Abstract

Die Effizienz einer Chemotherapie maligner Erkrankungen ist oft durch Resistenzen gegenüber der eingesetzten Zytostatika limitiert, die durch eine Vielzahl unterschiedlicher, parallel oder sequentiell auftretender Mechanismen vermittelt werden. Gegenstand der Erfindung ist die Verwendung eines Verfahrens zur Erfassung von Resistenz-Profilen mittels RNA aus Geweben bzw. Zelllinien mit der real time-RT-PCR-Technologie (durchführbar z.B. am Light Cycler, Roche Diagnostics GmbH). Es können so die Expressionen unterschiedlicher Gene, die an der Entstehung oder Verstärkung oder Verminderung von Resistenzen beteiligt sind, quantitativ nachgewiesen werden. Basierend darauf können dann z.B. Patienten-individuelle Resistenz-Profile erstellt werden, die die molekularbiologische Grundlage zur Auswahl geeigneter Zytostatika vor und auch während der jeweiligen Tumorchemotherapie bilden. Darüber hinaus können prognostisch die Erfolgschancen (Response) bestimmter chemotherapeutischer Regime abgeschätzt werden.
