Abstract

La présente invention concerne des compositions et méthodes pour la modification de gènes dans des cellules, et leurs utilisations. Elle concerne notamment des vecteurs comprenant a) une cassette encadrée par des séquences homologues à un gène cible ou par des sites de clonage destinés à l'insertion desdites séquences homologues, ladite cassette comprenant en 5' une unité bicistronique comprenant un gène de sélection positive et un gène marqueur liés fonctionnellement par un élément IRES, lesdits gènes étant dépourvus de promoteur, en aval de ladite unité bicistronique, une unité comprenant un gène de sélection négative lié à un promoteur et dont l'orientation transcriptionnelle est identique ou opposée à celle de l'unité bicistronique, et, des sites de recombinaison site-spécifique encadrant l'unité bicistronique et l'unité comprenant le gène de sélection négative lié à un promoteur, et b), un gène toxique situé en aval de ladite cassette et pourvu d'un promoteur.
