Abstract

 本発明は、アミノ酸の置換を有することにより非切断性となっている、CD40リガンドの非切断性機能的変異体、これをコードする核酸分子、この核酸分子を含むベクター、およびこのベクターで形質転換した細胞、ならびにこれらを含む医薬組成物に関する。また、本発明は、非切断性のCD40リガンド変異体を細胞上に発現させることにより、細胞の免疫反応性を改変する方法にも関する。

明 細 書
CD40リガンドの変異体およびその使用
技術分野
[0001] 本発明は、 CD40リガンド(CD40L)の非切断性変異体、これをコードする核酸分 子、該核酸分子を含むベクター、該ベクターによって形質転換された細胞に関する。 本発明はまた、該変異体により細胞の免疫反応性を改変する方法に関する。本発明 はさらに、 CD40陽性腫瘍、 CD40— CD40リガンド相互作用の異常に起因する疾 患、または CD40— CD40リガンド相互作用を強化することにより予防および Zもしく は治療が期待できる疾患を処置するための医薬、または、免疫増強剤、ならびにそ の製造方法にも関する。
背景技術
[0002] 動物の体内では、細胞同士が様々な方法で情報を伝達し合うことにより、協調して 種々の機能を担っている。中でも免疫系の細胞は局所で作用する機会が多ぐ細胞 間の情報伝達には細胞表面上に存在する種々のタンパク質分子を介した接触が重 要な役割を果たしている。力かるタンパク質分子の 1つに CD40L (CD154ともいう) 力 Sある。
CD40Lは、 TNFファミリーに属する約 39kDの II型膜タンパク質であり、活性化 CD 4+T細胞を始め、活性化 CD8+T細胞、好塩基球、肥満細胞、血小板などに発現し ている。 CD40Lは、 B細胞、マクロファージ、濾胞性樹状細胞などの抗原提示細胞、 上皮細胞、胸腺上皮細胞、血小板、線維芽細胞、および種々の癌細胞などに発現 する膜タンパク質である CD40と結合し、これを活性化する。
CD40Lの結合により活性化された CD40は、発現している細胞を活性化に導き、 B細胞においては、 CD23、 CD25、 CD80、 CD54、 MHCクラス IIの発現増強、凝 集の誘導、増殖の誘導、クラススィッチなどをもたらすことが、抗 CD40抗体を用いた 実験によって明らかにされている。また CD40陽性腫瘍細胞の一部 (膀胱癌、腎癌、 口腔癌など)では、 CD40Lの結合がアポトーシス誘導、細胞増殖抑制などをもたら すことが明らかになつている。 [0003] 一方、免疫系が腫瘍細胞を排除する能力を有することが分かっている。例えば、へ ルパー T細胞は、腫瘍細胞自身または抗原提示細胞との接触によって活性化される と、それ自身提示された腫瘍抗原を認識して腫瘍細胞と接触し Fas— FasL相互作 用によりアポトーシスに導くほか、 IL一 2、 IFN- yなどのサイト力インを放出すること により細胞傷害性 T細胞を活性化させ、当該細胞の腫瘍攻撃を補助する。しかしな がら、腫瘍細胞が免疫系による排除を回避する機構を有していることも分かっている 。かかる機構としては、例えば腫瘍細胞の免疫原性の欠如や、腫瘍細胞による免疫 系の抑制(免疫抑制分子の分泌、 T細胞シグナル伝達異常、癌細胞上の Fasリガンド 発現による T細胞アポトーシス誘導など)、腫瘍細胞上の免疫関連分子の欠失などが 考えられている。
[0004] 腫瘍免疫の要となる T細胞の活性化には、 T細胞レセプター (TCR)からの抗原刺 激の他に種々の補助刺激分子からの刺激が必要であることが明らかになつており、 T CRからの刺激のみを受けた場合、この T細胞は活性化しないば力りでなぐその後 同じ抗原刺激に不応答性となることが知られている。この機序は、免疫系構築の初期 段階で、自己抗原に対する免疫寛容を誘導するのに有用であるが、腫瘍に対する免 疫寛容をも誘導し得るため、腫瘍免疫には抑制的に作用する。力かる補助刺激分子 のうち、腫瘍免疫で最も重要なものの 1つに CD28があり、当該分子のリガンドは CD 80である。
[0005] CD40陽性の腫瘍細胞は、 CD80を発現していないことが多ぐしたがって MHCを 発現してレ、る場合には T細胞を不応答性にし、免疫系による排除を回避してレ、ると考 えられる。しかし、 CD40を刺激すると、力かる細胞も CD80を発現するので、免疫系 が認識できるようになる。このような効果を期待して、種々の可溶性 CD40Lまたは抗 CD40抗体を用いて CD40を刺激する試みがなされてきた力 これらの分子は細胞 膜上に発現される CD40リガンドほど有効ではなぐまた、肺に炎症反応を起こすな ど好ましくない生理的効果を惹起する可能性があるため(非特許文献 1参照)、臨床 的に使用できるものではな力 た。
[0006] かかる弊害を改善するために、 CD40Lをコードする遺伝子を腫瘍細胞に導入して 細胞表面上に発現させる試みがなされた (非特許文献 2、特許文献 1参照)。当該方 法によれば、細胞表面に発現した CD40Lが他の腫瘍細胞表面の CD40を次々と刺 激して CD80の発現を誘導するため、 CD80遺伝子を直接導入するより非常に効率 力 い。ところ力 上記の方法においては、ヒト腫瘍細胞にヒト CD40Lを導入しても in vivoでの発現および生物活性が十分ではなかった。そこで、ヒト CD40Lの代わりにマ ウス CD40Lを導入したところ、ヒト CD40リガンドを導入する方法に比べ比較的高レヽ 効果が認められた (非特許文献 2参照)。ヒト腫瘍細胞に導入したヒト CD40Lとマウス CD40Lとの間で力、かる違いが生じる原因は現在のところ不明である力 臨床試験で はマウス CD40リガンドを発現させる方法がとられ、ヒトの腫瘍治療においてある程度 の成果を収めている(非特許文献 3参照)。
[0007] し力、しながら、マウス CD40リガンドはヒトにとつて異種のタンパク質であり、抗原抗 体反応など望ましくない現象を惹起する恐れがあるとともに、動物実験においては、 上記方法を用いた場合、関節炎、浮腫、下痢などの炎症性病態の発症が認められて おり、ヒトにおいてもかかる副作用が生じる可能性があった。そこで、 CD40陽性腫瘍 の治療において、より副作用の危険の少ない方法が求められていた。
一方、非特許文献 4には、 111〜123位のアミノ酸を欠失させた非切断性のヒト CD 40L変異体を作製し、 CD40Lの可溶化体と膜結合型との機能的な違いを、 B細胞 の増殖能および IgE産生能について調査したところ、両者の間に実質的な違いが認 められな力 たことが記載されている。し力しながら、かかる変異体は、前記調査のた めに便宜的に作製されたに過ぎず、その応用やその他用途等については何ら言及 されていない。

