Abstract

L'invention porte sur des amidases, acides nucléiques codant pour elles et les procédés d'obtention et d'utilisation de ces polynucléotides and polypeptides. Selon un aspect l'invention porte sur des enzymes présentant une activité secondaire d'amidase c.-à-d. l'hydrolyse des amides, y compris des enzymes à activité de peptidase, de protéase et/ou d'hydantoinase. Selon un autre aspect les enzymes de l'invention peuvent servir à améliorer le goût d'aliments (par exemple fromage mûri par enzymes), à favoriser l'élimination de bactéries et champignons, à modifier et déprotéger des intermédiaires chimiques fins, à synthétiser des liaisons peptide, à exécuter des résolutions chirales, à hydrolyser la Céphalosporine C. Les enzymes de l'invention peuvent servir à produire de l'acide7-aminocéphalosporanique (7-ACA) et des antibiotiques semi-synthétiques de Céphalosporine dont la caphalothine, la céphaloridine et la céfuroxime. Les enzymes de l'invention peuvent en outre servir d'agents antimicrobiens, par exemple d'agents hydrolytiques de la paroi cellulaire. L'invention porte également sur un substrat fluorescent d'amidase constitué de 7-(E-D-2-aminoadipoylamido)-4-méthylcoumarine.
