック動物に抗原または抗原発現細胞を投与し、 所望のヒト抗体を取得してもよい [Proc. Natl. Acad. Sci. USA, 97卷、 722-727頁(200 0年)、国際公開 W096/33735、 WO97/07671, W097/13852, W098/37757参照]。 そのかわりに、リンパ球を in vitroで免疫化してもよい。宿主動物から得られた血清か ら、抗原に結合する画分を集め、精製することにより、ポリクローナル抗体を取得する ことができる。また、ハイプリドーマ細胞を形成するために、ポリエチレングリコールの ような適当な融合試薬を用いて、リンパ球を骨髄腫細胞と融合させることにより、モノ クローナノレ f几体を作製することができる (Goding, Monoclonal Antibodies: Principals a nd Practiceゝ 59- 103頁、 Academic pressゝ 1986年)。例えば本発明のモノクローナル
抗体は、ハイプリドーマ法 [Nature、 256卷、 495頁(1975年)]を用いても、組換え DNA 法 (Cabillyら、米国特許第 4816567号)を用いても作製することができる。

[0086] このようにして得られた本膜分子に対する各種抗体は、その特徴を生かすことがで きる様々な用途に使用可能である。本膜分子が成熟 DCに特異的に発現が見られる 分子であることを利用して、本抗体を蛍光性物質 (ローダミン、フルォレサミンなど)で 標識し、周知の FACSを用いて目的の DCを検出'分離したり、 mo-DCの in vitroでの 分ィ匕を確認することができる。さらに、本膜分子は DCの成熟化に伴って発現量が顕 著に上昇するタンパク質であることから、本抗体を用いて FACSにより未成熟 DCと成 熟 DCを分離することもできる。本膜分子の検出に関しては、 FACSでの検出にとどま るものではな!/、。例えばウェスタンブロッテイングにぉ ヽて本抗体を 1次抗体として作 用させることにより検出可能であることが予想されるし、タンパク質レベルでの発現確 認を行うことができる。また本抗体を固相 (ポリスチレンビーズ、マイクロタイターゥエル 表面、ラテックスビーズなど)に結合して不均一系で、あるいは均一系で、免疫学的反
応を行って相同な膜分子を検出、定量 (蛍光抗体法、 ELISA、ラジオィムノアツセィな どの方法使用)することができる。この場合、免疫学的反応は競合反応であってもよ V、し非競合反応であってもよ!/、。また 2つ以上の抗体 (モノクローンまたはポリクローン) を用いるサンドイッチ法による反応も使用できる。上記における検出、定量のために は、当業界で公知の 、ずれの免疫学的手法も用いることができる。
[0087] その他、本膜分子の機能を評価する用途にも使用可能である。成熟 DCは強力な A PCであり、 MHCクラス II分子を介した CD4陽性の T細胞を刺激活性ィ匕及び MHCクラス I分子を介した CD8陽性細胞障害性 T細胞を刺激活性ィ匕することが知られて ヽる。こ れら機能を制御し得るか否かを確認するために、ァロジェニック MLR (Mixed Lymphoc yte Reaction)での機能を抑制しうるカゝ否かの確認、抗原特異的に CTLを誘導した場 合にその機能を抑制しうる力否かの確認、さらに DCの抗原提示に関わる分子力否か の確認等の in vitroでのアツセィにも使用可能である。
[0088] 本発明の抗体はさらに、 in vivoで免疫応答を制御するために使用することもできる 。本膜分子群は、前述したとおり、副刺激に関与する分子であると予想され、 T細胞 活性ィ匕に関わる膜分子であると予想される。このため、本発明の抗体が本膜分子群 と T細胞上のリガンドとの結合を阻害する抗体であれば T細胞活性ィヒを抑制すること が期待される。このように本膜分子群の機能制御可能な抗体を作用させることで DC の機能さらには T細胞の機能を制御することによる免疫応答を制御し得ることが期待 される。