発明が解決しょうとする課題
[0009] 本発明は、 CD40陽性腫瘍、 CD40— CD40L相互作用の異常に起因する病態、 および CD40— CD40リガンド相互作用を強化することにより予防および/もしくは治 療が期待できる疾患を処置するための新規な分子、および、細胞の免疫反応性を改 変するための新規な方法の提供を目的とする。
課題を解決するための手段
[0010] 本発明者らは、上記目的を達成すべく鋭意検討を行う中で、ヒト細胞へのヒト CD40 Lの導入によって所望の効果が得られないのは、遺伝子導入によって発現した該 CD 40L力 S、生体内の酵素であるマトリックス 'メタ口プロテアーゼ(MMP)によって切断さ れ、細胞から脱離して可溶化体となるために、細胞表面上の発現が低く抑えられてい ると考えた。そして、かかる着眼点を基に、 MMPに切断されず、かつ、 CD40刺激活 性を保持したヒト CD40Lの変異体の作製を試みたところ、 CD40L中の特定のァミノ 酸を変異させた変異体が、ヒト腫瘍細胞においても高い発現率を示すとともに、上記 のような炎症性の副作用も少なぐし力も優れた CD40刺激効果を奏することを見出 し、本発明を完成するに至った。
[0011] すなわち本発明は、アミノ酸の置換を有することにより非切断性となっており、 CD4 0刺激活性を有する、 CD40リガンドの非切断性変異体に関する。
また本発明は、アミノ酸の置換が、切断予想部位および/またはその近傍に位置す る上記変異体に関する。
さらに本発明は、 CD40リガンドがヒト由来である上記変異体に関する。 さらにまた本発明は、配列番号 1〜4のいずれかのアミノ酸配列を有する変異体、 および該アミノ酸配列に任意の変異を有するが、なお該変異体と同等の機能を有す る変異体からなる群から選択される上記変異体に関する。
本発明はまた、上記の変異体をコードする核酸配列またはその相補配列からなる 核酸分子、その断片、およびこれらの核酸分子にストリンジェントな条件でハイブリダ ィズする核酸分子からなる群から選択される、核酸分子に関する。 本発明はさらに、配列番号 7〜: 10のいずれかの核酸配列を有する核酸分子、およ び該核酸配列に任意の変異を有するが、なお該核酸分子がコードする変異体と同 等の機能を有する変異体をコードする核酸分子からなる群から選択される、上記核 酸分子に関する。

