Abstract

Die Erfindung betrifft ein Verfahren zur enzymatischen Reduktion von Substraten mit molekularem Wasserstoff. Erfindungsgemäss wird ein Mediator mittels einer Hydrogenase mit Wasserstoff umgesetzt. Im besonderen kann dieses Verfahren angewandt werden, um chemische Mediatoren oder auch Cofactoren von Enzymen zu reduzieren. Haupteinsatzpunkt dieses Verfahrens liegt in der Reduktion von NADP+ zu NADPH bzw. Einsatz des im Patent beschriebenen Enzyms zur Cofaktorregenerierung von NADPH mit molekularem Wasserstoff. Vorteil dieses Verfahrens, gegenüber allen etablierten Verfahren zur Regenerierung von NADPH, liegt in der Verwendung des sehr kostengünstigen molekularen Wasserstoffs zur Generierung bzw. Regenerierung des Cofaktors und damit nicht der Bildung eines noch weiter abzutrennenden Nebenproduktes. In diesem Fall wird Wasser als Nebenprodukt gebildet. Als Katalysator Können verschiedene Aufreinigungen und Präparationen des Enzyms Pyrococcus furiosus eingesetzt werden.
