Abstract

A method of site-specifically modifying in a cell a target genomic DNA sequence consisting of first and second DNA strands to introduce a desired alteration, which desired alteration is of a single base-pair or a few base-pairs and is base-pair insertion, deletion, substitution or any combination thereof, the method comprising introducing into the cell a single-stranded targeting DNA molecule, wherein the targeting DNA molecule is 10-100 nucleotides in length and is of nucleotide sequence identical to nucleotide sequence within the first or second DNA strand of the target genomic DNA sequence except for the desired alteration and optionally one or more additional differences, wherein the differences are each insertion, substitution or deletion of a nucleotide, resulting in site-specific modification of the target genomic DNA to introduce the desired alteration.