さらにまた本発明は、腫瘍が、白血病、リンパ腫、急性骨髄性白血病、急性骨髄単 球性白血病、急性リンパ球性白血病、慢性骨髄性白血病、慢性骨髄単球性白血病 、慢性リンパ球性白血病、多発性骨髄腫、黒色腫、膝臓癌、前立腺癌、頭頸部癌、 乳癌、肺癌、大腸癌、胃癌、食道癌、卵巣癌、扁平上皮癌、肝細胞癌、胆管癌、中 皮腫および類表皮癌からなる群から選択され、 CD40— CD40リガンド相互作用の 異常に起因する疾患が、全身性エリテマトーデス、慢性関節リウマチ、 自己免疫性血 管炎、ァテローム性動脈硬化症および X連鎖型高 IgM症候群からなる群から選択さ れ、 CD40— CD40リガンド相互作用を強化することにより予防および/または治療 が期待できる疾患が、ウィルス感染症、細菌感染症、真菌感染症、寄生虫感染症か らなる群より選択される、上記使用に関する。
発明の効果 [0016] 本発明の変異体を細胞表面に発現させることにより、 CD40Lの可溶化体への開裂 が抑えられ、可溶化体の産生が顕著に減少するとともに、細胞上にとどまる CD40L の量を実質的に増やすことができる。これにより、少ない副作用でより効果的に CD4 0陽性腫瘍、ならびに X連鎖型高 IgM症候群や全身性エリテマトーデス(SLE)、慢 性関節リウマチ、自己免疫性血管炎などの自己免疫性疾患病およびァテローム性動 脈硬化などの CD40— CD40L相互作用の異常に起因する疾患を治療することが可 能となり、力かる難病に苦しむ多くの患者を救うことができる。さらに、本発明の変異 体による CD40— CD40L相互作用の増強は、免疫力の強化をもたらすので、種々 の感染症などの予防、治療効果が期待される。また、本発明の変異体は MMPによ る切断を受けないため、 CD40Lが細胞膜に結合した状態で長期間存在することが 可能になることから、これまでに十分ではなかった、 CD40Lの膜結合形態および可 溶化形態の生体内での機能の解明に貢献することができる。

本発明において、「CD40リガンド(CD40L)」は、 CD40、またはその相同体もしく は断片(以下 CD40等という)に結合し得るポリペプチドを意味する。本発明における CD40Lは、好ましくは天然の CD40L、その相同体、変異体、およびそれらの断片 であり、特に好ましくは野生型のヒト CD40Lである。野生型 CD40Lのアミノ酸配列は 、例えば EMBL受託番号 CAA78737 (ヒト)、同 CAA46448 (マウス)として開示されてい る。野生型ヒト CD40Lのアミノ酸配列を配列番号 5として示す。本発明において、 CD 40Lは好ましくは細胞膜上に発現しており、より好ましくは CD40等との結合に関与 する部分が細胞外に存在するように細胞膜を貫通してレ、る。

[0025] 上記活性は、例えば、本発明の変異体の CD40への結合能によって評価すること ができる。かかる結合能は、例えば、野生型 CD40Lに対する抗体の該変異体への 結合の程度を、免疫沈降法、免疫細胞化学、フローサイトメトリー法などの当業者に 周知の技法を用いて決定することにより評価することができる。また、上記活性は、 C D40Lの CD40への結合によって発現が増強される分子の発現量を決定することに より評価することができる。かかる分子の例としては、 CD23、 CD25、 CD54、 CD80 、 CD83、 CD86、 Fas、 MHCクラス IIなどが挙げられる。当該発現量の決定方法とし ては、例えば、当該分子に特異的な抗体を用いた、免疫沈降法、免疫細胞化学、フ ローサイトメトリー法などの技法が挙げられる。上記活性はまた、細胞の増殖活性を 測定することによって評価することもできる。増殖活性は、例えば、 [H3]—ゥリジンま たは [H3]—チミジンの取込みの測定などによって容易に評価することができる。上記 活性は、さらに B細胞の IgEの産生誘導を測定することなどによっても評価できる。上 記評価方法は、当業者であればレ、ずれも容易に行うことができる。

