Abstract

Beschrieben werden Expressionsvektoren zur gewebespezifischen Expression eines Gens, das für ein cytotoxisches oder 'Prodrug'-aktivierendes Protein codiert, wobei der Expressionsvektor folgende DNA-Sequenzen funktionell verknüpft umfaßt: a) ein oder mehrere gewebespezifische Enhancerelemente, beispielsweise den murinen oder humanen Tyrosinase-Enhancer; b) einen gewebespezifischen Promotor, beispielsweise den Tyrosinase-Promotor oder den 'Melanoma Inhibitory Activity' (MIA)-Promotor; und c) eine für ein cytotoxisches oder 'Prodrug'-aktivierendes Protein, beispielsweise die A-Kette des Diphterietoxins oder Herpes-Simplex-Virus Thymidinkinase (HSV-TK), codierende DNA-Sequenz. Diese Vektoren eignen sich zur Behandlung von Tumoren, beispielsweise des malignen Melanoms.