[0089] さらには本膜分子および本膜分子のリガンドの可溶ィ匕型分子 (すなわち、細胞外領 域に相当する分子)およびこれら分子に対する抗体にも免疫応答を制御する活性が 期待される。
[0090] 本発明の抗体類または本膜分子の可溶ィ匕型分子を用いて本膜分子のリガンドを取 得することも可能である。可溶ィ匕型本膜分子リガンドは直接本膜分子に作用し、 DC 上の本膜分子を介したシグナルを制御し得る。また、本膜分子のリガンドと本膜分子 の相互作用をモジュレートできる低分子物質や本膜分子のリガンド及び本膜分子に 関わる細胞内シグナル経路をモジュレートする低分子物質もシグナルの制御に有用 である。
[0091] 本発明の抗体類または本膜分子の可溶ィ匕型分子、本膜分子のリガンドの可溶ィ匕型 分子、上記低分子を治療に用いる場合には、例えば癌、自己免疫疾患、臓器移植、 感染症、アレルギーなどの疾患の治療に適用可能である。
[0092] 投与法および投与剤型は特に制限されないが、静脈、動脈内投与、筋内投与、経 口投与、座剤投与などであり、薬学的に許容可能な賦形剤や希釈剤と組合わせて経 口用または非経口用に処方することができる力 好ましくは非経口投与である。投与 は 1日あたり 1回力または数回に分けて行い、投与量は患者の重篤度、年齢、性別、 体重などの条件に応じて決定され、副作用を併発しない範囲である。
以下に本発明の実施例を記載するが、本発明はこれらの実施例によって限定され ないものとする。
実施例
[0093] ¾細 DCの細qflfの調製
細胞膜タンパク質の場合、元々発現量が低いこと、またハンドリングが難しいことが 挙げられる。これを解決する手段の一つとして、より純度の高い細胞膜を取得する方 法を確立することが必要である。細胞膜表面をコートし、これを均一に破砕することで 、比重の重くなつた細胞膜を密度勾配遠心により取得する方法で純度の高い細胞膜 を調製した [J. Biol. Chem.、 258卷、 10062-10072頁(1983年)]。
[0094] ターゲットとしている膜分子の発現量の少なさとハンドリングの難しさから、大量のマ ウス DCを集めるために骨髄細胞を GM-CSF存在下培養することで骨髄細胞由来未 成熟 DCを大量調製した。 LPS (リポポリサッカライド)刺激あり、なしを同数の細胞で実 施し、同数 (5 X 108細胞)の未成熟 DCと成熟 DCを得た。
[0095] 実施例 2 タンパク質の高感度検出と in gel digestion
Mannら [Anal. Chem.、 68卷、 850-858頁(1996年)]の手法に従って銀染色によりタ ンパク質を検出した。酵素消化法についてはトリプシンを用いた in gel digestion法 [A nal. Chem.、 224卷、 451- 455頁(1995年)]により行い、以下で使用する分析サンプル とした。
[0096] ¾細13 蹬タンパク皙の分画
実施例 1で得られた細胞膜より界面活性剤を用いて細胞膜タンパク質を可溶ィ匕した
後、コンカナノ リン Aセファロースにアプライし、界面活性剤含有緩衝液にて洗浄した 。これら素通り画分を ConA FTと称した。吸着画分はメチル -alpha-D-ダルコビラノシ ドと界面活性剤を含有した緩衝液で溶出した (C。nA EL)。 ConA FTは小麦胚ァダル チュンセファロースに再度アプライし、界面活性剤含有緩衝液にて洗浄した。これら 素通り画分を WGA FTと称した。吸着画分は N-ァセチルダルコサミンと界面活性剤を 含有した緩衝液で溶出した (WGA EL)。 ConA EL、 WGA EL、 WGA FTを分子同定 サンプルとした。これら画分を SDS-PAGEにて展開した後、実施例 2の手法によりタン パク質を検出し、短冊状に切ったゲルを in gel digestionして分析サンプルとした。
[0097] 実施例 4 LC/MSによる微量分析