[0071] 本発明はまた、本発明の変異体、核酸分子、ベクターおよび形質転換細胞からな る群から選択される成分を含む医薬組成物に関する。本発明の医薬組成物における 好ましい成分は、ベクターおよび/または形質転換細胞であるが、裸の核酸分子を含 むことができる。かかる成分は単独で使用してもよいが、一般には薬学的に許容でき る担体と共に使用される。そのような担体としては、水、生理食塩水、グルコース、ヒト アルブミン等の水性等張溶液が好ましい。更に、薬学的に通常使用される添加剤、 保存剤、防腐剤、香料、アジュバント等を添加することもできる。そのように調製した医 薬組成物は、治療すべき疾病に依存して適切な投与形態、投与経路によって投与 すること力 Sできる。
[0072] 投与形態としては、例えば、乳剤、シロップ剤、カプセル、錠剤、顆粒剤、注射剤、 インプラント、軟膏等が挙げられるが、これらに限定されない。注射剤やインプラントな ど、直接体内に注入または埋入する組成物は、好適には適切な滅菌処理を施す。本 発明の医薬組成物がベクターを含む場合は、通常成人一人当たり 1回に 108個のベ クタ一を、該組成物が形質転換細胞の場合は、通常成人一人当たり 1回に 109個の 細胞を投与するのがそれぞれ好ましいが、かかる用量は疾病の状態や標的細胞'組 織の性質によって適宜変更することができる。投与回数は、 1日 1回〜数回でよぐ投 与期間は 1日〜数ケ月以上にわたってもよぐ 1回〜数回の投与を 1クールとして、長 期にわたって断続的に複数のクールを投与してもよい。投与経路は、特に限定され ないが、非経腸投与が好ましぐこのうち特に好ましいのは、静脈内、筋肉内、動脈内 、皮下などへの注射、点滴注入、インプラントの埋入である。
[0073] 本発明の医薬組成物は、 CD40陽性腫瘍、または CD40— CD40リガンド相互作 用の異常に起因する疾患または CD40— CD40リガンド相互作用を強化することによ り予防、治療効果が期待される疾患の処置に好適に用いることができる。 CD40陽性 腫瘍の例としては、リンパ腫、 AML、 AMML、 ALL, CML、 CMML、 CLLなどの 白血病、多発性骨髄腫、黒色腫、勝臓癌、前立腺癌、頭頸部癌、乳癌、肺癌、大腸 癌、胃癌、食道癌、卵巣癌、扁平上皮癌、肝細胞癌、胆管癌、中皮腫および類表皮 癌などの腫瘍力 CD40— CD40リガンド相互作用の異常に起因する疾患の例とし ては、 X連鎖型高 IgM症候群、全身性エリテマトーデス(SLE)、慢性関節リウマチ、 自己免疫性血管炎などの自己免疫性疾患およびァテローム性動脈硬化などが挙げ られる。また、 CD40— CD40リガンド相互作用を強化することにより予防、治療効果 が期待される疾患の例としては、 HIVや HTLV_ 1、 CMVなどのウィルス感染症、 細菌感染症、真菌感染症、寄生虫感染症などの感染症などが挙げられる。
[0074] 本発明の医薬組成物は、 CD40— CD40リガンド相互作用の強化を介して免疫力 を増強することができるので、一般的な免疫増強剤として用いることもできる。例えば 、該医薬組成物を疾患に特異的なワクチンとともに対象に投与することにより、ヮクチ ンの効果を増強することが可能となる。本発明の医薬組成物の力かる使用は、免疫 機能が低下してレ、る対象、例えば AIDSなどの免疫機能の低下を伴う疾患に罹患し ている個体、老齢個体、被ストレス個体などにとりわけ有効である。
実施例
[0075] 本発明を、以下の実施例を用いてより具体的に説明するが、本発明はこれらの実 施例に限定されるものではない。
実施例 1 :野牛型ヒト CD40L (hCD40L)のクローニング
GenBank受託番号 L07414として登録された hCD40Lの cDNAの塩基配列情報を もとに、下記のプライマーを作製した。

ヒト末梢血から単核球をリンホセパール (IBL社製、カタログ番号 23010)に重層遠心 法で採取し、 RPMI1640培地 (シグマ社製)に 10%牛胎児血清 CIRH社製)を添カロ した培地にて、 OKT3抗体(ヤンセンファーマ社製、薬品コード 6399409A1029)ととも に 48時間インキュベーションし、活性化させた。該活性化細胞から定法に従い RNA を抽出し、これをテンプレートとして、以下の条件で RT—PCRを行った。

[0078] 実施例 2:発現ベクターへの hCD40Lのクローニング
プラスミド pCAccAngl (Yamauchi et al., J. Gene Med., 5(11):994- 1004, 2003参照) の EcoRI/Bglllサイトに上記で得た pGEM-T_hCD40Lの EcoRI/BamHI断片をクロー二 ングし、発現ベクター pCAcchCD40L (図 1中の「発現ベクター」に該当)を得た。すな わち、 50 μ 1の pGEM_T— hCD40Lを、 8 μ 1の 10 X NEBバッファー、 8 1の10 :63八 、 5 μ 1の EcoRI、 5 μ ΐの BamHIおよび 4 μ ΐの蒸留水とともに、および 2 μ 1 (2 μ gに丰目 当)の pCAccAnglを、 2 μ 1の 10 X NEBバッファー、 2 x l<D10 X BSA、 1 μ 1の EcoRI 、 Ι μ ΐの Bglllおよび 12 μ 1の 2回蒸留水とともに、それぞれインキュベートした後、 CI ΑΡで処理し、 hCD40Lの EcoRI/BamHI断片(約 800bp)および pCAccAnglの pCAcc/EcoRI/Bglll/CIAP断片(約 4. 7kb)を切り出し、それぞれ 10 μ 1の TEに溶解 した。そして、定法に従い、上記 EcoRI/BamHI断片を含む液 6 μ 1、 pCAcc/EcoRI/Bglll/CIAP断片を含む液 2 /i 1、 10 XT4DNAリガーゼバッファー 1 μ 1および T4DNAリガーゼ 1 μ 1 (合計 10 /i l)力 なる混合液を、 16°Cで 3時間インキュ ペートすることにより pCAcchCD40L (約 5. 5kb、図 1)を得た。得られたベクターを、製 造者のプロトコルに従レ、 TOP 10細胞に導入し、 EcoRI/BamHI断片が所望の位置お よび方向で挿入されたことを確認した。

