Abstract

Gegenstand der Erfindung ist ein Verfahren zur Identifizierung und Isolierung von Genomfragmenten mit Kopplungsungleichgewicht, wobei man bei nicht miteinander verwandten sowie bei miteinander verwandten Individuen Bereiche des Genoms isoliert, welche Kandidatengenbereiche enthalten, die sich im Kopplungsungleichgewicht mit ihrer engeren DNA-Umgebung befinden. Die im Vergleich zu anderen Verfahren frühere Durchführung des Klonierungsschritts ermöglicht eine mengenunabhängige Gewinnung der DNA-Fragmente und ersetzt weiterhin den in anderen Verfahren durchgeführten Methylierungsschritt. Außerdem wurde gefunden, dass die erfindungsgemäße Verwendung eines Pflanzenenzym eine weitaus höhere Spezifität des Verfahrens gegenüber solchen bewirkt, bei denen der Mut S,H,L-Komplex verwendet wird.