LC/MS(Applied Biosystems社製 QTRAP)を用いて、実施例 3で得たサンプルを分析 した。 95%溶液 A (0.1%ギ酸)および 5%溶液 B (0.08%ギ酸、 80%ァセトニトリル)で平 衡化した PepMap逆相カラム(内径 0.075mm X長さ 150mm) (LCパッキングス社製)に 力け、 180ナノリットル/分の流速で、 5分洗浄後、 67.5分かけて溶液 Bの比率を 50%ま で直線的に上げることにより順次溶出し MSに導入した。 MSに導入されたサンプルは 次の繰り返しサイクルでデータ取得を行い、その配列情報を得ることとした。 1. Full M S Scan:m/z=0〜2000のレンジでの親イオンの分子量の観測。 2. Zoom MS Scan: Full MS Scanで同定された親イオンの価数の観測。 3. MS/MS Scan: Zoom MS Scanで測 定された分子に Heガスをあてた場合に生じる娘イオンの観測。理論的な b,yシリーズ とどれだけの本数がどれだけの強度でマッチングするかをスコア化してランクをつける 同定法(SEQUESTアルゴリズム)により同定した [アメリカン ·ソサイァティ ·フォ一 ·マス 'スぺタトロメトリー、 5卷、 976- 989頁(1994年)]。
[0098] 実飾 15 本膜分子の同定
実施例 4の手法により同定された分子のうち、新規分子である本膜分子が同定され た。同定対象となったフラグメントは m/z=1470.67の 2価イオンであり、その MS/MSの パターンは本膜分子の部分アミノ酸配列(27残基) DTMSTSNQLPWPTVDGSTDMV SSDLQK (配列番号 5)に非常に良く帰属された。これら部分配列の non-redundant D NA databaseに対するホモロジ一サーチの結果、これら部分配列を有する塩基配列 は他に存在しないことが明ら力となった。
[0099] 実施例 6 本膜分子の遣伝子クローユング
実施例 5で同定された部分アミノ酸配列(配列番号 5)をマウス遺伝子データベース に対してサーチを実施したところ、唯一 N W199221が完全に一致する部分配列を有 する遺伝子であることが判明した。そこで、実施例 5で明らかとなった部分アミノ酸配 列に対応する遺伝子配列をもとにプライマーを合成し、マウス成熟 DCの cDNAライブ ラリーを铸型として RT- PCRを実施した。プライマーはそれぞれ 30merおよび 40merで 、センスプライマー 5し ATGAGTAAGACACTCAGAGGCAACCTGGAC -3' (配列番 号 6)とアンチセンスプライマー 5し CTAAGGAGAAATGTCTTTAGCTAGAAGTTCA GAGTCCAAA -3' (配列番号 7)を用いた。この結果、約 800bp弱の本膜分子の遺伝 子断片を取得した。この遺伝子断片をプローブとしてコロニーハイブリダィゼーシヨン により本膜分子の遺伝子を含むクローンを選抜し、塩基配列を決定した。推定 open r eading frame構造を配列番号 1に、推定されるアミノ酸配列を配列番号 2に示す。得ら れた遺伝子配列は既知遺伝子として報告のある NM#199221と同じであるものの、報 告とは 1塩基欠失していることから、読み枠がずれており、途中力 アミノ酸配列が異 なっていることが明らかとなった。またヒト単球由来榭状細胞の cDNAライブラリーを铸 型として同様の手法でヒトオルソログを取得した。プライマーはそれぞれ 28merおよび 30merで、センスプライマー 5し ATGTGCAGAAGGTGCAAGCCAGAGCTCA -3' (配 列番号 8)とアンチセンスプライマー 5'- CTAAGTGGCCATGTCTTTAGTCAGAGG TTC -3' (配列番号 9)を用いた。推定 open reading frame構造を配列番号 3に、推定 されるアミノ酸配列を配列番号 4に示す。得られた遺伝子は N W174892に一致する。
[0100] 本膜分子の推定アミノ酸配列の一次構造について Kyteと Doolittleの方法 (J. Exp.