本発明のヒト CD40L変異体の例である、 hCD40LMl〜4は、それぞれ配列番号 1〜4のアミノ酸配列を有し、野生型 hCD40Lと比較すると、 hCD40LMlは、 Glnll4 が Proに、 Lysl l5が Argに、 Glnl l8が Gluに、 Asnll9が Aspに、 hCD40LM2は、 Glnll4が Proに、 Lysl l5が Argに、 Aspl l7が Gluに、 Prol20が Serに、 hCD40LM3は 、 Glnll4が Proに、 Lysl l5が Argに、 Aspl l7が Gluに、 Glnll8が Gluに、 Asnl l9が Asp に、 Prol20が Serに、そして hCD40LM4は、 Glull2が Lysに、 Metll3が lieにそれぞ れ置換されている。上記変異体は、野生型 hCD40Lをコードする核酸分子に部位特 異的変異導入を行うことにより得た。変異導入部位は、表 1の通りである。

CAcchCD40L/ClaI (野生型)を、コスミド pWEAx_F/RGD (約 39401bp、 Hum. Gene Ther., 13(5): 613-626, 2002に記載)の Clalサイトにクローニングし、コスミド pAxCAhCD40L- F/RGDを得た。

Pro Natl. Acad. Sci. USA, 83: 8664-8668, 1986参照) を改変して作製し、 pAdexlの右端と左端を全長とし、左端は ntl〜357であり、右端の ファイバーの HIループには F/RGD変異が導入してある。本コスミドのバックボーンに はコスミド PWE15を用いており、 F/RGD変異(CDCRGDCFC)を含む HIループ部分( 166bp、配列番号 21)は、 pWEAx-F/RGDの場合と同様に、制限酵素サイト SacII、 Asel、 XhoI、 BamHI、 Mlul、 Sailと連なっている。発現カセットである Clal断片をコスミド の Clalサイトにクローユングした場合、 CAプロモーターの方向力 アデノウイルスゲノ ムに対して、右方向または左方向であるものが作られ得る力 本実施例においては、 すべて左方向のものを用いた(図 6参照)。

コスミド pAxCAhCD40L_F/RGDのアデノウイルスゲノムの右端を、 Padダイジェスト で切断した。また、変異型のコスミド pAxCAhCD40LMl、 pAxCAhCD40LM2、 pAxCAhCD40LM3、 PAxCAhCD40LM4, pAxCAhCD40LDlに関しては、アデノウィル スゲノムの左端と右端の両方を Paclダイジェストで切断した。得られた断片を、それぞ れ、 293細胞にトランスフエタトし、 Yoshida et al., Human Gene Therapy, 9(17): 2503-2515, 1998などに記載された通常の方法に従って、野生型の hCD40Lを有する 組み換えアデノウイルス AxCAhCD40L_F/RGD、ならびに、変異型の hCD40Lを持つ 組み換えアデノウイルス AxCAhCD40LMl、 AxCAhCD40LM2、 AxCAhCD40LM3、 AxCAhCD40LM4、 AxCAhCD40LDlをそれぞれ樹立した。ただし、本実施例では、 上記 Yoshida et al.に記載された DNA— TPCは使わず、コスミド DNAだけを 293糸田 胞にトランスフエタトして、アデノウイルスを作製した。

J. Cancer Res., 88: 296-305, 1997に記載さ れた pRxプラスミドから派生したものであり、ブラスチジン耐性マーカー遺伝子を有し ている。プラスミド pRxlbsrの模式図を図 7に示す。
野生型 hCD40L核酸を含むレトロウイルス発現プラスミド(pRxlhCD40L)は、 pGEM-T_hCD40Lから EcoRI/BamHIダイジェストで hCD40Lを切り出し、得られた hCD40Lの EcoRI/BamHIフラグメント、 pRxlbsrのサイトメガロウィルスェンハンサー C MVとレトロウイルスプロモーター dLTRを含む EcoRI/XhoIフラグメント(ntl〜2057、 2057bp)、および pRxlbsrの IRES-bsrを含む BamHI/XhoIフラグメント(nt2087〜6606、 4519bp)力 なる合計 3つのフラグメントをライゲーシヨンすることによって作製した。

