Abstract

L'invention concerne un procédé de construction d'une banque d'ADN consistant: (a) à ligaturer des ADN à domaine unitaire en combinaisons arbitraires; (b) à mélanger les ADN ainsi obtenus de sorte que la séquence terminale de chacun de ces ADN utilisés se chevauche avec la séquence terminale d'au moins un autre type d'ADN à domaine unitaire; puis, (c) à réaliser une amplification en chaîne de la polymérase en utilisant le mélange d'ADN à domaine unitaire ligaturés comme modèle, de manière à obtenir une banque d'ADN comprenant au moins deux clones. La présente invention concerne également un procédé permettant de construire une masse de gènes, consistant à ligaturer ensemble dans des ordres différents et à des longueurs variables une pluralité de fragments d'ADN codant pour des séquences d'acides aminés arbitraires.
