Abstract

L'invention concerne, dans un aspect, une bibliothèque (de codage) d'ARNi aléatoires ou semi-aléatoires. Dans un autre aspect, elle concerne des méthodes de construction de bibliothèques (de codage) d'ARNi aléatoires ou semi-aléatoires. Dans un autre aspect, l'invention concerne des systèmes de vecteurs utilisables dans la construction de bibliothèques d'ARNi et/ou pouvant exprimer des ARNi individuels ou des bibliothèques d'ARNi, tant de manière constitutive que de façon inductible. Dans un autre aspect, l'invention concerne une méthode d'utilisation d'une bibliothèque d'ARNi. La bibliothèque d'ARNi est introduite dans la population de cellules. La population de cellules est ensuite soumise à un processus de sélection d'une sous-population de cellules présentant une propriété comportementale, biochimique, chimique, fonctionnelle, moléculaire, morphologique, phénotypique ou physique différente de celle du reste de la population. A l'issue du processus de sélection, la sous-population de cellules peut être isolée, analysée et/ou clonée à loisir. Une telle analyse de la sous-population peut consister à identifier et séquencer les espèces d'ARNi responsables pour les différentes propriétés de la sous-population relativement au reste de la population. En variante, la sous-population peut subséquemment être analysée grâce à des dosages génomiques, protéomiques et/ou cellomiques. Lorsque de tels dosages génomiques, protéomiques et/ou cellomiques sont mis en oeuvre, la méthode de l'invention peut produire plusieurs produits bio-informatiques utiles. Des ARNi spécifiques identifiés par ce processus peuvent présenter un intérêt thérapeutique direct.