PRxlhCD40LM3bsr)は、上記と同様に、実施例 3で作製したクローニングベクター pGEM_T-hCD40LM3_lから EcoRI/Avrllダイジェストで得た hCD40LM3の前半部分( hCD40LM3- 1/EcoRI/AvrII)、 pGEM-T-hCD40LM3_2から Avrll/BamHIダイジェスト で得た hCD40LM3の後半部分(hCD40LM3-2/Avrn/BamII)、 pRxlbsrのサイトメガロ ウィルスェンハンサー(CMV)とレトロウイルスプロモーター dLTRを含む EcoRI/XhoI フラグメント(ntl〜2057、 2057bp)、および、 pRxlbsrの IRES- bsrを含む BamHI/XhoIフ ラグメント(nt2087〜6606、 4519bp)の合計 4つの部分をライゲーシヨンすることによつ て作製した。
[0099] 組み換えレトロウイルスの作製と細胞株の樹立は、前記「遺伝子治療ハンドブック」 などに記載されている方法に従って行った。すなわち、 pRxlhCD40Lbsr、
pRxlhCD40LMlbsr、 pRxlhCD40LM2bsr、 pRxlhCD40LM3bsrおよび
pRxlhCD40LM4bsrをェコトロピックレトロウイルス産生細胞である BOSC23にそれぞ れリポフエクシヨンにより導入し、得られたェコトロッピックレトロウイルスを含む培養上 清を用いて、 ci CRIP— P131細胞を培養し、アンフォト口ピックレトロウイルスを産生 する φ CRIP— P131細胞を、野生型および変異型 hCD40Lのそれぞれについて得 た。さらに、得られた各 Φ CRIP— P131細胞の培養上清を用いてヒト神経膠腫細胞 U251を培養することにより、 U251細胞にアンフォト口ピックレトロウイルスを感染させ 、ブラストサイジンによる薬剤選択を行うことにより、 hCD40Lの野生型および 4種の 変異型をそれぞれ安定に発現する U251細胞の亜株を樹立し、それぞれ、
U251/CD40L, U251/CD40LM1, U251/CD40LM2, U251/CD40LM3および
U251/CD40LM4とした。
[0100] 実施例 11:野牛型および栾異型 hCD40Lの細朐膜上での発現
レトロウイルス感染により作製した野生型 hCD40Lおよび変異型 hCD40LMl〜4 を発現するトランスフエクタントが、細胞膜上にそれぞれの遺伝子産物を実際に発現 しているかを、フローサイトメトリー法にて確認した。すなわち、各トランスフヱクタントを 、選択薬剤ブラストサイジンの存在下で培養後、トリプシン— EDTA溶液 5mlにより 5 分間室温で処理し、培養フラスコより剥離、回収した。回収した細胞は、 10%牛胎児 血清加 DMEM培地を等量添加し、 1500卬 m、 5分の遠心により洗浄した。細胞ペレ ットを再び培地で撹拌後、 5 X 104個の細胞を各チューブに入れ、陰性コントロールと してフィコエリスリン (PE)標識マウス IgGl (BD社製)を、陽性コントロールとして PE標 識抗ヒト CD40L抗体 TRAP— 1 (BD社製)をそれぞれ 10 μ 1添カ卩し、 4°Cで 45分間 反応させた。 3回遠心洗浄後、リン酸緩衝食塩水 PBSにヨウ化プロピジゥム lOOngを 添加し、フローサイトメーター(BD社製)にて解析した。その結果、図 8に示すように、 野生型ならびに 4種の変異型トランスフヱクタントの細胞膜上に機能的な hCD40Lの 発現を確認した。
[0101] 実施例 12:栾異型 hCD40Lによる可溶化体産牛の定量
実施例 10で作製したトランスフエクタント(U251/CD40L、 U251/CD40LM1, U251/CD40LM2, U251/CD40LM3および U251/CD40LM4)が可溶化体 hCD40Lを 産生するか否かについて解析した。各トランスフエクタントを 3 X 104個/ mlまたは 1 X 104個/ mlの細胞濃度で、 2日間 37°Cで培養した。細胞培養液を回収し 2000卬 mで 10分間遠心した後、 0. 45 μ πιのフィルターで濾過し、培養上清とした。
U251/CD40L, U251/CD40LM1および U251/CD40LM2については、 3 β Μの ΜΜΡ 阻害剤(カルビオケム社製、 GM 6001)または同量の DMSOのいずれかを添加し、 可溶化体産生に与える ΜΜΡの影響を評価した。
[0102] 可溶化体 hCD40Lの定量は Kato et al., J. Clin. Invest., 104: 947-955, 1999に記 載されたサンドイッチ ELISA法にて測定した。まず、 ELISA用 96穴プレート (Coster 社製)に、 5 μ g/mlの抗ヒト CD40L抗体 TRAP— 1を 50 μ 1ずつ添加し、 4°Cで 16 時間反応させた。 5%FCS加 PBSを 200 μ 1ずつ添加してブロッキングを行った後、 培養上清サンプノレを 100 μ ΐずつ添カ卩した。陽性コントロールとして、リコンビナント h CD40Lタンパク質 (Alexis社製)を lOngZmlの濃度を開始点として 2倍希釈液を順 次作製し 100 μ ΐずつ添加した。室温で 2時間反応後、非付着成分を 0. 05%Twee n加 PBSで 3回洗浄し、 2 μ § 1111のビォチン標識抗ヒト〇040し抗体810_ 24_ 31 ( eBioscience社製)を ΙΟΟ μ Ιずつ添加し、室温で 1時間反応させた。 0. 05%Tween カロ PBSで 4回洗浄後、西洋ヮサビペルォキシダーゼ(HRP)標識ストレプトアビジン (Amersham社製) 500倍希釈液を 50 μ 1ずつ添加し、室温で 1時間反応させた。 [0103] 0. 05%Tween加 PBSで 5回洗浄後、ペルォキシダーゼ発色基質溶液 (BD社製) を 100 /i lずつ添カ卩し、室温で 10分間反応させ、 2N硫酸溶液で発色反応を停止し た。発色溶液の吸光度 450nmと対照吸光度 570nmを 96穴プレートリーダーで測定 し、数値化した。測定値はコンピュータ一にて解析し、標準タンパク質の吸光度から 標準曲線および近似式を計算し、培養上清サンプノレの吸光度の値から可溶化体 hC D40L値を計算した。その結果を図 9と図 10に示す。野生型 hCD40Lのトランスフエ クタントから産生される可溶化体量に比べて、 4種の変異体のトランスフヱクタントでは 、いずれも可溶化体の産生が低下していた。また、野生型 hCD40Lのトランスフヱク タントから産生される可溶化体の量は MMP阻害剤によって顕著に減少した。