Med., 157卷、 105-132頁、 1982年)に従い、ハイド口パシープロット解析を行なった( 図 1)。その結果、本膜分子は N末端にシグナル配列を有する I型の細胞膜貫通タン ノ ク質であることが明ら力となった。本膜分子 (A)および (B)はそれぞれ (A) 209、 ( B) 201アミノ酸残基力もなり、ハイド口パシープロット解析結果から 17残基のシグナ ル配列、(A) 140残基、(B) 134残基の細胞外領域、 21残基の膜貫通領域、 (A) 3 1残基、(B) 29残基の細胞内領域を有していると予想される。さらにホモロジ一検索 並びにモチーフ検索の結果力 細胞外領域は (A) 2ケ所、(B) 0ケ所のァスパラギン
結合型糖鎖付加部位、 2ケ所のィムノグロブリン (Ig)ドメイン形成に必要なシスティン 残基を持つ 1つのィムノグロブリン様の構造を有していた。また、この細胞外の Igドメイ ンは V set構造であることが類推された。さらに、(A)、(B)ともに細胞内領域にシグナ ルを伝達し得るモチーフを有しないものの、(A)、(B)ともに膜貫通領域にリジン残基 を有することから、アダプター分子と会合していることが予想され、この会合したァダ プター分子を介してシグナルを伝達することが予想された。また、本膜分子は細胞外 に IgV setをひとつ有すること、細胞膜貫通領域にポジティブチャージを有するリジン 残基を有すること、などから構造的に TREM-1と相同ある 、は類似のファミリ一に属す ることが予想される。本マウス膜分子のマウス TREM-1に対するアミノ酸一致は 26%、 本ヒト膜分子のヒト TREM-1に対するアミノ酸一致は 26%、であり、 IgV setの形成に必 要なシスティン残基は保存されて 、る。
[0101] 施例 7 本蹬分早の可溶化型鉬.橼 体の作製
ノ、イド口パシープロット解析(図 2)の結果から、細胞外ドメインと考えられる領域とヒト IgGlの Fc領域を結合させた組換え体を作製した。なおヒ HgGlの Fc領域は各種細胞 表面上に存在する Fc gamma receptorへの結合を防ぐために 3ケ所のアミノ酸置換を 行った。具体的には細胞外領域として (A)の配列番号 1のアミノ酸 157位までを選択 した。この可溶化型組換え体のアミノ酸配列を配列番号 10に示す。 Fc領域でァミノ 酸置換を行った部位は配列番号 10に示す 177位 (Ala→Leu)、 178位(Glu→Leu)、 3 44位 (Asp→Val)〖こ相当する。この遺伝子を CMVプロモータ制御下 293T細胞で一過 性に発現させ、得られた培養上清カゝら Fc領域に結合性を有するプロテイン Aを結合 させた榭脂を用いたァフィ-テイク口マトグラフィにより発現産物を精製した(図 3)。

実施例 7に記載の組換え体を用いてマウス脾細胞上に本膜分子と結合する分子が 存在する力否かをフローサイトメーターを用いて検索した。具体的にはフィコエリスリ ン標識 F(ab')化抗ヒ HgG Fc領域抗体 (ベックマンコールター社製)を二次抗体として

該組換え体を認識させる。その結果、 CD3陽性 T細胞、 CD19陽性 B細胞、 NK1.1陽 性 NK細胞、 CD1 lc陽性榭状細胞など確認した全ての細胞にぉ ヽて本膜分子が結合 することが確認された。(図 4)
産業上の利用可能性
[0103] 本発明により、 DCを他の血球系細胞力 選択的に高い純度で分離することができ るのみならず、成熟 DCと未成熟 DCとの分離も可能とすることから、 DC療法への該細 胞の供給を可能とする。分離された DCは、抗原をパルスしたのち、再び患者に戻す ことにより癌ワクチンとして利用するなどの用途が期待される。 また、本膜分子に対す る抗体、あるいは本膜分子の可溶ィ匕型分子を用いて DCと T細胞との相互作用を阻害 することで免疫応答を制御し得ることも期待される。