[0104] 実施例 13:野牛型および栾異型 hCD40Lの CD40刺激活件
実施例 2、 4または 7でクローユングした野生型核酸分子 hCD40L、置換型核酸分子 hCD40LM3または欠失型核酸分子 hCD40LDlを、それぞれレトロウイルスベクターを 用いて U251細胞に遺伝子導入し、ブラストサイジン薬剤存在下で 2週間培養し耐性 株を選択した。これらの細胞をさらに抗 CD40L抗体と抗マウス IgG抗体結合磁気ビ ーズを室温で 1時間反応させ、 AutoMACS (ミルテニ一社製)を用いて CD40L陽性 細胞を選択分離した。各遺伝子導入細胞を培養用 6穴プレートに 1 X 104個培養後、 ヒト B細胞性白血病細胞(B— CLL)または B細胞性リンパ腫細胞(Ramos)を 5 X 104 個添加し 37°Cで 2日間培養した。培養後、白血病細胞またはリンパ腫細胞をそれぞ れ回収し FITC標識抗 CD95抗体、 FITC標識抗 CD54抗体、 PE標識抗 CD83抗 体、 PE標識抗 CD80抗体(いずれも BD社製)を各 5ml添加し 4°Cで 1時間反応後、 FACSにて抗原の発現量を解析した。その結果、図 11および 12に示すとおり、野生 型 CD40Lと置換型 CD40LM3はともに各抗原の発現を同程度誘導した力 欠失型 CD40LD1の発現誘導効果は前二者に比べて低ぐ生物活性が弱まることが確認さ れた。
[0105] 実施例 14 :野牛型および栾異型 hCD40Lのトランスフエクタントの牛体への移入およ びその畺 ,
野生型および変異型 hCD40Lの生体内での分布および各組織に与える影響を調 ベた。野生型 hCD40Lトランスフヱクタント (U251CD40L)または変異型 hCD40Lトラ ンスフエタタント (U251CD40LM3)を、培養フラスコからトリプシン一 EDTA溶液を用い て回収し、遠心洗浄後、 10万個の細胞をマトリジエル(BD社製) 100 /i 1に懸濁した。
BALB/cヌードマウス腹側部皮下に、 1 X 105個の各トランスフエクタントを 26G針付 注射筒を用いて移植した。
移植トランスフエクタントの生着 (腫瘤)は 1週間後に確認された。図 13に示すとおり 、移植 21日後に、野生型 hCD40Lトランスフエクタント移植マウスにおいて全身性の 血管浮腫(ェデ一マ)が認められたが、変異型 hCD40Lトランスフヱクタント移植マウ スでは認められなかった。
また、図 14に示すように、移植 28日後にぉレヽて、野生型 hCD40Lトランスフエクタ ント移植マウスの前肢および後肢に関節炎の所見が認められたが、変異型 hCD40L トランスフエクタント移植マウスでは認められなかった。
さらに、移植 28日後には、野生型 hCD40Lトランスフエクタント移植マウスにおいて 軽度の下痢症状が確認された力 変異型 hCD40Lトランスフエクタント移植マウスで は認められなかった。
[0106] 移植 35日後に、野生型 hCD40Lトランスフエクタント移植マウスおよび変異型 hCD 40Lトランスフエクタント移植マウスの肝臓、腎臓の組織学的解析を行った。深麻酔 下にて安楽死した移植マウスから肝臓および腎臓を摘出し、 4%パラホルムアルデヒ ドリン酸緩衝液(和光純薬製)中に浸潤させ、 4°Cで 24時間、組織固定を行った。各 固定組織はパラフィン包坦後、薄利切片を作製し、へマトキシリンーェォジン染色を 行った。その結果、図 15に示すように、野生型 hCD40Lトランスフエクタント移植マウ スでは、肝臓実質内および胆管周縁部に多数の炎症細胞の浸潤を確認できた(図 中の円で囲んだ部分を参照)。これに対して、変異型 hCD40Lトランスフヱクタント移 植マウスでは、図 16に示すように、明らかな炎症細胞の浸潤を認めなかった。腎臓に おいても糸球体の萎縮.損傷が野生型 hCD40Lトランスフヱクタント移植マウスでは 認められたが、変異型 hCD40Lトランスフエクタント移植マウスでは認められな力 た
[0107] 移植 35日後の野生型 hCD40Lトランスフエクタント移植マウスおよび変異型 hCD4 0Lトランスフエクタント移植マウスの末梢血中の可溶化体 hCD40L値を ELISAにて 測定した。各移植マウスの末梢血をエツペンドルフチューブに採取後、 lOOOOrpm で 5分間遠心し、その上清を血漿成分として回収した。血漿を 3倍に希釈後、実施例 12に記載の方法に従って可溶化体 hCD40L値を測定した。その結果、図 17に示す ように、野生型 hCD40Lトランスフエクタント移植マウスの血中可溶化体 hCD40L値 は 840pg/mlであった。これに対して正常非移植マウスおよび変異型 hCD40Lトラ ンスフヱクタント移植マウスの可溶化体 hCD40L値は検出限界値(20pgZml)以下 であった。
実施例 15:アデノウイルスベクターによる野牛型または栾異型 hCD40L遺伝子の導 A
実施例 9で作製したアデノウイルスベクター AxCAhCD40L-F/RGD、
AxCAhCD40LM3および AxCAhCD40LM4を用いて U251細胞に遺伝子導入を行レヽ 、細胞膜上での発現と可溶化体の産生を解析した。 U251細胞 1 X 105個に対して 各アデノウイスルベクター溶液を添加して 3日間培養した。アデノウイルス量は細胞 1 個あたり 10および 100粒子ユニット (pu)加えた。遺伝子導入細胞は培養上精液を 回収した後、 EDTA溶液で細胞を回収し、 3回遠心洗浄後、 CD40Lの発現をフロー サイトメトリー法で解析した。その結果、図 18に示すように、野生型、変異型ともに膜 上に CD40Lの発現が確認された。また図 19に示すように、培養上清中に産生され る可溶化体も、変異型の CD40LM3および CD40LM4で低下しており、これはレト ロウィルスベクターで遺伝子導入した場合と同様であった(図 9〜: 10参照)。

Claims

請求の範囲
[I] アミノ酸の置換を有することにより非切断性となっており、 CD40刺激活性を有する 、 CD40リガンドの非切断性変異体。
[2] アミノ酸の置換が、切断予想部位および/またはその近傍に位置する、請求項 1に 記載の変異体。
[3] CD40リガンドがヒト由来である、請求項 1または 2に記載の変異体。
[4] 配列番号:!〜 4のいずれかのアミノ酸配列を有する変異体、および該アミノ酸配列 に任意の変異を有するが、なお該変異体と同等の機能を有する変異体力 なる群か ら選択される、請求項:!〜 3のいずれかに記載の変異体。
[5] 請求項 1〜4のいずれかに記載の変異体をコードする核酸配列またはその相補配 列からなる核酸分子、その断片、およびこれらの核酸分子にストリンジェントな条件で ハイブリダィズする核酸分子からなる群から選択される、核酸分子。
[6] 配列番号 7〜: 10のいずれかの核酸配列を有する核酸分子、および該核酸配列に 任意の変異を有するが、なお該核酸分子がコードする変異体と同等の機能を有する 変異体をコードする核酸分子からなる群から選択される、請求項 5に記載の核酸分 子。
[7] 請求項 5または 6に記載の核酸分子を含むベクター。
[8] ウィルスベクターである、請求項 7に記載のベクター。
[9] ウィルスベクターがレトロウイルス、アデノウイルス、アデノ随伴ウィルス、ヘルぺスゥ イノレス、レンチウィルスおよびバキュロウィルスからなる群力、ら選択される請求項 8に 記載のベクター。
[10] ウィルスファイバーに RGD配列を含む、請求項 8または 9に記載のベクター。

腫瘍が、白血病、リンパ腫、急性骨髄性白血病、急性骨髄単球性白血病、急性リン パ球性白血病、慢性骨髄性白血病、慢性骨髄単球性白血病、慢性リンパ球性白血 病、多発性骨髄腫、黒色腫、瞎臓癌、前立腺癌、頭類部癌、乳癌、肺癌、大腸癌、 胃癌、食道癌、卵巣癌、扁平上皮癌、肝細胞癌、胆管癌、中皮腫および類表皮癌か らなる群から選択され、 CD40— CD40リガンド相互作用の異常に起因する疾患が、 全身性エリテマトーデス、慢性関節リウマチ、 自己免疫性血管炎、ァテローム性動脈 硬化症および X連鎖型高 IgM症候群からなる群から選択され、 CD40— CD40リガ ンド相互作用を強化することにより予防および Zまたは治療が期待できる疾患が、ゥ ィルス感染症、細菌感染症、真菌感染症、寄生虫感染症からなる群より選択される、 請求項 27に記載の使用。
